An evaluation of the homoeopathic drug proving of Naja Mossambica in the light of a doctrine of signatures analysis and a comparison between the proving symptons and the venom toxicology

These symptoms were compared to the toxicology of Naja mossambica venom as well as a doctrine of signatures analysis of the snake in order to expand and clarify the remedy picture. Existing knowledge of the venom toxicology gives a clear indication of the organs and body systems that the substance has an affinity for. Many poisonous substances used homoeopathically rely heavily on inferences made from the toxicology of the substance, as much of the gross pathology in the symptom picture cannot safely be elucidated in a proving. The aim of this study was to determine the sphere of action of Naja mossambica by utilising symptoms obtained from the proving and from the toxicology of the venom. This was done in order to determine the remedy's usefulness in a homoeopathic clinical setting by expanding our understanding of the substance and thereby facilitating the treatment of disease based on the law of similars.

A homoeopathic drug proving of Bitis atropos with a subsequent comparison to venom toxicology and related remedies

Submitted in partial compliance with the requirements of the Master’s Degree in Technology: Homoeopathy, Durban University of Technology. 2013. / This study was a homoeopathic drug proving of Bitis atropos 30CH (derived from Berg adder venom) with a subsequent comparison of the proving symptoms to known venom toxicology and existing remedies from the materia medica, that on repertorisation, yielded the greatest similarities in the Mental, General, Physical and unique symptomatology of Bitis atropos. Methodology : The proving was carried out in the form of a double-blinded, placebo controlled trial on healthy subjects who were administered the proving substance or placebo. The resultant influence of this substance on the health of provers (i.e. symptoms produced) was recorded in journal format and formed the materia medica and ultimately the clinical indications thereof according to the Law of Similars. Twenty eight healthy consenting provers who meet the inclusion criteria (Appendix B), were randomly split into two groups, one being the experimental group comprising 22 provers, and the other a placebo control group comprising 6 provers. This was further split between the researcher and co-researcher, each responsible for 11 provers receiving verum and 3 receiving placebo. The researchers and the individual provers were unaware of their respective group allocation and the provers were unaware of the identity of the proving substance. The fresh venom sourced from a wild, Berg adder, was processed according to the German Homoeopathic Pharmacopoeia (Appendix G) to produce the 30CH Homoeopathic potency thereof. Six lactose powders were dispensed to each prover (either placebo or verum) and taken sublingually three times a day or until the onset of symptoms. Symptoms were recorded by the provers in journals over 4 weeks and were closely supervised by the researcher. When the symptoms subsided, the combined journals were collected, collated, analysed, interpreted and validated. Accepted symptoms were converted to materia medica and Repertory format. Results : The proving yielded a total of 903 rubrics, of which 18 were newly created. The systems mostly affected were Dreams, Mind, Head and Eye. Comparison of proving symptoms to that of venom toxicology, as seen in case studies of envenomation by Bitis atropos, yielded similar results, as the sensations experienced in provers closely matched that of known venom toxicology. On repertorisation of the proving symptoms, the existing remedies that were closely related were Sepia officinalis, Lachesis mutus and Argentum nitricum. Further repertorisation of toxicological symptoms indicated a further relation to Belladonna, Natrum muriaticum and Hyoscyamus niger. Conclusion : Clearly observable signs and symptoms were produced by healthy provers in response to administration of Bitis atropos 30CH, in addition there was a significant degree of similarity between proving symptoms and that of known toxicology of the crude substance. The researcher identified Sepia officinalis, Lachesis mutis and Argentum nitricum as the three most similar existing homoeopathic remedies and a detailed comparison thereof was conducted. A further repertorisation of the toxicological symptoms of envenomation by the snake, yielded the remedies Belladonna, Natrum muriaticum and Hyoscyamus niger which were also compared to Bitis atropos.

The Squamata (Reptilia) of the Orange Free State, South Africa.

This study deals with the three orders of Squamata (Sauria, Amphisbaenia, Serpentes) of the Orange Free State, South Africa. Following an intensive systematic survey 4 492 specimens have been examined comprising 77 different species and subspecies of which 25% represent new records for the study area. Variation in taxonomically important characters as well as informa- tion on colour, size, habitat, breeding, predators and diet are discussed. Certain taxonomic changes have been made. Distribution maps have been compiled on the basis of eighth-degree-units. Distributional patterns have been analyzed to establish areas of relative homogeneity separated by transitional zones. A causal analysis of the main distribution patterns of the O.F.S. Squamata indicated that rainfall is the major environmental factor influencing distribution. Rozmnožovací strategie vybraných druhů hroznýšovitých hadů (Boidae) / Reproductive strategies in boid snakes

The important part of life history of species is number and size of offspring, which is limited by size of maternal investment. Family of boas (Boidae) comprises species with various maternal investments. The species of study was Cuban boa (Chilabothrus angulifer) having small litter size, but producing one of the biggest neonates from family Boidae. The aim of the thesis was to analyze relationships between life-history parameters and try to find possible evolutionary causes of this huge maternal investment. Results show, that the female's decision to reproduce is under "Capital breeder" strategy, when size of the female positively influence litter size and offspring size. Surprising result is that smaller (younger) females are producing more sons than daughters in accordance with "Local resource competition" hypothesis. Fitness of the young should increase with birth size. That was confirmed only in interspecific survival rate being very high in Cuban boa. Phylogenetic Relationships and Evolution of Snakes

Snakes represent an impressive evolutionary radiation of over 3,500 widely-distributed species, categorized into 515 genera, encompassing a diverse range of morphologies and ecologies. This diversity is likely attributable to their distinctive morphology, which has allowed them to populate a wide range of habitat types within most major ecosystems. In my first chapter, I provide the largest-yet estimate of the snake tree of life using maximum likelihood on a supermatrix of 1745 taxa (1652 snake species + 7 outgroup taxa) and 9,523 base pairs from 10 loci (5 nuclear, 5 mitochondrial), including previously unsequenced genera (2) and species (61). I then use this phylogeny to test hypotheses regarding heterogeneity in diversification rates and how this shaped overall patterns of snake diversity in Chapter 2. I also used the species-level phylogeny to test the evolution of habitat use in snakes, morphological variation, and whether distantly-related species exhibit morphological convergence in Chapter 3. Finally, in Chapter 4 I investigate how prehensile tails effect striking performance in arboreal snakes.

Estudos sobre eventos reprodutivos masculinos normalmente, não incluem qualquer análise microscópica, estando sujeitos a conclusões equivocadas. A histologia pode ser utilizada para caracterizar a condição testicular e ciclo espermatogênico em serpentes. Portanto, o uso dela é indispensável para o estudo do ciclo reprodutivo em serpentes. Este trabalho teve como objetivo realizar uma análise da morfologia do trato reprodutivo da serpente Tomodon dorsatus nas diferentes estações do ano. Para tanto, foram utilizados 50 espécimes de Tomodon dorsatus provenientes da coleção hepetológica do Instituto Butantan e 6 indíviduos vivos que foram eutanasiados posteriormente. O testículo, rim e ducto deferente foram mensurados e em seguida coletados fragmentos destinados para análise histológica. A largura do túbulo seminífero foi maior no outono (p=0,001); o epitélio obteve maior altura no verão (p=0,001). Os testículos estavam ativos ao longo do ano, porém, a espermiogênese obteve um pico no verão; no outono, a maioria dos indivíduos estava em espermiogênese ou iniciando a regressão; no inverno, alguns indivíduos estavam em recrudescência ou em regressão. Espermatozoides foram encontrados no lumem dos ductos deferentes em todas as estações do ano. Studies on reproductive cycle in the male do not included histological analysis, which can lead a erroneous conclusions. The histology is used to characterized the testicular conditions and spermatogenesis in snakes. Thus, the histology is pivotal to study the reproductive cycle in snakes. The aim of this study was analysed the morphology of the male reproductive tract in the snake Tomodon dorsatus in the different seasons of the year. For this purpose, 50 specimens of Tomodon dorsatus from the hepetological collection of the Butantan Institute and 6 living individuals were used and later euthanized. The testicular, kidney and vas deferens were measured and then fragments were collected for histological analysis. The width of the seminiferous tubule was greater in the autumn (p = 0.001); the epithelium had higher height in the summer (p = 0.001). The testicles were active throughout the year, however, the spermiogenesis peaked in the summer; in the fall, most individuals were either in espemiogenesis or initiating regression; in winter, some individuals are in recrudescence or regression. Orientador: Hussam El Dine Zaher / Resumo: Apostolepis Apostolepis é o gênero mais diversificado da Tribo Elapomorphini, composto por 40 espécies válidas, sendo 36 brasileiras e quatro que distribuem-se nas Guianas, Colômbia, Peru, Bolívia, Paraguai e Argentina. Apostolepis é reconhecido como um dos grupos taxonomicamente mais problemáticos de Dipsadidae, principalmente por causa de descrições de espécies com informações insuficientes, por vezes baseadas em variações intraespecíficas e em indivíduos anômalos. Mais de 50 táxons nominais são relacionados a Apostolepis, dos quais 21 foram descritos nos últimos 30 anos e 35 baseados apenas no holótipo; vários já sinonimizados a espécies descritas anteriormente. Por causa dos hábitos de vida as espécies do gênero não são abundantemente encontradas, fazendo com que muitas espécies ainda sejam minimamente conhecidas e outras conhecidas apenas pelo holótipo. Combinando todos esses fatores, foram gerados muitos equívocos na identificação das espécies, contribuindo para uma sucessão massiva de controvérsias taxonômicas e resultando no insatisfatório entendimento da evolução, ecologia e história natural das espécies do grupo. It is composed of 40 valid species, being 36 from Brazil and others distributed in the Guianas, Colombia, Peru, Bolivia, Paraguay and Argentina. The genus Apostolepis is recognized as one of the most problematically taxonomic groups of Dipsadidae because original species descriptions with insufficient information, sometimes based on intraspecific variations and anomalous specimens. Caracterização bioquímica, estrutural e funcional de uma L-aminoácido oxidase isolada de peçonha de Lachesis muta (Serpentes, Viperidae) / Purification, biochemistry and functional characterization of a new L-amino acid oxidase from Lachesis muta venom.

As peçonhas de serpentes contêm uma mistura complexa de substâncias farmacologicamente ativas, como metaloproteases, fosfolipases A2, serino-proteases, L-aminoácido oxidase (LAAO), além de outros importantes compostos sem ação enzimática. LAAOs são flavoproteínas que catalisam a desaminação oxidativa de L-aminoácidos e produzem o -cetoácido correspondente, com a concomitante liberação de amônia e peróxido de hidrogênio. A peçonha de Lachesis muta (L. muta) contém L-aminoácido oxidase, a qual pode contribuir com o envenenamento. Portanto, o objetivo deste trabalho é a purificação da L-amino acido oxidase de peçonha de Lachesis muta (LmLAAO) e a sua caracterização bioquímica, estrutural e funcional. Para isso, foram desenvolvidos dois protocolos distintos de purificação, os quais forneceram LmLAAO com grande pureza. No primeiro protocolo, 20 mg de peçonha bruta de L. muta foram submetidos a uma gel filtração em Sephacryl S100®. Das dez frações obtidas, a primeira fração apresentou atividade L-aminoácido oxidase e foi submetida a mais um passo cromatográfico em Mono Q®. A homogeneidade da fração com atividade L-aminoácido oxidase após a troca iônica foi comprovada por presença de banda única com 60,2 kDa em SDS-PAGE. O segundo protocolo de purificação foi uma sequência de três passos cromatográficos, na qual 200 mg de peçonha bruta de L. muta foram submetidos a gel filtração em Sephacryl S200®, seguido por interação hidrofóbica em Phenyl Sepharose® e Affi- gel Blue Gel®. Da mesma forma, a pureza da enzima obtida depois desses passos cromatográficos foi comprovada por presença de banda única com 64 kDa em SDS-PAGE. Em ambos os protocolos de purificação, LmLAAO manteve sua atividade enzimática. A massa molar de LmLAAO foi determinada por espectrometria de massas (MALDI-TOF) e apresentou valor de 60,85 kDa. Além disso, foi determinado o valor do ponto isoelétrico da LmLAAO como 5,1. A LmLAAO mostrou preferência catalítica por aminoácidos hidrofóbicos (L-Metionina L-Leucina e L-Fenilalanina) e apresentou perda de atividade catalítica quando submetida à altos valores de pH ou de temperatura. Os parâmetros cinéticos foram determinados e a constante de Michaelis-Menten para o substrato L-Leucina foi de 0,9737 mmol/L e a velocidade máxima de reação foi de 0,06345 mol peróxido de hidrogênio/min. A sequência N-terminal dos 40 primeiros resíduos da LmLAAO purificada foi determinada por degração de Edman e a sua estrutura primária completa foi deduzida da sequência do cDNA obtido da glândula de peçonha. Verificou-se uma grande identidade entre as sequências em aminoácidos da LAAO de L. muta e as de outros viperídeos. A estrutura da LmLAAO foi resolvida por substituição molecular usando as coordenadas atômicas da LAAO de Agkistrodon halys pallas (PDB 1REO). As atividades farmacológicas da LmLAAO foram determinadas in vivo e in vitro. A injeção da enzima (10 µg) não induziu edema de pata em camundongos, nem hemorragia (50 µg) e nem toxicidade sistêmica (100 µg). No entanto, provocou uma leve mionecrose (100 µg) e edema em músculo quadríceps de camundongo, aumentando a creatina quinase plasmática. In vitro, foram testadas as atividades citotóxicas da LmLAAO em células de carcinoma. Os dados obtidos mostram IC50 de 22,70 µg/mL, para linhagem AGS (carcinoma de estômago), e IC50 de 1,41 µg/mL linhagem MCF-7 (carcinoma de mama). Para a atividade antiparasitária, foi determinada uma IC50 de 2,22 µg/mL para a forma promastigota de Leishmania brasiliensis. No entanto, a LmLAAO (32 µg/mL) não apresentou toxicidade relevante para a forma tripomastigota de Tripanosoma cruzi. Concluindo, este trabalho descreve o isolamento e a caracterização estrutural e funtional de uma nova LAAO da peçonha de L. muta. A enzima mostrou efeitos antitumorais e leishmanicida, sem toxicidade sistêmica relevante, mas apresentou significativa ação edematogênica e miotóxica local. Este estudo é relevante não apenas por contribuir para uma melhor compreensão do papel da LAAO no envenenamento, mas também por demonstrar seu potencial biotecnológico como agente terapêutico. / Snake venoms comprise a complex mixture of pharmacologically active substances, such as metalloproteases, phospholipase A2, serine proteases and L-amino acid oxidases (LAAOs) other compounds showing no enzymatic activity. LAAOs are flavoproteins catalyzing the oxidative deamination of L-amino acids to produce the corresponding -keto acid with the concomitant release of ammonia and hydrogen peroxide. Lachesis muta (L. muta) venom contains L-amino acid oxidase which may contribute to the envenomation. The aim of this study is the purification of an L-amino acid oxidase from Lachesis muta venom (LmLAAO) and its structural and functional characterization. For that, two purification protocols were performed and both provided highly pure LmLAAO. In the first protocol, 20 mg of crude venom of L. muta were submitted to a gel filtration on Sephacryl S100® and yielded ten fractions, whose were tested for L-amino acid oxidase activity. The first fraction showed L-amino acid oxidase activity and it was submitted to a further chromatographic step on Mono Q®. The homogeneity of the fraction showing L-amino acid oxidase activity after ion exchange was confirmed by the presence of a single band corresponding to 60.2 kDa by SDS-PAGE. The second purification protocol was a sequence of three chromatographic steps, where 200 mg of crude L. muta venom were submitted to gel filtration on Sephacryl S200, followed by hydrophobic interaction on Phenyl Sepharose® and Affi-gel-Blue Gel. For the second protocol, the purity of LmLAAO was confirmed by the presence of a single band with 64 kDa as determined by SDS-PAGE. In both purification protocols LmLAAO kept its enzymatic activity. The molar mass of LmLAAO was determined by mass spectrometry (MALDI-TOF) and showed a value of 60.85 kDa. Moreover, the isoelectric point was 5.1. In addition, LmLAAO showed a catalytic preference for hydrophobic amino acids (L-methionine, L-leucina and L-phenylalanine) and lost its catalytic activity when subjected to high pH or temperature. The kinetic parameters for LAAO were determined and the Michaelis-Menten constant for the substrate L-leucine was 0.9737 mmol/L, and the maximum reaction velocity was 0.06345µmol hydrogen peroxide/min. Furthermore, the sequence of the first forty residues was determined by Edman degradation and the complete sequence of LmLAAO was resolved by cloning cDNA obtained from the venom glands. The amino acid sequence of LmLAAO showed a great identity with sequences of LAAOs from other viper snakes. The LmLAAO structure was solved by molecular replacement using the the atomic coordinates of the LAAO from Agkistrodon halys pallas (PDB 1REO). In addiction, LmLAAO pharmacological activities were determined in vivo and in vitro. Thus, LmLAAO (10 µg) did not induce paw edema in mice, neither hemorrhage (50 µg) nor systemic toxicity (100 µg). However, it caused a mild myonecrosis (100 µg) and edema in the quadriceps muscles of mice, increasing plasma creatine kinase. In vitro activities of LmLAAO in carcinoma cells was assayed. The IC50 of LmLAAO on AGS cell line (stomach cancer) was 22.70 µg / mL, and the IC50 of LmLAAO on MCF-7 cell line (breast carcinoma) was 1.41 µg/mL. Moreover, antiparasitic activity of LmLAAO was determined on promastigote of Leishmania brasiliensis and an IC50 of 2.22 µg/mL was found, whereas on trypomastigote form Trypanosoma cruzi LmLAAO showed no toxicity at doses of 32 µg/mL. In conclusion, this work reports the isolation and the structural and funtional characterization of a new LAAO from L. muta snake venom. The enzyme showed antitumoral and leishmanicidal effects, without relevant systemic toxicity, but presented a significant local edema inducing and myotoxic action. This study is relevant not only for contributing to a better understanding of LAAO role in envenomation, but also for demonstrating its biotechnological potential as a therapeutic agent.

Modelos reprodutivos em serpentes: estocagem de esperma e placentação em Crotalus durissus e Bothrops jararaca (Serpentes: Viperidae) / Reproductive patterns: sperm storage and placentation in Crotalus durissus and Bothrops jararaca (Serpentes: Viperidae)

As serpentes pitvipers Crotalus durissus e Bothrops jararaca apresentam um ciclo reprodutivo sazonal com um estágio de desenvolvimento folicular ativo (vitelogênese Tipo II) no final do verão e início de outono. A cópula ocorre no outono (março até maio) e a gestação na primavera até o início do verão quando os nascimentos acontecem (Dezembro até Março). Depois do parto as serpentes permanecem em estado de quiescência folicular, o que as impede de reproduzir na próxima estação do mesmo ano. Assim, cada fêmea madura da população reproduz-se a cada dois anos caracterizando um ciclo reprodutivo bienal. A morfologia do trato reprodutivo em ambas as espécies mostra ovários e ovidutos assimétricos. Cada oviduto é dividido em três regiões distintas, a saber o infundíbulo, o útero (posterior e anterior) e a vagina. A assincronia entre a vitelogênese e a fertilização promove uma estocagem de esperma obrigatória. Esse processo ocorre na porção posterior do útero, por meio de uma contração muscular. Estudos morfofisiológicos do oviduto da jararaca e da cascavel combinados, com o uso de microscopias de luz, transmissão e varredura nunca foram conduzidos antes. A anatomia macroscópica do útero depois da cópula é caracterizada por uma torção e uma contração da musculatura uterina lisa longitudinal, a qual forma uma espiral. O útero permanece visivelmente contraído até a ovulação em setembro (início de primavera). O desenvolvimento embrionário ocorre no útero. Cada ovo ou embrião é isolado por uma constrição que funciona como uma implantação. A íntima aposição epitélio corioalantóico com o epitélio uterino indica que a placenta é do tipo epiteliocorial. As especializações da alantoplacenta em ambas as espécies caracteriza a placenta como (Tipo II). Em ambas as serpentes o oviduto parece ser um órgão capaz de exercer funções diferentes como a retenção e estocagem de esperma, manutenção e transporte dos ovos, fertilização e manutenção do embrião (gestação e placentação uterina). Machos de Bothrops e Crotalus mostram dois testículos alongados que se comunicam bilateralmente na cloaca por meio dos ductos deferentes convolutos. O diâmetro mais largo do ducto deferente, na porção distal foi observado no verão e outono, devido a espermiogênese, que ocorre um pouco antes da cópula. Em ambas as espécies observam-se um aumento dos testículos na primavera e verão provavelmente relacionada a espermatogênese, a qual também não é sincronizada com a cópula (outono) e a fertilização (setembro/outubro). Todas estas estratégias fisiológicas e morfológicas, tanto em machos quanto em fêmeas das duas espécies de serpentes promovem a liberação simultânea dos gametas no início da primavera (setembro), de modo a tornar harmônico o ciclo reprodutivo dos machos e das fêmeas. / The neotropical pitvipers Crotalus durissus and Bothrops jararaca show a sazonal reproductive cycle with an active stage of follicular development (vitellogenesis Type II) in late austral autumn and winter. Mating occurs in early to middle autumn (March to May) and gestation occurs in spring up to early summer when birth takes place (December up to March). After parturition snakes show a follicular quiescence which prevents them to reproduce in the following season of the same year. Therefore they skip one reproductive season characterizing a biennial reproductive cycle. Morphology of the reproductive tracts in both species showed the presence of unpaired ovaries and oviducts. Each oviduct is divided in four distinct regions, namely infundibulum, posterior and anterior uterus, and vagina. The lack of synchrony between vitellogenesis and fertlization, makes female store sperm obligatory. Sperm storage in both snakes occurs in the posterior portion of the uterus by means of a uterine muscle contraction and convolution. Morphological studies of the oviduct of jararaca and rattlesnakes combining the use of LM, TEM and SEM has never been carried out before. Macroscopical anatomy of the uterus after mating is characterized by convolution and contraction of the longitudinal smooth musculature which formed a spiral. The uterus remains visibly convoluted until ovulation occurs in September (early spring). Embryo development occurs in the uterus; each egg or embryo is isolated by a uterine short constricted segment that probably works as an implantation chamber like a placenta. The intimate apposition of the uterine lining with the chorioallantois indicates that the placenta is epitheliochorial type. The allantoplacenta, in both species develop folds in uterine and embryonic epithelia characterizing the second category of placenta (Type II). In both the species the oviduct has demonstrated to be an organ capable of some different functions such as sperm retaining and storage, egg uptake and transport, fertilization, maintenance of the embryo (uterine gestation and placentation) and parturition. Males of Bothrops and Crotalus show two elongated testes which communicate bilaterally with the cloaca through the coiled deferent ducts. The largest diameter of deferent ducto, was observed in summer and autumn, due to intense spermiogenesis, just before the mating season. In both species an increase in size of the testes was observed in spring and summer probably related with seasonal spermatogenesis, which is asynchronous in relation to mating (autumn). All these physiological and morphological strategies both in male and female B. jararaca and C. Durissus promote simultaneous liberation of gamets in early spring (september), in such a way to make reproductive cycle harmonic in these crotaline pitvipers.

A hematopoese nas serpentes inicia-se durante a embriogênese e através dos processos de alterações da vida fetal. A primeira atividade eritropoética é extraembrionária, a partir de células mesodérmicas do saco vitelínico e durante o desenvolvimento embrionário torna-se intraembrionário. Em serpentes recém-nascidas e adultas, o principal foco hematopoético ocorre na medula óssea. O objetivo deste estudo foi caracterizar os diferentes estágios de maturação das células sanguíneas da serpente O. guibei, com base em estudos de microscopia, reações citoquímicas e aspectos ultraestruturais. Fragmentos de vértebras de serpentes recém-nascidas e adultas (n=11) foram coletados para obtenção da medula óssea que foi fixada em formol cálcio ou Bouin e processadas para histologia de rotina. Cortes histológicos, imprint de medula óssea e esfregaços sanguíneos foram corados com Rosenfeld, hematoxilina e eosina ou azul de metileno. As reações citoquímicas realizadas foram ácido periódico de Schiff (PAS), azul de toluidina (AT), Sudan Black B (SBB), benzidina peroxidase (PA) e fosfatase ácida (FA). Para a microscopia electrónica de transmissão (TEM), a medula óssea foi fixada em paraformaldeído a 4% + glutaraldeído 2%, em tampão Tyrode, pós fixados em tetróxido de ósmio a 1% e embebidos em resina Epon 812. A maior parte das células progenitoras de células sanguíneas foram identificadas em focos hematopoiéticos ativos na medula óssea de vértebras e costelas. As linhagens azurofílicas e linfoides foram morfologicamente similares aos de outros répteis. A linhagem granulocítica foi classificada como mieloblasto, promielócito, mielócito e granulócito maduro. Mielócitos podem ser diferenciados em basófilos com grânulos grandes, redondos e eletrondensidade homogênia ou heterófilos, com grânulos de tamanhos e formas variadas na análise MET. TB e PAS foram positivos nos grânulos imaturos e maduros basófilos. Por outro lado, heterófilos e azurófilos mostraram reação fortemente positiva para lípidos de SBB e BP. FA foi encontrado nos azurófilos em várias fases de maturação. As diferentes fases de eritrócitos foram classificadas como: proeritroblasto, eritroblasto basófilo, eritroblasto policromático, proeritrócitos e eritrócitos maduros. Os trombócitos apresentaram positivadade para PAS. As características dos trombócitos maduros e imaturos foram definidas através da TEM apresentando corpos densos, grânulos alfa, microtúbulos e sistema canalicular aberto. Concluindo, a medula óssea das costelas e vértebras é um importante foco hematopoético nas serpentes O. guibei recém-nascidas e adultas. Os aspectos morfológicos, citoquímicos e ultraestruturais são úteis para identificar e caracterizar os diferentes estágios de maturação das células sanguíneas / Hematopoiesis in snakes begins during embryogenesis and the process changes through fetal life. The first erytropoietic activity is extraembryonic from mesoderm cells of the yolk sac and during the embryonic development it becomes intraembryonic. In newborn and adult snakes, the main site of hematopoiesis occurs in the bone marrow. The aim of this study was to characterize different stages of blood cells maturation of O. guibei snakes, based on microscopic studies including cytochemical stains and ultrastructural features. Fragments of vertebrae of newborn and adult snakes (n= 11) were collected to obtain bone marrow that was fixed in Bouin or formol calcium and processed routinely for histology. Tissue sections, imprint of bone marrow and blood smears were stained with Rosenfeld, hematoxylin and eosin or methylene blue. The cytochemical reactions performed were periodic acid-Schiff (PAS), toluidine blue (AT), sudan black B (SBB), benzidine peroxidase (BP) and acid phosphatase (FA). For transmission electron microscopy (MET), bone marrow was fixed in paraformaldehyde 4% + glutaraldehyde 2% in Tyrode buffer, postfixed in 1% osmium tetroxide and embedded in Epon 812 resin. Most of progenitors of blood cells were identified in the active hematopoietic focus in bone marrow of vertebrae and ribs. The azurophilic and lymphocytic series were morphologically similar to those of other reptiles. Granulocytic lineage was classified as myeloblast, promyelocyte, myelocyte and mature granulocytes. Myelocyte can be differentiated into basophils, with large, round and electrondensity homogeneous granules or heterophils, with varied size and shapes granules in the TEM analysis. AT and PAS were positive in the immature and mature basophils granules. On the other hand, heterophils and azurophils showed strong positive reaction for lipids staining of SBB and BP. FA was found on azurophils in various stages of maturation. The different stages of erythrocytes were classified as: proerythroblast, basophilic erythroblast, polychromatic erythroblast, proerythrocyte and mature erythrocytes. Thrombocytics cells showed PAS positive. The characteristics of mature and immature thrombocytes were defined using TEM identifying the dense bodies, alpha granules, microtubules and open canalicular system. Concluding, the rib or vertebral bone marrow is an important hematopoietic site in the newborn and adult O. guibei snakes. The morphologic, cytochemical and ultrastructural characteristics are useful to identify and characterize different stages of maturation of blood cells
