The Phantom Menace: the F-4 in Air Combat in VietnamHankins, Michael W. 08 1900 (has links)
The F-4 Phantom II was the United States' primary air superiority fighter aircraft during the Vietnam War. This airplane epitomized American airpower doctrine during the early Cold War, which diminished the role of air-to-air combat and the air superiority mission. As a result, the F-4 struggled against the Soviet MiG fighters used by the North Vietnamese Air Force. By the end of the Rolling Thunder bombing campaign in 1968, the Phantom traded kills with MiGs at a nearly one-to-one ratio, the worst air combat performance in American history. The aircraft also regularly failed to protect American bombing formations from MiG attacks. A bombing halt from 1968 to 1972 provided a chance for American planners to evaluate their performance and make changes. The Navy began training pilots specifically for air combat, creating the Navy Fighter Weapons School known as "Top Gun" for this purpose. The Air Force instead focused on technological innovation and upgrades to their equipment. The resumption of bombing and air combat in the 1972 Linebacker campaigns proved that the Navy's training practices were effective, while the Air Force's technology changes were not, with kill ratios becoming worse. However, the last three months of the campaign introduced an American ground radar system that proved more effective than Top Gun in improving air-to-air combat performance. By the end of the Vietnam War, the Air Force and Navy overcame the inherent problems with the Phantom, which were mostly of their own making.

4-D Modeling of Displacement Vector Fields for Improved Radiation TherapyZachariah, Elizabeth 10 December 2010 (has links)
In radiation therapy, it is imperative to deliver high doses of radiation to the tumor while reducing radiation to the healthy tissue. Respiratory motion is the most significant source of errors during treatment. Therefore, it is essential to accurately model respiratory motion for precise and effective radiation delivery. Many approaches exist to account for respiratory motion, such as controlled breath hold and respiratory gating, and they have been relatively successful. They still present many drawbacks. Thus, research has been expanded to tumor tracking. This paper presents a spatio-temporal model for four dimensional CT reconstruction. The method begins with a set of initial CT projections and a simultaneously acquired breathing trace. Two methods are explored to model the spatial components: principal component analysis and a pseudoinverse matrix method. An iterative approach is used to match the simulated projections to the actual projections. The simulated projections and the initial projections are evaluated using Normalized Root Mean Square Error (NRMSE). The proposed method shows simulated projections and actual projections match, and as such the model is able to accurately predict the deformation.

Sous chargement, les matériaux dits ‘quasi-fragiles’ tels que les bétons et de nombreuses roches présentent souvent une perte de rigidité progressive, ainsi qu’un comportement anisotrope, asymétrique et hystérétique. Il est maintenant largement admis que ce comportement, fortement non linéaire et complexe, est lié principalement à la création, au développement et au jeu d’ouverture et fermeture des réseaux de microfissures accompagnés de glissement et de frottement entre lèvres de microdéfauts en contact.Dans l’optique d’une utilisation industrielle sérieuse, la construction d’un modèle d’endommagement simple, fiable sur le plan mathématique et thermodynamique et offrant une réponse pertinente pour les matériaux quasi-fragiles jusqu’à l’apparition d’une localisation de l’endommagement, constitue à l’heure actuelle un enjeu important. Il représente en effet une étape indispensable avant de pouvoir espérer modéliser de manière satisfaisante le comportement jusqu’à la ruine de nombreux ouvrages du génie civil. L’objectif du travail de thèse est d’améliorer les capacités prédictives d’un modèle simple d’endommagement par microfissuration existant qui prend en compte les effets unilatéraux liés à l’ouverture-fermeture des microfissures. La nouvelle approche proposée est basée sur l’introduction d’un second phénomène dissipatif susceptible d’être induit lors de la fermeture des microdéfauts : le glissement avec frottement des lèvres en contact. La modélisation de ce nouveau mécanisme dissipatif s’effectue à l’échelle macroscopique en ne faisant référence à l’échelle inférieure ou aux résultats de la micromécanique des milieux microfissurés que pour interpréter ou justifier certaines hypothèses. Une attention particulière est accordée à la continue différentiabilité du potentiel thermodynamique proposé. L’étude et la simulation de différents essais permettent d’illustrer la pertinence des choix retenus. / Under loading, materials called 'quasi-brittle' as concrete and many rocks often exhibit progressive loss of stiffness, as well as anisotropic behavior, and asymmetric hysteretic. It is now widely accepted that this behavior highly nonlinear and complex, is mainly related to the creation, development and set of opening and closing of microcracks accompanied networks and sliding friction between the lips of microdefects in contact. In the context of a serious industrial use, the construction of a damage model simple, thermodynamically and mathematically reliable and providing an appropriate response for quasi-brittle materials until the onset of localization damage, is currently an important issue. It is indeed a necessary step before adequately modelling behavior to the ruin of many civil engineering works. The objective of this thesis is to improve the predictive capabilities of an existing simple model of damage by microcracking which takes into account unilateral effects related to the opening and closing of microcracks. The new approach is based on the introduction of a second dissipative phenomenon may be induced when closing microdefects: frictional sliding of the lips of microcracks. Modeling of this new dissipative mechanism occurs at the macroscopic scale by only referring to the lower scale or the results of the micro environments that microcracked to interpret or justify certain assumptions. Particular attention is paid to the continuous differentiability of the thermodynamic potential proposed. The study and simulation of various tests enable to illustrate the relevance of the choices made.

Sex differences in astrocyte and microglia responses immediately following middle cerebral artery occlusion in adult miceMorrison, Helena W., Filosa, Jessica A. 12 1900 (has links)
Epidemiological studies report that infarct size is decreased and stroke outcomes are improved in young females when compared to males. However, mechanistic insight is lacking. We posit that sex-specific differences in glial cell functions occurring immediately after ischemic stroke are a source of dichotomous outcomes. In this study we assessed astrocyte Ca2+ dynamics, aquaporin 4 (AQP4) polarity, Sloop expression pattern, as well as, microglia morphology and phagocytic marker CD11b in male and female mice following 60 min of middle cerebral artery (MCA) occlusion. We reveal sex differences in the frequency of intracellular astrocyte Ca2+ elevations (F-(1,F-86) = 8.19, P = 0.005) and microglia volume (F-(1,F-40) = 12.47, P = 0.009) immediately following MCA occlusion in acute brain slices. Measured in fixed tissue, AQP4 polarity was disrupted (F-(5,F-86) = 3.30, P = 0.009) and the area of non-S100 beta immunoreactivity increased in ipsilateral brain regions after 60 min of MCA occlusion (F-(5,F-86) = 4.72, P = 0.007). However, astrocyte changes were robust in male mice when compared to females. Additional sex differences were discovered regarding microglia phagocytic receptor CD11b. In sham mice, constitutively high CD11b immunofluorescence was observed in females when compared to males (P = 0.03). When compared to sham, only male mice exhibited an increase in CD11b immunoreactivity after MCA occlusion (P = 0.006). We posit that a sex difference in the presence of constitutive CD11b has a role in determining male and female microglia phagocytic responses to ischemia. Taken together, these findings are critical to understanding potential sex differences in glial physiology as well as stroke pathobiology which are foundational for the development of future sex-specific stroke therapies. Läromedelsanalys : Möjligheten till begreppsinlärning och de matematiska förmågorna i förhållande till Lgr2011Karlsson, Louise January 2017 (has links)
Mot bakgrund av att svensk matematikundervisning visat sig vara strakt styrd av läromedel, vilket i sin tur visat sig påverka hur och om eleverna får möjlighet att utveckla matematiska förmågor. På grund av detta finns ett behov att bidra med kunskap om vilka förutsättningar läromedel ger att utveckla matematiska förmågor i förhållande till Lgr 2011. Den här studiens syfte är att bidra med kunskap om hur två läromedel tillverkade efter 2011, som sägs vara anpassade efter rådande läroplanen behandlar området procent. Studien genomfördes med läromedelsanalys som metod, där kriterierna för de utvalda läromedlen var att de skulle vara producerade efter 2011 och behandla området procent. Studiens resultat visar att uppgifterna i de analyserade läromedlen främst ger förutsättningar att utveckla de matematiska förmågorna resonemangsförmågan, metodförmågan, begreppsförmågan och den minst komplexa delen av problemlösningsförmågan samtidigt som kommunikationsförmågan ges mindre förutsättningar att utvecklas i förhållande till de andra förmågorna. Resultatet visar även att möjligheten till begreppsinlärning inom området procent är förenlig med det centrala innehållet i den nuvarande läroplanen för området procent i de analyserade uppgifterna.

Efecto de la testosterona en la captación de glucosa dependiente del transportador GLUT4 a través del eje de señalización CaMKII/AMPK en cardiomiocitosWilson Rodríguez, Carlos Antonio January 2011 (has links)
Memoria de título para optar al título profesional de Bioquímico / La testosterona es la principal hormona sexual masculina, responsable de múltiples
efectos en el organismo. Dentro de estos, se ha descrito que es capaz de producir hipertrofia
cardiaca tanto in vitro como in vivo. Una de las etapas claves en el desarrollo de la
hipertrofia del cardiomiocito consiste en el aumento del consumo de glucosa, como
estrategia para aumentar la producción de energía asociada al crecimiento hipertrófico. Si
bien la testosterona es capaz de producir hipertrofia cardiaca, no se ha descrito que además
aumente la captación de glucosa en cardiomiocitos.
Como hipótesis a este problema, se planteó que la testosterona aumenta la captación
de glucosa en cardiomiocitos a través del transportador de glucosa GLUT4. Para evaluar
esta hipótesis, se realizó cultivo primario de cardiomiocitos de ratas neonatas y se evaluó la
captación de glucosa por medio de la glucosa fluorescente 2-NBDG (2-(N-(7-nitrobenzo-2-
oxa-1,3-diazol-4-il) amino)-2-deoxiglucosa) y 2-deoxi-[3H] glucosa en cardiomiocitos
estimulados con testosterona. Adicionalmente, se evaluó la participación del transportador
de glucosa GLUT4, la proteína Akt, la proteína dependiente del complejo Ca2+/calmodulina
(CaMKII) y la proteína quinasa activada por AMP (AMPK) en el proceso.
Los resultados obtenidos a través de microscopía de epifluorescencia y centelleo
líquido, indican que la testosterona aumentó la captación de la glucosa 2-NBDG y 2-deoxi-
[3H] glucosa, respectivamente, alcanzando un valor máximo luego de 2 h de estímulo. La
inhibición selectiva del transporte de glucosa a través de GLUT4 con indinavir, bloqueó el
aumento en la captación de glucosa por testosterona, sugiriendo la participación de GLUT4
en el proceso. Adicionalmente, cardiomiocitos transfectados con el plasmidio GLUT4myceGFP
revelaron que la testosterona aumentó la translocación de GLUT4 a la membrana
plasmática. Mediante ensayos de Western blot se determinó que la testosterona aumentó la fosforilación de las proteínas CaMKII y AMPK (alcanzando un máximo en su fosforilación
luego de 15 y 60 min de estímulo, respectivamente), las que resultaron ser claves tanto en la
translocación de GLUT4 a la membrana plasmática, como en la captación de glucosa por
testosterona.
Finalmente, es posible concluir que la testosterona aumenta la captación de glucosa
a través de GLUT4 por medio de la activación de la vía de señalización CaMKII/AMPK.
Lo anterior revela el papel de la testosterona en la captación de glucosa en cardiomiocitos,
lo cual puede constituir un mecanismo de captación de glucosa destinada a la producción de
energía celular, síntesis de componentes necesarios para el crecimiento hipertrófico y otros
procesos anabólicos importantes para estas células. / Testosterone is the principal male sexual hormone, and plays a key role in the
development of male reproductive tissues. It has been reported that testosterone induces
cardiac hypertrophy, both in vivo and in vitro. One critical step during cardiac hypertrophy
development is the induction of glucose uptake to improve energy production during
cardiac growth. Although it is known that testosterone induces cardiac hypertrophy, its
effect on glucose uptake in cardiomyocytes remains unknown.
The hypothesis of this work is that testosterone increases GLUT4-dependent
glucose uptake by the activation of the CaMKII/AMPK signaling pathway. Experiments
were performed in primary cell culture of neonatal cardiomyocytes. To evaluate
testosterone-induced glucose uptake in cardiomyocytes, the fluorescent analog of glucose
2-NBDG (2-(N-(7-nitrobenz-2-oxa-1,3-diazol-4-yl)amino)-2-deoxyglucose) and 2-deoxy-
[H3] glucose were used. Moreover, participation of the glucose transporter GLUT4, as well
as the protein Akt, Ca2+/calmodulin complex dependent protein kinase (CaMKII) and
AMP-activated protein kinase (AMPK) were assessed.
Testosterone increased both 2-NBDG and 2-deoxy-[H3] glucose uptake (by
epifluorescence microscopy and liquid scintillation counting, respectively), reaching a peak
after 2 h of testosterone stimulation. Indinavir, a specific GLUT4 blocker, inhibited
testosterone-induced glucose uptake in cardiomyocytes. In addition, western blot assays
showed that testosterone increased phosphorylation levels of both CaMKII and AMPK,
after 15 and 60 min of stimulation, respectively. Moreover, both proteins were crucial for GLUT4 translocation to the plasma membrane and glucose uptake induced by testosterone
in cardiomyocytes.
Taken together, data showed that testosterone increases GLUT4-dependent glucose
uptake through CaMKII/AMPK signaling pathway activation. These findings suggest that
testosterone increases glucose uptake in cardiomyocytes, which could be necessary for
energy production required for cellular homeostasis, cardiomyocyte growth and other
important anabolic process in cardiac cells.

LPS previene la pérdida de viabilidad de fibroblastos cardiacos inducida por isquemia/reperfusión simulada : rol protector del receptor de tipo toll 4Queirolo Fuentes, Cristián Felipe January 2014 (has links)
Memoria para optar al título de Químico Farmacéutico / El daño ocasionado por la ocurrencia de un infarto cardiaco es complejo, generando la pérdida de viabilidad de las células cardiacas entre otros efectos deletéreos ocurridos tanto en el periodo isquémico de falta de oxígeno y nutrientes, así como también en la posterior reperfusión sanguínea. Frente a esta condición patológica los fibroblastos cardiacos (FC) son capaces de reaccionar, secretando y renovando la matriz extracelular; lo que los convierte en elementos celulares claves en la cicatrización y remodelamiento del tejido cardiaco dañado post-infarto al miocardio. Esto hace necesario el intentar regular la viabilidad de estas células para una correcta cicatrización y mantención de la función cardiaca. En relación a esto se ha reportado el efecto cardioprotector ejercido por el empleo de LPS en condiciones de daño celular causado por isquemia/reperfusión (I/R). Sin embargo, dicho efecto ha sido descrito principalmente en cardiomiocitos, por lo que su efecto en FC es aún desconocido.
Nuestro trabajo estudió la capacidad cito-protectora ejercida por LPS sobre FC de ratas neonatas sometidos a un modelo de I/R in vitro e indagó las vías transduccionales implicadas en este efecto. El tratamiento de los FC con LPS (1 μg/mL) durante la isquemia y reperfusión previno la pérdida de viabilidad inducida por I/R. Sin embargo, en el pre-condicionamiento con LPS durante 24 o 16 h, y en el tratamiento con LPS durante la isquemia o reperfusión, no se observó el efecto cito-protector. En esta misma línea, demostramos que el efecto cito-protector ejercido por LPS es mediado a través del receptor TLR4 vía PI3K/Akt y ERK1/2, ya que el empleo de TAK-242, Ly29002 y PD98059, inhibidores del receptor y de las vías transduccionales respectivamente, bloquearon completamente el efecto cito-protector. La activación de TLR4 por LPS previno, además, el procesamiento de la procaspasa 8 y 3 inducido por I/R. Conjuntamente, demostramos que las vías PI3K/Akt y ERK1/2 participaban en la prevención del procesamiento de la procaspasa 8 ejercido por LPS, pero no tuvieron efecto en la activación de la caspasa 3. Nuestros resultados dan cuenta del efecto cito-protector y antiapoptótico ejercido por LPS a través de TLR4 vía PI3K/Akt y ERK1/2 frente a la muerte de FC inducida por I/R / The damage caused by a myocardial infarct is complex, causing cardiac cell viability loss, among other deleterious effects that occur during the ischemia period, such as lack of oxygen and nutrients; as well as the posterior reperfusion with blood. In this situation, cardiac fibroblasts react by secreting proteins and renewing the extracellular matrix. These properties make them a key element in the scar and remodeling process of the injured cardiac tissue. Thus, the viability of these cells are required to preserve the correct functioning of the heart. In this regard, the use of LPS as a cytoprotector agent in cardiac Ischemia/reperfusion (I/R) has been reported. However such effect has been described mostly in cardiomyocytes cells, whereas its role in cardiac fibroblasts remains unknown.
Our work studied the LPS cytoprotector effect in neonate cardiac fibroblast exposed to an in vitro model of I/R, and transductional pathways involved in this process were explored. Incubation with LPS (1μg/mL) during both ischemia and reperfusion periods prevented the I/R-induced cell loss. However, LPS used only during preconditioning, ischemia or reperfusion did not induced cytoprotection. Furthermore we demonstrated that the LPS-dependent protective effects were completely abolished when TLR4 receptor (TAK-242), and PI3K/Akt and ERK1/2 (Ly29002 and PD98059, respectively) inhibitors were used. These data suggest that LPS-dependent cytoprotector effects are mediated through TLR4 receptor activation and PI3K/Akt and ERK1/2 signalling pathways. Additionally the activation of TLR4 by LPS prevented the cleave of procaspases 3 and 8 induced by I/R. Both PI3K/Akt and ERK1/2 were necessary for the prevention of the caspase 8 activation, but not of caspase 3. Our results conclude that LPS protects cardiac fibroblasts from I/R apoptosis by the activation of TLR4 and both PI3K/Akt and ERK1/2 signalling pathways / FONDECYT
