Dissecting Phenotypic Variation in Pigmentation using Forward and Reverse GeneticsHellström, Anders R January 2010 (has links)
Coat color and patterning phenotypes have been extensively studied as a model for advancing our understanding of the relationship between genetic and phenotypic variation. In this thesis, genes of relevance for pigment cell biology were investigated. The dissertation is divided in two parts. Forward genetics was used in the first part (Paper I and II) to identify the genes controlling the Silver and Sex-linked barring loci in chicken. In the second part, reverse genetics was employed to create a mouse line in which the PMEL17 protein is inactivated (Paper III). In Paper I, we report five mutations in SLC45A2 causing plumage color variants in both chicken and Japanese quail. Normal function of the SLC45A2 gene has previously been shown to be essential for the synthesis of both red/yellow pigment (pheomelanin) and brown/black pigment (eumelanin) in numerous species, including humans. The major discovery in this paper is the specific inhibition of pheomelanin in Silver chickens, whilst null mutations at this locus cause an almost complete absence of both pheomelanin and eumelanin. In Paper II, we report that Sex-linked barring in chickens is controlled by the CDKN2A/B tumor suppressor locus. The locus encodes two proteins, INK4B and ARF. The genetic analysis indicates that missense mutations in ARF or mutations in the promoter region of the ARF transcript are causing Sex-linked barring. In previous studies, mutations inactivating the CDKN2A/B tumor suppressor locus, have been shown to be responsible for familiar forms of human melanoma. Here we propose that these mutations in chicken CDKN2A/B cause the premature cell death of melanocytes as opposed to the cell proliferation and tumor growth associated with loss-of-function alleles in humans. In Paper III, we created a mouse line in which the PMEL17 protein is inactivated. Missense mutations in the gene encoding PMEL17 have previously been shown to be associated with reduced levels of eumelanin in epidermal tissues in several vertebrate species. The knockout mice are viable, fertile, and display no obvious developmental defects. The eumelanosomes within the melanocytes of these mice are spherical in contrast to the cigar-like shaped eumelanosomes present in wild-type animals. PMEL17 protein inactivation has only a subtle diluting effect on the coat color phenotype in four different genetic backgrounds. This suggests that other previously described alleles in vertebrates with more striking effects on pigmentation are dominant-negative mutations.

Animal models are crucial to improve the knowledge of the mechanisms underlying the different pathological processes. These models are also excellent tools to facilitate the research of new targets for the treatment of different diseases and to evaluate the benefit/risk ratio of the potential new treatments. We have focussed this research work in the study of a new potential targets for pain, cognitive and eating disorders using new animal models developed in our laboratory. We first investigated the effects of the interaction between cannabinoids and nicotine on cognitive processes and metabolism using different behavioural models and new experimental devices. In a second part of this work, we investigated new therapeutic targets for neuropathic pain and for this purpose we developed a new behavioural model to improve the study of the therapeutic potential and possible side-effects of novel compounds. / Los modelos animales son cruciales para mejorar el conocimiento sobre los mecanismos que constituyen la base de los diversos procesos patológicos. Estos modelos representan también excelentes herramientas para facilitar la investigación de nuevas dianas para el tratamiento de estas enfermedades y para evaluar el cociente beneficio/riesgo de los nuevos tratamientos potenciales. Este trabajo de investigación se encuentra centrado en el estudio de nuevos dianas terapéuticas para el dolor, los procesos cognitivos y los desórdenes alimentarios utilizando nuevos modelos animales desarrollados en nuestro laboratorio. En primer lugar, hemos investigado los efectos de la interacción entre los cannabinoinoides y la nicotina a nivel los procesos cognitivos y del metabolismo usando diversos modelos comportamentales y nuevos dispositivos experimentales. En una segunda parte de este trabajo, hemos estudiado nuevas dianas terapéuticas para el dolor neuropático y hemos desarrollado para este propósito un nuevo modelo comportamental que permite evaluar el potencial terapéutico y los posibles efectos secundarios de nuevos compuestos.

The aim of this thesis was to clarify the influence of the stress on the adrenergic and muscarinic receptors in the heart and in the lungs. Research was perform on rat hearts and lungs and on the hearts and lungs of the CRH KO mice. First, we assessed mRNA levels of all α- and β-adrenergic receptor and muscarinic receptor subtypes. Subsequently, we performed the radioligand-binding studies to determine densities of these receptors. We identified all three α1-adrenergic receptor subtypes in the rat lungs. In the lungs of WT mice, we found that the amount of α1-adrenergic and muscarinic receptors was sex-dependent. Densities of the former were higher in females and those of the latter were higher in males. There was no difference between males and females in β-adrenergic receptor density. As for CRH KO mice, the basal densities of studied receptors were lower than in CRH WT mice (except β1-adrenergic receptors in females). The main purpose of the thesis was to detect the immobilization-induced changes in the studied receptors in the kontrol (WT) and CRH KO mice. Short-term and long-term immobilization caused decrease in all α1-adrenergic receptor subtypes in females, whereas only α1A-adrenergic receptors decreased in males. A insuficiência cardíaca (IC) é uma síndrome clínica de alta incidência e mau prognóstico, caracterizada por fadiga, dispnéia e grande limitação aos esforços físicos. Essas alterações não estão apenas limitadas ao comprometimento cardíaco, mas em parte, são decorrentes também de alterações morfo-funcionais da musculatura esquelética. Para a dissertação foram utilizados camundongos com deleção dos genes para os receptores ?2A e ?2C adrenérgicos (KO) que desenvolvem cardiomiopatia induzida por hiperatividade simpática, associada à sinais clínicos de IC e 50% de mortalidade aos sete meses de idade. Foi objetivo desse estudo realizar a caracterização fenotípica do músculo esquelético por meio de avaliações funcionais e morfológicas em camundongos KO previamente ao desenvolvimento da IC (três meses de idade), e ao longo de sua progressão (cinco e sete meses de idade). Somente na faixa etária de sete meses de idades foi constatado o estabelecimento da miopatia muscular esquelética. Nessa fase, observou-se rarefação vascular, atrofia muscular, aumento na porcentagem de fibras glicolíticas e redução na atividade máxima da enzima citrato sintase, que em conjunto, contribuem para a antecipação da fadiga observada nesse modelo, e como conseqüência, para a redução da tolerância aos esforços. Os resultados sugerem que os camundongos KO apresentam alterações morfo-funcionais da musculatura esquelética semelhantes às observadas nos demais modelos de IC e em indivíduos portadores dessa síndrome. Portanto, sendo um ótimo modelo experimental para estudos de futuras estratégias terapêuticas que visem minimizar as alterações na musculatura esquelética decorrentes da IC / Heart failure (HF) is a clinical syndrome with high incidence and bad prognostic, characterized by fatigue, dyspnea, and increased intolerance to exercise. These changes are not only related to the cardiovascular tissue, but are at least in part, consequence of morphofunctional alterations in skeletal muscles. To the present study it was used mice lacking both ?2A/?2C AR subtypes (KO) which develop cardiomyopathy induced by sympathetic hyperactivity, associated to clinical signals of HF and 50% of mortality at seven months of age. The aim of the present study was characterize the phenotype of skeletal muscle by functional and morphological evaluations in KO before (three months of age) and during the HF progression (five and seven months of age). Skeletal muscle alterations due HF were observed only at seven months of age. The alterations were characterized by vascular rarefaction, muscular atrophy, increase in glycolitic fibers percentage and reduction of maximal activity of citrate synthase and contributed with early fatigue observed in this model, and consequently, exercise intolerance. The results of present study suggest that KO mice present morphofunctional changes in skeletal muscles in a similarly to others models of HF and in patients that have this syndrome. Therefore, consisting in an excellent experimental model to future studies related to therapeutic strategies to minimize the skeletal muscle changes due HF

An In-vivo Analysis of SLMAP Function in the Postnatal Mouse MyocardiumRehmani, Taha January 2017 (has links)
SLMAP is a tail anchored membrane protein that alternatively splices to generate three isoforms, SLMAP1, SLMAP2 and SLMAP3. Previous studies in our lab have shown that the postnatal cardiac-specific overexpression of SLMAP1 results in intracellular vesicle expansion and enhanced endosomal recycling. I generated a postnatal cardiac-specific knockout model using the Cre-Lox system to nullify all three SLMAP isoforms and further evaluate its role in the mouse myocardium. SLMAP knockdown and knockout mouse hearts were analyzed with western blotting and qPCR. I found that only SLMAP3 was nullified and phenotypic evaluation through echocardiography indicated that young and old SLMAP3 knockout animals showed no remarkable changes in cardiac function. Furthermore, challenge with stressor isoproterenol had a similar response to wildtype and knockout mice in cardiac structure and function. Surprisingly the level of expression of SLMAP1 and SLMAP2 was maintained in the myocardium from SLMAP3 deficient mice. Interestingly the machinery involved in endosomal recycling was not impacted by the loss of SLMAP3. These data indicate that loss of SLMAP3 does not alter cardiac structure and function in the postnatal myocardium in the presence of SLMAP1 and SLMAP2.

Characterization of Calcium Homeostasis Parameters in TRPV3 and CaV3.2 Double Null MiceMehregan, Aujan 19 December 2017 (has links)
In mammals, calcium influx is required for oocyte maturation and egg activation, as it supports the persistent calcium oscillations induced by fertilization. These oscillations are required for the initiation of embryo development. The molecular identities of the plasma membrane calcium-permeant channels that underlie calcium influx are not established. Among these channels, Transient Receptor Potential Vanilloid, member 3 (TRPV3) allows divalent cations, namely strontium (Sr2+) and calcium (Ca2+) with high permeability, into cells, and its expression pattern seems to predict an essential role in the initiation of development. Another channel that was identified to be expressed in oocytes/eggs is the low-voltage-activated T-type channel, CaV3.2. However, the ability to accurately probe the expression and function of these channels on Ca2+ homeostasis in mouse eggs is hindered by the lack of specific and known pharmacological agents and antibodies for these channels.
Here, we simultaneously knockout out these two Ca2+ influx channels in the mouse to explore the effects on Ca2+ homeostasis. We examined fertility rates, development, and morphological defects that arose from the double null pups. Next, we investigated the consequences on Ca2+ store content in immature and mature oocytes and eggs. We also examined the effects on fertilization-induced Ca2+ oscillations in response to in vitro fertilization and PLCz cRNA microinjection. We found that female mice null for these channels display drastic subfertility compared to the single knockout mice for these channels. Additionally, the Ca2+ store content is significantly diminished in double knockout eggs versus controls, as was the frequency of the fertilization-induced Ca2+ oscillations. These results suggest that these channels play a crucial role in Ca2+ influx during maturation and contribute to maintain Ca2+ oscillations post-fertilization. These null oocytes and eggs will be an important tool to perform electrophysiological studies to accurately measure the native current(s) of a specific channel(s) in eggs, and to identify the channel(s) that mediate Ca2+ during fertilization.

In the mid-eighties, the development of radioactive-ion beams enabled the exploration ofregions of the nuclear landscape away from the valley of stability. Close to the neutrondripline, in the light neutron-rich region, halo nuclei were observed. These nuclei exhibit asurprisingly large matter radius and a strongly clusterized structure. These two featurescan be explained by the weak binding of one or two neutrons which allows them to tunnelfar from the rest of the nucleons, surrounding the nucleus by a diffuse halo. These nuclearstructures have challenged the usual description of the nucleus, described as a compactmany-body object with nucleons piling up into well defined orbitals. Because they areshort-lived, these nuclei are often studied through reaction processes, such as elasticscattering, breakup and knockout. To infer precise information from the experimentaldata, an accurate reaction model coupled with a realistic description of the nucleus isneeded.Compared to other state-of-the-art methods, the eikonal approximation is very cheapfrom a computational viewpoint. This model assumes that the projectile-target relativemotion does not differ much from the initial plane wave. It also makes the adiabaticapproximation, which sees the internal coordinates of the projectile as frozen during thecollision. These two assumptions hold for reactions occurring at high energy, i.e. above60 MeV/nucleon, in which the deflection of the projectile by the target is small and thecollision time is brief.In this thesis, I focus on improvements of the eikonal approximation. First, I studythe extension of the validity of the eikonal model down to 10 MeV/nucleon, in the energyrange of the facilities HIE-ISOLDE at CERN and ReA12 at the upcoming FRIB. To thisend, I analyse different corrections to the eikonal approximation, which account for thedeflection of the projectile by the target. I assess their accuracy for the elastic-scatteringand breakup observables of one-neutron halo nuclei at 10 MeV/nucleon. Next, I developa dynamical correction to the eikonal approximation, which applies to both nuclear andCoulomb interactions while conserving the eikonal numerical cost. I study this correctionin the cases of breakup reactions of one-neutron halo nuclei on light and heavy targets.Then, I investigate which nuclear-structure information can be inferred from knockoutreactions of one-neutron halo nuclei. To do so, I conduct a sensitivity analysis of theirobservables to the nuclear structure of the projectile, described within a halo effectivefield theory.
