Association entre l'utilisation de Facebook et les marqueurs de stress psychologiques et physiologiques chez les adolescents

La création de Facebook est venue changer la façon dont les gens interagissent, mais nous en savons peu sur les impacts de Facebook sur la santé et le bien-être. À l’heure actuelle, environ 90 % des adolescents sont actifs sur Facebook et la majorité l’utilise tous les jours. Sachant que l’adolescence est une période critique du développement et que lors de cette période les adolescents sont particulièrement vulnérables aux effets du stress, il importe de comprendre les facteurs pouvant entrainer une augmentation des hormones de stress chez les adolescents. Le but du présent mémoire était donc d’étudier la relation entre l’utilisation de Facebook chez les adolescents et des marqueurs de stress psychologique et physiologique. Pour ce faire, nous avons mesuré les hormones de stress chez 88 adolescents (41 garçons, 47 filles) âgés entre 12 et 17 ans. Les adolescents devaient remplir le ‘Social Network Survey’, un questionnaire mesurant différents facteurs associés à l’utilisation de Facebook et le ‘Child Depression Inventory’, qui mesure les symptômes de dépression. Les résultats suggèrent que ce n’est pas le temps passé sur Facebook qui est en lien avec le stress psychologique et physiologique, mais plutôt la nature de l’utilisation de Facebook. Pour les filles, c’est le nombre d’amis sur Facebook qui est relié à des hauts niveaux de cortisol, tandis que pour les garçons c’est le fait de s’exposer sur Facebook. Cette étude est la première à démontrer une association entre la nature de l’utilisation de Facebook et les niveaux d’hormones de stress chez des adolescents. / The creation of Facebook can change the way people interact, but we know little about the impact of Facebook on health and well-being. As of today, 90 % of adolescents are active on Facebook and most of them connect everyday. Adolescence is a critical period in childrens’ development and during this period they are particularly vulnerable to stress hormones. It is therefore important to understand factors that may cause an increase in stress hormones in adolescents. The goal of this Masters thesis was to determine the relation between Facebook utilization in adolescents and psychological and physiological markers of stress. In order to do so, we measured stress hormones in 88 adolescents (41 boys, 47 girls) aged between 12 and 17 years old. Adolescents where asked to fill out the Social Network Survey, a questionnaire measuring different factors related to Facebook utilization and the Child depression Inventory, measuring symptoms of depression. Results suggest that it is not the time spent on Facebook that is related to psychological and physiological stress but the nature of Facebook utilization. For adolescent girls, it is the number of Facebook friends that is associated with high levels of cortisol, whereas in adolescent boys it is the fact of exposing one-self on Facebook that is associated with high levels of cortisol. This study is the first to show an association between Facebook utilization and stress hormones in adolescents.

O ensaio de mutagenicidade Salmonella/microssoma tem sido amplamente utilizado para a avaliação de amostras atmosféricas o qual é muito sensível na detecção de vários tipos de mutágenos, pois é realizado com diferentes linhagens de Salmonella typhimurium na presença e ausência de ativação metabólica exógena. O principal objetivo deste trabalho foi verificar a aplicabilidade da utilização de combinações seletivas de linhagens S. typhimurium na caracterização da mutagenicidade de amostras de material particulado total de ar coletado em Limeira, uma cidade do interior do estado de São Paulo sob influência de intenso tráfego, emissões industriais e da agricultura, incluindo o plantio de cana de açúcar. Foram avaliados seis extratos orgânicos obtidos com diclorometano por ultrasonicação, utilizando o método Salmonella/microssoma em microssuspensão na presença e ausência de ativação metabólica (mistura S9) em experimentos de dose-resposta, com quantidade máxima de 50 µg de material orgânico extraído (MOE) por placa. Os pares de linhagens utilizados foram: YG1041/TA98, para verificar a presença de nitrocompostos, como 1- nitropireno e/ou aminas aromáticas; YG5161/TA1538, altamente sensivel aos hidrocarbonetos policíclicos aromáticos (HPA) não substituídos como o benzo[a]pireno (BaP); e YG7108/TA1535, para a indicação de agentes alquilantes. Os resultados indicaram que os compostos que podem melhor explicar os efeitos observados são os nitro e/ou aminas aromáticas e não os HPA não substituídos como o BaP. O par YG7108/TA1535 forneceu respostas negativas para todas as amostras. As combinações seletivas de linhagens S. typhimurium (YG1041/TA98 e YG5161/TA1538) mostraram-se eficientes na caracterização da mutagenicidade de amostras de ar coletadas na cidade de Limeira, podendo ser utilizadas como uma importante ferramenta de auxílio do ensaio Salmonella/microssoma em microssuspensão em estudos de análises quimicas direcionadas pelo efeito biológico (ADEB). As potências mutagênicas observadas com a linhagem TA98 para a cidade de Limeira foram similares aquelas verificadas na Região Metropolitana de São Paulo. A comparação das respostas de mutagenicidade observadas para as amostras analisadas com o perfil de compostos puros, encontrados na literatura, não forneceu candidatos que podem representar a mistura. Mais estudos de mutagenicidade com compostos puros utilizando os pares de linhagens propostos (TA98/YG1041 e TA1538/YG5161) e análises quimicas de extratos fracionados são necessários para verificar se esta abordagem pode ser útil em estudos de ADEB, e consequentemente na descoberta de novos poluentes atmosféricos. / The atmospheric particulate matter is recognized as mutagenic. We tested six extracts obtained with methylene chloride by ultrasonication using the Salmonella/microsome microsuspension protocol in the presence and absence of rat liver S9 in dose response experiments, using 50 µg of extractable organic matter (EOM) as a maximum dose per plate. The following pairs of strains were used: YG1041/TA98, to verify the presence of nitro-compounds, as 1-nitropyrene and/or aromatic amines; YG5161/TA1538, highly sensitive to non-substituted polycyclic aromatic hydrocarbons (PAHs) as benzo[a]pyrene (BaP); and YG7108/TA1535, for the indication of alkylating agents. The results indicated that the compounds that can better explain the observed effect are the nitro and/or aromatic amines and not nonsubstituted PAHs such as BaP. The pair YG7108/TA1535 provided negative responses for all the samples. Selective combinations of S. typhimurium strains (YG1041/TA98 and YG5161/TA1538) proved efficient in the characterization of mutagenicity of airborne samples collected in the city of Limeira and can be used as an important tool for the Salmonella/microsome microsuspension assay in effect directed chemical analysis (EDA) studies. The mutagenic potencies observed with TA98 strain for the city of Limeira were similar to those verified in the Metropolitan Region of São Paulo. The comparison of the observed mutagenicity responses for the samples analyzed with the profiles of pure compounds found in the literature did not provide any candidates that could alone represent the mixture. More mutagenicity studies with pure compounds using the proposed pair of strains (TA98/YG1041 and TA1538/YG5161) along with chemical analysis of fractioned extracts are needed to verify if this approach can be useful in EDA studies, and consequently in the discovery of new air priority pollutants.

Non-invasive measurement of stress and pain in cattle using infrared thermography : a thesis submitted in partial fulfilment of the requirements for the degree of Doctor of Philosophy in Animal Science at Massey University, Palmerston North, New Zealand

The aim of this thesis was to validate the use of infrared thermography (IRT) to non-invasively measure stress and/or pain in cattle. The main approach was to measure changes in heat emitted from superficial capillaries around the eye (referred to as eye temperature) in response to various aversive husbandry procedures used routinely on farms. In addition, various exogenous challenges were given to investigate the role of the hypothalamic-pituitary-adrenal (HPA) axis and the autonomic nervous system (ANS) in regulating the eye temperature response. No evidence was found to support the hypothesis that an increase in eye temperature was due to HPA activity in cattle. A rapid drop in eye temperature occurred immediately after disbudding, an electric prod, startling and shouting. It is suggested that this was caused by the redirection of blood from the capillary beds via sympathetically-mediated vasoconstriction. Therefore, the role of the ANS was tested by measuring eye temperature, heart rate variability (HRV) and plasma catecholamine responses simultaneously. Somatic pain from disbudding and initial responses to surgical castration included a synchronised drop in eye temperature, increases in catecholamines and changes in HRV indicative of increased sympathetic activity. The role of the sympathetic nervous system was further confirmed by a drop in eye temperature that occurred following an epinephrine challenge. In contrast, deeper visceral pain from castration caused a more marked increase in eye temperature and changes in HRV indicative of increased parasympathetic tone. The underlying mechanism driving the increase in eye temperature is unknown; however, it is possible that it may be caused by vasodilation due to increased parasympathetic activity. These differences in ANS responses to different procedures, detected by IRT and HRV, may be due to the nature of the pain and the relative fear associated with the procedure. In summary, this research showed that during stress or pain, the heat emitted from superficial capillaries around the eye changes as blood flow is regulated under ANS control and these changes can be quantified using IRT. A combination of IRT and HRV is a non-invasive way to measure ANS activity and assess acute welfare impacts of husbandry practices in cattle. Further research using pharmacological inhibition and stimulation of the ANS activity would be beneficial to fully understand the underlying regulatory mechanisms of the eye temperature and HRV responses in cattle and other species during stress and/or pain. The full capability of IRT and HRV for detection of disease and emotional states and the effects of different intensities of pain, individual traits and previous experience also deserve attention.

The emotional motor system and gastrointestinal symptoms

There is a significant comorbidity between anxiety/depression and functional gastrointestinal syndromes, such as irritable bowel syndrome (IBS) and functional dyspepsia. The pathophysiological link between emotions and the gut is not known. A model of an emotional motor system (EMS) which reacts to interoceptive and exteroceptive stress has been proposed. EMS consists of specific brain structures including anterior cingulate cortex (ACC), amygdala, hippocampus and hypothalamus and mediates their communication to the rest of the body (including the gastrointestinal tract) through the hypothalamus-pituitary-adrenal (HPA) axis, the autonomic nervous system (ANS) and by a pain modulation system. The aim of this thesis was to test the EMS model by studying the relationship between symptoms of anxiety and depression and IBS-like symptoms in patients with recurrent unipolar depression, in patients with IBS and in a sample of a normal Swedish population. The peripheral limb of EMS (ANS, HPA axis and the pain modulations system) was tested in patients with IBS and control subjects. Spectral heart rate variability was used to investigate ANS function in patients with refractory IBS and in healthy controls. The HPA axis function was tested by a weight adjusted low dose dexamethasone suppression test in control subjects. The influence of catecholamine degradation on pain modulation was tested by analyzing val158met catechol-o-methyl transferase (COMT) polymorphism in patients with IBS and in control subjects. We found a significant relationship between symptoms of anxiety/depression and IBS-like symptoms in patients with recurrent unipolar depression, in patients with IBS and in a sample of the normal population. Interestingly, patients with recurrent unipolar depression in remission had no more IBS-like symptoms than controls, indicating that the gastrointestinal symptoms may resolve when depression is treated to remission. Patients with IBS have an increased mid-frequency power in rest and in supine position (after tilt test) compared to healthy controls indicating an increased sympathetic ANS drive. The symptoms of diarrhea and early satiety has in the litterature been associated to the stimulation of corticotropin releasing hormone (CRH) receptors and was also in our study related to HPA axis function tested by a low dose dexamethasone test. Interestingly both hypo- and hyperfunction of the HPA axis was related to these symptoms in control subjects. The val158met COMT polymorphism was associated to IBS-like symptoms. Control subjects with IBS-like symptoms (defined by the upper quartile in total GSRS-IBS score) had a higher frequency of the met/met and a significantly lower frequency of the val/met genotype. Also patients with IBS tended to have a lower frequency of the heterozygous val/met genotype so we conclude that this genotype may be protective against IBS/IBS like symptoms. In addition, the val/val genotype in patients with IBS was associated to diarrhea symptoms. Conclusions: Our results support the model of an emotional motor system in the genesis of functional gastrointestinal symptoms by the finding of the association of IBS-like symptoms and mood disturbances, and by finding alterations in the peripheral limbs of EMS (ANS, HPA axis and catecholamines) in subjects with IBS and IBS-like symptoms.

Neurocognitive and endocrine dysfunction in women with exhaustion syndrome

Stress has emerged as one of the most important factors to consider in psychiatric diagnoses and has become a common reason for long-term sick leave (LTSL). Roughly 50% of LTSL due to psychiatric diseases are thought to be associated with work-related stress. The demarcation towards major depression is disputed, and no international consensus exists for how to diagnose and rehabilitate these individuals. The Swedish National Board of Health has suggested the term “exhaustion syndrome” to integrate these individuals into stress-related disorders. Prominent features of this syndrome are fatigue, sleeping disorders, and cognitive dysfunction. The cognitive dysfunction may be due to an interaction between personality features, environmental factors, the biological effects of stress hormones, and dysfunction in key brain areas, notably the hippocampus and prefrontal cortex. A consistent feature of chronic stress is activation of the cortisol, or hypothalamic-pituitary-adrenal, axis, which may be linked to cognitive dysfunction. Increased glucocorticoid levels, mainly cortisol in humans, are known to impair memory performance. The aim of this thesis was to investigate whether patients with exhaustion syndrome exhibit specific alterations in an extensive set of biological, psychological and immunological variables. Patients in Study 1 had significant cognitive impairment for specific tasks assumed to tap frontal lobe functioning. In Study 2 anxiety prone, worrying, pessimistic individuals with low executive drive and a persistent personality type were more likely to develop exhaustion syndrome. Decreased reactivity was found on the pituitary level after corticotropin releasing hormone (CRH) in exhaustion syndrome patients. The cortisol/adrenocorticotropic hormone response to CRH was slightly higher in patients compared to controls, indicating increased sensitivity at the adrenal cortex level. No differences were found in hippocampal volume. In Study 3, functional imaging revealed a different pattern of brain activation in working memory tests in patients with exhaustion syndrome compared to healthy individuals and patients with depression. In summary, our data suggests an intimate link between personality and wellbeing, cognitive performance and neuroendocrine dysfunction, in exhaustion syndrome. We thus find similarities with major depression but also distinct differences between the exhaustion syndrome and major depression.

Caracterización de los cambios celulares y moleculares en el sistema cerebral del estrés durante la dependencia de morfina / Cellular and molecular changes in the stress-responsive system during morphine dependence

Morphine withdrawal increases the HPA axis activity, which is dependent on a hyperactivity of noradrenergic pathways (NTS-A2) innervating the PVN. In this Thesis we found that morphine withdrawal resulted in an increase in ACTH and corticosterone secretion and a neuronal activation in the PVN. Additionally, we found robust increases in CRF and AVP hnRNAs in the PVN, concomitantly with an increase in c-Fos expression. Morphine withdrawal activated ERK1/2 in PVN and NTS, which could be involved in the c-Fos expression. Morphine withdrawal induced an increase in TH mRNA levels in the NTS-A2, total TH protein in the NTS and TH phosphorylation at Ser31 in PVN and NTS-A2, which resulted in an augmentation of TH activity in the PVN. The enhancement in TH phosphorylated at Ser31 was blocked by SL327 in both nuclei. Finally, we have found that adrenalectomy eliminated the hyperactivity of noradrenergic pathways innervating the PVN during morphine withdrawal. / La abstinencia a morfina aumenta la actividad del eje HHA, que depende de la hiperactivación de las vías noradrenérgicas (NTS-A2) que inervan al PVN. En esta Tesis encontramos que la abstinencia a morfina induce la liberación de ACTH y corticosterona y la activación neuronal del PVN. Además, observamos un aumento en los niveles de hnRNA para CRF y AVP en el PVN, concomitantemente con un incremento en la expresión de c-Fos. El síndrome de abstinencia a morfina activó a ERK1/2 en PVN y NTS. También indujo un incremento en el mRNA para TH en el NTS-A2, proteína TH total y fosforilación de TH en su Ser31 en PVN y NTS-A2, que se tradujo en un aumento en su actividad enzimática. El aumento de TH fosforilada en Ser31 fue bloqueado por SL327. Finalmente, la adrenalectomía eliminó la hiperactividad de las vías noradrenérgicas que inervan al PVN durante la abstinencia a morfina.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A mutagenicidade do material particulado é atribuída primeiramente aos hidrocarbonetos policíclicos aromáticos (HPA). Investigamos a atividade mutagênica do material particulado (MP2,5) em amostras coletadas em três pontos da cidade do Rio de Janeiro. As coletas foram realizadas com auxílio de um amostrador de grande volume na Avenida Brasil, no campus da Universidade do Estado do Rio de Janeiro e no Túnel Rebouças em filtros de fibra de vidro. Metade de cada filtro foi submetido à extração por sonicação com o solvente diclorometano. Seis HPA foram identificados e quantificados por cromatografia gasosa com espectrometria de massa (GC/MS). Após a análise química as concentrações dos HPA obtidos foram correlacionados ao fatores físicos, além de ser realizado avaliação de risco para cada HPA estudado. Linhagens de Salmonella typhimurium (TA98 e derivadas TA98/1.8-DNP6, YG1021 e YG1024) foram utilizadas no ensaio de mutagenicidade e tratadas (10-50 g/placa) com extrato orgânico na presença e na ausência de metabolização exógena. Células de raiz de cebola foram tratadas com extratos orgânicos nas concentrações (5-25g/mL). A alta umidade encontrada no Túnel Rebouças pode ter influenciado na deposição de cinco dos seis HPA estudados em material particulado. Além disso, em diferentes condições de tráfego, motoristas de ônibus que cruzam a Avenida Brasil e o Rebouças túnel estão expostos ao risco induzidos por HPA na ordem de 10-6. Mutagenicidade foi detectada tanto na presença quanto na ausência de metabolização, para as linhagens YG1021 e YG1024 nos três pontos, sugerindo a presença de nitro e amino derivados de HPA. We investigated the mutagenic activity of particulate matter (PM2.5) samples collected from three sites in Rio de Janeiro. Samples were collected using a high-volume sampler at Avenida Brasil, the campus of the Rio de Janeiro State University, and Rebouças tunnel. Half of each filter was submitted to sequential extraction by sonication with dichloromethane. Six PAHs were quantified by gas chromatography/mass spectrometry (GC/MS). Salmonella typhimurium TA98 and the derivative strains TA98/1.8-DNP6, YG1021 and YG1024 used in mutagenicity assays were treated (10-50 g/plate) with and without exogenous metabolization. Onion root cells were treated with organic extracts concentrations (5-25g/mL). The high humidity detected in the Rebouças Tunnel may have influenced the deposition of five of the six PAHs studied in particulate matter. Moreover, in different traffic conditions can put the bus drivers that cross the Brasil Avenue and Rebouças tunnel at the risk of exposure induced by HPA in the order of 10-6. Independently of exogenous metabolization, mutagenicity was detected for strains YG1021 and YG1024 at all the sites, suggesting the presence of nitro and amino derivatives of PAHs. Rebouças tunnel presented the highest values for rev/g and rev/m3. These could be related to the fact that this long, enclosed passageway and restricts ventilation. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / A mutagenicidade do material particulado é atribuída primeiramente aos hidrocarbonetos policíclicos aromáticos (HPA). Investigamos a atividade mutagênica do material particulado (MP2,5) em amostras coletadas em três pontos da cidade do Rio de Janeiro. As coletas foram realizadas com auxílio de um amostrador de grande volume na Avenida Brasil, no campus da Universidade do Estado do Rio de Janeiro e no Túnel Rebouças em filtros de fibra de vidro. Metade de cada filtro foi submetido à extração por sonicação com o solvente diclorometano. Seis HPA foram identificados e quantificados por cromatografia gasosa com espectrometria de massa (GC/MS). Após a análise química as concentrações dos HPA obtidos foram correlacionados ao fatores físicos, além de ser realizado avaliação de risco para cada HPA estudado. Linhagens de Salmonella typhimurium (TA98 e derivadas TA98/1.8-DNP6, YG1021 e YG1024) foram utilizadas no ensaio de mutagenicidade e tratadas (10-50 g/placa) com extrato orgânico na presença e na ausência de metabolização exógena. Células de raiz de cebola foram tratadas com extratos orgânicos nas concentrações (5-25g/mL). This product is one of the main constituents of polycyclic aromatic hydrocarbons (PAH), which exhibit high toxicity. Therefore, this study aims at developing and validating a method for determining PAH (Benz(a)anthracene, Benzo(b)fluoranthene, Benzo(k)fluoranthene, Benzo(a)pyrene, and Dibenzo (a,h)anthracene) in PW by using magnetic solid phase extraction (MSPE) and high-performance liquid chromatography with fluorescence detector. We synthesized three new adsorbents: FeCt (with ferric chloride and water rich in natural organic matter); FeCc (with ferric chloride, distilled water, and coconut husk), and FeSt (with ferric sulfate and water rich in natural organic matter). The x-ray diffraction and infrared spectroscopy confirmed obtaining the hybrid materials. The best extraction conditions were obtained by using ultrasonic cleaning as a mixing mechanism with time set to 2.5 minutes for adsorption and 2.5 minutes for desorption, using 10 mL of ethyl acetate as an extractor and 20 mg of hybrid FeSt; the values of PAH recovery varied from 84.3 to 92.3 %. The capacity of reusing the material was assessed and the results exhibited a great recovery with up to 4 cycles of adsorption/desorption. The methodology we propose was applied to two PW samples. The recovery values for samples PWW1 (produced water at well 1) enriched with 100μg L-1 were of 81 – 104%, without the presence of B(a)P standard. As for the PAH concentrations in the sample, they varied from 0.63 to 1.66 μg L-1 for PWW1 from 0.54 to 1.62 μg L-1 for PWW2 (produced water at well 2). The validation of the method was performed for the following parameters: linearity, which exhibits a correlation coefficient (r) above 0.9943 within the interval of 0.5 - 25 μg L-1 ; accuracy and precision, with recovery values between 98.2 – 108.9 %, and coefficient of variation 0.64 – 6.72 % for the concentration levels of 3.8 and 15 μg L-1 (n=3), respectively. LODs varied from 0.012 and 0.02 μg L-1 and LOQs, from 0.036 to 0.06 μg L-1. / A indústria de petróleo e gás produz grandes quantidades de água produzida (AP), que é um efluente complexo, sendo que um dos seus principais constituintes são os Hidrocarbonetos Policíclicos Aromáticos (HPA), os quais apresentam elevada toxicidade. Assim, o presente trabalho tem por objetivo desenvolver e validar um método de determinação de HPA (Benzo(a)Antraceno, Benzo(b)Fluoranteno, Benzo(k)Fluoranteno, Benzo(a)Pireno e Dibenzo(ah)Antraceno) em AP utilizando a extração em fase sólida magnética (MSPE) e cromatografia líquida de alta eficiência com detector de fluorescência. Foram sintetizados três novos adsorventes FeCt (com cloreto férrico e água com alto teor de MON), FeCc (com cloreto férrico, água destilada e bagaço de coco) e FeSt (com sulfato férrico e água com alto teor de MON). A difração de raios X e a espectroscopia de infravermelho confirmaram a obtenção dos materiais híbridos. As melhores condições de extração foram obtidas em banho de ultrassom como mecanismo de agitação, com tempo estipulado em 2,5 min para a adsorção e 2,5 min para a dessorção, utilizando 10 mL de acetato de etila como extrator e 20 mg do híbrido FeSt. Os valores de recuperação dos HPA variaram de 84,3 – 92,3 %. A capacidade de reutilização do material foi avaliada e os resultados apresentaram ótima recuperação com até 4 ciclos de adsorção/dessorção. A metodologia proposta foi aplicada em duas amostras de AP e os valores de recuperação para a amostra de APP1 (água produzida poço 1), enriquecida com 100 μg L-1, foram de 81 – 104 %, sem a presença do padrão do B(a)P, e a concentração dos HPA na amostra variou de 0,63 – 1,66 μg L-1 para o APP1, e de 0,54 – 1,62 μg L-1 para o APP2 (água produzida poço 2). A validação do método foi realizada para os parâmetros: linearidade, com o coeficiente de correlação (r) acima de 0,9943 no intervalo de 0,5 - 25 μg L-1; exatidão e precisão, com valores de recuperação entre 98,2 - 108,9 % e coeficiente de variação 0,64 - 6,72 % para os níveis de concentração 3, 8 e 15 μg L-1 (n=3), respectivamente.
