Redressing clothing in the Hebrew Bible : material-cultural approachesWagstaff, Bethany Joy January 2017 (has links)
Despite the dynamic portrayal of clothes in the Hebrew Bible scholars continue to interpret them as flat and inert objects. They are often overlooked or reduced to background details in the biblical texts. However, this thesis will demonstrate that the biblical writers’ depictions of clothes are not incidental and should not be reduced to such depictions. This thesis employs a multidisciplinary approach to develop and challenge existing approaches to the clothing imagery in the Hebrew Bible. It will fall into two main parts. In the first part, I draw insights from material-cultural theories to reconfigure ways of thinking about clothing as material objects, and reassessing the relationships between people and objects. Having challenged some of the broader conceptions of clothing, I will turn to interrogate the material and visual evidence for clothing and textiles from ancient Syro- Palestinian and ancient West Asian cultures to construct a perspective of the social and material impact of clothing in the culture in which the biblical texts were constructed and formed. In the second part, I will examine the biblical writers’ depiction of clothing through two case studies: Joseph’s ketonet passim (Genesis 37) and Elijah’s adderet (1 Kings 19 and 2 Kings 2). These analyses will draw from the insights made in the first part of this thesis to reassess and challenge the conventional scholarly interpretations of clothing in these texts. In this thesis, I argue that clothes are employed in powerful ways as material objects which construct and develop the social, religious and material dimensions of the text. They are also intimately entangled in relationships with the characters portrayed by the biblical writers and can even be considered as extensions of the people with whom they are engaged. Clothes manifest their own agency and power, which can transform other persons and objects through their performance and movement in a biblical text.

Molecular Insights into Lymphoid Malignancy : Role of Transcription Factor BCL11B in T-cell Leukemia Genesis and Biochemical Characterization of DNA Binding Domain of RAG1Deepthi, R January 2017 (has links) (PDF)
The lymphoid tissues consist of distinct cell subpopulations of B and T cell lineages and possess complex signaling pathways that are controlled by a myriad of molecular interactions. During the fine-tuned developmental process of the lymphoid system, inappropriate activation of oncogenes and loss of tumor suppressor gene activity can push lymphocytes into uncontrolled clonal expansion, causing several lymphoid malignancies. V(D)J recombination is one such essential process, important for the proper development of the mammalian immune system. However, mistakes in normal V(D)J recombination can lead to deletion of tumor suppressor genes or activation of proto-oncogenes. In the first part of the study, the physiological and pathological roles of DNA binding domain of RAG1 have been characterized.
RAG (Recombination Activating Gene) complex consisting of RAG1 and RAG2, is a site specific endonuclease responsible for the generation of antigen receptor diversity. It cleaves a specific DNA sequence termed as recombination signal sequence (RSS), comprising of a conserved heptamer and nonamer. Recent studies have shown that RAGs can also act as a structure-specific nuclease by cleaving flaps, heterologous loops, bubbles, hairpins etc. Nonamer binding domain (NBD) of RAG1 plays a central role in the recognition of RSS during its sequence specific activity. To investigate its DNA binding properties, NBD of murine RAG1 was cloned, overexpressed and purified from E. coli. Electrophoretic mobility shift assays showed that NBD binds with high affinity to nonamer in the context of 12/23 RSS. However, it did not bind to heteroduplex DNA, irrespective of the sequence of the single-stranded region. Interestingly, when a nonamer was present next to a heteroduplex DNA, NBD exhibited robust binding. NBD binding was specific to thymines when single stranded DNA containing poly A, C, G and T were used. Biolayer interferometry studies showed that the observed poly T binding to NBD was robust with a binding constant of 0.45±0.16 µM. >23 nt was essential for NBD binding at homothymidine stretches. On a double-stranded DNA, NBD could bind to A:T stretches, but not G:C stretches or random sequences. Although NBD is indispensable for sequence-specific activity of RAGs, external supplementation of purified nonamer binding domain to NBD deleted cRAG1/cRAG2 did not restore the sequence specific activity, suggesting that the overall domain architecture of RAG1 is important for maintaining its properties. Therefore, we define the sequence requirements of NBD binding to double- and single-stranded DNA, which will have implications in generation of chromosomal rearrangement and genomic instability in lymphoid cells.
Genetic alterations are one of the hallmarks of lymphoid malignancies. Many genes involved in chromosomal abnormalities are known to play central roles in the development of normal lymphocytes. In the second part of the study, molecular mechanism associated with fragility of the transcription factor, B cell leukemia 11B (BCL11B) that drives malignant transformation of T-cells has been studied. BCL11B is a zinc finger protein transcription factor with multiple functions. It plays a key role in both development and subsequent maintenance of T-cells. BCL11B gene alterations are implicated in a number of diseases including T-cell malignancies. It acts as a haplo-insufficient tumor suppressor and loss of BCL11B allele leads to susceptibility to mouse thymic lymphoma and human T-ALL. Recent studies reveal heterozygous BCL11B mutations and deletions across each of the major molecular subtypes of T-ALL (15% of patients). Most of the BCL11B missense mutations identified so far affected the residues within BCL11B zinc finger domains of the exon 4. However, mechanism of generation of such specific mutations leading to altered functions of BCL11B remains to be explored.
In the present study, we address the potential mechanism of fragility of BCL11B gene during leukemia genesis. Firstly, we have evaluated different regions of BCL11B gene for presence of non-B DNA sequence motifs. Studies using non-B DB database reveal clustering of several non-B DNA forming motifs at the region spanning exon 4 of BCL11B gene. In order to biochemically evaluate the potential of non-B DNA structure formation, two different regions of exon 4 were PCR amplified and cloned. Using bisulfite modification assay we demonstrate that, single strandedness exists at both region I and II of BCL11B exon 4, when the region is present on a plasmid DNA. Bisulfite reactivity on chromosomal DNA confirmed existence of such altered DNA structures in the context of human genome. In vitro gel shift assays showed formation of both intra and intermolecular G-quadruplexes. Primer extension studies revealed that non-B DNA structures could block polymerization during replication on a plasmid, leading to DNA replication arrest. Extrachromosomal assays showed that non-B DNA structure motifs, in contrast to its mutants, blocked transcription leading to reduced expression of green fluorescent protein (GFP) within cells.
Many non-B DNA-forming sequences have been mapped to regions of common chromosomal breakpoints in human tumors, known as “hotspots”, which are associated with leukemia, lymphomas and genomic disorders. Thus, alternative DNA conformations are believed to contribute to mutations, deletions and other genetic instability, leading to the deregulation of cancer-related genes in malignant diseases such as leukemia and lymphoma. Activation induced cytidine deaminase (AID), is an essential enzyme involved in antibody diversification of immunoglobulin genes. However, aberrant AID expression in B- cell and non-B cell background is reported in various cancers including leukemia and lymphoma. AID activity requires single stranded DNA (ssDNA) as a substrate. Since activation induced cytidine deaminase (AID) deaminates cytosines when present on a single stranded DNA and its expression is deregulated in many cancers, we investigated the role of AID in BCL11B gene mutagenesis. We observed substantial AID expression in many T-cell leukemic cell lines. Thus, we hypothesize that AID might be targeted to single stranded DNA present at BCL11B exon 4 due to formation of non-B DNA structures such as G-quadruplexes causing AID mediated deamination, further leading to nucleotide alterations and the mutational signature observed at BCL11B exon 4 resulting in T-ALL. Based on our findings, we propose that single strandedness resulted due to formation of non-B DNA structures such as G-quadruplex DNA, triplex DNA or cruciform DNA during physiological processes like DNA replication and transcription at exon 4 of BCL11B, can act as the target for AID. Thus, our findings uncover a new possible link between non-B DNA structure motifs and AID expression in causing mutations at BCL11B exon 4 which could lead to T cell leukemia genesis.
BCL11B is a bifunctional transcriptional regulator that can act as a repressor and transactivator, and is known to differentially control the expression of specific genes in a context-dependent manner. In order to understand the transcriptional network involving BCL11B, it was cloned, overexpressed and purified from E. coli. To investigate the DNA binding properties of BCL11B protein, electrophoretic mobility shift assays were performed. Our results lead to identification of a specific sequence motif that is responsible for DNA binding. Competition experiments in presence of specific and nonspecific oligomers further confirmed the binding specificity.
Thus, in the present study, we have characterized the binding properties of nonamer binding domain of RAG1, emphasizing its pathological relevance in causing genomic instability in lymphoid cells. The study may help in better understanding of RAG induced genomic instability in lymphoid tissues and role of aberrant AID expression in inducing mutations at BCL11B Zinc finger domain, leading to its deregulation and culminating into T-cell leukemia

Previous issue date: 2013-04-22 / The thesis presents interpretations of Augustine of Hippo with regard to the first three chapters of genesis, the first book of the Bible. These interpretation are found in the works Literal Commentary on Genesis, concerning Genesis Against the Manicheans, Literal Commentary on Genesis, Unfinished, Books Xi-XIII of the Confessions, as well a brief exposition in Books XI-XIV of the City of God. Exposition of these Augustinian Commentaries seeks to demonstrate various interpretations made by author in one group of texts revealing a hermeneutics centered in the interpreter and not in established interpretive rules. In sequence there is described succinctly the evolution of textual hermeneutics during the modern period up to the reflections conducted by Martin Heidegger in the first half of the twenties. Based in Heidegger s existential commentaries on the Pauline Epistles (Galatians and I-II Thessalonians) and on the tenth Book of the Confessions, there is shown a return to a interpreter-based hermeneutics, such as practiced by Augustine. The concern manifested by Heidegger with regard to given bases foundations, existent in Dasein, foundations which can influence self-comprehension, constitute themselves in possibilities of explication as much for Augustinian interpretive variations, as for the existential approach applied by Heidegger to the Pauline epistles and Augustine s text / A tese apresenta as interpreta??es de Agostinho de Hipona acerca dos tr?s primeiros cap?tulos do G?nesis, o primeiro livro da B?blia. Les processus de la mise en scène. Polyphonie et complexité dans la création scénique / The process of performance creation. Polyphony and complexity in stage creationSiaud, Florent 15 December 2014 (has links)
A complex human reality, based on dialogues as well as power relations which are permanently being redefined, is at the heart of the process of performance creation. So as to theorize such processes, several studies have been building a corpus compiling the documents which are produced during meetings or rehearsals. However, such a transfer has proven questionable : as it is made of incomplete traces, such material is necessarily too incomplete to bespeak of the organic and polyphonic life of a performance in gestation. A first solution is to elect a decidedly intermedial approach of a performance’s archives : since various artistic disciplines interact in the process of a performance’s production, one may analyze it by actively comparing and contrasting the different media which are generated during its elaboration. As a complement to the first proposal, a second approach will lead the researcher to get involved into an in vivo observation of meetings and rehearsals so as to have at his disposal a more comprehensive research material. This epistemological clarification paves the way for an attempt to theorize the processes of stage creation. First, it appears that the stage or work space is as much of a physical receptacle for the artists’ research as it is a catalyst : it is in the course of getting to own this space collectively that a group of collaborators gives substance to the production. The creative space thus reveals a polyphonic dimension which is also true regarding time : since it involves an ensemble of artists, a creative process has no uniform chronological linearity ; it comprises a whole array of relations to time which are specific to each of the participants, and one has to bring these temporalities together to give birth to a performance that belongs to all. There is therefore a fundamentally social dimension to any staging process. As it is gathered in a given space and time, the small society which is formed around the stage director has to follow a creative process based on dialogue, where the suggestions of the different individuals coalesce to produce a prolific discourse whose strength and unity are guaranteed by the presence of the director.

It has as starting point the elementary school of the old farm Almas in the municipality of Carirà north of state. Our goal is to understand the functioning of that school and its interface with culture rooted in semiarid landscapes typically from northeast. Traversing these paths we find the answers to same questions of our research, especially that one which seeks to justify the reasons that made those schools the protagonists of rural primary educacion during the twentieth century in our state. Our aim is to study from the educational developments in Brazil, despite the lack of resources, the importance of those schools houses, recognizing that many of them have emerged as an alternative to the education of populations in which the absence of conditions made impossible the displacement to other locations. In this way we contribute from our questions with the understanding of other teaching units and reflections of misguided policies that have been promoted in rural primary school. Then a typically urban education program without however meet the real needs of those populations beyond the suffering caused by the lack of assistence even lived with a social setting aggravated by long periods of drought

This research investigated the use of Mathematics’ videos of the TV Escola by the
Mathematics teacher at the Municipal School of Recife. Statistical data on these
loans from the collection of media General Directorate for Technology in Education
and Citizenship - DGTEC indicated that since 2001, no Mathematics video was ask
by these professionals. We checked if the teachers had access to the videos of TV
Escola by other ways, such as the kits distributed in the schools or the internet, and
made questions if the Mathematics’ teachers who have access to the videos of TV
Escola use them as instruments of pedagogical mediation. The research was mainly
based on the theory of Instrumental Genesis developed by Pierre Rabardel as well
as in studies of José Manuel Moran, Joan Ferres and Juana Maria Sancho. In line
with these theorists, so that we can demonstrate the use of any technological artifact,
it is necessary that the teacher has incorporated didactically teaching processes,
positioning itself epistemologically forward to the possibilities and limitations inherent
in the artifacts and patterns of individual use. The method included questionnaires,
interviews, classroom´s observations and workshops collective. Among mathematics
teachers who answer the questionnaire, there are few who use the medias in their
teaching pedagogical and none of them claimed that had used the TV Escola videos
as teaching tools of mediation. In order to further analyze the reasons for any such
use, a study was made by the type: action-research with two teachers as well as
visits and interviews with managers and coordinators of the schools to which they are
linked. From the analysis of these data, it was identified the constraints and
limitations that are imposed on the Mathematics’ teachers practice and create
barriers process´s instrumentation and instrumentalization for the use of video in the
classroom The subjects claim to know the existence of this collection and recognize
its potential. The reasons for not using it were grouped in our research around seven
interrelated foci: infrastructure´s questions infrastructure issues, such as deficiencies
in the conditions of preservation, difficulties of access to the collection of TV Escola
and lack of information, , lack of specific training for the use that appropriate action;
number of students in the classroom; resistance, from the teacher regarding the use
of technology; belief that students' knowledge are not compatible with the use of
videos, lack of preparation time and experience in the classroom.
