Företaget som studien utförts på upplever ett problem vid hanteringen av underhållsarbete i fabriken. Processerna för felrapportering och beställning av reservdelar är tidskrävande och involverar arbete från flera olika parter. Merparten av allt underhållsarbete som utförs är av oplanerat slag och ett fungerande uppföljningsarbete för att komma till rätta med de pro-duktionsstörningar som äger rum i processen saknas. Syftet med examensarbetet har varit att skapa underlag för att förbättra företagets effektivitet genom att optimera underhållsarbetet med hjälp av förslag till ett underhållssystem och olika tillvägagångssätt för att effektivisera underhållsarbetet. Under arbetets gång har en underhållsloggbok utformats i Excel. Loggboken används till att logga alla underhållsarbeten som inträffar i produktionen. Dessa arbeten ligger till grund för merparten av de beräkningar som utförts under projektet. Beräkningarna syftade till att kartlägga var i processen flest fel inträffar och av vilken karaktär dessa fel är. För att påvisa potentialen i ett utvecklat underhållsarbete beräknades även vilka vinstmöjligheter som kan åstadkommas genom implementering av ett underhållssystem och nya arbetsmetoder. Studien resulterade i ett förslag till ett underhållssystem som anses lämpligt utefter företagets situation och problem. Utöver införandet av ett underhållssystem stod det även klart att fö-retaget behöver minska mängden oplanerade produktionsstopp. För att lyckas med detta är det viktigt att operatörerna involveras i underhållsarbetet och utför alla arbeten av enklare slag. Underhållsavdelningen kan då, tack vare mer frigjord tid fokusera på att finna grundor-saker till problem och utföra planerade underhåll för att minska mängden oplanerade under-håll. Att effektivisera underhållsarbetet och införa Total productive maintenance (TPM) är en tidskrävande process som kan ta många år. För att underlätta för företaget och minska risken för att trilla tillbaks i gamla vanor utformades en implementeringsmodell för TPM. Modellen är ämnad som ett framtida arbete för företaget i arbetet med att utveckla effektiviteten på företaget ytterligare. / The company in the study was experiencing a problem when it comes to dealing with maintenance-work. The processes for bug tracking and ordering of spare parts is time con-suming and involves work from several different people. Most of all maintenance work performed is unplanned and a functioning follow-up work is missing. The purpose of the thesis was to help improving the company's efficiency by optimizing the maintenance work. This was achieved by a proposal for a maintenance system and vari-ous approaches to improve the maintenance work. During the project a maintenance log book was been designed in Excel. The log book is used to log all maintenance activities that occur in the production. These logs are the basis for most of the calculations carried out during the project. The calculations aimed to identify where in the process most errors occur and what the na-ture of those errors are. To demonstrate the potential of a maintenance system the profit opportunities that can be achieved through the implementation of a maintenance system and new working methods were calculated. The study resulted in a proposal for a maintenance system that suited the company's needs, situation and problems. In addition to the introduction of a maintenance system, it was also clear that the company needs to reduce the amount of unplanned downtime. To achieve this it is important to involve the operators with maintenance work and they shall perform all the works of the simpler kind. The maintenance department can then, thanks to the re-leased time, focus on finding the root causes of problems and perform scheduled mainte-nance to reduce the amount of unplanned maintenance. Improving the maintenance work and introduce Total Productive Maintenance (TPM) is a time consuming process that might go on for years. In order to facilitate the work and re-duce the risk of falling back into old habits an implementation model was designed for the TPM-work. The model is intended as a future work for the company to facilitate the work of reaching even higher efficiency.

Foram realizadas pesquisas em documentações e registros sobre as necessidades de melhorias na usina de cana-de-açúcar no período de 1997 a 2003, época em que as preocupações para manter a organização, a limpeza, o bem estar no ambiente de trabalho e as conquistas no mercado eram necessárias para o crescimento industrial. Neste período foram analisadas todas as etapas de implantação do Programa 5 S, incluindo um programa de apoio para o 4 e 5 S, denominado POL – Programa de Organização e Limpeza. La presente investigación tiene como objetivo general: Implementar la metodología 5S para mejorar el desempeño laboral del área de producción de la Corporación Industrial DAAZUR E.I.R.L. Huancayo – Junín, 2017. La investigación es de tipo aplicada, apoyada en un diseño pre experimental, de tipo explicativo. Teniendo como resultados: el diagnóstico del estado actual de la empresa, en relación a los aspectos claves de la metodología de las 5S fue crítico con valores: Seiri = 18, Seiton = 20, Seiso = 18, Seiketsu = 17 y Shitsuke = 15 siendo el puntaje esperado 50 y en relación al desempeño laboral el resultado dio deficiente en un 60%, al implementar la metodología 5S el resultado mejoro notoriamente: Seiri = 48 mejorando 60%, Seiton= 48 mejorando un 56%, Seiso= 49 mejorando 62%, Seiketsu = 49 mejorando 64% y Shitsuke =49 mejorando 68%. Así mismo, la eficacia laboral de los trabajadores mejoró significativamente (p<0.05) de Deficiente 20% y Regular 80% a Excelente 100%, la eficiencia laboral de los trabajadores mejoró significativamente (p<0.05) de Deficiente 60% y Regular 40% a Excelente 100%, la calidad laboral de los trabajadores mejoró significativamente (p<0.05) de Deficiente 60% y Regular 40% a Excelente 100%, la economía laboral de los trabajadores mejoró significativamente (p<0.05) de Deficiente 100% a Excelente 100%. Conclusión: Al implementar la metodología 5S, se logró mejorar significativamente (p < 0.05) el desempeño laboral, en sus cuatro dimensiones Eficacia, Eficiencia, Calidad y Economía, de los trabajadores del área de producción de la Corporación Industrial DAAZUR E.I.R.L. Pasando el resultado del desempeño laboral de Deficiente 60% y Regular 40% a Excelente 100%. El cual se acepta con un 95% de confianza.

L’ARNr 5S est une petite molécule d'ARN codée par le noyau, qui est partiellement adressée dans les mitochondries dans les cellules humaines. Bien que le mécanisme d'importation a été étudié, la fonction de ce phénomène est mal comprise. Les données publiées suggèrent que l’ARNr 5S importé pourrait participer à la synthèse des protéines mitochondriales, éventuellement être associé aux mitoribosomes. Cependant, les études structurelles ne supportent pas ces observations. Pour comprendre le rôle de l’ARNr 5S dans les mitochondries humaines, nous avons exploité plusieurs approches. Nous avons montré que seule une petite fraction de l’ARNr 5S pourrait être associé à mitoribosomes humaines, insuffisante pour former un complexe stoechiométrique 1:1. Nous avons appliqué l’approche de chromatographie d'affinité à l’ARN MS2 pour identifier les partenaires protéiques de l’ARNr 5S importé. Les résultats suggèrent que l’ARNr 5S pourrait être associé à des protéines ribosomales et des facteurs mitochondriaux d'assemblage du ribosome mitochondrial. Cela nous a permis de formuler une hypothèse sur la participation de l'ARNr 5S dans la biogenèse mitoribosomale. Ces données ont été partiellement validées par des expériences de co-immunoprécipitation, bien que plusieurs expériences sont encore nécessaires pour valider la participation ARNr 5S dans cette voie. / 5S rRNA is a small nuclear-encoded RNA molecule, which is partially imported into mitochondria from cytoplasm in human cells. Although the import mechanism was studied in details, the functional significance of this phenomenon is poorly understood. Published data suggest that imported 5S rRNA might participate in mitochondrial protein synthesis, possibly associating with mitoribosomes. However, structural studies do not support these observations. We have exploited several approaches to figure out the function of 5S rRNA in human mitochondria. Our studies showed that only a minor part of 5S rRNA pool could be associated with human mitoribosomes, insufficient to form stoichiometric 1:1 complex. We applied MS2 affinity chromatography approach to identify protein partners of 5S rRNA in human mitochondria. The results suggested that 5S rRNA could associate with mitochondrial ribosomal proteins and mitochondrial ribosome assembly factors. This allowed us to formulate a hypothesis about possible participation of 5S rRNA in mitoribosome biogenesis. The data were partially validated by co-immunoprecipitation experiments, although subsequent studies are required to validate 5S rRNA involvement in this pathway.

On behalf of Skanska, the goal is to develop proposals for improving the work environment around the waste management to improve the sorting of construction waste to ultimately reduce costs and improve the order of the workplace. The study involves comparing two construction stages erected by Skanska with Riksbyggen contracting Sandåkern in Umeå. Why was there more residue in the second stage than in the first? What can be useful for future projects and what opportunities should we create in order to reach a continuous improvement on both the working environment and sorting of waste? Interviews are the basis for some of the conclusions about the differences in the amounts of waste at the two stages and are included as appendices, at the end of this thesis. A combined result from the questions asked during the introduction has resulted in an "Easy to do right" - template. Where the idea is that the template is to become a poster at the workplace in size A3. The poster will contain small, simple tips on how waste can be reduced and the work environment could be improved. This can be achieved by sorting through the materials available on site and keep the necessary things, which will improve the overview of what is available and fewer materials would break or disappear due to a messy environment. It is good to use both breaks and lunches to dispose of materials with the “feed-cart” and bring the cart back after the break. Let everyone participate in how to developing the workplace and what kind of daily routines it should have. It makes everyone become more committed to keep on doing it. A cable that hangs down from the ceiling or the floor could cause an injury, this need to be taken care of by securing it so no one will get hurt. Holes that may exist should also be covered so that no one will trip and fall. Sorting out the fractions will save money since then the waste company doesn’t have to charge extra for their sorting of the recycled materials. When everyone keeps a high level of cleanliness it reduces the spread of dust, clutter in the workplace and prevents materials from getting destroyed which would result in an increased amount of waste. The management of construction waste can be improved with simple and sound means, which will create a safer work environment and a quieter life. Through these small, simple means there is both the environment and money to save and a sense of coming home healthy and happy to ones family at the end of the work day. It is something that we all want to achieve, not only construction companies, but also individual to be happy with their work. Let's put our heads together and build a better and safer future for both nature and the individual. / Examensarbetet är utfört efter ett förslag från Skanska Hus med målsättningen att ta fram förslag till förbättringar av arbetsmiljön kring avfallshanteringen och med den kunna förbättra sorteringen av byggavfallet för att i slutändan kunna minska kostnader och få en bättre ordning på arbetsplatsen. Studien handlar om att jämföra två etapper som utfördes av Skanska med Riksbyggen som byggherre på Sandåkern i Umeå. Varför blev det mer avfall på den ena etappen än den andra? Vilka delar kan vi ta med oss till kommande projekt och vilka möjligheter bör vi hitta för att nå en ständig förbättring kring både arbetsmiljö och sortering av avfall? Intervjuer ligger till grund för några av de slutsatser kring skillnaderna i avfallsmängderna på de båda etapperna och finns med som bilagor. (Bilaga 1, 5, 6) Resultatet från de frågeställningar som ställs under inledningsdelen ligger till grund för ”Lätt att göra rätt” – mall. Tanken att mallen ska kunna bli en poster ute på arbetsplatsen i A3 storlek. Där presenteras små enkla tips om hur avfallsmängder kan reduceras och arbetsmiljön förbättras. Avfallsmängden kan minskas genom att sortera materialet på plats och endast försöka ha det mest nödvändiga kvar. Det ger en bättre överblick över vad som finns och material som går sönder eller minskas. Det är bra att använda tiden runt raster och luncher att frakta bort material i fodervagn Starke Arvid och ta med vagnen tillbaka efter rasten. Alla ska vara delaktiga i hur arbetsplatsen utformas och vilka rutiner som bör finnas. Det gör att alla blir mer engagerade för för gjorda överenskommelser. Att vara noga med säkerheten och arbetsmiljön, t.ex. genom att hänga upp kablar från golv och tak och täcka eventuella hål för undvikande av fallolyckor. Genom att sortera upp fraktioner väl sparar det pengar på eftersortering men även återanvändning av återvunnet material. Om alla håller städningen på en bra nivå minskar dammspridningen, oreda på arbetsplatsen och materialförstörelse som resulterar i ökad avfallsmängd. Hanteringen av byggavfall kan förbättras med enkla och sunda medel, vilket skapar en tryggare arbetsmiljö och en lugnare arbetsdag. Genom dessa enkla små medel finns det både pengar att hämta, en miljö att rädda och en känsla av att komma hem hel och glad till sin familj i slutet av arbetsdagen. Det är något som vi alla vill uppnå, inte bara byggföretaget utan även individen som ska trivas med sitt arbete. Om vi slår våra kloka huvuden ihop så bygger vi en bättre och tryggare framtid för såväl naturen som individen.

L'ARNr 5S est une partie intégrante du ribosome indispensable pour la synthèse des protéines. Cette fonction essentielle nécessite une régulation fine de l'expression des ARNr 5S en fonction des exigences cellulaires. L'ARNr 5S est codé par des gènes organisés en répétitions en tandem principalement situés dans les régions péricentromériques des chromosomes 3, 4 et 5 dans le génome Col-0 d'Arabidopsis thaliana. L’étude approfondie de l’organisation, la dynamique et la régulation épigénétique des gènes d’ARNr 5S reste cependant difficile du fait de l’absence d’assemblage de ces régions hautement répétées dans la séquence du génome d’Arabidopsis disponible dans TAIR10.Dans cette thèse, nous apportons de nouvelles informations sur les gènes d'ARNr 5S chez Arabidopsis. Nous avons confirmé les signatures d'ADN spécifiques par séquençage haut débit, identifié des polymorphismes spécifiques des régions chromosomiques portant des ADNr 5S et ainsi défini de nouvelles séquences de référence. Nous avons également étudié le statut épigénétique des gènes ARNr 5S, montrant que ces gènes sont globalement enrichis en marques répressives. Les loci situés sur les chromosomes 4 et 5 présentent également des modifications post-traductionnelles d’histones et des variants d’histone particuliers et caractéristiques d’une activité de transcription. Nous avons aussi montré que les loci d'ADNr 5S sont très dynamiques au sein de l'espèce Arabidopsis avec des variations entre différent écotypes du nombre de copies de gènes d’ARNr 5S et de l’importance relative de chaque cluster. En utilisant l'écotype Ler et des mutants spécifiques, nous montrons que le locus du chromosome 5 est une source majeure de réarrangements chromosomiques qui affectent l'organisation de la chromatine dans le noyau. Enfin, nous suggérons que des variations de l’état chromatinien des gènes d'ARNr 5S peuvent conduire à une différence de transcription dans les différents écotypes.Cette étude permet d’établir un rôle de l'organisation de la chromatine dans la régulation transcriptionnelle et l'organisation des gènes d'ARNr 5S. / 5S rRNA is an integral component of the ribosome and is essential for protein synthesis. 5S rRNA expression is therefore tightly regulated to suit the cellular requirements. 5S rRNA is encoded by gene arrays organized in tandem repeats mainly situated in the pericentromeric regions of chromosomes 3, 4 and 5 in the Arabidopsis thaliana Col-0 genome. Full genome assembly remains challenging in these highly repeated regions and impedes further investigation of their organization, dynamics and epigenetic regulation.In this thesis, we provide information on 5S rRNA genes in Arabidopsis thaliana. We confirmed specific DNA signatures by deep sequencing and define chromosome-specific polymorphisms and new reference sequences. We also studied the epigenetic status of the 5S rRNA genes showing that these genes are enriched in repressive marks whereas the loci on chromosome 4 and 5 also display peculiar histone modifications and variants characteristic of transcriptional activity. We report that 5S rDNA loci are highly dynamic within the Arabidopsis species with high level of variation in global copy number and cluster proportion between ecotypes. Using the Ler ecotype, in which reorganization events were recorded, and specific mutant backgrounds, we identified chromosome 5 rDNA locus as a major source of variation and observed altered chromatin organization in nuclear space as a consequence of 5S rDNA locus variation. Finally, we suggest that differences in chromatin states at the two main 5S rDNA loci can lead to differential usage of 5S rRNA genes indifferent ecotypes.The analysis provides evidence for a role of chromatin organization in transcriptional regulation and 5S rDNA organization.

The master thesis is focused on a proposal of project implementation of 5S method for production system of LAUFEN CZ, s.r.o. in Znojmo. The proposal reacts to current economic situation and also to the status of company. This is the second phase of project with focusing on the production and completion of bathroom furniture. In master thesis are evaluated correct process and methods for successful implementation of project.

The thesis analyses a problem of improving the work environment and all the sections of an organisation. In the first part, the genezis, methods, theory and practical examples of Kaizen philosophy and 5S method are presented. In the second part, there is an instruction to make a system of presenting proposals of employees and the possible forms of its realisation. I dealt with assessment and finacial valuation of the given proposals. Finally, I propose documents for keepimg standards and increasing efficiency of the 5S method in a certain workplace.

In this work, a study has been conducted as to whether the industrial method The Visual Workplace can be adapted for use in retail trade industry. This might be extra interesting for businesses that conduct online commerce or trade behind the counter as these types of companies are most similar to the example in this work. The company that represents the trade industry in this work is Kronans apotek AB. This pharmacy is currently experiencing deficiencies in their inventory management, which results in lack of quality assurance. Consequences of this are that the staff cannot know for sure if the product sought is found in the notified storage area which impairs efficiency and productivity. The Visual Workplace aims to visualize work standards and routines. In this way, activities that do not add any value are eliminated while at the same time problems and deviations are immediately visible. This study will explore how this industrial method can be adapted to Kronan's pharmacy. / Något som ofta används inom både industri och handel är lager. I detta arbete har en undersökning gjorts som om huruvida den industriella metoden Den visuella arbetsplatsen kan anpassas för användning inom handeln. Extra intressant kan detta vara för verksamheter som bedriver näthandel eller handel bakom disk då dessa typer av bolag mest liknar exemplet i detta arbete. Företaget som representerar handeln i detta arbete är Kronans apotek AB. Detta apotek upplever idag brister inom deras lagerhantering som resulterar i bristande kvalitetssäkring. Konsekvenser av detta är att personalen inte med säkerhet kan veta om eftersökt vara återfinns på aviserad lagerplats vilket försämrar effektivitet och produktivitet. Den visuella arbetsplatsen syftar till att visualisera arbetsstandarder och rutiner. På så sätt elimineras aktiviteter som inte tillför något värde samtidigt som att problem och avvikelser direkt blir synliga. Genom arbetets gång undersöks hur denna industriella metod kan anpassas till att bli applicerbar till Kronans apotek.
