Previous issue date: 2011 / Introdução: As biopróteses valvares cardíacas estão relacionadas a
eventos tromboembólicos, infecciosos e degenerativos. Seu desgaste é
atribuído principalmente à desnaturação do colágeno. O glutaraldeído,
método predominante de preservação de biopróteses, favorece o processo
de calcificação e limita sua durabilidade. Diversas técnicas tentam conter o
processo degenerativo das biopróteses. Objetivos: Avaliar o processo de
calcificação, in vivo, de heteroenxertos pulmonares valvados, preservados
em meios não aldeídico (L-HydroR). Métodos: 17 carneiros foram
submetidos à substituição do tronco da artéria pulmonar por enxerto tubular
valvado de pericárdio bovino. Os animais foram distribuídos em dois
grupos: Grupo L-HydroR (teste / n=14) e Grupo Glutaraldeído (controle /
n=3). Cerca de 150 dias pós implante, foram realizadas angiografia,
medidas hemodinâmicas e sacrifício. Os animais foram necropsiados e as
próteses submetidas a estudo anátomo-patológico, avaliação radiológica e
dosagem do cálcio por espectofotometria de absorção atômica. A análise
estatística foi obtida através dos testes exato de Fisher, T de Student ou
Mann-Whitney (significância: 5%). Resultados: O desempenho
hemodinâmico e angiográfico das próteses testadas foi semelhante nos dois
grupos. A avaliação radiológica, a macroscopia, a microscopia e a dosagem
de cálcio por espectrofotometria de absorção atômica mostraram maior
calcificação nas próteses do Grupo Glutaraldeído, quando comparadas às
próteses do Grupo L-HydroR (p=0,001). 07 animais do Grupo L-HydroR
(50%) apresentaram aderência das cúspides à parede do tubo (p=0,228).
Conclusões: As próteses preservadas em L-HydroR mostraram-se mais
resistentes à calcificação, quando comparadas às preservadas em
Glutaraldeído. / Introduction: The cardiac bioprostheses are related to
thromboembolic events, infectious and degenerative diseases. Wear is
mainly attributed to the denaturation of collagen. Glutaraldehyde, the
predominant method of preservation of bioprostheses, favors the
calcification process and limits their durability. Several techniques try to
contain the degenerative process of bioprostheses. Objectives: To evaluate
the process of calcification in vivo pulmonary valve heterografts preserved
in non-aldehydic (L-HydroR). Methods: 17 sheep underwent replacement
of the pulmonary artery valved tubular grafts of bovine pericardium. The
animals were divided into two groups: Group L-HydroR (test / n = 14) and
Group Glutaraldehyde (control / n = 3). About 150 days after implantation,
angiography was performed, hemodynamic measurements and sacrifice.
The mice were sacrificed and the prostheses subjected to a pathological
study, radiological evaluation and measurement of calcium by atomic
absorption spectrophotometry. Statistical analysis was obtained through the
Fisher's exact test, Student's t or Mann-Whitney test (significance: 5%).
Results: The angiographic and hemodynamic performance of prostheses
tested was similar in both groups. Radiological evaluation, the macroscopic
and microscopic measurement of serum calcium by atomic absorption
spectrophotometry showed increased calcification of the prosthesis
Glutaraldehyde Group, when compared to L-HydroR Group prosthesis (p
=0.001). 07 animals in Group L-HydroR (50%) had adherence of the leaflets
to the wall of the tube (p = 0.228). Conclusions: Prostheses preserved in LHydroR
were more resistant to calcification when compared with

CoordenaÃÃo de AperfeÃoamento de Pessoal de NÃvel Superior / A lipase do tipo B de Candida antarctica recombinante expressa em Pichia pastoris pela inserÃÃo de um DNA externo (LIPB) foi imobilizada em sÃlica mesoporosa nanoestruturada (SBA-15) pelo mÃtodo da adsorÃÃo (SBA-15-LIPB) e ligaÃÃo covalente (SBA-15-APTES-GA-LIPB e SBA-15-APTES-DVS-LIPB). A influÃncia do tempo de contato enzima-suporte e do pH do meio foram avaliados para a produÃÃo dos biocatalisadores. A estabilidade tÃrmica das enzimas imobilizadas foi avaliada e os resultados mostraram que o SBA-15-APTES-GA-LIPB possui maior estabilidade tÃrmica com um t1/2 = 36,91 min enquanto o derivado SBA-15-APTES-DVS-LIPB teve t1/2= 11,83 min e o SBA-15-LIPB teve t1/2= 8,5 min, a 50 ÂC. No entanto, o SBA-15-LIPB possui maior estabilidade em solventes orgÃnicos do que os biocatalisadores produzidos por ligaÃÃo covalente. Assim, o biocatalisador SBA-15-LIPB (CondiÃÃes otimizadas: 100 % de atividade recuperada em um tempo de contato de 1 hora, pH 7) foi aplicado em uma reaÃÃo modelo de esterificaÃÃo, que à a sÃntese de Ãsteres de cadeia curta, o butirato de metila e etila. Os resultados mostraram que, em condiÃÃes otimizadas de esterificaÃÃo (temperatura: 37 ᵒC, tipo de solvente: hexano para o butirato de metila e iso-octano para a sÃntese do butirato de etila, a concentraÃÃo de substrato: 0,2 M, proporÃÃo molar de substratos de 1: 1, tempo de reaÃÃo: 12 h), as conversÃes foram: 79,25% para a sÃntese de butirato de metila e 86,52% para a sÃntese de butirato de etila. O biocatalisador SBA-15-LIPB exibiu elevada atividade e estabilidade operacional na sÃntese do butirato de metila e butirato de etila, por esterificaÃÃo, depois de cinco e seis ciclos sucessivos de 12 horas cada, respectivamente. Os biocatalisadores SBA-15-APTES-GA-LIPB e SBA-15-APTES-DVS-LIPB foram aplicados em uma reaÃÃo modelo de hidrÃlise, que à a resoluÃÃo cinÃtica do acetato de feniletila. Contribuição do glutaraldeído como componente de sistemas adesivos dentinários / Influence of glutaraldehyde priming on bond strengths of an experimental adhesive system applied to wet and dry dentinRenato Cilli 10 August 2007 (has links)
Neste trabalho foram testadas as seguintes hipóteses nulas: 1) Não há diferença entre os valores de resistência de união entre resina restauradora e dentina quando uma solução aquosa de glutaraldeído é aplicada como primer dentinário antes do emprego do agente adesivo; 2) Não há diferença entre os valores de resistência de união, quando a aplicação do mesmo sistema adesivo se dá em condições de dentina úmida ou seca. Dentes humanos foram selecionados, e tiveram sua porção oclusal desgastada, expondo a parte média da dentina coronária para adesão. Quatro grupos foram formados, variando-se a aplicação ou não do primer de glutaraldeído e a condição de umidade dentinária: úmida ou seca. Após aplicação e fotopolimerização do sistema adesivo experimental, foram confeccionados blocos resina composta de 4mm de altura sobre a superfície oclusal e os dentes foram preparados para o teste de microtração para a avaliação da resistência de união. Os resultados obtidos foram submetidos à análise de variância a um critério e teste de Tukey (\'alfa\'\'=0,05). A aplicação do primer de glutaraldeído aumentou significativamente os resultados de resistência de união. Não houve diferença estatística entre o mesmo sistema experimental se aplicado em dentina seca ou úmida. A hipótese 1 não se confirma e aceita-se a hipótese 2. / This work tested the following null hypothesis: 1) There is no difference between resin-dentin bond strengths when a glutaraldehyde primer solution is applied onto demineralized dentin during adhesive bonding. 2) There is no difference between resin-dentin bond strengths when the same experimental adhesives systems are applied whether in dry or wet demineralized dentin conditions. Human, extracted upper third molars were selected. The oclusal surfaces were ground flat, and midcoronal dentin was exposed for bonding. Four groups of six teeth were formed. Two groups were designated for dry dentin technique and two for the wet one. After application and photopolymerization of the bonding system, a 4 mm-high resin core build-up was fabricated over the adhesive layer. Specimens were then prepared for microtensile bond test for bond strengths evaluation. The obtained data were submitted to ANOVA and Tukey test (\'alpha\'=0.05). Glutaraldehyde primer application significantly improved resin-dentin bond strength. No significant difference was observed when the same experimental adhesive system was applied whether in dry or wet dentin conditions. Such results require to reject the first null hypothesis and accept the second one.

Chemical Engineering / Aims of this research project was to add value to largely wasted South African sawdust by development of low-cost AC of high efficiency for removal of toxic Cr (VI), phenol and glutaraldehyde from dilute aqueous media. The main objectives of the research project were: a) To develop low-cost AC based on South African P. patula sawdust using economical physical superheated steam activation.Characterization of carbon samples for selection of optimum preparation conditions for development of low-cost AC of effective microporosity mesoporosity and surface functionality for enhanced adsorption capacity of Cr (VI) and/or phenol and/or glutaraldehyde from dilute aqueous solution. Acid-amine surface groups modification of optimally developed AC for further enhancement of adsorption capacity for mixed polarized glutaraldehyde molecules from aqueous solution. b) To evaluate the aqueous phase batch adsorption properties of developed AC for Cr (VI) and phenol and, of acid-amine modified developed AC for glutaraldehyde. Generation of breakthrough curves at optimum adsorption conditions for evaluation of column performance indicators at different process conditions, bed regeneration-reusability potential, and dynamic adsorption selectivity of developed AC for Cr (VI) from solution of base metals. Determination of column diffusion parameters; accurate mass transfer and empirical modelling of breakthrough data; determination of applicable RSSCT scaling equation; and optimization of breakthrough data for accurate RSSCT scale-up.
