Outra trata se a perspectiva internacional sindical em curso pode contribuir para a manutenção ou novas conquistas para os trabalhadores. Tais questões são vitais, especialmente em um mundo de intensa e ainda crescente globalização em todos os setores sociais. Evidenciou-se a indicação de respostas positivas às dúvidas acima, sugerindo que, não obstante a crise atual, também, do movimento sindical internacional, trata-se de uma estratégia exequível e mesmo indispensável ao trabalhador. A metodologia seguida, além das referências teóricas, lançou mão, basicamente, da pesquisa de campo por meio do estudo de fontes primárias e entrevistas junto aos principais protagonistas da trajetória internacional que em 2014 completa 30 anos. / The present dissertation is focused on the experience of the international trade union organization of workers since Mercedes-Benz Brazil in São Bernardo do Campo. I highlight since because the very nature of the process led to the global connection with employees located at industrial units in foreigner countries of the same transnational company. Among several political players as well as in the academic world, there is the controversial issue about the importance of the union movement not only in Brazil, but also internationally. In other words, the main issue is about the effectiveness of international trade unionism in the presence of the capital hegemonic globalization. On one hand concerns if it is possible that this practice means an alternative to the hegemonic model, even being, nowadays, in an incipient stage. On the other hand if the international trade union perspective can contribute to the maintenance or new achievements of the workers. Such issues are strongly important, mainly in a world of intense and growing globalization of all sectors of society. There was an indication of positive responses to both questions above, suggesting that, despite the current worldwide crisis, involving the international trade union movement as well, is an feasible strategy and even essential to the worker. The methodology, besides the theoretical references, dealt with field research collecting primary data and interviews with the main protagonists of the international trajectory, which completes 30 years in 2014.

Esta investigación tuvo como objetivo determinar el impacto de la Implementación del Sistema de Costos ABC en el crecimiento de la rentabilidad en la empresa Inversiones 2 A entre el año 2016 y 2017, el tipo de investigación fue aplicada, de nivel explicativo y el diseño cuasi experimental con series cronológicas con un solo grupo. Los principales resultados fueron la reducción del costo directo por kilogramo en la fabricación de la Harina de Maca Gelatinizada en un 24%, después de la implementación del Sistema de Costos ABC, siendo esta reducción significativa (p<=0,05). Así mismo se disminuyó el costo indirecto de fabricación por kilogramo en un 20%, dichas comparaciones se realizaron entre los años 2016 y 2017, siendo este último donde se realizó la implementación del Sistema de Costos ABC

Elaborou-se um estudo com o intuito de estabelecer uma avaliação quantitativa dos inter-relacionamentos entre as atividades. Pretendeu-se evidenciar como uma alteração em uma das atividades possa alterar o relacionamento entre todas as atividades, ainda que de forma quantitativa. Este método foi testado em uma empresa de bebidas, mais especificamente em seu setor de logística, pois este apresentava uma grande quantidade dos requisitos necessários para um teste piloto. Esta aplicação experimental permitiu a validação do método referido e o sistema de informações de custos que mais se adequou foi o ABC. Ressaltamos porém que os sistemas de custeio tradicionais, a teoria das restrições o método UP e o Target Costs também foram considerados como possíveis no fornecimento de informações para o teste do método. O método ao ser implementado, nesta empresa do ramo de bebidas, mostrou-se eficiente, porém será necessário o apoio de um melhor sistema de informações de custos, notadamente nos seus aspectos de envolvimento de pessoas responsáveis pelas informações e pelo direcionamento filosófico da empresa. / It was elaborated a study which establishes a quantitative valuation of the interrelationships among activities. It was intended to establish how a change on one of the activities could change the relationship among all activities, even though in a quantitative form. This method was testified in a beverage company, more specifically at it logistic area, because it presented a large quantity of the necessary requirements for a pilot test. This experimental application allowed the acknowledgement of the mentioned method and ABC was the information costs system more appropriate, however it might be pointed out that the Traditional costs system, the Theory of constraints, the UP costing method and the Target Costing also were considered as possibilities to supply information for the test. The method implementation, in a beverage company, showed it efficiency, but it will be necessary the support of a better costs information system, regarding its aspects of envolvement by information responsable people and philosophic company tendency.

L’Adrénoleucodystrophie liée à l’X est une maladie neurodégénérative rare due à des mutations dans le gène ABCD1. Ce gène code un demi-transporteur ABC peroxysomal, impliqué dans l’importation d’acides gras à très longue chaîne. Deux autres demi-transporteurs sont localisés dans la membrane peroxysomale : ABCD2 et ABCD3. La surexpression d’ABCD2 permet de compenser la déficience en ABCD1, ouvrant ainsi des perspectives thérapeutiques. Dans cette optique, l’objectif principal de ma thèse était d’étudier la fonction et la structure d’ABCD2, et plus largement des transporteurs ABC peroxysomaux.Les demi-transporteurs doivent au minimum se dimériser pour constituer un transporteur fonctionnel. Leur dimérisation alternative pourrait moduler leur spécificité de substrat. Afin de tester cette hypothèse, nous avons réalisé des constructions plasmidiques codant différents dimères chimériques, dont la fonctionnalité a été vérifiée par transfection transitoire dans deux modèles cellulaires (fibroblastes humains et levures). D’après nos résultats, ABCD1 et ABCD2 seraient fonctionnels quel que soit leur agencement dimérique. De plus, comme d’autres transporteurs ABC, les transporteurs ABC peroxysomaux pourraient s’oligomériser. En utilisant différentes techniques biochimiques (co-immunoprécipitation, sédimentation sur gradient de sucrose et électrophorèse en conditions natives), sur un modèle cellulaire surexprimant ABCD2-EGFP, nous démontrons qu’ABCD2-EGFP interagit avec ABCD1 et ABCD3, et que les transporteurs ABC peroxysomaux sont capables de s’oligomériser. Il reste désormais à déterminer les facteurs qui contrôlent cette oligomérisation et comprendre la valeur fonctionnelle de ces interactions. / X-linked Adrenoleukodystrophy (X-ALD) is a rare neurodegenerative disease caused by deficiency of the peroxisomal half-transporter ABCD1, implicated in very long chain fatty acids import. Two additional half-transporters are located in the peroxisomal membrane: ABCD2 and ABCD3. Over-expression of ABCD2 is known to compensate for ABCD1 deficiency, making ABCD2 a therapeutic target for X-ALD treatment. In this context, the main objective of my thesis was to investigate the function and the structure of ABCD2, and more broadly, of peroxisomal ABC transporters.Half-transporters must at least dimerize to form a functional transporter. Alternative dimerization could modulate substrate specificity. In order to test this hypothesis, we engineered plasmidic constructs encoding chimeric ABCD dimers, whose functionality has been evaluated by transient transfection in two cell models (human fibroblasts and yeasts). Our results show that, ABCD1 and ABCD2 are functional whatever their dimeric organization. Besides, like other ABC transporters, peroxisomal ABC transporters could oligomerize. By using a multi-technical approach (co-immunoprecipitation, velocity sucrose gradient and native polyacrylamide gel electrophoresis experiments) on stably transfected hepatoma cells expressing ABCD2-EGFP, we demonstrate that ABCD2-EGFP interacts with ABCD1 and ABCD3, and that peroxisomal ABC transporters oligomerize. The perspectives will consist in determining which factors control the oligomerization process and understanding the functional value of these interactions.

The focus of this study was to determine the impact of activity-based costing (ABC) on organizational performance. ABC can support the strategic management process and provide significant benefit to organizations. Empirical research has provided mixed results of ABC success, which has been the center of controversy since the 1980s. This examination utilized the contingency theory as a theoretical basis for the study. The researcher expected to find that ABC organizations succeed or fail to improve performance due to how well they fit their strategies, structures, and managerial accounting practices to the complex business environment. This study addressed the fundamental question does ABC improve organizational performance? The study found that management accounting practices could only significantly predict organizational performance, as measured by market price, EPS, and asset turnover. The results indicated that ABC organizations have higher asset turnover than non-ABC organizations. However, non-ABC organizations have higher market price and EPS than ABC organizations. The findings indicated no statistically significant differences in operating performance when either ABC or non-ABC accounting practices are used with a low-cost strategy. Finally, the results found no statistically significant differences in operating performance when ABC organizations use a strategy other than a low-cost strategy.

The study of signaling pathways that modulate multidrug resistance The theme of cancer cell resistance to anti-cancer drugs including the common mechanisms of resistance development and the theory of cancer stem cells was introduced in the Introduction to the doctoral thesis. The theoretical part was focused more deeply on the two topics - the role of ATP-binding cassette (ABC) transport proteins and chromosomal abnormalities in the development of cancer chemoresistance. The possible therapeutic potential for the treatment of cancer was stressed for both topics. The Results were composed of the commentaries on the five published works, which the author of the thesis conducted as the main author. The first work brought the evidence supporting the hypothesis of the existence of ABC gene expression profiles (signatures), which are common to multiple types of tumors and are associated with significant clinical consequences. These general ABC gene expression profiles could possibly form a new hallmark of cancer. The second work studied more closely a group of acute myeloid leukemia patients, who did not achieve complete cytogenetic remission after two attempts to maintain remission of the malignant disease. The main topic of this diploma thesis is ARE (resistance) proteins from the ABC-F family of the second class of ABC proteins. ARE proteins confer resistance to antibiotics that bind to a large ribosomal subunit and therefore inhibit proteosynthesis. One of the ARE proteins is the Lmr (C) protein, which is part of the linkomycin biosynthesis cluster of Streptomyces lincolnensis, and according to new results, Lmr (C) does not have to be just resistant protein but may have also regulatory function. We decided to study Sco0636, the closest homologue to Lmr (C) in Streptomyces coelicolor, which is a model organism in the study of secondary metabolism. Thanks to the production of color pigments, it is possible to monitor the effect of ARE proteins on secondary metabolism directly on the plates. I prepared the deletion mutant and the strain with constitutive expression of sco0636, and observed the effect on the phenotype. I followed the production of a blue asset and set a minimum inhibitory concentration to selected antibiotics, which bind to the ribosome. I have found that Sco0636 gives high resistance to tiamulin and so it has been named TiaA. The deletion of gene sco0636 accelerated production of actinorodine, and constitutive expression of this gene slowed down production. Keywords: ABC proteins,...

Purpose – The purpose of this study is to increase the knowledge of how the inventory can becontrolled and managed when purchasing in conjunction with an ABC-classification. To fulfilthe purpose, two research questions have been formulated: [1] How can the ABC classification be used to manage the purchasing process? [2] How can the ABC classification contribute to improved warehouse service? Method – The study was initiated with a feasibility study in order to shed light on the problemarea and its context which the study intends to investigate further. The feasibility study incombination with a literature review contributed to the formulation of the purpose and researchquestions. The study is designed in accordance with a case study with a singular analysis unit, in this case the purchasing unit in a business system. Furthermore, data collection wasgenerated via literature study, interview, and questionnaire. The interviews and thequestionnaires contributed to qualitative primary data which were intended to illustrate theusers’ knowledge and use of the studied phenomenon. Findings – It was found in the study that ABC classification is an effective method fordifferentiating purchasing and inventory management. This study demonstrates that aninterconnection of ABC classification and inventory service level is the most effective approachin order to control and minimize the occurrence of shortages, resulting in improved warehouseservice. Implications – The subject areas, which first and foremost are ABC classification andinventory management, are thoroughly researched as proven by existing literature, hencegenerating new knowledge to the fields are considered to be challenging. Despite the givencircumstances, this study adds knowledge to a certain extent, as the study presents andstrengthens existing research regarding how a combination of an ABC classification and acertain degree of service level has been shown to streamline the purchasing process andimprove the warehouse service as well as the inventory management. Limitations – This study only includes the users of the business system and the purchasingmodule provided by the case company, which can be considered to affect the generalizability ofthe study. The fact remains, however, that generalizability should also apply in other systemsin connection with purchasing, since the starting point of the theory and its area of applicationis so well known and generally accepted. För att besvara syftet har det brutits ner i två frågeställningar: [1] Hur kan ABC-klassificering användas för att styra inköp? [2] Hur kan ABC-klassificering bidra till förbättrad lagerservice? Metod – Studien initierades med en förstudie för att belysa det problemområde och desskontext, vilken studien ämnar undersöka. Förstudien i kombination med litteraturstudie bidrogtill formulering av studiens syfte och frågeställningar. Studien är av enfallsdesign med enanalysenhet, i det här fallet inköpsmodulen i ett affärssystem. Vidare genereradesdatainsamling via litteraturstudie, intervju samt enkät. I samband med intervju och enkäterhölls kvalitativa primärdata som avsåg åskådliggöra användarnas kunskap och användningav det studerade fenomenet. Resultat – ABC-klassificering visar sig vara en effektiv metod för differentiering av inköp ochlagerstyrning. Den här studien påvisar att en sammankoppling av artikelklassificering ochlagerservicenivå är det mest effektiva tillvägagångssättet i syfte att kontrollera och minimerauppkomsten av bristtillfällen som ger upphov till förbättrad lagerservice. Implikationer – Ämnesområdet kring ABC-klassificering och servicenivå är väl utforskatinom befintlig litteratur, vilket bidrar till mindre utrymme att generera ny kunskap till området.Trots given omständighet tillför studien i viss utsträckning kunskap, då genomförd studieframlägger och stärker befintlig forskning då en kombination av ABC-klassificering ochservicenivå har visat sig effektivisera inköpsarbetet. En sådan kombination visar sig i empirinha minimerat antal bristtillfällen och på så vis förbättrat lagerservicen. Begränsningar – Studien inkluderar enbart användare av inköpsmodulen som fallföretagettillhandahåller, vilket kan påverka studiens generaliserbarhet. Faktum kvarstår dock attgeneraliserbarhet även bör gälla i andra fall då inköp görs eftersom utgångspunkten för teorinoch dess användningsområde är så pass känt och allmänt godtaget.
