The Involvement of Ventral Tegmental Area Dopamine and CRF Activity in Mediating the Opponent Motivational Effects of Acute and Chronic Nicotine

A fundamental question in the neurobiological study of drug addiction concerns the mechanisms mediating the motivational effects of chronic drug withdrawal. According to one theory, drugs of abuse activate opposing motivational processes after both acute and chronic drug use. The negative experience of withdrawal is the opponent process of chronic drug use that drives relapse to drug-seeking and -taking, making the identification of the neurobiological substrates mediating withdrawal an issue of central importance in addiction research. In this thesis, I identify the involvement of the neurotransmitters dopamine (DA) and corticotropin-releasing factor (CRF) in the opponent motivational a- and b-processes occurring after acute and chronic nicotine administration. I report that acute nicotine stimulates an initial aversive a-process followed by a rewarding opponent b-process, and chronic nicotine stimulates a rewarding a-process followed by an aversive opponent b-process (withdrawal). These responses can be modeled using a place conditioning paradigm. I demonstrate that the acute nicotine a-process is mediated by phasic dopaminergic activity and the DA receptor subtype-1 (D1R) but not by tonic dopaminergic activity and the DA receptor subtype-2 (D2R) or CRF activity, and the opponent b-process is neither DA- nor CRF-mediated. I also demonstrate that the chronic nicotine a-process is DA- but not CRF-mediated, and that withdrawal from chronic nicotine (the b-process) decreases tonic but not phasic DA activity in the ventral tegmental area (VTA), an effect that is D2R- but not D1R-mediated. I show that a specific pattern of signaling at D1Rs and D2Rs mediates the motivational responses to acute nicotine and chronic nicotine withdrawal, respectively, by demonstrating that both increasing or decreasing signaling at these receptors prevents the expression of the conditioned motivational response. Furthermore, I report that the induction of nicotine dependence increases CRF mRNA in VTA DA neurons, and that blocking either the upregulation of CRF mRNA or the activation of VTA CRF receptors prevents the anxiogenic and aversive motivational responses to withdrawal from chronic nicotine. The results described in this thesis provide novel evidence of a VTA DA/CRF system, and demonstrate that both CRF and a specific pattern of tonic DA activity in the VTA are necessary for the aversive motivational experience of nicotine withdrawal.

Safetifying from interpersonal violence through Phasic Protective Sequencing : a classic grounded metatheory

This study generates a metatheory of interpersonal violence from an initial qualitatively highest scoring academic literature item and theoretically sampling over 200 additional literature items using classic grounded theory methodological analysis and synthesis. Danger emerged as the main concern of those experiencing interpersonal violence. Safetifying from interpersonal violence through the socio-psychophysical process of Phasic Protective Sequencing emerged as the resolution of the main concern. The metatheory of safetifying emerged in four parts: 1. The first was the socio-structural process of endangering with interpersonal violence through the interaction of the people and the elite resulting in endangerment (danger). 2. Danger precipitates the socio-psychological process of safetifying before interpersonal violence which involves awarenessing, setting up, and evaluating. 3. If the person is attacked, it precipitates the psycho-physical process of safetifying during interpersonal violence including resiliencing, finishing, and victoring. 4. The aftermath of an attack or the avoidance of an attack precipitates the psycho-social process of safetifying after interpersonal violence including reclaiming, relinquishing, and achieving. The metatheory of safetifying was shown to contribute uniquely to the field of interpersonal violence, to many related theories, and to have potential for therapeutic application. / Psychology / M.A. (Psychology)

Incentive motivation improves numerosity discrimination in children and adolescents

We recently showed that incentive motivation improves the precision of the Approximate Number System (ANS) in young adults. To shed light on the development of incentive motivation, the present study investigated whether this effect and its underlying mechanisms may also be observed in younger samples. Specifically, seven-year-old children (n = 23; 12 girls) and 14-year-old adolescents (n = 30; 15 girls) performed a dot comparison task with monetary reward incentives. Both age groups showed higher accuracy in a reward compared to a neutral condition and, similarly, higher processing efficiency as revealed by the drift rate parameter of the EZ-diffusion model. Furthermore, in line with the Incentive Salience Hypothesis, phasic pupil dilations—indicating the activation of the brain’s salience network—were greater in incentivized trials in both age groups. Together these finding suggest that incentive modulation improves numerosity discrimination in children and adolescents by enhancing the perceptual saliency of numerosity information. However, the observed reward anticipation effects were less pronounced in children relative to adolescents. Furthermore, unlike previous findings regarding young adults, the decision thresholds of children and adolescents were not raised by the monetary reward, which may indicate a more protracted development of incentive regulation of response caution than perceptual evidence accumulation.

ENERGY-EFFICIENT SENSING AND COMMUNICATION FOR SECURE INTERNET OF BODIES (IOB)

<p>The last few decades have witnessed unprecedented growth in multiple areas of electronics spanning low-power sensing, intelligent computing and high-speed wireless connectivity. In the foreseeable future, there would be hundreds of billions of computing devices, sensors, things and people, wherein the technology will become intertwined with our lives through continuous interaction and collaboration between humans and machines. Such human-centric ideas give rise to the concept of internet of bodies (IoB), which calls for novel and energy-efficient techniques for sensing, processing and secure communication for resource-constrained IoB nodes.As we have painfully learnt during the pandemic, point-of-care diagnostics along with continuous sensing and long-term connectivity has become one of the major requirements in the healthcare industry, further emphasizing the need for energy-efficiency and security in the resource-constrained devices around us.</p> <p>  </p> <p>  With this vision in mind, I’ll divide this dissertation into the following chapters. The first part (Chapter 2) will cover time-domain sensing techniques which allow inherent energy-resolution scalability, and will show the fundamental limits of achievable resolution. Implementations will include 1) a radiation sensing system for occupational dosimetry in healthcare and mining applications, which can achieve 12-18 bit resolution with 0.01-1 µJ energy dissipation, and 2) an ADC-less neural signal acquisition system with direct Analog to Time Conversion at 13pJ/Sample. The second part (Chapters 3 and 4) of this dissertation will involve the fundamentals of developing secure energy-efficient electro-quasistatic (EQS) communication techniques for IoB wearables as well as implants, and will demonstrate  2 examples: 1) Adiabatic Switching for breaking the αCV^2f limit of power consumption in capacitive voltage mode human-body communication (HBC), and 2) Bi-Phasic Quasistatic Brain Communication (BP-QBC) for fully wireless data transfer from a sub-6mm^3, 2 µW brain implant. A custom modulation scheme, along with adiabatic communication enables wireline-like energy efficiencies (<5pJ/b) in HBC-based wireless systems, while the BP-QBC node, being fully electrical in nature, demonstrates sub-50pJ/b efficiencies by eliminating DC power consumption, and by avoiding the transduction losses observed in competing technologies, involving optical, ultrasound and magneto-electric modalities. Next in Chapter 5, we will show an implementation of a reconfigurable system that would include 1) a human-body communication transceiver and 2) a traditional wireless (MedRadio) transceiver on the same integrated circuit (IC), and would demonstrate methods to switch between the two modes by detecting the placement of the transmitter and receiver devices (on-body/away from the body). Finally, in Chapter 6, we shall show a technique of augmenting security in resource-constrained devices through authentication using the Analog/RF properties of the transmitter, which are usually discarded as non-idealities in a digital transceiver chain. This method does not require any additional hardware in the transmitter, making it an extremely promising technique to augment security in highly resource-constrained scenarios. Such energy-efficient intelligent sensing and secure communication techniques, when combined with intelligent in-sensor-analytics at the resource-constrained nodes, can potentially pave the way for perpetual, and even batteryless systems for next-generation IoT, IoB and healthcare applications.</p>
