Dissection génomique, transcriptomique et chimique des leucémies myéloïdes aiguës

Les leucémies myéloïdes aiguës (LMA) consistent en un groupe de cancers agressifs causés par une accumulation de mutations génétiques et épigénétiques survenant dans les cellules souches ou progénitrices de la moelle osseuse. Il s’agit d’un groupe de maladies très hétérogène, caractérisé par un grand nombre de combinaisons d’altérations qui perturbent à la fois les voies de signalisation qui y sont exprimées, leur sensibilité aux différents traitements et le pronostic des patients. Le déploiement des technologies de séquençage de nouvelle génération au courant de la dernière décennie a permis l’exploration à une échelle sans précédent du paysage mutationnel et transcriptomique de différents cancers, incluant les LMA. Dans le cadre de nos travaux, nous avons voulu tester l'hypothèse selon laquelle les LMA se déclinent en plusieurs sous-groupes génétiques caractérisés chacun par des mutations distinctes et une expression génique dérégulée, ainsi qu’une réponse différentielle à des molécules qui pourraient représenter de nouvelles stratégies thérapeutiques. Nous avons testé cette hypothèse au sein de la cohorte Leucegene, qui comprend un grand nombre de LMA primaires analysées par le séquençage du transcriptome, et nous avons analysé les différences entre les différents sous-groupes en les analysant un à la fois. Cette étude des différents sous-groupes nous a permis de disséquer le profil génomique, transcriptomique et les sensibilités aux petites molécules de sept sous-groupes génétiques, représentant environ la moitié des cas de LMA de l’adulte. Notre approche a permis de découvrir plusieurs nouvelles mutations spécifiques aux différents sous-groupes, dont certaines ont été validées dans des cohortes indépendantes. Nous avons également confirmé que les gènes différentiellement exprimés dans les sous-groupes sont plus informatifs que les signatures d'expression non supervisées pour identifier les biomarqueurs de la maladie. Nous avons ainsi identifié dans la majorité des sous-groupes des gènes représentant un biomarqueur d'intérêt, ayant une pertinence fonctionnelle ou pronostique. Ces données ont également mené à des criblages chimiques ciblés qui ont identifié de nouvelles vulnérabilités dépendant du contexte génétique. Au-delà de ces observations, nos travaux pourraient avoir une portée translationnelle tandis que le séquençage de nouvelle génération est de plus en plus utilisé en clinique. La combinaison avec d’autres modalités de séquençage et l’incorporation de technologies émergentes aideront à poursuivre la dissection génomique, transcriptomique et chimique de la LMA et l’approche utilisée pourra même éventuellement s’appliquer à d’autres types de cancers. / Acute myeloid leukemias (AML) are a group of cancers caused by an accumulation of genetic and epigenetic mutations occurring in the stem or progenitor cells of the bone marrow. They represent a very heterogeneous group of diseases, characterized by a large number of combinations of alterations which disrupt to varying degrees key networks in these cells, their sensitivity to treatments and the prognosis of the patients. The deployment of next-generation sequencing technologies over the past decade has enabled exploration on an unprecedented scale of the mutational and transcriptomic landscape of various cancers, including AML. As part of our work, we tested the hypothesis according to which AMLs comprise several genetic subgroups, each characterized by distinct mutations and deregulated gene expression profiles, as well as a differential response to molecules that could represent novel therapies. We tested this hypothesis in the Leucegene cohort, which includes a large number of primary AMLs analyzed by transcriptome sequencing, which we explored one subgroup after the other, dissecting the genomic, transcriptomic or small molecule sensitivities profile of seven AML subgroups representing approximately half of adult AML cases. Our approach has allowed us to discover several new mutations specific to different subgroups, some of which have been validated in independent cohorts. We also confirmed that genes differentially expressed in subgroups are more informative than unsupervised expression signatures, and we identified genes representing potential biomarkers, or having a functional or prognostic relevance in the majority of subgroups. Generated data also led to targeted chemical screens performed on primary AML cells, which identified new context-dependent vulnerabilities. Beyond these observations, our work could have a translational scope while next-generation sequencing is paving its way in the clinic. The combination with other Omics and the incorporation of emerging technologies will help to further the multi-dimensional dissection of these groups and additional ones, as the presented approach could be applied to additional disease subsets and cancer types.

Contributions à la sonification d’image et à la classification de sons

L’objectif de cette thèse est d’étudier d’une part le problème de sonification d’image et de le solutionner à travers de nouveaux modèles de correspondance entre domaines visuel et sonore. D’autre part d’étudier le problème de la classification de son et de le résoudre avec des méthodes ayant fait leurs preuves dans le domaine de la reconnaissance d’image. La sonification d’image est la traduction de données d’image (forme, couleur, texture, objet) en sons. Il est utilisé dans les domaines de l’assistance visuelle et de l’accessibilité des images pour les personnes malvoyantes. En raison de sa complexité, un système de sonification d’image qui traduit correctement les données d’image en son de manière intuitive n’est pas facile à concevoir. Notre première contribution est de proposer un nouveau système de sonification d’image de bas-niveau qui utilise une approche hiérarchique basée sur les caractéristiques visuelles. Il traduit, à l’aide de notes musicales, la plupart des propriétés d’une image (couleur, gradient, contour, texture, région) vers le domaine audio, de manière très prévisible et donc est facilement ensuite décodable par l’être humain. Notre deuxième contribution est une application Android de sonification de haut niveau qui est complémentaire à notre première contribution car elle implémente la traduction des objets et du contenu sémantique de l’image. Il propose également une base de données pour la sonification d’image. Finalement dans le domaine de l’audio, notre dernière contribution généralise le motif binaire local (LBP) à 1D et le combine avec des descripteurs audio pour faire de la classification de sons environnementaux. La méthode proposée surpasse les résultats des méthodes qui utilisent des algorithmes d’apprentissage automatique classiques et est plus rapide que toutes les méthodes de réseau neuronal convolutif. Il représente un meilleur choix lorsqu’il y a une rareté des données ou une puissance de calcul minimale. / The objective of this thesis is to study on the one hand the problem of image sonification and to solve it through new models of mapping between visual and sound domains. On the other hand, to study the problem of sound classification and to solve it with methods which have proven track record in the field of image recognition. Image sonification is the translation of image data (shape, color, texture, objects) into sounds. It is used in vision assistance and image accessibility domains for visual impaired people. Due to its complexity, an image sonification system that properly conveys the image data to sound in an intuitive way is not easy to design. Our first contribution is to propose a new low-level image sonification system which uses an hierarchical visual feature-based approach to translate, usingmusical notes, most of the properties of an image (color, gradient, edge, texture, region) to the audio domain, in a very predictable way in which is then easily decodable by the human being. Our second contribution is a high-level sonification Android application which is complementary to our first contribution because it implements the translation to the audio domain of the objects and the semantic content of an image. It also proposes a dataset for an image sonification. Finally, in the audio domain, our third contribution generalizes the Local Binary Pattern (LBP) to 1D and combines it with audio features for an environmental sound classification task. The proposed method outperforms the results of methods that uses handcrafted features with classical machine learning algorithms and is faster than any convolutional neural network methods. It represents a better choice when there is data scarcity or minimal computing power.

Influence of the Active Screen Plasma Power during Afterglow Nitrocarburizing on the Surface Modification of AISI 316L

Active screen plasma nitrocarburizing (ASPNC) increases the surface hardness and lifetime of austenitic stainless steel without deteriorating its corrosion resistance. Using an active screen made of carbon opens up new technological possibilities that have not been exploited to date. In this study, the effect of screen power variation without bias application on resulting concentrations of process gas species and surface modification of AISI 316L steel was studied. The concentrations of gas species (e.g., HCN, NH3, CH4, C2H2) were measured as functions of the active screen power and the feed gas composition at constant temperature using in situ infrared laser absorption spectroscopy. At constant precursor gas composition, the decrease in active screen power led to a decrease in both the concentrations of the detected molecules and the diffusion depths of nitrogen and carbon. Depending on the gas mixture, a threshold of the active screen power was found above which no changes in the expanded austenite layer thickness were measured. The use of a heating independent of the screen power offers an additional parameter for optimizing the ASPNC process in addition to changes in the feed gas composition and the bias power. In this way, an advanced process control can be established.

This thesis explores how a displayed image and architecture interact together. This manifesto is analogically explored using parts of the human body such as the eyeballs as a structural analysis for the buildings with a projection of the cornea, the pupil, the retina, the optic nerves and the brain. This analysis follows a sequential order of capturing light, transcribing light into image, and displaying such image onto a screen. Both, the image and architecture are created parallel to each other respectively when conceiving an architectural idea in order to develop the idea into a building and then perceiving the architecture from such building. These steps are a cinematic approach using a video camera to record an experience of movement through the journey of a metro ride. This video is one of the tools used to edit an urban tissue of downtown Washington, D.C. The project becomes a Center for the Art of Moving Images exposing vectors of movements through its architecture. The building is manifested in a three-dimensional design where the site provides a sunken plaza 60 ft below street level perceived as a new floor in the city. The underground metro station transitions to the street surface through the use of this plaza in a very harmonious way. The result is a visual depth parallel to the perspectives perceived in movies where you see beyond the surface of the screen and in this case, beyond the surface of the city. / Master of Architecture

В ходе работы проведено формирование электрокаталитически активного слоя на поверхности рабочего электрода. Таким образом, каталитически активный слой с наибольшей чувствительностью сформирован на печатном электроде 3-в-1 с использованием многостенных углеродных нанотрубок (cMWCNT), электроосаждённым золотом и медь(II)-имидазольного каркаса, состоящего из иона меди(II) и 2-меркаптоимидазола и 2-метилимидазола. Проведены исследования селективности полученного каталитически активного слоя. / This work consists of 3 chapters and is devoted to the enzyme-free quantitative determination of creatinine, glucose, urea using copper(II)-imidazole frameworks. In the course of the work, the formation of an electrocatalytically active layer on the surface of the working electrode was carried out. Thus, the catalytically active layer with the highest sensitivity is formed on a 3-in-1 printed electrode using multi-walled carbon nanotubes (cMWCNT) electrodeposited with gold and copper(II)-imidazole framework consisting of copper(II) ion and 2-mercaptoimidazole and 2-methylimidazole. The selectivity of the obtained catalytically active layer has been studied.

Lösungs- und Ausscheidungsprozesse in silberhaltigen Glasschmelzen bei der thermischen Kontaktierung von multikristallinem Silicium

Silber-Glas-Dispersionen werden in der Herstellung von Solarzellen zur Ausbildung des Vorderseitenkontaktes großtechnisch seit vielen Jahren eingesetzt. Nichtsdestotrotz ist der Kontaktbildungsmechanismus bis jetzt nicht vollständig verstanden. In dieser Arbeit wurden Lösungs-, Transport- und Ausscheidungsprozesse von silberhaltigen Glasschmelzen in Silber-Glas-Dispersionen während der thermischen Kontaktierung von multikristallinem Silicium untersucht. Hierfür wurden systematisch Änderungen in der Zusammensetzung der Silber-Glas-Dispersionen sowie der hierfür verwendeten Gläser durch geführt, um Einzeleffekte zu separieren. Die Gläser wurden mit Silber zu Modelldispersionen verarbeitet und auf Siliciumwafer zu Herstellung von Solarzellen mittels Siebdruck abgeschieden und eingebrannt. Sowohl die Einzelkomponenten als auch die hergestellten Dispersionen wurden hinsichtlich ihrer thermisch aktivierten, linearen Schwindung untersucht. Die hergestellten Solarzellen wurden elektrisch sowie mittels FESEM in ihrer Mikrostruktur charakterisiert. Es konnten Zusammenhänge zwischen diesen Untersuchungen hinsichtlich der Silberlösung im Glas unter Oxidation, dem Silbertransport im Glas mittels Diffusion oder Konvektion sowie der Silberausscheidung unter Reduktion an der Waferoberfläche hergestellt und Einzeleffekte für den Silbertransport während des Einbrandes von Solarzellen identifiziert werden.

<b>Investigation of Additively Manufactured Silver Plated Stainless Steel Monolith Catalyst Beds</b>

<p dir="ltr">Additive manufacturing has introduced new possibilities for the design and manufacturing of monolith catalyst beds. Many hydrogen peroxide monolith catalyst beds are made of ceramics and washcoated through a complex process. However, creating a metal monolith bed with the tried-and-true silver catalyst could provide an alternative decomposition method for 90 wt.% hydrogen peroxide with easier manufacturing methods and similar or better decomposition efficiency. 91.2 wt.% hydrogen peroxide was decomposed with a lattice-type monolithic catalyst bed additively manufactured out of 316L stainless steel that was electroplated with pure silver. The variables investigated included the catalyst bed’s mass loading, chamber pressure, pressure drop, and length-to-diameter ratio (L/D). The catalyst bed had loadings of 0.1 lb<sub>m</sub>/s/inch<sup>2</sup>, 0.25 lb<sub>m</sub>/s/inch<sup>2</sup>, and 0.4 lb<sub>m</sub>/s/inch<sup>2</sup>. One catalyst bed configuration had an L/D of 2.6, while the other configuration had an L/D of 0.85. A modular throat controlled the chamber pressures for each catalyst bed loading case. The decomposition efficiency was calculated with the theoretical and expected characteristic velocity (c*) of the catalyst beds. The chamber pressures for the lowest bed loading and highest L/D ratio varied from 52 psia to 202 psia. The hydrogen peroxide decomposition efficiency was approximately 85% for the lowest chamber pressure and approximately 100% for the highest chamber pressure. The chamber pressures for the middle and highest bed loading and high L/D were 193 psia at the lowest to 325 psia at the highest. The decomposition efficiencies for all middle and highest bed loading tests with high L/D were 90% or higher for all tests. For all of the highest L/D tests, decomposition was also confirmed by observing videos of the exhaust plume, which was clear and showed no sign of flow channeling. For all of the highest L/D tests, the pressure drops in all of the middle bed loading cases were at or below 30% of the chamber pressure. The high chamber pressure, highest bed loading cases also had a pressure loss below 30% of the chamber pressure. The smallest L/D configuration performed significantly worse than expected, with efficiencies between 15-25% at chamber pressures between 33-75 psi. The silver electroplated on the stainless steel survived the 143 s of lifetime on the catalyst bed during testing with minimal to no silver loss determined by weight and visual inspection with a microscope post-test. The higher L/D catalyst bed tests prove that silver electroplated on to an additively manufactured stainless steel monolith is a viable method for creating a catalyst bed. More research is required to determine the lowest L/D possible, which resides somewhere between the two L/D cases studied, and higher bed loadings also require investigation.</p>

Beyond the electronic connection : the technologically manufactured cyber-human and its physical human counterpart in performance : a theory related to convergence identities

This thesis is an investigation of the complex processes and relationships between the physical human performer and the technologically manufactured cyber-human counterpart. I acted as both researcher and the physical human performer, deeply engaged in the moment-to-moment creation of events unfolding within a shared virtual reality environment. As the primary instigator and activator of the cyber-human partner, I maintained a balance between the live and technological performance elements, prioritizing the production of content and meaning. By way of using practice as research, this thesis argues that in considering interactions between cyber-human and human performers, it is crucial to move beyond discussions of technology when considering interactions between cyber-humans and human performers to an analysis of emotional content, the powers of poetic imagery, the trust that is developed through sensory perception and the evocation of complex relationships. A theoretical model is constructed to describe the relationship between a cyber-human and a human performer in the five works created specifically for this thesis, which is not substantially different from that between human performers. Technological exploration allows for the observation and analysis of various relationships, furthering an expanded understanding of ‘movement as content’ beyond the electronic connection. Each of the works created for this research used new and innovative technologies, including virtual reality, multiple interactive systems, six generations of wearable computers, motion capture technology, high-end digital lighting projectors, various projection screens, smart electronically charged fabrics, multiple sensory sensitive devices and intelligent sensory charged alternative performance spaces. They were most often collaboratively created in order to augment all aspects of the performance and create the sense of community found in digital live dance performances/events. These works are identified as one continuous line of energy and discovery, each representing a slight variation on the premise that a working, caring, visceral and poetic content occurs beyond the technological tools. Consequently, a shift in the physical human’s psyche overwhelms the act of performance. Scholarship and reflection on the works have been integral to my creative process throughout. The goals of this thesis, the works created and the resulting methodologies are to investigate performance to heighten the multiple ways we experience and interact with the world. This maximizes connection and results in a highly interactive, improvisational, dynamic, non-linear, immediate, accessible, agential, reciprocal, emotional, visceral and transformative experience without boundaries between the virtual and physical for physical humans, cyborgs and cyber-humans alike.
