Studies of UHT-plant fouling by fresh, recombined and reconstituted whole milk : a thesis presented in partial fulfilment of the requirements for the degree of Doctor of Philosophy in Food Engineering

The objective of this study was to investigate the effects of preheat treatments on fouling by fresh whole milk (FWM), recombined whole milk (RCB) and reconstituted whole milk (Recon) in the high-temperature heater of indirect UHT plants. Various preheat treatments prior to evaporation during milk powder manufacture were applied to skim milk powder (SMP, 75 °C 2 s, 85 °C, 155 s and 95 °C, 155 s) and whole milk powder (WMP, 95 °C, 33 s). These preheat treatments were so-called “evaporator preheat treatments”. Skim milk powder (SMP) and whole milk powder (WMP) were derived from the same original batch of pasteurised FWM to remove the effects of the variation in milk composition between different milk batches. These SMPs were recombined with anhydrous milk fat and water to prepare RCB, and WMPs were reconstituted with water to prepare Recon. Then, (homogenized) FWM, RCB and Recon were subjected to various preheat treatments (75 °C, 11 s, 85 °C, 147 s and 95 °C, 147 s) prior to UHT processing. These preheat treatments were so-called “UHT preheat treatments”. Temperature difference (hot water inlet temperature – milk outlet temperature) was taken as a measure of the extent of fouling in the high-temperature heater. The slope of the linear regression of temperature difference versus time (for two hours of UHT processing) was taken as fouling rate (°C/h). Increasing both evaporator and UHT preheat treatments resulted in increasing fouling rate and total deposit weight for all three whole milk types for several milk batches. In the case of FWM, there was no reduction in fouling rate with increasing UHT preheat treatment whether FWM was homogenized then preheated, preheated then homogenized or not homogenized at all. These findings, which are wholly consistent and well replicated, are in apparent conflict with the results of most previous comparable studies. Possible reasons for this are explained. Further investigations of the effects of homogenization relating to the role of whey protein on the surface of the fat globules showed that whey protein associated with the membrane covering the surface of fat globules for homogenized then preheated FWM, RCB and Recon and that association increased with increasing heating process stage. The increasing association of whey protein with the milk fat globules membrane with increasing severity of heating process stage became faster when preheat treatment was more severe: the association of whey protein plateaued on intermediate temperature heating when the milks were preheated at 75°C, 11 s and on preheating when the milks were preheated at 95°C, 147 s. In the case of FWM, the thickness of the membrane covering the surface of fat globules for homogenized then preheated FWM, which increased with the severity of heating process stage, was greater than the thickness of the membrane in preheated then homogenized FWM. Preheating then homogenization resulted in the greater interfacial spreading of small molecules on the surface of fat globules, i.e. whey protein or small molecules from the disintegration of casein micelles during preheating. Possible basic mechanisms for UHT fouling in the high-temperature heater include: the reduction in the solubility of calcium phosphate and the deposition of protein as fat-bound protein and non-fat-bound protein. When non-fat-bound protein in milk plasma deposited, it could be a carrier for the deposition of mineral, such as, the precipitate of calcium phosphate in the casein micelles or the deposition of complexes between whey protein and casein micelles.

The study was carried out at the Laboratory of Animal Breeding Value Establishment and Selection of Lithuanian Veterinary Academy in the time of postgraduate studies from 2003 to 2005. During the last seven years Lithuanian red and red-and-white controlled dairy cattle breeds average yield increased 41.7%, milk fat production - 50.9%, milk proteins production – 46.1%. Between milk fat and milk proteins was found medium positive correlation coefficient (rp =0.36-0.56; p<0.01), which show milk fat increasing has positive effect to milk proteins increasing. Correlation coefficient between milk production and milk fat, milk proteins is very high (rp =0.87-0.96; p<0.01). The analysis of Variance showed the breed influence on reproductive traits of cow to be statistically reliable (p<0.001). The breed had the greatest influence on cows age during calving. The influence of bulls line on many traits of cows exterior is statistically reliable (p<0,001).

Tiriamasis darbas atliktas Lietuvos sveikatos ir mokslų universitete ir Lietuvos pieninių galvijų fermose 2011 – 2014 metais. Darbą sudaro 49 puslapių, 12 lentelių, 8 paveikslai. Baigiamojo darbo tikslas: įvertinti somatinių ląstelių skaičių karvių piene, išnalizuoti jų kitimo priežastis, nustatyti jų ryšį su produktyvumu. Baigiamojo darbo uždaviniai: Įvertinti somatinių ląstelių skaičių karvių piene, karvių pieno kiekio ir sudėties (riebumo, baltymingumo, laktozės) rodiklius, negenetinių veiksnių įtaką karvių produktyvumui ir SLS piene, genetinių veiksnių įtaką karvių produktyvumui ir SLS piene. Ištirtos 9 veislių karvės, trijuose skirtinguose ūkiuose. Iš viso buvo ištirtos 726 karvės Tyrimo metu buvo nagrinėta genetinių ir negenetinių veiksnių įtaka karvių pieno somatinių ląstelių skaičiui (tūkst./cm3), pieningumui (kg), riebumui (proc.), baltymingumui (proc.) bei laktozės kiekiui (proc.). Įvertinus tirtų karvių SLS piene nustatyta, kad vidutinis SLS kiekis piene buvo 220,65±10,477 tūkst./cm3. 43,09 proc. karvių piene SLS buvo mažesnis nei 100 tūkst./cm3, 68,92 proc. – svyravo nuo 100 iki 200 tūkst./cm3, 31,08 proc. - viršijo 200 tūkst./cm3, rodo, kad trečdalis tirtų karvių sirgo tešmens uždegimu. Įvertinus tirtų karvių pieno kiekio ir sudėties rodiklius nustatyta, kad vidutinis tirtų karvių pieningumas 19,78±0,192 kg, pieno riebumas buvo 4,61±0,022 proc., baltymingumas – 3,62±0,011 proc., laktozės kiekis piene - 4,50±0,005 proc. Goal of work: to evaluate somatic cells count in cows’ milk, to analyse their causes of changes and to determine their relationship with productivity. Goal of thesis: to evaluate the somatic cells count in cows’ milk, to determine milk yield and composition (fat, protein, lactose). The analysis is based on 726 dairy cows from different breeds (9 breeds) in three farms. During the study some data have been analysed influence of the genetic and non–genetic factors such as somatic cell count (SCC) in milk (thousand./cm3), milk yield (kg), milk fat (%) and protein (%) milk lactose content (%) and cow origin data. The estimation of somatic cells count in cows’ milk shows that average of SCC was 220.65±10.477 thousand/cm3. The 43.09 percent of cows’ SCC in milk was less than 100 thousand/cm3, 68.92 percent – ranged from 100 to 200 thousand/cm3 and 31.08 percent had more than 200 thousand/cm3. The evaluation of cows’ milk yield and composition traits showed that the average of cows’ milk yield was 19.78 ±0.192 kg, milk fat – 4.61 ±0.022 percent, the amount of protein – 3.62 ±0.011 percent, lactose content of milk – 4.50 ±0.005 percent. Phenotypic relationships between milk protein percentage, reproductive performance and body condition score in Irish dairy cattle : a thesis presented in partial fulfilment of the requirements for the degree of Master of Sciences (MSc) at Massey University, Palmerston North, New Zealand

A positive phenotypic correlation between milk protein percentage and reproductive performance in dairy cattle, especially during early lactation has been recently reported. The objective of this study was to quantify the relationship between milk protein percentage and different measures of fertility in Irish, seasonal calving, dairy cattle using data from experiments comparing strains of Holstein-Friesian cows under different feeding systems. The relationships between body condition score, milk production and fertility were also investigated. The data used in this study consisted of 584 lactation records over a 5-yr period. Principal component analysis and logistic regression was used to study the relationship between milk protein percentage and fertility performance of the cow. Greater milk protein percentage during the first 60 days post-calving was associated with better reproductive performance. The probability of a cow being submitted in the first 21 days of the breeding season increased with increased milk protein percentage during early lactation. Similarly, the probability of a cow becoming pregnant to its first service or to the whole breeding season also increased. Cows were classified as either high or low milk protein percentage based on their protein percentage over the whole lactation. Cows in the high milk protein group had a 7% greater conception rate compared to cows in the low protein percentage group. In conclusion, cows with higher protein percentage, especially during early lactation are submitted earlier in the breeding season, and have a higher conception rate. Physiologically, the shortage of glucose caused by negative energy balance restricts the synthesis of milk protein in the udder. On the other side, negative energy balance also causes the reduction of IGF-I, LH and oestradiol, which consequently delay the ovarian follicular development and finally reduces fertility. Therefore, there is a biological explanation for the association between milk protein percentage and fertility performance.

Among them, casein has been used for this purpose. This study aims to determine peptide profile and amino acid sequence of bioactive peptides derived from Moxotó goat milk casein hydrolysis, using proteolytic enzymes such as trypsin, pepsin, papain and a protease extracted from Penicillium aurantiogriseum URM 4622. Enzymatic hydrolysis was performed using a statistical experimental design, which independent variables were pH, enzyme substrate (E: S), temperature and reaction time in Moxotó goat milk casein hydrolysis. Hydrolysis products were visualized in SDS-PAGE electrophoresis. Hydrolysates subjected to ultrafiltration (cut off 3000Da) were used to determine biological properties. Antioxidant activity was evaluated by ABTS + [2,2 '-Azin-bis (3-ethylbenzothiazoline) 6-sulfonic acid] method, using the pool of peptides (permeate <3000Da; retentate >3000Da). Antimicrobial activity was determined by the Clinical and Laboratory Standards Institute. Binding through the zinc solubility of zinc in the sample by Mass Spectrometry Inductively Coupled Plasma. Peptide profile and amino acid sequences were determined by mass spectrometry MALDI-TOF-MS/MS. The best hydrolysis degree (38.27%) was obtained with pepsin enzyme pH 3.0, 1:100 E: S, temperature of 40°C and 5 hours time reaction. However, a high hydrolysis degree precluded the use of these hydrolysates for bioactive peptides obtention. Casein tryptic hydrolysates demonstrated antioxidant activity up to 3242.3 μmol.L-1TROLOX/mg peptide in retentate (> 3000Da). By papain use, we obtained an activity up to 2329.6 μmol.L-1TROLOX /mg of peptide in permeate (<3000Da). The peptides produced by P. auratiogriseum protease action showed activity from 843.17 to 2587.30 μmol.L-1of Trolox / mg of 10 and 29% peptides hydrolysates, respectively, which were compatible with natural antioxidants, such as C vitamin and α-tocopherol. Goat casein tryptic hydrolysates demonstrated antimicrobial activity against Enterococcus faecalis ATCC 6057, Escherichia coli ATCC 2508, Klebisiela pneumoniae ATCC 29665, Bacillus subtilis ATCC 6633. Casein hydrolysates showed IC50 of 4.46 mg/g for zinc binding. Mass spectrometry MALDI TOF MS\MS allowed the visualization of caprine casein peptides in permeate (<3000Da), ranging from 568 to 2923 Da. It also showed LLYQEPVLGPV and HPINHQGLSPEVPNENLLR amino acids sequences for αs1 and β-casein, respectively, from casein tryptic hydrolysates; LLYQEPVLGPV sequences of β-casein, the NPWDQVK αs2 NENLL-casein and-casein in the αS1 casein hydrolysates by papain use; LLYQEPVLGPVRGPFPI β-casein sequence from casein hydrolysates obtained by the use of P. aurantiogriseum URM 4622 protease. The casein hydrolysates bioactive properties are referent to the prevalence of hydrophobic amino acids, which possibility of their use as bioactive peptides. / Proteínas lácteas apresentam grande potencial na produção de peptídeos bioativos. A caseína se destaca na produção de bioativos. Entretanto, a sua bioatividade só é conseguida após hidrólise enzimática, ao qual se produz substâncias benéficas à saúde quando incorporados em alimentos ou produtos farmacêuticos. O objetivo deste trabalho foi o de caracterizar e a avaliar atividades biológicas e determinando o perfil peptídico e a sequência de aminoácidos dos peptídeos bioativos obtidos a partir de hidrólise da caseína do leite de cabra Moxotó, usando enzimas proteolíticas tripsina, pepsina, papaína e protease extraída de Penicillium aurantiogriseum URM 4622. A hidrólise enzimática foi realizada através de planejamento experimental estatístico, os quais as variáveis independentes foram pH, relação enzima substrato (E:S), temperatura e tempo de reação na hidrólise da caseína do leite de cabra Moxotó. Os produtos de hidrólise foram visualizados em eletroforese SDS‑PAGE. Os hidrolisados submetidos à ultrafiltração (Cut off 3000Da) foram utilizados para determição das propriedades biológicas. A atividade antioxidante foi avaliada no pool de peptídeos, permeado (<3000Da) e retentado (>3000Da) usando-se o método ABTS+ [2,2’-azino-bis (3-etilbenzotiazolin) 6-ácido sulfônico]. A atividade antimicrobiana foi determinada pelo método do Clinical and Laboratory Standards Institute. O carreamento de zinco através da solubilidade do zinco na amostra através da Espectrometria de Massa em Plasma Indutivamente Acoplado. O perfil peptídico e as sequências de aminoácidos foram determinados por espectrometria de massa MALDI-TOF-MS/MS. O melhor grau de hidrólise (38,27%) foi obtido com a enzima pepsina usando pH 3,0, E:S de 1:100, 40ºC e 5 horas de reação. Entretanto, um alto grau de hidrólise impossibilitou o uso desses hidrolisados para obtenção de peptídeos bioativos. Os hidrolisados trípicos da caseína mostraram atividade antioxidante de até 3.242,3μmol.L-1TROLOX/mg de peptídeo no retentado (>3000Da). Com o uso da papaína, obteve-se uma atividade de até 2.329,6μmol.L-1TROLOX/mg de peptídeo no permeado (<3000Da). Os peptídeos produzidos pela ação da protease produzida por P. auratiogriseum apresentaram atividade de 843,17 a 2.587,30μmol.L-1de TROLOX/mg de peptídeos para os hidrolisados com 10 e 29% respectivamente, os quais foram compatíveis a antioxidantes naturais, como a vitamina C e α-tocoferol. Os hidrolisados trípticos da caseína caprina apresentaram atividade antimicrobiana frente a Enterococcus faecalis ATCC 6057, Escherichia coli ATCC 2508, Klebisiela pneumoniae ATCC 29665, Bacillus subtilis ATCC 6633. Os hidrolisados de caseína mostraram IC50 de 4,46mg/g para carreamento de zinco. A espectrometria de massa MALDI TOF MS permitiu visualizar os peptídeos no permeado (<3000Da) da caseína caprina, na faixa de 568 a 2.923 Da. Mostraram as sequências LLYQEPVLGPV e HPINHQGLSPEVPNENLLR e da β- e αs1-caseina para hidrolisados trípticos da caseína; as sequências LLYQEPVLGPV da β-caseína, NPWDQVK da αs2-caseina e NENLL da αS1-caseína nos hidrolisados da caseína com o uso da papaína e a sequência LLYQEPVLGPVRGPFPI da β-caseina no hidrolisado da caseína com o uso da protease produzida por P. aurantiogriseum URM 4622. As propriedades bioativas dos hidrolisados da caseína referem-se à prevalência de aminoácidos hidrofóbicos, o que possibilita o uso destes como peptídeos bioativos.

Food-related diseases, including food allergies and food intolerances, are on the rise worldwide. According to the World Health Organisation (WHO), allergic diseases in the economically advanced countries of the WHO have become the largest child environmental epidemic. Dysregulation of immune tolerance is the basic mechanism involved in the development of food allergy. It is believed that the risk of developing allergies is more related to other lifestyle factors such as a diet, physical activity or obesity. Food allergies in children and adult populations vary both in the spectrum of triggering foods and also their symptoms and their severity. This diploma thesis deals with the evaluation of clinical manifestations in infants with allergic colitis and their overall nutritional status, anthropometry, blood count and antibodies against cow's milk proteins. Allergic colitis is one of the most common manifestations of infant allergy to cow`s milk protein with gastrointestinal involvement. The theoretical part summarizes the anthropometric evaluation of growth and development, infant nutrition, the most important laboratory indicators of nutritional status. The part is generally devoted to food allergies, mainly allergy to cow's milk protein. The practical part deals with the evaluation of a sample of examined patients in gastroenterology outpatient clinics and nutritional outpatient clinic of the Department of Pediatrics and Adolescent Medicine of the 1st Faculty of Medicine, Charles University and General Hospital in Prague. The observed group of infants with allergic colitis includes infants exclusively breastfed, breastfed by a mother following a non-dairy diet, infants on an extensive hydrolyzate or on an amino acid formula. Pequenos e médios laticínios têm grandes dificuldades no planejamento da produção para maximizar o lucro. Além disso, a forma atual de precificar o leite cru no Brasil desestimula o produtor a melhorar sua qualidade, pois valoriza mais seu volume do que seus componentes sólidos. A composição do leite cru é muito importante para os laticínios, pois ela afeta diretamente o rendimento de produção e a qualidade dos produtos lácteos. Técnicas de otimização, como programação linear (PL), ajudam a resolver problemas relacionados à decisão do mix de produtos, além de fazer análise econômica dos recursos. Em razão disso, foi desenvolvido em PL um modelo para maximização da margem de contribuição total (MCT = receita total das vendas - custos e despesas diretas totais) e precificação do leite cru através da determinação do mix ótimo de produtos lácteos. O modelo foi simulado em cenários diários de alta (Janeiro) e baixa (Julho) disponibilidade de matéria prima comparando as soluções ótimas com resultados reais de um laticínio do sudeste do Brasil. Foram realizadas análises de sensibilidade dos componentes nutritivos de dois tipos de leite cru de diferentes composições (LC1, leite cru proveniente de um fornecedor, e LC50, leite cru proveniente de 50 fornecedores) para determinar seus preços-sombra. Foram observados preços-sombra para o quilograma da caseína em Janeiro e Julho e para o litro do LC50 em Julho. A MCT ótima e os preços calculados de ambos os tipos de leite cru foram maiores em Janeiro devido à maior disponibilidade de matéria prima. Em ambos os cenários, os preços calculados de ambos os tipos de leite cru foram maiores que os praticados pelo laticínio e o lucro antes de juros, impostos, depreciação e amortização (LAJIDA) diário real foi maior que o ótimo. A embaladora foi um recurso limitante atuante na produção do queijo Minas Frescal nos mixes ótimos. A relação entre margem de contribuição unitária (MCU = preço unitário de venda - custo e despesa diretos unitários) e quantidade necessária de matéria prima por unidade de produto e a disponibilidade de recursos são determinantes na definição do mix de produtos lácteos e da MCT do laticínio. A precificação do leite cru pelo modelo proposto remunera o produtor em função da quantidade de seus componentes. O laticínio pode maximizar seu LAJIDA planejando melhor seu mix de produtos com PL e remunerando melhor seus fornecedores em função da qualidade do leite cru. / Small and medium-sized dairies face great difficulties in production planning to maximize profit. In addition, the current way of pricing the raw milk in Brazil discourages producers to improve its quality because its volume values more than its solid components. The raw milk composition is very important for the dairies because it directly affects the yield and quality of dairy products. Optimization techniques, such as linear programming (LP), aid solve problems related to the decision of the product mix and perform economic analyzes of resources. For this reason, a model was developed in LP to maximizing total contribution margin (TCM = total sales revenues - total variable costs and expenses) and pricing the raw milk by determining the optimal mix of dairy products. The model was simulated in two daily scenarios of high (January) and low (July) availability of raw material comparing the optimal solutions with actual results from a dairy plant in Southeastern Brazil. Sensitivity analyzes of the nutritional components of two kinds of raw milk of different compositions (RM1 and RM50) were performed to determine their shadow-prices. Shadow-prices were observed for the kilogram of casein in January and July and for the liter of RM50 in July. The optimal TCM and the calculated prices of both types of raw milk were higher in January due the increased availability of raw materials. In both scenarios, the calculated prices of both types of raw milk were higher than those paid by the dairy plant and the actual daily earnings before interests, taxes, depreciation and amortization (EBITDA) were greater than the optimum ones. The wrapper was an active limiting resource in the production of Frescal Minas cheese in optimal mixes. The relationship between unit contribution margin (UCM = unit sales price - unit variable cost and expense) and required amount of raw material per unit of output and resource availability are crucial in defining the mix of dairy products and the dairy TCM. The proposed raw milk pricing model pays the producer by the quantity of its components. The dairies can maximize their EBITDA planning their product mixes with LP and remunerating their suppliers based on the quality of raw milk.
