Tim Burton est un réalisateur américain qui crée des personnages atypiques, par leur nature, leurs caractéristiques physiques ou mentales. De son premier court-métrage professionnel Vincent à son dernier long-métrage Frankenweenie, il divertit, mais donne sa vision du monde à travers un portrait de la société, du monde réel, qui s'oppose au monde fantastique. Ce dernier correspond au cinéma, mais aussi au rêve, et lui permet de sortir de son quotidien. En créant ses mondes, il s'adresse à un public particulier, qui peut s'identifier dans ses personnages exclus, détachés de la société. Le personnage burtonien est souvent naïf. Il se trouve dans une situation initiale où il n'a pas vécu d'aventures. Il cherche à sortir de son monde, définitivement ou provisoirement. À plusieurs reprises durant le film, il se déplace entre un monde d'origine et un monde à atteindre. Ces déplacements, associés à des ellipses temporelles, des jeux de couleurs et de lumières ou la construction d'une relation avec un allié lui permettent de changer de statut, quel que soit le monde dans lequel il se trouve à la fin du film. Le but de ce mémoire est donc de mettre en avant les caractéristiques récurrentes du personnage principal burtonien à travers trois films qui constituent mon corpus : Edward Scissorhands (Burton, 1990), Charlie and the Chocolate Factory (Burton, 2005) et Alice in Wonderland (Burton, 2010). Il s'agit par la suite de vérifier si cette figure se retrouve dans le reste de la filmographie du réalisateur. The ecclesiological offices of bishop, priest and deacon holds center stage in the Church of Sweden and many other churches of our time. These offices are often legitimized by references to their scriptural heritage in the New Testament. There is certainly a linguistic connection between “bishop” and the Greek word ἐπίσκοπος, between “priest” and πρεσβύτερος and between “deacon” and διάκονος. The possible connection in meaning between each pair of words is far from obvious, however. Therefore, in this study I investigate what these words meant in their original context.  The purpose of the study is to contribute to the understanding or the function of ἐπίσκοπος according to First Timothy. The research question posed; What knowledge can be obtained in regard to this function and the requirements for becoming an ἐπίσκοπος according to 1 Tim 3:1-7? The study is divided into three parts. The first part surveys current research regarding the three Pastoral Epistles, with a focus on 1 Tim. The second part reviews current research on institutionalization in the Corpus Paulinum. Margret MacDonalds socio-historical study of Institutionalization in the Pauline and deutero-Pauline writings is particularly important. In addition, I review Berger and Luckmanns theory about the social construction of reality alongside Max Weber’s theory about routinized charisma. The third part of the study is a textual analysis of 1 Tim 3:1-7. I conclude that the central theme in 1 Tim 3:1-7 is the position (office or function) of ἐπισκοπή and not the person ἐπίσκοπος. An understanding of ἐπίσκοπος must therefore be consistent with one´s construal of ἐπισκοπή. It is argued that ἐπισκοπή is an institutionalized form of leadership with strong parallels in Greek-Roman moral philosophy. The institution described in 1 Tim 3:1-7 has apparently become quite fixed. Given the Greek-Roman parallels, that institution seems to contain a hierarchical and authoritarian form of leadership – legitimized by moral excellence. Further, 1 Tim 3:1-7 bears no witness to any specifically Christian innovations in leadership.

Para tanto, faremos um cotejo entre as duas obras à luz das teorias de Elizabeth Sewell (2015), Wim Tigges, (1988) e Susan Stewart (1980) que alicerçam o gênero literário nonsense. Lançaremos mão da versão literária Lewis Carroll: The Complete Fully Illustrated Works de 1995 da editora Gramercy, contendo toda a obra de Lewis Carroll com ilustrações originais. Isso, a fim de viabilizar e facilitar a comparação com as figuras presentes no texto literário, quando necessário, para investigar se houve aproximação na representação visual feita pelo diretor.

This master thesis deals with proposal of Tim Hortons' communications strategy, Canada's largest chain of fast food restaurants, when entering the Czech market. The main theme is the analysis of existing communications strategy and the subsequent setting of the marketing communication plan for 2011. The thesis is divided into two parts - theoretical and practical. Theoretical part characterizes the marketing and commercial communications and provides the basic theory of the stages of marketing communication plan planning. The practical part introduces company Tim Hortons, its marketing initiatives and then the concrete steps to build a communication strategy for the Czech market. Based on comparison of Tim Hortons brand positioning with its main competitors it is created the right communication campaign, its timing, budget and usage of appropriate media. Finally, the thesis summarizes the results of the analysis and the key pillars of the proposed marketing communication plan, which are positioning, creative and media strategy, sponsorship and campaign management.

Investigation into the molecular mechanisms of import of mitochondrial small Tim proteins

Protein import is essential for the biogenesis of mitochondria, as the majority (99%) of mitochondrial proteins are synthesised in the cytosol and thus, have to be imported into mitochondria for their function. The biogenesis of many cysteine-containing proteins of the mitochondrial intermembrane space (IMS), such as members of the small TIM and Cox17 families, is regulated by their thiol-disulphide redox state. Only the Cys-reduced precursors can be imported into mitochondria, whereas oxidised forms cannot. Their import and oxidative folding in the IMS is driven by the IMS disulphide relay system, known as mitochondrial import and assembly (MIA) pathway, whose central components are the oxidoreductase Mia40 and the sulphydryl oxidase Erv1. Currently, little is known about how the MIA precursors are maintained in the cytosol in an import-competent form, and whether they interact with the translocase of the outer membrane (TOM complex) to enter the IMS. In addition, the MIA-mediated protein folding events occurring in the IMS that lead to the generation of fully oxidised substrates are still under investigation. Using Tim9 and yeast as models, studies presented in this thesis showed that Tim9 binding to the mitochondrial outer membrane (OM) does not depend on the receptors of the TOM complex, and occurs without regard to the redox state of the precursor proteins. In addition, it is shown that the oxidised and reduced precursors share the same binding site on the OM, and that this binding site is not important for the translocation process across the OM (Chapter 3). Studies in this thesis investigated the role of the cytosolic thioredoxin and glutaredoxin systems in the biogenesis of mitochondria. Firstly, in vivo studies provided the evidence that the cytosolic thioredoxin system but not the glutaredoxin system is required for growth of yeast cells under respiratory conditions. Secondly, in vivo studies provided the first proof that the Trx system is required for the biogenesis of small Tim proteins. In vitro studies confirmed that the Trx1 system facilitates import of small Tim proteins into isolated mitochondria by maintaining the precursors in a reduced and therefore competent form (Chapter 5). Finally, in vitro studies showed that Mia40 is able to promote the full oxidation of Tim9. Efficient release of Tim9 from Mia40 required the presence of all cysteine residues of Tim9, as effective oxidation and concomitant release from Mia40 failed upon mutation of single cysteine residues. Finally, the study showed that reduced glutathione resolved rapidly the Mia40-Tim9 mixed-disulphide complexes, probably accelerating and/or promoting the Tim9 oxidative (Chapter 4).

Critical Role of Tim-3 Mediated Autophagy in Chronic Stress Induced Immunosuppression

Background: Psychological and physical stress can either enhance or suppress immune functions depending on a variety of factors such as duration and severity of stressful situation. Chronic stress exerts a significantly suppressive effect on immune functions. However, the mechanisms responsible for this phenomenon remain to be elucidated. Autophagy plays an essential role in modulating cellular homeostasis and immune responses. However, it is not known yet whether autophagy contributes to chronic stress-induced immunosuppression. T cell immunoglobulin and mucin domain 3 (Tim-3) has shown immune-suppressive effects and obviously positive regulation on cell apoptosis. Tim-3 combines with Tim-3 ligand galectin-9 to modulate apoptosis. However, its impact on autophagy and chronic stress-induced immunosuppression is not yet identified. Results: We found remarkably higher autophagy level in the spleens of mice that were subjected to chronic restraint stress compared with the control group. We also found that inhibition of autophagy by the autophagy inhibitor 3-methyladenine (3-MA) significantly attenuated chronic stress-induced alterations of pro-inflammatory and anti-inflammatory cytokine levels. We further elucidated that 3-MA dramatically inhibited the reduction of lymphocyte numbers. Moreover, chronic stress dramatically enhanced the expression of Tim-3 and galectin-9. Inhibition of Tim-3 by small interfering RNA against Tim-3 significantly decreased the level of autophagy and immune suppression in isolated primary splenocytes from stressed mice. In addition, α-lactose, a blocker for the interaction of Tim-3 and galectin-9, also decreased the autophagy level and immune suppression. Conclusion: Chronic stress induces autophagy, resulting with suppression of immune system. Tim-3 and galectin-9 play a crucial regulatory role in chronic stress-induced autophagy. These studies suggest that Tim-3 mediated autophagy may offer a novel therapeutic strategy against the deleterious effects of chronic stress on the immune system.
