Regulation of Leaf Margin Development by TOOTH/MIR160A in Arabidopsis Thaliana

TOOTH/MIR160A regulates leaf margin outgrowth in Arabidopsis thaliana Unlike animals, a striking aspect of the plant development is that they have evolved a flexible pattern of post embryonic development. This exposes them to the challenges of many biotic and abiotic signals throughout their life. So, plants have to evolve/regulate various mechanisms to modulate their growth and development for accomplishing a successful life cycle in the prevailing environmental conditions. Auxin is involved in the initiation of lateral organs at the meristem and serration development along the leaf margin (Bilsborough et al., 2011, Hay et al., 2006). These two developmental mechanisms share common molecular players. For example, CUC2 is required for the boundary formation at the SAM and also is shown to be essential for serration formation at the leaf margin. Similarly, tth shows increased leaf serration phenotype as well as defects in the positioning of flowers at the meristem. This demonstrates the functional significance of TTH-regulated ARFs in controlling auxin mediated developmental pathways. Leaves originate as small lumps of undifferentiated cells at the flanks of the shoot apical meristem which undergo several rounds division and expansion to generate the mature leaf with characteristic size, shape and leaf margin. Both, endogenous as well as environmental factors modulate the growth and development of a leaf. This is evident from the plasticity in leaf form, observed during the life time of a single plant, as well as from the diversity among closely related species living in different habitats. It is well known that pathways controlling leaf form are subjected to the effects of selection and adaptation. Leaf margin is a key feature of the final leaf shape and it contributes to the abundant diversity in leaf form. Leaf margin architecture varies quite significantly from smooth or entire margin to margins with large outgrowths (lobed margins). The evolution and ecological advantages of this diversity is a subject of intense investigation. It also provides a wonderful system to study the mechanistic details of iterative generation of repeated units, which is a common feature in producing many biological shapes. Recent advances in molecular technologies and the availability of genomic resources ushered the identification of new factors involved in leaf margin development. Our current knowledge of this developmental programme is that CUC2 establishes auxin maxima at the leaf margin by reorienting an auxin efflux carrier PIN1 which ultimately results in serration outgrowth (Bilsborough et al., 2011, Hay et al., 2006). A few missing links in this pathway are the mechanistic details of CUC2 function in reorienting PIN1 and the molecular details of auxin mediated serration outgrowth. Forward genetic screens have been valuable in characterizing a genetic pathway even in the post genomic era. An EMS mutagenesis screen was performed in this context to identify novel factors that can improve our understanding of this intricate mechanism. tooth was identified in the M2 population based on its increased leaf serration phenotype. Genetic analysis showed that tth phenotype is due to a monogenic recessive mutation. Along with increased leaf serration, tth also shows various developmental defects such as aberrant phyllotaxy, narrower cotyledons and narrower leaves. Positional cloning and sequencing analysis showed a G to A transition at the AT2G39175 locus which codes for MIR160A. The mutation is at the 7th base position of the mature miRNA sequence. Functional characterization of miRNAs by isolating mutations is hampered by their small genomic sizes. Till now, only a few miRNAs have been characterized by mutational analysis in plants (Allen et al., 2007, Baker et al., 2005, Cartolano et al., 2007, Chuck et al., 2007, Knauer et al., 2013, Nag et al., 2009, Nikovics et al., 2006). miR160-ARF10 regulatory module is shown to be required for leaf blade out growth and serration, but not leaf complexity in tomato (Hendelman et al., 2012). miR160 is coded by 3 loci in Arabidopsis, MIR160A, B and C. All three loci encode identical mature miRNA that targets 3 Auxin response factors, ARF10, 16 and 17. ARFs are the effector molecules of auxin mediated developmental programmes. Genetic analysis showed that enhanced serration outgrowth in tth is due to the up-regulation of its target genes. Here, we have identified a miRNA that negatively regulates serration outgrowth by repressing ARF10, 16 and 17 whose functional significance in regulating leaf margin development was not known previously. Extensive genetic interaction studies have shown that TTH acts in parallel to SAW-BP and MIR164-CUC pathways in regulating leaf margin development. We have also shown that CUC2 and PIN1 are absolutely essential for serration development in tth. CUC2 establishes a pattern required for the expression of ARF10 at the leaf margin. In the absence of CUC2, downstream effector molecules such as ARFs can not perform their function. arf10-2 arf16-2 could reduce, but not suppress serration outgrowth in various mutants suggesting their functional redundancy with other ARF family members. CUC2 establishes auxin maxima at the leaf margin that triggers the degradation of AUX/IAA repressors thereby relieving ARF proteins which mediate serration outgrowth. Whereas, TTH acts at the post transcriptional level for maintaining normal ARF transcript levels Role of SPYINDLY in Arabidopsis leaf margin development SPYNDLY encodes an O-linked N-acetyl glucosamine transferase that acts as a negative regulator of GA response. Consistent with its role in GA response, spy mutants show several GA dependent phenotypes such as early flowering and hyper branched trichomes. spy mutants also show several GA independent phenotypes such as aberrant phyllotaxy and smooth leaf margin. We have studied its role in regulating Arabidopsis leaf serration development. Reporter analysis of ARF10::GUS and CUC2::GUS in spy-3 revealed that SPY is not involved in establishing serration pattern. The spy-3 leaves did not show any defects during the early stages of serration development, but the mature leaves display smooth leaf margin indicating that SPY function is required for serration outgrowth. As shown in the present study, TTH regulated ARFs are also involved in serration outgrowth. Analysis of leaf margin phenotype in tth spy-3 showed that SPY activity is not required for ARF mediated serration outgrowth. Similar genetic interaction studies with SAW-BP pathway mutants showed that leaf margin out growth mediated by meristematic genes is not dependent on SPY function. Genetic interaction studies with MIR164-CUC pathway genes showed that SPY is required for serration outgrowth in these mutants. Interestingly, the cuc2-3 mutant is defective at both patterning and outgrowth of serration. The spy-3 could suppress serration out growth in cuc2-D suggesting that CUC2 mediated serration out growth is dependent on SPY activity. Protein-protein interaction studies between SPY and CUC2 are in progress to demonstrate whether SPY directly interacts with CUC2 or CUC2 derived signal to regulate serration out outgrowth. It is interesting to examine how mutations at SPY locus can abolish serration out growth mediated by CUC2, but does not affect the serration pattern, even though CUC2 is reported to be essential for both the patterning and outgrowth of serration.

As such, the author will be able to draw appropriate and informed recommendations on the prospects of the doctrine in African context. In other words, the study proceeds from the approach that 'diversity' alone is not enough to guarantee application of margin of appreciation. Rather, a variety of factors come into consideration while weighing whether margin of appreciation should be granted to states. Comparative risk assessment of carcinogens in alcoholic beverages using the margin of exposure approach

Alcoholic beverages have been classified as carcinogenic to humans. As alcoholic beverages are multicomponent mixtures containing several carcinogenic compounds, a quantitative approach is necessary to compare the risks. Fifteen known and suspected human carcinogens (acetaldehyde, acrylamide, aflatoxins, arsenic, benzene, cadmium, ethanol, ethyl carbamate, formaldehyde, furan, lead, 4-methylimidazole, N-nitrosodimethylamine, ochratoxin A and safrole) occurring in alcoholic beverages were identified based on monograph reviews by the International Agency for Research on Cancer. The margin of exposure (MOE) approach was used for comparative risk assessment. MOE compares a toxicological threshold with the exposure. MOEs above 10,000 are judged as low priority for risk management action. MOEs were calculated for different drinking scenarios (low risk and heavy drinking) and different levels of contamination for four beverage groups (beer, wine, spirits and unrecorded alcohol). The lowest MOEs were found for ethanol (3.1 for low risk and 0.8 for heavy drinking). Inorganic lead and arsenic have average MOEs between 10 and 300, followed by acetaldehyde, cadmium and ethyl carbamate between 1,000 and 10,000. All other compounds had average MOEs above 10,000 independent of beverage type. Ethanol was identified as the most important carcinogen in alcoholic beverages, with clear dose response. Some other compounds (lead, arsenic, ethyl carbamate, acetaldehyde) may pose risks below thresholds normally tolerated for food contaminants, but from a cost-effectiveness point of view, the focus should be on reducing alcohol consumption in general rather than on mitigative measures for some contaminants that contribute only to a limited extent (if at all) to the total health risk.

An extensive real-time range simulation capability is being developed to ensure successful communications with the autonomous X-33 vehicle. This paper provides an overview of the various levels of simulation, integration, and test being developed to support the X-33 extended range subsystems. These subsystems include the flight termination system, L-band command uplink subsystem, and S-band telemetry downlink subsystem.

Facies variability in deep water channel-to-lobe transition zone : Jurassic Los Molles Formation, Neuquen Basin, Argentina

This study focuses on the facies changes from the lower slope to toe-of-slope to basin floor over a 10 km outcrop belt, in down-dip and oblique-strike directions to the basin margin. The Jurassic Los Molles Formation in Neuquen Basin, Argentina represents the slope and basin floor of basin margin clinoforms, coeval with the shallow water and fluvial deposits named Las Lajas and Challaco formations respectively. The shallow and deep water deposits are diachronously linked in an Early-Mid Jurassic source-to-sink system developed in a back-arc basin during the incipient development of the Andes Mountains. Satellite images, high resolution panorama pictures and measured sections were used to correlate and interpret the spatial variability and overall geometry of the base of slope to basin floor units. The observations of this study refine the model for the channel-to-lobe transition zone with increase recognition and quantification of facies and architecture variability. The Los Molles basin margin was coarse grained and was ideal to observe changes in the geometry and depositional facies of channel-to-lobe deposits from updip to downdip continuous over an 8 km outcrop belt. The described channel-to-lobe transition zone clearly shows a downdip change in bed boundaries from dominantly erosive to non-erosional (bypass) to depositional and with a range of distinct facies changes. In the transition zone the sand to shale ratio is high (N:G: 65-70 %), with gutter casts and deep scours, with a high degree of amalgamation, gravel lags, mud rip-up clasts and laterally migrating beds. Within the same depositional unit (deep water lobe), at the base of the slope, the dominant sandstone beds change from amalgamated structureless and normal graded sandstone beds in the channelized lobe axis to parallel laminated and normally graded in the channelized lobe off-axis areas. Similar facies changes have been observed along proximal to distal direction. The lateral change of the dominant structures in the beds indicates changes in the flow regime and depositional style. / text

隨著台灣股票市場的自由化與國際化,信用交易與外資在台股的交易比重不斷的增加。本文旨在探討股價指數、信用交易指標之融資餘額與外資之關連性,利用Johansen共整合向量分析與誤差修正模型,以日資料進行實證分析研究,樣本期間為1998年1月2日起自2003年6月30日止,共1419個樣本觀測值。本研究結果如下:一、第t期的融資餘額與第t-2期的股價指數共整合程度最高,顯示股價指數領先融資餘額兩期,為其領先指標。二、同期的股價指數與外資持股共整合程度最高,且股價指數會受到前一期外資持股變量的影響,顯示外資在市場上的動作的確會造成其他投資人的跟進。三、同期的融資餘額與外資持股共整合程度最高,顯示兩者呈現同方向的變動,且外資持股會受到融資餘額前一期變量的影響,且方向相反。 / This paper examines the relationship between stock index, balance of margin trading, and stock holding of foreign investors. Using daily data from January 2 1998 through June 30 2003 we investigate the interactions among the three variables through Johansen cointegration analysis and error correction model. It is found that (1) balance of margin trading of time t and stock index of time t-2 have the highest level of cointegration, which means stock index leads the balance of margin trading for two days. (2) stock index and stock holding of foreign investors of the same time t have the highest level of cointegration, and stock index of time t is affected by the stock holding of foreign investors of time t-1. (3) balance of margin trading and stock holding of the same time t have the highest level of cointegration, and the stock holding of foreign investors at time t is adversely influenced by balance of margin trading of time t-1.

Essays on the Determinants and Measurement of Subjective Well-Being

This thesis consists of four self-contained essays in economics, all concerned with different aspects of subjective well-being. The abstracts of the four studies are as follows. Beyond Income: The Importance for Life Satisfaction of Having Access to a Cash Margin. We study how life satisfaction among adult Swedes is influenced by having access to a cash margin, i.e. a moderate amount of money that could be acquired on short notice either through own savings, by loan from family or friends, or by other means. We find that cash margin is a strong and robust predictor of life satisfaction, also when controlling for individual fixed effects and socio-economic conditions, including income. Decomposing Variation in Daily Feelings: The Role of Time Use and Individual Characteristics. I explore the potential of using time-use data for understanding variation in affective well-being. Using the Princeton Affect and Time Survey, I decompose variation in daily affect into explained and unexplained within- and between person variation. Time use is found to mostly account for within-variation. Hence, its explanatory power is largely additive to that of individual characteristics. The explanatory power of time use is small, however. Activities only account for 1–7% of the total variation and this is not increased much by adding contextual variables. The Association Between Life Satisfaction and Affective Well-Being. We estimate the correlation between life satisfaction and affect — two conceptually distinct dimensions of subjective well-being. We propose a simple model that distinguishes between a stable and a transitory component of affect, and which also accounts for measurement error in self-reports of both variables, including current-mood bias effects on life satisfaction judgments. The model is estimated using momentarily measured well-being data, from an experience sampling survey that we conducted on a population sample of Swedes aged 18–50 (n=252). Our main estimates of the correlation between life satisfaction and long-run affective well-being range between 0.78 and 0.91, indicating a stronger convergence between these variables than many previous studies that do not account for measurement issues. Do OLS and Ordinal Happiness Regressions Yield Different Results? A Quantitative Assessment. Self-reported subjective well-being scores are often viewed as ordinal variables, but the conventional wisdom has it that OLS and ordered regression models (e.g. ordered probit) produce similar results when applied to such data. This claim has rarely been assessed formally, however, in particular with respect to quantifying the differences. I shed light on this issue by comparing the results from OLS and different ordered regression models, in terms of both statistical and economic significance, and across data sets with different response scales for measuring life satisfaction. The results are mixed. The differences between OLS, probit and logit estimates are typically small when the response scale has few categories, but larger, though not huge, when an 11-point scale is used. Moreover, when the error term is assumed to follow a skewed distribution, larger discrepancies are found throughout. I find a similar pattern in simulations, in which I assess how different methods perform with respect to the true parameters of interest, rather than to each other.

X-ray Coherent Scatter Imaging for Intra-operative Margin Detection in Breast Conserving Surgeries

<p>One of the challenges facing clinical practice today is intra-operative margin detection in breast conserving surgeries (BCS) or lumpectomy procedures. When a surgeon removes a breast tumor from a patient during a BCS procedure, the surgically excised tissue specimen is examined to see whether it contains a margin of healthy tissue around the tumor. A healthy margin of tissue around the tumor would indicate that the tumor in its entirety has been removed. On the other hand, if cancerous tissue is at the surface of the specimen, that would indicate that the tumor may have been transected during the procedure, leaving some residual cancerous tissue inside the patient. The most effective intra-operative real-time margin detection techniques currently used in clinical practice are frozen section analysis (FSA) and touch-prep cytology. These methods have been shown to possess inconsistent accuracy, which result in 20% to 30% of BCS patients being called back for a repeat BCS procedure to remove the residual tumor tissue. In addition these techniques have been shown to be time-consuming--requiring the operating room team to have to wait at least 20 minutes for the results. Therefore, there is a need for accurate and faster technology for intra-operative margin detection. </p><p>In this dissertation, we describe an x-ray coherent scatter imaging technique for intra-operative margin detection with greater accuracy and speed than currently available techniques. The method is based on cross-sectional imaging of the differential coherent scatter cross section in the sample. We first develop and validate a Monte Carlo simulation of coherent scattering. Then we use that simulation to design and test coherent scatter computed tomography (CSCT) and coded aperture coherent scatter spectral imaging (CACSSI) for cancerous voxel detection and for intra-operative margin detection using (virtual) clinical trials. Finally, we experimentally implement a CACSSI system and determine its accuracy in cancer detection using tissue histology. </p><p>We find that CSCT and CACSSI are able to accurately detect cancerous voxels inside of breast tissue specimens and accurately perform intra-operative margin detection. Specifically, for the task of individual cancerous voxel detection, we show that CSCT and CACSSI have AUC values of 0.97 and 0.94, respectively. Whereas for the task of intra-operative margin detection, the results of our virtual clinical trials show that CSCT and CACSSI have AUC values of 0.975 and 0.741, respectively. The gap in spatial resolution between CSCT and CACSSI affects the results of intra-operative margin detection much more than it does the task of individual cancerous voxel detection. Finally, we also show that CSCT would require on the order of 30 minutes to create a 3D image of a breast cancer specimen, whereas CACSSI would require on the order of 3 minutes. </p><p>These results of this work show that coherent scatter imaging has the potential to provide more accurate intra-operative margin detection than currently used clinical techniques. In addition, the speed (and therefore low scan duration: 3 min) of CACSSI, along with its ability to automatically classify cancerous tissue for margin detection means that coherent scatter imaging would be much more cost-effective than the clinical techniques that require up to 20 minutes and a trained pathologist. With the cancerous voxel detection accuracy of a 0.94 AUC and scan time of on the order of 3 minutes demonstrated for coherent scatter imaging in this work, coherent scatter imaging has the potential to reduce healthcare costs for BCS procedures and rates of repeat BCS surgeries. The accuracy for CACSSI can be considerably improved to match CSCT accuracy by improving its spatial resolution through a number of techniques: incorporating into the CACSSI reconstruction algorithm the ability to differentiate noise from high frequency signal so that we can image with higher frequency coded aperture masks; implementing a 2D coded aperture mask with a 2D detector; or acquiring additional angles of projection data.</p> / Dissertation

A study of coverage optimized planning incorporating models of geometric uncertainties for prostate cancer

A fundamental challenge in the treatment planning process of multi-fractional external-beam radiation therapy (EBRT) is the tradeoff between tumor control and normal tissue sparing in the presence of geometric uncertainties (GUs). To accommodate GUs, the conventional way is to use an empirical planning treatment volume (PTV) margin on the treatment target. However, it is difficult to determine a near-optimal PTV margin to ensure specified target coverage with as much normal tissue protection as achievable. Coverage optimized planning (COP) avoids this problem by optimizing dose in possible virtual treatment courses with GU models directly incorporated. A near-optimal dosimetric margin generated by COP was reported to savvily accommodate setup errors of target and normal tissues for prostate cancer treatment. This work further develops COP to account for (1) deformable organ motion and (2) delineation uncertainties for high-risk prostate cancer patients. The clinical value of COP is investigated by comparing with two margin-based planning techniques: (i) optimized margin (OM) technique that iteratively modifies PTV margins according to the evaluated target coverage probability and (ii) fixed margin (FM) technique that uses empirically selected constant PTV margins. Without patient-specific coverage probability estimation, FM plans are always less immune to the degraded effect of the modeled GUs than the COP plans or the OM plans. Empirical PTV margins face more risks of undesirable target coverage probability and/or excessive dose to surrounding OAR. The value of COP relative to OM varies with different GUs. As implemented for deformable organ motions, COP has limited clinical benefit. Due to optimization tradeoffs, COP often results in target coverage probability below the prescribed value while OM achieves better target coverage with comparable normal tissue dose. For delineation uncertainties, the clinical value of COP is potentially significant. Compared to OM, COP successfully maintains acceptable target coverage probability by exploiting the slack of normal tissue dose in low dose regions and maximally limiting high dose to normal tissue within tolerance.
