Dans les savanes d'Afrique de l'Ouest, les moustiques Anopheles coluzzii et An. gambiae, qui transmettent le paludisme, pullulent en saison des pluies et disparaissent en saison sèche. Les mécanismes permettant le maintien des populations d'Anophèles d'une année sur l'autre restent inconnus et difficiles à mettre en évidence sur le terrain. Au cours de ce travail, nous avons étudié, par des approches comparatives et expérimentales, la plasticité physiologique et morphologique des femelles An. coluzzii et An. gambiae exprimée en réponse aux conditions déshydratantes de la saison sèche. Des analyses métabolomiques, protéomiques, morphométriques et respirométriques, nous ont permis de mettre en évidence une grande variabilité des profils exprimés entre les espèces et entre les différentes populations d'An. coluzzii. / In dry savannahs of West Africa, the malarial mosquitoes Anopheles coluzzii and An. gambiae swarm during the rainy season but almost disappear at the onset of the dry season. Interestingly, the ecological and physiological bases enhancing the survival of these insects during the desiccating conditions of the dry season remain unknown, and hard to understand with field studies. In the present work, we conducted experimental and comparative studies to analyse the physiological and morphological plasticity of female An. coluzzii and An. gambiae mosquitoes in response to the environmental conditions of the dry season. Metabolomic, proteomic, morphometric and gas exchange analyses highlighted a wide range of responses to the dry season conditions in these species, and enables distinguishing the two considered species.

Parameters that characterizes quality internal building environment come forth these days. One of the main reasons for the choice of the topic of this thesis is to ensure the quality of the internal environment and well-being during the usage of the buildings. Prevention of the excessive wall dampness is one of the basic conditions of a healthy and quality building environment. The thesis verifies reliability and compares the efficiency of the selected damp-proof materials and its joints. The first part deals with the influences that are reasons for the dampness of the buildings, earmark of an interest area, introducing of the valid legislation focused on the building redevelopment, consideration and evaluation of the building constructions in light of the dampness. The next part of the thesis sets main and partial targets and consecutive steps leading to fulfilment of the targets. Selection of the appropriate redevelopment procedures, materials and joints of the selected materials selected for the experiment realization follows. As this area is relatively large, we selected mechanical redevelopment methods reaching the walls and three most commonly used materials and its joints within the framework of the selected methods. There are three main steps set in the thesis: preparation of the methodology and experiment realization, measurement optimization and measurement within the laboratory and on the real buildings, comparison and evaluation of the measured values. Experimental part of the thesis took place between the years 2008 - 2011. Experiment realization comprises of material acquisition, building of the test samples and application of the single redevelopment measures. The next step includes measurement on the individual samples on the fixed measuring points. Measurements are evaluated through the mathematical statistical methods. Efficiency of the particular materials are compared and evaluated.

The effects of lime on the decomposition of buried human remains. A field and laboratory based study for forensic and archaeological application.

The inclusion of lime in burials is observed in historical and archaeological records, in contemporary mass graves and forensic cases. Clearly there are controversies within the literature and there is a general misconception of the effects of lime on decomposition. Recent casework in Belgium and the UK involving the search for human remains buried with lime, have demonstrated the need for a more detailed understanding of the effect of different types of lime on cadaver decomposition and its micro-environment. Field and laboratory experiments using pigs as human body analogues were undertaken to obtain a better understanding of the taphonomic processes that govern lime burials. The changes observed in the experiments were related back to archaeological parallels in which white residues have been found. The combined results of these studies demonstrate that despite conflicting evidence in the literature, hydrated lime and quicklime both delay the initial stages of the decay process but do not arrest it completely. The end result is ultimately the same: skeletonisation. Furthermore this study stresses the importance of the specific microenvironment in taphonomic research and highlights the need for chemical analysis of white residues when encountered in a burial. Not all white powder is lime. White residue could be identified as calcium carbonate, building material, body decomposition products, minerals or degraded lead. This study has implications for the investigation of clandestine burials and for a better understanding of archaeological plaster burials. Knowledge of the effects of lime on decomposition processes also have bearing on practices involving the disposal of animal carcasses and potentially the management of mass graves and mass disasters by humanitarian organisation and DVI teams. / Arts and Humanities Research Council (AHRC) and the University of Bradford

Influence of Physiological State, Prolonged Dry Storage, and Passage through Simulated Digestion on the Survival and Gene Expression of Salmonella enterica sv. Tennessee

Salmonella enterica serotypes have been linked to outbreaks associated with low water activity foods. The ability of biofilm forming pathogens, such as Salmonella, to survive thermal and chemical processes is improved; it is unclear if biofilms will also improve survival to desiccation and gastric stresses. The purpose of this study was to quantify the effect of physiological state (planktonic versus biofilm) and prior exposure to desiccation on Salmonella survival and gene expression after passage through an in-vitro digestion model. Cells of Salmonella enterica serotype Tennessee were deposited onto membranes for planktonic cells or on glass beads to create biofilms. The cells were subsequently dried at room temperature and stored in dried milk powder (aw = 0.3) for up to 30 days. Salmonella survival was quantified by serial dilution onto brilliant green agar before desiccation, after desiccation, after 1-day storage and after 30-day storage. At each sampling both physiological states were tested for survival through a simulated gastrointestinal system. RNA was extracted at the identical time points and relative gene expression determined for genes associated with stress response (rpoS, otsB), virulence (hilA, hilD, invA, sipC) and a housekeeping gene 16S rRNA using quantitative real-time PCR. The physiological state and length of storage effected the survival and gene expression of Salmonella within the desiccated milk powder environment and after passage through an in-vitro digestion system (p<0.05). Larger numbers of S. Tennessee were recovered by plate counts for biofilm cells, compared to planktonic cells. However, the numbers of 16S rRNA gene copies were not significantly different suggesting entry of S. Tennessee into a viable but non-culturable state. Prolonged storage in dry milk powder was not associated with increased cross-protection to gastric stress. Increased expression of stress response genes rpoS and otsB correlated with survival, indicating cross protection of low water activity and acid stress. Increased expression of virulence-associated genes was seen in cells exposed to short periods of dry storage, suggesting an increased virulence potential. / Master of Science in Life Sciences

Ce mémoire reconstitue l’histoire des théories, modèles et hypothèses qui ont été formulés, principalement en archéologie, afin d’expliquer la naissance de l’agriculture qui correspond, du point de vue chronologique, à la transition (ou révolution) néolithique. Ces schèmes explicatifs sont décrits chronologiquement depuis la naissance de l’archéologie préhistorique, dans la première moitié du XIXe siècle, jusqu’à maintenant. Ils sont classifiés en fonction des principales écoles qui ont joué un rôle prédominant en archéologie préhistorique depuis son origine, soit : l’évolutionnisme multilinéaire, l’école culturelle-historique, le processualisme, le néodarwinisme et le postprocessualisme. Les théories spécifiques (dites de « niveau mitoyen » ou « régionales ») qui ont été avancées par ces écoles sont présentées dans leur ordre chronologique, soit (principalement) : les théories de la dessiccation ou de l’oasis, des flancs de colline, de la périphérie, du stress démographique, du festin compétitif, de la révolution des symboles, etc. Ce mémoire pose enfin les jalons d’une théorie multifactorielle qui intègre ou synthétise les principaux facteurs qui ont pu influer sur la naissance de l’agriculture et sur la transition néolithique. / This dissertation reconstructs the history of theories, models and hypotheses which have been proposed in order to explain the birth of agriculture, that is the neolithic transition or revolution. These explanatory schemes are described chronologically from the birth of prehistoric archaeology, in the first half of the 19th century, until today. They are classified according to the main schools which have played a leading role in prehistoric archaeology since its birth: multilinear evolutionism, the culture-historical school, processualism, neodarwinism and postprocessualism. The specific theories (also called “middle-range” or “regional”) which have been proposed by these schools are presented the chronological order: the theories of desiccation, oases, hilly flanks, periphery, population stress, competitive feasting, revolution of symbols, etc. Finally, this dissertation lays the ground for a multifactorial theory integrating or synthesizing the main factors which may have influenced the birth of agriculture and the neolithic transition.

Pleurostima purpurea (Velloziaceae) é uma espécie rupícola encontrada em afloramentos rochosos do estado do Rio de Janeiro, sudeste do Brasil. Por apresentar notável capacidade de tolerar a dessecação vegetativa, este estudo teve o intuito de esclarecer detalhes sobre a estratégia ecofisiológica utilizada por tais plantas frente à desidratação do ambiente e as vantagens associadas ao processo. Para tanto foi analisada a dinâmica de trocas gasosas, o conteúdo de pigmentos fotossintéticos e o conteúdo relativo de água de tecidos foliares durante os processos de dessecação e reidratação de indivíduos adultos cultivados. Comportaram-se como homeohídricas típicas economizadoras de água sob condição de seca moderada que, quando agravada, assumiram o comportamento de pecilohídricas e peciloclorófilas. A suspensão da irrigação provocou o fechamento estomático sob conteúdo relativo de água foliar acima de 90%, levando a restrições sobre a transpiração e assimilação líquida de carbono até o estabelecimento da anabiose. Durante este processo, houve um atraso na diminuição do CRAfoliar em relação ao CRAplanta-solo. A degradação das clorofilas acompanhou a diminuição do CRAfoliar, o qual alcançou o valor médio mínimo de 17% sem incorrer na abscisão das folhas, enquanto sua resíntese somente se iniciou após a total reidratação dos mesmos tecidos. Durante todo este processo o conteúdo de carotenóides manteve-se estável. 12 horas de reidratação do solo de cultivo foram suficientes para a retomada respiratória. Um balanço positivo de CO2 foi observado a partir da 36ª hora de reidratação quando o grau de abertura estomática passou a ser maior do que o observado para o grupo controle. Comparando-se com as condições iniciais, 84 horas de reidratação foram suficientes para a recuperação fotossintética mesmo com uma recuperação apenas parcial do conteúdo de clorofilas. Assim como os tecidos foliares, suas raízes também devem ser tolerantes à dessecação. / Pleurostima purpurea (Velloziaceae), a rupicolous flowering species found in rock outcrops in Rio de Janeiro state, southeastern Brazil, show notable capacity to endure vegetative desiccation. This study had the intent to show more details about the ecophysiological strategy used by these plants facing a drying ambient and the advantages associated with the process. For this, it was analised the gas exchanges, photosynthetic pigments content, and relative water content of leaf tissues during desiccation and rehydration of cultivated individuals. They behaved typicaly as homeohydrics water savers under moderated drought condition that, when aggravated, prosecute with poikilohydric and poikilochlorophyllous behavior. After watering suspension, stomatic closing was observed under a relative leaf water content up to 90% that resulted in gas exchanges restrictions until the anabiosis. During this process, it was observed a delay in the leaf relative water content (RWCleaf) decrease comparing to the plant-soil relative water content (RWCplant-soil). The chlorophylls degradation acompanished the RWCleaf decrease, which achived the minimum average value of 17% without incur in leaves abscission, while its resynthesis just begun after the full leaves rehydration. During all of this process, the carotenoids content remained stable. Twelve hours of soil rehydration were sufficient for the respiration retake. A positive carbon balance was observed since the 36th hour of rehydration, when the degree of stomatic opening became bigger than that observed for the control group. Comparing with inicial conditions, 84 hours of rehydration were sufficient to photosynthetic recovery, even just with a parcial recovery of chlorophyll content. As well as the leaf tissues, its roots might be desiccation tolerants.

Etude expérimentale et comparative de la myrmécochorie : le cas des fourmis dispersatrices Lasius niger et Myrmica rubra / Experimental and comparative study of myrmecochory: the case of seed-disperser ants Lasius niger and Myrmica rubra

Ce travail porte sur la dispersion des graines par les fourmis et se divise en deux parties : l’une expérimentale (Chapitres 1, 2 & 3) et l’autre synthétique (Chapitres 4 & 5). L’approche expérimentale a consisté en une exploration en conditions de laboratoire des comportements des fourmis à deux étapes du processus de myrmécochorie: à la source de graines et dans le nid. Des graines des plantes myrmécochores Viola odorata et Chelidonium majus ont été présentées aux fourmis Lasius niger et Myrmica rubra. Chaque étape de la séquence myrmécochorique a généré une variabilité des comportements propre à chacun des quatre couples fourmis-graines. L’élaiosome n’attire pas les fourmis à distance. Les fourmis suivent toujours la même séquence de comportements : antennation, manipulation et prise des graines. Le nombre d’antennations et de manipulations avant la prise de graines peut être considéré comme un indicateur de l’« hésitation » des fourmis à prendre les graines. L’espèce à tendance carnivore Myrmica rubra a été plus rapide et efficace dans la prise de graines que l’espèce éleveuse de pucerons Lasius niger. Parallèlement, les fourmis ont moins antenné, moins manipulé et plus pris de graines de Chelidonium majus, ce qui montre un intérêt particulier pour cette espèce. Un jour après l’expérience, toutes les graines des deux espèces se trouvaient dans les déchets à l’extérieur du nid, avec au moins la moitié des élaiosomes consommés (Chapitre 1). Lors du passage des graines à l’intérieur du nid, les fourmis Myrmica rubra ont également montré une capacité à traiter les graines rapidement, en montrant une dynamique d’arrachage de l’élaiosome et de rejet des graines hors du nid plus rapide. Le taux d’arrachage de l’élaiosome a été influencé par l’espèce de graine, plus important pour les graines de Chelidonium majus. Nous avons montré qu’une proportion variable de graines rapportées au nid (moins de la moitié) étaient déposées directement au contact des larves, les autres étant traitées ailleurs par les ouvrières ou laissées temporairement à l’abandon dans le nid. Par ailleurs, les dynamiques de rejet des items hors du nid ont curieusement été peu influencées par l’espèce de graine. Pour une graine, le fait de ne plus avoir d’élaiosome diminue le nombre moyen d’ouvrières qui la contacte simultanément. Parallèlement, même si la réponse n’est pas de type « tout ou rien », l’absence d’élaiosome accroit aussi la probabilité qu’une graine a d’être rejetée. (Chapitre 2). Nous avons isolé expérimentalement le paramètre de dessiccation des graines afin mesurer son influence sur le taux de prises. La dessiccation progressive des graines réduit les taux de prises par les fourmis Myrmica rubra. La réhydratation des mêmes graines leur permet de retrouver une attractivité et donc une valeur fonctionnelle prolongée. Les graines de Viola odorata perdent presque toute attractivité après 4 jours de dessiccation et leur réhydratation ne rétabli que faiblement leur attractivité. A l’inverse, les graines de Chelidonium majus gardent un tiers de leur attractivité après un mois de dessiccation et leur réhydratation restaure presque entièrement leur attractivité (Chapitre 3). La synthèse bibliographique a permis de dresser un aperçu des principales caractéristiques des fourmis dispersatrices de graines myrmécochores. Certains traits « généralistes » rendent les rencontres entre fourmis et graines très probables : leur ubiquité et diversité taxonomique, leur régime alimentaire omnivore, et leur fourragement « diffus » et opportuniste. Les fourmis possèdent des traits qui les rendent uniques par rapport aux autres insectes : le fourragement au sol, la capacité à transporter de la nourriture, ainsi que la nidification. Certains traits des fourmis ont une influence considérable sur la dispersion des graines : leur taille, les préférences de régime alimentaire, la phénologie, la capacité d’apprentissage et la fréquence de déménagement des nids. Nous développons également l’hypothèse que la rapidité et l’efficacité du traitement des graines par les fourmis seraient une conséquence d’un comportement hygiénique des fourmis à tendance carnivore, habituées à gérer des proies périssables (Chapitre 4). Nous avons dressé pour la première fois une liste des espèces de plantes myrmécochores et potentiellement myrmécochores des régions d’Europe tempérée (260 spp.). Nous montrons que ces dernières sont majoritairement herbacées, et ont tendance à fleurir plus précocement que les autres espèces. La proportion de graines myrmécochores comprises entre 1 et 3 mm et entre 0,6 et 10 mg est plus importante que dans le reste de la flore (Chapitre 5). / This study concerns seed dispersal by ants and is divided in two parts: one experimental (chapters 1, 2 & 3), and one synthetic (chapters 4 & 5). Experimental work consisted in a series of laboratory experiments, in which ant behaviour was studied at two stages of the dispersal process: at the seed source and inside the nest. Seeds of the myrmecochorous plants Viola odorata and Chelidonium majus were presented to two ant species: Lasius niger and Myrmica rubra. Each stage of the myrmecochory sequence generated a variability of behaviours for each of the four ant-seed pairs. The elaiosomedo not attract seed at a distance. Ants followed always the same behavioural sequence: antennations, manipulations, and removal. The number of antennations and manipulations before removal can be considered as a measure of ants’ “hesitation” to remove seeds. The carnivorous species Myrmica rubra was faster and more efficient at taking seeds than the aphid-tending Lasius niger. At the same time, ants antennated and manipulated less Chelidonium majus seeds, which shows a particular interest for this seed species. One day after the experiment, all seeds of both species were located outside the nest in the refuse piles. At least half of their elaiosomes had been consumed (chapter 1). Inside the nest, Myrmica rubra also showed a great ability to treat seeds quickly, i.e. quicker dynamics of elaiosome removal and seed rejection outside the nest. Elaiosome removal rates were influenced by seed species (higher for Chelidonium majus). We showed that a variable proportion of seeds (less than half) was directly deposited in contact with larvae. The rest of the seeds were handled elsewhere by workers, or left temporarily unattended in the nest. Dynamics of seed rejection outside the nest were curiously little influenced by the seed species. For a seed, to loose the elaiosome decreased the number of workers manipulating it. At the same time, even if the rejection response is not automatic, the absence of elaiosome increases the probability for a seed to be rejected (Chapter 2). We isolated the desiccation parameter in order to measure its influence on seed removal rates. Progressive seed desiccation reduced Myrmica rubra removal rates. Rehydration of the same seeds restored their attractiveness, thereby prolonging their functional life. Viola odorata seeds lost almost all their attractiveness after 4 days of desiccation, and rehydration only restored a reduced part of their attractiveness. On the contrary, Chelidonium majus seeds kept one third of their attractiveness after one month of desiccation, and recovered almost all their attractiveness after rehydration (Chapter 3). The bibliographic review allowed us to compile an outline of the main features of seed-disperser ants. Some generalist features highly increase the probability that ants encounter seeds: their ubiquity and taxonomic diversity, omnivorous diet and their opportunistic “diffuse” foraging. Among insects, ants have unique traits that make them broad dispsersers: ground foraging, the ability to transport items, and nesting behaviour. Some other traits have a great influence on the seed dispersal system: the ant body size, their diet preferences, the phenology of the colony, the learning, and the frequence of nest relocation. We also develop the hypothesis according to which, handling efficiency of ants is a byproduct of hygienic behaviour of carnivorous oriented species, since they are used to manage perishable preys (Chapter 4). We compiled the first list of myrmecochorous and potentially myrmecochorous plants species of European temperate regions (260 spp.). We show that these plants are mainly herbaceous. They also tend to flower earlier than the whole flora. The proportions of myrmecochorous seeds having a size range between 1 and 3 mm, and a weight range between 0.6 and 10 mg are higher than in the rest of the flora (Chapter 5).

Structure and reactivity of a biological soil crust from a xeric sandy soil in Central Europe

The investigation was designed to explore the structure, composition and activity of a biological soil crust on an acidic, sandy soil from a temperate climate. The crust covers several hundreds of square meters on the hilltop of a large terminal moraine. The conjugate alga <i>Zygogonium ericetorum</i> forms the essential matrix for the crust, a dense web of algal filaments with interspersed lichens and mosses. The crust is composed of three layers, with an uppermost layer consisting nearly entirely of a dense algal mat. In lower layers, a parasitic fungus, penetrating the algal cells, is another important component of the crust community. In this soil crust, photosynthetic and respiratory activity is stabilized at low water activities.
