Efeitos da eletroestimulação neuromuscular durante a imobilização nas propriedades mecânicas do músculo esquelético / Effects of neuromuscular electric stimulation during the immobilization in the mechanical properties of the skeletal muscle.

A estimulação elétrica neuromuscular (EENM) é um importante recurso utilizado na medicina esportiva para acelerar processos de recuperação. O objetivo deste estudo foi analisar os efeitos da EENM durante a imobilização do músculo gastrocnêmio, em posições de alongamento (LP) e encurtamento (SP). Para tanto, 60 ratas fêmeas jovens WISTAR foram distribuídas em seis grupos e acompanhadas durante 7 dias: controle (C), eletroestimuladas (EE), imobilizadas em encurtamento (ISP), imobilizadas em alongamento (ILP), imobilizadas em encurtamento e eletroestimuladas (ISP+EE) e imobilizadas em alongamento e eletroestimuladas (ILP+EE). Para a imobilização, o membro posterior direito foi envolvido por uma malha tubular e ataduras de algodão juntamente à atadura gessada. A EENM foi utilizada com uma freqüência de 50 Hz, 10 minutos por dia, totalizando 20 contrações em cada sessão. Após 7 dias os animais foram submetidos à eutanásia e os músculos gastrocnêmios foram retirados para a realização do ensaio mecânico de tração em uma máquina universal de ensaios (EMIC®). A partir dos gráficos carga versus alongamento foram calculadas as seguintes propriedades mecânicas: alongamento no limite de proporcionalidade (ALP), carga no limite de proporcionalidade (CLP), alongamento no limite máximo (ALM), carga no limite máximo (CLM) e rigidez. As imobilizações SP e LP promoveram reduções significativas (p<0,05) nas propriedades de ALP, CLP, ALM e CLM sendo mais acentuada, principalmente, no grupo ISP. Quando utilizada a EENM, houve acréscimo significativo (p<0,05) destas propriedades somente no grupo ISP. Já, em relação à rigidez, foi observada redução significativa (p<0,05) somente do grupo C para o grupo ISP. Quando utilizada a EENM, a rigidez do grupo ILP+EE foi significativamente (p<0,05) maior e mais próxima do grupo C que a do grupo ISP+EE. Neste modelo experimental, a imobilização dos músculos em alongamento atrasou a queda das propriedades e a estimulação elétrica, contribuiu para a manutenção das propriedades mecânicas durante o período de imobilização, principalmente no grupo ILP+EE. / The neuromuscular electric stimulation (NMES) is an important tool used in sport medicine to accelerate the recovery process. The objective of this study was to analyze the effects of NMES during the immobilization of the gastrocnemius muscle, in lengthened (LP) and shortened positions (SP). Sixty young female rats WISTAR were distributed into six groups and followed for 7 days: control (C), electric stimulation (ES), immobilized in shortened (ISP), immobilized in lengthened (ILP), immobilized in shortened and electric stimulation (ISP+ES) and immobilized in lengthened and electric stimulation (ILP+ES). For the immobilization, the right hind limb was involved by a tubular mesh and cotton rolls and plaster. NMES was used in a frequency of 50 Hz, 10 minutes a day, totaling 20 contractions in each session. After 7 days the animals were killed and their gastrocnemius muscles of the right side were submitted to a mechanical test in traction in an universal test machine (EMIC®). From the curves load versus elongation the following mechanical properties were obtained: elongation in the yield limit (EPL), load in the yield limit (LPL), elongation in the ultimate load (EUL), ultimate load (UL) and stiffness. The immobilizations SP and LP promoted significant reductions (p<0,05) in the properties of EPL, LPL, EUL and UL being more accentuated mainly in group ISP. When used NMES, there was significant increment (p<0,05) of such properties only in group ISP. As for stiffness, significant reduction was observed (p<0,05) only of the group C for group ISP. When the NMES was used, the stiffness of group ILP+EE was significantly (p<0,05) higher and closer to group C than for group ISP+EE. We conclude that in this experimental model the immobilization of the muscles in the lengthened position delayed the reduction of the properties and the electric stimulation contributed to the maintenance of the mechanical properties during the immobilization period, mainly for group ILP+ES.

Análise das respostas musculares adaptativas frente a protocolos de alongamento e exercício excêntrico, aplicados após desuso do membro posterior de ratas em desenvolvimento pós-natal / Analysis of adaptive muscular responses to stretching and eccentric exercises applied after immobilization of the hindlimb in female rats in postnatal development

O presente estudo analisou as mudanças e adaptações geradas pelos protocolos de alongamento e exercício excêntrico nos músculos sóleo e plantar, após 10 dias de imobilização do membro posterior de ratas em desenvolvimento pós-natal. Para isso, 45 ratas da raça Wistar, com 21 dias de idade, foram divididas em 10 grupos: grupo imobilizado (GI); grupo imobilizado e treinado excentricamente por 10 dias (GITE(10)) ou 21 dias (GITE(21)); grupo imobilizado e alongado por 10 dias (GIAL(10)) ou 21 dias (GIAL(21)); grupo anestesiado (GA); grupo controle de 21 dias de idade (GC(zero)); grupo controle do imobilizado de 31 dias de idade (GC(Imob)); grupo controle de 10 dias com 41 dias de idade (GC(10)); grupo controle de 21 dias com 52 dias de idade (GC(21)). A imobilização foi aplicada por 10 dias no membro posterior direito dos animais, a fim de manter a posição de encurtamento dos músculos sóleo e plantar. Posteriormente, as ratas do GITE(10) e GITE(21) passaram pelo período de treino excêntrico em esteira declinada, por 40 minutos. Enquanto os animais do GIAL(10) e GIAL(21) foram submetidos ao alongamento mantido por 40 minutos, através da fixação da dorsiflexão máxima permitida pelo tornozelo. Após os protocolos, os fragmentos dos músculos sóleo e plantar foram retirados e submetidos às reações histoquímica, imunoistoquímica e bioquímica. Análise qualitativa e quantitativa (isoformas de MHC, proporção de fibras, diâmetro menor e relação capilar/fibra) foram realizadas. Para análise estatística foi usado o Modelo Linear de Efeitos Mistos com nível de significância de 5% e Intervalo de Confiança de 95%. A partir da análise temporal do desenvolvimento muscular dos animais, foram observadas características de imaturidade dos músculos sóleo e plantar (fibras arredondadas, núcleos volumosos, reatividade do tecido), principalmente em animais com 21 e 31 dias de idade. Aumento progressivo do diâmetro das fibras e da relação capilar/fibra, em ambos os músculos, foram observados. O predomínio de FT1 e maior expressão da MHCI já estavam evidentes no músculo sóleo, mesmo de animais com 21 dias de idade. O músculo plantar apresentou predomínio de FT2D na análise da mATPase, e a avaliação das isoformas indicou predomínio da MHCIId e MHCIIb. O procedimento de imobilização desencadeou alta reatividade do tecido, redução do diâmetro de todos os tipos de fibras, mudança na proporção das fibras (redução de FT1 no sóleo e aumento de FT2D no plantar) e na expressão das isoformas de MHC, e queda do número de capilares transversais. A aplicação do alongamento e exercício excêntrico provocou adaptações importantes como o aumento do diâmetro das fibras, modificação da proporção das fibras e aumento da relação capilar/fibra, em ambos os músculos. Da análise morfológica, o músculo sóleo apresentou maior reatividade, quando comparado ao músculo plantar, com grande quantidade de núcleos centralizados e halo basofílico. O treino excêntrico foi capaz de intensificar as respostas hipertrófica e angiogênica, principalmente no período de 21 dias de reabilitação. Portanto, o desenvolvimento muscular normal foi acompanhado por adaptações importantes que correspondem ao avançar da idade juntamente com o aumento da demanda funcional nessa fase. O dispositivo de imobilização foi eficaz para promover o desuso e encurtamento dos músculos avaliados. Diferenças nítidas entre a resposta adaptativa dos músculos sóleo e plantar são observadas em ambos os protocolos de reabilitação no animal bebê. / This study analyzed changes and adaptations in the soleus and plantar muscles induced by stretching and eccentric exercise protocols applied after 10 days immobilization of the hindlimb in developing female rats. Fortyfive Wistar female rats, 21 days old, were divided into 10 groups: Immobilized (IG); Immobilized and trained eccentrically for 10 days (IEG(10)) or 21 days (IEG(21)); Immobilized and stretched for 10 days (ISG(10)) or 21 days (ISG(21)); Anesthetized (AG); Control, 21 days old (CG(zero)); Control of immobilized group, 31 days old (CG(Immob)); Control group of 10 days, 41 days old (CG(10)) or 21 days, 52 days old (CG(21)). The animals had the right hindlimb immobilized for 10 days keeping the soleus and plantar mucles in a shortened position. Rats in the IEG(10) and IEG(21) groups were further submitted to eccentric training in a declining threadmill for 40 minutes, while the ones in groups ISG(10) and ISG(21) were stretched for 40 minutes by fixing the maximum ankle dorsiflexion. Fragments of the soleus and plantar muscles were removed and analyzed by histochemical, immunohistochemical and biochemical assays, after sacrifice of the animals. Qualitative and quantitative analysis were made of Myosin Heavy-Chain (MHC) isoforms, fiber proportion, minimum diameter and capillary/fiber ratio. Data was statistically analyzed by the method of Mixed Effects Linear Models with a significance level of 5% and confidence interval of 95%. Temporal analysis of muscle development suggested immaturity of soleus and plantar muscles (rounded fibers, voluminous nuclei and tissue reactivity) specially in animals aged from 21-31 days. Progressive increase of fiber diameter and the capillary/ fiber ratio was also observed. Predominance of FT1 fibers and higher MHCI expression were already evident in the soleus muscle even in 21 days old animals. Analysis of mATPase showed that FT2D fibers were predominant in the plantar muscle as were the isoforms MHCIId and MHCIIb. Immobilization produced high tissue reactivity, diameter reduction in all fiber types, changes in fiber proportion (reduction of FT1 in the soleus and increase of FT2D numbers in the plantar muscles) and MHC isoform expressions and decreased number of transversal capillars. Both, stretching and eccentric exercises induced important adaptations like increased fiber diameter, modified fiber proportions and increased capillary/ fiber ratios in the soleus and plantar muscles. Morphological analysis indicated that the soleus was more reactive as compared to the plantar muscle, showing large quantities of centralized nuclei and a basophilic halo. Eccentric training induced greater hypertrophic and angiogenic responses, specially during the 21 days rehabilitation. Thus, normal muscular development in this phase was accompanied by major adaptations, which correspond to advancing age and increased functional demand. The immobilization apparatus was efficient in preventing use and promoting shortening of the muscles evaluated. Sharp differences between adaptive responses of soleus and plantar muscles were observed in both rehabilitation protocols applied to baby animals.

De nos jours, les biocapteurs électrochimiques constituent des outils rapides, fiables, rentables et sans marquage pour la détection sur site des polluants environnementaux et l’évaluation des critères de sécurité alimentaire et pour la détection des résidus militaires sur le terrain. Nous avons développé des biocapteurs d’affinité basés sur des récepteurs biologiques pour la détection électrochimique de l'estradiol dans des échantillons environnementaux et de molécules odorantes dans l'adultération de viande de porc. Des biocapteurs sensibles, sélectifs, rapides, simples reproductible, stables et efficaces ont été élaborés. En outre, une autre stratégie basée sur des nanomatériaux intégrés a été employée pour élaborer un capteur biomimétique pour la détection d’explosifs, qui a montré de bonnes qualités dans la protection de l'environnement. A la fin du manuscrit, certaines perspectives sont discutées / Nowadays, electrochemical biosensors are rapid, reliable, cost-effective and label-free tools for in field monitoring of environmental pollutants, for assessing the food safety criterion and for detection of the territory military residues. We developed biological receptors based affinity biosensors for electrochemical detection of estradiol in environment samples and odorant molecules in pork meat adulteration. The sensitive, selective, rapid, simple, repetitive, stable and effective biosensors were produced. In addition, another strategy entrapment based on integrated nano-material was used to produce a biomimetic sensor for explosives detection, which has shown good valuable in environmental protection and homeland security. Développement d'une plateforme de criblage pour la recherche de nouvelles molécules anti-infectieuses : applications à Pseudomonas aeruginosa. / Glycoarray technology development for new anti-infective molecules discovering : applications to Pseudomonas aeruginosa

Pseudomonas aeruginosa (PA) est l’un des principaux germes impliqués dans les maladies nosocomiales et est aussi la principale cause de mortalité et morbidité des patients atteints de la mucoviscidose malgré l’utilisation massive d’antibiotiques. Dans la lutte contre PA, une alternative aux antibiotiques est l’inhibition de ses facteurs de virulence notamment ceux impliqués dans l’adhésion et la formation du biofilm via des interactions de type sucres/protéines. Ces protéines sont appelées lectines (PA-IL, PA-IIL, FliD). L’objectif de ce travail est la recherche de molécules inhibitrices (glycoclusters) de ces lectines impliquées dans la virulence de PA. Compte tenu du grand nombre de glycoclusters à tester et des faibles quantités de matériels biologiques disponibles, un outil de criblage innovant a été développé (glycoarray) à partir d’une lame de verre microstructurée et fonctionnalisée chimiquement afin d’immobiliser de manière organisée et ordonnée les glycoclusters. La méthode d’immobilisation choisie est la méthode d’immobilisation spécifique par hybridation de l’ADN appelée DDI : DNA Directed Immobilization. Sur ces glycoarrays, 3 méthodes indépendantes (lecture de fluorescence directe, IC50 et Kd) de mesure des interactions glycoclusters/lectines ont été mises au point et validées par une étude comparative donnant un classement similaire des glycoclusters pour leur affinité vis-à-vis des lectines Il faut noter que ces mesures faites sur glycoarrays ne consomment que quelques picomoles de glycoclusters comparées aux méthodes classiques (ITC, ELLA, RMN, …) qui nécessitent des micromoles de produits. A l’aide de ces glycoarrays, un criblage d’une bibliothèque d’une centaine de glycoclusters multivalents, de différentes topologies, charges et linkers a permis d’identifier deux structures montrant une très forte affinité vis-à-vis des lectines de PA. Ces glycoclusters sont actuellement en test in vitro et in vivo. Ces études d’interactions sur DDI-glycoarray ont été étendues à d’autres agents pathogènes tels que les bactéries Burkholderia ambifaria, Viscum album ou contre le virus de la grippe. Dans le futur, pour mieux appréhender les mécanismes d’interactions sucres/protéines, il serait intéressant de pouvoir suivre en temps réel ces interactions en utilisant des systèmes de détection sans marquage tel que, par exemple, la résonance plasmonique de surface. Aussi, le dernier chapitre donne les prémices d’une adaptation de la méthode DDI sur glycoarray sur surface d’or. / Pseudomonas aeruginosa (PA) is one of the predominant bacterium encountered in nosocomial infections. PA infections often lead to chronic inflammation and eventually to death despite aggressive antibiotic therapy. A promising approach is to inhibit the virulence factors of PA such as PA-IL, PA-IIL, FliD (lectins). Therefore, there is a great interest for studying carbohydrate/lectin interactions in order to design new treatments. The goal of this work is the research for inhibitory molecules (glycoclusters ) of these lectins involved in the virulence of PA. An innovative screening tool for studying carbohydrate/lectin interactions has been developed (glycoarray). Glycoarray are microstructured glass-slides, chemically functionalized in order to immobilize, organized and orderly, glycoclusters at the surface. The immobilization method is the specific immobilization method based on DNA hybridization called DDI (DNA Directed Immobilization). This miniaturized analytical biosystem allows multiplex test performed in one single microwell. Moreover, three independent methods of affinity measurement (direct fluorescence read-out, IC50 and Kd) have been developed and validated by a comparative study giving a similar ranking of glycoclusters for their affinity towards PA-IL. These measurements on glycoarrays consume only a few picomoles glycoclusters compared to conventional methods (ITC, ELLA...) that require micromoles of products. Using these glycoarrays, the screening of a library of hundreds of glycoclusters presenting different topologies, multivalencies, charges and linkers led to the identification of two structures showing a very strong affinity for PA lectins. These glycoclusters are currently in vitro assay and in vivo. These interaction studies on DDI-glycoarray were extended to other pathogens such as Burkholderia ambifaria bacteria, Viscum album or against the influenza virus. In the future, to better understand the mechanisms of sugar / protein interactions, it would be interesting to monitor in real time the interactions using label-free detection systems such as, for example, the surface plasmon resonance (SPR). Développement d’électrodes poreuses pour un bioréacteur pilote

Dans ce mémoire nous discutons le développement de l’électrode de travail d’un bioréacteur électrochimique, dispositif permettant de synthétiser suivant un procédé dit de « Chimie Verte » des substances chimiques à haute valeur ajoutée. L’électrode de travail étant le siège de la synthèse électrocatalytique en jeu, l’optimisation de sa structure a été étudiée dans le but de maximiser l’aire de sa surface active. L’élaboration d’électrodes macroporeuses hautement organisées et de taille définie par les dimensions du prototype du réacteur pilote, a pu être obtenue en utilisant la méthode de Langmuir-Blodgett pour assembler le cristal colloïdal servant de template. La formation de ce dépôt organisé de colloïdes est suivie de l’électrodéposition du matériau d’électrode puis de la dissolution du template afin de révéler la structure macroporeuse. L’immobilisation de l’intégralité du matériel bio-électrocatalytique à l’intérieur des pores a été investiguée dans le but de prévenir la pollution du milieu contenant le produit final d’électrosynthèse par un des constituants redox et d’augmenter la durée de vie du dispositif. Ainsi, des couches ultra-minces de silice électrogénérée et des matrices de polymère électrodéposé ont été étudiées dans le but de préserver et d’optimiser l’activité enzymatique du système qu’elles encapsulent. Une attention particulière a été portée sur la qualité des dépôts au sein des structures poreuses. La procédure d’immobilisation des protéines rédox dans les matrices de silice et de polymère a été en outre associée à un jeu de construction moléculaire qui a permis par l’instauration de diverses interactions électrostatiques, de retenir toutes les espèces responsables de la catalyse à la surface de l’électrode. Enfin, dans le but d’intensifier les réactions catalytiques responsables de la synthèse à réaliser, des nano-particules d’ormodifiées par une couche monomoléculaire d’un médiateur redox ont été incorporées aux différents matériaux d’immobilisation permettant de ce fait d’augmenter les interfaces d’échanges électrochimiques entre matériau conducteur et biomolécules. L’insertion de ces nano-objectscombinée à la nanostructuration du matériau d’électrode a permis de multiplier par plus de 170 fois l’intensité des réactions enregistrées. / The present work deals with the development of the working electrode of an electrochemicalbioreactor. This device enables the green synthesis of high added value chemical compounds. As theelectrochemical synthesis is located at the interface of the working electrode, structural optimizationof this reactor key component is required in order to maximize the available active surface area.Elaboration of highly organized macroporous gold electrodes with a size required by the pilot reactordimensions were obtained with the Langmuir-Blodgett method that was used to assemble a colloidalcrystal as a template. The elaboration of the organized colloidal deposit is first followed by theelectrodeposition of the electrode material, then by the dissolution of the template. The immobilization of the complete bio-electrochemical system inside the electrode pores was investigated in order to prevent pollution of the final product medium by one of the catalytic chaincomponent. This also improves the device life time. Subsequently electrogenerated ultra-thin silicalayers and electrodeposited polymer matrices were studied in order to preserve and optimize the catalytic activity of the redox proteins. In order to enhance the electrocatalytic synthesis, mediatormodified gold nanoparticules were incorporated in the different immobilization matrices. This allowed to increase the area of the electrochemical interface. The combination of the nano-objectincorporation and electrode nano-structuring intensified by a factor of 170 the catalytic process. / Die vorliegende Arbeit beschäftigt sich mit der Entwicklung einer Arbeitselektrode für einenelektrochemischen Bioreaktor, der die umweltfreundliche Synthese von wertvollen chemischenKomponenten ermöglicht. Da die elektrochemische Synthese an der Oberfläche der Arbeitselektrodestattfindet, ist es nötig, den strukturellen Aufbau der Schlüsselkomponente des Reaktors zuoptimieren und die aktive Oberfläche der Elektrode zu erhöhen. Mit Hilfe der Langmuir-BlodgettTechnik wurden kolloidale Kristalle erzeugt, die als Template dienten, um hochgeordnetemakroporöse Goldelektroden, deren Dimensionen von dem Pilotreaktor bestimmt wurden,herzustellen. Nach dem Erzeugen von geordneten kolloidalen Filmen wurde der Zwischenraumzwischen den Partikeln mittels elektrochemischer Abscheidung gefüllt und das Templateanschließend chemisch aufgelöst. In der Folge wurde die Immobilisierung des komplettenbioelektrochemischen Systems im Poreninnenraum untersucht, mit dem Ziel eine Verunreinigung desReaktionsmediums durch eine der katalytischen Komponenten zu verhindern. Die Lebensdauer derElektrode kann so zusätzlich erhöht werden. Es wurde untersucht, inwieweit durch elektrogenerierteultra-dünne Silikaschichten oder durch Elektroabscheidung erzeugte Polymerfilme die katalytischeAktivität der Redoxproteine erhalten und weiter optimiert werden kann. Goldnanopartikel, die miteinem Mediator modifiziert wurden, wurden in die jeweilige Immobilisationsschicht integriert, mitdem Ziel die Effizienz der elektrokatalytischen Synthese zu erhöhen. Auf diese Weise konnte dieaktive elektrochemische Oberfläche der Elektrode weiter erhöht werden. Die Kombination aus einernanostrukturierten Elektrode und Nanoobjekten die in die Immobilisationsschicht eingebettetwurden, führte zu einer Signalerhöhung des katalytischen Prozesses um mehr als eineGrössenordnung.

La nueva normalidad a raíz del COVID-19 exige un cambio en todos los sectores productivos a nivel mundial. En este sentido el sector construcción no se exime de establecer nuevos procedimientos al costo de obras, tales como las obras de vivienda multifamiliares, para lo cual se presenta una guía con la finalidad de medir el impacto de la pandemia. El proceso para la elaboración de la guía tiene como inicio el diagnóstico de la variación de costos, para lo cual se tuvo información de tres obras de viviendas multifamiliares. El análisis de los tres casos de estudio se realiza a las partidas con mayor relevancia de la especialidad de estructuras para, posteriormente evaluar su comportamiento entre las etapas pre y post inmovilización social, pudiendo de esa forma determinar el impacto por la pandemia. Teniendo en cuenta que esta investigación es de nivel explicativo, por analizar la causa y el efecto, la rigurosidad estadística está presente con el apoyo de los expertos que dieron sus opiniones en base a la guía, lo que permite mejorar la metodología y otros elementos para la perfección de esta. Determinar el impacto en los costos de la especialidad de estructuras para obras de viviendas multifamiliares trae consigo el análisis a los recursos: mano de obra, materiales, equipos y subcontratos. Asimismo, la variación en sus costos se relación con la implementación de lineamientos para la vigilancia y prevención y control de la salud de los trabajadores con riesgo de exposición establecidos mediante Resolución Ministerial Nº 448.2020.MINSA. / The new normality as a result of COVID-19 demands a change in all productive sectors worldwide. In this sense, the construction sector is not exempt from establishing new procedures and modifications to the cost of constructions, such as multifamily housing works, for which a guide is presented in order to measure the impact of the pandemic. The process for the elaboration of the guide has started with the diagnosis of the variation of costs, for which information was obtained from three multifamily housing projects. The analysis of the three case studies is carried out on the most relevant items of the specialty of structures in order to evaluate their behavior between the pre and post social immobilization stages, thus being able to determine the impact of the pandemic. Considering that this research is of explanatory level, by analyzing the cause and effect, the statistical rigor is present with the support of the experts who gave their opinions based on the guide, which allows improving the methodology and other elements for the perfection of this one. Determining the impact on the costs of the specialty of structures for multifamily housing projects involves the analysis of resources: labor, materials, equipment and subcontracts. Biochemical processes for Balsamic-styled vinegar engineering

These challenges include the ongoing drought, bush fires, climate change and several liquor amendment bills aimed at reducing alcohol consumption and alcohol outlets in South Africa. It is therefore critical for the wine industry to expand and find alternative ways in which sub-standard or surplus wine grapes can be used to prevent income losses and increase employment opportunities. Traditional Balsamic Vinegar (TBV) is a geographically and legislative protected product produced only in a small region in Italy. However, the methodology can be used to produce similar vinegars in other regions. Balsamic-styled vinegar (BSV), as defined in this thesis, is a vinegar produced by partially following the methods of TBV while applying process augmentation techniques. Balsamic-styled vinegar is proposed to be a suitable product of sub-standard quality or surplus wine grapes in South Africa. However, the production of BSV necessitates the use of cooked (high sugar) grape must which is a less favourable environment to the microorganisms used during fermentation. Factors that negatively affect the survival of the microorganisms include low water activity due to the cooking, high osmotic pressure and high acidity. To counteract these effects, methods to improve the survival of the non-Saccharomyces yeasts and acetic acid bacteria used are essential. The primary aim of this study was to investigate several BSV process augmentation techniques such as, aeration, agitation, cell immobilization, immobilized cell reusability and oxygen mass transfer kinetics in order to improve the performance of the microbial consortium used during BSV production. The work for this study was divided into four (4) phases. For all the phases a microbial consortium consisting of non-Saccharomyces yeasts (n=5) and acetic acid bacteria (n=5) was used. Inoculation of the yeast and bacteria occurred simultaneously. The 1st phase of the study entailed evaluating the effect of cells immobilized by gel entrapment in Ca-alginate beads alongside with free-floating cells (FFC) during the production of BSV. Two Ca-alginate bead sizes were tested i.e. small (4.5 mm) and large (8.5 mm) beads to evaluate the effects of surface area or bead size on the overall acetification rates. Ca-alginate beads and FFC fermentations were also evaluated under static and agitated (135 rpm) conditions. The 2nd phase of the study involved studying the cell adsorption technique for cell immobilization which was carried-out using corncobs (CC) and oak wood chips (OWC), while comparing to FFC fermentations. At this phase of the study, other vinegar bioreactor parameters such as agitation and aeration were studied in contrast to static fermentations. One agitation setting (135 rpm) and two aeration settings were tested i.e. high (0.3 vvm min−1) and low (0.15 vvm min−1) aeration conditions. Furthermore, to assess the variations in cell adsorption capabilities among individual yeast and AAB cells, the quantification of cells adsorbed on CC and OWC prior- and post-fermentation was conducted using the dry cell weight method. The 3rd phase of the study entailed evaluating the reusability abilities of all the matrices (small Ca-alginate beads, CC and OWC) for successive fermentations. The immobilized cells were evaluated for reusability on two cycles of fermentation under static conditions. Furthermore, the matrices used for cell immobilization were further analysed for structure integrity by scanning electron microscopy (SEM) before and after the 1st cycle of fermentations. The 3rd phase of the study also involved the sensorial (aroma and taste) evaluations of the BSV’s obtained from the 1st cycle of fermentation in order to understand the sensorial effects of the Ca-alginate beads, CC and OWC on the final BSV. The 4th phase of the study investigated oxygen mass transfer kinetics during non-aerated and aerated BSV fermentation. The dynamic method was used to generate several dissolved oxygen profiles at different stages of the fermentation. Consequently, the data obtained from the dynamic method was used to compute several oxygen mass transfer parameters, these include oxygen uptake rate ( 𝑟𝑟𝑂𝑂2 ), the stoichiometric coefficient of oxygen consumption vs acid yield (𝑌𝑌𝑂𝑂/𝐴𝐴), the oxygen transfer rate (𝑁𝑁𝑂𝑂2 ), and the volumetric mass transfer coefficients (𝐾𝐾𝐿𝐿𝑎𝑎). During all the phases of the study samples were extracted on weekly intervals to evaluate pH, sugar, salinity, alcohol and total acidity using several analytical instruments. The 4th phase of the study involved additional analytical tools, i.e. an oxygen µsensor to evaluate dissolved oxygen and the ‘Speedy breedy’ to measure the respiratory activity of the microbial consortium used during fermentation. The data obtained from the 1st phase of the study demonstrated that smaller Ca-alginate beads resulted in higher (4.0 g L-1 day−1) acetification rates compared to larger (3.0 g L-1 day−1) beads, while freely suspended cells resulted in the lowest (0.6 g L-1 day−1) acetification rates. The results showed that the surface area of the beads had a substantial impact on the acetification rates when gel entrapped cells were used for BSV fermentation. The 2nd phase results showed high acetification rates (2.7 g L-1 day−1) for cells immobilized on CC in contrast to cells immobilized on OWC and FFC, which resulted in similar and lower acetification rates. Agitated fermentations were unsuccessful for all the treatments (CC, OWC and FFC) studied. Agitation was therefore assumed to have promoted cell shear stress causing insufficient acetification during fermentations. Low aerated fermentations resulted in better acetification rates between 1.45–1.56 g L-1 day−1 for CC, OWC and FFC. At a higher aeration setting, only free-floating cells were able to complete fermentations with an acetification rate of 1.2 g L-1 day−1. Furthermore, the adsorption competence data showed successful adsorption on CC and OWC for both yeasts and AAB with variations in adsorption efficiencies, whereby OWC displayed a lower cell adsorption capability compared to CC. On the other hand, OWC were less efficient adsorbents due to their smooth surface, while the rough surface and porosity of CC led to improved adsorption and, therefore, enhanced acetification rates. The 3rd phase results showed a substantial decline in acetification rates on the 2nd cycle of fermentations when cells immobilized on CC and OWC were reused. While cells entrapped in Ca-alginate beads were able to complete the 2nd cycle of fermentations at reduced acetification rates compared to the 1st cycle of fermentations. The sensory results showed positive ratings for BSV’s produced using cells immobilized in Ca-alginate beads and CC. However, BSV’s produced using OWC treatments were neither ‘liked nor disliked’ by the judges. The SEM imaging results further showed a substantial loss of structural integrity for Ca-alginate beads after the 1st cycle fermentations, with minor changes in structural integrity of CC being observed after the 1st cycle fermentations. OWC displayed the same morphological structure before and after the 1st cycle fermentations which was attributed to their robustness. Although Ca-alginate beads showed a loss in structural integrity, it was still assumed that Ca-alginate beads provided better protection against the harsh environmental conditions in contrast to CC and OWC adsorbents due to the acetification rates obtained on both cycles. The 4th phase data obtained from the computations showed that non-aerated fermentations had a higher 𝑌𝑌𝑂𝑂/𝐴𝐴, 𝑟𝑟𝑂𝑂2 , 𝑁𝑁𝑂𝑂2 and a higher 𝐾𝐾𝐿𝐿𝑎𝑎 . It was clear that aerated fermentations had a lower aeration capacity due to an inappropriate aeration system design and an inappropriate fermentor. Consequently, aeration led to several detrimental biochemical changes in the fermentation medium thus affecting 𝐾𝐾𝐿𝐿𝑎𝑎 and several oxygen mass transfer parameters which serve as a driving force. Overall, it was concluded that the best method for BSV production is the use of cells entrapped in small alginate beads or cells adsorbed on CC under static and non-aerated fermentations. This conclusion was based on several factors such as cell affinity/cell protection, acetification rates, fermentation period and sensorial contributions. However, cells entrapped in Ca-alginate beads had the highest acetification rates. The oxygen mass transfer computations demonstrated a high 𝐾𝐾𝐿𝐿𝑎𝑎 when Ca-alginate beads were used under static-non-aerated conditions compared to fermentations treated with CC. Therefore, a fermentor with a high aeration capacity needs to be designed to best suit the two BSV production systems (Ca-alginate beads and CC). It is also crucial to develop methods which can increase the robustness of Ca-alginate beads in order to improve cell retention and reduce the loss of structural integrity for subsequent cycles of fermentation. Studies to define parameters used for upscaling the BSV production process for large scale productions are also crucial.

Efekti primene različitih postupaka remedijacije na imobilizaciju teških metala u sedimentu / The effects of remediation treatments application on the sediment heavy metal immobilization

<p>Predmet izučavanja ove disertacije, osim poređenja različitih metoda procene rizika na&nbsp;osnovu karakterizacije kvaliteta sedimenta vodotokova Vojvodine, bilo je i ispitivanje&nbsp;mogućnosti imobilizacije te&scaron;kih metalau sedimentu primenom različitih remedijacionih&nbsp;tehnika kao i određivanje njihove efikasnosti (npr. solidifikacija/stabilizacija silikatnim&nbsp;materijalima, portland cementom, kalcijum-oksidom itd.) u zavisnosti od brojnih faktora.&nbsp;Osim pseudo-ukupnog sadržaja metala određene su i specifičnije frakcije metala sa posebnim<br />osvrtom na određivanje potencijalno biodostupnih frakcija.</p><p>Rezultati su ukazali da je naosnovu holandskih preporuka sediment pojedinih vodotokova&nbsp;Vojvodine (Begej, DTD-kanal,Nadela, Sava-&Scaron;abac) zagađen metalima (klasa 4) i da je&nbsp;neohodno izmuljivanje i remedijacija. Prema USEPA i kanadskim preporukama, neki uzorci&nbsp;su potencijalno toksični, ali nije zabeležena akutna toksičnost za test vrste u pornoj vodi. Za&nbsp;neke uzorke, i pored visoke pseudo-ukupne koncentracije (klasa 4), nije potvrđena&nbsp;potencijalna toksičnost na osnovu odnosa kiselog volatilnog sulfida i simultano ekstrahovanih&nbsp;metala. Dok se ne primene skuplje metode remedijacije i tretmana sedimenta, najbolje re&scaron;enje&nbsp;je izolovano skladi&scaron;tenje na specijalnim&nbsp; deponijama. Naredni koraci bi trebali da budu&nbsp;usresređeni na kontrolu i sprečavanje zagađenja kako bi se obezbedilo da revitalizacija ovih&nbsp;vodotoka ima trajni pozitivan uticaj na životnu sredinu i nesmetanu i bezbednu plovidbu, kao&nbsp;i na primenu određenih remedijacionih metoda. Poređenje rezultata koji su dobijeni različitim&nbsp;procenama kvaliteta sedimenta pokazalo je da ponekad nije dovoljan jedan pristup i da je&nbsp;potrebno je uključiti metode procene biodostupnosti, biotestove i aspekt radioaktivnosti. U&nbsp;uzorku sedimenta koji je kori&scaron;ćen za ispitivanje efikasnosti remedijacionih tretmana,&nbsp;pokazano je da cink, nikal i olovo imaju visok rizik po okolinu, jer seprocenat metala u&nbsp;izmenljivoj i karbonatnoj fazi kretao od 40.1 do 45.2%. Cr i Cd imaju umeren rizik, dok je&nbsp;bakar prisutan u ovim frakcijama u koncentraciji koja predviđa nizak rizik (5.3%). Ovo je u&nbsp;skladu i sa rezultatima analize porne vode i simultano ekstrahovanih metala i kiselog&nbsp;volatilnog sulfida.</p><p>Istraživanje prikazano u radu je dalo odgovor na pitanje da li remedijacija može uspe&scaron;no da&nbsp;ukloni zagađenje, u smislu imobilizacije metala u stanje kada oni vi&scaron;e neće biti opasnost po&nbsp;okolinu. Svi primenjeni tretmani doveli su dosmanjenja procenta kumulativno izluženih&nbsp;metala iz S/S sme&scaron;a, ali nijedna sme&scaron;a tretiranog sedimenta i ispitivanih imobilizacionih&nbsp;agenasa ne pripada grupi inertnog otpada ukoliko se kumulativne izlužene koncentracije&nbsp;metala porede sa koncentracijama koje za otpad propisuje Evropska Unija (2003/33/EC).&nbsp;Ukoliko nam je cilj dobijanje nehazardnog otpada onda moramo primeniti u tretmanu&nbsp;sedimenta kontaminiranog metalima najmanje 30% imobilizaconih agenasa (cementa,&nbsp;kalcijum-oksida). Najveća efikasnost u imobilizaciji metala postignuta je kori&scaron;ćenjem&nbsp;cementa i kalcijum-oksida u sme&scaron;i (30% cementa i 10% kalcijum-oksida) i primenom termičkog tretmana na vi&scaron;oj temperaturi (1100<sup>0</sup>C) sa glinom (20% gline) &scaron;to je zaključeno na osnovu koeficijenata difuzije i indeksa izlužljivosti. Dominantan mehanizam izluživanjametala iz ovih sme&scaron;a je difuzija.</p> / <p>This work is concerned with the comparison of the different methods of metal risk assessment&nbsp;in sediments based on examination of the qualityof water courses in Vojvodina. Besides, it&nbsp;deals with the possibility of applying remediation treatments of contaminated sediment and&nbsp;with the efficiency of the different methods, that is the techniques, for immobilization of&nbsp;sediment heavy metals by applying S/S and thermal treatments. A dominant mechanism is&nbsp;proposed for the process of leaching ofthe metals from treated mixtures.</p><p>Comparison of the results obtained by the different methods of sediment quality assessment&nbsp;showed that in some cases one approach to solving this problem is not &nbsp;sufficient. Metal&nbsp;concentrations in particular sediment samples (the Begej, the DTD Canal, the Nadela, the&nbsp;Sava at &Scaron;abac) indicate the presence of contamination, the analyzed samples being of Class 4.&nbsp;According to the Dutch regulations, a sedimentof &nbsp;Class 4 is of unacceptable quality and&nbsp;requires urgent intervention in the sense of sediment dredging, disposal into special depots,&nbsp;and, if possible, remediation. For some samples, despite a high pseudo-total concentration&nbsp;(Class 4), no potential toxicity was confirmed onthe basis of the ratioof the acidic volatile&nbsp;sulfide and simultaneously extracted metals. The results showed that, apart from chemical&nbsp;analyses, biological tests are also necessary, but also sequential extraction analysis, which can&nbsp;more clearly define the way of metal binding to the particular sediment fractions, to allow a&nbsp;more reliable prediction of metal mobility, potential toxicity and bioavailability. In the&nbsp;analysis of sediment quality it is also necessary toinclude the aspect of radioactivity, as it has&nbsp;been shown that the results of this analysis can, not only confirm some of the results of the&nbsp;other analyses, but also indicate the sediment age, origin of contamination and potential&nbsp;toxicity.The subsequent steps should be directed to the control and prevention of&nbsp;contamination, in order to ensure that the water course reviatalization has a lasting positive&nbsp;effect on the environment, an unimpared and safe navigation and application of remediation&nbsp;methods.</p><p>In the sediment sample (Class 4) that was usedfor the examination of the efficiency of it&nbsp;appeared that zinc, nickel and lead exhibit a high risk, as the percentages of these metals in&nbsp;the exchangeable and carbonate phases were in the range from 40.1 &nbsp;to 45.2%. On the other&nbsp;hand, chromium and cadmium exhibited a moderaterisk, whereas copperin these fractions&nbsp;was present at the levels corresponding to a low risk (5.3%). This is alsoin agreement with&nbsp;the results of pore water analysis and simultaneously extracted metals and acid volatile&nbsp;sulphide examinations.</p><p>The investigations presented in this work provided an answer to the question whetehr the&nbsp;remediation can successfully remove the contamination in the sense of the immobilization of&nbsp;metals in a state that will not be harmful tothe environment. All the treatments applied&nbsp;yielded a decrease of the percentage of cumulatively leached metals from the S/S mixtures,&nbsp;but none of the mixtures of treated sediments with the tested immobilization agents belongs to&nbsp;the group of inert materials if the cumulative leached concentrations are compared with the&nbsp;concntrations for the wastes given by the EU legislation (2003/33/EC). Ifthe goal is to obtain a non-hazardous material it is necessary to treat the contaminatedsediment with at least 30% of the immobilizing agent (cement or calcium oxide). Based on the diffusion coefficients and leachability index, the highest immobilization efficiency was achieved using a mixture of cement and calcium oxide (30% of cement and 10% of CaO) and by applying thermal treatment at a higher temperature (1100<sup>0</sup>C) with clay (20%). A dominant mechanism of leaching metals from these mixtures is diffusion.</p>
