Desenvolvimento de antígeno vacinal para Staphylococcus spp na prevenção da piodermite canina / Development of vaccinal antigen to Staphylococcus spp in the canine pyoderma prevention

A piodermite bacteriana afeta cães de todas as idades, podendo ser recorrente por toda a vida. O principal agente etiológico é o Staphylococcus pseudintermedius, que é um micro-organismo saprófita da pele, agindo como oportunista frente a alguma fragilidade na barreira imunológica. A imunização dos cães através de uma vacina eficaz tem sido um desafio, pois a vacina comercial atualmente utilizada em clínicas veterinárias é de uso terapêutico e não tem caráter preventivo. Um dos problemas da vacina de uso terapêutico para piodermite é a dependência de um bom diagnóstico diferencial em relação a outras doenças de pele ou sistêmicas que tem como sequelas a erupção cutânea. Os animais que recebem a vacina terapêutica e não a preventiva são os que já apresentam problemas de pele reincidentes e são tratados concomitantemente com antibióticos. O objetivo do presente trabalho é desenvolver um antígeno vacinal com possibilidade de aplicação por via intradérmica de modo a tratar preventivamente a piodermite canina causada por Staphylococcus spp:. A via de administração intradérmica tem como finalidade estimular uma resposta imune sistêmica, principalmente por ativação das células dendríticas da derme, de modo a estimular estas células apresentadoras de antígeno no local onde ocorrem as infecções piodérmicas. Os resultados apresentados neste trabalho demonstraram que a inoculação intradérmica foram estatisticamente igual, quando comparada com a inoculação intraperitoneal, no que se refere à inibição por anticorpos: da atividade hemolítica, da atividade da catalase, da atividade da coagulase e da atividade citotóxica, bem como na reação antígeno-anticorpo determinado por ensaio imunoenzimático. Este trabalho também demonstra que a melhor formulação do antígeno estafilocóccico foi preparado a partir de sobrenadante de cultura diafiltrado por filtração tangencial com \"cut off\" de 5.000 Da. Outras preparações utilizando somente células bacterianas ou associação de sobrenadante com células bacterianas, apesar de produzirem antigenicidade, apresentaram títulos inferiores do que quando empregado somente sobrenadante de cultura diafiltrado. / The bacterial pyoderma affects dogs of all ages and can be recurrent for a lifetime. The main etiologic agent is Staphylococcus pseudintermedius, which is a saprophytic organism of the skin, acting as an opportunistic opposite to some weakness in the immune barrier. The immunization of dogs through an effective vaccine has been a challenge, since currently it has been used commercially in veterinary clinics is therapeutic and has a preventive character. One of the problems of the therapeutic use of the vaccine for pyoderma is dependent on a good differential diagnosis in relation to other skin diseases or systemic consequences which is the rash, so treatment often does not reach its goal. Animals receiving this vaccine are those who already have skin problems again and are receiving concomitant antibiotics. The objective of this study is to develop a vaccine antigen with the possibility of implementing a preventive treatment for canine pyoderma caused by Staphylococcus spp: through pre-clinical studies, for use in young and adults, preferably before they develop any type of skin disorder primary or secondary. The administration route is intradermal, stimulating an immune response primarily by dendritic cells to become a barrier to microorganisms, as well as systemic, by the production of antibodies, to last for at least one year until needed in the body booster vaccination. The results presented here demonstrated that the intradermal inoculation was statistically equivalent when compared to intraperitoneal inoculation with regard to inhibition by antibody: of the hemolytic activity, catalase activity, the coagulase activity and cytotoxic activity, as well as antigen-antibody reaction determined by enzyme immunoassay. This work also demonstrates that the best estafilococcico antigen formulation was prepared from the culture supernatant diafiltered by tangential filtration with \"cut off\" of 5,000 Da. Other preparations using only the bacterial cells or the association of supernatant with bacterial cells, although producing antigenicity, showed lower titles than when used dialyzed culture supernatant alone.

Avaliação de moléculas bioativas produzidas por isolados actinomicetos contra cocos Gram positivos de origem clínica / Characterization of bioactive molecules produced by actinomycetes isolated against clinical cocos gram positive

Os actinomicetos são bactérias Gram positivas caracterizadas por sua habilidade em formar hifas, são amplamente distribuídos no ambiente e conhecidos pela diversidade na produção de moléculas biologicamente ativas. O presente trabalho teve por objetivo avaliar a atividade de compostos produzidos por quarenta isolados de actinomicetos contra isolados clínicos de Enterococcus sp, Staphylococcus aureus e Staphylococcus epidermidis. O perfil de suscetibilidade das amostras clínicas foi avaliado empregando a técnica de disco difusão em ágar. A atividade antimicrobiana dos actinomicetos foi avaliada pela técnica da dupla camada. Os isolados que apresentaram atividade foram cultivados em caldo amido caseína à temperatura de 30ºC por sete dias, com agitação constante. Após o crescimento, a cultura foi filtrada para obtenção do extrato bruto. A atividade antibiótica do extrato foi avaliada através da técnica de difusão em poço. O isolado que apresentou maior espectro de ação foi selecionado para otimização dos compostos. A otimização da produção dos compostos com atividade antibiótica foi realizada através da avaliação de curva de produção, variação da fonte de carbono, tempo de incubação, pH tamponado e pH não tamponado. No ensaio de sobrecamada os isolados 50 e 8S apresentaram atividade contra a 90% das amostras de microrganismos clínicos de Staphylococcus sp. e Enterococcus sp. No ensaio de difusão em poço o isolado 50 apresentou maior atividade antibiótica que o isolado 8S. Na otimização do extrato as melhores condições de produção foram: 72 h de crescimento, fonte de carbono amido e sem tamponamento de pH. Não foi observada influência de biomassa na produção dos compostos. A cromatografia em camada delgada revelou a presença de duas bandas com fator de retenção de 0,28 (Rf1) e 0,57 (Rf2). / Actinomycetes are Gram positive bacteria, characterized by their ability to form hyphae. They are widely distributed in the environment and known for their diversity in producing biological active molecules. This study aimed to evaluate the activity of compounds produced by forty isolates of Actinomycetes against clinical isolates of Enterococcus sp, Staphylococcus aureus and Staphylococcus epidermidis. The susceptibility profile of the samples was evaluated using the disk diffusion technique in agar. The antimicrobial activity of actinomycetes was assessed by means of the double layer. Isolates that showed activity were grown in starch casein broth at a temperature of 30 ºC for seven days, with constant agitation. After growth the culture was filtered to obtain a crude extract. The antimicrobial activity of the extract was evaluated by the well diffusion technique. The actinomycete that showed activity against most of the test samples was selected for optimization(s) of the compound(s) production. The optimization of the production was performed by evaluating: growth curve, the use of different carbon source, changes in the incubation time, and culture media with buffered and unbuffered pH. In the overlay assay isolates 50 and 8S presented activity against most of Staphylococcus sp. and Enterococcus sp samples. In diffusion assay isolate 50 showed higher antibiotic activity than the isolated 8S. Compiling the results the best production conditions were: 72 h of growth, carbon source starch without pH buffering at 30°C. There was no effect of biomass (s) compound (s) activity. The thin layer chromatography revealed the presence of two bands with a retention factor of 0.28 (Rf1) and 0.57 (Rf2).

Caracterização genotípica de cepas de Staphylococcus aureus recuperadas de alimentos, mãos de manipuladores de alimentos e veiculadas por formigas / Genotypic characterization of strains of Staphylococcus aureus recovered from food, hands of manipulators of food and run by ants

A intoxicação alimentar causada por toxinas de Staphylococcus aureus é uma das principais causas de Doenças Transmitidas por Alimentos, principalmente em locais onde se preparam grandes quantidades de refeições, sendo o homem considerado a principal fonte de contaminação dos alimentos por este patógeno, pois de 30 a 50 % das pessoas sadias são portadoras desse microrganismo. O presente trabalho teve como objetivo comparar geneticamente cepas de S. aureus isoladas das mãos de manipuladores de alimentos, com cepas da mesma bactéria, recuperadas de alimentos e veiculadas por formigas. As cepas recuperadas dos manipuladores foram obtidas a partir das mãos destes utilizando-se a técnica de suabe e as cepas recuperadas de alimentos e veiculadas por formigas pertenciam à coleção de culturas do Laboratório de Microbiologia e Biologia Molecular do Departamento de Prática de Saúde Publica da Faculdade de Saúde Publica da USP. Todas as amostras foram submetidas às técnicas de PCR para verificação da presença de plasmídios e do gene sea. As análises microbiológicas revelaram que 18,4% dos manipuladores albergavam S. aureus em suas mãos. A confirmação da espécie aureus por PCR revelou somente 15,4% das cepas recuperadas dos manipuladores e 62,5% e 100% das recuperadas de alimentos e veiculadas por formigas, respectivamente. O perfil plasmidial revelou a existência de 2 grupos de cepas com perfis semelhantes e a PCR para o gene sea, que codifica a produção da enterotoxina SEA, demonstrou sua presença em 78,6% das cepas. A caracterização das cepas por ERIC-PCR evidenciou que todas as cepas apresentaram uma porcentagem de semelhança maior que 84% e que, as cepas de maior similaridade, apresentaram cerca de 90% de semelhança. Conclui-se que é de extrema importância a confirmação da espécie por método molecular, uma vez que este apresenta maior especificidade e sensibilidade do que os métodos microbiológicos; cepas isoladas de manipuladores, alimentos e veiculadas por formigas apresentavam certas semelhanças em relação ao perfil plasmidial, entretanto, a presença de plasmídio não esteve ligada necessariamente à presença do gene sea, normalmente relacionados; a técnica de ERIC-PCR constitui ferramenta importante para caracterização genética de cepas e elucidação de surtos de DTAs. / The food poisoning caused by toxins of Staphylococcus aureus is one of the main causes of illnesses transmitted by foods, mainly in places where there is prepare of great amounts of meals, since human being is considered the main source of contamination of foods for this microrganism, therefore of 30% to 50 % of the healthy people are S. aureus carriers. The present work had as objective to compare genetically strains of S. aureus isolated from hands of food handlers, with strains of the same bacteria, recovered from foods and carried by ants. Strains recovered from food handlers were obtained by technique of swab and strains recovered from foods and carried by ants belong to collection of cultures of the Laboratory of Microbiology and Molecular Biology of Department of Practical of Public Health of College of Public Health - USP. All strains were submitted to PCR for verification of the presence of plasmids and sea gene. The microbiological analyses had observed that 18,4% of the food handlers were S. aureus carriers. The confirmation of aureus species by PCR had shown only 15.4% of strais recovered from food handlers and 62.5% and 100% recovered from foods and propagated for ants, respectively. The plasmidial profile revealed presence of 2 groups of strains with similar profiles and PCR for verification of sea gene, that it codifies the production of SEA enterotoxin, demonstrated that 78,6% of strains were positive. The caracterization of strains by ERIC-PCR revealed that all of them had similarity above 84% and, strains that were very similar, presented 90% of similarity. It is concluded that it´s extremely important confirmation of species by molecular methods, since performed higher especificity and sensitivity than microbiological methods; strains isolated from food handlers, foods and carried by ants presented relative similarities in relation to the plasmidial profile, however, plasmid presence was not linked, necessarily, to presence of sea gene, normally related; ERIC-PCR is important tool to genetic diferentiating strains and to solve outbreaks.

Rôle inflammatoire des plaquettes sanguines lors du sepsis / Inflammatory platelet’s roles during sepsis

Depuis quelques années, les plaquettes sanguines sont reconnues comme un véritable élément de l’immunité. En effet, de nombreux rôles clés lors du processus inflammatoire sont attribués aux plaquettes, tels que la détection d’un signal, la libération de nombreux immunomodulateurs ou une forte interaction avec les autres cellules immunitaires. Le constat clinique d’une corrélation entre le taux plaquettaire et la mortalité du sepsis permet de relever l’importance de cette cellule dans la physiopathologie du sepsis. Ainsi ce travail doctoral a eu pour objectif de mettre en relief la part inflammatoire des plaquettes lors des interactions entre les facteurs de virulence de Staphylococcus aureus (S.aureus) que sont différentes exotoxines ou de souches de S. aureus issues de bactériémies de patients septiques. Un modèle expérimental de sepsis murin nous a permis de comprendre la composante inflammatoire jouée par les plaquettes ainsi que leurs implications dans la dysfonction pulmonaire issue lors du sepsis. Enfin, nous avons évalué la modulation de molécules antiplaquettaires, et particulièrement de l’acide acétylsalicylique, sur les nombreux rôles pathogéniques des plaquettes durant cette pathologie. / During some years, platelets are recognized as an key element of immunity. In fact, platelets play several roles, as signal detection, immunomodulator release and interaction with other immunity cells. The clinical significance of a correlation between platelet rate and sepsis mortality reveals the importance of this cell in the pathophysiology of sepsis. Thus, the purpose of this doctoral work was to highlight the inflammatory role of platelets in the interactions between the virulence factors of Staphylococcus aureus (S. aureus) that are different exotoxins or strains of S. aureus from bacteremia of septic patients. An experimental model of murine sepsis allowed us to understand the inflammatory component played by platelets as well as their implications in pulmonary dysfunction resulting from sepsis. Finally, we evaluated the modulation of antiplatelet molecules, particularly acetylsalicylic acid, on the numerous pathogenic roles of platelets during this pathology.

Fatores de virulência e genes regulatórios agr de Staphylococcus aureus e outras espécies coagulase positivas isoladas de mastites bovina e ovina / Virulence factors and regulatory genes (agr) of Staphylococcus aureus and other coagulase-positive species isolated from bovine and ovine mastitis

A mastite é um sério problema em rebanhos leiteiros com consideráveis conseqüências econômicas. Embora alguns patógenos possam causar essa doença, Staphylococcus aureus é o mais importante agente etiológico em mastites bovinas e ovinas. O surgimento de linhagens resistentes a diferentes antibióticos e a presença de múltiplos fatores de virulência dificultam o tratamento das infecções causadas por essa espécie. Neste estudo, foram analisados 70 isolados de estafilococos coagulase positivos de origem animal: 52 provenientes de amostras de leite de bovinos portadores de mastite subclínica, do óstio dos tetos desses animais e de insufladores de três fazendas de exploração leiteira do Estado de São Paulo. Os outros 18 isolados foram provenientes de ovinos da região do agreste do Estado de PE, sendo 6 de casos de mastite clínica e 12 de casos de mastite subclínica. Por meio da eletroforese de campo pulsado (PFGE), pôde-se concluir que houve alta diversidade genética entre as linhagens de S. aureus isoladas das diferentes fontes analisadas das três fazendas do Estado de SP. No entanto, um perfil clonal comum foi identificado nas amostras de leite de duas dessas fazendas e, em uma terceira fazenda, houve a predominância de outro perfil clonal de S. aureus entre os isolados de leite bovino proveniente de animais portadores de mastite subclínica. Em relação às outras espécies coagulase positivas, todas originadas de óstios bovinos, foi possível determinar os perfis clonais dos isolados S. schleiferi spp coagulans e S. delphini e identificar a ocorrência de dois perfis distintos entre as linhagens de S. intermedius. Os isolados S. aureus provenientes de ovinos mostraram baixa diversidade genética com três perfis clonais comuns aos casos de mastite clínica e subclínica. Observou-se, em relação à combinação dos fatores de virulência de bovinos e ovinos, a ocorrência do gene cflA em 100% das amostras, o que indica o seu envolvimento no processo de colonização da glândula mamária, independente do hospedeiro. Dois clones bovinos destacaram-se por apresentar multiplicidade dos genes codificadores dos fatores de virulência. No entanto, houve pouca ocorrência dos genes relacionados ao processo toxigênico entre os isolados dessa espécie animal, ao contrário das cepas provenientes de ovinos. Em relação ao sistema regulador agr, o grupo II foi associado aos perfis clonais que apresentaram maior combinação dos genes codificadores dos fatores de virulência em isolados S. aureus provenientes de bovinos. Já o grupo III e agr negativos, aos isolados portadores de poucos desses genes. A presença do gene eta, pouco freqüente em linhagens animais, foi associada tanto com cepas bovinas agr negativas como do grupo II. Assim como nos isolados bovinos, o grupo II de agr, em ovinos, também foi relacionado com o perfil que apresentou maior combinação dos fatores de virulência, incluindo a associação dos genes sec e tst, mas também com perfis portadores de poucos genes de adesinas ou de nenhum gene para exotoxinas. O mesmo foi observado para o grupo I em linhagens ovinas. As cepas resistentes à oxacilina (BORSA), provenientes de isolados bovinos e de insufladores, apresentaram concentração inibitória mínima ≥1 - ≤ 16 µg/ml com múltipla resistência a outros antimicrobianos, mas foram negativas para o gene mecA, o que indica que sejam portadoras de outros mecanismos de resistência aos beta-lactâmicos, diferente da produção de PBP2a. / Mastitis is a serious problem in dairy herds with considerable economic consequences. Although some pathogens may cause this disease, Staphylococcus aureus is the most important etiologic agent in bovine and ovine mastitis. The appearance of antibiotic resistant strains and the presence of multiple factors of virulence, make the treatment of the infections caused by these species very difficult. In this study, we analyzed 70 isolates of coagulase positive Staphylococcus sp of animal origin: 52 from milk samples of subclinic mastitis, bovine teats and milking linners collected in three dairy farms from São Paulo State. The other 18 isolates were from the agreste region ovine in the State of Pernanbuco, with 6 strains from cases of clinic mastitis and 12 cases of subclinic mastitis. By pulsed-field electrophoresis (PFGE), we found high genetic diversity among S. aureus strains from different sources analyzed in the three farms from São Paulo State. However, a common S. aureus cloning profile was identified in the milk samples from two farms and, the third farm, presented a predominance of different profile from two previous farms. The six coagulase positive isolates from bovine teats were identified as S. schleiferi spp coagulans (1), S. delphini (1) and S. intermedius (4). This last specie presented two distinct profiles among strains. The ovine S. aureus strains showed a low genetic diversity presenting three common profiles in both clinic and subclinic mastitis. The cflA gene occurred in 100% of the bovine and ovine strains, showing its involvement in the process of colonization of the mammary glands independent of host species. Two bovine clones was notable for presenting multiplicity of virulence factors genes. However, there was a low occurrence of toxin encoding genes among bovine strains, unlike the ovine strains. The group II agr regulatory systems of S. aureus from bovine strains was associated to a wider combination of the virulent factors encoding genes. However, group III and negative agr was associated to few combination genes. Although the gene eta is, in general, less frequent in animal strains, in this study it was associated in both agr negative and group II agr bovine strains. In ovine, agr group II, as in the bovine isolates, it was also related to a wider combination of virulence factors, including the association of sec and tst genes. It was also found a few genes encoding adhesion and exotoxins in this group, occurring the same in agr group I in ovine strains. The borderline oxacillin-resistant Staphylococcus aureus (BORSA) strains isolated from bovine ostium, milk and milking apparatus line presented minimum inhibitory concentration ≥1 - ≤ 16 µg/ml with multiple resistance to other antimicrobial, but they were negative to the mecA gene, suggesting they are carrying other resistance mechanisms to beta-lactam antibiotics, different from the production of PBP2a.

Identification of antibiotic-resistant staphylococci and epidemiological typing of methicillin-resistant Staphylococcus aureus by Fourier transform infrared spectroscopy

Staphylococci strains are among the most widespread multidrug-resistant nosocomial pathogens in Canada. Rapid and accurate identification and epidemiological typing of methicillin-resistant S. aureus (MRSA) and its discrimination from coagulase-negative staphylococci (CNS) and glycopeptide-intermediate S. aureus (GISA) are crucial for appropriate therapy and for monitoring and limiting intra- and inter-hospital spread of epidemic MRSA strains. Although pulsed-field gel electrophoresis and polymerase chain reaction methods for the identification of MRSA are reliable, they are technically demanding, time-consuming and inappropriate for routine clinical diagnosis. Moreover, no reliable method exists for discrimination of epidemic MRSA from sporadic MRSA and from GISA strains. The objective of the research described in this thesis was to investigate whether Fourier transform infrared (FTIR) spectroscopy could be used to distinguish MRSA from methicillin-susceptible S. aureus, borderline oxacillin-resistant S. aureus (BORSA), CNS, including methicillin-resistant CNS, and GISA. The application of FTIR spectroscopy for epidemiological typing of Canadian epidemic MRSA (CMRSA) strains as well as their discrimination from sporadic MRSA was also assessed. FTIR spectra were recorded from intact stationary-phase cells grown on Universal Medium (UM™) and deposited and dried on a ZnSe optical window, normalized, and converted to first-derivative spectra. Various chemometric approaches were employed to cluster the different phenotypes of staphylococci species and to subtype five CMRSA strains based on the similarity of their infrared spectral fingerprints in narrow spectral regions selected by visual inspection and by employing a singularvalue decomposition (SVD) algorithm. Pairwise separation of MRSA from MSSA, BORSA, CNS, MRCNS, and GISA was accomplished by using principal component analysis (PCA), self-organizing maps (SOM), and the K-nearest neighbors (KNN) algorithm. These chemometric techniques were also successfully employed for epidemiological typing of the five CMRSA strains and their discrimination from sporadic MRSA strains using a combination of different optimal spectral regions selected by SVD. These results demonstrate that FTIR spectroscopy has considerable potential as a rapid method for the identification of different phenotypes of staphylococci and epidemiological typing of MRSA.

Bakterijų jautrumo antimikrobinėms medžiagoms tyrimai žaliuose paukštienos produktuose / Investigations on bacterial susceptibility to antimicrobial agents in raw poultry products

Darbo tikslas – ištirti Lietuvoje realizuojamų gyvūninės kilmės žalių paukštienos produktų (kepenų) užterštumą atspariomis antimikrobinėms medžiagoms Escherichia coli rūšies bei Staphylococcus genties bakterijų padermėmis. Darbas atliktas Lietuvos sveikatos mokslų universitete, slėnio „Nemunas“ atviros prieigos centro mikrobiologinių tyrimų laboratorijoje. Iš paukštienos subproduktų (kepenų) buvo išskirtos ir identifikuotos 209 bakterijų padermės. Paplitimo dažnis E. coli: kalautienoje – 60 proc., vištienoje – 62,2 proc.; stafilokokų: kalakutienoje – 90 proc., vištienoje – 87,6 proc. Nustatytas šių bakterijų jautrumas įvairiems antibiotikams naudojant kokybinį tyrimų metodą, duomenis interpretuojant pagal EUCAST klinikinius lūžio taškus. E. coli išskirtos iš paukštienos subproduktų pasižymėjo dažniausiu atsparumu ampicilinui (64,2 proc.), ciprafloksacinui (25,4 proc.), sulfametoksazoliui-trimetoprimui (37,7 proc.), aztreonamui (23,9 proc.), cefoksitinui (22,4 proc.) bei kur kas retesniu atsparumu gentamicinui (6 proc.). Staphylococcus genties bakterijos dažniausiai buvo atsparios sulfametoksazoliui-trimetoprimui (57,4 proc.), penicilinui G (54,5 proc.), eritromicinui (28,7 proc.). [toliau žr. visą tekstą] / The aim of this study was to investigate contamination of raw puoltry products (liver) sold in retail markets by antimicrobial resistant Escherichia coli and Staphylococcus. The research was carried out at Microbiological Research Laboratory of open access Center “Nemunas” of Lithuanian University of Health Sciences. Two hundred and nine isolates from poultry liver were isolated and identified. The prevalence of E. coli was 60% in turkey and 62,2% in chicken, while staphylococci were found in 90% in turkey and 87,6% in chicken liver samples. The susceptibility of these bacteria to various antibiotics was estimated by using antibiotic disc method; the data obtained was interpreted according to clinical breakpoints presented by EUCAST. E. coli isolated from poultry were the most frequently resistant to ampicillin (64,2%), ciprofloxacin (25,4%), sulfamethoxazole-trimethoprim (37,7%), aztreonam (23,9%), cefoxitin (22,4%) and with a lesser extent to gentamicin (6%). Staphylococci demonstrated frequent resistance to sulfamethoxazole-trimethoprim (57,4%), penicillin G (54,4%) and erythromycin (28,7%). Slightly lower resistances were observed to tetracycline (13,9%), cefoxitin (11,9%), gentamicin (8,9%) and ciproflaxicin (7,9%). Staphylococcus aureus prevalence was determined in 55.6% of turkey and in 19,6% of chicken liver products.
