Der Nachweis der Telomeraseaktivierung in Tumorgeweben und dem Tumor benachbarten Geweben von Plattenepithelkarzinomen im Kopf-Hals-Bereich

Plattenepithelkarzinome der Kopf-Hals-Region sind durch einen aggressiven Krankheitsverlauf mit häufigem Auftreten von Lokalrezidiven und Zweitkarzinomen gekennzeichnet. Molekulare Veränderungen des Tumorrandes und der Tumor benachbarten Gewebe könnten in Ergänzung zur Histopathologie eine kurative chirurgische Tumorextirpation bestätigen. Wir untersuchten die Telomeraseaktivität in verschiedenen Entfernungen vom Tumorzentrum um ihre Verwendung als molekularen Marker zu prüfen. Wir untersuchten 80 kryokonservierte Gewebeschnitte von 40 Patienten mit einem Plattenepithelkarzinom im Kopf-Hals-Bereich, davon 33 karzinomfreie Tumorrandgewebe. 50% der 20 karzinomhaltigen Gewebe, 28 % der karzinomfreien Tumorrandgewebe und 35 % der tumorfernen Proben waren telomerasepositiv. Der Vergleich ergab keinen statistisch signifikanten Unterschied. Es konnte keine sichere Korrelation zwischen der Telomeraseaktivität in den untersuchten Geweben mit dem weiteren Krankheitsverlauf gefunden werden, allerdings zeigte die Telomerasepositivität in karzinomfreien Tumorrändern die Tendenz, mit einer besseren Prognose zu korrespondieren. Unsere Befunde sollten an einer größeren und homogeneren Patientengruppe überprüft werden. / Squamous cell carcinomas of the head-neck region are characterised by an aggressive course of disease and a strong tendency to loco-regional recurrence and second primary tumours. As a complement to histopathology, molecular changes of tumour marginal and tumor distant tissue may confirm curative surgical tumour extirpation. We tested telomerase activity at different distances to the tumour centre of squamous cell carcinomas to consider the use for molecular marker. We examined 80 cryo-conserved tissue samples from 40 patients with a squamous cell carcinoma of the head-neck region, among them 33 carcinoma-free margin tissues. 50 % of the carcinoma centres, 28 % of the carcinoma-free margin tissues and 35% of the tumour distant samples were telomerase-positive. The comparison showed no statistically significant difference. We have found no reliable correlation between telomerase activity in the carcinoma-free margin tissues and the further course of disease, but telomerase positivity in carcinoma-free tumour margins tended to correlate with a better prognosis. Confirmation of the results in a larger and more homogeneous patient group is needed.

Développement d'un refroidisseur-regroupeur quadripolaire radiofréquence pour PIPERADE et mesure de la demi-vie de 17F / Developpement of a radio-frequency quadrupole cooler and buncher for PIPERADE and half-life measurement of 17F

La future installation SPIRAL2 du GANIL, à Caen, permettra de produire une gamme étendue de noyaux exotiques avec des intensités très importantes. Cependant, ces faisceaux ne pourront pas être directement utilisés pour réaliser certaines études de haute précision et devront d'abord être purifiés. C'est pour réaliser ce travail que des équipes du CENBG, du MPIK (Heidelberg), du CSNSM, du LPC Caen, du GANIL et de l'IPNO développent un double-piège de Penning dans le cadre du projet PIPERADE. Ce double-piège nécessitant un travail de mise en forme préalable du faisceau (diminution de l'émittance transverse et mise en paquet), le CENBG est en charge de la réalisation d'un refroidisseur-regroupeur quadripolaire radiofréquence : le GPIB. C'est ce développement qui a constitué la majeure partie de mon travail de thèse, notamment en ce qui concerne les simulations de ce refroidisseur-regroupeur dont les résultats ont permis de trouver une méthode innovante pour la mise en paquet et de valider sa conception mécanique. Nous disposons également d'une source d'ions afin de pouvoir tester le GPIB et le double-piège et il m'a fallu la remonter, la comprendre et la caractériser pour que ces tests soient ensuite possibles. Dans un second temps j'ai aussi participer à l'analyse de l'expérience E622S menée au GANIL et qui avait pour but de déterminer précisément la demi-vie de 17F. Ce travail n'a pas permis d'améliorer la précision sur la demi-vie de 17F mais nous avons cependant quelques doutes sur les 2 précédentes mesures et sur leur détermination du taux de contamination de leurs échantillons. / The future SPIRAL2 installation of GANIL, at Caen, will produce large range of exotic nuclei with very high intensities. Nevertheless, these beams could not be used directly for some high precision studies and will have to be purified first. To achieve this work, teams of CENBG, MPIK (Heidelberg), CSNSM, LPC Caen, GANIL and IPNO develop a double Penning trap in the framework of the PIPERADE project. Because this double Penning-trap needs some shaping work (reduction of transverse emittance and bunching), the CENBG team is in charge of the realisation of a radio-frequency quadrupole cooler and buncher : the GPIB. This developpement work was the main part of my PhD work, especially for the simulations of this cooler buncher whose results lead us to find a new bunching method and allowed us to approve its mechanical design. We also have an ion source to be able to test both GPIB and Penning trap and I had to reassemble it, to understand it and to characterise it before these tests could be achieved. In a second time I also took part to the analysis of the E622S experiment which aimed to determined precisely the 17F half-life. This work did not lead to a more precise determination of this half-life but we now have some doubts concerning the 2 last measurements and their way to determine the contamination rate of their radioactive samples.

Charge-Storage mechanisms in polymer electrets

In view of the importance of charge storage in polymer electrets for electromechanical transducer applications, the aim of this work is to contribute to the understanding of the charge-retention mechanisms. Furthermore, we will try to explain how the long-term storage of charge carriers in polymeric electrets works and to identify the probable trap sites. Charge trapping and de-trapping processes were investigated in order to obtain evidence of the trap sites in polymeric electrets. The charge de-trapping behavior of two particular polymer electrets was studied by means of thermal and optical techniques. In order to obtain evidence of trapping or de-trapping, charge and dipole profiles in the thickness direction were also monitored. <br><br> In this work, the study was performed on polyethylene terephthalate (PETP) and on cyclic-olefin copolymers (COCs). PETP is a photo-electret and contains a net dipole moment that is located in the carbonyl group (C = O). The electret behavior of PETP arises from both the dipole orientation and the charge storage. In contrast to PETP, COCs are not photo-electrets and do not exhibit a net dipole moment. The electret behavior of COCs arises from the storage of charges only. <br><br> COC samples were doped with dyes in order to probe their internal electric field. COCs show shallow charge traps at 0.6 and 0.11 eV, characteristic for thermally activated processes. In addition, deep charge traps are present at 4 eV, characteristic for optically stimulated processes. <br><br> PETP films exhibit a photo-current transient with a maximum that depends on the temperature with an activation energy of 0.106 eV. The pair thermalization length (rc) calculated from this activation energy for the photo-carrier generation in PETP was estimated to be approx. 4.5 nm. The generated photo-charge carriers can recombine, interact with the trapped charge, escape through the electrodes or occupy an empty trap. <br><br> PETP possesses a small quasi-static pyroelectric coefficient (QPC): ~0.6 nC/(m²K) for unpoled samples, ~60 nC/(m²K) for poled samples and ~60 nC/(m²K) for unpoled samples under an electric bias (E ~10 V/µm). When stored charges generate an internal electric field of approx. 10 V/µm, they are able to induce a QPC comparable to that of the oriented dipoles. Moreover, we observe charge-dipole interaction. Since the raw data of the QPC-experiments on PETP samples is noisy, a numerical Fourier-filtering procedure was applied. Simulations show that the data analysis is reliable when the noise level is up to 3 times larger than the calculated pyroelectric current for the QPC. <br><br> PETP films revealed shallow traps at approx. 0.36 eV during thermally-stimulated current measurements. These energy traps are associated with molecular dipole relaxations (C = O). On the other hand, photo-activated measurements yield deep charge traps at 4.1 and 5.2 eV. The observed wavelengths belong to the transitions in PETP that are analogous to the π - π* benzene transitions. The observed charge de-trapping selectivity in the photocharge decay indicates that the charge detrapping is from a direct photon-charge interaction. Additionally, the charge de-trapping can be facilitated by photo-exciton generation and the interaction of the photo-excitons with trapped charge carriers. These results indicate that the benzene rings (C6H4) and the dipolar groups (C = O) can stabilize and share an extra charge carrier in a chemical resonance. In this way, this charge could be de-trapped in connection with the photo-transitions of the benzene ring and with the dipole relaxations. <br><br> The thermally-activated charge release shows a difference in the trap depth to its optical counterpart. This difference indicates that the trap levels depend on the de-trapping process and on the chemical nature of the trap site. That is, the processes of charge detrapping from shallow traps are related to secondary forces. The processes of charge de-trapping from deep traps are related to primary forces. Furthermore, the presence of deep trap levels causes the stability of the charge for long periods of time. / Angesichts der Bedeutung der Ladungsspeicherung in Polymerelektreten für viele Anwendungen, wie z.B. in elektromechanischen Wandler, ist es das Ziel dieser Arbeit, zum Verständnis der zugrundeliegenden Mechanismen der kurz- und langfristigen Ladungsstabilisierung beizutragen sowie mögliche Haftstellen zu identifizieren. Ladungs- und Entladungsprozesse in Elektreten geben Hinweise auf Ladungshaftstellen. Diese Prozesse wurden mit thermischen und optischen Methoden bei gleichzeitiger Messung von Ladungs- und Polarisationprofilen untersucht. Die experimentellen Untersuchungen der vorliegenden Arbeit wurden an Polyethylenterephthalat (PETP) und an Cyclischen-Olefin Copolymeren (COC) durchgeführt. <br><br> PETP ist ein Photoelektret und weist in der Carbonylgruppe (C = O) ein Dipolmoment auf. Die Elektreteigenschaften ergeben sich sowohl aus der Orientierungspolarisation als auch aus der Ladungsspeicherung. Im Gegensatz zu PETP ist COC kein Photoelektret und zeigt auch keine Orientierungspolarisation. Deshalb folgen die Elektreteigenschaften des COC ausschließlich aus der Ladungsspeicherung. Die COC-Proben wurden mit Farbstoffen dotiert, um das innere elektrische Feld zu untersuchen. Diese Systeme zeigen flache Ladungshaftstellen bei 0,6 und 0,11 eV, die durch thermisch stimulierte Prozesse entladen werden sowie tiefe Haftstellen bei 4 eV, die optisch stimuliert werden können. <br><br> PETP-Filme zeigen einen transienten Photostrom mit einem Maximalwert ( jp), der von der Temperatur mit einer Aktivierungsenergie von 0,106 eV abhängt. Der thermische Paarabstand (rc) kann für die Photoladungsgeneration in PETP auf ca. 4,5 nm abgeschätzt werden. Die Photoladungsträger können rekombinieren, mit den gespeicherten Ladungen interagieren, über die Elektroden entkommen oder eine leere Haftstelle einnehmen. <br><br> PETP zeigt einen kleinen quasi-statischen pyroelektrischen Koeffizienten (QPC) von ca. 0,6 nC/(m²K) für nicht polarisierte Proben, ca. 60 nC/(m²K) für polarisierte Proben und ca. 60 nC/(m²K) für nicht polarisierte Proben mit Vorspannung (E ~10 V/µm). Wenn die gespeicherten Ladungen ein internes elektrisches Feld von ca. 10 V/µm generieren können, sind sie in der Lage, einen QPC herbeizuführen, der vergleichbar mit dem von orientierten Dipolen ist. Es ist außerdem möglich, eine Ladungs-Dipol-Wechselwirkung zu beobachten. Da die QPM-Daten von PETP auf Grund des geringen Signals verrauscht sind, wurde ein numerisches Fourier-Filterverfahren angewandt. Simulationen zeigen, dass eine zuverlässige Datenanalyse noch bei einem Signal möglich ist, dessen Rauschen bis zu 3-mal größer ist als der berechnete pyroelektrische Strom. <br><br> Messungen der thermisch stimulierten Entladung von PETP-Filmen ergaben flache Haftstellen bei ca. 0,36 eV, welche mit der Dipolrelaxation der Carbonylgruppe (C = O) assoziiert sind. Messungen der photostimulierten Entladung ergaben tiefe Haftstellen bei 4,1 und 5,2 eV. Die beobachteten Wellenlängen entsprechen Übergängen in PETP analog den π - π* Übergängen in Benzol. Die beobachtete Selektivität bei der photostimulierten Entladung lässt auf eine direkte Wechselwirkung von Photonen und Ladungen schließen. Einen zusätzlichen Einfluß auf die Entladung hat die Erzeugung von Photo-Exzitonen und deren Wechselwirkung mit den gespeicherten Ladungsträgern. Diese Ergebnisse deuten darauf hin, dass die Phenylringe (C6H4) und die Dipolgruppen (C = O) eine zusätzliche Ladung in einer chemischen Resonanz stabilisieren und miteinander teilen können. Daher kann die gebundene Ladung auch durch einen Photoübergang im Benzolring oder durch eine Dipolrelaxation freigesetzt werden. <br><br> Die mittels thermisch stimulierter Entladung bestimmte Tiefe der Haftstellen unterscheidet sich deutlich von den mittels photostimulierter Entladung gemessenen Werten. Flachere Haftstellen werden bei der thermisch stimulierten Entladung gefunden und können sekundären Kräften zugeordnet werden. Die tieferen Haftstellen sind chemischer Natur und können primären Kräften zugeordnet werden. Letztere sind für die Langzeitstabilität der Ladung in Polymerelektreten verantwortlich.

Investigation into the Mechanism(s) which Permit the High-Rate, Degradation of PAHS and Related Petroleum Hydrocarbons in Sequencing Batch Reactors by Attached Cells in a Controlled Mixed Bacterial Community.

A stable mixed culture, deposited as ATCC 55644, previously shown to degrade petroleum hydrocarbons at relatively high concentrations was used as the source of inoculum. This culture was grown in Stanier’s minimal media, either in the presence of different concentrations of naphthalene, nitrobenzene and toluene (NNT) or naphthalene and toluene (NT) as the sole source of C and/or N. Results showed that the majority of the strains isolated from the mixed culture were able to grow in the presence of NNT or NT. A total of 20 different isolates were isolated from the mixed culture. Individual isolates were grown in Stanier’s minimal medium containing a single hydrocarbon as the source of carbon or carbon and nitrogen. Only one strain was found to grow solely in the presence of nitrobenzene as the source of C and N. Most of the other isolates were able to grow in the presence of naphthalene, toluene, acenaphthene, anthracene, fluoranthene and phenanthrene, n-dodecane, hexadecane, n-pentadecane, n-tetradecane, and n-octadecane. Planktonic and immobilized cells of the controlled mixed culture (ATCC 55644) were grown in separate Sequential Batch Reactors (SBR) using Stanier's media, to which naphthalene, nitrobenzene and toluene were added as the sole source of C and/or N. Biodegradation was determined by measuring the residual hydrocarbon in the SBR and the amount of trapped volatile organic carbon (VOC) and the evolved CO2. Gas chromatography data showed that immobilized cells were able to degrade NNT faster than the planktonic cells. This observation was confirmed by CO2 evolution. Over time the loading of hydrocarbon was significantly increased from a starting level of 400 ppm (Naphthalene), 100 ppm (Nitrobenzene), and 500 ppm (toluene), to a final level of 3000 ppm (Naphthalene), 400 ppm (Nitrobenzene), and 1600 ppm (toluene). While increasing nutrient loading, the frequency of re-feeding with hydrocarbons was changed from an initial re-feeding every 60 hrs to a final re-feeding frequency of 18 hrs. The experiments clearly showed that the attached, mixed microbial community was able to effectively and rapidly degrade high concentrations of hydrocarbons. This demonstrated the practical advantages of employing attached, mixed microbial cultures in a SBR.

Ausência do receptor Toll-Like 2 ocasionou a formação de lesões periapicais mais extensas e com maior número de osteoclastos em camundongos / Silence of toll-like receptor 2 promoted superior size of periapical lesion and number of osteoclasts in mice

O objetivo deste trabalho foi caracterizar a formação e progressão de lesões periapicais induzidas experimentalmente em dentes de camundongos knockout para receptores toll-like 2 (TLR2 KO) comparados a animais wild-type (WT). As lesões periapicais foram induzidas nos primeiros molares inferiores de 28 camundongos WT e de 27 camundongos TLR2 KO. Decorridos 7, 21 e 42 dias da indução da lesão periapical, os animais foram submetidos à eutanásia em câmara de CO2, as mandíbulas foram removidas e submetidas ao processamento histotécnico. A seguir, cortes representativos foram corados com hematoxilina e eosina (HE), para descrição do tecido pulpar e das regiões apical e periapical, em microscopia óptica convencional, e mensuração da área das lesões periapicais, em microscopia de fluorescência. Espécimes sequenciais foram avaliados por meio de: histoenzimologia para a atividade da TRAP, para identificação de osteoclastos; coloração de Brown & Brenn, para localização de bactérias; e imunoistoquímica, para identificação de marcadores da osteoclastogênese (RANK, RANKL, OPG). Os resultados numéricos obtidos da análise morfométrica da extensão da área das lesões periapicais e do número de osteoclastos foram submetidos à análise estatística por meio dos testes não-paramétricos de Mann-Whitney e Kruskal-Wallis, utilizando o software SAS (Statistical Analysis System) for Windows versão 9.1.3. O nível de significância adotado foi de 5%. Os resultados da coloração de Brown & Brenn e Imunoistoquímica foram expressos de maneira qualitativa. O grupo de animais WT apresentou diferença significante na extensão da área das lesões periapicais e no número de osteoclastos entre os períodos experimentais de 7 e 42 dias (p<0,05) e entre 21 e 42 dias (p<0.05). Por outro lado, no grupo de animais TLR2 KO, as diferenças para a extensão da área das lesões periapicais e número de osteoclastos foram encontradas entre os períodos experimentais de 7 e 21 dias (p<0,05) e entre 7 e 42 dias (p<0,05). Quando os períodos dos grupos foram comparados entre si, foram encontradas diferenças estatísticas entre todos os períodos experimentais, tanto para a análise morfométrica da extensão da área das lesões periapicais, quanto para o número de ostoclastos (p<0,05). A análise descritiva do tecido pulpar e das regiões apical e periapical, por meio da coloração de HE, bem como da localização das bactérias, por meio da coloração de Brown & Brenn, não mostrou diferenças entre os dois grupos de animais. Com relação à Imunoistoquímica, as marcações foram semelhantes entre os dois grupos de animais, exceto para as marcações de RANK, as quais não foram encontradas nas lesões periapicais do grupo de animais TLR2 KO. A partir das metodologias empregadas e dos resultados obtidos pode-se concluir que na ausência do TLR2, os animais desenvolveram lesões periapicais significantemente maiores (com maior presença de osteoclastos) quando comparados aos animais WT, sugerindo o importante papel desse receptor na resposta imune e inflamatória do organismo no sentido de combater a infecção do sistema de canais radiculares e dos tecidos perirradiculares. / The aim of the present study was to characterize the formation and progression of periapical lesions experimentally induced in the teeth of toll-like receptors 2 knockout (TLR2 KO) mice compared to wild-type (WT) mice. Periapical lesions were induced in the lower first molars of 28 WT and 27 TLR2 KO mice. After 7, 21 and 42 of periapical lesion induction, the animals were euthanized in a CO2 chamber, and the mandibles were removed and subjected to histotechnical processing. Representative histological sections were stained with hematoxylin and eosin (HE) for description of the features of the pulp tissue and the apical and periapical regions under conventional optical microscopy, and for determination of the size of the periapical lesions under fluorescence microscopy. Sequential specimens were evaluated by: TRAP histo-enzymology for identification de osteoclasts; Brown & Brenn staining for localization of bacteria; and immunohistochemistry for identification of osteoclastogenesis markers (RANK, RANKL, OPG). Data from the morphometric evaluation of the size of periapical lesions and the number of osteoclasts were subjected to statistical analysis by the nonparametric Mann-Whitney and Kruskal-Wallis tests, using the SAS (Statistical Analysis System) software for Windows version 9.1.3. A significance level of 5% was set for all analyses. Data from the Brown & Brenn staining and immunohistochemical analysis were displayed qualitatively. The group of WT mice presented statistically significant difference in the periapical lesion size and number of osteoclasts between the 7- and 42-day experimental periods (p<0.05) as well as between 21 and 42 days (p<0.05). On the other hand, in the group of TLR2 KO mice, significant differences in the periapical lesion size and number of osteoclasts were found between the 7- and 21-day experimental periods (p<0.05) as well as between 7 and 42 days (p<0.05). Comparison of the periods within each group revealed statistically significant differences among all experimental periods for the morphometric evaluation of the size of the periapical lesions and number of osteoclasts (p<0.05). Descriptive analysis of pulp tissue and apical and periapical regions by HE staining and localization of bacteria by Brown & Brenn staining did not show significant differences between the two groups of animals. The immunohistochemical results showed similar immunostaining in both groups of animals, except for RANK expression, which was not observed in the periapical lesions of the TLR2 KO mice. Based on the employed methodology and the obtained results it may be concluded that in the silence of TLR2, the animals developed superior size of periapical lesions (with higher presence of osteoclasts) compared to WT animals, suggesting the important role of this receptor during the immune and inflammatory response against the infection of root canal system and periapical tissues.

Photoinduced hole trapping in single semiconductor quantum dots at specific sites at silicon oxide interfaces

Blinking dynamics of CdSe/ZnS semiconductor quantum dots (QD) are characterized by (truncated) power law distributions exhibiting a wide dynamic range in probability densities and time scales both for off- and on-times. QDs were immobilized on silicon oxide surfaces with varying grades of hydroxylation and silanol group densities, respectively. While the off-time distributions remain unaffected by changing the surface properties of the silicon oxide, a deviation from the power law dependence is observed in the case of on-times. This deviation can be described by a superimposed single exponential function and depends critically on the local silanol group density. Furthermore, QDs in close proximity to silanol groups exhibit both high average photoluminescence intensities and large on-time fractions. The effect is attributed to an interaction between the QDs and the silanol groups which creates new or deepens already existing hole trap states within the ZnS shell. This interpretation is consistent with the trapping model introduced by Verberk et al. (R. Verberk, A. M. van Oijen and M. Orrit, Phys. Rev. B, 2002, 66, 233202).

Étude des interactions PET - Eau minérale dans les eaux embouteillées au Liban et approches analytiques des risques sanitaires / Study of PET interaction with mineral water in lebanese bottled water and analytical approaches of health risks

L'évaluation de l'inertie des matières plastiques au contact de denrées alimentaires est régie par le règlement européen N°10/2011. Au Liban, l'absence de législations relatives à l'usage des matières en plastiques a conduit à des emballages non contrôlés au niveau de la production et du stockage, ce qui peut induire un risque sanitaire pour le consommateur. L'objectif de ce travail a été d'apprécier les phénomènes de migration des contaminants du PET vers l'eau. Pour cela, l'analyse de certains migrants potentiels a été initiée tels que des COV et des SV afin de vérifier la conformité de leur teneur par rapport aux LMA dans les règlementations internationales. Le dosage des aldéhydes dans l'eau a été effectué par dérivation avec la 2,4-DNPH, suivie d'une étape de préconcentration sur une cartouche en C18, puis séparation et quantification par HPLC-UV. L'analyse des composés SV a été réalisée par SPME-GC-FID sur une fibre en PDMS/DVB. L'analyse des BTEX et du styrène a été réalisée par HS-Trap-GC-FID. Ces méthodes ont été validées conformément à la norme NF T 90-210 avec des LQ inférieures aux LMA. L'analyse des COV dans l'eau embouteillée et dans le PET a été réalisée par HSTrap- GC-MS. La présence de FA, d'AA, de DEP et des BTEX dans l'eau embouteillée en PET a été confirmée. Il a été mis en évidence que la température et l'exposition solaire influencent la migration des aldéhydes. Les concentrations d'AA trouvées après incubation à 40ºC ne sont pas négligeables et peuvent modifier les propriétés organoleptiques de l'eau. L'analyse du PET montre la présence des hydrocarbures, des aldéhydes, des alcools, et des cétones / The inertness’ value of plastics that are on contact with foodstuffs is governed by the European rule N°10/2011. In Lebanon, the lack of the Rules and Regulations of plastics’ usage led for a non-controlled packing in production as in storage, which will cause a sanitary risk to the consumer and to the population. The purpose of this research was to appreciate the migration’s phenomena of contaminants from the PET to the water. For this, there was an analysis of potential migrants such as VOC and SV to verify their tenor’s conformity relative to the MCL in international regulations. The aldehydes’ dosage in the water was performed by the derivation with the 2,4-DNPH, followed by a concentration on a C18 cartridge, then separation and quantification was done by HPLC-UV. The SV compounds’ analysis was realized by SPME-GC-FID method on a PDMS / DVB fiber. The BTEX and the styrene’s analysis was done by HS-trap-GC-FID method. These methods were validated in accordance with standard NF T 90-210 with a LQ less than the MCL. The volatile compounds in the bottled water and in the PET were analyzed by HS-trap-GC-MS method. The presence of FA, AA, DEP and BTEX in the bottled water with the PET was confirmed. It was highlighted that temperature and sun exposition influence the aldehydes’ migration. The acetaldehyde’s concentration after incubation at 40ºC wasn’t negligible and can modify the water’s organoleptic properties. Molecular and functional analysis of cardiac diversification by cell specific translatomic approaches in Drosophila Melanogaster / Analyses moléculaires et fonctionnelles de la diversification cardiaques par des approaches translatomiques cellules-spécifiques chez la Drosophile

Le cœur humain est un organe composé de différents types cellulaires tels que les cardiomyocytes, les fibroblastes, les muscles lisses et les cellules endothéliales. Ces cellules se diversifient grâce à des mécanismes moléculaires spécifiques en acquérant leurs propriétés fonctionnelles spécifiques. L’embryon de Drosophile est un modèle simple et adapté pour étudier la diversification des cellules cardiaques et leurs propriétés spécifiques. Le but du projet est d’améliorer notre connaissance sur les acteurs moléculaires qui contrôlent la diversification des cellules cardiaques. Pour atteindre cet objectif nous avons appliqué la méthode TRAP-rc ("rare cell Translation Ribosome Affinity Purification") suivie du séquençage ARN pour identifier les ARN messagers en cours de traduction spécifiques des cellules cardiaques Tin et Lb (Tin CBs et Lb CBs) à deux stades de développement corrélés avec la morphogenèse du cœur embryonnaire. Dans une première analyse focalisée sur l'analyse des données issues des TRAP-Seq sur cellules Tin nous avons mis en évidence que CAP et MSP-300 sont impliqués dans la migration des cardioblasts pendant la fermeture du cœur. En parallèle, nous avons également identifié deux autres gènes impliqués dans la morphogenèse, kon-tiki et dGrip qui semblent contrôler la cohésion des CBs au cours de la migration. En outre, nous avons trouvé qu'au stade 16, environ 60% des gènes enrichis sont communs entre les populations Tin et Lb. Parmi ces gènes, Src42, sqa et flr participent à la régulation du cytosquelette d'actine et nos analyses ont permis de démontrer qu'ils avaient également des fonctions dans la morphogenèse cardiaque. Nous avons également identifié des groupes de gènes plus spécifiques à chacune des populations ciblées. Une catégorie fonctionnelle fortement associée à la population Lb, comprend les gènes qui régulent l'épissage des ARN messagers et certains de ces gènes semblent être requis au cours de la morphogenèse cardiaque. Enfin, nous avons comparé nos données de TRAP-seq cardiaque avec des données de TRAP-Seq issues du muscle somatique (de l'équipe), et ainsi identifié près de 90 gènes qui présentent des isoformes protéiques spécifiques à chaque tissu notamment impliquées dans la formation de l'unité contractile sarcomérique. Ceci suggère que des mécanismes d'épissage spécifiques sont mis en place dans différents types cellulaires pour moduler les fonctions de certaines protéines musculaires. A travers ce projet, nous avons identifié de nouveaux acteurs généraux de la migration collective des cardioblastes au cours de la fermeture du cœur mais également de nouveaux gènes potentiellement impliqués dans l’acquisition des propriétés spécifiques dessous populations cardiaques Tin et Lb et de tissus musculaires distincts. Nous espérons que les données générées permettront dans le futur de mieux comprendre les mécanismes de la cardiogenèse des vertébrés ainsi que l’étiologie de maladies cardiaques. / Cardiac cells diversification is required for the formation of a functional heart. Human heart is a multi-lineage organ that develops through progressive diversification of progenitors derived from different heart fields. This process is underlined by numerous changes in the expression of a repertory of genes that allow cells to acquire their own identity and functions. The Drosophila embryo is a relatively simple model to study the diversification of cardiac cells and their properties. The goal of this project is to identify the repertories of genes that control the formation of different types of cardiac cells. To reach this objective we applied Translation Ribosome Affinity Purification (TRAP) method followed by RNA sequencing in order to identify mRNA engaged in translation specific to two cardiac cell types (Tinman (Tin) and Labybird (Lb) expressing cells), at two different time windows. We obtained a list of enriched genes for the different types of cardiac cells and time points. In a first part, we focused our attention on the Tindatasets and found that two genes, CAP and Msp300, are involved in cardioblasts migration during the heart closure. Then we identified two other candidate genes kontiki and dGrip that seem to contribute to maintain cohesion between CBs during heartmorphogenesis. Moreover by comparing our spatial datasets, we found that for the same time point, around 60% of Tin CBs enriched genes are common with Lb CBs enriched population and within this group we identified evolutionary conserved genes such as Src42, flr and sqa known to be involved in the cytoskeleton organization and in the actinpolymerization and depolymerisation. Our premiminary analyses show that they seem to be required for correct cardiac morphogenesis. We also identified sets of genes more specific for each cardiac cell population. Indeed, Lb CBs datasets show that in early stage there is the enrichment of genes mostly involved in transcriptional regulation and RNA splicing and some of these genes (prp8 and prp38) are involved in cardiac development. In parallel, we compared our TRAP-Seq dataset in the cardiac system with the TRAP-seqon muscle cells, and identified close to 90 genes that present cardiac or muscular specific isoforms. It is known that the alternative splicing, by increasing proteins diversity, contributes to the acquisition of specific cell properties. Furthermore, some cardiomyopathies are associated to defects in the alternative splicing of genes encoding sarcomeric proteins that we found in our dataset such as Tropomyosin and Zasp52. With this project, we have identified new actors of collective cardioblast migration and a set of genes with potential role in the acquisition of individual properties of Tin and Lbcardiac cells or of specific type of muscle tissue. We hope that our data could provide new insights into the genetic control of vertebrate cardiogenesis and into etiology of cardiac diseases.

This diploma thesis deals with a design and construction of an embeded camera trap with a video recording and night vision support, which is able to self adapt to surroundings. The device is based on the Raspberry Pi platform and configurable by touch screen control with a user-friendly graphical interface. Also the web-application for easy remote administration was developed in Ruby On Rails framework. The camera trap was built and successfully tested under the real conditions. There are also typical application cases and requirements for camera trap construction described in this thesis.

Facies Analysis, Sequence Stratigraphy and Paleogeography of the Middle Jurassic (Callovian) Entrada Sandstone: Traps, Tectonics, and Analog

The late Middle Jurassic (Callovian) Entrada Sandstone has been divided into two general facies associations consisting primarily of eolian sandstones in eastern Utah and "muddy" redbeds in central Utah. Sedimentary structures within the redbed portion are explained by the interfingering of inland sabkha, alluvial, and eolian depositional systems. A complete succession from the most basinward facies to the most terrestrial facies in the Entrada Sandstone consists of inland sabkha facies overlain by either alluvial or eolian facies. Where both alluvial and eolian facies interfinger, alluvial facies overlain by eolian facies is considered a normal succession. Sequence boundaries, often identified by more basinward facies overlying more landward facies, are observed in the Entrada Sandstone and are extrapolated for the first time across much of Utah, including both the eolian-dominated and redbed-dominated areas. Using these sequence boundaries as well as recent tephrochronologic studies, three time correlative surfaces have been identified in the Entrada. Based on the facies interpretations at each surface, five paleogeographic reconstructions and five isopach maps have been created, illustrating two major intervals of erg expansion and the location of the Jurassic retroarc foreland basin's potential forebulge. Eolian (erg-margin) sandstones pinch-out into muddy redbeds creating combination traps, as evidenced by dead oil (tar) and bleached eolian sandstone bodies within the Entrada. The Entrada Sandstone is a world-class analog for similar systems, such as the Gulf of Mexico's Norphlet Sandstone, where eolian facies grade into muddy redbed facies.
