Previous issue date: 2016 / This work aims to analyze the sectorial index of BMF &Bovespa in an unprecedented
way analysing the chances of integration and financial contagion using the
methodology of trends and common cycles; correlating them, if possible, with the
facts and economic theories. The co-integration test and the common cycles test
confirm the existence of an equilibrium relation among the variables in both long and
short term. The existence of these relation supports the application of Granger
causality test to identify which variables can react with each other and in which
directions it occurs. This information is particularly important in stock markets
analysis when mounting strategies for asset allocation. Tests results enabled the
theoretical correlation between statistics, facts and economic theory. / Esse trabalho visa analisar os índices setoriais da BMF&Bovespa de uma forma
inédita ao analisar as hipóteses de integração e de contágio financeiros utilizando a
metodologia de tendências e ciclos comuns, correlacionando-as, se possível, com os
fatos e teorias econômicas. O teste de co-integração e o teste de ciclos comuns
confirmam a existência da relação de equilíbrio entre as variáveis tanto no longo
como no curto prazo. A existência dessas relações corrobora a aplicação do teste de
causalidade de Granger que identifica quais variáveis podem reagir entre si e em
que direções isso ocorre. Esta informação é particularmente importante na análise
de mercados de ações quando se montam estratégias para a alocação de ativos. Os
resultados dos testes possibilitam a correlação teórica entre os resultados
estatísticos, os fatos e a teoria econômica.

The cJUN NH2-terminal kinase pathway in mammary gland biology and carcinogenesisGirnius, Nomeda A. 08 March 2018 (has links)
The cJUN NH2-terminal kinase (JNK) pathway responds to environmental stresses and participates in many cellular processes, including cell death, survival, proliferation, migration, and genome maintenance. Importantly, genes that encode components of the JNK signaling pathway are frequently mutated in human breast cancer, but the functional consequence of these mutations in mammary carcinogenesis is unclear.
Anoikis – suspension-induced apoptosis – has been implicated in oncogenic transformation and tumor cell metastasis. Anoikis also contributes to lumen formation during mammary gland development and epithelial cell clearance during post-lactational involution. JNK is known to contribute to certain forms of cell death, but the role of JNK during anoikis was unclear. I examined the requirement of JNK in anoikis and discovered that JNK promotes cell death by transcriptional and post-translational regulation of pro-apoptotic BH3-only proteins. This conclusion suggested that JNK signaling may contribute to mammary gland remodeling during involution. Indeed, JNK deficiency in mammary epithelial cells disrupted the remodeling program of gene expression and delayed involution. Finally, I sought to understand the importance of JNK in mammary carcinogenesis. I found that JNK loss in the mammary epithelium was sufficient for genomic instability and tumor formation. Moreover, JNK loss in a model of breast cancer resulted in significantly accelerated tumor development. Collectively, these studies advance our understanding of the JNK pathway and breast biology, and provide insight that informs the design of therapeutic approaches that target the JNK signal transduction pathway.

Physico-chemical characteristics and quantitative structure-activity relationships of PCBsAndersson, Patrik January 2000 (has links)
<p>The polychlorinated biphenyls (PCBs) comprise a group of 209 congeners varying in the number of chlorine atoms and substitution patterns. These compounds tend to be biomagnified in foodwebs and have been shown to induce an array of effects in exposed organisms. The structural characteristics of the PCBs influence their potency as well as mechanism of action. In order to assess the biological potency of these compounds a multi-step quantitative structure-activity relationship (QSAR) procedure was used in the project described in this thesis.</p><p>The ultraviolet absorption (UV) spectra were measured for all 209 PCBs, and digitised for use as physico-chemical descriptors. Interpretations of the spectra using principal component analysis (PCA) showed the number of ortho chlorine atoms and para-para substitution patterns to be significant. Additional physico-chemical descriptors were derived from semi-empirical calculations. These included various molecular energies, the ionisation potential, electron affinity, dipole moments, and the internal barrier of rotation. The internal barrier of rotation was especially useful for describing the conformation of the PCBs on a continuous scale.</p><p>In total 52 physico-chemical descriptors were compiled and analysed by PCA for the tetra- to hepta-chlorinated congeners. The structural variation within these compounds was condensed into four principal properties derived from a PCA for use as design variables in a statistical design to select congeners representative for these homologue-groups. The 20 selected PCBs have been applied to study structure-specific biochemical responses in a number of bioassays, and to study the biomagnification of the PCBs in various fish species.</p><p>QSARs were established using partial least squares projections to latent structures (PLS) for the PCBs potency to inhibit intercellular communication, activate respiratory burst, inhibit dopamine uptake in synaptic vesicles, compete with estradiol for binding to estrogen receptors, and induce cytochrome P4501A (CYP1A) related activities. By the systematic use of the designed set of PCBs the biological potency was screened over the chemical domain of the class of compounds. Further, sub-regions of highly potent PCBs were identified for each response measured. For risk assessment of the PCBs potency to induce dioxin-like activities the predicted induction potencies (PIPs) were calculated. In addition, two sets of PCBs were presented that specifically represent congeners of environmental relevance in combination with predicted potency to induce estrogenic and CYP1A related activities.</p>

Previous issue date: 2011-05-31 / The closing price of a stock is the most important reference for the fair value of the company. Given its broad use in several contracts and in situations of asymmetrical information, brokers, traders and fund managers alike have incentives to manipulate it. This essay studies the intraday records of all transactions in the BMF&BOVESPA stock exchange between april 4th 2006 and march 31st 2011, and finds evidence of distortions in both volume and volatility at the end of the day, as well as price reversals in the next morning. It is also shown that the magnitude of the distortions are in line with such incentives. / O preço de fechamento de uma ação é a referência mais importante de seu valor. Dada sua ampla utilização em diversos contratos e em problemas de assimetria de informação, corretores, traders e gestores de fundos têm incentivos a interferir em sua formação. Este estudo analisa o registro intradiário de todas as transações de ações na BMF&BOVESPA entre 03 de abril de 2006 e 31 de março 2011, e econtra no fechamento um volume atípicamente alto, acompanhado de retornos muito voláteis que são em grande parte revertidos no dia seguinte. Adicionalmente, mostra-se que a magnitude destes fenômenos estão em linha com os incentivos para a distorção.

Bedeutung des p53-Signalwegs für Apoptoseaktivierung und Zellzyklusarrestregulation durch das p14 ARF TumorsuppressorgenOverkamp, Tim 08 November 2012 (has links)
BH3-only Proteine, eine pro-apoptotische Untergruppe der Bcl-2 Proteinfamilie, sind zentrale Mediatoren von apoptotischen Signalen durch die Regulierung intrinsischer Apoptose-signalwege. Unsere Arbeitsgruppe hat vor kurzem gezeigt, dass Apoptose, die durch den p14ARF Tumorsuppressor induziert wird über die p53-abhängige Aktivierung des BH3-only Proteins Puma/Bbc3 vermittelt wird. Interessanterweise induziert p14ARF aber auch in p53 defizienten Zellen Zellzyklusarrest und Apoptose. Die dahinterliegenden Signalwege sind jedoch nicht bekannt. In dieser Arbeit berichten wir, dass das BH3-only Protein Bmf (Bcl-2 modifying factor) beim p14ARF-induzierten Zelltod in p53 defizienten Zellen eine wichtige Rolle spielt. Expression von p14ARF führt zu einer Induktion der PERK Kinase, daran anschließender Phosphorylierung von eIF2α sowie Aktivierung der stromabwärts liegenden Transkriptionsfaktoren ATF4 und CHOP. Diese Signalkaskade ist normalerweise Teil einer zellulären Antwort auf fehl- oder ungefaltete Proteine im Endoplasmatischen Retikulum (ER), der sogenannten ‘unfolded protein response’ (UPR), die zum einen durch verminderte Translationsinitiation und Hochregulierung von Chaperonen die Menge der fehlgefalteten Proteine reduzieren soll. Allerdings induziert p14ARF keinen ER Stress, sondern den PERK‒CHOP Signalweg. Die Transkriptionsfaktoren ATF4 und CHOP binden direkt in der Promotorregion von bmf und sind für dessen transkriptionelle Regulation verantwortlich. Unsere Daten zeigen, dass der PERK‒eIF2α‒ATF4‒CHOP Signalweg eine wesentliche Rolle bei der Induktion von Apoptose durch p14ARF spielt. Dieser Weg könnte ein Sicherungsmechanismus sein, der es den Zellen auch nach Verlust von p53 erlaubt Apoptose einzuleiten, nachdem p14ARF durch Onkogene hochreguliert wurde. / BH3-only proteins, a pro-apoptotic subgroup of the Bcl-2 family of proteins, are central mediators of apoptosis signals by regulating the intrinsic apoptosis pathway. We have recently shown, that apoptosis triggered by the p14ARF tumour suppressor protein is mediated by the p53-dependent activation of the BH3-only protein Puma/Bbc3. Nevertheless, expression of p14ARF in p53-family deficient cells is capable of inducing both cell cycle arrest and apoptosis, but the signalling pathways initiated remain elusive. Here, we report that the BH3-only protein Bmf (Bcl-2 modifying factor) is involved in cell death in p53-deficient cells triggered by p14ARF. Expression of p14ARF leads to the induction of the PERK kinase, subsequent phosphorylation of eIF2α and activation of transcription factors ATF4 and CHOP. This signalling cascade is usually part of the ‘unfolded protein response’ (UPR), which is activated upon ER stress to reduce the amount of misfolded proteins by reduction of global protein translation initiation and upregulation of chaperones. Of note, p14ARF does not induce ER stress but activates the PERK‒CHOP pathway. ATF4 and CHOP transcription factors directly bind to the promotor region of bmf and induce its transcription. These data suggest that the PERK‒eIF2α‒ATF4‒CHOP signalling pathway may play a substantial role in mediating p14ARF-triggered apoptosis. This pathway could play the role of a ‘fail-safe’ mechanism that allows cells, even after loss of p53, to undergo apoptosis induced by upregulation of p14ARF by oncogenes.

Études in vivo et in vitro sur le potentiel néphroprotecteur de plantes médicinales anti-diabétiques de la pharmacopée traditionnelle des Cris de la Baie-JamesLi, Shilin 08 1900 (has links)
Notre équipe a identifié le thé Labrador [Rhododendron groenlandicum L. (Ericaceae)] comme une plante potentiellement antidiabétique de la pharmacopée traditionnelle des Cris de la Baie James orientale. Dans la présente étude, nous avons évalué les effets néphroprotecteurs potentiels de la plante.
De la microalbuminurie et de la fibrose rénale ont été développées chez des souris alimentées avec une diète grasse (DG). Le R. groenlandicum améliore d’une façon non-significative la microalbuminurie, avec des valeurs de l’aire sous la courbe (ACR) diminuant de 0.69 à 0.53. La valeur de la fibrose rénale qui était, à l’origine, de 4.85 unités arbitraires (UA) dans des souris alimentées à la DG, a chuté à 3.27 UA après avoir reçu un traitement de R. groenlandicum. Le R. groenlandicum a réduit la stéatose rénale de presque la moitié alors que l’expression du facteur de modification Bcl-2 (Bmf) a chuté de 13.96 UA à 9.43 UA.
Dans leur ensemble les résultats suggèrent que le traitement avec R. groenlandicum peut améliorer la fonction rénale altérée par DG.
Dans l’étude subséquente, notre équipe a identifié 17 espèces de la forêt boréale, de la pharmacopée traditionnelle des Cris de la Baie James orientale, qui ont présenté des activités biologiques prometteuses in vitro et in vivo dans le contexte du DT2.
Nous avons maintenant examiné ces 17 extraits afin d’identifier lesquels possèdent un potentiel cytoprotecteur rénale en utilisant des cellules Madin Darby Canine Kidney (MDCK) mises à l’épreuve dans un médium hypertonique.
Nous concluons que plusieurs plantes antidiabétiques Cris exercent une activité de protection rénale qui pourrait être pertinente dans le contexte de la néphropathie diabétique (ND) qui affecte une proportion importante des Cris. La G. hispidula et la A. balsamea sont parmi les plantes les plus puissantes dans ce contexte et elles semblent protectrices principalement en inhibant la caspase 9 dans la voie de signalisation apoptotique mitochondriale.
Finalement, nous avons utilisé une approche de fractionnement guidée par un test biologique pour identifier les fractions actives et les composés de A. balsamea avec un potentiel de protection rénale in vitro dans des cellules MDCK mises au défi avec un médium hypertonique.
La fraction d’hexane (Hex) possède le potentiel le plus élevé parmi toutes les fractions de solvant contre les dommages cellulaires induits par le stress hypertonique. Dans des études précédentes, trois composés purs ont été identifiés à partir de la fraction Hex, à savoir, l’acide abiétique, l’acide déhydroabiétique et le squalène. L’acide abiétique se distinguait par son effet puissant dans le maintien de la viabilité des cellules MDCK (AnnV-/PI-) à un niveau relativement élevé (augmentation de 25.48% relative au stress hypertonique, P<0.0001), ainsi qu’une réduction significative (diminution de 20.20% par rapport au stress hypertonique, P<0.0001) de l’apoptose de stade précoce (AnnV+/PI-). L’acide abiétique peut donc servir à normaliser les préparations traditionnelles d’A. balsamea et à trouver des applications potentielles dans le traitement de la néphropathie diabétique.
Les trois études ont été intrinsèquement liées les unes aux autres, par conséquent, nous avons réussi à identifier R. groenlandicum ainsi que A. balsamea comme nouvelles plantes prometteuses contre la néphropathie diabétique. Nous croyons que ces résultats profiteront à la communauté crie pour la gestion des complications diabétiques, en particulier la néphropathie diabétique. En parallèle, nos données pourraient faire avancer l'essai clinique de certaines plantes médicinales de la pharmacopée traditionnelle des Cris de la Baie James orientale du Canada. / Our team has identified Labrador tea [Rhododendron groenlandicum L. (Ericaceae)] as a potential antidiabetic plant from the traditional pharmacopeia of the Eastern James Bay Cree. In the present study, we assessed the plant’s potential renoprotective effects.
Microalbuminuria and renal fibrosis was developed in high fat diet (HFD)-fed mice. Meanwhile, there was a tendency for R. groenlandicum to improve microalbuminuria, with the values of area under the curve (ACR) reducing from 0.69 to 0.53. Renal fibrosis value was originally 4.85 arbitrary units (AU) in HFD-fed mice, dropped to 3.27 AU after receiving R. groenlandicum treatment. R. groenlandicum reduced renal steatosis by nearly one half whereas the expression of Bcl-2-modifying factor (Bmf) diminished from 13.96 AU to 9.43 AU.
Taken altogether, the results suggest that R. groenlandicum treatment can improve renal function impaired by HFD.
In the following study, our team has identified 17 Boreal forest species from the traditional pharmacopeia of the Eastern James Bay Cree that presented promising in vitro and in vivo biological activities in the context of type 2 diabetes (T2D).
We now screened the 17 plants extracts for potential renal protective activity using Madin Darby Canine Kidney (MDCK) cells challenged with hypertonic medium. Cytoprotective potential as well as underlying mechanisms were investigated.
We conclude that several Cree antidiabetic plants exert renal protective activity that may be relevant in the context of diabetic nephropathy that affects a significant proportion of Cree diabetics. G. hispidula and A. balsamea are amongst the most powerful plants in this context and they appear to exert their modulatory effect primarily by inhibiting caspase 9 in the mitochondrial apoptotic signaling pathway.
We used a bioassay-guided fractionation approach to identify active fractions and compounds of A. balsamea with renal protective potential in vitro in MDCK cells challenged with hypertonic medium.
The hexane (Hex) fraction possessed the highest potential among all solvent fractions against cell damage induced by hypertonic stress. In previous studies, three pure compounds were identified from the Hex fraction, namely, abietic acid, dehydroabietic acid and squalene. Abietic acid stood out by its strongest effect in maintaining MDCK cell viability (AnnV-/PI-) at a relatively high level (25.48% increase relative to hypertonic stress, P<0.0001) as well as a significant reduction (20.20% decrease relative to hypertonic stress, P<0.0001) of early stage apoptosis (AnnV+/PI-). Abietic acid may thus serve to standardize A. balsamea traditional preparations and find potential applications in the treatment of diabetic nephropathy.
The three studies were inherently associated with each other, as a result we had successfully identified R. groenlandicum as well as A. balsamea as novel medicinal plants against diabetic nephropathy. We believe such findings will benefit the Cree community for the management of diabetic related complications, especially diabetic nephropathy. Meanwhile, our data might advance the clinical trial of certain medicinal plants from the traditional pharmacopeia of the Canadian Eastern James Bay Cree.
