Nop53 e uma protena nucleolar, conservada evolutivamente e essencial na levedura Saccharomyces cerevisiae para a biogênese da subunidade maior do ribossomo, 60S. O principal fenotipo causado pela repressão da expressão de Nop53 e o acumulo do intermedi ario de processamento de pre-Rrna, 7S, que tambem e substrato do complexo exossomo na formação do rRNA maduro 5:8S. Nop53 interage diretamente com a subunidade do exossomo Rrp6 e com a subunidade Mtr4 do co-ativador do exossomo TRAMP. O objetivo principal deste trabalho foi o de analisar como a interação entre Nop53 e o exossomo pode modular a atividade deste ultimo. Para isso, foram utilizados metodos bioqumicos, geneticos e de biologia molecular. Os resultados mostrados aqui demonstram que a depleção de Nop53 faz com que mais protenas ribossomais, principalmente da subunidade maior, sejam co-imunoprecipitadas com o core do exossomo, sugerindo que Nop53 possa ter um papel na liberação do exossomo da subunidade pre-60S depois da formação do rRNA maduro 5:8S. Esta hipotese foi conrmada atraves da separação de complexos por centrifugação em gradiente de glicerol, que mostrou a presenca de subunidades do exossomo em complexos maiores na ausência de Nop53, provavelmente correspondendo a partculas pre-ribossomais. Co-imunoprecipitação de RNA com o exossomo na ausência de Nop53 tambem conrmou uma maior associação deste complexo com o pre-rRNA 7S. Como tambem mostrado aqui, alem de interagir com Rrp6, Nop53 interage com subunidades do core do exossomo e a superexpressão de uma destas subunidades, Rrp43, complementa parcialmente a ausência de Nop53 na celula. Estes resultados levaram a conclusão de que Nop53 pode recrutar o exossomo para a partcula ribossomal pre-60S para a maturação do pre-rRNA 7S a 5:8S, e atue tambem na liberação do exossomo, possivelmente atraves de sua interação com a helicase Mtr4. / Abstract Nop53 is a nucleolar, conserved and essential protein in the yeast Saccharomyces cerevisiae, involved in the biogenesis of the large ribosomal subunit 60S. The main phenotype of the depletion of Nop53 in yeast cells is the accumulation of the prerRNA processing intermediate 7S, which is also the substrate of the exosome complex for the formation of the mature rRNA 5:8S. Nop53 directly interacts with the exosome subunit Rrp6, and with the subunit Mtr4 of the TRAMP complex, an exosome co-activator. The main objective of this work was the analysis of the interaction between Nop53 and the exosome and the identication of the mechanism through which Nop53 regulates the exosome activity. The results shown here demonstrate that the depletion of Nop53 leads to a more stable association of the exosome with the pre-60S ribosome particle, as determined by co-immunoprecipitation of proteins with one of the exosome core subunits, and by fractionation of complexes through glycerol gradients. These results suggested that Nop53 could play a role in the release of the exosome after the formation of the mature rRNA 5:8S. This hypothesis was conrmed through the co-immunoprecipitation of pre-rRNA 7S with the exosome in the absence of Nop53. In addition to the interaction with the exosome subunit Rrp6, as shown here, Nop53 also interacts with core subunits of the complex. Interestingly, overexpression of one of these subunits, Rrp43, partially complements the depletion of Nop53. These results led to the conclusion that Nop53 may recruit the exosome to the pre-60S particle for the maturation of the pre-rRNA 7S to the mature 5:8S, but Nop53 may also be involved in the release of the exosome, possibly through its interaction with the helicase Mtr4.

Utopické prvky Christianie: Hledání ideální společnosti / Utopian Elements of Christiania: Search for an Ideal SocietyMedová, Lenka January 2011 (has links)
Presented diploma thesis is based on seven-months fieldwork in the Danish commune of Christiania. It sustains a heritage of utopian ideas of a contraculture of the 60s on which it has been created. The thesis examines which aspects of utopia are presented here and asks about the character of these utopian visions. It focuses on the question how the dream, that locals wanted to implement, is still presented in their minds, and how they perceive its realization. I used examples of street art, that is plentifully presented here, as a mediator of ideological background of this community. Its symbolism allowed me to approach values, ideas and world-views of people who already have been living in this area for almost forty years. I used the principles of visual anthropology to reveal meanings contained in visual manifestations in a public space. My research is focused only on people who moved to Christiania in 70s that means those who were present here in the times of forming this community, and who were a part of revolting atmosphere of the 60s and of its ideals. Methods used during my fieldwork are a nonparticipant observation, informal, semi-structural and photographic interviews

Totalität und Ganzes versus Ausschnitt und Detail : Normbewahrung und Normveränderung im deutschsprachigen roman- und literaturtheoretischen Diskurs der 60er JahreMetzler Widmark, Cornelia January 2005 (has links)
This study is a thematic-descriptive investigation of the reproduction and transformation of norms in the theoretical discourse on the novel during the 1960s. Primary literature consists of articles and essays published in West German literary and cultural journals 1959-1967. The term ‘discourse’ is applied partly in accordance with Busse/Hermanns/Teubert (1994), the term ‘theory of the novel’ chiefly in accordance with Lämmert (ed. 1984). ‘Ideology’ is not used in the sense of ‘false ideology’ but rather as an umbrella term for various types of value-related statements. From this, the theory-of-the-novel discourse is perceived as an aesthetic-ideological discourse, containing statements directed at the contemporary novel which have clear programmatic function and significant thematic width. The objective of the investigation is to show that specific comprehensive thematic fields – Werteverlust (breakdown and loss of values), Subjektproblematik (‘problematisation of the concept of the subject’), Sprachproblematik (language related problems) and Realitätszerfall (reality loss, breakdown of the reality concept) – bear discursive significance as regards the discussion of literary norms during the 1960s, and that this discussion realises itself as two aesthetic-ideological discourses competing for interpretative precedence. The major issues are: Which reiterated patterns of argumentation, i.e. norm-related categories, concepts and rhetorical patterns, are used in the discourses for diagnoses and programmatic imperatives? How are the comprehensive thematic fields accentuated? What is treated, postulated or set aside as ‘truth’? How - based on the above – is the novel formulated as a ‘problem’ (‘crisis of the novel’)? The investigation confirms that the comprehensive thematic fields are particularly central to the theoretical discussion of literature in the 1960s. This manifests itself as a discursive re-evaluation process which may be characterised as a conflict between an ‘aesthetic-conservative discourse’ and a ‘discourse of change’ (‘Veränderungsdiskurs’) where the right to define and evaluate the novel in terms of literature is at stake. It is in the collision between these two discourses and their largely incompatible concepts of literature that the novel discursively becomes a ‘problem’. The discourses are maintained by specific reiterated patterns of argumentation which in the investigation are subsumed under the following headings: die negative Modernität (negative modernity), das bloß Moderne (phenomena of ‘fashionable character’, simply expressing trends) and das Überzeitliche und das Ganze (the timeless and the totality); respectively die traditionelle, bürgerliche Gesellschaft (traditional bourgeois society), die technisch-sprachliche Realität (technolinguistic reality) and der subjektive, sprachliche Realitätsausschnitt (‘subjective language based slice of reality’). The first group of argumentation patterns is linked to universal, ‘eternal’ and essential categories and inherited norms, ethical-aesthetical educational grounding and a ‘rhetoric of the spirit’ or of ‘mankind’, oriented around a specific reception of German Classicism and Idealism, a downgrading of the present and an upgrading of the past. The other group embraces an incipient constructivism, contextually bound and societal categories and norms as well as implicitly critical programmes of enlightenment, devaluing the past and ‘acknowledging’ rather than criticising the present. In doing so they tend rather to realise a rhetoric of the linguistic and political reality and of more modest programmatic proposals.

Metafiction, historiography, and mythopoeia in the novels of John FowlesBuchberger, Michelle Philips January 2009 (has links)
This thesis concerns the novelist John Fowles and analyses his seven novels in the order in which they were written. The study reveals an emergent artistic trajectory, which has been variously categorized by literary critics as postmodern. However, I suggest that Fowles's work is more complex and significant than such a reductive and simplistic label would suggest. Specifically, this study argues that Fowles's work contributes to the reinvigoration of the novel form by a radical extension of the modernist project of the literary avant-garde, interrogating various conventions associated with both literary realism and the realism of the literary modernists while still managing to evade a subjective realism. Of particular interest to the study is Fowles's treatment of his female characters, which evolves over time, indicative of an emergent quasi-feminism. This study counters the claims of many contemporary literary critics that Fowles's work cannot be reconciled with any feminist ideology. Specifically, I highlight the increasing centrality of Fowles's female characters in his novels, accompanied by a growing focus on the mysterious and the uncanny. Fowles's work increasingly associates mystery with creativity, femininity, and the mythic, suggests that mystery is essential for growth and change, both in society and in the novel form itself, and implies that women, rather than men, are naturally predisposed to embrace it. Fowles's novels reflect a worldview that challenges an over-reliance on the empirical and rational to the exclusion of the mysterious and the intuitive. I suggest that Fowles's novels evince an increasingly mythopoeic realism, constantly testing the limits of what can be apprehended and articulated in language, striving towards a realism that is universal and transcendent.

Pepper's Lonely Hearts Club Band, and in the Teatro do Absurdo (The Absurd Theater). We have the art as the expression of the spirit of that time in the development of the human thinking, which we treat here manifested in the daily life of each time. So we intend to close this paperwork showing some aspects that make possible the convergence of all these elements in the XXI century, tracing along the way the consequences of this way of thinking and dealing with the world. / O presente trabalho presta-se a traçar, a estabelecer uma relação entre o pensamento de Francis Schaeffer ao movimento da contracultura; o movimento contracultural da década 60 principalmente relativo aos anos de 67-69 com os beatniks, o Flower Power, Timothy Leary e seus desdobramentos mais marcantes; e, por fim, as expressões contraculturalistas no uso das drogas, no álbum dos The Beatles, Sgt. Fonction de la protéine cellulaire RISP (Reinitiation Supporting Protein) dans la reinitiation de la traduction chez les plantes / Functional role of the Reinitiation Supporting Protein (RISP) in plant translation initiation and reinitiationMancera-Martinez, Eder Alberto 24 November 2014 (has links)
Chez Arabidopsis, la protéine RISP est détournée par le virus CaMV pour assurer, ensemble avec la protéine virale TAV, la traduction de son ARN polycistronique. RISP a été identifiée comme une cible de la voie de signalisation de TOR et il a été montré que sa phosphorylation est requise pour promouvoir la réinitiation de la traduction activée par TAV. Les résultats que j’ai obtenus suggèrent que RISP, lorsqu’elle n’est pas phosphorylée, intervient ensemble avec eIF3, au niveau du complexe de pré-initiation 43S pour recruter le complexe ternaire grâce à l’interaction entre RISP et la sous-unité b du facteur eIF2. Il s’est avéré que RISP a la capacité, lorsqu’elle est phosphorylée, d’interagir non seulement avec la protéine ribosomique eL24 mais également avec eS6. Nos résultats indiquent que la liaison entre les sous-unités ribosomiques 60S et 40S sous l’effet de RISP, est régulée par la voie de TOR et qu’elle joue un rôle important dans le contrôle de la réinitiation de la traduction. / Many factors are required to recruit the tRNAi and a 60S ribosomal subunit to the 40S ribosomal subunit preinitiation complex. This recruitment is normally strictly limited during reinitiation of translation if factors recruited during the primary translation event are shed from 40S. However, factor retention can occur during long ORF translation if the CaMV viral factor TAV is present. RISP is a downstream target of TOR and found either within the 43S preinitiation complex, if bound to eIF3, and/or attached to 60S, if phosphorylated by TOR. We show here that RISP interacts with subunit b of eIF2 before phosphorylation. Critically, TOR activation up-regulates phosphorylation of both RISP and eS6 as well as the binding of both factors. Importantly, eS6-deficient plants are less active in TAV-mediated reinitiation and are thus less susceptible to CaMV infection. It is attractive to propose that eS6 phosphorylation contributes to retention and re-use of 60S during 40S scanning.
