O emprego das técnicas de posicionamento de alta-precisão e de precisão, com recurso ao Sistema de Posicionamento Global, têm se consolidado como solução definitiva para a obtenção das coordenadas de pontos sobre a superfície terrestre, nas mais diversas aplicações, em particular aquelas envolvidas na realização do cadastro em áreas urbanas e construção da base de dados associada aos Sistemas de Informação Geográfica utilizados na gestão territorial. Os receptores GPS armazenam as informações dos satélites inicialmente em formato próprio, porém a maioria dos usuários não usa esse formato para trabalho, e sim o formato RINEX. Sendo este um formato importante ao trabalhar com GPS, os softwares proprietários que geralmente acompanham os receptores e fazem a transformação de seu formato próprio para o RINEX, processam as observáveis de forma exclusiva e fechada, não permitindo alteração de seu código-fonte (o que configura uma caixa preta). Este trabalho visa elaborar e implementar um programa com código-fonte aberto para leitura e manipulação de arquivos de efemérides transmitidas e precisas do Sistema de Posicionamento Global GPS e aplicá-lo ao posicionamento de satélites artificiais, e ainda realizar um estudo do comportamento do mesmo no posicionamento geodésico de pontos na superfície terrestre. Considerando-se que a posição terrestre dos satélites influencia diretamente no cálculo da posição de pontos sobre a superfície, esse estudo verifica que é possível obter coordenadas dos satélites a partir das efemérides transmitidas de forma coerente, e isto é feito utilizando-se o processamento que o programa faz ao manipular corretamente os arquivos RINEX. O programa implementado é capaz de lidar com os arquivos de formato RINEX e SP3, lendo, extraindo e utilizando informações para uma aplicação de posicionamento terrestre de satélites artificiais GPS, podendo ainda realizar outras aplicações como o posicionamento geodésico de pontos adaptando seu código-fonte para outros modelos. Esse programa poderá ser utilizado em benefício dos profissionais que atuam em diversas aplicações que envolvam o posicionamento geodésico por satélites, proporcionando flexibilidade de trabalho, pois o programa terá código-fonte aberto.

N-myc oncogene expression in neuroblastoma is dependent on Sp1 and Sp3

Regulation of N-myc oncogene expression is an important determinant of the biological behavior of neuroblastoma. The N-myc promoter contains several potential binding sites for transcription factors of the Sp1 family. Mutation of a CT-box motif contained within a 26 base pair region required for N-myc downregulation by retinoic acid decreased basal transcriptional activity and altered DNA-protein interactions of the promoter, while mutations flanking this motif did neither. On gel shift this region generated 3 specific DNA-protein complexes that were reliant on wild type sequence of the core CT element within it. Both Spl and Sp3 bound to the wild type probe as distinct complexes in specifically retarded bands, while neither protein was present on mutated sequences. Lysates from Drosophila S2 cells expressing exogenous Sp1 and Sp3 proteins were able to reproduce the gel shift complexes seen with neuroblastoma nuclear extract. Transient transfections of S2 cells showed that individually or together, Sp1 and Sp3 were able to trans-activate a N-myc CT-box-containing luciferase reporter construct in a dose-dependent manner. Conversely, transfection of CT-box oligonucleotide was able to decrease endogenous N-myc expression in neuroblastoma cells. Together these results suggest that the CT-box element serves a critical functional role, and in the basal state allows for N-myc transactivation by Spl and Sp3.

Neutron transport in hexagonal reactor cores modeled by trigonal-geometry diffusion and simplified P3 nodal methods

The superior advantage of a nodal method for reactor cores with hexagonal fuel assemblies discretized as cells consisting of equilateral triangles is its mesh refinement capability. In this thesis, a diffusion and a simplified P3 (or SP3) neutron transport nodal method are developed based on trigonal geometry. Both models are implemented in the reactor dynamics code DYN3D. As yet, no other well-established nodal core analysis code comprises an SP3 transport theory model based on trigonal meshes. The development of two methods based on different neutron transport approximations but using identical underlying spatial trigonal discretization allows a profound comparative analysis of both methods with regard to their mathematical derivations, nodal expansion approaches, solution procedures, and their physical performance. The developed nodal approaches can be regarded as a hybrid NEM/AFEN form. They are based on the transverse-integration procedure, which renders them computationally efficient, and they use a combination of polynomial and exponential functions to represent the neutron flux moments of the SP3 and diffusion equations, which guarantees high accuracy. The SP3 equations are derived in within-group form thus being of diffusion type. On this basis, the conventional diffusion solver structure can be retained also for the solution of the SP3 transport problem. The verification analysis provides proof of the methodological reliability of both trigonal DYN3D models. By means of diverse hexagonal academic benchmark and realistic detailed-geometry full-transport-theory problems, the superiority of the SP3 transport over the diffusion model is demonstrated in cases with pronounced anisotropy effects, which is, e.g., highly relevant to the modeling of fuel assemblies comprising absorber material.

Les activités catalytiques de complexes du ruthénium et de l'iridium ont été examinées dans trois thématiques. De nouvelles phosphines chirales bifonctionnelles à groupement acide et les complexes métalliques correspondants ont été préparés. Nous avons montré que le nouveau ligand (S)-Sulfo-binepine est très efficace pour l'hydrogénation énantiosélective de cétones aromatiques catalysée par le ruthénium et l'hydrogénation énantiosélective de cétiminesaromatiques en présence de catalyseurs de l'iridium. Sur la base d'études mécanistiques, un mécanisme de sphère externe a été proposé pour l'hydrogénation de cétones. Dans le cas de l'hydrogénation de cétimines, les intermédiaires-clés ont été obtenus, ce qui a permis de proposer deux chemins réactionnels compétitifs pour expliquer les effets sur l'énantiosélectivité. Les fonctionnalisations d'amines cycliques sur l'atome d'azote et les carbones en α et β de l'hétéroatome ont été réalisées grâce à des processus de transfert d'hydrogène assistés par des catalyseurs du ruthénium et l'iridium à ligand phosphinesulfonate. / In this doctoral thesis, the catalytic activities of new ruthenium and iridium complexes have been examined in three major topics. New chiral phosphine-containing acidic bifunctional ligands and correspondingmetalcomplexes have been prepared. (S)-Sulfo-binepine was shown to be a very efficient novel ligand for ruthenium-catalyzed enantioselective hydrogenation of aryl ketones and iridium-catalyzed enantioselective hydrogenation of arylketimines. Based on mechanistic studies, outer-sphere mechanism was proposed for ketone hydrogenation. For ketimine hydrogenation, the key intermediate and resting species have been obtained, allowing the proposal of two competitive reaction pathways to explain the effects on enantioselectivity. N- and(α,β)-C-functionalization of cyclic amines have been achieved via borrowing hydrogen processes assisted by ruthenium and iridium phosphinesulfonate catalysts.
