It is necessary to clarify the mode of action through which it has caused rodent neoplasia and verify its relevance to humans. Herein, two experiments were developed to verify the initiating and promoting potentials of diuron in a 23- and a 21-week long mouse skin carcinogenesis protocol. In one, dimethylsulphoxide (DMSO) was the solvent for the herbicide; in the other, acetone was the alternative solvent in order to verify if DMSO had inhibitory influence on a potential cutaneous carcinogenic activity. The adopted schedule for the tumor promoting agent 12-O-tetradecanoylphorbol 13-acetate (TPA) resulted in skin ulcers, evidencing the need for careful selection of TPA dose levels and frequency of application in this model. In both studies diuron did not exert any influence on the skin carcinogenesis process, in contrast with results already reported in the literature.

Foram obtidos diagramas de difração a baixo ângulo para os sistemas: DPL, DPL - pireno, DPL - 3, 4- benzopireno e DPL - 1, 2, 5, 6 - dibenzantraceno. Estes diagramas, subseqüentemente, foram analisados usando-se as intensidades das reflexões lamelares. A distribuição de densidade eletrônica normal ao plano da bicamada foi calculada pela síntese de Fourier, com resolução de 7.5 Aº. No caso do pireno, o mapa de densidade eletrônica mostra uma leve perturbação da estrutura na região hidrofóbica, próxima ao grupo glicerol. Este resultado concorda com as experiências de NMR realizadas por Vanderkooi e colaboradores (J. Chem. Phys., 63, 3662, 1975). Para os sistemas de DPL - 3,4 - benzopireno a distorção na região da queda abrupta existente no centro de simetria do perfil de densidade eletrônica é similar àquela observada para as amostras de DPL - pireno. A incorporação do 1, 2, 5, 6 - dibenzantraceno dentro das bicamadas afeta bastante o empacotamento das cadeias, por perturbações ao longo das cadeias de carbono e na região do centro da bicamada correspondente aos grupos terminais / Abstract: Polycyclic aromatic hydrocarbons were incorporated into hydrated dipalmitoyl lecithin (DPL) multilayers in order to detect structural changes induced in the host model membrane. Low angle patterns were obtained for the systems DPL, DPL - Pyrene, DPL - 3, 4 - Benzpyrene and DPL - 1, 2, 5, 6 - Dibenzanthracene. These patterns were subsequently analyzed using the intensities of the lamelar reflections. The electron density distribution normal to the plane of the bilayer w calculated, by Fourier synthesis, with a resolution of 7.5 Aº. In the case of Pyrene, the electron density map shows a slight perturbation of the structure in the hydrophobic region, close the glycerol group. This is in agreement with NMR experiments reported by Vanderkooi et al (J. Chem. Phys. 63, 3662, 1975). For the DPL - 3, 4 - Benzpyrene system the distortion of the central trough is similar to that observed for the DPL - Pyrene samples. Substituição de floculante comercial por biofloculante de Moringa oleífera Lamarck no setor sucroenergético /

Uma etapa fundamental é a clarificação do caldo, onde são adicionados polieletrólitos sintéticos a base de acrilamida, objetivando a rápida sedimentação de impurezas presentes no caldo de cana na produção de açúcar e etanol. Considerando-se que a molécula constituinte deste insumo pode ficar retida no cristal de açúcar e na levedura, sua utilização deve ser controlada, uma vez que apresenta ações cancerígenas e neurotóxicas ao ser humano. Um bioexrato que pode ser promissor e que apresenta ação coagulante, é o de semente de Moringa oleífera Lamarck. Neste contexto, o objetivo do trabalho foi comparar os reflexos do emprego de extrato de semente de moringa, com diversas concentrações do polímero à base de acrilamida comercial, como auxiliares de sedimentação na clarificação do caldo de cana, avaliando se ao final do processo ocorre a permanência de resíduos de acrilamida e a qualidade na produção de açúcar e etanol. O extrato de moringa (13mg/L), apresentou características tecnológicas e viabilidade fermentativa, semelhantes comparado ao polímero. O emprego de dosagens superiores a 1,5mg/L de polímero, acarreta redução dos teores de açúcares. Para produção de açúcar o extrato de semente de moringa, apresentou as características dentro do estabelecido para o açúcar VHP, sendo que a pol, a umidade e a cor foram similares ao polímero (1,5mg/L). A key step is the sugar cane juice clarification, where synthetic acrylamide based polyelectrolytes are added, aiming a rapid sedimentation of impurities present in sugarcane juice, for sugar and ethanol production. Considering that the constituent molecule of this input can be retained in the sugar crystal and in the yeast, its use must be controlled, since it presents carcinogenic and neurotoxic actions to the human being. A bioextract that may be promising and exhibits coagulant action is the one of Moringa oleifera Lamarck seed. In this context, the objective of this study was to compare the use of Moringa seed extract with various concentrations of the commercial acrylamide based polymer as sedimentation aids, in the clarification of sugarcane juice, evaluating at the end of the process the permanence of acrylamide residues and the quality of sugar and ethanol production. The moringa extract (13mg/L) presented technological characteristics and fermentative viability similar to the polymer. The use of dosages higher than 1.5mg/L of polymer, leads to reduction of sugar content. For sugar production, the Moringa seed extract presented characteristics within the established for the VHP sugar, being Pol, humidity and color similar to the polymer (1.5mg/L). The Moringa seed extract presents potential for its use as a flocculant, being equivalent to the acrylamide polymer of 1.5 mg/L. By quantifying the contents of acrylamide in the sugar produced and in the yeast separated from the fermentation process, it was found that the residues of this biomolecule were below the limit of detection. / Mestre

Aplicando-se a técnica AgNOR, o objetivo principal foi correlacionar as análises quantitativa (contagem) e qualitativa (morfológica) dos pontos nucleares prata-positivos em células uroteliais de casos controles e neoplásicos, de baixo e alto grau de malignidade. Este estudo compreendeu setenta (70) exames cito lógicos pareados com seus respectivos histopatológicos de carcinomas uroteliais primários vesicais (n=44) e de controles (n=26). As avaliações citológicas foram realizadas em lâminas de sedimento urinário, previamente coradas pela hematoxilina-eosina, descoradas, impregnadas pela prata coloidal, através da técnica AgNOR (PLOTON et aI., 1986) e analisadas com microscopia óptica comum, em objetiva de 40x. Na citologia, os casos positivos para células neoplásicas foram classificados conforme a graduação citológica de KOSS (1992) em: tumores de baixo grau de malignidade (n=19), correspondendo aos graus 1 e 1-2 da classificação histológica de KOSS e tumores de alto grau (n=25), correspondendo aos graus 2, 2-3, e 3. Houve significância estatística entre as diferenças das médias de pontos contados nos casos de neoplasia e nos controles (p<0,001- teste de Mann- Withney) e entre os três grupos juntos (p<0,001- teste de Kruskal-Wallis). Na interseção das curvas da distribuição dos pontos AgNORs, no grupo controle no tumoral (valores logaritmados), determinamos o valor crítico de corte de 2,96 pontos AgNORs. A especificidade da técnica é 96,29%, a sensibilidade é 97,77%, e a acurácia, 97,22%. Pela morfologia das AgNORs há diferença significante entre os casos tumorais e os controles (p<0,001) e entre os de alto e baixo grau de malignidade (p=0,001), aplicando-se dois (2) testes de Fisher. Além disso, a análise discriminatória, pelo método de Jackknive, classifica corretamente 84% dos casos com as variáveis independentes: número médio de AgNORs, o diferencial da variação máxima e mínima de AgNORs para cada caso e o padrão morfológico (homogêneo e heterogêneo). As variáveis dependentes foram: ausência de neoplasia, neoplasia de baixo grau e neoplasia de alto grau. Concluímos, portanto, que a técnica AgNOR, em citologia oncótica urinária, é auxiliar no diagnóstico diferencial das lesões ~esicais, usando-se os critérios quantitativo e qualitativo / Abstract: The oncocytic urinary cytology is a reliable toeI in diagnosis and folow-up of bladder tumor treatment as well as in the research of more agressive forms of bladder cancer. According to the literature it is of great value especially in patients who continue shedding identifiable cancer cells in urinary material after treatment. They are at very high risk for development of invasive bladder cancer (Koss, 1992). AgNOR technique may be helpful for the cytological differential diagnosis of bladder urothelial tumors, especially between those with low malignancy degree and the reactive urothelial cells. This study consisted of paired cytohistological examinations of patients with (n=44) and without (n=26) primary vesical urotelial carcinoma. Evaluations were performed with urinary sediment slides, previously stained by hematoxylin-eosin, unstained, impregnated by colloidal silver (AgNOR technique) and analyzed under common optic microscope (40 x). Control (non neoplastic) and neoplastic cases with low and high malignancy degree were investigated. Our principal aim was to correlate quantitative (counting) and morphological qualitative (subjective) analyses of silver-positive nuclear dots in both cases. The averages of the counted dots showed a statistically significant difference in both cases (p<0, 001 Mann-Withney's test). The grafic of AgNORs distribution of the neoplastic cases is similar to a logarithmic curve (normal log). The critical cut-off value of 2,96 AgNORs dots was determined in the intersection of the distribution curves of AgNOR dots in contrQI, and tumoral groups. There was 2,9% of falses negatives cases below this value and 2,9% of falses positives above it. AgNOR morphology was classified into homogeneous and' heterogeneous pattern. Control group presented the former in all cases. In the neoplastic group with low malignancy degree, 15/19 cases had the homogeneus pattern and in those with high degree, 15/25 had the heterogeneous pattern. According to AgNOR morphology, there are significant differences between tumoral and' control cases (p<0,001) and between those with low and high malignancy degree (p=0,001-Fisher's tests).Thus, AgNOR technique can be useful to aid in the differential diagnosis in oncotic urinary cytology by using qualitative and quantitative criteria / Mestrado / Anatomia Patologica / Mestre em Ciências Médicas

Para estudar o envolvimento dos genes NAT2 em tumores na população brasileira da região de Campinas, avaliamos seis polimorfismos (C282T; T481C; G590A; A803G; G857A; G191A) em câncer de tireóide e quatro (T481C; G590A; A803G e G857A) em câncer de próstata. Em um estudo de caso-controle, comparamos 126 indivíduos com câncer de próstata com 101 indivíduos controles com hiperplasia prostática benigna pareados para idade e condições de dieta e exposição ambiental; 139 indivíduos com câncer de tireóide (112 CP e 27CF) e 179 controles também adequadamente pareados. A análise foi feita pela extração de DNA com base no sangue periférico, PCR e restrição enzimática. Os polimorfismos T481C (76.24%) e A803G (59.41%) apareceram com maior freqüência entre os pacientes controles com hiperplasia prostática benigna do que nos com câncer da próstata (60.32% e 45.60%, p=0.0152 e 0.0186, respectivamente). Ao contrário, G857A foi mais freqüente entre os pacientes com câncer da próstata (18.4%) do que nos controles com hiperplasia prostática benigna (5.94%; p=0.0044). Assim, a presença do polimorfismo NAT2T481C reduziu o risco de câncer da próstata (OR=2.115; 95% C.I=1.155-3.872). Da mesma forma, a presença do polimorfismo NAT2A803G reduziu o risco de câncer da próstata (OR=1.973; 95%C.I=1.120-3.474; p=0.0186). Ao contrário, a presença do polimorfismo G857A aumentou o risco para câncer da próstata mais de quatro vezes (OR=4.095; 95%C.I=1.551-10.812). A presença de um fenótipo de acetilação lenta aumentou o risco para câncer de próstata em 24 vezes (OR=24.145;95%CI=1.416-411.63). Nos carcinomas da tireóide, observamos que mutações pontuais de tipo A803G apareceram com maior freqüência entre os casos de carcinomas (46.76%) do que nos controles (31.84%) (p=0.0069), enquanto que mutações pontuais de tipo G191A e C282T foram mais freqüentes nos controles (25,70% e 68.16% dos casos, respectivamente) do que nos casos (5,04% e 33,81%, respectivamente) (p=0,0001). Assim, a herança do polimorfismo A803G representou um risco de 1.8 vezes maior de desenvolver carcinoma diferenciado da tireóide (OR= 1.880; 95% IC= 1.189-2.973). Por outro lado, a herança dos polimorfismos G191A e C282T do gene NAT2 representou um fator de proteção de cerca de 80,6% contra o risco de desenvolver um carcinoma diferenciado da tireóide (OR=0.153; 95% IC=0.067-0.352 e OR=0.239; 95% IC=0.149-0.382, respectivamente). Em conclusão, nossos dados mostram que polimorfismos do gene NAT2 estão associados ao risco para o desenvolvimento tanto dos tumores de próstata quanto de tireóide, podendo vir a ser importantes marcadores de susceptibilidade para tais doenças em nossa população / Abstract: N-acetyltransferases (NAT), which are polymorphic in the population, metabolize important carcinogens such as different kinds of meat and tobacco products that have been directly implicated in the tumor initiation process. In order to investigate the role of NAT2 polymorphisms in the susceptibility to cancer in the Brazilian population from our region, we studied 6 polymorphisms (C282T; T481C; G590A; A803G; G857A; G191A) in differentiated thyroid cancer and 4 polymorphisms (T481C; G590A; A803G e G857A) in prostate cancer. We conducted a case-control prospective study comparing 126 prostate cancer to 101 benign prostatic hyperplasia patients paired for age, diet and environmental exposure; 139 thyroid cancer patients (112 papillary carcinomas and 27 follicular carcinomas) and 179 paired controls. Analyses were performed in DNA extracted from peripheral blood using the polymerase chain reaction-based restriction fragment length polymorphism method. We observed T481C (76.24%) and A803G (59.41%) polymorphisms with higher frequency among control patients than in prostate cancer cases (60.32% e 45.60%, p=0.0152 e 0.0186, respectively). On the contrary, G857A polymorphisms was more frequent among prostate cancer patients (18.4%) than in the benign hyperplasia control partients¿ group (5.94%; p=0.0044). Therefore, the presence of NAT2T481C polymorphism reduced the risk of prostate cancer (OR=2.115; 95% C.I=1.155-3.872). Likewise, the presence of NAT2A803G reduced the risk of prostate cancer (OR=1.973; 95%C.I=1.120-3.474; p=0.0186). On the contrary, the presence of G857A increased the risk for prostate cancer more than 4 times (OR=4.095; 95%C.I=1.551-10.812). The presence of a low acetylation phenotype increased the risk for prostate cancer more than 24 times (OR=24.145;95%CI=1.416-411.63). Regarding thyroid cancer, we observed that point mutations like A803G appears more frequently among thyroid carcinomas (46.76%) than in controls (31.84%) (p=0.0069), while G191A and C282T polymorphisms were more frequent among controls (25,70% and 68.16% of the cases, respectively) than among thyroid cancers (5,04% e 33,81%, respectively) (p=0,0001). Therefore, the inheritance of an A803G polymorphism represents an 1.8 times higher risk to thyroid cancer development (OR= 1.880; 95% IC= 1.189-2.973). On the other hand, the inheritance of G191A and C282T NAT2 polymorphisms represents a protection around 80,6% against the risk of thyroid cancer development (OR=0.153; 95% IC=0.067-0.352 and OR=0.239; 95% IC=0.149-0.382, respectively). In conclusion, our data demonstrate that NAT2 gene polymorphisms are associated to the risk to both prostate and thyroid cancer, suggesting they could become useful molecular markers of susceptibility to these tumors in our population / Mestrado / Clinica Medica / Mestre em Clinica Medica

Lupinus albus (L. albus), conhecido popularmente como tremoço branco, vem sendo usado na medicina popular, em forma de sementes cruas, como regulador de níveis de glicemia e colesterol. Embora descrito na literatura que suas proteínas e alcaloides isolados têm potencial para isto, não se tem qualquer informação científica em relação à eficiência de suas sementes cruas. O objetivo deste trabalho foi avaliar os possíveis efeitos, citotóxicos, genotóxicos e antigenotóxicos, mutagênicos e antimutagênicos, carcinogênicos e anticarcinogênicos e também o potencial imunomodulador do extrato aquoso de sementes cruas de L. albus, por meio de ensaios de micronúcleo em medula óssea, Focos de criptas aberrantes em cólon, contagem diferencial de leucócitos, níveis de glicemia e colesterol do soro de camundongos da espécie Mus musculus, bem como determinar e quantificar, por meio de cromatografia gasosa, alcaloides e ácidos graxos desse extrato. Foi avaliado nos camundongos seus efeitos nas concentrações equivalente a 3 sementes/dia (como utilizado na medicina popular) e 9 sementes/dia. A análise fitoquímica do extrato revelou a presença do alcaloide lupanina, dos ácidos graxos oleico, linoleico, palmítico e esteárico. Porém não foi possível observar, por meio de ensaios bioquímicos, os efeitos hipoglicêmicos de L. albus em ambos os protocolos seguidos. Foi observado que os níveis de colesterol apresentaram melhora, para o protocolo com administração da DMH. Lupinus albus (L. albus), popularly known as white lupine, has been used in popular medicine in form of raw seeds, as a regulator of blood glucose and cholesterol. Although literature describes that their isolated proteins have the this potential, there is no scientific information regarding the efficiency of their raw seeds. The aim of this study was to evaluate the possible genotoxic, antigenotoxic, mutagenic, antimutagenic, anticarcinogenic and carcinogenic effects, and also the immune modulatory potential of aqueous extract of raw seeds of L. albus, by means of bone marrow micronucleus, aberrant crypt foci in colon, leukocyte counting, blood glucose and serum cholesterol levels of mice Mus musculus, as well as to determine and quantify alkaloids and fatty acids of these extracts, by gas chromatography. The effects of concentrations equivalent to 3 seeds/day (as used in popular medicine) and 9 seeds/day were evaluated in mice. Chemical analysis of the extract showed the presence of the alkaloid lupanine and oleic, linoleic, palmitic and stearic fatty acids. However, according to biochemical assays, it was not possible to observe hypoglycemic effects of L. albus in both followed protocols. It was observed that cholesterol levels showed an improvement in the protocol using DMH. No cytotoxic effects were observed in the MN assay in bone marrow, but genotoxic and mutagenic effects were recorded in highest concentration of the tested extract. Antigenotoxic and antimutagenic effects were also observed, demonstrating a preventive action for the two concentrations tested due to the promotion of harm reduction for pre-extract treatment. O MTT mostrou que o selênio não foi tóxico em qualquer uma das concentrações testadas. O ensaio de proliferação celular mostrou que o selênio não interfere com a proliferação celular nas três concentrações testadas. Em contraste, quando as três concentrações foram combinadas com a doxorrubicina, uma diminuição significativa na taxa de proliferação celular foi observada. A taxa de apoptose foi significativamente maior no selênio (500 ng/mL) e doxorrubicina. A expressão CASP9 foi aumentada e expressão BCL-XL diminuiu no selênio (500 ng/mL) e o grupo de doxorrubicina. HT29 cells were cultured and treated with selenium at concentrations of 5, 50 and 500 ng/mL with or without the damage-inducing agent doxorubicin. These cells were then evaluated for cytotoxicity (MTT), cell proliferation and in situ apoptosis induction. To evaluate gene expression, only the cells treated with 500 ng/mL of selenium were used. RNA was extracted from these cells, and the expressions of CASP9, BCL-XL and APC were analyzed by the RT-PCR method. The GAPDH gene was used as a reference gene.
