Les techniques non-conventionnelles de lithographie ont fait depuis deux décennies une entrée remarquée dans les sciences de l'ingénierie. Elles sont considérées aujourd'hui comme un enjeu majeur pour le développement des dispositifs. L'objectif principal de cette thèse est d'explorer de nouvelles voies pour la conception des micro&nanabiocapteurs en procédant à des impressions de poly(pyrrole) (PPy) à haute résolution par microtamponnage assisté par polymérisation catalytique (nanoCP-CCP) sur des substrats polymériques (poly(téréphtalate) d'éthylène (PETE), Copolymère d'oléfine cyclique (COC), polyétheréthercétone (PEEK), poly(éthylène 2,6-naphtalate (PEN) et le polyamide (PI)). Dans un premier temps, nous avons mis au point différentes techniques d'impression (greffage par impression, impression adressée et impression directe) et des conditions de polymérisation pour moduler les caractéristiques de PPy micro et nano-structurés, afin de contrôler la taille, la forme et les propriétés électriques désirées. Nous avons constaté que les paramètres les plus importants qui influent sur le processus d'impression surtout à l'échelle nanométrique sont: a) Le rapport des concentrations des réactifs pour le procédé de polymérisation qui comprend le Py-silane, nitrate d'argent (AgNO3), le chlorure du fer (III)/ le chlorure de Lithium (FeCl3/LiCl). b) Les paramètres physiques de la machine GeSIM; la pression d'impression, le niveau de contact, le temps d'encrage du tampon polydiméthylesiloxane (PDMS), et le temps d'impression. Finalement, on est arrivé à fabriquer des nanofils de PPy (PPy-NF) de 747±12,2 nm de largeur, 114±8 nm de hauteur et avec une séparation de 573±13,4 nm entre deux PPy-NF consécutifs. Ces films micro et nano-structurés ont été caractérisées par microscopie électronique à balayage (SEM), microscopie à force atomique (AFM) et spectrométrie de photon-électrons induits par rayons X (XPS). Dans une deuxième partie, on a développé des immunocapteurs à base de PPy-NF sensible aux biomarqueurs interleukine 8 et 6. Pour cela, différentes stratégies ont été adoptées pour immobiliser les anticorps spécifiques à ces deux biomarqueurs. Ces immunocapteurs ont été caractérisés par la méthode de spectroscopie d'impédance électrochimique (EIS). Les résultats obtenus par rapport à la sensibilité et la sélectivité sont très satisfaisants avec des limites de détection de l'ordre de quelques pg/L pour les deux immunocapteurs développés / Non-conventional lithography techniques have made for the two last decades a huge impact in the engineering sciences. They are now regarded as a main challenge for the development of the devices. The objective of this thesis is to explore new alternative possibilities for designing micro & nano biosensors based on poly(pyrrole) (PPy) high resolution microprinting attended by catalytic polymerization (nanoCP-CCP) on substrates Polymer (poly (terephthalate) ethylene (PETE), cyclic olefin copolymer (COC), polyetheretherketone (PEEK), poly (ethylene 2,6-naphthalate (PEN), and polyamide (PI)). In a first step We have developed various printing techniques (grafting printing, addressed printing and direct printing) and polymerization conditions to modulate the characteristics of PPy micro and nano-structured in order to control the size, shape and Electrical desired properties. We found that the most important parameters that affect the printing process especially at the nanoscale are: a.) The ratio of the concentrations of reagents for the polymerization process which includes the Py-silane, nitrate silver (AgNO3), iron chloride (III) / lithium chloride (FeCl3 / LiCl). b) The physical parameters of the GeSIM machine; the printing pressure, contact level, the inking time stamp of polydimethylsiloxane (PDMS), and printing time. Finally, we got to manufacture PPy nanowires (PPy-NW) 747 ± 12.2 nm wide, 114 ± 8 nm in height and with a separation of 573 ± 13.4 nm between two consecutive PPy-NW. These micro and nano-structured films were characterized by scanning electron microscopy (SEM), atomic force microscopy (AFM) and electron-photon spectroscopy induced by X-rays (XPS). In the second part, we have developed a PPy-NW-based immunosensors sensitive to interleukin 8 and 6 biomarkers. For this, different strategies have been adopted to immobilize antibodies specific to these two biomarkers. These immunosensors were characterized by electrochemical impedance spectroscopy method (EIS). The results obtained in relation to the sensitivity and selectivity are very satisfactory with the security detection limits of a few pg / L for both developed immunosensors

Des composés de type 3-arylpyrroles appelés pyakols ont montré une activité antimitotique sur des cellules tumorales murines, avec en particulier un effet sur les microtubules. Ce type d’activité biologique présentant un intérêt important en cancérologie, le travail présenté dans ce manuscrit est consacré à l’étude de ces hétérocycles. La première partie a pour objectif de développer une stratégie de synthèse permettant d’accéder efficacement au composé chef de file (pyakol I), basée sur la cyclisation d’intermédiaires α-amino-ynols catalysée par des complexes d’or. L’évaluation de l’activité biologique du pyakol I sur le cycle cellulaire et le cytosquelette de diverses lignées tumorales humaines a été réalisée. Les premiers résultats ont révélé une action originale du pyakol I sur le cytosquelette, provoquant une désorganisation du réseau de microtubules et un défaut de positionnement du fuseau mitotique. La séquence réactionnelle a ensuite été validée en l’appliquant pour la réalisation de modulations autour du motif 3-arylpyrrole ainsi que pour l’obtention de molécules marquées. La deuxième partie concerne un travail de méthodologie basée sur la réaction de cyclisation à l’or pour la synthèse de nouveaux pyrroles trifluorométhylés polysubstitués. La stratégie utilisant le trifluoroacétaldéhyde comme substrat de départ permet d’accéder à divers 3-trifluorométhylpyrroles avec de bons rendements, dans des conditions douces. / A family of 3-arylpyrroles named pyakols have shown antimitotic properties on murine cell lines, displaying in particular an effect on microtubules. Given the interest of these properties in cancerology, this work is focused on these heterocycles. The objective of the first part was to develop a synthetic strategy based on the gold-catalysed cyclisation of α-amino-ynols intermediates in order to access the lead (Pyakol I). Then, the evaluation of the biological activity of this molecule on the cell cycle and on the cytoskeleton of various human tumoral cell lines was carried out. The first results revealed an original effect on the organization of the microtubules network and the positioning of the mitotic spindle. The developed strategy was then validated by modulating the 3-arylpyrrole moiety on diverse positions, and used for the synthesis of labelled derivatives. The second part of this manuscript focused on the development of a methodology to synthesize new polysubstituted 3-trifluoromethylpyrroles, based on the gold-catalyzed cyclisation reaction. Using trifluoroacetaldehyde as building-block, various trifluoromethylated pyrroles were obtained in mild conditions with good yields.

Les pyrrole-2-aminoimidazoles (P-2-AIs) sont des métabolites secondaires spécifiques des éponges marines. Ils sont principalement rencontrés dans les familles d’éponge Agelasidae, Halichondridae et Axinellidae. Ainsi l’étude chimique de l’éponge marine Axinella donnani dans le but d’isoler de nouveaux composés P-2-AIs a-t-elle permis un élargissement des connaissances au niveau de la diversité de la famille ainsi qu’une meilleure compréhension de leur biogénèse. Deux lots de cette éponge ont été étudiés afin de mener à l’isolement de 29 composés : parmi lesquels six P-2-AIs dimères nouveaux, un nouveau composé de la famille des manzacidines et un nouveau dimère 3,5-dibromotyrosinique. Les structures de ces nouvelles molécules ont déterminées à l’aide d’expériences RMN 1D et 2D. Une étude de leur stéréochimie absolue a aussi été menée à l’aide de dichroïsme circulaire. Par ailleurs la réalisation d’une chimiothèque d’analogues du dispacamide A a permis l’obtention de 29 analogues différemment substitués qui seront testés sur de nombreuses cibles biologiques afin d’évaluer leurs activités. / Pyrrole-2-aminoimidazoles (P-2-AIs) are secondary metabolites specifically encountered in marine sponges. They are mainly found in Agelasidae, Halichondridae and Axinellidae families. Thus, the chemical study of the marine sponge Axinella donnani has led to the extension of the current knowledge about the diversity of the family and a better understanding of their biogenesis. Two batches of the sponges have been studied to lead to the isolation of 29 compounds: among them six P-2-AIs new dimers, a new mazacidin molecule and a new 3,5-bromotyrosine dimer. Their structures have been determined with 1D and 2D NMR spectroscopy experiments. Their absolute configuration has also been studied with circular dichroïsm. Additionally, 29 analogues of dipacamide A have been synthetized in order to test these molecules on various biological targets.

Este trabalho descreve a preparação e caracterização de biocátodos para biocélula a combustível Etanol e Metanol//O2 utilizando a enzima lacase (trametes versicolor) num sistema de transferência eletrônica mediada (TEM). Na primeira etapa do trabalho, os resultados de cinética enzimática com a enzima lacase em solução e imobilizada sobre tecido de carbono mostraram que os vários parâmetros experimentais (pH, temperatura, estabilidade) analisados devem ser considerados, a fim de se obter atividade máxima com os biocatalisadores. Além disso, em relação aos testes cinéticos e de estabilidade, pode-se inferir que o dendrímero PAMAM pode ser empregado como um bom agente imobilizante na preparação de bicátodos para biocélula a combustível enzimática. Na segunda etapa do trabalho, uma semibiocélula Etanol//O2 foi testada e os eletrocatalisadores testados foram o verde de metileno (VM) e o azul de meldola (AM). Os testes de potência mostraram a importância da presença do mediador ABTS e do eletrocatalisador (VM) para melhorar o desempenho do dispositivo. Na terceira etapa do trabalho, eletrodos com diferentes mediadores (ABTS, ferro porfirina, ferroceno, complexo de ósmio e complexo de rutênio) e com polipirrol eletropolimerizado na superfície do eletrodo foram testados numa semibiocélula Metanol//O2. Os testes de semibiocélula Etanol e Metanol//O2 com transferência eletrônica mediada mostraram que os biocátodos preparados com o dendrímero PAMAM e com os diferentes eletrocatalisadores e mediadores, se mostraram capazes de gerar densidades de potência competitivas em relação aos valores encontrados na literatura. / This work describes the preparation and characterization of biocathodes for Ethanol and Methanol//O2 biofuel cell using the enzyme laccase (trametes versicolor) enzyme and mediated electron transfer (MET). Investigation of the enzymatic kinetics of the enzyme laccase in solution and immobilized onto carbon platforms showed that the analyzed experimental parameters (pH, temperature, and stability) must be considered for maximum activity to be achieved. The kinetic and stability tests revealed that PAMAM dendrimers constitute very good immobilization agent to prepare biocathodes for enzymatic biofuel cell. The second part of this work, dealt with Ethanol//O2half-cell using methylene green (MG) ormeldola blue (MB) as electrocatalyst. The power test evidenced that it is important to have ABTS as mediator and an electrocatalyst, to ensure that the device performs better. The third part of this work evaluated electrodes with distinct mediators (ABTS, iron porphyrin, ferrocene, osmium complex, and ruthenium complex) and containing electropolymerized polypyrrole on their surface in a Methanol//O2half-cell. Ethanol and Methanol//O2 half-cell tests with mediated electron transfer showed that the biocathodes prepared with PAMAM dendrimers, electrocatalyst, and distinct mediators generated competitive power densities as compared with literature data.

Desenvolvimento de novas plataformas poliméricas para detecção eletroquímica de amônia e uréia / Development of new polymeric platforms for ammonia and urea electrochemical detection

Este trabalho apresenta o desenvolvimento de (bio)sensores baseados em plataformas poliméricas convencionais e nanoestruturadas. Os sensores estudados contém poli(pirrol) como transdutor eletroquímico, responsável pela oxidação de amônia a 0,35 V. Como a enzima urease hidrolisa cataliticamente uréia a amônia, após a imobilização da enzima sobre o poli(pirrol), obtém-se um biossensor para detecção indireta de uréia, via amônia. A detecção de amônia pelo poli(pirrol) foi otimizada em termos da densidade de carga de poli(pirrol) presente no sensor, e do potencial de trabalho. Então verificou-se a ação do polímero poli(5-amino-1-naftol) na redução do sinal dos interferentes ácidos úrico e ascórbico. Após a otimização do sensor-base, estudou-se a influência do tipo de imobilização da urease ao substrato polimérico na sensibilidade e estabilidade do biossensor de uréia. Concluiu-se que a pouca quantidade de enzima imobilizada através das metodologias estudadas era o fator limitante da sensibilidade de detecção, e então implementou-se a nanoestruturação do poli(pirrol), para aumentar a área superficial disponível para a imobilização da urease. Preparou-se filmes de poli(pirrol) nanoestruturado (macroporoso e nanofios), e a compreensão dos fenômenos observados só foi possível ao aliar características geométricas e propriedades físico-químicas intrínsecas de sistemas nanoestruturados. / This work presents the development of (bio)sensors based on conventional and nanostructured polymeric platforms. The analyzed sensors contain poly(pyrrole) as electrochemical transducer, responsible for ammonia oxidation at 0.35 V. As the enzyme urease catalytically hydrolyses urea to ammonia, after enzyme immobilization onto poly(pyrrole), a biosensor for urea indirect detection, via ammonia, is obtained. Ammonia sensing by poly(pyrrole) was optimized in terms of both charge density of poly(pyrrole) present in the sensor and working potential. In sequence, poly(5-amino-1-naphtol) role as interferents (uric and ascorbic acids) signal blocker was verified. After optimization of the base-sensor, the influence of urease immobilization method on both detection sensitivity and biosensor durability was investigated. It was found that the small amount of enzyme immobilized using the compared methods limited detection sensitivities, and then nanostructuration of poly(pyrrole) was implemented, in order to increase the surface area available for urease immobilization. Nanostructured poly(pyrrole) films (macroporous and nanowires) were prepared, and the fully understanding of the observed phenomena was only possible combining geometric characteristics and physical-chemical properties related to nanostructured systems.

Conductive Nanocrystalline Cellulose Polymer Composite Film as a Novel Mediator in Biosensor Applications

Recent biosensors using glucose oxidase enzyme to detect glucose (“blood sugar”) were made with intrinsic conducting polymers such as poly pyrrole (PPY) to mediate the reaction. PPY coated electrodes were difficult to employ via eletropolymerization because PPY is only soluble in solvents potentially damaging to enzymes. Nano crystalline cellulose – poly pyrrole (NCC-PPY) colloid circumvents this by forming natural, enzyme compatible, and hydrophilic films mechanically bound to electrodes using easy-to-disperse colloids. NCC-PPY was studied as mediator to investigate use in biosensor applications. Using NCC-PPY film casted on microfabricated interdigitated electrodes, a glucose biosensor with sensitivity factor of 20 was achieved. NCC-PPY showed enhanced catalysis with no enzyme inactivation and a total current of 2mA. Enhanced sensitivity was attributed to resistance changes of doped PPY, redox mediation, and compatibility of cellulose with enzyme.

Different substituted thiophene and pyrrole monomers namely hexamethylene (bis-3-thiopheneacetamide) (HMTA), N-(4-(3-thienylmethylene)-oxycarbonylphenyl)maleimide (MBThi), 2,4,6-Tris-(4-pyrrol-1-yl-phenoxy)-[1,3,5]triazine (TriaPy), 2,4,6-Tris-(thiophen-3-ylmethoxy)-[1,3,5] triazine (TriaTh), and N-(2-(thiophen-3-yl)methylcarbonyloxyethyl) maleimide (NMT) were synthesized. The chemical structures of the monomers were characterized by Nuclear Magnetic Resonance (1H-NMR and 13C-NMR) and Fourier Transform Infrared (FTIR) Spectroscopies. Electrochemical behavior of the monomers in the presence and absence of comonomers were studied by cyclic voltammetry. Subsequently, monomers were homopolymerized and copolymerized via electrochemical methods and the resultant polymers were characterized by FTIR, Scanning Electron Microscopy (SEM) and conductivity measurements. Second part of the study was devoted to investigate one of most interesting property of conducting polymers, the ability to switch reversibly between the two states of different optical properties, &ldquo / electrochromism&rdquo / . In recent years there has been a growing interest in application of conducting polymers in electrochromic devices. Thus, electrochromic properties of the synthesized conducting polymers were investigated by several methods like spectroelectrochemistry, kinetic and colorimetry studies. Spectroelectrochemistry experiments were performed in order to investigate key properties of conjugated polymers such as band gap, maximum absorption wavelength, the intergap states that appear upon doping and evolution of polaron and bipolaron bands. Switching time and optical contrast of the homopolymers and copolymers were evaluated via kinetic studies. Results implied the possible use of these materials in electrochromic devices due to their good electrochromic properties.
