Cette thèse s’intéresse aux phénomènes d’hydratation de composés organiques à l’échelle moléculaire. Des méthodes basées sur une fonction d’onde multi-déterminantale sont capables de rendre compte des phénomènes de hydratation avec une précision approchant la réalité expérimentale. Or, ces méthodes sont limitées par la taille du système. L’utilisation de la DFT semble indispensable à une étude de complexes, même pour un nombre limité de molécules d’eau. Il s’avère que ces méthodes ne prennent pas en compte les interactions de nature dispersive. Des corrections empiriques ont été proposées récemment pour palier à ce problème. Cependant, ces corrections ne s’appliquent qu’à l’énergie et sur la géométrie des complexes hydratées, la fonction d’onde n’étant pas affectée par la correction. D’autres alternatives pour la prise en compte des effets de dispersion reposent sur l’emploi de méthodes hybrides fonction d’onde/DFT. Ceci peut s’effectuer en introduisant une séparation de portée dans le traitement des interactions électroniques. L’un des objectifs de cette thèse consiste à proposer une nouvelle méthode double hybride à séparation de portée permettant une bonne description des phénomènes d’hydratation. L’autre objectif de cette thèse consiste à utiliser des outils topologiques permettant la prédiction de composés organiques hydraté par l’étude du potentiel électrostatique moléculaire et la caractérisation de ces interactions non covalentes par la théorie AIM. / This thesis deals with hydration phenomena of organic compounds at the molecular scale. The Schrodinger equation considered within the Born-Oppenheimer approximation and within a non-relativistic context contains all the physics necessary to describe in particular the micro-solvation of organic compounds. Methods that are based on a multi-determinant wave function are able to account for micro-hydration phenomena with a precision approaching the experimental reality. These methods are limited by the size of the system. The use of DFT seems necessary for a study of complexes, even for a limited number of water molecules. It turns out that these methods do not take into account dispersive interactions. Empirical corrections have recently been proposed to address this problem. However, these corrections apply only to the energy and to the geometry of the hydrated complexes, the wave function not being affected by the correction. Other alternatives for taking into account dispersion effects using double-hybrid methods should thus be considered. This can be done by introducing a range separation on the electronic interactions. There are two main objectives in this thesis. The first one is to propose a new double-hybrid method with range separation allowing a satisfactorily description of the hydration phenomena at the molecular scale. The second objective consists in using topological tools allowing the prediction of hydrated organic compounds using the electrostatic molecular potential and the characterization of these non-covalent interactions by the "Atoms in molecules" theory.

In this thesis, Sebilwane is the subject of a narratological investigation. The point of departure of this study is based on the fact that a narratological text consists of three levels: the history, the composition, the usage of words which are recognisable in the style of the author. The epic-poem is not the subject of a verse-technical investigation and description. The narratological model is adapted to the aim of this study. The historical level regarded in principle as the original level prior to the material's exposure to a viewpoint and it is interpreted. The four narrative elements that are investigated are: the events, the characters/actors, time and place. In Sebilwane there are main events identified by the criteria of: (a) change, (b) cause, and (c) result. The characters/actors have been described and classified according to: (a) aim, (b) supporter, (c) patron, (d)helper and patron,and (e) opponent. In as far as historical time is concerned, it has been concluded that the events occurred: (a) in the remote past, and (b) stretched it to a 24 hour period. The actors/persons find themselves in a rural area which can be comparable to Botlokwa which is lying within the borders of Lebowa. The composition of the information which is given in the historical level, gives the shape of the author's aim. Here, what is important, are the functions which are described by the elements themselves. Then the idea of the theme comes clearly in this part and it is therefore identified as the main - and sub-theme. The third level concerns the usage of words; the information now gets a personal or subjective selection. Therefore, only a short passage is to be selected for stylistic analysis. The analytic model which is effected here is Kerkhoff's. AFRIKAANS : In hierdie verhandeling word Sebilwane aan 'n narratologiese ondersoek onderwerp. Die uitgangspunt van hierdie studie is dat 'n narratologiese teks uit drie lae bestaan: die geskiedenis, die samestelling, die verwoording wat in die styl van die outeur kenbaar is. Hierdie epiese gedig word nie verstegnies ondersoek en beskryf nie. Die narratologiese model is vir die doel van hierdie studie aangepas. Die geskiedenislaag word in beginsel as die oorspronklike laag beskou voordat die gegewens vanuit 'n bepaalde gesigspunt bekyk en weergegee word. Die vier vertelelemente wat ondersoek word, is die gebeurtenisse, die karakters/akteurs, tyd en plek. In Sebilwane is die kerngebeurtenisse geïdentifiseer deur die kriteria van (a) verandering, (b) oorsaak en (c) afloop. Die karakters/akteurs is beskryf en geklassifiseer volgens (a) doelstelling, (b) begunstigde, (c) begunstiger, (d) helper en (e) teëstaander. Wat die tyd betref, speel die gebeure (a) histories in die verre verlede af, en (b) strek dit oor 'n 24 uur tydperk. Die akteurs/mense bevind hulle in 'n landelike gebied wat waarskynlik Botlokwa is wat binne Lebowa geleë is. Die samestelling van die gegewens wat in die geskiedenislaag gegee is, gee aan die doestelling van die outeur gestalte. Daarvan gaan dit hier om die funksies wat aan die elemente toegesê word. Neben der f&uuml;r SMS charakteristischen schnellen &Uuml;bermittlung von Textnachrichten bietet Instant Messaging weitere Vorteile: es ist f&uuml;r den Sender auf den ersten Blick ersichtlich, ob der Gespr&auml;chspartner f&uuml;r eine Kommunikation zur Verf&uuml;gung steht. Neben Textnachrichten k&ouml;nnen aber auch Informationen wie Bilder, Videos oder andere Formate &uuml;bermittelt werden. <br> <br> Auch f&uuml;r Organisationen ist ein Einsatz von Instant Messaging sehr interessant. Implementation of Community-Based Lifestyle Programs for Individuals with Type 2 Diabetes Mellitus in Southwest and Central Virginia: Formative and Process Evaluation

Type 2 Diabetes Mellitus (T2D) is a major public health issue. Diabetes prevalence is growing and is the 7th leading cause of death in the US. Virginia has a slightly higher prevalence than the national average. Community-based diabetes lifestyle management programs that include a physical activity component are effective at improving glycemic control and influencing lifestyle behavior changes among people with T2D. The Balanced Living with Diabetes (BLD) program uses an active learning approach to improve glycemic control and healthful lifestyles. The Lifelong Improvements through Fitness Together (LIFT) program uses behavioral strategies to improve functional fitness, participant engagement, and program adherence. Participant retention is a challenge for community-based program. Participants may start the program, but then fail to complete the program and/or adhere to program recommendations. Two pilot studies were conducted to evaluate strategies for increasing participant retention and improving outcomes. The first evaluated the implementation and impact of a retention plan implemented in BLD programs conducted from 2015-2017. There were more participants returned to the reunion class session prior the development of the retention plan. There were improvements in some health behavior and self-efficacy indicators in programs that used the retention materials, however, impacts on outcomes were mixed. Preliminary findings showed that the retention plan could be a tool for providing additional support to participants, however strategies for dissemination of the retention plan needs to be reevaluated. The second study evaluated the impact on participant engagement and program outcomes when the LIFT program is incorporated with the BLD program. There was an increase in participant retention, self-efficacy and one health behavior for physical activity and health behaviors related to diet in BLD+LIFT programs. Extension Agents are willing to implement the BLD+LIFT programs, however, clarity of program logistics is needed prior to implementation. Extension Agents indicated that implementation of these programs in locations with older adults that have greater disability may not be the best locations. Having larger studies on the effect of incorporation of LIFT with the BLD with older adult populations that have fewer co-morbidities are needed to determine the impact of addition of the LIFT program with the BLD program on program outcomes. / Doctor of Philosophy / Type 2 Diabetes is a chronic disease in which the body does not use insulin as it should or does not produce enough insulin. The Centers for Disease Control and Prevention (CDC) indicates that the prevalence of diabetes was 23.1 million among US adults in 2015. The prevalence of diabetes in Virginia is slightly higher than the national average. Community-based diabetes lifestyle management programs that include a physical activity component are effective in improving glycemic control and influencing lifestyle behavior changes among people with T2D. The Balanced Living with Diabetes (BLD) program uses an active learning approach to influence better glycemic control and healthful lifestyles. Expanding the conduct of BLD programs that incorporate the LIFT program can be effective in improving glycemic control and increasing physical activity, however, working with organizations that service people that have diabetes with fewer health conditions that limit physical activity may be more effective.

A number of strict requirements were however set for such a claim. For a considerable time a claim has been recognised in English and German law even where a substitute was not hired and where the article was used for pleasure purposes. It is submitted that this should also be the position in South African law because the loss of the use of a thing per se has an independent value. It is further submitted that the interest on capital value method (as per English law) can be used as starting-point in the determination of quantum. Standing on the toes of giants : social movement theory and the case of the learning organisation

The thesis examines two interrelated topics; the perception of management theory in general, and the efficacy of the learning organisation in particular. The purpose of this thesis, therefore, is to compare the rhetoric espoused in the learning organisation literature with a survey of senior managers in large, commercial organisations. The results revealed a positive disposition towards business and management theory in general. The survey also demonstrated that leadership is considered the most important variable in the success of a company. This is facilitated through the attraction and retention of the appropriate staff and creating sufficient space for them to operate. Learning is embedded by anticipating the future, learning from the past and enabling good communication. However, the latter is balanced through change management. This is derived through legitimate authority and a reliance on planning. Finally, a strategy of experimentation is balanced by challenging industry rules. Themes involving politics, corporate alignment and corporate longevity were found to have relatively little convergent validity. The third section of the questionnaire reveals a positive disposition towards the learning organisation. The contribution of this thesis is in three areas. First, it is the only work to evaluate the anti-guru school. Second is the development of a syncretic model of learning organisation concept using structural equation modelling. Finally, it explains and examines the largely misunderstood concept of management fashions.

Conception et synthèse d'inhibiteurs de la NO Synthase inductible à visée thérapeutique / Conception and synthesis of NO Synthase inducible inhibitors with therapeutic aim

Depuis sa découverte, le monoxyde d'azote n'a pas cessé d'intéresser la communauté scientifique. Sa biosynthèse est catalysée par les NO Synthases, qui est une famille de trois isoenzymes. La NOS neuronale et la NOS endothéliale sont constitutives et produisent du NO impliqué dans la neurotransmission et la vasodilatation respectivement. La NOS inductible quant à elle produit du NO impliqué dans la réponse immunitaire innée. Une surproduction de monoxyde d'azote est impliqué dans de nombreuses pathologies comme les maladies neuro-dégénératives ou les maladies chroniques à composantes inflammatoires. L'inhibition de iNOS et nNOS présente donc un grand intérêt thérapeutique. Cependant l'enjeu est de développer des inhibiteurs hautement sélectifs, afin de préserver les fonctions vitales de eNOS.Nous avons choisi de synthétiser des composés constitués d'un analogue de substrat tel que thiocitrulline et S-alkyl-isothiocitrulline auquel est ajouté, par un lien peptidique ou hétérocyclique, un résidu susceptible de se lier dans le canal d'accès du substrat, région moins conservée du site actif pouvant fournir des interactions spécifiques. La synthèse de ces composés a été réalisée sur support solide selon une méthode d'ancrage par la chaîne latérale développée au laboratoire, ou pour certains composés, selon une méthode classique d'ancrage par l'amine alpha, ces deux méthodes offrant un fort potentiel en chimie combinatoire. Les différents composés synthétisés ont été testés sur les trois isoformes recombinantes.Un second travail est brièvement exposé dans ce manuscrit. La synthèse de cyclopeptides basés sur le motif RGD impliqué dans la liaison aux intégrines a été réalisée selon un mode de cyclisation développé au laboratoire faisant intervenir la formation d'un pont guanidine qui sera ensuite diversement substitué. L'étude RMN et l'évaluation de leur activité biologique sur les récepteur opioïdes semble montrer un influence de la substitution du pont sur leur conformation et leur activité. / Since its discovery, Since its discovery, the nitric oxide did not stop interesting the scientific community. Its biosynthesis is catalyzed by NO synthases, a family of three isoenzymes. Neuronal NOS and endothelial NOS are constitutive and produce NO involved in neurotransmission and vasodilation process respectively. Inducible NOS produces NO involved in the innate immune response. An overproduction of nitric oxide is involved in many diseases such as neurodegenerative diseases or chronic diseases with inflammatory components. Thus, its inhibition should be of high therapeutical interest. However, it is necessary to develop highly selective inhibitors to preserve the vital functions of eNOS.We chose to synthesize compounds constituted of one substrate analogue as thiocitrulline or S-alkyl-isothiocitrulline which linked by a peptide bond or heterocycles to a residue able to bind into the substrate access channel, a less conserved region into the active site were specific interactions could be established. The synthesis of these compounds was performed on solid support according to an anchoring method through the side chain developed at the laboratory or, for some compounds, according to a conventional anchoring method through the alpha amine. These approaches are particulary interesting for a combinatory chemistry approach. All compounds were tested on the three recombinant isoforms.A second work is outlined in this manuscript. It consist of synthesizing cyclopeptides based on RGD motif involved in the binding to integrins and enkephalins analogues. The cyclisation method was developed in the lab and involves the formation of a guanidine bridge diversely substituted. NMR studies and biological evaluations on opioid receptor suggests that the diverse bridge substitutions could modulate the conformation and activity of the peptides.
