Occupations et logiques policières: la police communale de Bruxelles pendant les première et deuxième guerres mondiales, 1914-1918 et 1940-1944

En tant que pays occupé pendant les deux conflits mondiaux, la Belgique s’avère être un laboratoire pour étudier le phénomène des occupations pendant le XXe siècle. Pour la bureaucratie étatique, ces occupations posent la question de leur positionnement face à une dissociation entre Etat et Nation. La comparaison diachronique de la police communale de Bruxelles – à travers l’angle organisationnel et à travers sa pratique dans l’espace social – a permis de dégager plusieurs thèses. <p>Le développement des appareils administratifs a pris de telles dimensions dans le XIXe siècle que l’occupant est obligé de trouver un modus vivendi avec les institutions existant sur les territoires occupés, lui-même étant incapable de gérer seul les pays sous son contrôle. Cette constellation donne une marge de manœuvres importante à la police locale, l’institution qui fait l’objet de notre étude.<p>Pendant les deux guerres, la police est soumise à un processus de réformes visant à améliorer son fonctionnement :centralisation du commandement, spécialisation d’unités, élargissement géographie des compétences d’intervention… Ces changements s’inspirent d’une part d’idées ambiantes en Belgique et d’autre part de projets réalisés en Allemagne dans les deux périodes procédant la guerre.<p>L’intégration de l’appareil policier communal à l’intérieur d’un régime d’occupation est facilitée par le professionnalisme de celui-ci qui contraste fortement avec la pratique des polices auxiliaires pour lesquelles l’ordre patriotique et/ou idéologique peut prendre le dessus sur le ‘maintien d’ordre classique’. Exploring the construction of white male identity in selected novels by J.M. Coetzee

Coetzee's own experience of living in apartheid South Africa provides the backdrop for novels infused with sardonic irony and rich metaphoric systems. In modes of metafiction that emphasize the destructive and violent nature of language, he optimizes his unique oeuvre to interrogate global, national and domestic power relations. This dissertation relies on psychoanalytical theories that examine microstructures of power within the individual, and in his domestic domain. Each of Coetzee's chief protagonists carries a secret related to a dysfunctional mother/son relationship. This hampers their psychosocial dynamics, their masculinity and sexuality. As they respectively strive toward an elusive new life they confront patriarchal power structures that speak on behalf of individuals, '[whose] descent into powerlessness [is] voluntary' (Coetzee 2007: 4-5). Coetzee's constructed white males perform their several identity roles in milieux that span divergent phases of colonial history. His critique points to white patriarchal hegemonic ideological discourses that bespeak the self/other dichotomy in a postcolonial world where the language of dominance supports an oppressive status quo. / English Studies / M.A. (English)

Analysis of global gene expression profiles and invasion related genes of colorectal liver metastasis

Die Leber ist das am häufigsten von Metastasen betroffene Organ und kann daher als Modellorgan für metastatische Invasion dienen. Aus diesem Grund war es das Ziel dieser Dissertation Genexpressionsprofile zu verstehen und metastasierungs- sowie invasionsassoziierte Gene zu identifizieren. Differentielle Genexpression wurde in drei Systemen überprüft: Einem syngenen Mausmodell, einem Xenograftmodell sowie in fünf Gewebeproben von Patienten. Genexpressionprofile des syngenen Mausmodells und der Patientenproben zeigten, dass man die Invasionsfront als Ganzes betrachten, um möglichst viele über-lappende Gene zu finden. Globale Genexpressionstudien, die auf den Wirtsteil der Invasionsfront zeigten bemerkenswerte Überrepräsentation z. B. der „GO-terms“ „extrazelluläre Matrix“, Zellkommunikation“, „Antwort auf biotischen Stimulus“, Strukturmolekülaktivität“ und „Zellwachstum“. Marker der Aktivierung hepatischer Sternzellen überrepräsentiert in der invasionsfront, was die Durchführbarkeit einer Analyse differentieller Genexpression im genomweiten Rahmen anzeigt. Globale Genexpressionsstudien, auf den Tumorzellen in der in vitro Situation, in vivo und in der Invasionsfront zeigten insgesamt einen Anstieg zellulärer Spezialisierung von der in vitro zur Invasionsfront. Sezernierte proangiogenetische Chemokine zeigten eine Hochregulation in der Invasionsfront. Das beta catenin Gen war in der Invasionsfront 9.6 fach erhöht im Vergleich zur in vitro Situation. Die Überprüfung der transkriptionellen Aktivierung von beta catenin über die Prüfung der Promotoraktivität zeigte einen 18.4 fachen Anstieg in den Tumorzellen der Invasionsfront. Weiterhin war die Promotoraktivität (an Hand der Aktivität der mRNA des Alkalischen Phosphatase Reportergens) im Tumorinneren 3.5 fach höher als in der Zellkultur, was für einen transkriptionellen Mechanismus der beta catenin Regulation zusätzlich zu den posttranslationalen Mechanismen spricht. / Liver is most frequently populated by metastases and may therefore serve as a model organ for metastatic invasion. So the aim of this thesis is to understand the gene expression profiles and identify metastasis and invasion related genes. Differential gene expression was examined in three systems: A syngeneic mouse model, a xenograft model and five clinical specimens. Gene expression profiles of a syngenic mouse model and human clinical specimen revealed that the invasion front should be considered as a whole to find more overlapping potential target genes. Global gene expression studies on the host part of the invasion front, revealed a pronounced overrepresentation of GO-terms (e.g. “extracellular matrix”, “cell communication”, “response to biotic stimulus”, “structural molecule activity” and “cell growth”). Hepatic stellate cell activation markers were over-represented in the invasion front demonstrating the feasibility of a differential gene expression approach on a genome wide scale. Global gene expression studies of the tumor cells in vitro, in vivo and tumor part of the invasion front revealed an overall increase of cellular specialization from in vitro to the invasion front. Secreted angiogenic cytokines were found to be up regulated in the invasion front. Beta catenin gene of “cell adhesion” GO term was elevated 9.6 fold in invasion front compared to in vitro. Evaluation of transcriptional up-regulation of beta catenin by promoter activity showed an 18.4 fold increase in the tumor cells of the invasion front as compared to those from the faraway tumor. Promoter activity assessed by soluble human placental alkaline phosphatase reporter gene mRNA was 3.5 fold higher in the inner parts of the tumor than in vitro cells indicating a transcriptional mechanism of beta catenin regulation in addition to the posttranslational regulatory mechanisms.

Systems biology approaches to somatic cell reprogramming reveal new insights into the order of events, transcriptional and epigenetic control of the process

Die Reprogrammierung somatischer Zellen hat sich kürlich als leistungsfähige Technik für die Herstellung von induzierten pluripotenten Stammzellen (iPS Zellen) aus terminal differenzierten Zellen bewährt. Trotz der großen Hoffnung, die sie speziell im Bezug auf patientenspezifische Stammzelltherapie darstellt, gibt es viele Hindernisse auf dem Weg zur Anwendung in der Humanmedizin, die sich von niedrigen Effizienzen bei der technischen Umsetzung bis hin zur unerwünschten Integration von Onkogenen in das menschliche Genom erstrecken. Aus diesem Grund ist es unabdingbar, unser Verständnis der zugrundeliegenden Prozesse und Mechanismen zu vertiefen. Durch neue Datengewinnungsmethoden und stetig wachsende biologische Komplexität hat sich der Denkansatz der Systembiologie in den letzten Jahrzehnten stark etabliert und erfährt eine fortwährende Entwicklung seiner Anwendbarkeit auf komplexe biologische und biochemische Zusammenhänge. Verschiedene mathematische Modellierungsmethoden werden auf den Reprogrammierungsprozess angewendet um Engpässe und mögliche Effizienz-Optimierungen zu erforschen. Es werden topologische Merkmale eines Pluripotenznetzwerkes untersucht, um Unterschiede zu zufällig generierten Netzen und so topologische Einschränkungen des biologisch relevanten Netzwerkes zu finden. Die Optimierung eines Booleschen Modells aus einem selbst kuratierten Netzwerk in Bezug auf Genexpressionsdaten aus Reprogrammierungsexperimenten gewährt tiefgreifende Einblicke in die ersten Schritte und wichtigsten Faktoren des Prozesses. Der Transkriptionsfaktor SP1 spielt hierbei eine wichtige Rolle zur Induktion eines intermediären, transkriptionell inaktiven Zustands. Ein probabilistisches Boole''sches Modell verdeutlicht das Zusammenspiel epigenetischer und transkriptioneller Kontrollprozesse zusammen, um Pluripotenz- und Zelllinien-Entscheidungen in Reprogrammierung und Differenzierung zu treffen. Erklärungen für die geringe Effizienz werden versucht. / Somatic Cell Reprogramming has emerged as a powerful technique for the generation of induced pluripotent stem cells (iPSCs) from terminally differentiated cells in recent years. Although holding great promises for future clinical development, especially in patient specific stem cell therapy, the barriers on the way to a human application are manifold ranging from low technical efficiencies to undesirable integration of oncogenes into the genome. It is thus indispensable to further our understanding of the underlying processes involved in this technique. With the advent of new data acquisition technologies and an ever-growing complexity of biological knowledge, the Systems Biology approach has seen an evolution of its applicability to the elaborate questions and problems of researchers. Using different mathematical modeling approaches the process of somatic cell reprogramming is examined to find out bottlenecks and possible enhancements of its efficiency. I analyze the topological characteristics of a pluripotency network in order to find differences to randomly generated networks and thus deduce constraints of the biologically relevant network. The optimization of a Boolean model from a curated network against early reprogramming gene expression profiles reveals profound insights into the first steps and most important factors of the process. The transcription factor SP1 emerges to play an important role in the induction of an intermediate, transcriptionally inactive state. A probabilistic Boolean network (PBN) illustrates the interplay of transcriptional and epigenetic regulatory processes in order to explain pluripotency and cell lineage decisions in reprogramming and differentiation. Explanations for the low reprogramming efficiencies are tried.

Bioinformatics of eukaryotic gene regulation

Die Aufklärung der Mechanismen zur Kontrolle der Genexpression ist eines der wichtigsten Probleme der modernen Molekularbiologie. Detaillierte experimentelle Untersuchungen sind enorm aufwändig aufgrund der komplexen und kombinatorischen Wechselbeziehungen der beteiligten Moleküle. Infolgedessen sind bioinformatische Methoden unverzichtbar. Diese Dissertation stellt drei Methoden vor, die die Vorhersage der regulatorischen Elementen der Gentranskription verbessern. Der erste Ansatz findet Bindungsstellen, die von den Transkriptionsfaktoren erkannt werden. Dieser sucht statistisch überrepräsentierte kurze Motive in einer Menge von Promotersequenzen und wird erfolgreich auf das Genom der Bäckerhefe angewandt. Die Analyse der Genregulation in höheren Eukaryoten benötigt jedoch fortgeschrittenere Techniken. In verschiedenen Datenbanken liegen Hunderte von Profilen vor, die von den Transkriptionsfaktoren erkannt werden. Die Ähnlichkeit zwischen ihnen resultiert in mehrfachen Vorhersagen einer einzigen Bindestelle, was im nachhinein korrigiert werden muss. Es wird eine Methode vorgestellt, die eine Möglichkeit zur Reduktion der Anzahl von Profilen bietet, indem sie die Ähnlichkeiten zwischen ihnen identifiziert. Die komplexe Natur der Wechselbeziehung zwischen den Transkriptionsfaktoren macht jedoch die Vorhersage von Bindestellen schwierig. Auch mit einer Verringerung der zu suchenden Profile sind die Resultate der Vorhersagen noch immer stark fehlerbehafted. Die Zuhilfenahme der unabhängigen Informationsressourcen reduziert die Häufigkeit der Falschprognosen. Die dritte beschriebene Methode schlägt einen neuen Ansatz vor, die die Gen-Anotation mit der Regulierung von multiplen Transkriptionsfaktoren und den von ihnen erkannten Bindestellen assoziiert. Der Nutzen dieser Methode wird anhand von verschiedenen wohlbekannten Sätzen von Transkriptionsfaktoren demonstriert. / Understanding the mechanisms which control gene expression is one of the fundamental problems of molecular biology. Detailed experimental studies of regulation are laborious due to the complex and combinatorial nature of interactions among involved molecules. Therefore, computational techniques are used to suggest candidate mechanisms for further investigation. This thesis presents three methods improving the predictions of regulation of gene transcription. The first approach finds binding sites recognized by a transcription factor based on statistical over-representation of short motifs in a set of promoter sequences. A succesful application of this method to several gene families of yeast is shown. More advanced techniques are needed for the analysis of gene regulation in higher eukaryotes. Hundreds of profiles recognized by transcription factors are provided by libraries. Dependencies between them result in multiple predictions of the same binding sites which need later to be filtered out. The second method presented here offers a way to reduce the number of profiles by identifying similarities between them. Still, the complex nature of interaction between transcription factors makes reliable predictions of binding sites difficult. Exploiting independent sources of information reduces the false predictions rate. The third method proposes a novel approach associating gene annotations with regulation of multiple transcription factors and binding sites recognized by them. The utility of the method is demonstrated on several well-known sets of transcription factors. RNA interference provides a way of efficient down-regulation of gene expression. Difficulties in predicting efficient siRNA sequences motivated the development of a library containing siRNA sequences and related experimental details described in the literature. As all above, Ven. Ju Zan served as an active coordinator between the central government and Buddhist sanga, tried his best to avoid the potential contradiction during the adaptation of Chinese Buddhism to socialism. Confronted with the inevitable social-political reformation launched by the powerful communist regime, Ju Zan did not fully surrender his faith, but did whatever he can to make Buddhism survive from the dramatic changes. / As the Korean War broke out in 1950, Chinese government decided to send a voluntary army to the front line and mobilized the whole country to support the war. Ven. Ju Zan reinterpreted the Mahayana doctrine, emphasized the concept of compassionate killing and repayment of kindness based on patriotism, justified the legitimacy of Buddhist participation in the Resist America Aid Korea Movement. And the Buddhists successfully proved their patriotic and political loyalty to the socialist regime. / The main purpose of this research is to survey how Chinese Buddhism adopted itself to socialism though the case study of Ven. Ju Zan during the founding period of People' Republic of China. Since the communists assumed power, the new government carried out a series of socialist reform in order to reconstruct the whole society. Buddhists also have to make adjustment for survival. / To adapt the ideological change after 1949, many progressive Buddhist scholars tried to link up Buddhism and Marxism, created a type of new Buddhist doctrine system with the name of Buddho-Marxist Syncretism. The main concern of these syncretists was clarifying the validity of Buddhism in the socialist era. Although Ven. Ju Zan believed that Buddhism and Marxism are consistent, he noticed the potential risk of excessive demonstration would blur the boundary between them. Then Ju Zan persuaded all the Buddhists should pay more attention to the Self Remolding Movement. / Ven. Ju Zan played a distinctive role as the leader during this adapting process. The innovation of Chinese Buddhism after 1949 can be regarded as the legacy of the Buddhist reform initiated by Master Taixu in the Republic period. Ju Zan is an excellent follower of Tai Xu and greatly influenced by him. Ju Zan is also an activist with critical consciousness. After the death of Mater Tai Xu in 1947 and the triumph of Chinese Communist Party in the civil war, he decided to cooperate with the communist regime and carry out the refonn of Buddhism. O Fórum Gaúcho de Saúde Mental e os argumentos sobre a reforma psiquiátrica no Rio Grande do Sul : relações sociais e princípios de justificação

Ce mémoire est a comme sujet les arguments produits dans les litiges qui impliquent les conceptions sur la santé mentale, présente dans les justifications et les actions des acteurs sociaux impliqués dans les discussions relatives à la maladie mentale au Rio Grande de Sul, dans un contexte de Contre-Réforme Psychiatrique. Cette étude vise à déterminer si c'est à partir de certaines relations sociales parmi les acteurs sociaux que les justifications et les actions sont produites. L'objet de cette étude est composé par les relations sociales et les arguments produits par les membres du Forum Gaucho de Santé Mentale à Porto Alegre. On utilise l’analyse relationnelle ainsi que la référence théorique de Michel Foucault et la sociologie pragmatique de Luc Boltanski. Il a été constaté qu’à partir d’une mise en cause sur la manière dont la configuration du Forum Gaucho de la Santé Mentale se concentre sur les principes de la justification employée par les acteurs sociaux appartenant au groupe, qu'il y a des idées différentes sur la santé mentale au sein du Forum et qu’il existe une prévalence de certains principes de la justification dans le discours une fois qu’il y a une prévalence de certains acteurs sociaux qui dépeint le discours. Pour bien mener cette investigation, dix représentants du Forum Gaucho de la santé mentale à Porto Alegre ont été interviewés et on a utilisé comme procédure méthodologique l'observation, ainsi que l'interview avec l'utilisation d'un script écrit. Après l'achèvement des entretiens, on a fait l'analyse de contenu. / Esta dissertação tem como tema os argumentos produzidos nas disputas envolvendo as concepções sobre saúde mental, presentes nas justificações e ações dos atores sociais envolvidos nas discussões referentes à Reforma Psiquiátrica, no Rio Grande de Sul, em um contexto de Contra-Reforma Psiquiátrica. O presente estudo tem como finalidade verificar se é a partir de determinadas relações sociais entre os atores sociais que as justificações e as ações são produzidas. O objeto deste estudo são as relações sociais e os argumentos produzidos pelos integrantes do Fórum Gaúcho de Saúde Mental de Porto Alegre. Utiliza-se a análise relacional, bem como o referencial teórico de Michel Foucault e a sociologia pragmática de Luc Boltanski. Foi verificado, a partir do questionamento de como a configuração do Fórum Gaúcho de Saúde Mental incide sobre os princípios de justificação mobilizados pelos atores sociais pertencentes ao grupo, que há concepções diferenciadas sobre saúde mental dentro do Fórum e há a prevalência de determinados princípios de justificação no discurso porque há uma prevalência de determinados atores sociais que veiculam este discurso. Para a realização desta pesquisa, foram entrevistados dez representantes do Fórum Gaúcho de Saúde Mental de Porto Alegre e utilizou-se como procedimento metodológico a observação, bem como a entrevista com o uso de um roteiro escrito. Após a realização das entrevistas, efetuou-se a análise de conteúdo. / The subject of this thesis is composed by the arguments produced in disputes involving conceptions about mental health present in the justifications and actions of social actors involved in discussions relating to Psychiatry Reformation in Rio Grande de Sul, in a context of Psychiatry Counter-Reformation. This study aims to check whether it is from certain social relations among the social actors that justifications and actions are produced. The object of this study is composed by the social relations and the arguments produced by members of the Forum Gaucho of Mental Health in Porto Alegre. It is used the relational analysis as weel as the theoretical reference to Michel Foucault and to the Luc Boltanski’s pragmatic sociology. It was found from the question of how the configuration of the Forum Gaucho of Mental Health focuses on the principles of justification employed by social actors belonging to the group, that there are different ideas about mental health within the Forum and there is the prevalence of certain principles of justification in the speech because there is a prevalence of certain social actors who portrayed the speech. To carry out this survey, ten representatives of the Forum Gaucho of Mental Health in Porto Alegre were interviewed and it was used as a methodological procedure the observation, as well as the interview using a script writing. After the nterview completion, it was the analysis of content.

Presentations of masculinity in a selection of male-authored post-apartheid novels

Thesis (MA (English Literature))--University of Stellenbosch, 2005. / In this thesis I examine the presentations of masculinity in several novels published in the post-apartheid period in South Africa, that is, the period after 1994. The novels under discussion are all male-authored texts and include novels by J M Coetzee (1999), André Brink (2000), Phaswane Mpe (2001), K Sello Duiker (2001), Zakes Mda (2002) and Damon Galgut (2003). In the introduction theoretical issues regarding masculinity are discussed on the basis of Morrell (2001) and a broad framework for the thesis is outlined. Subsequently the presentation of masculinity is analysed in each of the respective novels under discussion. Issues such as a definition of masculinity (or rather, masculinities), the interaction between men as friends, as colleagues; as well as issues such as heterosexuality and homosexuality are discussed. What perspectives does the author provide on masculinity? How do the male characters experience the new South Africa? What is the nature of their interaction with the female characters in the novels? Another aspect dealt with is the repression of homosexual desire for another man and the way in which it is suppressed beneath a macho façade. In the conclusion the different perspectives are compared and similarities and differences are briefly pointed out. In the end, an important question that comes to mind is: Do these men present a different type of masculinity emerging in the period after liberation, or do they merely (as depicted by their authors) perpetuate the patriarchal masculinities associated with the period before 1994?
