A Study of the Three Poets in The Hai-Wai-Chi Society

Guo, Chiou-shien 06 September 2007 (has links)
Abstract The Hai-Wai-Chi Society, an overseas organization derived from the ¡§Chi Society¡¨ based in Shanghai, was initiated and founded by Hsu Fu-Yuan, one of the six poets of the Chi Society. Towards the end of the Hung Kung reign of the late Ming dynasty, in the wake of the defeat of the Sungchiang uprising staged by the society, Hsu continued to work for the recovery of the Ming legacy against the ruling Qing regime. At first, he left Shanghai to follow Emperor Lungwu in Fujian. After Emperor Lungwu was defeated, he proceeded to Choushan to follow King Lu and cooperated with Chang Huang-Yen. Later on, after Choushan was seized by the Qing army, Hsu accompanied King Lu under the shelter of Koxinga in Xiamen. It was at this point that the society came to its apex. When Koxinga took hold of Taiwan following his defeat at Nanjing battle, the society members began to enter the island along with the soldiers. Therefore, the literature of Hai-Wai-Chi Society is viewed as the beginning of Han Taiwanese literature. Taiwan Shih Cheng by Lien Heng refers to Hsu Fu-Yuan, Chang Huang-Yen, Lu Jo-Teng, Sheng Chuan-Chi, Chao Tsung-Lung, Chen Shih-Ching as ¡§The Six Poets of Hai-Wai-Chi Society.¡¨ This paper, entitled A Study of Three Poets in the Hai-Wai-Chi Society, is intended to follow the same way of thinking in its exploration of Hsu Fu-Yuan, Chang Huang-Yen, and Lu Jo-Teng through the existing anthology of the society. Meanwhile, the relevant literature concerning the other three poets will be reviewed for necessary references. The Hai-Wai-Chi Society is characterized by its composition of core members, mainly loyalists to King Lu, who followed Koxinga to Taiwan as a consequence of the fall of Choushan. Mourning over the end of the Ming empire, intellectuals of high morality and talent assembled to form a society of poetry to express their emotions toward the fatherland. The members of the Hai-Wai-Chi Society were actively involved in the fight against the Qing court. In spite of successive defeats, they would live and die in exile overseas rather than surrender as subjects of the Qing regime. The Introduction aims to explain the motivation of this study, the meaning and formation of The Hai-Wai-Chi Society, with a literature review of the three poets. The first chapter explores the public opinions of The Dong-Lin Party¡]ªFªLÄÒ¡^ and the Restoration Society¡]´_ªÀ¡^ in the Late Ming period, and the poetry of patriotism by Chen Tzu-Lung and Hsia Wang-Chun of The Hai-Wai-Chi Society, to be defined in this study as two typical kinds of heritage of The Hai-Wai-Chi Society. The second chapter gives an analysis of the historical background of South Ming royalists fighting against the Qing regime and the formation and development of The Hai-Wai-Chi Society. The third chapter is focused on the Tiao-Huang-Tang Manuscript by Hsu Fu-Yuan to study his overseas poems that express his uncompromising anti-Qing mentality and noble patriotic integrity. The fourth chapter represents the social reality of war-ridden Kinmen during the Koxinga period based on Niao-Yi Poetry and Liu-An Anthology by Lu Jo-Teng. The fifth chapter deals with Chi-Ling Grass, Ping-Tsa Collection, and Tsai-Wei Recitation by Chang Huang-Yen to distinguish him as a great noble Ming loyalist who had fought against the Qing power for 19 years overseas. The Conclusion sums up the poetry by the three poets as: (1) a reflection of history with a national concern, (2) an expression of opposition against invasion with a firm anti-Qing determination, and (3) a kind of oceanic literature with a universal sympathy.

Development of the CRISPR nuclease Cas9 for high precision mammalian genome engineering

Hsu, Patrick David January 2014 (has links)
Recent advances in genome engineering technologies based on the CRISPR-associated RNA-guided endonuclease Cas9 are enabling the systematic interrogation of genome function. Analogous to the search function in modern word processors, Cas9 can be guided to specific locations within complex genomes by a short RNA search string. Using this system, DNA sequences within the endogenous genome and their functional outputs are now easily edited or modulated in virtually any organism of choice. Cas9-mediated genetic perturbation is simple and scalable, empowering researchers to elucidate the functional organization of the genome at the systems level and establish causal linkages between genetic variations and biological phenotypes. To facilitate successful and specific Cas9 targeting, we first optimize the guide RNAs (sgRNA) to significantly enhance gene editing efficiency and consistency. We also systematically characterize Cas9 targeting specificity in human cells to inform the selection of target sites and avoid off-target mutagenesis. We find that SpCas9 tolerates mismatches between guide RNA and target DNA at different positions in a sequence-dependent manner, sensitive to the number, position and distribution of mismatches. We also show that Cas9-mediated cleavage is unaffected by DNA methylation and that the dosage of Cas9 and sgRNA can be titrated to minimize off-target modification. Additionally, we provide a web-based software tool to guide the selection and validation of target sequences as well as off-target analyses. We next demonstrate that Cas9 nickase mutants can be used with paired guide RNAs to introduce targeted double-strand breaks. Because individual nicks in the genome are repaired with high fidelity, simultaneous nicking via appropriately offset guide RNAs can reduce off-target activity by over 1,500-fold in human cells. In collaboration with researchers at the University of Tokyo, we further identified a PAM-interacting domain of the Cas9 nuclease that dictates Cas9 target recognition specificity. Finally, we present protocols that provide experimentally derived guidelines for the selection of target sites, evaluation of cleavage efficiency and analysis of off-target activity. Beginning with target design, gene modifications can be achieved within as little as 1-2 weeks. Taken together, this work enables a variety of genome engineering applications from basic biology to biotechnology and medicine.

許學夷《詩源辯體》研究 / A Study of Hsu Hsueh-yi's Shih yuan pien t'i

謝明陽, Hsieh, Ming Yang Unknown Date (has links)

Simple models for predicting dune erosion hazards along the Outer Banks of North Carolina [electronic resource] / by Lauren McKinnon Wetzell.

Wetzell, Lauren McKinnon. January 2003 (has links)
Title from PDF of title page. / Document formatted into pages; contains 84 pages. / Thesis (M.S.)--University of South Florida, 2003. / Includes bibliographical references. / Text (Electronic thesis) in PDF format. / ABSTRACT: Hurricane hazards result from the combined processes of wind, waves, storm surge, and overwash (Lennon et al., 1996). Predicting the severity of these hazards requires immense effort to quantify the processes and then predict how different coastal regions respond to them. A somewhat simpler, but no less daunting task is to begin to predict the hazards due to potential erosion of barrier islands. A four-part scale has been developed by Sallenger (2000) to provide a framework for understanding how barrier islands might respond during extreme storm events. These four regimes describe how beach and dune elevations interact with surge and wave runup. This study will produce estimates of potential hazards through combining lidar surveys of dune elevation with modeled elevations of storm water levels. Direct measurements of maximum wave heights during hurricanes are rare. / ABSTRACT: We evaluated three simple equations proposed by Kjerfve (1986), Young (1988), and Hsu (1998) to forecast the maximum wave height (Hmax) generated by three 1999 hurricanes. Model results were compared to wave data recorded by the National Oceanic and Atmospheric Administration (NOAA) wave rider buoys. The radius of maximum winds, wind speed, forward velocity, distance from buoy to the storm's eye-wall (r), and buoy's position relative to the quadrant of the storm (Q) were found to have significant and direct roles in evaluating recorded hurricane induced wave heights (H) and thus, were individually examined for each comparison. The implications of the r and Q on H were assessed when determining the overall effectiveness of the modelers' equations. Linear regression analyses tested the accuracy of each modeled prediction of the Hmax, comparing it to the observed wave heights. Three statistical criteria were used to quantify model performance. / ABSTRACT: Hsu's model was the most reliable and useful forecasting technique. Despite the predictive skill of Hsu's model, direct observations of the maximum wave conditions, when available and appropriate, are preferred as inputs for SWAN, a 3rd generation shoaling wave model. Outputs from SWAN are used to calculate the empirical relationships for wave runup. For our test case, pre and post-storm topographies were surveyed as part of a joint USGS-NASA program using lidar technology. These data sets were used to calculate changes in the elevation and location of the dune crest (Dhigh) and dune base (Dlow) for the North Carolina Outer Banks. We hindcast potential coastal hazards (erosional hot spots) using the pre-storm morphology and modeled wave runup and compare those estimates to the measured results from the post-storm survey. / ABSTRACT: Links among the existing topography and spatial variations in wave runup were found to be 95% correlated for the north-south and east-west facing barrier islands. Application of Sallenger's (2000) four-part Storm Impact Scale to the pre-storm Dhigh elevation survey and wave runup extremes (Rhigh and Rlow) were found to accurately predict zones of overwash and showed potential to forecast the inundation regime. / System requirements: World Wide Web browser and PDF reader. / Mode of access: World Wide Web.

Shaping the Future Past: Finding History, Creating Identity in the Kwan Hsu Papers

Donnelly, Lisa Chere' 01 January 2012 (has links)
Dr. Kwan Hsu was neither a superstar nor a celebrity. Her name does not come up in conversations about important contributors to her field of biophysics nor is she instantly recognizable for her contributions to Portland State University's international program or the state of Oregon's business ties with China. Yet she was a contributor, a cog-in-the-wheel, at the very least, in all of these areas and more. She was a peripheral member of a well-known Chinese family, but few in the United States know of or perhaps have interest in, but otherwise, she had no great connections or family ties to generate interest in her story. How does one process a collection for a woman who does not meet the traditional criteria for excellence or success or public interest for an archive? Where is the value to the larger historical narrative of our time in preserving the memories of someone who was non-remarkable, or, conversely, someone who may be even too unique to contribute to that greater narrative? These are the questions I wrestled with when I first came to this collection. As my research progressed, I realized that I faced more questions, and that to come to any understanding that might answer them, I was going to have to research the history of archives and archival processes. Science, the Cold War, Communist China, women, the immigrant experience, all of these issues became part of my thesis, however shallowly I was able to investigate them. Questions of identity and historiography, of power and discourse were explored. In the end, what I found was that a collection that on the outside looked unimpressive and unenlightening, could indeed be very valuable, and provide insight into any number of areas of current interest in historical research. This is that story.
