Expression und Funktion der Chemokinrezeptoren CXCR4 und CXCR7 in der Schwannzelllinie RN22

Schwannzellen sind die myelinisierenden Zellen des peripheren Nervensystems, die auch eine Rolle bei Entzündungs- und Regenerationsprozessen spielen. Diese Arbeit beschäftigt sich mit der Bedeutung der Chemokinrezeptoren CXCR4 und CXCR7 bei der SDF-1-abhängigen Signalübermittlung in der Schwannzelllinie RN22. Mittels PCR, Western-Blotting und FACS erfolgte zunächst ein Expressionsnachweis der Chemokinrezeptoren. Anschließend wurde mittels Western-Blotting festgestellt, dass eine Behandlung mit SDF-1 zu einer Aktivierung der intrazellulären Signalkinasen Erk1/2, p38 und Akt führt. Für PKC ζ/λ wurde kein Effekt beobachtet. Durch Einsatz der spezifischen Antagonisten für CXCR4 (AMD3100) und CXCR7 (CCX733) konnten diese Effekte blockiert werden. Die Ergebnisse legen nahe, dass beide Rezeptoren ihren Liganden binden müssen, damit ein intrazellulärer Effekt auftritt. Ähnliches gilt auch für die SDF-1-abhängige Migration von RN22-Zellen, die ebenfalls bereits durch einen der beiden Antagonisten unterbunden werden kann.

Etude de l'impact clinique et immunologique d'un traitement par lénalidomide dans la maladie de Kaposi liée au VIH / Clinical and immunological impact of lenalidomide in patients with HIV-associated Kaposi's sarcoma receiving antiretroviral therapy

L'objectif de cette thèse menée dans le cadre de l'essai clinique ANRS 154 Lenakap a été d'étudier l'impact clinique et immunologique d'un traitement par lénalidomide dans la maladie de Kaposi liée au VIH (MK-VIH) et d'identifier de nouveaux biomarqueurs de la pathologie, notamment à travers l'étude du trio de chimiokine et récepteurs CXCL12/CXCR4-CXCR7. A l'heure actuelle aucun traitement curatif de la MK-VIH n'est disponible. Le lénalidomide, un immunomodulateur pléïotrope, ciblant différentes anomalies rencontrées dans la MK constitue une perspective thérapeutique dans cette indication. L'interprétation de la réponse clinique des patients au traitement au cours de l'essai clinique ANRS 154 Lenakap est difficile, notamment en raison de discordances entre les scores d'évaluation utilisés. Cependant, nos résultats ont montré une bonne tolérance du lénalidomide chez les patients inclus, infectés par le VIH et traités par antirétroviraux. Aucune interaction médicamenteuse pharmacologiquement ou cliniquement significative n'a été détectée chez les patients, ouvrant de nouvelles perspectives pour la prise en charge de ces derniers, y compris dans d'autres indications, tels que le myélome multiple et les syndromes myélodysplasiques. Nous avons également mis en évidence l'impact du TNF-α, de l'IFN-γ et de l'IL-10 dans la progression de la MK-VIH.L'ensemble des mécanismes physiopathologiques de la MK n'est pas encore élucidé et nous ne disposons actuellement d'aucun biomarqueur, de suivi d'évolution, ou de réponse au traitement dans cette indication. Des données de la littérature suggèrent une implication du trio CXCL12/CXCR4-CXCR7 dans la physiopathologie de la MK-VIH. Nos analyses immunohistochimiques et par immunofluorescence, couplées à la mise au point d'une technique de quantification sur lames numérisées ont permis de mettre en évidence la présence augmentée des protéines CXCL12/CXCR4-CXCR7 dans les lésions de MK. Les corrélations positives retrouvées entre les protéines du trio et la présence du virus sous forme latente, la prolifération cellulaire et à la présence du facteur de croissance VEGF, suggèrent de possibles effets autocrines et paracrines du trio à l'origine d'une propagation tissulaire du virus et d'effets prolifératif et pro-angiogénique dans le MK. Cette étude suggère pour la première fois le rôle de biomarqueur tissulaire, témoin du processus physiopathologique de la MK, pour le trio CXCL12/CXCR4-CXCR7. En revanche, CXCL12 ne semble pas être un biomarqueur plasmatique à la fois de la physiopathologie ou de la progression de la MK. Enfin, cette étude confirme l'état pro-inflammatoire des patients infectés par le VIH et rapporte une immunomodulation particulière chez les patients MK-VIH avec des taux plasmatiques de TNF-alpha, IL-6, IFN-gamma et IL-10 augmentés, peut-être à l'origine du développement et de la progression de la maladie. / The objective of my PhD works, conducted as part of the clinical trial ANRS 154 Lenakap, was to evaluate the clinical and immunological impact of lenalidomide in patients with HIV-associated Kaposi's sarcoma (AIDS-KS) and to identify new biomarkers of disease, particularly through the study of the trio CXCL12/CXCR4-CXCR7 So far, no cure for the AIDS-KS is available. Lenalidomide, an oral immunomodulating agent targeting various anomalies observed in KS is a therapeutic perspective in this indication. Evaluation of clinical response to treatment in the ANRS 154 Lenakap clinical trial was difficult, especially because of discrepancies observed between assessments scores used to evaluate this parameter. However, lenalidomide was well tolerated in patients infected with HIV and treated with antiretroviral drugs. We detected no pharmacologically or clinically significant drug interactions between lénalidomide and antiretroviral drugs, opening new perspectives for the treatment of HIV-positive patients, including other indications such as multiple myeloma and myelodysplastic syndromes. We also highlighted the impact of TNF-alpha, IFN-gamma and IL-10 in the progression of AIDS-KS. All pathophysiological mechanisms of KS are not yet elucidated, and so far, no biomarker is available to monitor evolution, or response to treatment in this indication. Some data suggest an involvement of the trio CXCL12/CXCR4-CXCR7 in the pathophysiology of KS. Our immunohistochemical and immunofluorescence analysis, coupled to a technique for quantification of digitized slides, have allowed us to demonstrate the over-expression of CXCL12, CXCR4 and CXCR7 proteins in KS cutaneous lesions. Moreover, we reported for the first time the simultaneous in situ up-regulation of CXCL12, CXCR4 and CXCR7 in AIDS- and classic-KS. These deregulations correlated with lesion severity, latent viral load, proliferation and angiogenesis. This suggests a possible autocrine and paracrine effects of the trio leading to the virus propagation, the cells proliferation and the angiogenic process observed in KS. Our results further indicate that the trio could be used in KS rather as a histologic than a circulating biomarker. Finally, this study confirms the pro-inflammatory state of HIV-infected patients and highlights a specific immune modulation in AIDS-KS patients with increased TNF-α, IL-6, IFN-γ and IL-10 plasma levels. This microenvironment may participate in the disease progression.

Etude de la régulation du gène codant le récepteur de chimiokine CXCR4 dans le système de la ligne latérale postérieure du poisson-zèbre (danio rerio) / Study of the regulation of the gene encoding the chemokine receptor CXCR4 in the zebrafish (danio rerio) posterior lateral line system

La ligne latérale postérieure embryonnaire du poisson-zèbre est composée d'un ensemble d'organes sensoriels, appelés neuromastes, qui permet au poisson de détecter les mouvements de l'eau. Le primordium qui génère la ligne latérale postérieure embryonnaire migre de la tête vers l'extrémité de la queue le long d'une piste de cellules sécrétrices de SDF-1, et dépose des groupes de cellules précurseurs des neuromastes. Cette migration dépend de la présence de CXCR4, le récepteur de SDF-1, dans la région en tête du primordium et de la présence du second récepteur de SDF-1, CXCR7, dans la région en queue du primordium. L'objectif de ma thèse est d'identifier des régulateurs de l'expression de cxcr4b au sein du primordium. Nous avons montré que l'inactivation du récepteur des strogènes ESR1 induit l'expression ectopique de cxcr4b dans les cellules de queue du primordium alors que sa surexpression induit une réduction du domaine d'expression de cxcr4b, suggérant que ESR1 agit comme un répresseur de cxcr4b. Cette découverte expliquerait pourquoi les strogènes diminuent la capacité métastatique des cellules du cancer du sein strogéno-dépendants. L'inactivation de ESR1 conduit aussi à l'extinction de l'expression de cxcr7b dans les cellules de queue du primordium, cet effet étant toutefois indirect et induit par la signalisation ectopique SDF-1/CXCR4 dans ces cellules. L'inactivation et la surexpression de ESR1 provoquent toutes deux une migration défectueuse du primordium, confirmant l'importance de ce récepteur dans le contrôle de la migration dépendante de SDF-1. Nous avons aussi montré qu'un effecteur majeur de la signalisation Wnt canonique, LEF-1, contribue au contrôle de l'expression de cxcr4b et de cxcr7b dans les cellules en tête du primordium. Nous montrons que la prolifération cellulaire, qui assure une taille constante du primordium en dépit des dépositions successives de cellules, est réduite en absence de LEF-1, et que cela conduit à une ligne latérale postérieure incomplète. / The zebrafish embryonic posterior lateral line is componed by a set sense organs, called neuromasts, allowing the fish to detect the water movements. The primordium that generates the embryonic posterior lateral line of zebrafish migrates from the head to the tip of the tail along a trail of SDF-1-producing cells, and deposits cell groups that will become the neuromasts. This migration critically depends on the presence of the SDF-1 receptor CXCR4 in the leading region of the primordium and on the presence of a second SDF1 receptor, CXCR7, in the trailing region of the primordium. The aim of my thesis is to identify regulators of the cxcr4b expression within the primordium. Here we show that inactivation of the estrogen receptor ESR1 results in ectopic expression of cxcr4b throughout the primordium, whereas ESR1 overexpression results in a reciprocal reduction in the domain of cxcr4b expression, suggesting that ESR1 acts as a repressor of cxcr4b. This finding could explain why estrogens significantly decrease the metastatic ability of ESR-positive breast cancer cells. ESR1 inactivation alsoleads to extinction of cxcr7b expression in the trailing cells of the migrating primordium; this effect is indirect, however, and due to the down-regulation of cxcr7b by ectopic SDF-1/CXCR4 signaling in the trailing region. Both ESR1 inactivation and overexpression result in aborted migration, confirming the importance of this receptor in the control of SDF-1-dependent migration. We also showed that a major effector of the canonical Wnt signaling, LEF-1, contributes to the control of both cxcr4b and cxcr7b expression in the leading cells of the primordium. We show that cell proliferation, which ensures constant primordium size in spite of sucessive rounds of cell deposition, is reduced upon lef1 inactivation, leading in a truncated posterior lateral line.

Etude des mécanismes impliqués dans le processus métastatique dans le cancer colique humain : implication de l'axe CXCL12/CXCR4/CXCR7 / Study of the mechanisms involved in the metastatic process in human colic cancer : implication of the CXCL12 / CXCR4 / CXCR7 axis

Malgré les diagnostics précoces et les avancées thérapeutiques, le taux de mortalité chez les patients diagnostiqués d’un CCR au stade métastatique reste très élevé. L’objectif de ce travail a été d’étudier le rôle de la chimiokine CXCL12 et ses deux récepteurs CXCR4 et CXCR7 dans processus métastatique. J’ai comparé l’expression de la chimiokine CXCL12 dans des tumeurs coliques humaines avec les tissus sains associés puis je me suis intéressée aux mécanismes de régulation de cette expression et plus particulièrement la régulation épigénétique. J’ai montré que le promoteur de CXCL12 est méthylé dans 35% des CCR et qu’un défaut d’acétylation des histones du promoteur entraîne la perte d’expression de CXCL12. Des enzymes impliquées dans la régulation des mécanismes épigénétiques, potentiellement liées à ce défaut d’acétylation ont été identifiées par l’analyse des tumeurs par PCR Array et parmi ces facteurs, j’ai identifié l’histone acétyl-transférase PCAF dont l’expression est diminuée dans les tumeurs. Enfin, pour comprendre le rôle respectif de CXCR4 et de CXCR7 dans la dissémination métastatique, j’ai invalidé l’expression du gène des récepteurs dans la lignée colique humaine SW480 par Crispr-Cas9, puis j’ai comparé la capacité migratoire des cellules in vitro et leur potentiel métastatique in vivo. L’induction d’une perte d’expression du récepteur CXCR7 n’a pas eu d’impact sur le développement des métastases pulmonaire et hépatique in vivo, mais a entraîné une baisse de la migration in vitro. / Despite early cancer detection and therapeutic advances, the mortality rate in patients diagnosed with CRC at the metastatic stage remains very high. The aim of this work was to study the role of the chemokine CXCL12 and its two receptors CXCR4 and CXCR7 in the metastatic process. I compared the expression of the chemokine CXCL12 in human colon tumors with the associated healthy tissues, then I focused on the mechanisms regulating this expression and more particularly the epigenetic regulation. I have shown that the CXCL12 promoter is methylated in 35% of the CCR and that a lack of histone acetylation of the CXCL12 promoter causes the loss of its expression. Enzymes involved in the regulation of epigenetic mechanisms, potentially related to this acetylation defect, were identified by Array PCR on tumors and among these factors, the histone acetyl transferase PCAF, whose expression is decreased in tumors, was identified. Finally, to understand the respective role of CXCR4 and CXCR7 in metastatic spread, I invalidated the expression of both receptor genes in the human colonic line SW480 by Crispr-Cas9, and then compared the migratory capacity of the cells in vitro and their metastatic potential in vivo. Inducing a loss of expression of CXCR7 receptor did not have an impact on the development of pulmonary and hepatic metastases in vivo, but resulted in a decrease in in vitro migration.

Das Chemokine SDF-1α und sein Rezeptor CXCR4 sind in eine Vielzahl biologischer Prozesse, wie der Organogenese, der Hämatopoese und der Immunantwort involviert. Die Entdeckung des alternativen SDF-1α-Rezeptors CXCR7 bewirkte eine erneute Untersuchung der Funktion des SDF-1-Systems in diesen Vorgängen. CXCR7 ist in der Lage, je nach Gewebe- oder Zelltyp, als \"Scavenger\"-Rezeptor, Modulator des CXCR4 oder selbstständig aktiver Rezeptor zu agieren. In dieser Arbeit wurde untersucht, inwiefern beide Rezeptoren im Verlauf der Entwicklung der Muskulatur exprimiert werden, welche Aufgabe sie dabei übernehmen und ob sich Rückschlüsse auf die Muskelregeneration daraus ableiten lassen. Hierfür erfolgten in vitro-Untersuchungen an C2C12-Zellen und die anschließende Analyse der Expression von CXCR4, CXCR7 und SDF-1α in der sich entwickelnden Gliedmaßenmuskulatur von Wildtyp- und mdx-Mäusen. Die Untersuchung von C2C12-Zellen zeigte in allen Differenzierungsstadien eine detektierbare Menge von CXCR4 und eine zunehmende Expression des CXCR7. Die Behandlung der Zellen mit SDF-1α führte zu einer Phosphorylierung von Erk1/2 und PKCζ/λ und hemmte gleichzeitig deren Differenzierung. Nach einer Blockierung des CXCR4 mit seinem pharmakologischen Antagonist AMD3100 oder nach Hemmung der Expression durch spezifische siRNA blieb die Aktivierung des Signalweges aus und der hemmende Einfluss des SDF-1α auf die Differenzierung verschwand vollständig. Im Gegensatz dazu blieben nach der pharmakologischen Blockierung oder durch siRNA vermittelten Expressionshemmung des CXCR7 alle SDF-1α induzierten Effekte vollständig erhalten. Eine Untersuchung des Signalweges am dritten Tag der Differenzierung zeigte keine Aktivierung von Erk1/2. Ebenso blieb Erk1/2 nach einer Hemmung der Expression des CXCR4 unphosphoryliert. Im Gegensatz dazu fand nach einer Hemmung der Expression des CXCR7 mit spezifischer siRNA erneut eine Aktivierung des Signalweges statt. Weiterhin konnte in vivo festgestellt werden, dass die Expression des CXCR4 in der Muskulatur während der embryonalen Entwicklung am höchsten ist und bereits kurz nach der Geburt stark abnimmt, wenn die sekundäre Muskelentwicklung abgeschlossen ist. Die Expression des CXCR7 hingegen steigt perinatal an und bleibt bis zum Erwachsenenalter bestehen. Zusammengefasst zeigen diese Ergebnisse, dass CXCR4 aktiv das Signalgeschehen von SDF-1α in der Myogenese vermittelt und somit zur Differenzierungshemmung von Myoblasten beiträgt. CXCR7 hingegen wirkt als passiver SDF-1α-Scavenger und induziert somit durch eine Modulierung der extrazellulären SDF-1α-Konzentration die Differenzierung. In Übereinstimmung mit der Rolle des SDF-1α-Systems bei der Muskelentwicklung, konnte eine kontinuierliche SDF-1α- Expression im Bindegewebe um pränatale und im Endomysium von postnatalen und adulten Muskelfasern festgestellt werden. Diese SDF-1α-Expression stieg ebenso wie die CXCR4-Expression bei der Analyse der Muskulatur von dystrophin-defizienten Mäusen an und zeigte somit, dass dieses System auch für die Proliferation von Muskelvorläuferzellen in der regenerativen Muskulatur eine wichtige Rolle spielt. Bemerkenswerter Weise hatte diese Muskeldystrophie keinen Einfluss auf die Expression des CXCR7 und beeinflusst vermutlich dessen Funktion über einen anderen Mechanismus. Durch die offensichtlich enge Kontrolle von Muskelentwicklung und Regeneration durch CXCR4, CXCR7 und deren Liganden SDF-1α, bilden diese ein vielversprechendes therapeutisches Ziel für bestimmte Muskelerkrankungen. / The chemokine, SDF-1α, and its receptor, CXCR4, are assumed to control a multitude of biological processes such as organogenesis, haematopoesis, and immune responses. The previous demonstration that SDF-1α additionally binds to the chemokine receptor, CXCR7, currently urges a re-evaluation of the function of the SDF-1 system in these processes. In fact, depending on the tissue and cell type, CXCR7 either acts as a scavenger receptor, a modulator of CXCR4 or an independent active receptor. This thesis is dedicated to answer the following questions: Are both SDF-1α receptors expressed during muscle development? What is the actual function of these receptors during myogenesis? Is there a role of the SDF-1 system in muscle regeneration? To adress these issues both in vitro studies with the myoblast cell line, C2C12, as well as in vivo analyses on the expression of CXCR4, CXCR7 and SDF-1α in developing and regenerating limb muscles have been performed. At all stages of differentiation, C2C12 cells exhibited measurable amounts of CXCR4. In addition, in the course of differentiation C2C12 cells showed increasing expression levels of CXCR7. Treatment of the cells with SDF-1α resulted in the phosphorylation of Erk1/2 and PKCζ/λ and subsequently blocked their myogenic differentiation. Following inactivation of CXCR4 either by its antagonist, AMD3100, or by specific siRNA, SDF-1α failed to activate both pathways and in addition no longer inhibited the myogenic differentiation of C2C12 cells. By contrast, inactivation of CXCR7 remained without effects on SDF-1α-induced cell signalling and resulting inhibitory effects on myogenic differentiation. Interestingly, SDF-1α also failed to activate Erk1/2 signalling in differentiated C2C12 cells. Cell signalling in differentiated C2C12 cells was, however, restored following inhibition of CXCR7 expression, but not following inhibition of CXCR4 expression. The in vivo analysis further revealed that in limb muscles expression of the CXCR4 is highest during embryonic development with a decrease in expression levels shortly after birth when secondary muscle development is completed. Vice versa, expression levels of CXCR7 increased perinatally and remained high into adulthood. In summary, these findings unravel that CXCR4 actively mediates SDF-1α-signalling during myogenesis. The findings further provide evidence that CXCR7 acts as a scavenger receptor during myogenesis which controls myogenic differentiation by modulating extracellular SDF-1α concentration. In further agreement with a major role of SDF-1α in muscle development, SDF-1α is continously expressed by the endomysium of postnatal and adult muscle fibers. Moreover, expression of SDF-1α as well as CXCR4 is massively increased in muscles of dystrophin-deficient mice further implying that the SDF-1 system plays an equally important role during muscle development and regeneration. The pivotal role of SDF-1α in muscle development and regeneration points to the SDF-1 system as a promising therapeutical target for certain muscle diseases.

Eine Untersuchung des Signalweges am dritten Tag der Differenzierung zeigte keine Aktivierung von Erk1/2. Ebenso blieb Erk1/2 nach einer Hemmung der Expression des CXCR4 unphosphoryliert. Im Gegensatz dazu fand nach einer Hemmung der Expression des CXCR7 mit spezifischer siRNA erneut eine Aktivierung des Signalweges statt. Weiterhin konnte in vivo festgestellt werden, dass die Expression des CXCR4 in der Muskulatur während der embryonalen Entwicklung am höchsten ist und bereits kurz nach der Geburt stark abnimmt, wenn die sekundäre Muskelentwicklung abgeschlossen ist. Die Expression des CXCR7 hingegen steigt perinatal an und bleibt bis zum Erwachsenenalter bestehen. Zusammengefasst zeigen diese Ergebnisse, dass CXCR4 aktiv das Signalgeschehen von SDF-1α in der Myogenese vermittelt und somit zur Differenzierungshemmung von Myoblasten beiträgt. CXCR7 hingegen wirkt als passiver SDF-1α-Scavenger und induziert somit durch eine Modulierung der extrazellulären SDF-1α-Konzentration die Differenzierung. In Übereinstimmung mit der Rolle des SDF-1α-Systems bei der Muskelentwicklung, konnte eine kontinuierliche SDF-1α- Expression im Bindegewebe um pränatale und im Endomysium von postnatalen und adulten Muskelfasern festgestellt werden. Diese SDF-1α-Expression stieg ebenso wie die CXCR4-Expression bei der Analyse der Muskulatur von dystrophin-defizienten Mäusen an und zeigte somit, dass dieses System auch für die Proliferation von Muskelvorläuferzellen in der regenerativen Muskulatur eine wichtige Rolle spielt. Bemerkenswerter Weise hatte diese Muskeldystrophie keinen Einfluss auf die Expression des CXCR7 und beeinflusst vermutlich dessen Funktion über einen anderen Mechanismus. Durch die offensichtlich enge Kontrolle von Muskelentwicklung und Regeneration durch CXCR4, CXCR7 und deren Liganden SDF-1α, bilden diese ein vielversprechendes therapeutisches Ziel für bestimmte Muskelerkrankungen. / The chemokine, SDF-1α, and its receptor, CXCR4, are assumed to control a multitude of biological processes such as organogenesis, haematopoesis, and immune responses. The previous demonstration that SDF-1α additionally binds to the chemokine receptor, CXCR7, currently urges a re-evaluation of the function of the SDF-1 system in these processes. In fact, depending on the tissue and cell type, CXCR7 either acts as a scavenger receptor, a modulator of CXCR4 or an independent active receptor. This thesis is dedicated to answer the following questions: Are both SDF-1α receptors expressed during muscle development? What is the actual function of these receptors during myogenesis? Is there a role of the SDF-1 system in muscle regeneration? To adress these issues both in vitro studies with the myoblast cell line, C2C12, as well as in vivo analyses on the expression of CXCR4, CXCR7 and SDF-1α in developing and regenerating limb muscles have been performed. At all stages of differentiation, C2C12 cells exhibited measurable amounts of CXCR4. In addition, in the course of differentiation C2C12 cells showed increasing expression levels of CXCR7. Treatment of the cells with SDF-1α resulted in the phosphorylation of Erk1/2 and PKCζ/λ and subsequently blocked their myogenic differentiation. Following inactivation of CXCR4 either by its antagonist, AMD3100, or by specific siRNA, SDF-1α failed to activate both pathways and in addition no longer inhibited the myogenic differentiation of C2C12 cells. By contrast, inactivation of CXCR7 remained without effects on SDF-1α-induced cell signalling and resulting inhibitory effects on myogenic differentiation. Interestingly, SDF-1α also failed to activate Erk1/2 signalling in differentiated C2C12 cells. Cell signalling in differentiated C2C12 cells was, however, restored following inhibition of CXCR7 expression, but not following inhibition of CXCR4 expression. The in vivo analysis further revealed that in limb muscles expression of the CXCR4 is highest during embryonic development with a decrease in expression levels shortly after birth when secondary muscle development is completed. Vice versa, expression levels of CXCR7 increased perinatally and remained high into adulthood. In summary, these findings unravel that CXCR4 actively mediates SDF-1α-signalling during myogenesis. The findings further provide evidence that CXCR7 acts as a scavenger receptor during myogenesis which controls myogenic differentiation by modulating extracellular SDF-1α concentration. In further agreement with a major role of SDF-1α in muscle development, SDF-1α is continously expressed by the endomysium of postnatal and adult muscle fibers. Moreover, expression of SDF-1α as well as CXCR4 is massively increased in muscles of dystrophin-deficient mice further implying that the SDF-1 system plays an equally important role during muscle development and regeneration. The pivotal role of SDF-1α in muscle development and regeneration points to the SDF-1 system as a promising therapeutical target for certain muscle diseases.

Le cancer colorectal métastatique est l’une des premières causes de décès par cancer dans les pays occidentaux, malgré le développement récent de nouveaux traitements ciblés. L’amélioration de la survie des patients passe par une meilleure compréhension des mécanismes moléculaires impliqués dans la progression tumorale et la formation des métastases. Compte tenu de l’importance du rôle des axes mTOR/HIF1α et CXCL12/CXCR4/CXCR7 dans le processus métastatique, nos objectifs ont été : i) d’analyser de façon extensive le statut de CXCL12 dans une collection de polypes et de tumeurs coliques de tous stades et phénotypes en comparaison avec la muqueuse saine, puis de comprendre les mécanismes régulant l’expression de la chimiokine dans les cellules tumorales ; ii) d’étudier in vitro le rôle de l’hypoxie dans la régulation de la signalisation induite par CXCL12 via CXCR4 et CXCR7 et l’intérêt d’une inhibition des axes mTOR/HIF-1α et CXCL12/CXCR4/CXCR7 en particulier sur les capacités migratoires des cellules tumorales.Nous avons montré que l’extinction du gène CXCL12 est un évènement précoce et systématique au cours de la cancérogenèse colique et que cette perte d’expression pourrait être régulée par un mécanisme d’acétylation au niveau des histones. Une expression différentielle de CXCR4 et CXCR7 au sein des tumeurs du colon a été mise en évidence. L’augmentation d’expression de CXCR7 dans les métastases par rapport aux stades précoces montre l’importance de cet axe dans le processus métastatique. Le maintien de l’expression à la surface cellulaire de CXCR4 en normoxie pendant au moins 24h après un bref passage en hypoxie n’avait pas encore été décrit. Ceci pourrait expliquer le « homing » des cellules tumorales circulantes dans les niches métastatiques selon un gradient de CXCL12. L’utilisation combinée d’irinotécan et de chalcone permettant d’inhiber la migration des cellules tumorales est une approche originale in vitro. Enfin, nous avons initié le développement de modèles métastatiques de cancer du colon par greffe orthotopique sur le caecum de souris NUDE dans l’objectif de tester de nouvelles approches thérapeutiques ciblées in vivo. / Despite the recent development of new targeted chemotherapies, metastatic colorectal cancer is still one of the leading causes of cancer related deaths in western countries. A better understanding of metastatic process would improve survival. Since the role of mTOR/HIF1α and CXCL12/CXCR4/CXCR7 axes in metastasis formation, our objectives were: i) to analyze extensively CXCL12 status in a collection of polyps and colon tumors whatever stages and phenotypes; ii) to study the role of hypoxia in CXCL12/CXCR4/CXCR7 signaling pathway in vitro and on tumor cells migration. We have shown that the CXCL12 extinction is a systematic early event during colorectal carcinogenesis. CXCL12 loss expression may be regulated by histone acetylation mechanism. There is a differential CXCR4 and CXCR7 expression in colon tumors. Increased CXCR7 expression in metastasis compared to early stages underlines the importance of this axis in metastatic process. We have shown for the first time that CXCR4 expression remained stabilized at the cell membrane 24 hours after a transient passage in hypoxia. It could explain circulating cells are attracted in metastatic niches under CXCL12 gradient. Drug combinations with chalcone and irinotecan are an original approach for inhibiting cell migration in vitro. Finally, we have initiated the development of a metastatic model of colon cancer with orthotopic colon human tumors xenograft in NUDE mice to test new therapeutic approaches in vivo.

Die Rolle der Chemokinrezeptoren CXCR3 und CXCR7 bei den tumorogenen Einflüssen von CXCL11

Das Chemokin CXCL11/I-TAC wirkt über die beiden Chemokinrezeptoren CXCR3 und CXCR7 und kontrolliert die Entwicklung verschiedener Organe, sowie die Hämatopoese, Angiogenese und die Lymphozytenwanderung. Bekannte pathophysiologische Funktionen des CXCL11 Systems umfassen des Weiteren die Kontrolle von Immunantworten, die Entstehung und Progression von Autoimmunerkrankungen, sowie die Metastasierung von verschiedenen Tumoren. Trotz dieser weitreichenden Einflüsse und der wachsenden Bedeutung des CXCL11 Systems bei Krebserkrankungen ist bis heute nur unzureichend geklärt über welche Rezeptoren die Einflüsse von CXCL11 vermittelt werden. Dieser Aspekt wird dabei durch die Tatsache erschwert, dass der humane CXCR3 Rezeptor in 3 Splicevarianten existiert: CXCR3A, CXCR3B und in der trunkierten Form CXCR3alt. Dabei wirkt CXCR3A tumorfördernd, indem er Tumorzellmigration und Zellproliferation stimuliert. CXCR3B dagegen hemmt Tumorwachstum und Tumorzellmigration, wie auch Angiogenese. In dieser Arbeit sollte die Lokalisation und das Expressionsmuster der Rezeptoren CXCR7 und CXCR3, mit seinen beiden Varianten CXCR3A und CXCR3B, bei fünf ausgewählten humanen Tumorzelllinien analysiert und hinsichtlich ihrer Beteiligung bei Tumorzellmigration und Proliferation verglichen werden. Mit Hilfe des Boyden-Chamber-Assay konnte gezeigt werden, dass CXCL11 die Migration aller Tumorzelllinien stimuliert. Die chemotaktische Antwort der einzelnen Tumorzelllinien auf CXCL11 zeigte dabei eine unterschiedliche Sensitivität für den spezifischen CXCR3-Antagonisten AMG487 und den spezifischen CXCR7- Antagonisten CCX771 und wird somit in den einzelnen Tumorzellen durch unterschiedliche Rezeptoren geregelt. Mittels Western-Blot und Immunzytochemie wurden die Proteinlevel und die subzelluläre Lokalisation der beiden CXCL11- Rezeptoren CXCR3 und CXCR7 in den fünf verschiedenen Tumorzelllinien genauer betrachtet. Die immunzytochemische Analyse bestätigte, dass alle untersuchten Tumorzelllinien CXCR3 und CXCR7 ko-exprimieren. Beide Rezeptoren liegen sowohl in der äußeren Zellmembran vor, als auch intrazellulär. Interessanterweise führte AMG487 in bestimmten Tumorzelllinien zu einer Potenzierung des chemotaktischen Einflusses von CXCL11, was vermutlich auf der Hemmung des antitumorogenen Signals von CXCR3B beruht. Für diese Annahme spricht ebenfalls die Beobachtung, dass CXCL4, welches selektiv an CXCR3B bindet, die chemotaktische Wirkung von CXCL11 auf die entsprechenden Tumorzellen hemmte. Letztendlich ergab die quantitative real-time-PCR Analyse, dass in allen Tumorzelllinien der Gehalt an CXCR3B mRNA im Vergleich zu dem der CXCR3A mRNA deutlich höher ist. Insgesamt zeigen diese Untersuchungen, dass das Signal von CXCL11 zellspezifisch entweder über CXCR3 oder CXCR7, oder beide zusammen vermittelt wird.
