Prandato, Emeline 08 October 2013 (has links) (PDF)

DEVELOPMENT OF NOVEL MULTI-RESPONSIVE MATERIALS CHARACTERIZED BY POTENTIAL CONTROLLED RELEASE PROPERTIES

Chikh Alard, Ibaa 05 December 2018 (has links) (PDF)
With the emergence of novel and more effective drug therapies, increased importance is being placed upon the methods by which these drugs are being delivered to the body. In conventional drug delivery systems, there is very little control over the release of drug. The effective concentration at the target site can be achieved by intermittent administration of grossly excessive doses, which, often results in constantly, unpredictable variations in plasma concentrations, with the risk of reaching levels below or above the therapeutic range leading to marked side effects. A plethora of formulation strategies mainly based on polymeric/lipid nanoparticles, are described in literature. Even though these systems are therapeutically advantageous in comparison to conventional systems, they remain insensitive to the changing metabolic states of the body although the symptoms of most metabolic diseases follow a rhythmic pattern.A more appropriate and effective approach of managing some of these conditions lies in the chronotherapy. This approach allows for pulsed or self-regulated drug delivery which is adjusted to the staging of biological rhythms, since the onset of certain diseases exhibits strong circadian temporal dependence. In order to reach the objective of mimicking the biophysical and biochemical processes of pathological states, many innovations in material design for drug delivery systems (DDS) that are able to release the therapeutic payload-on-demand were done to release the therapeutic agent only when it is required, according to the physiological need. The development of multidisciplinary research teams has brought huge advantages in the design, fabrication and utilization of such smart systems, especially in the pharmaceutical field. Interestingly, numerous smart polymeric materials exhibit a response to a specific stimulus. A step further, the elaboration of purpose-built monomers can give rise to compounds with tunable sensitivities or multi-stimuli responsiveness. These smart polymers demonstrate an active responsiveness to environmental (or external) signals and change their physicochemical properties as designed (e.g. conformation, solubility, shape, charge or size). As far as the stimuli are concerned, they consist of physical (e.g. temperature, ultrasound, light, electricity, magnetic or mechanical stress), chemical (e.g. pH, ionic strength) and biological signals (e.g. enzymes, biomolecules). Due to the intrapersonal variabilities which may make internal stimuli hazardous, externally controlled systems rely on externally applied stimuli that are produced by stimuli-generating devices, which results in pulsed drug delivery. This type of delivery may be rapid and allows a transient release of a determined amount of drug within a short period of time immediately after a pre-determined off-release period. A novel strategy for the formation of multi-stimuli responsive materials endowed with pH, magnetic and light sensitivity was achieved. The approach relied on the incorporation of magnetic tetrahalogenoferrate(III) anions along a polymeric backbone based on poly(2-(N,N-dimethylamino) ethyl meth-acrylate) (PDMAEMA). Starting from the same PDMAEMA, quaternized pending amine groups with various halide derivatives gave rise to magnetic materials after anion metathesis. Measuring the magnetic susceptibility of these materials exhibited that the magnetic susceptibility increased as the substituted group size decreased (become smaller) which was apparently related to the steric hindrance around the ionic pendants. Additionally, a good correlation between the magnetic susceptibility and ferric content was found. Additional experimental and theoretical Raman analyses allowed the determination of the nature of the magnetic species constituting the materials. This strategy further offers the opportunity to tailor the magnetic response through partial ammonium salt formation. In order to merge the magnetic properties of ferric-based materials with another stimuli-responsive functionality, random copolymers containing DMAEMA (D) with diazobenzene (A) unit were prepared. So, three copolymers PDA were synthesized (with targeted D/A ratios 4/6 (PDA4), 6/4 (PDA6) and 8/2 (PDA8)). Meanwhile, different degrees of amine quaternization (10, 50 and 100 %) were applied, which led to the following polymeric salts PDAX/Y where X = 4, 6, 8 (referring to the percentage of the DMAEMA unit) and Y = 10, 50 and 100 (referring to the percentage of quaternized amine groups). Finally, the aforementioned materials were converted into magnetic polymers by anion exchange. As a result, magnetic responses correlated well with amount of iron oxide in these compounds and the amount of ionic pending groups along the backbone. Moreover, the remaining tertiary amines conferred pH sensitivity to the polymers whereas the diazobenzene units ensured light responsiveness through the well-established trans-to-cis isomerization. In order to functionalize these materials in the pharmaceutical field, an intelligent delivery system was prepared. Firstly, an attempt to formulate riboflavin-5’-phosphate sodium (RPS) loaded on PDA8 microspheres was made using double emulsion evaporation method. Meanwhile, prednisolone (PRD) microspheres were prepared using s/o/w emulsion technique. Subsequently, coating systems of cochineal red tablets were developed. These tablets were coated with polymer solution (using each of three types of copolymers: PDA8, PDA6, and PDA4) until the desired percentage of the coating was achieved (10, 15, and 20 % w/w). The cumulative release profiles of cochineal red tablets coated with PDA8, PDA6, and PDA4 showed a pH-sensitive release behavior. The release in the neutral media (pH ≈ 7.0) was very slow (less than 3 % after one hour). Then, after changing the pH to 1.2, an increase in the release of cochineal was observed. Furthermore, the cumulative release of cochineal red was at the highest value for the PDA8 and the lowest for PDA4 depending on the percentage of PDMAEMA moieties. Moreover, by increasing the percentage of the coating from (10, 15 to 20 % w/w), the cumulative release of cochineal decreased. Therefore, the copolymer PDAX can be used for controlling the release of drug by changing the pH value. Finally, the cochineal tablets coated with PDA6 (10 %) showed features of light sensitivity. The release of cochineal red from coated tablets was only due to the switching in the conformational trans/cis isomerization of azobenzene moieties upon irradiation, which was confirmed by comparing the release of coated tablets with uncoated tablets upon irradiation.

The method is based on the Griess-Llosvay reaction and measuring of the absorbance at 540 nm using a green LED integrated into the μFBA built in the urethane-acrylate resin. Initially, the natural nitrite ion content of natural dairy sample product is determined in the micro-mixing chamber, while the nitrate is reduced to nitrite in the micro-column coupled to the micro cadmium sponge and then determined as in the micro-chamber nitrite. content of nitrate ions was estimated from the difference between the first and second determination. The analytical curve for nitrate and nitrite was linear in the work range of 10.0 100.0 μg L 1 with a correlation coefficient of 0.992 and 0.998, respectively. The limit of detection and relative standard deviation were estimated at 0.39 μg L 1 and < 1.7 % (n = 5) for nitrite and, 0.41 μg L 1 and < 1.3 % (n = 5) for nitrate. Comparing with the reference methods, no statistically significant differences were observed when applying the paired t-test at a 95% confidence level. The accuracy was assessed through recovery test (97.7 to 102.9 %). The proposed microsystem employed in-line cadmium sponge microcolumn presented satisfactory portability, robustness, flexibility, low-cost device and reduced chemicals consumption compared to recent methods.

Jahny, Karsten 30 August 2001 (has links)
This paper describes the synthesis of a reactive amphiphilic polyurethane, and its use as an emulsifier for the emulsion polymerization of styrene. The colloid properties and the structure of the emulsifier in the aqueous phase have been investigated. When the acrylic polymerization is carried out with hydrophobic monomers, we obtain a composite particle structure. The variation of particle size and particle size distribution as a function of the reaction parameters was measured by light scattering methods and Flow-fieldflow-fractionation (F-FFF) . Through investigation by dilatometry and reaction calorimetry it was possible to compare the polymerization process with that of common emulsion polymerization. Solid State NMR analysis allowed us to develop a core-shell model for the composite particle structure, and to determine the presence of an interphase layer between core and shell. Atomic Force Microscopy (AFM) methods were used to characterize the surface of the films, and tapping mode AFM was used to characterize the polymer phase separation on the micro scale.

Aluminium oxide - poly(ethylene-co-butylacrylate) nanocomposites : synthesis, structure, transport properties and long-term performance

Nordell, Patricia January 2011 (has links)
Polymer nanocomposites are promising materials for dielectrical use in high voltage applications and insulations. This work presents a study of nanocomposites based on poly(ethylene-co-butyl acrylate) with two different comonomer compositions and two different aluminium oxide nanoparticles. The nanoparticles were either untreated, or surface-treated with two different silanes, aminopropyl triethoxy silane and octyltriethoxy silane. The best level of dispersion was found for the polymer with 13 wt. % of butyl acrylate (EBA-13) whereas the low melt viscosity of the polymer with 28 wt. % of butyl acrylate (EBA-28) resulted in insufficient mixing with uneven dispersion as a result. Octyltriethoxy silane-treated particles were best dispersed in the polymer. The nanoparticles acted as nucleation agents in EBA-28, increasing the crystallization temperature by several degrees. Studies of the water uptake in the nanocomposite materials showed the effect of the enormous interfacial surfaces and great number of polar groups present on the nanoparticle surfaces. For the well-dispersed nanomaterials, the water sorption data could be modeled by a single Fickian equation, whereas materials that contained a sizeable fraction of large nanoparticle agglomerates showed a two stage sorption process, first a fast process associated with the saturation of the polymer phase and second, a slow diffusion process due to water sorption of large particle agglomerates. The long-term performance and interaction between the nanoparticles and the phenolic antioxidant (Irganox 1010) was investigated by differential scanning calorimetry in order to assess the oxidation induction time (OIT); the latter being proportional to the concentration of efficient antioxidant. It was found that the stabilizer was adsorbed to the untreated Al2O3 nanoparticles, resulting in a significant reduction in OIT. However, silanization of the nanoparticles resulted in an increase in OIT, compared to the materials containing untreated particles. Furthermore, it was shown that the stabilizer was not irreversibly adsorbed to the particles, allowing a gradual release of stabilizer with ageing time. / QC 20110128

Dielectric properties of poly(ethyelene-co-butyl acrylate) filled with Alumina nanoparticles

Jäverberg, Nadejda January 2011 (has links)
In this work dielectric properties of the poly(ethylene-co-butyl acrylate)filled with alumina nanoparticles are evaluated. These nanocomposite materialswere manufactured at the department of Fibre and Polymer Technology,KTH.This study is limited to the properties of general importance for the AC applications. The dielectric permittivity of the nanocomposite materials wasstudied as a function of filler size, filler content, coating, temperature and airhumidity used for conditioning of the samples. The ultimate goal with thisproject is to describe the influence of material composition, temperature andair humidity on the dielectric properties and model these dependencies. In this thesis the experimental setup for voltage endurance testing of thenanocomposites, namely studying applied voltage frequency dependence ofpartial discharges in electrical trees, with a possibility of following electricaltreeing optically, was developed and described. The dielectric spectroscopy measurements were performed on thoroughly driednanocomposites - so-called dry DS study. It was shown that the experimentaldata can be fitted with Havriliak-Negami approximation, which justifiesthe correctness of the measurement results. It has been shown that addingnanoparticles to the EBA matrix changes the low frequency dispersion significantlyfor the dried samples. It was also indicated that the particle coatingused has very low impact on the resulting permittivity of the thoroughly driedsamples. From the dry DS studies it was suggested that the main cause ofthe scattering in data between the dry samples is most likely the influenceof the material inhomogeneity and possibly the moisture absorption. Thisleads to a possibility of using dielectric spectroscopy as a tool for probing thedispersion of nanoparticles in the polymer matrix. The dielectric spectroscopy measurements were also carried out on the nanocompositesconditioned in the environments with different humidity levels of air inorder to study the influence of absorbed water on the dielectric permittivity- so-called wet DS study. From the wet study it was shown that for the wetsamples the amplitude of the loss peak is defined by the filler size, filler contentand coating used; while its position in frequency domain is determinedby the coating and the humidity level used for conditioning. / QC 20110315

P(EMA-co-HEA)/SiO2 hybrid nanocomposites for guided dentin tissue regeneration: structure, characterization and bioactivity

Vallés Lluch, Ana 15 December 2008 (has links)
Vallés Lluch, A. (2008). Synthèse contrôlée et auto-organisation de glycopolymères amphiphiles à greffons polymères mésogènes, destinés à la vectorisation de principes actifs / Controlled synthesis and self-assembly of amphiphilic glycopolymers with polymeric mesogen grafts, in view of drug delivery applications

Ferji, Khalid 08 October 2013 (has links)
Ces nano-objets pourront être testés ultérieurement pour la formulation d'un nouveau type de vecteurs de principes actifs / New graft glycopolymers with well-defined parameters [dextran-g-poly(diethylene glycol cholesteryl ether acrylate) (Dex-g-PADEGChol)] have been prepared in four steps using the "grafting from" strategy. Challenge of this work arises from the combination for the first time of a hydrophilic, biocompatible/biodegradable polysaccharide backbone with mesogen hydrophobic polymeric grafts. Controlled growth of the grafts (PADEGChol) was obtained using ATRP initiated in homogeneous medium from a dextran derivative (DexAcBr). In order to find the best polymerization conditions, homopolymerization of ADEGChol monomer was investigated using an initiator model and various catalytic systems CuIBr/(PMDETA or OPMI) in two solvents (Toluene and THF). The amphiphilic properties of such glycopolymers were evaluated and their mesomorphic properties have been studied by thermal polarizing optical microscopy, differential scanning calorimetry and X-ray scattering. Using transmission electron microscopy and dynamic light scattering, vesicular morphology called "polymersome" was observed in aqueous medium when DMSO was used as co-solvent. These polymersomes could be tested as new drug delivery systems

Estudo da dinâmica molecular em copolímeros em bloco compostos de poli(metacrilato de metila), poli(ácido acrílico) e poli(acrilato de chumbo) por técnicas de ressonância magnética nuclear e análise térmica / Study of Molecular Dynamics in Copolymers of Poly (methyl methacrylate), poly (acrylic acid) and Poly (acrylate lead) by nuclear magnetic resonance and thermal analyses

Silva, André Luis Bonfim Bathista e 07 July 2009 (has links)
This thesis involved the study of molecular dynamics in triblock copolymers consisting of poly (methyl methacrylate) (PMMA), poly (acrylic acid) (PAA) and Poly (lead acrylate) (PAPb) by solid-state Nuclear Magnetic Resonance (NMR) and thermal analyses (DSC and DMTA). These block copolymers were synthesized in order to obtain materials not only presenting higher refractive index but also offering good radiological protection, which are useful for individual and combined applications. For the study of these materials, the samples were prepared with various compositions, including those in pure form containing only a block, resulting from the combination of two blocks, and the triblock with different relative amounts of PAPb, ranging from 1 to 40%. In the case of PMMA, the molecular dynamics is well known, characterized by a β relaxation, which involves more specifically the ester side-group dynamics occurring within a broad temperature range centered around the room temperature, and the glass transition, predominantly involving the main chain dynamics that occur for temperatures around 100oC. Due to large extension of these two dynamical events in wide temperature ranges, they merge at temperatures below 100oC. The fact that the β relaxation occurs for the PMMA around the room temperature, confers to it one of its most important features, toughness. However, due to the presence of the hydroxyl group in PAA, which allows the occurrence of hydrogen links between different branches, the β relaxation in this material is strongly suppressed, making it brittle and not suitable for the desired applications. In the case of the triblock copolymer, it is also expected that PAPb may also interfere with the mechanical properties of the final material due to its involvement in the full molecular dynamics of the copolymer. For this study, several samples were prepared with different relative amounts of PMMA, PAA and PAPb, in order to understand the molecular dynamics of these materials, individually and collectively, and select the most suitable ones for the desired technological applications. To characterize and study these samples, we employed thermal analyses (DSC and DMTA), which give quick and important data on the macroscopic dynamics, and basic and advanced solid-state NMR methods, which provide more detailed information about he molecular dynamics. These studies indicated that PAA, necessary along the synthesis route used for the insertion of PAPb in the triblock copolymers, is an undesirable element in the final product, because it is always acting on the suppression of β relaxation. Although the proposition of these new materials for the ophthalmic area is not very appropriate, since the amount of PAPb necessary to obtain good refraction indexes would be necessarily large (over 5%, when they become brittle), the samples with small amounts of PAPb (up to 5%, when they present good mechanical properties) allowed a significant reduction of about 50% of the incident x-rays. Finally, using solid-state NMR methods, including modern Exchange experiments and thermal analyses (DSC and DMTA), it was possible to understand in detail the individual and cooperative motions of the main chain and side groups. The most import result obtained indicate that, despite being important inserting PAPB and PAA in the block copolymer for improving the desired optical and x-ray shielding properties, the presence of these blocks tend to suppress the β relaxation, reducing thesample mechanical properties.
