Previous issue date: 2013-08-05 / Changes in the results of erythrocyte standards require decisions and guide the conduct of clinical regarding prognosis and treatment of any diseases. The use of modern equipment that make the blood cell count used to calculate the RDW (red cell distribution width) and its fractions RDW-CV and RDW-SD, which provide a quantitative assessment of heterogeneity of each erythrocyte. Given the limited information available in the literature about the RDW for the domestic cat, this study aims to compare the values of RDW, with MCV and reticulocytes from 40 (n=14males, n=26 females) healthy mongrel cats, with age ranging from 1.2 to 5.5 years (3.0 ± 1.2) were used. The animals were kept in individual cages all over the experiment. Blood samples were taken directly from the external jugular vein and separated into two tubes: one containing anticoagulant EDTA (ethylene diamine tetra acetic acid) to 10% for the complete blood count and the second without anticoagulant for biochemical tests. The results showed that there is a direct relationship between values of RDW-SD and RDW-CV and an inverse correlation between RDW-CV with MCV. There was no relation between RDW and reticulocyte aggregates or punctate. There was no relation between sex and RDW as well. RDW values determined in this experiment can be used as a resource for future research and also as reference values for the feline species. / As alterações nos resultados dos padrões eritrocitários demandam decisões e norteiam a conduta do clínico quanto ao prognóstico e a terapêutica de eventuais patologias. A utilização de equipamentos modernos que fazem a contagem celular sanguínea permite calcular a amplitude de distribuição dos eritrócitos ou RDW (red cell distribution width) e suas frações RDW-CV e RDW-SD, que provêem uma avaliação quantitativa da heterogenicidade de cada eritrócito. Diante da escassa informação disponível na literatura científica sobre os valores de RDW para o gato doméstico, este estudo se propõe a comparar os valores de RDW com o VCM e reticulócitos de 40 gatos domésticos hígidos, sem raça definida, com idade variando de 1,2 a 5,5 anos (3,0±1,2), sendo 14 machos e 26 fêmeas provenientes do gatil da instituição. O sangue foi colhido diretamente da veia jugular externa e separado em dois tubos: um contendo anticoagulante EDTA (ácido etileno diamino tetra acético) a 10%, para a realização do hemograma e o segundo, sem anticoagulante, para as provas bioquímicas de triagem. Os resultados encontrados mostraram que existe uma relação direta significativa entre os valores de RDW-SD e RDW-CV e uma correlação inversa entre os valores de RDW-CV o VCM. Não houve correlação entre os valores de RDW e reticulócitos agregados ou ponteados. O sexo não interfere no diâmetro das hemácias. Os valores de RDW determinados neste experimento podem ser usados como fonte de consulta para futuras pesquisas e também como valores de referência da espécie felina.

Função esplênica e eventos de adesão celular em Anemia Falciforme e em Esferocitose Hereditária / Splenic function and cellular adhesion events in Sickle Cell Anemia and in Hereditary SpherocytosisFerreira, Priscilla Carnavale Gomes 02 March 2018 (has links)
As Anemias Hemolíticas compreendem um grupo de doenças em que há redução acentuada na sobrevivência dos glóbulos vermelhos circulantes e a medula óssea não é capaz de compensação, mesmo aumentando sua produção, o que causa anemia desde os primeiros anos da vida da pessoa. Dentre as doenças deste grupo, a Anemia Falciforme (SCA) e a Esferocitose Hereditária (HS) destacam-se por se tratarem de enfermidades com defeitos genéticos intrínsecos das células vermelhas (RBCs) que geram complicações multissistêmicas agudas e crônicas em seus portadores. Por vias patofisiológicas distintas, reticulócitos e respectivas hemácias defeituosas de tais doenças, falciformes e esferócitos, são continuamente aprisionados e fagocitados no baço, importante órgão de destruição de células velhas e/ou defeituosas via hemólise extravascular, o que leva progressivamente à disfunção e eventual perda da função esplênica. O objetivo desse trabalho é avaliar o papel do baço em relação à habilidade e ao fenótipo adesivos de reticulócitos (ret) e eritrócitos (erit) em pacientes com SCA e HS, com e sem função esplênica preservada. Amostras de sangue de 37 pacientes (22 SCA and 15 HS) com função esplênica e 19 pacientes (13 SCA e 6 HS) sem ela foram avaliadas. Ainda, sangue de 22 crianças com SCA foi coletado em estudo longitudinal dos 6 e 29 meses de vida. Todas as amostras de sangue foram analisadas quanto à função esplênica (Contagem de células PIT e de corpúsculos de Howell-Jolly - HJB), quanto ao perfil imunofenotípico celular (em % e em média de intensidade de fluorescência - MFI) e quanto à habilidade de adesão das células vermelhas à laminina e à linhagem celular endotelial HMEC-1. A análise da transição da perda de função esplênica demonstrou que a mesma se intensificou a partir dos 3 anos de idade (PIT: r=0,8; p<0,0001; HJB: r=0,7; p<0.0001). Quanto à imunofenotipagem celular, a contagem PIT se correlacionou positivamente, principalmente com os marcadores CD147 (%ret: r=0,6; p<0,0001; MFIret: r=0,6; p<0,0001; %erit: r=0,7; p<0,0001; MFIerit: r=0,6; p<0,0001), LuBCAM (%ret: r=0,5; p=0,004; MFIret: r=0,6; p<0,0001; %erit: r=0,6; p<0,0003; MFIerit: r=0,4; p<0,004) and CD58 (%ret: r=0,4; p=0,006; MFIret: r=0,5; p<0,0013; %erit: r=0,4; p<0,009; MFIerit: r=0,6; p<0,0001). Na comparação entre ausência ou presença do baço, a perda de sua função exerceu influência no aumento da expressão de adesão de RBCs em SCA, principalmente CD147 (%ret: p=0,002; MFIret: p=0,003; %erit: p<0,0001; MFIerit: p=0,005), LuBCAM (%ret: p=0,0001; MFIret: p<0,0001; %erit: p<0,0001; MFIerit: p<0,0001) e CD58 (%ret: p=0,007; MFIret: p=0,006; %erit: p=0,003; MFIerit: p=0,0004), embora a adesão celular tenha diminuído em pacientes HS esplenectomizados. Na comparação entre as doenças, pacientes HS com o baço apresentaram maior freqüência de adesão celular em relação aos SCA, notavelmente em relação ao LuBCAM (%ret: p=0,0008; MFIret: p=0,03; %erit: p<0,0001; MFIerit: p=0,0002), CD58 (%ret: p=0,0009; %erit: p=0,003) e CD44 (%ret: p=0,009; %erit: p<0,003). No entanto, as amostras SCA sem função esplênica tiveram maior expressão de adesão celular para CD147 (%ret: p=0,006; MFIret: p=0,02; %erit: p=0,02), LuBCAM (%ret: p=0,004; MFIret: p<0,0001), CD36 (%ret: p=0,0002; MFIret: p=0,01), CD242 (%ret: p=0,0008; %erit: p=0,05) e CD49d (%ret: p=0,04). Em relação ao Ensaio de Adesão in vitro, na ausência de baço, os RBCs SCA apresentaram maior adesividade à laminina do que os RBCs SCA com função esplênica preservadaem todas as taxas de fluxo de tensão de cisalhamento empregadas (0,5 dyne/cm2: p=0,01; 1 dyne/cm2: p=0,02; 2 dynes/cm2: p=0,03; 3 dynes/cm2: p=0,03; 5 dynes/cm2: p=0,04 e 7 dynes/cm2: p=0,03). Especialmente, reticulócitos de pacientes sem baço apresentaram maior adesividade à HMEC-1 em baixas tensões de cisalhamento (1 dyne/cm2) em ambas as doenças (SCA: p=0,03; HS: p=0,03). Por fim, reticulócitos apresentaram maior habilidade adesiva à células endoteliais em indivíduos SCA do que em pacientes HS, com (0,5 dyne/cm2: p=0,04; 1 dyne/cm2: p=0,03) ou sem baço (0,5 dyne/cm2: p=0,02; 2 dynes/cm2: p=0,01; 3 dynes/cm2: p=0,03; 5 dynes/cm2: p=0,02 e 7 dynes/cm2: p=0,03). Nossos resultados indicam que embora pertençam ao grupo de Anemias Hemolíticas, as patofisiologias e evoluções clínicas distintas de SCA e de HS levam a padrões imunofenotípicos diferentes de expressão da adesão celular. Na SCA, a ausência de função esplênica teria direta relação com o aumento do fenótipo pró-adesivo e com a adesividade de RBCs SCA, o que traz sérias consequências clínicas aos pacientes, enquanto na HS sem baço, de maneira geral, os eventos de adesão celular são minimizados, embora ainda apresentem reticulócitos e eritrócitos adesivos circulantes após a esplenectomia. / Hemolytic Anemias comprise a group of diseases in which there is marked reduction in the survival of circulating erythrocytes and the bone marrow is not capable of compensation, even by increasing its production, which causes anemia from the first years of the person\'s life on. Among the diseases of this group, Sickle Cell Anemia (SCA) and Hereditary Spherocytosis (HS) stand out for being diseases with intrinsic genetic defects of red blood cells (RBCs) that generate acute and chronic multisystemic complications in their patients. By distinct pathophysiological pathways, reticulocytes and these disease\'s respective defective erythrocytes, sickle and spheroid ones, are continuously trapped and phagocytosed in the spleen, important organ of destruction of old and/or defective cells via extravascular hemolysis, which progressively leads to dysfunction and eventual loss of splenic function. The objective of this study was to evaluate the role of the spleen in relation to the reticulocyte (ret) and erythrocyte (eryt) adhesive ability and adhesion phenotype in patients with SCA and HS, with and without preserved splenic function. Blood samples from 37 patients (22 SCA and 15 HS) with splenic function and 19 patients (13 SCA and 6 HS) without it were evaluated. Still, blood from 22 children with SCA was collected in a longitudinal study from 6 to 29 months of age. All blood samples were analyzed for splenic function [pitted cells (PIT) and Howell-Jolly bodies (HJB) counting], for the cellular immunophenotypic profile (in % and in mean fluorescence intensity - MFI) and for the adhesive ability of RBCs to laminin and to endothelial cell line HMEC-1. Analysis of the splenic function loss transition showed that it intensified from 3 years of age on (PIT: r=0.8, p<0.0001; HJB: r=0.7, p<0.0001). Regarding the cellular immunophenotyping, PIT count correlated positively, mainly with CD147 markers (%ret: r=0.6, p<0.0001; MFIret: r=0.6, p<0.0001; %eryt: r=0.7, p<0.0001; MFIeryt: r=0.6, p<0.0001), LuBCAM (%ret: r=0.5, p=0.004; MFIret: r=0.6, p<0.0001; %eryt: r=0.6, p<0.0003; MFIeryt: r=0.4, p<0.004) and CD58 (%ret: r=0.4, p=0.006; MFIret: r=0.5, p<0.0013; %eryt: r=0.4, p<0.009; MFIeryt: r=0.6, p<0.0001). In the comparison between spleen absence or presence, the loss of its function exerted influence on the increase of RBCs adhesion expression in SCA, mainly on CD147 (%ret: p=0.002; MFIret: p=0.003; %eryt: p<0.0001; MFIeryt: p=0.005), LuBCAM (%ret: p=0.0001; MFIret: p<0.0001; %eryt: p<0.0001; MFIeryt: p<0.0001) e CD58 (%ret: p=0.007; MFIret: p=0.006; %eryt: p=0.003; MFIeryt: p=0.0004), although cell adhesion has been decreased in splenectomized HS patients. In the comparison between diseases, HS patients with spleen showed higher cell adhesion frequency compared to SCA, notably in relation to LuBCAM (%ret: p=0.0008; MFIret: p=0.03; %eryt: p<0.0001; MFIeryt: p=0.0002), CD58 (%ret: p=0.0009; %eryt: p=0.003) and CD44 (%ret: p=0.009; %eryt: p<0.003). However, SCA samples without splenic function had higher cell adhesion expression for CD147 (%ret: p=0.006; MFIret: p=0.02; %eryt: p=0.02), LuBCAM (%ret: p=0.004; MFIret: p<0.0001), CD36 (%ret: p=0.0002; MFIret: p=0.01), CD242 (%ret: p=0.0008; %eryt: p=0.05) and CD49d (%ret: p=0.04). Concerning the in vitro Adhesion Assay, in the spleen absence, SCA RBCs showed greater adhesiveness to laminin than SCA RBCs with preserved splenic function did at all shear stress flow rates applied (0.5 dyne/cm2: p=0.01, 1 dyne/cm2: p=0.02, 2 dynes/cm2: p=0.03, 3 dynes/cm2: p=0.03, 5 dynes/cm2: p=0.04 and 7 dynes/cm2:p=0.03). Especially, reticulocytes from patients without spleen showed higher adhesiveness to HMEC-1 at low shear stresses (1 dyne/cm2) in both diseases (SCA: p=0.03; HS: p=0.03). Finally, reticulocytes showed greater adhesion ability to endothelial cells in SCA subjects than in HS patients, with (0.5 dyne/cm2: p=0.04 and 1 dyne/cm2: p=0.03) or without spleen (0.5 dyne/cm2: p=0.02, 2 dynes/cm2: p=0.01, 3 dynes/cm2: p=0.03, 5 dynes/cm2: p=0.02 and 7 dynes/cm2: p=0.03). Our results indicate that although both diseases belong to the Hemolytic Anemias group, SCA and HS distinct pathophysiologies and clinical evolution lead to different immunophenotypic patterns of cell adhesion expression. In SCA, the absence of splenic function may have a direct relation with the increase of SCA RBCs proadhesive phenotype and adhesiveness, which brings serious clinical consequences to the patients, whereas in HS without spleen, in general, cellular adhesion events are minimized, although they still present adhesive circulating reticulocytes and erythrocytes after splenectomy.

Erythropoietin is a glycoprotein which stimulates the erythropoiesis, clinically used for the treatment of renal anaemia. The biological and chromatographic methods for the potency evaluation of recombinant human erythropoietin (rhEPO) in pharmaceutical products were validated in the present work. The normocythaemic mice bioassay was carried out in female BALB/c strain, with multiple injections schedule and reticulocytes counted by automated flow cytometry. The dose-response curve was linear (r2=0.9708) in the concentration range of 1 to 64 IU/mL. A reversed-phase liquid chromatography method (RP-LC) was developed and
validated using a Jupiter C4 column (250 mm x 4.6 mm), maintained at ambient temperature (25°C). The mobile phase A consisted of 0.1% trifluoroacetic acid (TFA) and mobile phase B consisted of 0.08% TFA:acetonitrile (30:70, v/v), run in linear gradient: 0.1-60 min, 0.1-100% of B at flow rate of 0.5 mL/min with detection at 280 nm. The chromatographic separation was obtained within 60 min and it was linear (r2=0.9997) in the concentration range of 10-150 μg/mL. The procedures were validated evaluating parameters such as, the specificity, linearity, precision, accuracy, robustness, and limit of quantitation, as well as, the limit of detection evaluated in the validation of the chromatographic method. The methods were applied for the potency evaluation of the alfa and beta rhEPO in pharmaceutical products, which were analysed by the chromatographic method and compared to the bioassay showing mean differences between the estimated potencies of 11.2% higher for the RP-LC. The alternative established represents a contribution towards the replacement of the animals improving the quality control and assuring the safety and efficacy of the biological product. / Eritropoietina é uma glicoproteína que estimula a eritropoiese e é usada clinicamente para o tratamento de anemias associadas à falência renal crônica. No presente trabalho foram validados métodos biológico e cromatográfico para a avaliação de potência de eritropoietina humana recombinante (rhEPO) em produtos farmacêuticos. O ensaio biológico foi validado em camundongos normocitêmicos, fêmeas, da linhagem BALB/c, com protocolo de injeções múltiplas e contagem automatizada dos reticulócitos por citometria de fluxo. vermelhas. A eritropoetina humana recombinante (rhEPO) produzida em laboratório e usada
como medicamento pode apresentar as isoformas alfa e beta, chamadas de alfaepoetina e
betaepoetina humana recombinante. O tratamento com rhEPO melhora bastante a vida de
pacientes anêmicos e/ou em diálise, reduzindo a necessidade de transfusão de sangue. A
grande demanda por este biofármaco justifica o esforço de pesquisa não só para conhecimento
de seus mecanismos de ação e desenvolvimento de processos de produção, como também
para controle da qualidade do produto. É de grande importância o desenvolvimento de
métodos de análise que diminuam a margem de erro e aumentem a precisão e a confiabilidade
dos resultados, além de reduzir o tempo de liberação para o mercado.
O ensaio de determinação da potência da alfaepoetina humana recombinante
produzida por Bio-Manguinhos é realizado pelo método de camundongos normocitêmicos,
com contagem de reticulócitos por microscopia. Estemétodo de contagem é demorado, pouco
preciso e desgastante para o analista. O objetivo deste trabalho é estudar o uso do método de
citometria de fluxo na contagem de reticulócitos para o ensaio de potência da alfaepoetina
humana recombinante. Visando a implantação do método no Departamento de Controle de
Qualidade (DEQUA) de Bio-Manguinhos, o desenvolvimento deste trabalho teve também
como objetivo comparar a sua eficácia em relação aoprocesso atualmente utilizado, e a
realizar os testes necessários à sua validação.
Os camundongos foram inoculados com três doses diferentes de amostra ou material
de referência e os reticulócitos presentes no sangue coletado foram marcados com o corante
fluorescente laranja de tiazol (Retic-COUNT). Os reticulócitos foram contados em citômetro
de fluxo FACSCalibur e seu percentual foi utilizadopara a determinação da potência de
acordo com os cálculos da Farmacopéia Européia 6ª edição. Para a comparação entre os
métodos, vinte lotes de alfaepoetina humana recombinante produzidos por Bio-Manguinhos
tiveram sua potência determinada pela contagem de reticulócitos por citometria e
microscopia. Para a determinação da repetitividade e da precisão intermediária, foi utilizado
um único lote de alfaepoetina humana recombinante. Para avaliar a concordância entre os
métodos de citometria de fluxo e microscopia, foi utilizado o teste estatístico de Bland-Altman, com comparação dos vinte resultados obtidoscom os mesmo lotes, analisados a
partir das mesmas amostras de sangue.
Os resultados apresentaram boa repetitividade e precisão intermediária e mostraram
que os métodos são equivalentes no que diz respeitoà determinação da potencia da rhEPO
produzida por Bio-Manguinhos. No entanto, em funçãoda alta probabilidade de erros
associados ao método de microscopia, a introdução da citometria de fluxo mostra-se uma
alternativa bastante promissora para suprir a demanda de testes com precisão,
reprodutibilidade e maior velocidade de liberação de lotes. / Erythropoietin is the hormone responsible for the regulation of the red cells’
production. The recombinant human erythropoietin (rhEPO) produced in laboratory and used
as a drug exists in the alpha and beta isoforms, called alpha epoietin and beta epoietin. The
treatment with rhEPO improves the life of anemic patients and/or on dialysis, reducing the
need for blood transfusion. The high demand for this biopharmaceutical product justifies the
research effort not only for the knowledge of its mechanisms of action and development of
production processes, but also for the product quality control. The development of quality
control methods able to reduce errors and increase the accuracy and the reliability of the
results, along with a reduction in the release timeto the market, is of great importance.
The assay to determine the biological activity of the recombinant human alpha
epoietin produced in Bio-Manguinhos is performed bythe normocythaemic mice method and
reticulocyte counting by microscopy. This counting method is time consuming, imprecise and
stressful to the analyst. The present study aimed to study the use of the flow cytometric
method for reticulocyte counting in the biological activity assay for the human alpha epoietin.
Intending to implement this new method in the Quality Control Department of
Bio-Manguinhos, this work also compared its effectiveness with the currently used method,
and the tests required to its validation were performed.
The mice were inoculated with three different dosesof sample and reference material,
and the reticulocytes on the collected blood were stained with the fluorescent dye thiazole
orange (Retic-COUNT). The reticulocytes were counted in a FACSCalibur flow cytometer
and their percentage was used for the biological activity determination in accordance with the
European Pharmacopoeia 6
th
edition. To compare the methods, the biological activity of
twenty batches of recombinant human alpha epoetin produced by Bio-Manguinhos was
determined trough both cytometry and microscopy. Tothe determination of repeatability and
intermediate precision, only a single batch of recombinant human alpha epoetin was used. For
the evaluation of agreement between both methods, the Bland-Altman test was used to the
compare the twenty results obtained from the same batches, analyzed from the same blood
samples.
The results showed good repeatability and intermediate precision and indicated that
the two methods are equivalent when it concerns thedetermination of the rhEPO biological
activity. However, due to the high probability of errors associated with the microscopy
method, the introduction of the flow cytometry method seems to be a promising alternative to
meet the demand for analyses with precise and reproducible results, along with a good speed
in releasing batches for the market.

The chromatographic method for the potency evaluation of recombinant human erythropoietin (rhEPO) in pharmaceutical products was validated in the present work. A size-exclusion liquid chromatography method (SE-LC) was validated using a BioSep-SEC-S
2000 column (300 mm x 7,8 mm I.D.), maintained at ambient temperature (25°C). The mobile phase consisted of 0.001 M monobasic potassium phosphate, 0.008 M dibasic sodium phosphate and 0.2 M sodium chloride buffer, pH 7.4, run isocratically at a flow rate of 0.5 mL/min with detection at 214 nm. The chromatographic separation was obtained within 30 min and it was linear in the concentration
range of 5-150 μg/mL (r2=0.9991). Validation parameters were evaluated such as sensitivity, precision, accuracy, detection limit, quantitation limit and robustness. The specificity was evaluated by the peak purity of the Recombinant Human Erythropoietin Biological Reference Preparation of European Pharmacopoeia (rhEPO-SBR) storage under stress conditions. The proposed method was
applied for the analysis of erythropoietin in pharmaceutical products, evaluating also the dimmers and
high-molecular-mass forms. Moreover, was performed the analysis of erythropoietin by reversedphase liquid chromatography (RP-LC), evaluating the sulphoxides and deamidates forms. The correlation with the methods was established, showing mean differences between the estimated
potencies of 2.50% lower for the bioassay, and 9.29% higher for the RP-LC, compared with the sizeexclusion
liquid chromatography method, with significative correlation, conform calculated for the Pearson s correlation coefficient (r=0,9629 e r=0,9422, respectively). The alternative established represents a contribution towards the reduction or replacement of
the animals improving the quality control and assuring the safety and efficacy of the biological product. / A eritropoietina é uma proteína endógena que estimula a eritropoiese. No presente trabalho foi validado método cromatográfico por exclusão molecular (CL-EM) para a avaliação de potência de eritropoietina humana recombinante (rhEPO) em produtos farmacêuticos, empregando coluna BioSep-SEC-S 2000

determinação da pureza do pico da Eritropoietina Humana Recombinante Substância Biológica de Referência da Farmacopéia Européia (rhEPO-SBR) submetida à degradação sob condições forçadas. O método proposto foi utilizado para análise de eritropoietina em produtos farmacêuticos,
determinando-se as formas diméricas e de alta massa molecular. Paralelamente, efetuaram-se análises por cromatografia líquida em fase reversa (CL-FR), determinando-se as formas de sulfóxidos/desamidados. Estabeleceu-se correlação entre os métodos, demonstrando que os produtos
farmacêuticos forneceram diferenças médias 2,50% menores pelo bioensaio, e 9,29% maiores para CL-FR, em relação ao método por cromatografia líquida por exclusão molecular, com correlação significativa, conforme calculado pelo coeficiente de correlação de pearson (r=0,9629 e r=0,9422,
respectivamente). Desse modo, pesquisou-se alternativa no contexto da redução ou substituição do uso de animais,
contribuindo para aprimorar o controle da qualidade, garantindo a segurança e eficácia terapêutica do
produto biológico.

Função esplênica e eventos de adesão celular em Anemia Falciforme e em Esferocitose Hereditária / Splenic function and cellular adhesion events in Sickle Cell Anemia and in Hereditary SpherocytosisPriscilla Carnavale Gomes Ferreira 02 March 2018 (has links)
As Anemias Hemolíticas compreendem um grupo de doenças em que há redução acentuada na sobrevivência dos glóbulos vermelhos circulantes e a medula óssea não é capaz de compensação, mesmo aumentando sua produção, o que causa anemia desde os primeiros anos da vida da pessoa. Dentre as doenças deste grupo, a Anemia Falciforme (SCA) e a Esferocitose Hereditária (HS) destacam-se por se tratarem de enfermidades com defeitos genéticos intrínsecos das células vermelhas (RBCs) que geram complicações multissistêmicas agudas e crônicas em seus portadores. Por vias patofisiológicas distintas, reticulócitos e respectivas hemácias defeituosas de tais doenças, falciformes e esferócitos, são continuamente aprisionados e fagocitados no baço, importante órgão de destruição de células velhas e/ou defeituosas via hemólise extravascular, o que leva progressivamente à disfunção e eventual perda da função esplênica. O objetivo desse trabalho é avaliar o papel do baço em relação à habilidade e ao fenótipo adesivos de reticulócitos (ret) e eritrócitos (erit) em pacientes com SCA e HS, com e sem função esplênica preservada. Amostras de sangue de 37 pacientes (22 SCA and 15 HS) com função esplênica e 19 pacientes (13 SCA e 6 HS) sem ela foram avaliadas. Ainda, sangue de 22 crianças com SCA foi coletado em estudo longitudinal dos 6 e 29 meses de vida. Todas as amostras de sangue foram analisadas quanto à função esplênica (Contagem de células PIT e de corpúsculos de Howell-Jolly - HJB), quanto ao perfil imunofenotípico celular (em % e em média de intensidade de fluorescência - MFI) e quanto à habilidade de adesão das células vermelhas à laminina e à linhagem celular endotelial HMEC-1. A análise da transição da perda de função esplênica demonstrou que a mesma se intensificou a partir dos 3 anos de idade (PIT: r=0,8; p<0,0001; HJB: r=0,7; p<0.0001). Quanto à imunofenotipagem celular, a contagem PIT se correlacionou positivamente, principalmente com os marcadores CD147 (%ret: r=0,6; p<0,0001; MFIret: r=0,6; p<0,0001; %erit: r=0,7; p<0,0001; MFIerit: r=0,6; p<0,0001), LuBCAM (%ret: r=0,5; p=0,004; MFIret: r=0,6; p<0,0001; %erit: r=0,6; p<0,0003; MFIerit: r=0,4; p<0,004) and CD58 (%ret: r=0,4; p=0,006; MFIret: r=0,5; p<0,0013; %erit: r=0,4; p<0,009; MFIerit: r=0,6; p<0,0001). Na comparação entre ausência ou presença do baço, a perda de sua função exerceu influência no aumento da expressão de adesão de RBCs em SCA, principalmente CD147 (%ret: p=0,002; MFIret: p=0,003; %erit: p<0,0001; MFIerit: p=0,005), LuBCAM (%ret: p=0,0001; MFIret: p<0,0001; %erit: p<0,0001; MFIerit: p<0,0001) e CD58 (%ret: p=0,007; MFIret: p=0,006; %erit: p=0,003; MFIerit: p=0,0004), embora a adesão celular tenha diminuído em pacientes HS esplenectomizados. Na comparação entre as doenças, pacientes HS com o baço apresentaram maior freqüência de adesão celular em relação aos SCA, notavelmente em relação ao LuBCAM (%ret: p=0,0008; MFIret: p=0,03; %erit: p<0,0001; MFIerit: p=0,0002), CD58 (%ret: p=0,0009; %erit: p=0,003) e CD44 (%ret: p=0,009; %erit: p<0,003). No entanto, as amostras SCA sem função esplênica tiveram maior expressão de adesão celular para CD147 (%ret: p=0,006; MFIret: p=0,02; %erit: p=0,02), LuBCAM (%ret: p=0,004; MFIret: p<0,0001), CD36 (%ret: p=0,0002; MFIret: p=0,01), CD242 (%ret: p=0,0008; %erit: p=0,05) e CD49d (%ret: p=0,04). Em relação ao Ensaio de Adesão in vitro, na ausência de baço, os RBCs SCA apresentaram maior adesividade à laminina do que os RBCs SCA com função esplênica preservadaem todas as taxas de fluxo de tensão de cisalhamento empregadas (0,5 dyne/cm2: p=0,01; 1 dyne/cm2: p=0,02; 2 dynes/cm2: p=0,03; 3 dynes/cm2: p=0,03; 5 dynes/cm2: p=0,04 e 7 dynes/cm2: p=0,03). Especialmente, reticulócitos de pacientes sem baço apresentaram maior adesividade à HMEC-1 em baixas tensões de cisalhamento (1 dyne/cm2) em ambas as doenças (SCA: p=0,03; HS: p=0,03). Por fim, reticulócitos apresentaram maior habilidade adesiva à células endoteliais em indivíduos SCA do que em pacientes HS, com (0,5 dyne/cm2: p=0,04; 1 dyne/cm2: p=0,03) ou sem baço (0,5 dyne/cm2: p=0,02; 2 dynes/cm2: p=0,01; 3 dynes/cm2: p=0,03; 5 dynes/cm2: p=0,02 e 7 dynes/cm2: p=0,03). Nossos resultados indicam que embora pertençam ao grupo de Anemias Hemolíticas, as patofisiologias e evoluções clínicas distintas de SCA e de HS levam a padrões imunofenotípicos diferentes de expressão da adesão celular. Na SCA, a ausência de função esplênica teria direta relação com o aumento do fenótipo pró-adesivo e com a adesividade de RBCs SCA, o que traz sérias consequências clínicas aos pacientes, enquanto na HS sem baço, de maneira geral, os eventos de adesão celular são minimizados, embora ainda apresentem reticulócitos e eritrócitos adesivos circulantes após a esplenectomia. / Hemolytic Anemias comprise a group of diseases in which there is marked reduction in the survival of circulating erythrocytes and the bone marrow is not capable of compensation, even by increasing its production, which causes anemia from the first years of the person\'s life on. Among the diseases of this group, Sickle Cell Anemia (SCA) and Hereditary Spherocytosis (HS) stand out for being diseases with intrinsic genetic defects of red blood cells (RBCs) that generate acute and chronic multisystemic complications in their patients. By distinct pathophysiological pathways, reticulocytes and these disease\'s respective defective erythrocytes, sickle and spheroid ones, are continuously trapped and phagocytosed in the spleen, important organ of destruction of old and/or defective cells via extravascular hemolysis, which progressively leads to dysfunction and eventual loss of splenic function. The objective of this study was to evaluate the role of the spleen in relation to the reticulocyte (ret) and erythrocyte (eryt) adhesive ability and adhesion phenotype in patients with SCA and HS, with and without preserved splenic function. Blood samples from 37 patients (22 SCA and 15 HS) with splenic function and 19 patients (13 SCA and 6 HS) without it were evaluated. Still, blood from 22 children with SCA was collected in a longitudinal study from 6 to 29 months of age. All blood samples were analyzed for splenic function [pitted cells (PIT) and Howell-Jolly bodies (HJB) counting], for the cellular immunophenotypic profile (in % and in mean fluorescence intensity - MFI) and for the adhesive ability of RBCs to laminin and to endothelial cell line HMEC-1. Analysis of the splenic function loss transition showed that it intensified from 3 years of age on (PIT: r=0.8, p<0.0001; HJB: r=0.7, p<0.0001). Regarding the cellular immunophenotyping, PIT count correlated positively, mainly with CD147 markers (%ret: r=0.6, p<0.0001; MFIret: r=0.6, p<0.0001; %eryt: r=0.7, p<0.0001; MFIeryt: r=0.6, p<0.0001), LuBCAM (%ret: r=0.5, p=0.004; MFIret: r=0.6, p<0.0001; %eryt: r=0.6, p<0.0003; MFIeryt: r=0.4, p<0.004) and CD58 (%ret: r=0.4, p=0.006; MFIret: r=0.5, p<0.0013; %eryt: r=0.4, p<0.009; MFIeryt: r=0.6, p<0.0001). In the comparison between spleen absence or presence, the loss of its function exerted influence on the increase of RBCs adhesion expression in SCA, mainly on CD147 (%ret: p=0.002; MFIret: p=0.003; %eryt: p<0.0001; MFIeryt: p=0.005), LuBCAM (%ret: p=0.0001; MFIret: p<0.0001; %eryt: p<0.0001; MFIeryt: p<0.0001) e CD58 (%ret: p=0.007; MFIret: p=0.006; %eryt: p=0.003; MFIeryt: p=0.0004), although cell adhesion has been decreased in splenectomized HS patients. In the comparison between diseases, HS patients with spleen showed higher cell adhesion frequency compared to SCA, notably in relation to LuBCAM (%ret: p=0.0008; MFIret: p=0.03; %eryt: p<0.0001; MFIeryt: p=0.0002), CD58 (%ret: p=0.0009; %eryt: p=0.003) and CD44 (%ret: p=0.009; %eryt: p<0.003). However, SCA samples without splenic function had higher cell adhesion expression for CD147 (%ret: p=0.006; MFIret: p=0.02; %eryt: p=0.02), LuBCAM (%ret: p=0.004; MFIret: p<0.0001), CD36 (%ret: p=0.0002; MFIret: p=0.01), CD242 (%ret: p=0.0008; %eryt: p=0.05) and CD49d (%ret: p=0.04). Concerning the in vitro Adhesion Assay, in the spleen absence, SCA RBCs showed greater adhesiveness to laminin than SCA RBCs with preserved splenic function did at all shear stress flow rates applied (0.5 dyne/cm2: p=0.01, 1 dyne/cm2: p=0.02, 2 dynes/cm2: p=0.03, 3 dynes/cm2: p=0.03, 5 dynes/cm2: p=0.04 and 7 dynes/cm2:p=0.03). Especially, reticulocytes from patients without spleen showed higher adhesiveness to HMEC-1 at low shear stresses (1 dyne/cm2) in both diseases (SCA: p=0.03; HS: p=0.03). Finally, reticulocytes showed greater adhesion ability to endothelial cells in SCA subjects than in HS patients, with (0.5 dyne/cm2: p=0.04 and 1 dyne/cm2: p=0.03) or without spleen (0.5 dyne/cm2: p=0.02, 2 dynes/cm2: p=0.01, 3 dynes/cm2: p=0.03, 5 dynes/cm2: p=0.02 and 7 dynes/cm2: p=0.03). Our results indicate that although both diseases belong to the Hemolytic Anemias group, SCA and HS distinct pathophysiologies and clinical evolution lead to different immunophenotypic patterns of cell adhesion expression. In SCA, the absence of splenic function may have a direct relation with the increase of SCA RBCs proadhesive phenotype and adhesiveness, which brings serious clinical consequences to the patients, whereas in HS without spleen, in general, cellular adhesion events are minimized, although they still present adhesive circulating reticulocytes and erythrocytes after splenectomy.

traitement empirique couramment recommandé en médecine aviaire lors
d’hémorragie ou d’anémie. L’objectif principal de cette étude était d’évaluer
les effets de ce traitement sur l’anémie chez les oiseaux de proie. Deux types
d’individus ont été utilisés : des crécerelles d’Amérique (Falco sparverius) où
une anémie par perte de sang externe aiguë a été créée (deux phlébotomies
de 20-40 % du volume sanguin total à un intervalle de 6 h) et des oiseaux de
proie sauvages de différentes espèces souffrant d’anémies diverses.
L’ensemble des oiseaux a été subdivisé aléatoirement en groupe traitement
(fer dextran 10 mg/kg IM) et contrôle (NaCl 0,9% IM). Un suivi dans le temps
a été réalisé afin d’étudier leur récupération de l’anémie, la présence d’effets
secondaires au traitement et l’impact d’une administration de fer sur ces
réserves. Aucune différence significative n’a été observée entre les deux
groupes en ce qui concerne les signes cliniques, l’hématocrite, le
pourcentage des polychromatophiles/réticulocytes, la densité cellulaire et le
fer de la moelle osseuse, la créatine kinase et le fer plasmatique. La majorité
des crécerelles ont présenté une myosite au site d’injection du fer. Nos
résultats suggèrent qu’une administration de 10 mg/kg de fer dextran IM n’a
pas d’effet sur l’érythropoïèse des rapaces souffrant d’anémie par perte de
sang externe aiguë, qu’elle provoque une légère inflammation au site
d’injection et qu’elle n’influence pas les réserves de fer. Le comptage des
réticulocytes en anneau et des polychromatophiles semble être deux

volume at 6 hours interval) and other various species of wild birds of prey
suffering from different types of anemia. All subjects were randomized into a
treatment (iron dextran 10 mg/kg IM) or a control (NaCl 0,9 % IM) group.
Monitoring was carried out to evaluate the evolution of the anemia, presence
of side effects and impact of an iron administration on their iron reserve. No
significant differences were observed between the two treatment groups for
clinical signs, packed cell volume, the percentage of
reticulocytes/polychromatophilic erythrocytes, bone marrow cellularity and
iron, plasmatic iron and creatine kinase. Most kestrels had a myositis at the
iron injection site. Our results suggest that an IM injection of 10 mg/kg iron
dextran has no effect on raptor erythropoiesis after an acute external blood
loss anemia, that it has no effect on iron reserve, and that it can cause mild
inflammation at the injection site. The polychromatophilic erythrocytes and the
