Takotsubo cardiomyopathy – an unexpected complication in spine surgery

Introduction: Takotsubo cardiomyopathy is an apical ballooning syndrome, which can be triggeredby stress. Only few case reports describe the onset of Takotsubo as a complication of neurosurgery procedures. Clinical presentation: A case of a 53 year-old female with a spinal neurinoma and surgery-associated Takotsubo cardiomyopathy is demonstrated. The patient developed typical signs of a myocardial infarction with circulation depression and ST elevation, but normal cardiac enzymes at the end of surgery. Cardiac catheterization and levocardiography confirmed the absence of any critical coronary disease but the presence of a typical apical ballooning and midventricular hypokinesis. The patient recovered completely under supportive conservative and cardiological therapy, showing regular left ventricular pumpfunction. Conclusion: Interventions in neurosurgery and perioperative care should be kept as stress free as possible. Due to the possibility of neurogenic mechanisms related to cardiomyopathy, Takotsubo cardiomyopathy as an entity of stress-induced complications should be taken into consideration.

Análise da atividade metabólica de bactérias persistentes após os procedimentos endodônticos de desinfecção: estudo molecular baseado em RNA e DNA / Metabolic activity analysis of persistent bacteria after endodontic disinfection procedures: RNA- and DNA-based molecular study

Micro-organismos persistentes que permanecem viáveis e em níveis elevados nos canais radiculares após os procedimentos endodônticos podem influenciar no sucesso do tratamento endodôntico. O objetivo deste estudo foi avaliar a quantidade e a atividade metabólica de bactérias persistentes utilizando métodos moleculares baseados em rDNA e rRNA. Foram selecionados 15 pacientes com infecções endodônticas primárias assintomáticas. Amostras microbiológicas foram coletadas dos canais radiculares após a cirurgia de acesso (S1), após o preparo químico-cirúrgico realizado com Sistema Reciproc e NaOCl 2.5% (S2) e após medicação intracanal com Ca(OH)2 por 14 dias (S3). As amostras dos canais radiculares foram submetidas à extração de DNA e RNA. O RNA foi submetido à reação de transcrição reversa para confecção de DNA complementar (cDNA). O efeito dos procedimentos endodônticos na redução bacteriana foi determinado por qPCR baseada em rDNA, utilizando iniciadores universais para a região 16S rRNA do domínio Bacteria e iniciadores espécie-específicos para Bacteroidaceae [G-1] sp oral taxon 272. A atividade metabólica de bactérias totais e Bacteroidaceae [G-1] sp oral taxon 272 foi calculada pela razão rRNA/rDNA baseados nos dados dos ensaios de qPCR. Os dados foram analisados pelo teste de Wilcoxon para análise quantitativa e teste de McNemar para comparação da taxa de detecção dos métodos, com nível de significância de 5%. Todas as amostras S1 foram positivas para bactérias totais, com uma mediana de 1,87 x 105 cópias de rDNA por amostra. Após o preparo químico-cirúrgico houve uma redução significativa de rDNA de bactérias totais (mediana 7,86 x 104, P = 0,01). Entretanto, não houve redução de rDNA bacteriano após a medicação intracanal quando comparada à etapa anterior (mediana 7,97 x 104, P > 0,05). Os resultados da razão rRNA/rDNA revelaram que houve uma redução do metabolismo de bactérias totais em S2 (mediana 1, P = 0,04) e um aumento do metabolismo bacteriano em S3 (mediana 2, P = 0,04). Na análise de Bacteroidaceae [G-1] sp oral taxon 272, o tratamento endodôntico não influenciou na redução nem no metabolismo bacteriano. Concluiu-se que o número total de bactérias foi drasticamente reduzido após o preparo químico-cirúrgico, assim como seu metabolismo. Entretanto, o metabolismo bacteriano aumentou após o uso da medicação intracanal com Ca(OH)2. Bacteroidaceae [G-1] sp. oral taxon 272 permaneceu metabolicamente ativo após o preparo químico-cirúrgico e medicação intracanal, participando, assim, da composição da microbiota persistente. / High levels of microorganisms that persist viable in root canals after endodontic procedures may influence endodontic treatment outcome. This study aimed to evaluate the amount and the metabolic activity of persistent bacteria using rRNA- and rDNA-based molecular methods. Fifteen patients with primary endodontic infections were selected. Microbiological samples were taken from root canals after access cavity (S1), after chemomechanical preparation with Reciproc System and 2.5% NaOCl (S2) and after intracanal medication with calcium hydroxide for 14 days (S3). DNA and RNA were extracted from root canal samples, and cDNA was synthetized using reverse transcription reaction. The effect of endodontic procedures on bacterial reduction was determined by rDNA-based qPCR using universal primers for 16S rRNA Bacteria domain and species-specific primers to Bacteroidaceae [G-1] sp oral taxon 272. The metabolic activity of total bacteria and Bacteroidaceae [G-1] sp oral taxon 272 was calculated by rRNA/ rDNA ratio estimated by qPCR. The data was analyzed by the Wilcoxon test for quantitative analysis and McNemar test for comparison of the detection rate of the methods, with 5% significance level. All S1 samples were positive for total bacteria, with a median value of 1.87 x 105 bacterial cells. After chemomechanical preparation a significant reduction of bacterial rDNA was observed (median 7.86 x 104, P = 0.01). Nevertheless, there was no bacterial rDNA reduction after intracanal medication when compared to S2 samples (median 7.97 x 104, P > 0.05). The rRNA/ rDNA ratio showed a reduction of total bacteria metabolism in S2 (median 1, P = 0,04), and an increase of bacterial metabolism in S3 (median 2, P = 0,04). Bacteroidaceae [G-1] sp oral taxon 272 analysis showed that endodontic treatment did not impact the amount and the metabolic activity of this taxon. In conclusion, the number and metabolism of total bacteria were severely reduced after chemomechanical preparation. On the other hand, the bacterial metabolism increased after intracanal dressing with Ca(OH)2. Bacteroidaceae [G-1] sp. oral taxon 272 remained metabolically active after chemomechanical preparation and intracanal dressing thus participating in the composition of the persistent microbiota.

Periodontite apical induzida em cães: efeito do tratamento endodôntico. Estudo microbiológico, tomográfico e de microscopia confocal

Este trabalho teve o objetivo de avaliar a capacidade da bactéria Enterococcus faecalis em induzir periodontite apical no modelo experimental canino; verificar se o preparo químico-mecânico afeta a sobrevivência dessa bactéria no sistema de canais radiculares e avaliar o reparo desses dentes por meio de radiografia periapical e tomografia computadorizada cone beam. Dentes com e sem tratamento endodôntico foram avaliados por meio de microscopia confocal de varredura a laser. Para tal, foram utilizados 2 cães. A bactéria Enterococcus faecalis foi inoculada nos canais de 4 pré-molares superiores, 11 prémolares inferiores e 9 incisivos superiores. As câmaras pulpares foram seladas e, após 60 dias, os canais das raízes distais dos pré-molares e de 7 incisivos superiores foram submetidos a tratamento endodôntico em sessão única e os canais das raízes mesiais dos pré-molares e 2 incisivos superiores foram deixados sem tratamento (controle). Amostras microbiológicas foram feitas antes e após o preparo químico-mecânico. O reparo foi avaliado após 6 meses do tratamento mediante radiografias periapicais e por tomografia computadorizada cone beam. A comparação entre as imagens obtidas após o período experimental pelos 2 métodos foi feita por medições da área em mm2 de cada lesão encontrada, utilizando o software ImageTool Os resultados mostraram que a presença de periodontite apical crônica foi verificada em todos os dentes inoculados, independentemente da colonização pela bactéria Enterococcus faecalis ou pela flora mista. O preparo químico-mecânico reduziu significativamente o número de bactérias no interior dos canais radiculares (p<0.05). Os resultados radiográficos e tomográficos demonstraram lesões de menor diâmetro nos dentes tratados endodonticamente em comparação ao grupo controle (p<0.05). A comparação entre os métodos demonstrou diferença estatística entre eles sendo evidenciada áreas radiolúcidas maiores utilizando a tomografia cone beam em comparação com a radiografia periapical (p<0.05). Concluiu-se que a bactéria Enterococcus faecalis induziu periodontite apical crônica similarmente à flora mista; o tratamento endodôntico reduziu o número de bactérias cultiváveis significativamente embora sem relação com o reparo radiográfico. Os dentes tratados em sessão única apresentavam lesões menores em comparação aos não-tratados e as áreas das lesões observadas na TC cone beam foram maiores do que as áreas encontradas nas radiografias periapicais. A microscopia confocal e o método proposto neste estudo se mostraram eficazes para determinar, qualitativamente, a viabilidade bacteriana e a distribuição de ácidos nucléicos bacterianos dentro dos túbulos dentinários. Houve diferença entre a dentina infectada in vitro com o padrão de infecção in vivo, caracterizada pela presença de biofilmes aderidos à parede do canal radicular. / The aims of this study were to evaluate the Enterococcus faecalis ability to induce apical periodontitis in dogs root canals, to verify the bacterias ability to survive to the cleaning and shaping procedures and to assess the healing of the induced apical periodontitis by periapical radiograph and cone beam computed tomography. Also, endodontically treated and non-treated teeth were evaluated by confocal laser scanning microscope. Two mongrel dogs were used in the experiment. Enterococcus faecalis strain was inoculated into the root canals of 4 maxillary premolars, 11 mandibular premolars and 9 maxillary incisors. After 60 days the root canals of the distal roots of the mandibular and maxillary pre-molars and 7 maxillary incisors were endodontically treated. The premolars mesial root canal and 2 maxillary incisors were used as control (no treatment). Microbiologic samples were done after and before the cleaning and shaping procedure. The healing was evaluated after 6 months by periapical radiographs and cone beam computed tomography. The comparison between obtained images after the experimental period by the two methods was done using measures of the lesion area in mm2 with ImageTool software. The results showed the presence of chronic apical periodontitis in every inoculated teeth, with Enterococcus faecalis or mixed infection. The cleaning and shaping procedures reduced the number of bacteria of the root canals (p<0.05). The radiographic and tomographic results showed the lower diameter lesion in endodontically treated teeth than in the control group (p<0.05). The comparison between the methods showed statistical difference and greater radiolucent areas were evident in cone beam computed tomography images (p<0.05). It is possible to conclude that the bacteria Enterococcus faecalis induced chronic apical periodontitis as well as the mixed microflora; the endodontic treatment reduced the number of cultivable bacteria in a significant way, but with no relation to the radiographic healing. The treated teeth in only one session presented smaller lesions in comparison to the non-treated teeth and the lesions area in cone beam computed tomography were bigger than the areas found in periapical radiograph. The confocal microscopy and the proposed method of this study showed to be efficient to determine the bacterial ability and the distribution of bacterial nucleic acids inside the dentin tubules. There was difference between in vitro infected dentin with the in vivo infection pattern, which presents biofilm attached to the root canal walls.

O tratamento endodôntico é constituído por uma sequência de fases que devem ser respeitadas e conduzidas com o mesmo grau de importância para o sucesso da terapia endodôntica. Atenção especial é dada a fase do o preparo biomecânico do canal radicular, que pode ocasionar extrusão de restos pulpares, tecido necrosado, rapas de dentina e substâncias químicas auxiliares através do forame apical, provocando uma reação inflamatória ou flare-up. Com o intuito de se obter um pós-operatório com êxito, realizou-se um estudo com o objetivo de avaliar, ex vivo, a quantidade de material extruído através do forame apical durante o preparo do canal radicular pelas seguintes técnicas: manual Step-Back, mecanizada rotatórias Crown Down e Free Tip Preparation. Trinta incisivos inferiores foram distribuídos aleatoriamente em 3 grupos (n=10), de acordo com as seguintes técnicas de instrumentação: G1- técnica manual Step Back (controle); G2- Crown Down; G3- Free Tip Preparation. Todos os grupos foram instrumentados a 0,5mm aquém do forame apical. O material extruído foi coletado em um Dispositivo Coletor de Material Extruído(DCME), confeccionado especificamente para essa finalidade e a extrusão apical foi calculada pela determinação da massa de material sólido extruído. Os valores obtidos foram analisados pelo teste de Kruskal- Wallis e o teste complementar de Dunn`s Multiple, com P=0.05, como nível de significância estatística. Os resultados mostraram que a técnica Step-Back promoveu maior quantidade de material extruído através do forame apical; e que não houve diferença estatística significante entre as técnicas Crown Down e Free Tip Preparation. Pode-se concluir também que houve extrusão de material sólido através do forame apical em todas as técnicas de instrumentação estudadas. / Endodontic treatment is constituted of a sequence of stages that must be respected and performed with the same degree of importance in order to ensure the success of endodontic therapy. Special attention is paid to the stage of biomechanical preparation of the root canal, which may cause the extrusion of pulp remainders, necrotic tissue, dentin scrapings and auxiliary chemical substances through the apical foramen, causing an inflammatory reaction or flare-up. With the intention of achieving successful post-operative period, a study was conducted with the aim of making an ex vivo evaluation of the quantity of material extruded through the apical foramen during root canal preparation, by means of the following techniques: manual Step-Back, mechanized rotary Crown Down and Free Tip Preparation. Afterwards, the teeth were randomly assigned to three equal groups (n=15) according to post length, as follows: G1 manual Step-Back technique (control); G2 - Crown Down; G3 - Free Tip Preparation. All the groups were instrumented to 0.5 mm short of the apical foramen. The extruded was collected in an Extruded Material Collecting Device (EMCD), made specifically for this purpose, and apical extrusion was calculated by determining the mass of solid material extruded. The values obtained were analyzed by the Kruskal-Wallis test and complementary Dunns Multiple test, with P=0.05, as the level of statistical significance. The results showed that the Step-Back technique promoted the largest amount of material extruded through the apical foramen; and that there was no statistically significant difference between the Crown Down and Free Tip Preparations. It could also be concluded that there was extrusion of solid material through the apical foramen in all the studied instrumentation techniques used.

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Eugenia involucrata DC. and Handroanthus chrysotrichus (Mart. ex DC) Mattos are native species with economic, ecological and silvicultural. They have problems with their spread via seeds, because they are recalcitrant and lose viability within a few weeks after collection. In addition, there is little information about the propagation of these species. Therefore, the general objective of this study was to evaluate methodologies which may contribute to micropropagation of E. involucrata and H. chrysotrichus. For multiplication of E. involucrata were tested different sources (BAP, KIN, 2iP and TDZ) and concentrations (0, 16 and 32 μM) of cytokinins added to the nutrient medium MS/2. Also, we tested the effect of TDZ (16 or 32 μM) combined with GA3 (0; 10; 20 and 40 μM) added in medium MS/2, on in vitro multiplication of nodal segments. Even for E. involucrata, we examined the effect of NAA (0; 0,5 and 1 μM) combined with TDZ (0; 16 and 32 μM), added to the medium MS/2. Finally, for E. involucrata, we tested the effect of different concentrations of TDZ (0; 16 and 32 μM) added to the nutrient medium MS/2, on the multiplication of shoot apices of seedlings germinated in vitro. For H. chrysotrichus were tested different types of explants (shoot apical segments and epicotyl) and nutrient media (WPM, MS, WPM/2 and MS/2) in the presence and absence of activated charcoal as well as the effect of BAP (0; 2; 4; 8 and 16 μM) on in vitro multiplication of this species. For in vitro multiplication of nodal segments of E. involucrata the cytokinins tested are dispensable; cytokinins favor the growth of bacterial contamination, high levels of contamination reduce in vitro survival and establishment of nodal segments; the in vitro establishment for this species is impaired by the occurrence of leaf chlorosis and swelling of the explants. As for H. chrysotrichus, the shoot apical segments show the highest development in the in vitro survival and establishment than of epicotyl ; there is a high in vitro swelling of the explants in the absence of growth regulators; in the in vitro establishment in medium WPM/2 in the presence of 0,05 μM shoot apical segments are more efficient under low concentrations of BAP; there isgreater calli formation in epicotyl with increase the concentration of BAP, the opposite was true for the shoot apical segment. / Eugenia involucrata DC. e Handroanthus chrysotrichus (Mart. ex DC) Mattos são espécies florestais nativas com importância econômica, silvicultural e ecológica. Possuem problemas quanto a sua propagação via sementes, pois estas são recalcitrantes, perdendo a viabilidade em poucas semanas após a coleta. Além disso, existem poucas informações sobre a propagação vegetativa dessas espécies. Considerado o exposto, o objetivo geral do presente estudo foi avaliar metodologias que possam contribuir para a micropropagação in vitro de E. involucrata e de H. chrysotrichus. Para a multiplicação de E. involucrata foram testadas diferentes fontes (BAP, CIN, TDZ e 2iP) e concentrações (0; 16 e 32 μM) de citocininas adicionadas ao meio nutritivo MS/2. Também foi testado o efeito de TDZ (16 e 32 μM) combinado a GA3 (0; 10; 20 e 40 μM), acrescentados em meio MS/2, na multiplicação in vitro de segmentos nodais. Ainda para E. involucrata, foi avaliado o efeito de ANA (0; 0,5 e 1 μM) combinado a TDZ (0; 16 e 32 μM), acrescidos ao meio MS/2. Por fim, para esta espécie, testou-se o efeito de diferentes concentrações de TDZ (0; 16 e 32 μM) adicionadas ao meio MS/2, na multiplicação de segmentos apicais caulinares obtidos de plântulas germinadas in vitro. Para H. chrysotrichus, foram testados diferentes tipos de explantes (segmento apical caulinar e epicótilo) e meios nutritivos (WPM, MS, WPM/2 e MS/2), na presença e ausência de carvão ativado, bem como o efeito de BAP (0; 2; 4; 8 e 16 μM) na multiplicação in vitro dessa espécie. Para multiplicação in vitro de segmentos nodais de E. involucrata, as citocininas testadas são dispensáveis, uma vez que favorecem o crescimento de contaminação bacteriana. Elevados índices de contaminação acarretam reduzida sobrevivência e estabelecimento in vitro de segmentos nodais; o estabelecimento in vitro para essa espécie é comprometido pela ocorrência de clorose foliar e intumescimento dos explantes. Já para H. chrysotrichus, o segmento apical caulinar apresenta desenvolvimento superior ao do epicótilo na sobrevivência e no estabelecimento in vitro; há elevado intumescimento in vitro dos explantes na ausência de fitorreguladores. No estabelecimento in vitro em meio WPM/2 na presença de 0,05 μM de ANA, os segmentos apicais caulinares são mais eficientes sob concentrações reduzidas de BAP; há maior calogênese em epicótilos à medida que aumenta a concentração de BAP, ocorrendo o contrário para o segmento apical caulinar.

Effect of cultural practices and selected chemicals on flowering and fruit production in some mango (Mangifera indica L.) cultivars

Although mango (Mangifera indica L.) has been studied for many years, numerous problems still elude researchers. The objectives of the current trials were to study the effects of some cultural practices (fruit thinning, panicle/ bud/ renewal/ post-harvest pruning) and chemicals (Corasil.E, potassium nitrate/urea, paclobutrazol) on various vegetative, floral, yield and quality parameters. The study meant to address problems of both South African and Ethiopian mango growers. The thinning (on ‘Sensation’) and pruning (on ‘Tommy Atkins’ and ‘Keitt’) experiments were conducted for two seasons (2001-2003) at Bavaria Fruit Estate in South Africa. Effects of Paclobutrazol and Potassium nitrate were studied on ‘Tommy Atkins’ during 2002-2003 season at Upper Awash Agro-industry Enterprise in Ethiopia. Complementary effects of paclobutrazol and potassium nitrate on floral induction were studied in growth chamber experiments at the experimental farm of University of Pretoria on ‘Tommy Atkins’ and ‘Keitt’ mango. Where fruit on ‘Sensation’ were thinned to one and two fruit per panicle, a significant increase was obtained for most of the fruit quantitative parameters. The treatments where one fruit and two fruit per panicle were retained and 50% panicles removed, produced a significant increase in size of the fruit, fruit qualitative parameters and fruit retention percentage. Corasil.E produced very small sized fruit with a considerable percentage of “mules” (fruit without seed). Trees subjected to severe thinning intensities showed earlier revival of starch reserves and better vegetative growth. Applications of paclobutrazol (1- (4-chlorophenyl) –4,4-dimethyl-2- (1,2,4- triazol-1-yl) pentan-3-ol) at rates of 5.50 and 8.25 g a.i. per tree, both as a soil drench and spray applications, on ‘Tommy Atkins’ mango were effective in suppressing vegetative growth as compared to the control. Consequently, the trees from these treatments had higher total non-structural carbohydrate in their shoots before flowering which led to higher results of percentages of shoots flowering, number of panicles produced, percentages of hermaphrodite flowers, yield and quality of the fruit. Trees that received panicle pruning (during full bloom) treatment at the point of apical bud attachment, were observed to be induced for synchronized re-flowering and also attained more fruit per panicle. On the other hand, trees on which renewal pruning (early in the season) and post-harvest pruning (especially for early cultivars) treatments were applied, have been observed for the development of an adequate number of productive inflorescences. Post-harvest pruning treatments also resulted in greater vegetative growth on both cultivars. The responses to pruning treatments were greater especially in ‘Tommy Atkins’ than ‘Keitt’. The trend for the interaction of duration and chemicals in Tommy Atkins and Keitt mango cultivars revealed the possible floral induction complementary effect of PBZ after the trees were induced only for 15 days at 10/15 oC temperature. Higher potassium nitrate concentrations especially in combination with urea (5 litre solution of 4% KNO3+0.5 g urea tree-1 and 5 litres of 4% KNO3+1 g urea tree-1 ) produced higher results for most of the flowering and yield parameters in ‘Tommy Atkins’. / Thesis (DPhil (Horticulture))--University of Pretoria, 2006. / Plant Production and Soil Science / unrestricted

Caractérisation du lien entre croissance et patterning dans la morphogenèse chez Arabidopsis / Linking patterning to growth changes during morphogenesis in Arabidopsis shoot meristem

Le contrôle moléculaire du patterning au cours des processus développementaux est aujourd’hui bien décrit chez les organismes multicellulaires. A l’inverse, la contribution de la croissance dans l’émergence des patterns reste peu explorée, et est souvent réduite à un rôle passif. Au cours de cette thèse, j’ai étudié cette question en utilisant le méristème apical caulinaire (MAC) d’Arabidopsis comme modèle. Le méristème est un groupe de cellules en divisions situé à l’extrémité de toutes les tiges et les branches et qui génère tous les organes aériens de la plante selon un patron stéréotypé, aussi appelé phyllotaxie. Dans une première partie, j’ai étudié comment la croissance de la tige pouvait influencer le patron phyllotactique. Plus précisément, en découplant dépôt de la cellulose dans la paroi et l’orientation des microtubules, j’ai montré que le patron de phyllotaxie devenait bimodal en raison de l’induction d’une torsion lors de la croissance de la tige. Dans une seconde partie, j’ai analysé le lien entre forme du MAC et expression génétique. En particulier, j’ai pu corréler l’expression d’un gène maître : SHOOTMERISTEM LESS (STM) au degré de courbure dans le MAC. De plus, en utilisant des approches de micromécaniques, j’ai aussi pu montrer que l’expression de STM pouvait être induite par le patron de contraintes localement généré par la courbure. Pour finir, j’ai aussi étudié comment la taille du méristème influence la robustesse du pattern de phyllotaxie sur la tige en modulant la fréquence d’initiation des organes. L’ensemble de ce travail met ainsi en avant le rôle de la croissance dans le patterning, notamment via des mécanismes de rétrocontrôles géométriques et mécaniques. / The molecular mechanisms behind the emergence of patterns during developmental processes have been well described in multicellular organisms. However, the contribution of growth in patterning is still poorly understood; growth is often seen as a passive output of the activity of the patterning signals. In this PhD, I have studied the relation between growth and patterning using the shoot apical meristem of Arabidopsis as a model system. The meristem is a group of dividing cells located at the tip of every stems and branches that generates all the aerial organs of the plant following a typical spatio-temporal pattern also called phyllotaxis. In a first part, the influence of post-meristematic growth on phyllotaxis was assessed. More precisely, by uncoupling cellulose deposition from the orientation of the microtubule array, I showed that the resulting stem torsion induces the emergence of a new and robust bimodal phyllotactic pattern. In a second part, the relation between meristem shape and gene expression was analyzed. More precisely, I correlated the expression of a master regulatory gene: SHOOT MERISTEMLESS (STM) to tissue curvature in the boundary domain that separates the emerging organ from the meristem. Furthermore, I showed that STM expression can be induced by micromechanical perturbations thus suggesting that shape-derived mechanical stresses in the meristem boundary contribute to STM expression. Finally, I also studied how meristem size can influence the robustness of the pattern of phyllotaxis along the stem through a modulation of the frequency of organ initiation. Altogether, this work highlights the important contribution of growth in patterning, notably thanks to the existence of geometrical and mechanical feedbacks.

Constrição celular apical durante a invaginação do placóide do cristalino em galinhas. / Apical cell constriction during chicken lens placode invagination.

O cristalino de vertebrados se origina a partir da invaginação do ectoderme que recobre a vesícula óptica. A invaginação epitelial em diversos modelos é causada pela constrição celular apical, mediada pela contração apical de actina e miosina II e regulada pela GTPase RhoA. Neste trabalho nós investigamos se a invaginação do cristalino em embriões de galinha ocorre devido à constrição celular apical e se este evento é controlado por RhoA. Actina filamentosa e miosina II são expressas na porção apical do cristalino durante a invaginação. Quando a polimerização de actina é inibida por Citocalasina D, o cristalino não invagina, sugerindo que a constrição celular apical poderia contribuir para a invaginação do cristalino. RhoA também é expressa durante o desenvolvimento do cristalino, mas a inibição de RhoA, por eletroporação da forma dominante-negativo, não impediu a invaginação do placóide do cristalino, não alterou a distribuição de miosina II na porção apical do cristalino nem sua ativação, indicando que a invaginação do cristalino independe de RhoA. / Vertebrate lens derives from invagination of the ectoderm that overlies optic vesicles. Epithelial invagination in many model systems is driven by apical cell constriction, mediated by actin and myosin II contraction regulated by GTPase RhoA. Here we investigate the possibility that chick lens placode invagination could also be driven by apical cell constriction and controlled by RhoA. We show that actin and myosin II are expressed at lens apical side during lens invagination. Actin polymerization inhibition by in ovo Cytochalasin D treatment prevents lens placode invagination, suggesting that lens placode invagination could be driven by apical cell constriction. RhoA GTPase is also expressed at apical portion of lens placode and during lens invagination. However, when we overexpressed by electroporation the dominant-negative RhoA in the pre-lens ectoderm invagination was not affected. Furthermore, dominant-negative RhoA didnt affect myosin II apical localization nor myosin II phosphorilation, indicating that in lens invagination this process is not regulated by GTPase RhoA.

Transport d’iode par le transporteur de sodium/acide monocarboxylique SMCT1

Le transporteur de Na+/ acide monocarboxylique sensible à l’ibuprofène (SMCT1) est exprimé dans la membrane apicale de plusieurs épithélia. Son rôle physiologique dans la glande thyroïde reste cependant obscur mais on présume qu’il pourrait agir comme un transporteur apical d’iode nécessaire pour la synthèse des hormones thyroïdiennes. Récemment, on a montré que SMCT1 possède un courant de fuite anionique sensible à [Na+]e qui permettrait de transporter l’iode de façon électrogénique. Cependant, un efflux d’iode sensible à l’ibuprofène, mais indépendant de la [Na+]e a été aussi observé sur des cultures primaires des thyrocytes porcins, suggérant un autre mécanisme de transport d’iode par SMCT1. Ce travail vise à comprendre les caractéristiques de ce genre de transport en utilisant comme modèle d’expression les ovocytes de Xenopus laevis. Les résultats obtenus des essais de captation d’iode radioactif montrent que SMCT1 présente un transport d’iode sensible à l’ibuprofène de l’ordre de 30nmol/ovocyte/h. Si ce transport est non saturable en iode (0-100 mM), il nécessite du Na+ dans la solution externe. En effet, le remplacement du Na+ extracellulaire par le NMDG inhibe complètement le transport. En outre, on s’est intéressé à exclure la possibilité de différents artefacts. En ayant trouvé que la grande majorité de l’iode radioactif se trouve dans la partie soluble de l’ovocyte, on exclut une liaison non spécifique de l’iode à la membrane cellulaire. Cependant, une bonne proportion de l’iode transporté pourrait être liée à des protéines à l’intérieur de l`ovocyte. En effet, on observe une réduction du transport d’iode dans les ovocytes exprimant SMCT1 de 81,6 ± 2 % en présence de 2 % BSA dans la solution extracellulaire. Également, on écarte la possibilité que le transport d’iode soit le résultat de la surexpression de protéines de transport endogènes dont les canaux chlore. Le transport d’iode semble spécifique à l’expression de SMCT1 et de manière intéressante à l’expression d’un autre transporteur de monocarboxylates, MCT1. L’analyse de l’ensemble des essais, y compris le fait que l’amplitude du transport observé est 20 fois plus grande que celle du courant de fuite nous mène à proposer que SMCT1 puisse transporter l’iode de façon électroneutre. Cependant, le mécanisme par lequel ceci est accompli n’est pas évident à identifier. L’utilisation d’un autre modèle cellulaire serait surement utile pour répondre à cette question. / Ibuprofen sensitive, Sodium Monocarboxylate Transporter (SMCT1) is expressed in the apical membrane of diverse epithelia. Its physiological role in the thyroid remains however unknown, but it has been proposed that SMCT1 could act as an apical iodide transporter required for the main function of the gland: the thyroid hormone synthesis. We previously reported that SMCT1 exhibit a [Na+]e sensible anionic leak current that could account for the electrogenic transport of iodide. However, an iodine efflux sensitive to ibuprofen but independent of [Na+]e, was also observed in primary cultures of porcine thyrocytes, suggesting another mechanism of iodine transport mediated by SMCT1. This work aims to understand the characteristics of this type of transport using Xenopus laevis oocytes as an SMCT1 expression system. By realising 125I uptakes, we found that SMCT1 transports iodide in an ibuprofen sensitive manner (30nmol/oocyte/h). While nonsaturable uptake iodide kinetics were observed, SMCT1 iodide transport was Na+ dependent as shown by the transport reduction when the [Na+]e is replaced by NMDG. The possibility of artifacts, such as non specific binding and the overexpression of endogenous proteins, was analysed. By observing that the vast majority of the radioactive iodide is found in the soluble portion of the oocyte, we excluded non-specific binding of iodide to the cell membrane. However, it is believed that most of the iodide entering the cell is not free and must be bound to some intracellular proteins. Indeed, there is a significant reduction of SMCT1-mediated iodide transport when 2% BSA is present at the extracellular solution. Furthermore, the lack of iodide transport when overexpressing other proteins than SMCT1, precludes the possibility of an overexpression of endogenous transport proteins like chloride channels for example. In fact, the transport of iodide appears to be specific to the expression of SMCT1 and interestingly of another monocarboxylate transporter MCT1. The analysis of all trials, including the fact that the amplitude of the observed transport is 20 times larger than the leak current lead us to propose that SMCT1 can carry iodide in an electroneutral manner. However, the mechanism by which this is accomplished is not easy to identify and future experiments will be necessary to determine whether this transport is observed in other SMCT1 expression systems.

The Effect of Chlorhexidine Gluconate as an Endodontic Irrigant on the Apical Seal: Long-term Results

The purpose of this study was to determine whether chlorhexidine gluconate (0.12%), used as an endodontic irrigating solution would affect the apical seal of three root canal cements. One hundred extracted human single-canal teeth were divided into 9 experimental groups of 10 teeth each, in addition to a positive and negative control group of 5 teeth each. The teeth were decoronated at the level of the cementoenamel junction, accessed, instrumented to a Master Apical File #50, irrigated with either sterile saline, 5.25% NaOCl or 0.12% chlorhexidine gluconate, and dried using paper points. Obturation was accomplished using lateral condensation and one of three endodontic sealers: Roth's 811, AH26, or Sealapex. Post-obturation apical leakage was measured at 270- and 360-day observation periods using the fluid filtration method. Using the mixed-model repeated-measures ANOVA test with Tukey's HSD multiple comparison procedure, the results showed the saline-Sealapex combination had significantly more leakage (p<0.05) than either the Peridex-Sealapex or saline-Roth's combinations at 270 days. No other significant differences were noted between any sealer-irrigant combination at 270 days. The saline-Sealapex combination had significantly more leakage than the saline-Roth's combination at 360 days. No other significant differences were noted at 360 days. Under the conditions of this study, chlorhexidine gluconate irrigant did not adversely affect the apical seal of three root canal cements at 270 and 360 days.
