The early blight disease, caused by the fungus Alternaria solani is one of the most important disease of this culture. The control is usually done with fungicides that can cause the production of fruits contaminated with pesticide. The use of homeopathy has been a practice for disease control, and an alternative to reduce these chemical drugs. The objective of this study was to evaluate the fungitoxic activity against A. solani, the disease control of early blight and the effect on plant growth of tomato.of homeopathic drugs Propolis, Isotherapic of A. solani and Isotherapic of ash in at dinamizations 6, 12, 30 and 60CH, and Sulphur, Silicea terra, Staphysagria, Phosphorus, Ferrum sulphuricum and Kali iodatum at dinamizations 6, 12, 30 and 100CH. In vitro assays was investigate the antimicrobial activity of the treatments on mycelial growth, sporulation and germination of spores of A. solani. The results indicated that Sulphur 100CH and Staphysagria 100CH reduced the mycelial growth compared with the control treatment wint distilled water and water-etanol solution. Propolis, 6, 30 and 60CH and Ferrum sulphuricum 6 and 30CH inhibited the sporulation, Isotherapic of A. solani in 6CH, Isotherapic of ash at 6CH, and Ferrum sulphuricum 6 and 30CH reduced the spores germination of A. solani. The data also indicated that the distilled water at 60 and 100CH reduced the mycelial growth. In the second phase, anassay was conducted in a greenhouse with Propolis, Sulphur and Ferrum sulphuricum. The work was carried out in pots, and at 19 days after transplanting of seedlings, the 6 ths. leaves were treated and 72 hours after the treatment, the 6ths. and 7ths. leaves were inoculated with A. solani. The severity were evaluated 7, 10, 14 and 16 days after inoculation and was calculated the area under the disease progress curve (AUDPC). Sulphur, 12 and 30 CH, Ferrum sulphuricum at 6, 12 and 30CH and Propolis in all dinamizations reduced the AUDPC of early blight. Sulphur 60CH and water-etanol solution 10% had systemic effect in the induction of resistance. Propolis at 30 and 60CH increased the amount of root, Ferrum sulphuricum 60CH and Sulphur in 30CH increased the root dry weight, while Propolis on 30 and 60CH, Ferrum sulphuricum in 6, 12, 30 and 60CH and Sulphur in all dinamizations increased the mass of the shoot. The results indicate that the homeopathic drugs tested have potential for control of early blight in the tomato, by direct actions on the pathogen and host. / O tomate (Lycopersicon esculentum Mill) é a segunda hortaliça mais consumida no Brasil. A doença pinta preta, causada pelo fungo Alternaria solani é uma das mais importantes. O tratamento normalmente é feito com fungicidas o que ocasiona a produção de frutos contaminados por resíduos de agrotóxicos. O uso da homeopatia tem sido uma prática relevante no controle de doenças, e uma alternativa para reduzir esses contaminantes. O objetivo deste trabalho foi avaliar a ação dos medicamentos homeopáticos Propolis, Isoterapico de A. solani e Isoterapico de cinza, nas dinamizações 6, 12, 30 e 60CH, e Sulphur, Silicea terra, Staphysagria, Phosphorus, Ferrum sulphuricum e Kali iodatum nas dinamizações 6, 12, 30 e 100CH, na atividade fungitóxica sobre A. solani, no controle da doença pinta preta, e no crescimento das plantas de tomate. Foi realizado ensaio in vitro visando investigar a atividade antimicrobiana dos tratamentos no crescimento micelial, esporulação e germinação de esporos de A. solani. Os resultados indicaram que Sulphur em 100CH e Staphysagria em 100CH reduziram o crescimento micelial comparando com os controles água destilada e solução hidroalcoólica. Na inibição da esporulação, Propolis em 6, 30 e 60CH e Ferrum sulphuricum em 6 e 30CH diferiram dos controles. Isoterapico de A. solani em 6CH, Isoterapico de cinza em 6CH e Ferrum sulphuricum em 6 e 30CH reduziram a germinação de esporos de A. solani. Os dados indicaram também que a água destilada, quando dinamizada em 60 e 100CH, reduziu o crescimento micelial. Na segunda fase, foi realizado ensaio em casa de vegetação com ambiente controlado com os medicamentos Propolis, Sulphur e Ferrum sulphuricum. O trabalho foi realizado em vasos e, aos 19 dias após o transplante das mudas, as 6ªs folhas foram tratadas e 72 horas após, as 6ªs e 7ªs folhas foram inoculadas com A. solani. Foram feitas avaliações da severidade com 7, 10, 14 e 16 dias após a inoculação e com os dados calculada a área abaixo da curva de progresso da doença (AACPD). Ao término foram avaliadas variáveis de crescimento. Sulphur em 12 e 30 CH, Ferrum sulphuricum em 6, 12 e 30CH e Propolis em todas as dinamizações reduziram a AACPD da pinta preta, Sulphur em 60CH e solução hidroalcóolica 10% apresentaram efeito sistêmico na indução de resistência. Propolis em 30 e 60CH incrementaram o volume de raiz, Ferrum sulphuricum em 60CH, Sulphur em 60CH e Propolis em 30CH incrementaram a massa seca da raiz, enquanto que Propolis em 30 e 60CH, Ferrum sulphuricum em 6, 12, 30 e 60CH e Sulphur em todas as dinamizações incrementaram a massa da parte aérea. Os resultados indicam que os medicamentos homeopáticos testados têm potencial para controle de pinta preta no tomateiro, por ação direta no patógeno e no hospedeiro.

Preparados em altas diluições para o manejo fitossanitário e pós-colheita do tomateiro / High dilution preparations to tomato crop and post-harvest management

Synthetic organic pesticides that are used to manage tomato crops have been harmful to human being and environment as well. Fruits of tomato are very sensitive to handling and its quality is highly influenced by the crop system and post-harvest practices. The objective of this research was to evaluate the effect of high dilution preparations to manage pest and disease, and post-harvest conservation of tomatoes under organic production systems. Experiments were conducted as follow: a) Two experiments under field conditions; b) one experiment in greenhouse; c) one bio-assay concerning post-harvesting; d) twelve bio-assays in vitro conditions. The experiments were conducted in the Lages Experimental Station area of EPAGRI and at Centro Agroveterinario of University of Santa Catarina State, SC. Treatments consisted of high dilutions of Staphysagria, Arsenicum album, Sulphur at 12CH (centesimal hahnemaniana dilution), and Arnica montana, Sulphur, tomato, and jua (Solanum aculeatissimum), nosodes at 12 and 24DH. Bourdoux mixture at 0.3%, Cupric formulation EEC at 50 ppm, and Bacillus thuringiensis were included as standard treatments. Data were from field experiments consisted of septoriosis incidence (Septoria lycopersici), insect occurrence, damage caused by Agathomerus sellatus, yields, and fruit quality attributes. Data analysis was performed throughout statistical package. Results showed that Staphysagria 12CH and Arnica Montana 12DH could increase tomato fruit weigh. Damage caused by tomato caterpillars were reduced by spraying Sulphur 12CH and Bacillus thuringiensis. B. thuringiensis reduced also the incidence of septoriosis. High dilution preparation interfered to insect occurrence and reduced the damage of Agathomerus sellatus in the stages of larvae and adults. Tomato plants treated by jua nosodes at 12 and 24CH increased the SST (° Brix) contents, which gives better qualities of processing and palatability, as well. In the greenhouse experiment, it was evaluated the treatments of Arnica montana, Sulphur, tomato and jua nosodes, all at 12 and 24DH dinamization. Bourdoux mixture at 0.3% and cupric formulation EEC at 50 ppm were considered as standard treatments. It was observed that tomato nosodes at 12DH and 24DH supressed the septorioris incidence. In the post-harvest bio-assay, fruits were treated with Calcarea carbonica at 6, 12, 24CH dinamizations. The evaluation was concerning chemical contents and physical attributes. Calcarea carbonica post-harvest treatment did not affected the acidity, SST (° Brix), hardness or the weight losses of tomato fruits. However, Calcarea carbonica 24CH delayed the fruit maturation in such way that the kind sauce fruit was reduced. In the laboratory bio-assays the follow high dilution preparations were studied: Staphysagria, Arsenicum album, and Nitricum acidum at 6, 12, 25, 30, 50, 60, 80, and 100CH 11 dinamizations. All preparations were tested in two media combined by two application methods into Petry dishes with colonies of the fungus Alternaria solani. Bioassay was studied by measuring the diameter of the colonies. The measurements were made from 24h until the 10th day after install the assays. It was observed that the media with basic composition were more suitable to screening high dilution preparations against A. solani before to test in the field. The application method of high dilution preparations over the fungus colony was better than incorporated one. The use of high dilution preparations has the potential effect to replace pesticide used on tomato crops and it would be a suitable management strategy to fill legal requirements for organic production systems. / O tomateiro (Solanum lycopersicum L.) sob sistema de produção convencional está constantemente sujeito à pragas e doenças. Pesticidas orgânicos sintéticos usados para seu manejo podem causar graves pertubações ao meio ambiente e a saúde humana. Frutos de tomateiro também são sensíveis ao manuseio e sua qualidade é afetada pelo sistema de cultivo e pelas práticas de conservação pós-colheita. O objetivo deste trabalho foi avaliar o efeito de preparados em altas diluições no manejo fitossanitário e pós-colheita da cultura do tomateiro sob sistema orgânico de produção. Foram conduzidos (a) dois experimentos em condições de campo, (b) um experimento em casa de vegetação, (c) um ensaio de conservação pós-colheita em laboratório e (d) doze bioensaios in vitro. Os experimentos foram conduzidos na Estação Experimental da EPAGRI de Lages e, no Centro de Ciências Agroveterinárias da UDESC. Os tratamentos avaliados nos experimentos a campo foram os preparados em altas diluições de Staphysagria, Arsenicum album, Sulphur na dinamização 12CH (centesimal hahnemaniana); Arnica montana, Sulphur, nosódios de tomateiro e de juá (Solanum aculeatissimum), nas dinamizações 12 e 24DH (decimal hahnemaniana). Calda bordalesa 0,3%, Calda cúprica EEC a 50 ppm e Bacillus thuringiensis foram incluídos em doses ponderais e considerados tratamentos padrões. Nos experimentos a campo avaliou-se a incidência de septoriose (Septoria lycopersici), ocorrência e danos por insetos, produtividade e atributos químicos de qualidade dos frutos. A análise dos dados mostrou que os preparados Staphysagria 12CH e Arnica montana 12DH proporcionaram a maior peso de frutos. Danos causados por brocas foram reduzidos com a aplicação de Sulphur 12CH igualando a Bacillus thuringiensis. B. thuringiensis também reduziu a incidência de septoriose. Observou-se que os preparados em altas diluições interferiram na ocorrência de insetos em plantas de tomateiro e reduziram os danos causados por Agathomerus sellatus, tanto na fase larval quanto na fase adulta. Tomateiros tratados com nosódio de juá 12 e 24DH tiveram incremento no teor de SST (° Brix) dos frutos, atributo que melhora a palatabilidade e a qualidade para a industrialização. No experimento em casa de vegetação, avaliou-se Arnica montana, Sulphur, nosódios de tomateiro e de juá, nas dinamizações 12 e 24DH. Calda bordalesa 0,3% e calda cúprica EEC a 50 ppm também foram incluídas em doses ponderais e considerados tratamentos padrões. Observou-se que o nosódio de tomateiro na 12DH e 24DH suprimiram a incidência de septoriose nas plantas tratadas. No ensaio em laboratório de conservação pós-colheita, frutos foram tratados com Calcarea carbonica nas dinamizações 6, 12, 24CH. Avaliou-se atributos químicos e físicos dos frutos. Calcarea carbonica em tratamento pós-colheita não interferiu na acidez, SST (° Brix), firmeza e perda de massa fresca dos frutos. Embora, Calcarea9 carbonica 24CH retardasse a formação de frutos do tipo molho. Nos estudos in vitro foram avaliados os preparados Staphysagria, Arsenicum album e Nitricum acidum nas dinamizações 6, 12, 25, 30, 50, 60, 80 e 100CH, em meios de cultivo básico e enriquecido e, em dois métodos de aplicação. Nos bioensaios foi avaliado o diâmetro das colônias do fungo Alternaria solani iniciando-se 24 horas após a instalação do ensaio e estenderam-se até o décimo dia. Observou que meios de cultivo com composição básica foram mais adequados para o screening de preparados em altas diluições e suas dinamizações, em estudos in vitro com A. solani. A aplicação dos preparados em altas diluições sobre a colônia do fungo foi o método mais adequado para estudos in vitro. Preparados em altas diluições tem potencial para serem eficazes na substituição de agrotóxicos que são utilizados na cultura do tomateiro, e poderiam ser adequados para preencherem requisito legal em sistemas orgânicos de produção

Conselho Nacional de Desenvolvimento Científico e Tecnológico / The tomato (Lycopersicon esculentum Mill) is an important crop in the world and an important product for trade "in nature and for industry. The amount of fungicides used in tomato crop for control of diseases is high and contributes significantly to production costs of tomato, besides the high risk of intoxication of the applicator and environmental problems. So, the use of disease forecasting systems based on mathematical models to manage the applications of fungicides for foliar diseases control in tomato may to reduce production costs, by decreasing the number of fungicide applications and machinery operation during the culture cycle. The objective was to test the systems for predicting occurrence of late blight (Phytophthora infestans) and early blight (Alternaria solani) on irrigated tomato under two climatic conditions and estimate the cost of deployment. Three experiments were conducted, the first experiment in the spring of 2007, in Santa Maria - RS. The other two were accomplished in Cristalina - GO. There were two seasons of transplanting (01/04/08 and 03/05/08). Used the hybrid U2006 (UNILEVER), with growth habit determined. The spacing between the rows of plants was 1.3 m between plants in rows of 0,3 m. The randomized block design with three replications was used, each plot of 5,2 m wide, consisting of 4 rows of plants with 5 m long. The systems for predicting disease FAST, to early blight, and Blitecast to late blight were used. Irrigation was performed by spraying in Santa Maria and sprinkler, surface drip and subsurface in Cristalina-GO. The results showed that the use of disease forecasting systems is the most appropriate way to control diseases in tomato, as with the use of disease forecasting systems that can significantly reduce the number of fungicide applications in relation the calendar, based on weekly applications in areas with conditions generally unfavorable to the development of disease. Considering the scenarios of reductions of the number of fungicide applications (30, 43 and 65%), compared to the weekly application, in climatic conditions unfavorable to disease development, it is possible to recover the costs of implementing the system prediction in all scenarios. / A cultura do tomate (Lycopersicon esculentum Mill) é uma das mais expressivas culturas no cenário agrícola mundial, constituindo importante produto para o comércio in natura e indústria de extratos. O volume de fungicidas utilizado na cultura do tomateiro no controle das principais doenças é elevado e contribui significativamente nos custos de produção de tomate, além do elevado risco de intoxicação do aplicador e dos problemas ambientais. Nesse sentido, utilização de sistemas de previsão e aviso, baseados em modelos matemáticos para gerenciar o manejo das aplicações de fungicidas para o controle das principais doenças foliares no tomateiro poderá auxiliar na redução dos custos de produção, através da diminuição do número de aplicações de fungicidas e de operações de máquinas durante o ciclo de desenvolvimento da cultura. O objetivo do trabalho foi testar os sistemas de previsão de ocorrência de requeima e alternaria em tomateiro industrial irrigado sob duas condições climáticas e estimar o seu custo de implantação. Foram realizados três experimentos, o primeiro experimento no segundo semestre de 2007, em Santa Maria RS. Os outros dois foram realizados no município de Cristalina GO. Realizaram-se duas épocas de transplante (01/04/08 e 03/05/08). Utilizou-se a cultivar híbrida U2006 (UNILEVER), de hábito de crescimento determinado. O espaçamento entre as fileiras de plantas foi de 1,3 m e entre as plantas nas fileiras de 0,3 m. Utilizou-se o delineamento experimental de Blocos ao Acaso com três repetições, sendo cada parcela de 5,2 m de largura, composta de 4 fileiras de plantas com 5 m de comprimento. Utilizaram-se os sistemas de previsão de doenças FAST, para mancha de alternaria, e BLITECAST, para requeima. A irrigação foi realizada por aspersão em Santa Maria e por aspersão, por gotejamento superficial e subsuperficial em Cristalina. Os resultados mostraram que o uso de sistemas de previsão de doenças é a maneira mais adequada para o controle de doenças na cultura do tomateiro, pois com o uso de sistemas de previsão de doenças se consegue reduzir significativamente o número de aplicações de fungicidas, em relação ao calendário semanal, em regiões com condições geralmente desfavoráveis ao desenvolvimento de doenças. Considerando-se os cenários de reduções do número de aplicação de fungicidas (30, 43 e 65%), em relação ao calendário semanal de aplicação, condições climáticas desfavoráveis ao desenvolvimento de doenças, é possível recuperar os custos de implantação do sistema de previsão em todos os cenários.

EFFICACY OF ORGANICALLY CERTIFIABLE MATERIALS AND NATURAL COMPOUNDS AGAINST FOLIAR HEMIBIOTROPHIC AND NECROTROPHIC FUNGI IN CANTALOUPE AND TOMATO

Kentucky reported a solid 13.1% growth in certified organic land from 1997 to 2002. The relative lack of research on disease management practices in Kentucky consistent with organic regulations is an issue that needs to be addressed to provide more reliable information to local farmers. Thus, the first objective of this research was to investigate the potential disease control obtained with natural, organically certifiable spray materials against Colletotrichum orbiculare in vitro and in vivo. The second objective was to test certifiable spray materials in combinations to identify synergistic interactions. The third objective was to evaluate Organic Material Review Institute (OMRI)-certified materials for managing Septoria leaf spot and early blight in tomato under field conditions. The fourth objective was to evaluate chitosan-based products against C. orbiculare in vitro and in vivo. Essential oils, Trilogy®, and Actinovate®, failed to suppress C. orbiculare in vitro as well as cucurbit anthracnose. Bicarbonate salts, Regalia®SC, Sonata®, copper based-products, lime sulfur and water-soluble chitosan showed high antifungal activity in vitro. Bicarbonate salts, Sonata®, Serenade Max®, Soil Gard 12G®, copper based-products and lime sulfur reduced anthracnose disease severity in vivo. In the synergism experiments only a limited number of mixtures showed synergistic interactions, but even in those cases, the effect was not consistent between experiments. The main response obtained was antagonism. In field experiments the most effective fungicides for managing Septoria leaf spot and early blight of tomato were copper-based fungicides. None of the biological-based products (Sonata® and Serenade Max®)), plant-based extracts (Trilogy® and Regalia® SC), chitosan, ammonium bicarbonate nor horticultural lime sulfur provided a significant reduction in disease severity. For the fourth objective, water-soluble chitosan with a molecular weight between 3 to 10 kDa (80 and 85% deacetylated) showed the highest antifungal activity among all chitosan-based products evaluated in vitro. Also, combining the in vitro and in vivo results suggest that the antifungal activity of chitosan-based products is molecular weight- and concentration-dependent. These results provide a significant advance in the evaluation of the efficacy of OMRI-certified materials and natural materials to help organic farmers in Kentucky and the USA to manage diseases.

Entretanto, apesar de existirem diversos benefícios na redução ou até eliminação do uso de defensivos, a grande diversidade de doenças em tomateiro (Lycopersicon esculentum Mill.), capazes de limitar a produção, torna necessária a busca por alternativas viáveis, eficientes e tecnicamente comprovadas. Dentre os organismos mais estudados, bactérias têm sido relatadas como agentes de biocontrole capazes de atuar por meio de mecanismos como antibiose, parasitismo, competição e indução de resistência. O presente trabalho teve como objetivos selecionar bactérias do filoplano do tomateiro, baseado em uma estratégia de seleção in vivo, verificando se há um método de isolamento que permita obter antagonistas eficientes no controle da pinta-preta, causada por Alternaria solani, requeima por Phytophthora infestans, mancha-bacteriana pequena por Pseudomonas syringae pv. tomato e mancha-bacteriana por Xanthomonas vesicatoria. Objetivou ainda estudar a possibilidade dos mecanismos de antibiose e indução de resistência serem responsáveis pelo controle destas doenças e se testes de antibiose in vitro são adequados como critério de seleção. Caracterizar aspectos biológicos dos antagonistas que podem otimizar sua aplicação como agente de biocontrole. Determinar a quais produtos antimicrobianos os isolados são insensíveis, visando fornecer subsídios para o desenvolvimento de meios semi-seletivos e estudos de dinâmica populacional. Verificar sua compatibilidade com antibióticos e fungicidas registrados para o controle de enfermidades do tomateiro, a fim de inseri-lo no sistema de manejo integrado e estudar a eficiência de antagonistas selecionados em condições de campo. Os resultados demonstram que, em folíolos mais jovens, os métodos de isolamento que visam obter bactérias da população total e da superfície do filoplano, foram os que permitiram obter a maioria dos antagonistas. O único obtido de folíolos mais velhos foi proveniente da população capaz de habitar sítios protegidos do filoplano e/ou resistir a fatores de estresse. Não se observou relação entre características biológicas dos antagonistas e dos patógenos testados. Nos testes de antibiose com os antagonistas selecionados, o isolado UFV-STB 6 foi capaz de produzir compostos voláteis e inibir a germinação de cistos de Phytophthora infestans, o que possivelmente deve estar envolvido no controle da requeima. O isolado UFV-IEA 6 produziu quitinase, havendo uma tendência em reduzir a taxa de crescimento de Alternaria solani por compostos voláteis. Ficou demonstrado que os testes de antibiose in vitro são inadequados como critério para seleção de agentes de biocontrole do filoplano de tomateiro. A caracterização dos melhores antagonistas demonstrou que três são bactérias Gram-positivas, em forma de bastonete, e uma Gram-negativa, pleiomórfica. Dentre as Gram-positivas todas são anaeróbias facultativas e uma forma endósporos. Nenhum antagonista foi capaz de causar reação de hipersensibilidade (HR) em fumo e produzir pigmento fluorescente in vitro. Os períodos de geração calculados a partir das curvas de crescimento revelaram que três isolados são capazes de se multiplicar rapidamente em meio de cultura, o que é uma característica desejável. Os resultados obtidos a partir dos antibiogramas, mostraram que existem antibióticos que podem ser utilizados para elaboração de meios semi-seletivos, adequados a cada antagonista e os testes de compatibilidade com antibióticos e fungicidas utilizados na cultura do tomateiro revelaram que os antagonistas podem ser expostos aos fungicidas benomyl, enxofre, dimetomorph e tiofanato-metílico. Verificou-se também a inadequação de se utilizarem compostos antimicrobianos em meio de cultura para isolamento de agentes bacterianos de controle biológico, uma vez que os antagonistas selecionados foram sensíveis à maioria dos produtos testados. Os testes com as enzimas indicadoras do estado de indução de resistência, β-1,3-glucanases, Fenilalanina amônia-liase (PAL), Peroxidases (PO), Polifenoloxidases (PPO) e Lipoxigenases (LOX), indicaram que o isolado UFV-IEA 6 foi capaz de promover aumento significativo na atividade das PO, evidenciando a possibilidade do antagonista agir como indutor de resistência. Esse parece ser o primeiro caso que se tem conhecimento de uma bactéria não fitopatogênica do filoplano induzindo resistência na mesma cultura de onde foi obtida. Os testes com os dois antagonistas em condições de campo demonstraram que UFV-STB 6 foi o mais eficiente em reduzir a severidade da requeima no terços médio e superior das plantas, enquanto UFV-IEA 6, somente no terço superior. Houve tendência na redução do progresso da septoriose por UFV-STB 6 e capacidade em diminuir o número de frutos com sintomas de requeima. Os resultados demonstram o potencial de uso dos agentes de biocontrole selecionados para as doenças da parte aérea de tomateiro estudadas. / Farmers are increasingly adapting environmentally less aggressive management systems for food production and there are many benefits in reducing or even eliminating pesticide use. Due to a large number of production limiting diseases on tomato (Lycopersicon esculentum), it is desirable to find technically viable and proven alternatives for their control. Among the microorganisms, bacteria have been reported as biocontrol agents capable of acting through antibiosis, parasitism, competition and induced resistance. The present study aimed at selecting the tomato phylloplane bacteria, based on in vivo isolation strategy, and to determine if this method permits obtaining efficient antagonists to control Alternaria solani leaf spot, Phytophthora infestans blight, small bacterial leaf spot caused by Pseudomonas syringe pv tomato, and bacterial leaf spot caused by Xanthomonas vesicatoria. The study also aimed at determining mechanisms of action, such as antibiosis and induced resistance, involved in disease control, and to determine if the antibiosis tests are sufficient selection criteria. The study also included biological characterization of the antagonists that may optimize their use. To help develop selective or semi-selective media for population dynamic studies, insensitivity of selected isolates to some antimicrobial compounds was also determined. The compatibility of select antagonists with antibiotics and fungicides registered for control of tomato diseases was elucidated so that the antagonist can be inserted in the integrated management. Field studies were done to determine the efficiency of select antagonists. The isolation method that obtain total bacterial population from the phylloplane of the young leaflets permitted obtaining maximum number of antagonists. The only one isolate obtained from the older leaflets originated from the population capable of inhabiting protected sites of phylloplane and/or that resist stress factors. There was no relation between biological characteristics of the antagonists and of the pathogen tested. In the antibiosis testes, the isolate UFV-STB 6 produced volatile compounds that inhibited germination of P. infestance cyst and may be involved in the control of blight. The isolate UFV-IEA 6 produced chitinase and showed a tendency to reduce A. solani growth by the volatile compounds. In vitro antibiosis testes were inadequate criteria to select biocontrol agents from tomato phylloplane. The characterization of promising antagonists showed that three were Gram positive bacilli and one was gram negative pleiomorphic bacteria. Among the Gram positives all were facultative anaerobes and the one formed endospores. None of the antagonists caused hypersensitive reaction (HR) in tobacco, and did not produce fluorescent pigment in vitro. The generation period calculated from the growth curve revealed that three isolates are capable of multiplying rapidly in the culture media, which is a desirable characteristic. The antibiograms showed that there are antibiotics that can be used for elaboration of semi-selective media for each of antagonists and the compatibility testes with antibiotics and fungicides used on tomato crop revealed that the antagonists can be exposed to fungicides such as benomyl, sulfur, dimethomorph and thiophante-methyl. Many antimicrobial compounds were inhibitory to the selecte antagonists in culture media used for isolation of bacterial biocontrol agents, therefore were inadequate for use in selective media. The analysis of enzymes involved in induced resistance, like -1,3-gluconase, phenylalanine ammonia lyase (PAL), peroxidase (PO), polyphenol oxidase (PPO) and lipoxigenase (LOX), showed that the isolate UFV-IEA 6 was capable of increasing PO activity, showing the possibility of being a resistance inducer . This appears to be the first case of a non-pathogenic phylloplane bacterium inducing resistance in a plant of origin. The field testes with two antagonists, UFV-STB 6 was more efficient in reducing the blight severity in the middle and upper third of the plant, while UFV-IEA 6 only in the upper third. The latter isolate also showed a tendency for reducing the Septoria leaf spot progress and the number of fruits with the blight symptoms. Os componentes epidemiológicos da resistência, período de incubação (PI), número de lesões (NL), a severidade, taxa de expansão das lesões (TEL), período latente (PL) e o número de esporos por área foliar afetada (NEAA) foram quantificados nos terços inferior, médio e superior de plantas dos cultivares Aracy (resistente), Delta (resistência intermediária), Desirée(suscetível) e Bintje (suscetível) inoculadas com Alternaria solani numa concentração de 10 3 conídios.mL -1 . No cultivar Aracy constatou-se maior PI, redução do NL, da TEL e conseqüentemente da severidade em todos os terços avaliados. Já o menor NL, TEL, e severidade no terço médio explicam a resistência moderada do cultivar Delta. O cultivar Desirée, classificado como suscetível, apresentou, no terço superior, NL e severidade similar ao cultivar Aracy. O PL e o NEAA não foram adequados para a avaliação da resistência em condições de casa de vegetação e, provavelmente, foram influenciados pela falta de molhamento foliar. Todos os componentes foram influenciados pela idade dos tecidos. Maior PI e menores valores de NL, TEL, severidade e AACPD foram observados no terço superior, comprovando a menor suscetibilidade desses tecidos. Foram efetuados estudos histológicos do processo infeccioso de A. solani nas diferentes cultivares e terços usando a técnica de diafanização de tecidos. Foi quantificado o número de conídios germinados, número de apressórios, número de penetrações diretas e número de sítios de penetração exibindo células com reação de hipersensibilidade (HR). Não se constatou mecanismos de resistência que pudessem ser associados com os níveis de resistência dos cultivares estudados, nem com a maior resistência dos tecidos jovens. Constatou-se uma relação positiva entre o número de sítios de penetração mostrando HR e a resistência do cultivar Aracy. Houve associação desse componente com a resistência dos tecidos jovens. A expressão de genes homólogos à glucanase básica de tabaco (gluB) e à chitinase básica de feijão (chiB) como resposta a infecção de A. solani, foi analisada nos terços inferior e superior dos cultivares Bintje e Aracy, mediante a técnica de "Northern Blotting". Um aumento na expressão de transcritos a gluB e chiB foi constatado no terço inferior do cultivar Aracy, mas não em Bintje. Esse padrão de expressão sugere um possível envolvimento de isoformas destes genes com a resistência de Aracy a A. solani. Essa expressão diferenciada, não foi observada em folhas do terço superior desse cultivar sugerindo um padrão de regulação gênica dependente da idade dos tecidos, e o envolvimento de outros mecanismos bioquímicos na resistência dos tecidos jovens. / The resistance components of four potato cultivars to Alternaria solani were studied in three different levels: 1) quantification of epidemiological components related to resistance in cultivars Aracy, Delta, Desirée, and Bintje, and the effect of age-related resistance in these components; 2) histological analysis of the infectious process in these cultivars, aiming at identifying possible structural mechanisms related to resistance; 3) differential expression of genes homologous to gluB (basic glucanase of tobacco) and chiB (basic chitinase of bean) that encode pathogenesis-related proteins (PR) and the correlation between gene expression levels and cultivar/tissue age resistance levels. The epidemiological components related to resistance, incubation period (PI), number of lesions (NL), early blight severity, lesion expansion rate (TEL), latent period (PL), and the number of spores produced by lesion area (NEAA) were quantified in the lower, middle, and upper parts of cultivars Aracy (resistant), Delta (intermediate resistance), Desirée (susceptible), and Bintje (susceptible) inoculated with a suspension of 103 conidia of A. solani.mL-1. Longer PI, reduced NL, low TEL, and low severity were recorded in Aracy in all parts of the plant. For 'Delta', the reduced NL, low TEL and severity on the middle part of the plant corroborates its intermediate resistance level. Desirée is a susceptible cultivar, but NL and severity on the upper part did not differ from Aracy. The PL and NEAA were not informative when assessing resistance under greenhouse conditions. Probably, PL and NEAA were influenced by the shortage or lack of leaf wetness. All components were influenced by tissue age. Longer PI and smaller values of NL, TEL, and severity were observed in the upper leaves, indicating higher resistance of these tissues. Histological analyses of the infectious process of A. solani in different cultivars were carried out by tissue discoloration. The number of germinated conidia, number of apressoria, number of direct penetrations, and number of penetration sites with cells exhibiting hypersensitive reaction (HR) were quantified. No mechanisms of resistance were associated with cultivar resistance levels or with tissue age. For cultivar Aracy there was a direct relationship between number of penetration sites showing HR and cultivar resistance. The number of sites with HR was also associated with resistance of young tissues. The expression of gluB and chiB in response to A. solani infection was analyzed in the lower and upper parts of 'Bintje' and 'Aracy' plants by Northern blotting analysis. An increase in the level of transcripts related to gluB and chiB was recorded in the lower part of Aracy, but not in Bintje. Differential expression suggests the involvement of isoforms of these genes in Aracy resistance. Differential expression was not observed in leaves of the upper part of Aracy suggesting an age-dependent gene regulation process and/or the involvement of other biochemical mechanisms associated with young tissue high resistance level. / Tese importada do Alexandria

Yield and quality responses of Egyptian white garlic (Allium sativum L.) and wild garlic (Tulbaghia violacea Harv.) to nitrogen nutrition

Allium sativum and Tulbaghia violacea are some of the most important medicinal plants used by South African traditional healers for the treatment of flu, fever, cold, tuberculosis, asthma and many more diseases. However, growth, yield and quality are constrained by excessive and under fertilization. This study was carried out to determine, firstly, the effect of N source (ammonium sulphate and calcium nitrate) on yield and quality of A. sativum and T. violacea plants. Secondly, to determine the best season for harvesting T. violacea and lastly, to determine the antifungal effects of A. sativum and T. violacea plant extracts against plant pathogens Altenaria solani and Sclerotium rolfsii. Both plants were treated with both N sources applied as topdressing treatments at a total of 0, 50, 100, 150 and 200 kg.ha-1, divided into three applications at three week (A. sativum) and three month (T. violacea) intervals. A. sativum plants were sampled at 54, 82, 112, 140 and 175 days after planting (DAP) while, T. violacea plants were sampled monthly for ten months. Parameters recorded were growth analysis, yield and bioactivity for both plant species. Both nitrogen sources improved plant growth and yield of A. sativum and T. violacea plants. Calcium nitrate at 150 kg•ha-1 and ammonium sulphate at 200 kg•ha-1 produced the highest at 24 t•ha-1 and 27 t•ha-1, respectively. Ammonium sulphate improved bioactivity of leaves with the highest bioactivity recorded at 82 and 112 DAP. Yield obtained from the autumn harvest was not affected by N source. Ammonium sulphate and calcium nitrate at 200 kg•ha-1 produced the highest yields of 23.6 t•ha-1 and 23.5 t•ha-1, respectively. In contrast, yield obtained from the winter harvest was affected by N source at 200 kg•ha-1, with significantly better yield of 30.8 t•ha-1 with calcium nitrate compared to 27.4 t•ha-1 with ammonium sulphate. Crude extracts of T. violacea bulbs that were treated with ammonium sulphate significantly inhibited the growth of plant pathogenic fungi, whereas extracts from plants treated with calcium nitrate showed low bioactivity. Extracts from plants grown with ammonium sulphate at 100 kg•ha-1 were more effective in controlling growth of plant pathogens when compared to other N levels. The minimum inhibitory concentration (MIC) effects of A. sativum against S. rolfsii and A. solani were at 0.01 mg•mL-1. The MIC of T. violacea extracts against A. solani was at 0.006 mg•mL<Sup>-1. The MIC of T. violacea extracts were better than previously reported in literature. Epidemiology of early blight on potatoes in South Africa

Early blight (Alternaria solani Sorauer)is a major foliar disease of potatoes in most growing regions of the world and is underestimated in South Africa. This project studies the epidemiology and control of the disease in South Africa. A decision support system (DSS) for early blight in South Africa was developed and evaluated in field trials. This early blight DSS is the first such system to be developed in South Africa and once incorporated with the late blight model, will represent innovative technology for use in the South African potato industry. Trends in weather variables and concentrations of airborne conidia of A. solani were monitored. Distinct seasonal variation was noted. Peaks in spore concentration coincided with periods favourable for spore formation and dispersal; most notable was diurnal periodicity and interrupted wetting periods. The results obtained from these measurements will be useful in improving early blight DSSs for southern Africa. Isolates of A. solani from various potato-growing regions in South Africa were characterized using virulence, vegetative compatibility (VC) and random amplified microsatellite (RAMS) primers. Neither the virulence assays nor VC tests sufficiently characterised the population. Analysis of RAMS profiles revealed 27% genetic diversity among the isolates. This value is similar to diversity values obtained by previous authors studying A. solani, however, it is relatively high for an asexually reproducing fungus. There was no evidence for geographical clustering of isolates, indicating that isolates are widespread across South Africa. A survey on control practices and grower perceptions of early blight in South Africa was conducted using a questionnaire. These questionnaires were distributed to growers from 10 potato-growing regions in South Africa. Results highlighted the most popular control methods and cultivars in the South African potato industry. The majority of respondents indicated that they would use an accurate, cost-effective early blight DSS, and that more research is necessary on early blight in South Africa. A survey on control practices and grower perceptions of early blight in South Africa was conducted using a questionnaire. These questionnaires were distributed to growers from 10 potato-growing regions in South Africa. Results highlighted the most popular control methods and cultivars in the South African potato industry. The majority of respondents indicated that they would use an accurate, cost-effective early blight DSS, and that more research is necessary on early blight in South Africa. Estimated crop losses ranged from 1% - 60%, with an average of approximately 20%. Tomato yield and early blight incidence in response to cultivar and irrigation management practices in the Giyani Municipality

Thesis (M. Agricultural Management (Plant Production)) -- University of Limpopo, 2020 / Tomato is a major vegetable crop produced in Limpopo province by subsistence and commercial farmers. Biotic and abiotic factors such as diseases and drought affect its productivity and yield negatively. The most economically important tomato disease in Limpopo is early blight, caused by Alternaria solani. Early blight affects tomato growing under stressful conditions such as drought. The main aim of this study was to evaluate water management in tomato production and its effect on tomato yield, early blight occurrence and severity under smallholder farming systems in Giyani. A survey study and field experiments on early blight occurrence and severity were conducted in the Greater Giyani municipality in Limpopo province. Firstly, a survey of early blight was conducted in tomato fields selected from three service centres namely Guwela, Hlaneki and Mhlava Willem. Twenty five potential tomato grower’s farms were randomly selected for survey based on the location and production management strategies. A second study was carried out by establishing field experiments at two farms in the same area, namely: A hi tirheni Mqekwa and Duvadzi farms. The experiments were laid out in a randomized complete block design (RCBD) in a split plot arrangement, with irrigation as the main plot treatment and tomato variety as the subplot treatment. Each treatment was replicated four times. The irrigation treatments consisted of full irrigation up to field capacity and deficit irrigation. The tomato varieties planted were HTX14, Rodade, STAR9006 and Commander, and were irrigated with a drip irrigation system. The overall survey results showed that Guwela s/c had high disease incidence and the lowest disease severity as compared to the other two service centers. The lowest disease incidence was observed at Hlaneki s/c whilst Mhlava willem s/c had the highest disease severity. The two water treatments (full and deficit irrigation) in both farms have shown no significant pairwise differences among xx the volumetric water content means. There was no significant difference (P≥0.05) in early blight disease severity in both farms. Results shows that, all tomato varieties did not differ significantly (P≥0.05) in yield observed between treatments at Duvadzi and A hi tirheni Mqekwa Farm. Deficit irrigation can be used effectively to produce high yields in tomato production provided the cropping season window does not coincide with the hottest time of the year. Full and deficit irrigation did not have influence on the early blight disease incidence on tomato plants. Key words: Alternaria solani; disease incidence; disease severity and drip irrigation
