Future tense: lessons from the best and worst cases in Afghanistan from Pakistan's perspective

Approved for public release; distribution is unlimited / Reissued 5 May 2017 with typographic corrections to degree, abstract. / Broadly, this thesis argues that success in Afghanistan—for Afghans and their neighbors alike—entails establishing state-building measures, supremacy of law and human rights, rapid economic growth, and strong Afghan National Security Forces who can encourage the Afghan public to accept the practices of stable democracy and good neighborly relations. To fix some of the myriad variables in Afghanistan's likely state in 2026, which will mark the end of the transformation decade following the U.S. withdrawal in 2016, the proposed project installs three hypothetical scenarios: The Good, The Bad, and The Ugly. These scenarios do not predict the future in Afghanistan but rather shed light on the factors and variables that will shape the postwar period. Examining such scenarios may allow strategic planners to develop alternative measures for complex situations. Sustained political stability and the will to reform by the Afghan establishment, fortified by consistent international financial and military support for at least 10 to 15 or more years, would likely be the key to success in the Afghanistan end game. Otherwise, Afghanistan will slip back into a situation much like the one that emerged once after the Soviet withdrawal—extremism, war, and instability. Epidemiological study on antimicrobial use and non-compliance with withdrawal times in broiler farms in Vietnam

L’utilisation d’antimicrobiens chez les animaux de consommation est une source de préoccupation importante pour la santé publique à travers le monde en raison de ses impacts potentiels sur l’émergence de micro-organismes résistants aux antimicrobiens et sur la présence de résidus antimicrobiens néfastes dans la viande. Cependant, dans les pays en développement, peu de données sont disponibles sur les pratiques d’utilisation des antimicrobiens à la ferme. Par conséquent, une étude épidémiologique transversale a été menée de juin à août 2011 dans des élevages de poulets de chair situés dans le sud du Vietnam, ayant pour objectifs de décrire la prévalence d’utilisation des antimicrobiens ajoutés à l’eau de boisson ou aux aliments à la ferme, et de tester les associations entre les caractéristiques des fermes et la non-conformité avec les périodes de retrait recommandés sur l’étiquette des produits. Un échantillon d’accommodement de 70 fermes a été sélectionné. Les propriétaires des fermes ont été interrogés en personne afin de compléter un questionnaire sur les caractéristiques des fermes et les pratiques d’utilisation d’antimicrobiens. Au cours des 6 mois précédant les entrevues, il a été rapporté que la colistine, la tylosine, l’ampicilline, l’enrofloxacine, la doxycycline, l’amoxicilline, la diavéridine et la sulfadimidine ont été utilisés au moins une fois dans les fermes échantillonnées, avec une fréquence descendante (de 75.7% à 30.0%). D’après deux scénarios de risque basés sur la comparaison de la période de retrait recommandée sur l’étiquette du produit et celle pratiquée à la ferme, de 14.3% à 44.3% des propriétaires de ferme interrogés n’ont pas respecté la période de retrait recommandée sur l’étiquette au moins une fois au cours des 6 derniers mois, et ce pour au moins un antimicrobien. Les facteurs de risque associés (p<0.05) avec une non-conformité avec la période de retrait recommandée sur l’étiquette pour au moins un des deux scénarios sont les suivants : élever des oiseaux qui n’appartiennent pas tous à des races d’origine asiatique, vacciner contre la bronchite infectieuse, avoir utilisé plus de 6 différents antimicrobiens à la ferme au cours des 6 derniers mois, et utiliser un mélange d’aliments fait maison et commerciaux. Nos résultats soulignent l’importance d’utiliser les antimicrobiens de façon judicieuse et en respectant les temps de retrait officiels, afin de protéger le consommateur contre les risques pour la santé causés par une exposition à des niveaux nocifs de résidus antimicrobiens. / Antimicrobial use in food-animal husbandry is an important public health concern worldwide due to its potential impact on the emergence of drug-resistant microbes and its harmful residues in meat. However, in developing countries, few data are available on farm drug use practices. Therefore a cross-sectional epidemiological study was conducted on broiler chicken farms in Southern Vietnam from June 2011 to August 2011 with the aim of both describing prevalence of antimicrobials added to feed or water at the farm level and ascertaining any associations between farm characteristics and non-compliance of antimicrobial withdrawal times on labels. A convenient sample of 70 broiler farms was surveyed via personal interviews with farm owners using a questionnaire pertaining to farm characteristics and drug use practices. Over the 6-month period prior to the interviews, colistin, tylosin, ampicillin, enrofloxacin, doxycyclin, amoxicillin, diaveridin, and sulfadimidin were used at least once in descending frequency (from 75.7% to 30.0%) by the farms surveyed. Following two risk scenarios based on the comparison of recommended label withdrawal times with actual withdrawal times practiced during this period, between 14.3% and 44.3% of farmers did not comply with on-label withdrawal times for at least one antimicrobial. Risk factors associated (p<0.05) with non-compliance with on-label withdrawal times in at least one risk scenario were: raising birds other than Asian-indigenous bird breeds only, vaccinating against infectious bronchitis, using more than 6 different antimicrobials on a farm during the last 6 months prior to the interview, and mixing home-made and commercial feed. Our results underline the importance of using antimicrobials judiciously and respecting official withdrawal times in order to protect the consumer from the health risks caused by exposure to harmful levels of antimicrobial residues.

The aim of the thesis is to be a comprehensive analysis of the non-competition clause in Czech Labour law. The reason for my research is to examine the non-competition clause, compare and contrast its advantages and disadvantages and to make a conclusion, whether the current legislation meets the requirements of both contracting parties. The thesis is composed of four chapters, each of them dealing with different aspects of non-competition clause, its history and its current problems. Chapter One is introductory and describes the competition in general and in Labour law. The chapter is subdivided into two parts. Part One describes the difference between the competition during the employment relationship and after termination of the employment. Part Two describes the basic terminology and the current state of a non-competition clause. Right of a member state to leave the EU in context of the Brexit Abstract One year has passed since the United Kingdom officially announced its intention to withdraw from the European Union membership. Brexit as this withdrawal is often called means the turning point not only for the view of the concept of the ever closer European integration, but it also presents feasibility of a new option for the EU Member States. It is astonishing that in spite of popular attention to this topic there is an absence of serious academic research dealing with the right of a Member State to withdraw in all its wide aspects. Therefore, this thesis aims at the goal of describing the problem of the withdrawal in the most complex way and not only in the light of the recent Brexit. For understanding the present situation, it seems to be necessary, at least in the limited way, to introduce the right of withdrawal as the external aspect of the sovereignty ultima ratio which even in the historical period before the explicit incorporation in the Lisbon Treaty had to exist. A abstinência ao etanol está associada ao transtorno de ansiedade. Várias estruturas e sistemas de neurotransmissores estão envolvidos na mediação deste efeito, entretanto, tais substratos ainda não foram completamente elucidados. Em modelos animais, estudos mostram aumento significativo da neurotransmissão nitrérgica em resposta à abstinência do consumo crônico de etanol. O Núcleo Dorsal da Rafe (NDR) é uma importante região envolvida na gênese da ansiedade e possui grande quantidade da isoforma neuronial da enzima Sintase do Óxido Nítrico (nNOS). Assim, o objetivo deste estudo foi verificar se a inibição da transmissão nitrérgica no NDR atenua os efeitos ansiogênicos induzidos pela abstinência do etanol. Com essa finalidade, ratos Wistar machos foram submetidos a tratamento crônico (21 dias) ou agudo (1 dia) com etanol 6%(v/v) seguido da suspensão abrupta do tratamento por 48hs. Após as 48hrs os animais receberam uma injeção intra-NDR de L-NAME (100nmoles/0,2µL; inibidor não seletivo da NOS) ou 7-NI (7-Nitro-Indazol; 5 nmoles/0,2µL; inibidor seletivo para nNOS) e 5 min. depois foram testados no Labirinto em Cruz Elevado (LCE). Em resumo, os dados mostraram que a abstinência de 48hs. do tratamento crônico com etanol promoveu efeitos ansiogênicos significativos, evidenciados pela redução do número de entradas e tempo gasto nos braços abertos do LCE, o qual foi atenuado pelo tratamento intra-NDR com L-NAME mas não com 7-NI, sugerindo o envolvimento do NO no NDR na ansiedade induzida pela abstinência do etanol / The ethanol withdrawal is linked to the disorder of anxiety. Several structures and neurotransmitter systems are involved in mediating this effect, however, such substrates have not yet been elucidated. In animal models, studies show significant increase in neurotransmission nitrérgica in response to abstinence from chronic consumption of ethanol. The Dorsal Raphe Nucleus (DNR) is an important region involved in the genesis of anxiety and has large amounts of the enzyme Sintase neuronial isoform of nitric oxide (nNOS). The aim of this study was to verify that the inhibition of transmission in the DNR nitrérgica attenuates the anxiogenic effects induced by ethanol withdrawal. For this purpose, male Wistar rats were subjected to chronic treatment (21 days) or acute (1 day) with 6% ethanol (v / v) followed by the abrupt suspension of treatment for 48 hours. 48hrs after the animals received an intra-DNR L-NAME (100nmoles / 0.2 L, non-selective inhibitor of NOS) or 7-NI (7-Nitro-indazol, 5 nmol / 0,2 L; selective inhibitor for nNOS ) and 5 min. then were tested in the elevated plus-maze (EPM). Data obtained showed that ethanol withdrawal elicited significant \"anxiety-like\" behaviors, as revealed by the decrease in the number of entries into and time spent onto the open arms of the EPM., which was attenuated by treatment with intra-DNR L-NAME but not with 7-NI. These findings are indicative of the involvement of the DNR \"anxiety-like\" behaviors induced by withdrawal of ethanol.

Ilusão da máscara côncava em pacientes em síndrome de abstinência de álcool leve e moderada / Hollow face illusion in patients with mild and moderate alcohol withdrawal syndrome.

O uso lesivo de bebidas alcoólicas é o terceiro maior fator mundial de risco de doenças e incapacitação. O álcool pode causar várias alterações no Sistema Nervoso Central (SNC), dentre elas, a diminuição do processamento de informações visuais. Alguns estudos avaliam as alterações nos processos perceptuais durante a Síndrome de Abstinência do Álcool (SAA) por meio da ilusão visual da máscara côncava. Essa ilusão exemplifica o fenômeno ilusório de inversão visual da profundidade de uma máscara humana oca, que é percebida como convexa. Foi encontrado um possível desequilíbrio entre os componentes bottom-up e top-down da percepção visual provocado pelo álcool, capaz de prejudicar a capacidade do indivíduo de perceber essa ilusão. Entretanto, a presente pesquisa investiga uma hipótese alternativa, e sugere que a redução na frequência de respostas de inversão pode resultar de mudanças de critérios para emitir as repostas e não em alterações perceptuais. Para isso, foram utilizados os parâmetros da Teoria da Detecção de Sinal (TDS) aplicada à psicofísica. Participaram da pesquisa 20 indivíduos saudáveis, 20 com SAA leve e 20 com SAA moderada. Eles realizaram duas tarefas experimentais de observação monocular dos lados côncavo e convexo, alternadamente, de uma máscara da face humana de tamanho reduzido. Na tarefa de confidence rating, foram julgadas a concavidade ou convexidade da máscara e o grau de certeza nas respostas, com certeza ou com dúvida. Na tarefa de escolha forçada entre duas alternativas (2AFC), as máscaras foram apresentadas aos pares alternados e o participante identificava o lado côncavo. A análise dos resultados revelou que os indivíduos com SAA moderada apresentaram maiores escores para os índices de sensibilidade R-index, Az, e da, na tarefa de confidence rating, apontando para uma capacidade significativamente maior de identificação e discriminação dos lados côncavo e convexo da máscara. Isto reflete um prejuízo significativo na capacidade destes indivíduos em perceber a ilusão da máscara côncava. Não houve diferença entre os grupos de indivíduos saudáveis e com SAA leve. Entretanto, os grupos com SAA apresentaram um maior grau de certeza em seus julgamentos comparativamente ao grupo controle. Resultados semelhantes para o grupo com SAA moderada foram encontrados na tarefa de escolha forçada para os índices d e taxa de acerto, indicando maior capacidade desses indivíduos em discriminar os dois lados da máscara. Todos os participantes apresentaram critérios de decisão moderados na tarefa de confidence rating. Os indivíduos saudáveis, na tarefa de escolha forçada, se mostraram tão capazes quanto os com SAA moderada na discriminação dos dois lados da máscara. As análises apontam para um possível desequilíbrio, já relatado em estudos anteriores, entre os componentes bottom-up e top-down da percepção visual, provocado pelo álcool, que impede o SNC de corrigir hipóteses perceptuais ambíguas. / The harmful use of alcoholic beverages is the third largest worldwide risk factor for illness and disability. Alcohol can cause several changes in the Central Nervous System (CNS), among them, the decrease in the processing of visual information. Some studies evaluate changes in perceptual processes during Alcohol Withdrawal Syndrome (AWS) through the hollow face illusion. Its exemplifies the illusory visual depth inversion of a hollow human mask, which is perceived as convex. A possible imbalance was found between the bottomup and top-down components of visual perception caused by alcohol, which could impair the individual\'s ability to perceive the illusion. However, the present research investigates an alternative hypothesis, and suggests that the reduction in the frequency of inversion responses may result from changes in the criteria to issue responses rather than on perceptual changes. The parameters of the Signal Detection Theory (SDT) applied to psychophysics allow this analysis. Twenty healthy subjects, 20 with mild AWS and 20 with moderate AWS participated in the study. They performed two experimental tasks of monocular observation of concave and convex sides, alternately, of a reduced size human face mask. In the task of confidence rating, the concavity or convexity of the mask and the degree of certainty in the answers were judged: certainly or with doubt. In the task of forced choice between two alternatives (2AFC), the masks were presented in alternating pairs and the participant identified the concave side. The analysis of the results revealed that individuals with moderate AWS presented higher scores for the sensitivity index scores R-index, Az, and da, pointing to a significantly greater capacity of identification and discrimination of the concave and convex sides of the mask. This reflects a significant impairment in the ability of these individuals to perceive the hollow face illusion. There was no difference between healthy individuals and with mild AWS. However, all groups with AWS presented a greater degree of certainty in their judgments compared to the control group. Similar results for the group with moderate AWS were found in the task of forced choice to the indices d and hit rate, indicating a greater ability of these individuals to discriminate both sides of the mask. Healthy subjects, on the task of forced choice, showed themselves to be as capable as those with moderate AWS in discriminating both sides of the mask. However, it supposed that these results were from unexpected strategies used by these individuals in their judgments, impairing the internal validity of the results, and represented an important research bias. The analyzes point to a possible imbalance, already reported in previous studies, between the bottom-up and top-down components of visual perception, caused by alcohol, which inhibits the CNS from correcting ambiguous perceptual hypotheses.

Estudo de depleção residual de benzoato de emamectina em filés de peixe e estimativa de período de carência / Depletion study of emamectin benzoate in fish fillet and determination of withdrawal period

O benzoato de emamectina, um antiparasitário da classe das avermectinas, é amplamente utilizado em diversos países para o tratamento de infecções causadas por helmintos intestinais e ectoparasitas, os quais estão relacionados a grandes prejuízos na produção aquícola. Apesar de sua eficácia ser conhecida na aquicultura, seu uso não está regulamentado no Brasil. Diante da demanda nacional por uma maior variedade de medicamentos veterinários regulamentados para uso em espécies aquícolas, o presente projeto visou estimar um período de carência para o antiparasitário benzoato de emamectina, a partir de um estudo experimental de depleção residual desse fármaco em tilápia (filé com pele em proporção natural), a espécie de maior importância comercial para a produção aquícola no Brasil. Para tanto, fez-se necessárias as seguintes etapas: (i) incorporar o benzoato de emamectina na ração através da técnica de revestimento polimérico em equipamento de leito fluidizado; (ii) desenvolver e validar métodos analíticos para a determinação do benzoato de emamectina na ração e em filé de tilápia empregando cromatografia líquida de alta eficiência acoplada à espectrometria de massas (LC- MS/MS); e (iii) realizar um ensaio com tilápias para avaliar a depleção residual de emamectina B1a no filé. O revestimento da ração com 1,5% de etilcelulose permitiu incorporar o benzoato de emaectina na ração sem alterar a estrutura física da mesma, o que torna o processo vantajoso, visto que a ração mantém suas características nutricionais e de flutuabilidade. Além disso, o processo foi homogêneo (CV de 2,1%) e não apresentou taxa de lixiviação quantificável do fármaco para a água, contribuindo para uma maior segurança em relação à dose de tratamento a ser administrada e em relação ao risco potencial ao meio ambiente. A emamectina B1a foi extraída da ração por processo sólido-líquido e do filé de tilápia pelo método QuEChERS modificado. Os métodos foram avaliados mediante os seguintes parâmetros: seletividade, curva analítica, linearidade, precisão, exatidão, limite de detecção (LD) e de quantificação (LQ). Todos os parâmetros avaliados se encontraram conformes às recomendações dos guias de validação tomados como referências, qualificando os métodos como adequados para os objetivos propostos no presente trabalho. Para avaliar a depleção de emamectina B1a no filé de tilápia, os peixes receberam o fármaco via ração em dose de 50 ?g/kgPV/dia durante sete dias consecutivos. As curvas de depleção residual de emamectina B1a se ajustaram ao modelo exponencial de primeira ordem. Tomando como referência o limite máximo de resíduo de 100 ?g/kg recomendado pelo Codex Alimentarius, não houve a necessidade de se propor um período de carência mínimo para o abate dos peixes, considerando as condições experimentais empregadas. / Emamectin benzoate, an antiparasitic of the class of avermectins, it is widely used in several countries for the treatment of infections caused by intestinal helminths and ectoparasites, which are related to great losses in aquaculture production. Although its known effectiveness for aquaculture, its use is not regulated in Brazil. The national demand for a greater variety of regulated veterinary drugs for use in aquaculture species,the current project that aimed to evaluate a withdrawal period for the antiparasitic emamectin benzoate, based on an experimental study of residual depletion of this drug in tilapia ( fillet with skin in natural proportion) the species of major commercial importance for aquaculture production in Brazil. To do so, it was necessary to: (i) incorporate emamectin benzoate into the feed through the polymer coating technique in fluidized bed equipment; (ii) to develop and validate the analytical methods for the determination of emamectin benzoate in the feed and in tilapia fillet using high-performance liquid chromatography coupled to mass spectrometry (LC-MS / MS) and (iii) perform a tilapia test to evaluate the depletion of emamectin B1a in fillet. The feed coating with 1.5% ethylcellulose allowed to incorporate the emaectin benzoate into the feed without altering the physical structure, which makes the process advantageous, since the feed retains its nutritional and buoyancy characteristics. In addition, the process was homogeneous (CV 2,1%) and did not present quantifiable leaching rate of the drug into the water, contributing to a greater safety in relation to the dose of treatment to be administered and in relation to potential risk to the environment. Emamectin B1a was extracted from the feed by solid-liquid process and from tilapia fillet by the modified QuEChERS method. The methods were evaluated using the following parameters: selectivity, analytical curve, linearity, precision, accuracy, limit of detection (LD) and quantification (LQ). All the evaluated parameters were in accordance with the recommendations of the validation guides taken as references, qualifying the methods as adequate for the objectives proposed in the present work. To evaluate the depletion of emamectin B1a tilapia fillet, the fish received the drug via feed at a dose of 50 ?g / kgPV / day for seven consecutive days. The residual depletion curves of emamectin B1a were fitted to the first order exponential model. Taking into account the maximum residue limit of 100 ?g / kg recommended by the Codex Alimentarius, there was no need to propose a minimum withdrawal period for the slaughtering of fish, considering the experimental conditions used.

Após as 48hrs os animais receberam uma injeção intra-NDR de L-NAME (100nmoles/0,2µL; inibidor não seletivo da NOS) ou 7-NI (7-Nitro-Indazol; 5 nmoles/0,2µL; inibidor seletivo para nNOS) e 5 min. depois foram testados no Labirinto em Cruz Elevado (LCE). Em resumo, os dados mostraram que a abstinência de 48hs. do tratamento crônico com etanol promoveu efeitos ansiogênicos significativos, evidenciados pela redução do número de entradas e tempo gasto nos braços abertos do LCE, o qual foi atenuado pelo tratamento intra-NDR com L-NAME mas não com 7-NI, sugerindo o envolvimento do NO no NDR na ansiedade induzida pela abstinência do etanol / The ethanol withdrawal is linked to the disorder of anxiety. Several structures and neurotransmitter systems are involved in mediating this effect, however, such substrates have not yet been elucidated. In animal models, studies show significant increase in neurotransmission nitrérgica in response to abstinence from chronic consumption of ethanol. The Dorsal Raphe Nucleus (DNR) is an important region involved in the genesis of anxiety and has large amounts of the enzyme Sintase neuronial isoform of nitric oxide (nNOS). The aim of this study was to verify that the inhibition of transmission in the DNR nitrérgica attenuates the anxiogenic effects induced by ethanol withdrawal. For this purpose, male Wistar rats were subjected to chronic treatment (21 days) or acute (1 day) with 6% ethanol (v / v) followed by the abrupt suspension of treatment for 48 hours. 48hrs after the animals received an intra-DNR L-NAME (100nmoles / 0.2 L, non-selective inhibitor of NOS) or 7-NI (7-Nitro-indazol, 5 nmol / 0,2 L; selective inhibitor for nNOS ) and 5 min. then were tested in the elevated plus-maze (EPM). Data obtained showed that ethanol withdrawal elicited significant \"anxiety-like\" behaviors, as revealed by the decrease in the number of entries into and time spent onto the open arms of the EPM., which was attenuated by treatment with intra-DNR L-NAME but not with 7-NI. These findings are indicative of the involvement of the DNR \"anxiety-like\" behaviors induced by withdrawal of ethanol.

Medida de topografia de superfície usando a técnica de deslocamento de fase / Measurement of Surface Topography using the Phase Shift Technique.

Neste trabalho, medimos o perfil 3D de superfícies (microtopografia) utilizando uma técnica de interferometria óptica: Phase-Shi,ftzng (Deslocamento de Fase). Utilizamos um interferômetro do tipo Twyman-Green para produzir fi.guras de interferência da superfície analisada. Essas imagens foram armazenadas usando-se uma câmera CCD ligada à um microcomputador. Para obter a microtopografia, calculamos o Mapa de Fase a partir das imagens digitalizadas usando um programa de microcomputador. Posteriormente um outro programa removeu a ambiguidade da função tangente (unwrapping), pela Técnica do Autômato Celular, usada no cáiculo do Mapa de Fase. Então efetuamos os cálculos para determinar a microtopografia da superfície. Depois fizemos a análise da microtopografia, levantando informações relevantes para a sua caracterização. Analisamos objetos com alta refletividade (espelhos planos e redes de Ronchi) e obtivemos bons resultados. Também comparamos alguns dos resultados obtidos com a técnica de Deslocamento de Fase com os resultados obtidos pela análise de Franjas de Igual Espessura. / In this work we measured the 3D profile of surfaces (microtopography) using a optical interferometric technique: Phase-Shifting. We used a interferometer of type Twyman-Green to produce interferograms from analyzed surface. These images was captured using a CCD camera that was linked to a microcomputer. To obtain a microtopography, we calculated the Phase Map using the digitalized images and a software of microcomputer. Then another program removed the wrapping of tangent fuction, using the Cellular-Automata Technique, that was used to calculate the Phase Map. So we calculated the microtopography of the surface. After we did the analyses of the microtopography, find out some important informations of its description. We studied objects with high reflectivity (plane mirrors and Ronchi ruting) and we obtained good results. Also we compared some results with that obtained by analyses of Fringes of Equal Thickness\'
