Estimating and Modeling Transpiration of a Mountain Meadow Encroached by Conifers Using Sap Flow Measurements

Mountain meadows in the western USA are experiencing increased rates of conifer encroachment due to climate change and land management practices. Past research has focused on conifer removal as a meadow restoration strategy, but there has been limited work on conifer transpiration in a pre-restoration state. Meadow restoration by conifer removal has the primary goal of recovering sufficient growing season soil moisture necessary for endemic, herbaceous meadow vegetation. Therefore, conifer water use represents an important hydrologic output toward evaluating the efficacy of this active management approach. This study quantified and evaluated transpiration of encroached conifers in a mountain meadow using sap flow prior to restoration by tree removal. We report results of lodgepole pine transpiration estimates for an approximate 1-year period and an evaluation of key environmental variables influencing water use during a dry growing season. The study was conducted at Rock Creek Meadow (RCM) in the southern Cascade Range near Chester, CA, USA. Sap flow data were collected in a sample of lodgepole pine and scaled on a per-plot basis to the larger meadow using tree survey data within a stratified random sampling design (simple scaling). These estimates were compared to a MODIS evapotranspiration (ET) estimate for the meadow. The 1-year period for transpiration estimates overlapped each of the 2019 and 2020 growing seasons partially. The response of lodgepole pine transpiration to solar radiation, air temperature, vapor pressure deficit, and volumetric soil water content was investigated by calibrating a modified Jarvis-Stewart (MJS) model to hourly sap flow data collected during the 2020 growing season, which experienced below average antecedent winter precipitation. The model was validated using spatially different sap flow data in the meadow from the 2021 growing season, also part of a dry year. Calibration and validation were completed using a MCMC approach via the DREAM(ZS) algorithm and a generalized likelihood (GL) function, enabling model parameter and total uncertainty assessment. We also used the model to inform transpiration scaling for the calibration period in select plots in the meadow, which allowed comparison with simple scaling transpiration estimates. Average total lodgepole pine transpiration at RCM was estimated between 220.57 ± 25.28 and 393.39 ± 45.65 mm for the entire campaign (mid-July 2019 to mid-August 2020) and between 100.22 ± 11.49 and 178.75 ± 20.74 mm for the 2020 partial growing season (April to mid-August). The magnitude and seasonal timing were similar to MODIS ET. The model showed good agreement between observed and predicted sap velocity for the 2020 partial growing season (RMSE = 1.25 cm h-1), with meteorological variables modulating early growing season sap flow and volumetric soil water content decline imposing transpiration decrease in the late growing season. The model validation performed similarly to calibration in terms of performance metrics and the influence of meteorological variables. The consistency of the declining volumetric soil water content effect during the late growing season between periods could not be evaluated due to an abridged validation period. Overall, the implementation GL-DREAM(ZS) showed promise for future use in MJS models. Lastly, the model derived transpiration estimates for the 2020 partial growing season showed some of the potential utility in using the MJS model to scale sap flow at the study locale. It also highlights some of the key limitations of this approach as it is executed in the present study.

Phylogeny and Visual System Evolution in Sap Beetles (Coleoptera: Cucujoidea: Nitidulidae)

Coleoptera is currently the most diverse lineage of life on Earth with more than 400,000 described species. The group also contains unbelievable ecological and morphological variation that has been evolving since the early Carboniferous (>350 MYA). The group provides the perfect playground to test evolutionary hypotheses with large-scale phylogenies. First, a newly developed bioinformatics tool is presented that allows for the critical assessment of the stability of phylogenetic nodes with even minor changes in taxon sampling (i.e. a single taxon jackknifed). The tool, TANOS, is capable of performing tests on large genomics-scale datasets. The case study included is a widely used phylogenomics effort to resolve the higher level relationships of all Insecta. The specific beetle group chosen for the remaining products is the superfamily Cucujoidea, and more specifically the family Nitidulidae, or sap beetles. Several taxonomic and nomenclatural projects were necessary to provide a more stable classification and begin the process of establishing the group as a model system for questions concerning feeding behavior evolution. Several new species of the genus Carpophilus were described from both the West Indies and the New World tropics and subtropics. Several type designations were also needed for many species in the subfamily Carpophilinae. In addition, the fossil diversity of Nitidulidae was reviewed with the addition of the oldest known member of the Cillaeinae from amber (Dominican). With the reviewed fossil fauna, as well as many more from across the Cucujoidea more broadly, a divergence-time estimated phylogeny was generated for >250 species of the superfamily. The phylogeny, based on the combination of multiple published Sanger datasets and supplemented with newly generated data for a further 50 species, was used to date multiple feeding shifts in the group. Specifically, the clade ages were used to compare with published clade ages for the corresponding food resource each clade is known to exploit. A significant relationship was recovered between age of the beetle clade and the corresponding food resource clade. Interestingly, it was found that clades that shifted to a food resource first, in the absence of existing beetle competitors, were significantly more diverse over subsequent lineages that shifted to the same resource. A feeding ecology of specific importance, at least to many groups of nitidulid, is anthophily, or flower-visiting. An extreme variation in eye size is observable across the family and so multiple factors including; sex, day-night activity, and feeding behavior were tested in a phylogenetic context. No sexual dimorphism nor variation consistent with activity period was found. Relative eye size of flower visiting species was significantly larger than that of all other feeding behaviors in sap beetles. This correlation was also tested in a phylogenetic context, with the increase in morphological investment in vision repeatedly shown to correspond to shifts in flower associated behavior. In an attempt to investigate the interplay between behavior, morphology, and genetics in the same process, opsin data was captured as part of a large targeted enrichment sequencing effort (AHE) across Nitidulidae and their relatives. The first phylogenomic estimate was generated for the group, with a taxon sampling of 192, and data sampling depth of 703 loci. A newly updated pipeline was developed for AHE data and is presented herein. Topological results support the recent idea of a non-monophyletic Cucujoidea. Results also demonstrate multiple classification issues within the Nitidulidae with several new subfamilies/tribes necessary to maintain diagnosable monophyletic groups in the family. The vast majority of sampled genera were supported, with the exception of Brachypeplus and Pallodes. Preliminary opsin diversity across the group is much greater than previously estimated. Opsin copy number is potentially plastic even within individual genera, although consistently duplications are associated with taxa that visit flowers. It is clear that the complexity of cucujoid visual systems increases with anthophilous behavior.

In the course of the study, an analysis was made of the most well-known information systems on the market, with the help of which it is possible to optimize the sales process, as well as the processes of pre-sales operations. The implemented systems for interaction with the company's customers from the point of view of sales analyzed the weaknesses of the company and provided solutions by optimizing processes.

Development of intermonoploid somatic hybrids of potato and their molecular analysis based on polymorphism for retroelement Tst1

Inbred lines for hybrid crop production have been a mainstay of plant breeding. Biotechnological approaches to hasten the process are available including anther culture to halve the genome and protoplast fusion to create hybrids between incompatible partners. We applied these techniques to potato to evaluate their potential for breeding highly heterozygous, cross-pollinating species. Four families of monoploids (2n=1x=12), developed from diploid hybrids with diverse genomic constitutions but heavily favoring Solanum phureja, a primitive cultivated potato, were used in electrofusion experiments to create intermonoploid somatic hybrids (SH). The "monoploid sieve" results in the survival of only those gametes free of lethal and deleterious genes but generates sterile sporophytes, necessitating protoplast fusion for SH development. From six intermonoploid electrofusion combinations, 276 plants were regenerated over 6-9 months. Fusion conditions were optimized. Ploidy was determined by flow-cytometry and SH confirmed by microsatellite analysis. Field evaluations over three years revealed that intermonoploid SH were inferior to cultivars. Dihaploids derived by anther culture of a tetraploid intermonoploid SH were reduced in vigor with an increase in homozygosity, while 2x X 2x sexually derived populations had better yield than the SH, suggesting that producing SH introduced or eliminated factors required for productivity. Molecular analysis of the SH was conducted to examine genomic stability through protoplast isolation and plant regeneration. Sequence specific amplified polymorphism (S-SAP) represents a hybrid system incorporating amplified fragment length polymorphism (AFLP) technology in conjunction with the use of a defined genomic sequence, e.g., retrotransposon display (RD) when the defined sequence is anchored into a consensus sequence of a retrotransposon such as the long terminal repeat (LTR) sequence of Tst1. Parental monoploids, SH and various Solanaceae were evaluated by RD. Fluorescently-labeled retrotransposon-based primers were used in the ALFexpress automated fragment analyzer system. Eleven probes from RD were created for Southern blot analysis and used to verify taxonomic relationships between selected Solanaceae. Blots of intermonoploid somatic hybrids confirmed hybridity and occasional loss of genomic fragments. No activation or replication of retrotransposons was detected. Sequencing of inter-retrotransposon amplified polymorphism (IRAP) and S-SAP fragments revealed that all fragments had the expected Tst1 retroelement and/or the AFLP adaptor sequence. BLAST analysis identified 4 of the 17 fragments sequenced as part of the chloroplast genome, a tobacco anther-specific gene, repetitive DNA, and the phytochrome F gene. / Ph. D.

Evidence for a learning effect in short-wavelength automated perimetry.

No / Purpose To document the magnitude of any learning effect for short-wavelength automated perimetry (SWAP) in patients with either ocular hypertension (OHT) or open-angle glaucoma (OAG) who are experienced in standard automated perimetry (SAP). Participants Thirty-five patients (22 with OHT and 13 with OAG) who had previously undergone at least 3 threshold SAP visual field examinations with the Humphrey Field Analyzer (HFA; Carl Zeiss Meditech Inc., Dublin, CA), and 9 patients with OHT who had not previously undertaken any form of perimetry. Methods Each patient attended for SWAP on 5 occasions, each separated by 1 week. At each visit, both eyes were examined using Program 24-2 of the HFA; the right eye was always examined before the left eye. Main Outcome Measures (1) Change over the 5 examinations, in each eye, of the visual field indices Mean Deviation (MD), Short-term Fluctuation (SF), Pattern Standard Deviation (PSD), and Corrected Pattern Standard Deviation. (2) Change in each eye between Visits 1 and 5 in proportionate Mean Sensitivity (pMS) for the central annulus of stimulus locations compared with that for the peripheral annulus thereby determining the influence of stimulus eccentricity on any alteration in sensitivity. (3) Change between Visits 1 and 5 in the number and magnitude of the Pattern Deviation (PD) probability levels associated with any alteration in sensitivity. Results The MD, SF, and PSD each improved over the 5 examinations (each at P<0.001). The improvement in pMS between Visits 1 and 5 was greater for the peripheral annulus than for the central annulus by approximately twofold for the patients with OAG. Considerable variation was present between patients, within and between groups, in the number of locations exhibiting an improving sensitivity between Visits 1 and 5 by 1 or more PD probability levels. Conclusions Care should be taken to ensure that, during the initial examinations, apparent field loss with SWAP in patients exhibiting a normal field by SAP is not the result of inexperience in SWAP. Apparently deeper or wider field loss in the initial examinations with SWAP compared with that exhibited by SAP in OAG also may arise from inexperience in SWAP.

De största hindren är begränsade resurser och brist på teknisk expertis, och studien betonar behovet av att balansera strategisk planering med snabb anpassning och extern expertis.

A firm’s information requirements can only be understood by examining their Information System (IS) through Organizational, People, Technical and Strategic perspective.  Current study focuses on the value of Information System implementation for Swedish Wind Power Industry (SWPI) that has reached to more than one billion dollars in recent decade. A well-known integrated IS i.e. Enterprise Resource Planning (ERP) used by Swedish Wind Power Industry (SWPI) for decision making activities is not without its challenges, that consumes a significant portion of their capital and resources for implementation. Exploratory and explanatory research conducted for triangulation, through quantitative and qualitative methods, with empirical data of survey, multiple case studies, interviews and extensive literature review, to highlight the problems and solutions for implementing ERP in SWPI. Several, IS are highlighted with their functionalities and critical success and failure factors. Cette thèse analyse les formes et les effets de l’utopisme qui sous-tend le projet politique du SAP (parti social-démocrate suédois) entre 1932 et 1986. Dans le cadre d’une approche conceptuelle du politique nous nous interrogeons sur le pouvoir des idées, des symboles et de l’imaginaire dans la constitution d’un roman national original, mais aussi à travers la mise en oeuvre de l’action publique. A partir d’un important corpus inédit de discours et d’écrits rédigés par les trois leaders qui se sont succédés à la tête du gouvernement pendant un demi-siècle(Hansson, Erlander, Palme), la thèse s’efforce de mettre à jour « la cristallisation sémantique » qui a structuré le lexique politique du SAP. En son coeur figure la métaphore du Folkhem qui fonctionne comme un mythe mobilisateur pour la nouvelle société, avant de se transformer en élément essentiel de l’identité nationale et de la culture politique de la Suède. Autour du Folkhem, se noue le contrat social suédois, et la notion se transforme en source etressource de légitimité politique. L’histoire des récits du Folkhem permet de mieuxcomprendre les origines du modèle suédois et les modalités récentes de son évolution. Par son extrême plasticité, le Folkhem continue de nos jours à faire l’objet d’appropriations politiques nombreuses et diverses, confirmant ainsi son rôle fondateur d’institution de sens. / This thesis relates the forms and effects of the utopianism that underlies the SAP (Swedish Social-Democratic Party) political project between 1932 and 1986. The power of ideas,symbols and imaginary in politics is our theoretical base. The political discourse of the SAP leaders and the importance of words in the creation of public policy are studied. Our corpus consists of a vast archive material. Guided by utopianism, the leaders express their will to create a new society. The Folkhem political metaphor, narrative at the heart of the political glossary of the SAP, is in the centre of this utopianism. The notion is used as a mobilizingmyth for the new society and it works as a social contract for the Swedes. The Folkhem narratives help us understand the Swedish model and contemporary Sweden. By its extreme plasticity and by being part of the Swedish identity, the notion is still a source of political legitimacy and continues to produce meaning.

This subject of interest in this paper is the ideas expressed and formulated in the making andinauguration of the Branting-monument at Norra Bantorget in Stockholm. The initiative for a monument honoring the late Social Democratic leader Hjalmar Brantingwas taken at the Swedish Social Democratic Party Congress in 1928. In 1935 the party board decided to give the task of designing the monument to the much renowned artist Carl Eldh. A national fund-raising campaign for the monument was also initiated. In1942 the model of the monument was completed, but because of the war it was stored in a shelter. On June 2, 1952 the monument was inaugurated with great ceremony. Old working movement songs and anthems were played and sung and speeches were made by the Prime Minister Tage Erlander, Stockholm City Council President Carl Albert Andersson, and three foreign guests: Salomon Grumbach, Camille Huysmans and Paul LöbeIn the Social Democratic daily press reporting on the inauguration and in the speeches from the inauguration one can track several important recurring themes. It is clear that they were eager to define Branting as a prudent reformist and anti-communist. It also points out that both Branting was a big Swede that the whole nation stood behind, but also a great European and internationalist. But it also adds time to let different people describe Branting on a personal level, thus both strengthening the heroic image of Branting and making the image of Branting more personal. Even liberal newspapers adopted much of this rhetoric. The only newspaper that in the true sense took a fairly critical stance in relationship to the inauguration was the communist Ny Dag.
