Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / Alpinia zerumbet, conhecida popularmente como colÃnia no Nordeste do Brasil, Ã uma planta medicinal usada amplamente na medicina popular na forma de chÃs e infusÃes para o tratamento de doenÃas cardiovasculares, como a hipertensÃo arterial. O intenso consumo popular dessas infusÃes levou-nos a avaliar o perfil toxicolÃgico e genotoxicolÃgico do extrato aquoso e do Ãleo essencial das folhas de A. zerumbet. Esse estudo foi avaliado pelos ensaios de curta duraÃÃo in vivo e in vitro. Inicialmente foi avaliada a citotoxicidade e o efeito hemolÃtico in vitro, porÃm nÃo houve resposta tÃxica. A DL50 encontrada para o extrato aquoso foi >5 g/Kg, demonstrando que os princÃpios ativos do extrato apresentam baixa toxicidade. Os estudo de genotoxicidade foram realizados in vivo. Os animais foram tratados, por via oral, com trÃs doses do extrato aquoso (2 g/Kg, 3,5 g/Kg e 5 g/Kg) e com a dose de 400 mg/Kg do Ãleo essencial. ApÃs 24h e 48h, o sangue perifÃrico e a medula Ãssea foram coletados. No ensaio do cometa nÃo houve detecÃÃo de nenhum cometa de grau elevado e as anÃlises estatÃsticas demonstraram um P<0,01 (grau de significÃncia: P<0,05) para todas as amostras em relaÃÃo ao controle positivo e um P>0,05 (grau de significÃncia: P<0,05) para as amostras em relaÃÃo ao controle negativo. No ensaio do micronÃcleo, todas as doses do extrato aquoso e do Ãleo essencial tiveram diferenÃa estatisticamente significante em relaÃÃo a ciclofosfamida, um antineoplÃsico citotÃxico, com um P<0,001 e um P>0,05 em relaÃÃo ao controle negativo (significÃncia: P<0,05). Todos esses resultados indicam que o extrato aquoso e o Ãleo essencial das folhas de A. zerumbet nÃo apresentam aÃÃes citotÃxicas nem genotÃxicas nos modelos testados. / Alpinia zerumbet, known ordinarily as colony in Northwestern Brazil, is a medicinal plant widely used in popular medicine as tea and infusions for the treatment of intestinal and cardiovascular illnesses, such as hypertension. Due to the high levels of consumption of such infusions, we have sought to evaluate the toxicological and genotoxicological profile of the tea and essential oil made from A. zerumbet leaves. This study has been evaluated by short term in vivo and in vitro trials. We initially evaluated the cytotoxicity and the hemolytic effect in vitro; however, there was no toxic response. The DL50 found for the tea was of >5 mg/Kg, which demonstrates that the active principles of the tea present low toxicity. The genotoxicity trials were carried out in vivo. The animal received treatment orally, with three doses of the tea (2 g/Kg, 3,5 g/Kg and 5 g/Kg) and with a dosage of 400 mg/Kg of the essential oil. Peripheral blood and bone marrow were collected after 24 and 48 hours. In the comet trial no high level comets were detected and the statistical analyses demonstrate a P<0,01 (significance: P<0,05) for all samples when compared to the positive control and a P<0,05 (significance: P<0,05) for samples in relation to the negative control. In the micronucleus test, all doses of the tea and essential oil presented a statistically significant difference in relation to cyclofosfamide, with a P < 0.001 and a P > 0.05 compared with the negative control (significance: P<0,05). All those results indicate that the tea and essential oil made from A. zerumbet leaves do not present cytotoxic or genotoxic action in the tested models.

Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / Alpinia zerumbet, conhecida popularmente como ColÃnia, no Nordeste do Brasil, à uma planta medicinal usada amplamente na medicina popular na forma de chÃ, sendo tradicionalmente utilizada para o tratamento da hipertensÃo arterial. O objetivo desse estudo foi avaliar a seguranÃa e o potencial genotÃxico do chà de A. zerumbet em voluntÃrios sadios. O ensaio clÃnico consistiu de um estudo duplocego, controlado por placebo, randomizado e paralelo, com 36 voluntÃrios (18 homens e 18 mulheres), adultos. Os quais foram aleatoriamente distribuÃdos em dois grupos: ColÃnia, constituÃdo por 24 voluntÃrios, e Placebo, composto por 12 sujeitos. Os voluntÃrios foram tratados durante 28 dias ininterruptos com 540 mL de chà de ColÃnia ou Placebo. AvaliaÃÃes clÃnica e laboratorial foram realizadas no prÃ-estudo, durante o perÃodo de tratamento, bem como apÃs o encerramento do estudo. A genotoxicidade do chà de colÃnia, por sua vez, foi investigada mediante o emprego do teste do cometa. O Ãndice de massa corpÃrea (IMC) foi de 25,300Â2,918 Kg/ cm2 para o grupo ColÃnia no prÃ-estudo e 25,289Â2,965Kg/ cm2 no pÃs-estudo. No grupo Placebo o IMC foi de 25,179Â2,564 no prÃ-estudo e de 24,961Â2,409 no pÃs-estudo. As funÃÃes hematolÃgica, hepÃtica, renal e metabÃlica foram analisadas, antes, durante (14 e 28 dia) e apÃs o estudo atravÃs dos exames laboratoriais, os quais nÃo evidenciaram sinais de toxicidade. CefalÃia, sonolÃncia, dor abdominal, vÃmito, flatulÃncia e poliÃria foram os eventos adversos atribuÃdos a ingestÃo do chà nos dois grupos. Pelo teste do cometa, nÃo foram observados danos (p<0,05) nos linfÃcitos perifÃricos dos voluntÃrios tratados com o chà de colÃnia. Os estudos de toxicologia clÃnica e genotoxicidade nÃo evidenciaram nenhuma toxicidade nos voluntÃrios tratados por 28 dias ininterruptos com o chà de ColÃnia. / Alpinia zerumbet, popularly known as Colonia in the Northeast of Brazil, is a medicinal plant widely used in the popular medicine as a tea, being traditionally used to treat arterial hypertension. The objective of this study was to evaluate the safety and genotoxic potential of the A. zerumbet tea in healthy volunteers. Clinical trial consisted of a double-blind study, placebo controlled, randomized and parallel, with 36 adult volunteers (18 men and 18 women), which were randomly distributed in two groups: Colonia, consisted of 24 volunteers, and Placebo, consisted of 12 subjects. The volunteers were treated for 28 consecutive days, with 540 mL of Colonia or Placebo tea. Clinical and laboratorial evaluations were performed during the pre-study and treatment, as well as during the closure of the study. The genotoxicity of the Colonia tea was investigated by the comet assay. The body mass index (BMI) was 25.300Â2.918 Kg/ cm2 for the Colonia group in the pre-study and 25.289Â2.965Kg/ cm2 in the end of the study. For the Placebo group, BMI was 25.179Â2.564 in the pre-study and 24.961Â2.409 after the study. The blood, liver, kidney and metabolic functions were analyzed before, during (14th and 28th day) and after the study by the evaluation of laboratorial exams, which did not evidence signs of toxicity. Headache, sleepiness, abdominal pain, vomit, flatulence and polyuria were the adverse events attributed to the ingestion of the tea in both groups. The comet assay did not show damage in peripheral lymphocytes of the volunteers treated with the Colonia tea (p<0.05). The clinical toxicology and genotoxicity studies did not evidence any toxicity in the volunteers treated for 28 consecutive days with the Colonia tea.

This plant is popularly known as colony and is widely used in folk medicine for its diuretic and antihypertensive properties. The aim of this study was to investigate whether the aqueous extract of leaves of A. zerumbet (EAAz) had vasodilator activity in rat aorta pre-contracted with phenylephrine and to elucidate its mechanism of action. Male Wistar rats (250-300 g) from the vivarium of UFC, were killed by cervical dislocation and the thoracic aorta removed and dissected. The aortic rings (4-5 mm) were mounted in organ cameras, and containing Krebs solution aerated with carbogen at 37 ° C to measure changes in isometric tension. The endothelium of the aortic rings was removed mechanically to study their involvement in the vasodilator effect of EAAz. To study the involvement of nitric oxide (NO), cGMP, prostanoid, potassium channels activated by Ca2+, reactive oxygen species and activation of enzymes kinases Src and PI3 kinase, the preparations were treated respectively with L-NAME (100 µM ), ODQ (10 µM), indomethacin (10 µM), carybdotoxin (100 nM) plus apamin (100 nM), MnTMPyP (100 µM), catalase (500 U / ml), PEG-catalase (500 U/ ml), SOD (500 U / ml) and PP2 (20 µM), wortmannin (300 nM). The vasodilatory effect of EAAz (0.05, 0.15, 0.5, 1.5, 15 and 50 µg / mL), acetylcholine (10-8 - 10-5 M) and sodium nitroprusside (10-8 M ) were evaluated in the aorta of rats pre-contracted with phenylephrine (10-8 - 3x10-8 M) before and after treatment with the specific inhibitors. EAAz induced vasodilation was dependent and endothelium mediated by NO via cGMP as the vascular relaxation was abolished in the presence of the methyl ester of NG-nitro-L-arginine and ODQ. Vasodilation was also reduced by MnTMPyP (mimetic cellular permeant SOD), catalase polyethylene glycol, PP2 (Src kinase inhibitor) and wortmannin (inhibitor of phosphoinositide 3-kinase). The route of production of ROS induced EAAz was significantly prevented by the action of MnTMPyP and PEG-catalase. There was also the inhibition of the kinase ROS/PI3 / Src kinase that are involved in the activation of the eNOS. The EAAz caused an endothelium-dependent relaxation via NO / cGMP with the possible involvement of ROS and Src kinase and phosphoinositide 3-kinase-dependent Akt. Taking us to the conclusion that the vascular relaxation induced by EAAz could explain the traditional use of the tea leaves of A. zerumbet for the treatment of hypertension. / A Alpinia zerumbet é uma planta comumente encontrada na região do nordeste do Brasil. Essa planta é conhecida popularmente por colônia e é muito utilizada na medicina popular por suas propriedades diuréticas e anti-hipertensivas. O objetivo do presente estudo foi investigar se o extrato aquoso das folhas de A. zerumbet (EAAz) apresentava atividade vasodilatadora em aorta de rato pré-contraída com fenilefrina e, elucidar o seu mecanismo de ação. Ratos machos Wistar (250 a 300 g), provenientes do biotério da UFC, foram sacrificados por deslocamento cervical e a aorta torácica removida e dissecada. Os anéis da aorta (4 a 5 mm) foram montados em câmeras orgânicas, contendo solução de Krebs e aerados com carbogênio a 37°C, para as medidas de variações na tensão isométrica. O endotélio dos anéis da aorta foi removido mecanicamente para estudar o seu envolvimento no efeito vasodilatador do EAAz. Para estudar o envolvimento do óxido nítrico (NO), GMPc, prostanóides, canais de potássio ativados por Ca2+, espécies reativas do oxigênio e a ativação das enzimas quinases Src e PI3 quinase, as preparações foram tratadas, respectivamente, com L-NAME (100 µM), ODQ (10 µM), indometacina (10 µM), caribdotoxina (100 nM) mais apamina (100 nM), MnTMPyP (100 µM), catalase (500 U/ml), PEG-catalase (500 U/ml), SOD (500 U/ml) e PP2 (20 µM), Wortmannin (300 nM). O efeito vasodilatador do EAAz (0,05; 0,15; 0,5; 1,5; 15 e 50 µg/mL), acetilcolina (10-8 - 10-5 M) e nitroprussiato de sódio (10-8 M) foram avaliados em aorta de ratos pré-contraída com fenilefrina (10-8 - 3x10-8 M), antes e após tratamento com os inibidores específicos. A vasodilatação induzida pelo EAAz mostrou-se dependente do endotélio e mediada pelo NO via GMPc já que o relaxamento vascular foi abolido na presença do éster metílico de NG-nitro-l-arginina e do ODQ. A vasodilatação também foi reduzida pelo MnTMPyP (permeante celular mimético da SOD), polietilenoglicol catalase, PP2 (inibidor da quinase Src) e Wortmannin (inibidor da fosfoinositideo 3-quinases). A via de produção de EROs induzida pelo EAAz foi inibida significativamente pela ação do MnTMPyP e PEG-catalase. Ocorreu também a inibição da via EROs/PI3 quinase/Src quinase que estão envolvidas na ativação da eNOS. O EAAz causou um relaxamento dependente do endotélio via NO/GMPc com o possível envolvimento de EROs e das Src quinase e fosfoinositideo 3-quinase dependente de Akt. Dessa forma, pode-se concluir que o relaxamento vascular induzido pelo EAAz poderia explicar o tradicional uso do chá das folhas de A. zerumbet para o tratamento da hipertensão arterial.

A widely used animal model involves repeated administration of ketamine (TEC) that induces the disease symptoms as well as causes oxidative damage. The Alpinia zerumbet is a plant whose essential oil (OEAZ) has shown important antipsychotic effect in previous studies of our research group. Based on this statement the present study aimed to determine the effects of OEAZ in the prevention and reversal of positive symptoms (pre-pulse inhibition test -PPI), negative (social interaction -SI) and cognitive (maze Y) induced by repeated administration of CET and to assess neurochemical changes as part of its mechanism of action: Reduced Glutathione - (GSH); Nitrite (indirect measurement of nitric oxide); Levels of brain-derived neurotrophic factor (BDNF); Interleukin-6 (IL-6) and to study the possible adversities related to the oil effect, extrapyramidal events (catalepsy), changes in body weight (BW). For the prevention protocol, male mice were pretreated with OEAZ 100 and 200 mg / kg (OEAZ 100 and OEAZ 200) or Olanzapine 2 mg / kg (OLANZA) or saline (SAL) for 7 days, from 8 ° to 14 ° CET was added. The reversal protocol the animals were pretreated with CET the 1st to the 7th day and 8 ° to 14 ° OEAZ 100 and 200 or OLANZA or SAL were added. CET caused deficit in the IPP, social interaction and the Labyrinth Y, reduced GSH levels, nitrite and BDNF, and increased IL-6. The CET has reduced immobility in catalepsy and increased PC of animals. The OEAZ 100 was able to prevent the PPI deficits caused, SI and Maze Y. OEAZ 200 increased the time SI. In reversing the OEAZ 100 and 200 increased the index in PPI and Y maze. OLANZA improved the index in the PPI, SI. 100 OEAZ prevented by increasing GSH levels in the CPF (prefrontal cortex) and CE (striatum) and did not alter the HC (hippocampus) in reversal, increased the CPF, HC, EC. The OEAZ 200 was not significant in preventing, but reverted low GSH levels in all three areas. In nitrite levels OEAZ 100 and 200 is not prevented, but reversed increasing the HC and CE Regarding BDNF only OEAZ 100 and OLANZA were able to increase the levels in the prevention and reversal, the Olaza only reversed. Both doses of OEAZ and OLANZA significantly reduced IL-6 levels. In the evaluation of possible side effects OEAZ 100 and 200 slightly increased immobility time of animals. On Pc OLANZA increased sharply, while the OEAZ 100 and 200 increased and reduced respectively in preventing and acted contrary to the reversal. Overall the OEAZ presented a similar performance to antipsychotic, preventing and reversing the positive, negative and cognitive symptoms. Its mechanism involves dopaminergic pathways, neurotrophic and antioxidants. / A esquizofrenia é um transtorno psiquiátrico crônico, caracterizada pela presença de sintomas positivos, negativos e cognitivos. O modelo animal utilizando Cetamina (CET) é capaz de induzir a sintomatologia da doença, assim como provoca danos oxidativos com administrações repetidas. Os antipsicóticos tratam a sintomatologia, mas provocam efeitos colaterais como excessivo ganho de peso, comprometendo a adesão dos pacientes ao tratamento. Dada a necessidade de novas terapias a Alpinia zerumbet é uma planta cujo óleo essencial (OEAZ) vem apresentando importante efeito antipsicótico em publicações prévias de nosso grupo de pesquisa. Baseado nesta afirmativa o presente trabalho objetivou determinar os efeitos do OEAZ na prevenção e reversão dos sintomas positivos (teste de inibição pré-pulso - IPP), negativos (interação social - IS) e cognitivos (labirinto em Y) induzidos pela administração repetida de CET (25mg/kg), bem como avaliar alterações neuroquímicas como parte de seu mecanismo de ação: Glutationa reduzida – (GSH); Nitrito (medida indireta de óxido nítrico); Níveis do fator neurotrófico derivado do cérebro (BDNF); Interleucina- 6 (IL-6) e estudar os possíveis adversidades relacionadas ao efeito do óleo: eventos extrapiramidais (Catalepsia), alterações no peso corporal (PC). No protocolo de prevenção para simular um episódio agudo de esquizofrenia, camundongos machos foram previamente tratados com OEAZ 100 e 200 mg/kg (OEAZ 100 e OEAZ 200) ou Olanzapina 2 mg/kg (OLANZA) ou Salina (SAL) durante 7 dias, do 8° ao 14° dia CET foi acrescentada no tratamento. No protocolo de reversão (manutenção do tratamento) os animais foram previamente tratados com CET do 1° ao 7° dia e do 8° ao 14° dia o OEAZ 100 e 200 ou OLANZA ou SAL foram acrescentados de acordo com o grupo experimental. CET causou déficit na IPP, Interação social e no Labirinto Y, reduziu os níveis de GSH, Nitrito e BDNF, e aumentou o de IL-6. A CET reduziu a imobilidade na Catalepsia e aumentou o PC dos animais. O OEAZ 100 foi capaz de prevenir os déficits causados na IPP, IS e labirinto Y. O OEAZ 200 aumentou o tempo de IS. Na reversão o OEAZ 100 e 200 aumentaram o índice na IPP e labirinto –Y. OLANZA melhorou o índice na IPP, IS. OEAZ 100 preveniu aumentando os níveis de GSH no CPF (córtex pré-frontal) e CE (corpo estriado) e não alterou no HC (Hipocampo), na reversão, aumentou no CPF, HC, CE. O OEAZ 200 não foi significativo na prevenção, mas reverteu os baixos níveis de GSH nas três áreas. Nos níveis de Nitrito OEAZ 100 e 200 não preveniram, mas reverteram aumentando no HC e C.E. Quanto ao BDNF apenas o OEAZ 100 e a OLANZA foram capazes de aumentar os níveis na prevenção e reversão, a OLAZA só reverteu. Ambas as doses de OEAZ e OLANZA reduziram significativamente os níveis de IL-6. Na avaliação dos possíveis efeitos colaterais OEAZ 100 e 200 aumentaram discretamente o tempo de imobilidade dos animais. No PC a OLANZA aumentou acentuadamente, enquanto o OEAZ 100 e 200 aumentou e reduziu respectivamente na prevenção e atuaram de forma contrária na reversão. No geral o OEAZ apresentou uma atuação antipsicótica semelhante a OLANZA, prevenindo e revertendo os sintomas positivos, negativos e cognitivos DA esquizofrenia. Seu mecanismo envolve vias dopaminérgicas, neurotróficas e antioxidantes.

It is used for medicinal purposes to treat hypertension and has been studied in relation to its pharmacological properties. This study aimed to analyze the pharmacological effect of hexanic (HFEOAz), chloroformic (CFEOAz), and methanolic (MFEOAz) fractions of the essential oil of Alpinia zerumbet (EOAz) in the vascular reactivity in vitro and cardiovascular parameters in vivo. The research project was approved by the Ethics Committee on Animal Research, of the Federal University of Ceará, under protocol numbers 55/10 and 18/2011, according to the standards of good laboratory practice involving the use of experimental animals. For all experiments male Wistar rats were used. Experiments of vascular reactivity were conducted in organ bath, with preparations of isolated rat aorta with intact endothelium and desnuded, kept in carbogen and Krebs-Henseleit solution with a concentration in mmol/L: NaCl: 118,0; KCl: 4,7; KH2PO4: 1,2; MgSO4.7H2O; 1,2; NaHCO3: 15,0; CaCl2: 2,5 e Glicose: 5,5. Was observed variation in isometric tension and used different specific inhibitors to analyze the mechanism of action of the vasodilator effect. We analyzed blood pressure indirectly by tail plethysmography in rats submitted to chronic hypertension by inhibition of nitric oxide by the administration of L-NAME, obtaining the systolic, diastolic and mean blood pressure, heart rate, beyond variation in body weight. It was found that the EOAz, HFEOAz, CFEOAz MFEOAz induced relaxation of aortic rings pre-contracted with Phenylephrine (1 mol/L), of dose-dependent manner, with the smallest of the EC50 MFEOAz (150.45 mg/mL). The administration by gavage of 100 mg/kg EOAz, HFEOAz, CFEOAz MFEOAz reduced blood pressure in hypertensive rats by the model of chronic inhibition of nitric oxide, an effect that was greater than the negative control with distilled water and less than the positive controls with Captopril and Nifedipine. The MFEOAz (0.1 - 3000 µg/mL) concentration dependently relaxed Phenylephrine (1 mol/L) and KCl (80 mmol/L) contracted rings with intact or denuded endothelium. Studies of calcium mobilization showed that FMOEAz inhibits the influx of Ca2+ from the extracellular environment and interferes with the contraction-induced Ca2+ release from intracellular stocks by Phenylephrine (1 mol/L) or Caffeine (30 mmol/L). The 4-aminopyridine (1 mmol/L) and Iberiotoxina (30 nmol/L) increased the EC50 of MFEOAz but do not interfere in its final vasodilator effect. The pre-incubation with L-NAME (100 mol/L), ODQ (10 mol/L), Indomethacin (10 mol/L), Atropine (1 mol/L), Catalase (500 U/mL), SOD (300 U/mL), Wortmannin (0.5 mol/L), Tetraethylammonium (10 mmol/L), Apamin (1 mol/L); Caribdotoxin (15 nmol/L) and Glibenclamide (10 mol/L) did not interfere with the relaxation induced by MFEOAz. It was concluded that the EOAz, HFEOAz, CFEOAz and MFEOAz have hypotensive effect in vivo and vasodilator effect in vitro, and that the mechanism of action of MFEOAz probably involves the antagonism of calcium channels voltage-dependent, the calcium channel operated by receptor, also interfering in the release of calcium from intracellular stores. / A Alpinia zerumbet é uma planta da família Zingiberaceae, denominada popularmente “colônia” no Nordeste do Brasil. É utilizada com fins medicinais no tratamento de hipertensão e tem sido estudada em relação as suas propriedades farmacológicas. Esta pesquisa teve por objetivo analisar o efeito farmacológico das frações hexânica (FHOEAz), clorofórmica (FCOEAz) e metanólica (FMOEAz) do óleo essencial da Alpinia zerumbet (OEAz) na reatividade vascular in vitro e nos parâmetros cardiovasculares in vivo. O projeto de pesquisa foi aprovado pela Comissão de Ética em Pesquisa Animal, da Universidade Federal do Ceará, sob os protocolos n 55/10 e n 18/2011, de acordo com as normas de boas práticas que envolvem o uso de animais experimentais. Para todos os experimentos foram utilizados ratos Wistar machos. Realizaram-se experimentos de reatividade vascular no banho de órgãos, com preparações de aorta isolada de rato, com endotélio íntegro e desnudo, mantidos em carbogênio e solução de Krebs-Henseleit com concentração em mmol/L: NaCl: 118,0; KCl: 4,7; KH2PO4: 1,2; MgSO4.7H2O; 1,2; NaHCO3: 15,0; CaCl2: 2,5 e Glicose: 5,5. Foi observada a variação da tensão isométrica e utilizados diferentes fármacos inibidores específicos para análise do mecanismo de ação do efeito vasodilatador. Analisou-se a pressão arterial indireta por pletismografia de cauda em ratos submetidos ao modelo de hipertensão por inibição crônica do óxido nítrico, através da administração do L-NAME, obtendo-se a pressão arterial sistólica, diastólica e média, frequência cardíaca, além da variação do peso corporal. Constatou-se que o OEAz, FHOEAz, FCOEAz e FMOEAz induzem relaxamento de anéis aórticos pré-contraídos com Fenilefrina (1 mol/L), de forma dependente da dose, sendo a menor CE50 a da FMOEAz (150,45 g/mL). A administração por gavagem de 100 mg/kg de OEAz, FHOEAz, FCOEAz e FMOEAz reduziu a pressão arterial em ratos hipertensos pelo modelo de inibição crônica do óxido nítrico, um efeito que foi superior ao controle negativo com água destilada e inferior aos controles positivos com Captopril e Nifedipina. A FMOEAz (0,1 - 3000 g/mL) induz relaxamento dependente da dose em anéis aórticos pré-contraídos com Fenilefrina (1 mol/L) ou KCl (80 mmol/L), com endotélio intacto ou desnudo. Os estudos de mobilização de cálcio mostraram que a FMOEAz inibe o influxo de Ca2+ do meio extracelular, bem como interfere na contração induzida pela liberação de Ca2+ dos estoques intracelulares pela Fenilefrina (1 mol/L) ou Cafeína (30 mmol/L). A 4-aminopiridina (1 mmol/L) e a Iberiotoxina (30 nmol/L) aumentam a CE50 da FMOEAz, mas não interferem no seu efeito vasodilatador final. A pré-incubação com L-NAME (100 mol/L), ODQ (10 mol/L); Indometacina (10 mol/L), Atropina (1 mol/L), Catalase (500 U/ml), SOD (300 U/mL); Wortmannina (0,5 mol/L), Tetraetilamônio (10 mmol/L), Apamina (1 mol/L); Caribdotoxina (15 nmol/L) e Glibenclamida (10 mol/L) não interferiram no relaxamento induzido pela FMOEAz. Concluiu-se que o OEAz, FHOEAz, FCOEAz e FMOEAz possuem efeito hipotensor in vivo e vasodilatador in vitro, e que o mecanismo de ação da FMOEAz, provavelmente, envolve o antagonismo aos canais de cálcio dependentes de voltagem, aos canais de cálcio operados por receptor, interferindo também na liberação de cálcio dos estoques intracelulares.

Schizophrenia is a syndrome characterized by the presence of positive, negative and cognitive symptoms. The current therapy does not manage satisfactorily the negative and cognitive symptoms. One of the most relevant models for the study of schizophrenia in a preclinical approach is the administration of ketamine. In this sense the repeated administration of the drug can largely mimic the symptoms of schizophrenia in rodents. The Alpinia zerumbet is a plant whose essential oil (EOAZ) has shown significant antipsychotic effect as evidenced in previous studies of our research group. Based on this statement the present study aimed to determine the effects of EOAZ against positive, negative and cognitive schizophrenia induced by acute and repeated administration of ketamine-like symptoms. For this it was used male Wistar rats treated with EOAZ 100 or 200 mg/kg alone or following the administration of ketamine. In repeated administration protocol ketamine was applied for five days and in the subsequent five days the animals received both ketamine and EOAZ 100 or 200 mg/kg. One group of animals was treated with the atypical antipsychotic risperidone and other received saline (control). After treatment the animals were subjected to behavioral assessments for the determination of positive symptoms (locomotor activity) , negative (social interaction) and cognitive (Y maze and object recognition memory) . The results showed that acute administration of ketamine was not able to cause changes in all models used , especially in relation to recognition memory of objects that was even enhanced by acute drug administration. The EOAZ at both doses was able to reverse those changes. Repeated administration of ketamine was able to mimic more accurately the changes related to schizophrenia , and in this model the administration of EOAZ was superior to risperidone , particularly in working memory. Therefore, the results of this study show that EOAZ may be a new pharmacological approach in treating negative and cognitive symptoms of schizophrenia. / Antipsicóticos representam o alicerce no tratamento da esquizofrenia, um transtorno mental grave, incapacitante, com elevada carga social. A esquizofrenia é uma síndrome caracterizada pela presença de sintomas positivos, negativos e cognitivos. A terapia atual não trata de maneira satisfatória os sintomas negativos e cognitivos. Um modelo relevante para o estudo da esquizofrenia em uma abordagem pré-clínica é o da administração de cetamina. Neste sentido a administração repetida desta droga é capaz de mimetizar grande parte dos sintomas da esquizofrenia em roedores. A Alpinia zerumbet é uma planta cujo óleo essencial (OEAZ) vem apresentando importante efeito antipsicótico como evidenciado em publicações prévias de nosso grupo de pesquisa. Com base nesta afirmativa o presente trabalho objetivou determinar os efeitos do OEAZ contra sintomas positivos, negativos e cognitivos tipo esquizofrenia induzidos pela administração aguda e repetida de cetamina. Para tanto foram utilizados ratos Wistar machos tratados com OEAZ 100 ou 200 mg/kg sozinho ou após a administração de cetamina. No protocolo de administrações repetidas a cetamina foi administrada por cinco dias e por mais cinco dias os animais receberam cetamina e OEAZ 100 ou 200 mg/kg. Um grupo de animais foi tratado com o antipsicótico atípico risperidona e outro recebeu solução salina (controle). Após os tratamentos os animais foram submetidos a avaliações comportamentais para a determinação de sintomas positivos (atividade locomotora), negativos (interação social) e cognitivos (labirinto em Y e memória de reconhecimento de objetos). Os resultados mostraram que a administração aguda de cetamina não foi capaz de causar alterações em todos os modelos utilizados, principalmente em relação à memória de reconhecimento de objetos que foi inclusive melhorada pela administração aguda da droga. O OEAZ em ambas as doses foi capaz de reverter estas alterações. A administração repetida da cetamina foi capaz de mimetizar de forma mais fiel as alterações relacionadas à esquizofrenia, sendo que neste modelo a administração do OEAZ foi superior à da risperidona, principalmente na memória de trabalho. Portanto, os resultados do presente estudo mostram que o OEAZ pode ser uma nova abordagem farmacológica no tratamento dos sintomas negativos e cognitivos da esquizofrenia

The angiotensin I converting enzyme (ACE) can increase blood pressure through the conversion of angiotensin I to the potent vasoconstrictor, angiotensin II. Thus, inhibition of ACE activity is considered a useful therapeutic approach to combat hypertension. Alpinia zerumbet, a herb abundant in northeastern Brazil, is commonly used in folk medicine due to its diuretic and hypotensive properties. Moreover, several studies reported for this species antihypertensive activity. Thus, Alpinia zerumbet can be an important source of ACE inhibitors. Knowing this, we decided to investigate the extracts of the rhizomes of this species in the search for compounds ACE inhibitors. Prospecting chemical hexane extract of the rhizome allowed the isolation of five substances: A stilbene, named (E) -3,5-dimethoxystilbene, unprecedented in the species in this study, a labdane diterpene skeleton called (E) -15.16-bisnorlabda- 8,11-diene-13-one; one chalcone called 2’,4’-dihydroxy-6’-methoxy-3’-(3’’-methyl-6’’- methyl - ethyl -2’’-cyclohexenyl) chalcone; a steroid called β-sitosterol, commonly found in plants, and an aromatic hydrocarbon class kavapirone, called 5,6- dehidrokawain. In the structural determination of individual compounds, spectrometric techniques were employed dimensional analysis [nuclear magnetic resonance (NMR) 1H and 13C and two dimensional techniques (1H-1H COSY, HSQC, HMBC, NOESY). Inhibition angiotensin converting enzyme, antioxidant potential, inhibition of acetilcolinestarase and larvicidal tests of ethanol and hexane extracts of the rhizomes of Alpinia zerumbet were tested. The two extracts showed potential for inhibition of angiotensin converting enzyme and antioxidant potential. Only the hexane extract showed larvicidal activity with LC50 823.1 ppm. None of the extracts studied showed potential inhibition of the enzyme acetylcholinesterase. / A hipertensão, uma das doenças mais comuns em todo o mundo, é uma doença crônica onde a pressão sanguínea elevada pode afetar a saúde. A enzima conversora de angiotensina I (ECA) pode aumentar a pressão arterial através da conversão da angiotensina I no potente vasoconstritor angiotensina II. Sendo assim, a inibição da atividade da ECA é considerada uma abordagem terapêutica útil para combater a hipertensão. Alpinia zerumbet, uma planta aromática abundante no nordeste do Brasil, é comumente utilizada na medicina popular devido suas propiedades diuréticas e hipotensivas. Além disso, vários estudos relatam atividade antihipertensiva para esta espécie. Dessa forma, Alpinia zerumbet pode ser uma importante fonte de inibidores de ECA. Sabendo disso, resolveu-se investigar os extratos dos rizomas desta espécie na busca de compostos inibidores de ECA. A prospecção química do extrato hexânico do rizoma permitiu o isolamento de cinco substâncias: Um estilbeno denominado (E)-3,5-dimetoxiestilbeno, inédito na espécie em estudo; um diterpeno de esqueleto labdano denominado (E)-15,16-bis-norlabda-8,11-dieno-13-ona; uma chalcona denominada 2’,4’-dihidroxi-6’-metoxi-3’-(3’’-metil-6’’-metil-etil-2’’-ciclohexenil)chalcona; um esteróide denominado β-sitosterol, comumente encontrado em plantas; e um hidrocarboneto aromático da classe kavapirona denominado 5,6-dehidrokawain. Na determinação estrutural dos compostos isolados, foram empregadas técnicas espectrométricas de análise unidimensionais [ressonância magnética nuclear (RMN) de 1H e 13C, e técnicas bidimensionais (1H-1H-COSY, HSQC, HMBC, NOESY). Também foram realizados os testes de inibição da enzima conversora de angiotensina, antioxidante, de inibição da enzima acetilcolinestarase e larvicida dos extratos hexânicos e etanólicos dos rizomas de Alpinia zerumbet. Os dois extratos apresentaram potencial de inibição da enzima conversora de angiotensina e antioxidante. Somente o extrato hexânico apresentou atividade larvicida, com CL50 823,1 ppm. Nenhum dos extratos em estudo apresentou potencial de inibição da enzima acetilcolinesterase.

Due to the high levels of consumption of such infusions, we have sought to evaluate the toxicological and genotoxicological profile of the tea and essential oil made from A. zerumbet leaves. This study has been evaluated by short term in vivo and in vitro trials. We initially evaluated the cytotoxicity and the hemolytic effect in vitro; however, there was no toxic response. The DL50 found for the tea was of >5 mg/Kg, which demonstrates that the active principles of the tea present low toxicity. The genotoxicity trials were carried out in vivo. The animal received treatment orally, with three doses of the tea (2 g/Kg, 3,5 g/Kg and 5 g/Kg) and with a dosage of 400 mg/Kg of the essential oil. Peripheral blood and bone marrow were collected after 24 and 48 hours. In the comet trial no high level comets were detected and the statistical analyses demonstrate a P<0,01 (significance: P<0,05) for all samples when compared to the positive control and a P<0,05 (significance: P<0,05) for samples in relation to the negative control. In the micronucleus test, all doses of the tea and essential oil presented a statistically significant difference in relation to cyclofosfamide, with a P < 0.001 and a P > 0.05 compared with the negative control (significance: P<0,05). All those results indicate that the tea and essential oil made from A. zerumbet leaves do not present cytotoxic or genotoxic action in the tested models. / Alpinia zerumbet, conhecida popularmente como colônia no Nordeste do Brasil, é uma planta medicinal usada amplamente na medicina popular na forma de chás e infusões para o tratamento de doenças cardiovasculares, como a hipertensão arterial. O intenso consumo popular dessas infusões levou-nos a avaliar o perfil toxicológico e genotoxicológico do extrato aquoso e do óleo essencial das folhas de A. zerumbet. Esse estudo foi avaliado pelos ensaios de curta duração in vivo e in vitro. Inicialmente foi avaliada a citotoxicidade e o efeito hemolítico in vitro, porém não houve resposta tóxica. A DL50 encontrada para o extrato aquoso foi >5 g/Kg, demonstrando que os princípios ativos do extrato apresentam baixa toxicidade. Os estudo de genotoxicidade foram realizados in vivo. Os animais foram tratados, por via oral, com três doses do extrato aquoso (2 g/Kg, 3,5 g/Kg e 5 g/Kg) e com a dose de 400 mg/Kg do óleo essencial. Após 24h e 48h, o sangue periférico e a medula óssea foram coletados. No ensaio do cometa não houve detecção de nenhum cometa de grau elevado e as análises estatísticas demonstraram um P<0,01 (grau de significância: P<0,05) para todas as amostras em relação ao controle positivo e um P>0,05 (grau de significância: P<0,05) para as amostras em relação ao controle negativo. No ensaio do micronúcleo, todas as doses do extrato aquoso e do óleo essencial tiveram diferença estatisticamente significante em relação a ciclofosfamida, um antineoplásico citotóxico, com um P<0,001 e um P>0,05 em relação ao controle negativo (significância: P<0,05). Todos esses resultados indicam que o extrato aquoso e o óleo essencial das folhas de A. zerumbet não apresentam ações citotóxicas nem genotóxicas nos modelos testados.

Para avaliar a eficiência micorrízica em substratos usados na aclimatização de Alpinia purpurata (alpinia) e Zingiber spectabile (sorvetão) e a tolerância de plantas de alpinia micorrizadas, ao ataque de nematóides, foram realizados dois experimentos. O primeiro foi inteiramente casualizado, em fatorial, com quatro tratamentos de inoculação (sem ou com Gigaspora albida, Glomus etunicatum e Acaulospora longula), três substratos (solo; solo:vermicomposto e solo:areia:vermicomposto) com oito repetições. Após 100 dias não houve interação entre FMA e substratos relacionada ao crescimento das duas espécies, porém o substrato solo:vermicomposto beneficiou a produção vegetal. No segundo experimento, plantas de alpinia micorrizadas foram inoculadas com Meloidogyne arenaria. Após 270 dias plantas infestadas com o nematóide e associadas com G. etunicatum e com A. longula tiveram maior massa seca aérea e área foliar em relação aos demais tratamentos. A micorrização com G. albida e A. longula reduziu o número de galhas, enquanto o número de ovos do nematóide foi menor no tratamento com G. albida. A presença de M. arenaria afetou a colonização micorrízica. Concluiu-se que na fase de aclimatização, substratos orgânicos favorecem a formação de mudas vigorosas de alpinia e sorvetão, dispensando a inoculação com FMA, e que a micorrização de A. purpurata com A. longula induz tolerância a M. arenaria, constituindo alternativa para diminuir os prejuízos da meloidoginose

CaracterizaÃÃo da aÃÃo vasodilatadora do extrato aquoso obtido de folhas Alpinia zerumbet K.SCHUM em aorta de ratos / Characterization of vasorelaxant effects of aqueous extract obtained from leaves of Alpinia zerumbet K. SCHUM in rat aorta

nÃo hà / A Alpinia zerumbet à uma planta comumente encontrada na regiÃo do nordeste do Brasil. Essa planta à conhecida popularmente por colÃnia e à muito utilizada na medicina popular por suas propriedades diurÃticas e anti-hipertensivas. O objetivo do presente estudo foi investigar se o extrato aquoso das folhas de A. zerumbet (EAAz) apresentava atividade vasodilatadora em aorta de rato prÃ-contraÃda com fenilefrina e, elucidar o seu mecanismo de aÃÃo. Ratos machos Wistar (250 a 300 g), provenientes do biotÃrio da UFC, foram sacrificados por deslocamento cervical e a aorta torÃcica removida e dissecada. Os anÃis da aorta (4 a 5 mm) foram montados em cÃmeras orgÃnicas, contendo soluÃÃo de Krebs e aerados com carbogÃnio a 37ÂC, para as medidas de variaÃÃes na tensÃo isomÃtrica. O endotÃlio dos anÃis da aorta foi removido mecanicamente para estudar o seu envolvimento no efeito vasodilatador do EAAz. Para estudar o envolvimento do Ãxido nÃtrico (NO), GMPc, prostanÃides, canais de potÃssio ativados por Ca2+, espÃcies reativas do oxigÃnio e a ativaÃÃo das enzimas quinases Src e PI3 quinase, as preparaÃÃes foram tratadas, respectivamente, com L-NAME (100 ÂM), ODQ (10 ÂM), indometacina (10 ÂM), caribdotoxina (100 nM) mais apamina (100 nM), MnTMPyP (100 ÂM), catalase (500 U/ml), PEG-catalase (500 U/ml), SOD (500 U/ml) e PP2 (20 ÂM), Wortmannin (300 nM). O efeito vasodilatador do EAAz (0,05; 0,15; 0,5; 1,5; 15 e 50 Âg/mL), acetilcolina (10-8 - 10-5 M) e nitroprussiato de sÃdio (10-8 M) foram avaliados em aorta de ratos prÃ-contraÃda com fenilefrina (10-8 - 3x10-8 M), antes e apÃs tratamento com os inibidores especÃficos. A vasodilataÃÃo induzida pelo EAAz mostrou-se dependente do endotÃlio e mediada pelo NO via GMPc jà que o relaxamento vascular foi abolido na presenÃa do Ãster metÃlico de NG-nitro-l-arginina e do ODQ. A vasodilataÃÃo tambÃm foi reduzida pelo MnTMPyP (permeante celular mimÃtico da SOD), polietilenoglicol catalase, PP2 (inibidor da quinase Src) e Wortmannin (inibidor da fosfoinositideo 3-quinases). A via de produÃÃo de EROs induzida pelo EAAz foi inibida significativamente pela aÃÃo do MnTMPyP e PEG-catalase. Ocorreu tambÃm a inibiÃÃo da via EROs/PI3 quinase/Src quinase que estÃo envolvidas na ativaÃÃo da eNOS. O EAAz causou um relaxamento dependente do endotÃlio via NO/GMPc com o possÃvel envolvimento de EROs e das Src quinase e fosfoinositideo 3-quinase dependente de Akt. Dessa forma, pode-se concluir que o relaxamento vascular induzido pelo EAAz poderia explicar o tradicional uso do chà das folhas de A. zerumbet para o tratamento da hipertensÃo arterial. / The Alpinia zerumbet is a plant commonly found in the northeastern region of Brazil. This plant is popularly known as colony and is widely used in folk medicine for its diuretic and antihypertensive properties. The aim of this study was to investigate whether the aqueous extract of leaves of A. zerumbet (EAAz) had vasodilator activity in rat aorta pre-contracted with phenylephrine and to elucidate its mechanism of action. Male Wistar rats (250-300 g) from the vivarium of UFC, were killed by cervical dislocation and the thoracic aorta removed and dissected. The aortic rings (4-5 mm) were mounted in organ cameras, and containing Krebs solution aerated with carbogen at 37  C to measure changes in isometric tension. The endothelium of the aortic rings was removed mechanically to study their involvement in the vasodilator effect of EAAz. To study the involvement of nitric oxide (NO), cGMP, prostanoid, potassium channels activated by Ca2+, reactive oxygen species and activation of enzymes kinases Src and PI3 kinase, the preparations were treated respectively with L-NAME (100 ÂM ), ODQ (10 ÂM), indomethacin (10 ÂM), carybdotoxin (100 nM) plus apamin (100 nM), MnTMPyP (100 ÂM), catalase (500 U / ml), PEG-catalase (500 U/ ml), SOD (500 U / ml) and PP2 (20 ÂM), wortmannin (300 nM). The vasodilatory effect of EAAz (0.05, 0.15, 0.5, 1.5, 15 and 50 Âg / mL), acetylcholine (10-8 - 10-5 M) and sodium nitroprusside (10-8 M ) were evaluated in the aorta of rats pre-contracted with phenylephrine (10-8 - 3x10-8 M) before and after treatment with the specific inhibitors. EAAz induced vasodilation was dependent and endothelium mediated by NO via cGMP as the vascular relaxation was abolished in the presence of the methyl ester of NG-nitro-L-arginine and ODQ. Vasodilation was also reduced by MnTMPyP (mimetic cellular permeant SOD), catalase polyethylene glycol, PP2 (Src kinase inhibitor) and wortmannin (inhibitor of phosphoinositide 3-kinase). The route of production of ROS induced EAAz was significantly prevented by the action of MnTMPyP and PEG-catalase. There was also the inhibition of the kinase ROS/PI3 / Src kinase that are involved in the activation of the eNOS. The EAAz caused an endothelium-dependent relaxation via NO / cGMP with the possible involvement of ROS and Src kinase and phosphoinositide 3-kinase-dependent Akt. Taking us to the conclusion that the vascular relaxation induced by EAAz could explain the traditional use of the tea leaves of A. zerumbet for the treatment of hypertension.
