Efecto del ibuprofeno y celecoxib sobre la fertilidad de ratas hembras

Objetivo: Determinar si el Ibuprofeno y Celecoxib disminuyen la fertilidad de ratas hembras. Diseño: Estudio experimental. Lugar: Bioterio de la Facultad de Medicina de la Universidad Nacional Mayor de San Marcos. Material biológico: Ratas Sprague-Dawley adultos de 3 – 4 meses de edad, con peso 150 ±20 g. no preñadas. . Métodos: Se realizó el seguimiento del ciclo estral del total de ratas, y se distribuyó aleatoriamente, en 3 grupos: Grupo Control (n=7): Suero fisiológico 0.1ml/100g; Grupo Ibuprofeno (n=7): Ibuprofeno 120 mg/kg/12h; Grupo Celecoxib (n=8): Celecoxib 20 mg/kg/12h. El tratamiento fue iniciado en diestro y se reunieron con los machos en estro del mismo ciclo, solo hasta cumplir 10 días. Luego se mantuvieron 10 días sin tratamiento y solas, al concluir 20 días fueron eutanizadas y expuestos los úteros. Principales Medidas de Resultados: se usó el porcentaje gestación y se evaluó el número de fetos a través de la media y el porcentaje de reducción. Resultados: En la evaluación de % de gestación, el ibuprofeno tuvo 28.6% y celecoxib 62.5% que fueron menores en comparación con el control 85.7%; en la evaluación del numero de fetos, el % de reducción fue en el ibuprofeno 76.2%, y en el celecoxib 59.9%, en relación al grupo control. Sin diferencia significativa entre los grupos ibuprofeno y celecoxib, tanto en el porcentaje de gestación como en el número de fetos. Conclusión: La administración por vía intragástrica de Ibuprofeno 120 mg/kg/12h y celecoxib 20 mg/kg/12h modifica negativamente la fertilidad en ratas hembras normales. No se encontró diferencia significativa entre Ibuprofeno 120 mg/kg/12h y celecoxib 20 mg/kg/12h sobre la fertilidad en ratas hembras normales. Main Outcome Measures: We used the pregnancy rate and number of fetuses evaluated through the average and the percentage reduction. Results: The evaluation of % of gestation, ibuprofen was 28.6% and celecoxib was 62.5%, who were under the control compared with 85.7% in the assessment of the number of fetuses, the % reduction was 76.2% for ibuprofen, and 59.9% for celecoxib compared to the control group. No significant difference between ibuprofen and celecoxib groups, both in the pregnancy rate and the number of fetuses. Conclusion: The intragastric administration of ibuprofen 120 mg/kg/12h and celecoxib 20 mg/kg/12h adjust negatively normal fertility in female rats. O esclarecimento do papel de prostaglandinas na reabsorÃÃo Ãssea periodontal tem contribuÃdo para a utilizaÃÃo mais racional de fÃrmacos inibidores das ciclooxigenases disponÃveis no mercado. Nesse sentido, torna-se necessÃrio o estudo da seguranÃa, nÃo obstante sua eficÃcia, destes agentes antiinflamatÃrios nÃo esteroidais, como celecoxib ou diclofenaco potÃssico. No presente estudo, utilizou-se um modelo de periodontite induzida por corpo estranho em ratos descrito na literatura, com o objetivo de avaliar a capacidade do celecoxib e do diclofenaco potÃssico. Observou-se que a inserÃÃo cirÃrgica do fio de nÃilon no modelo estudado induziu a perda Ãssea alveolar de forma significante aos 11 dias. Celecoxib nas doses diÃrias de 3, 9 e 27 mg/kg foi capaz de inibir a perda Ãssea alveolar de forma significante e dose-dependente (64%, 53% e 75,4%, respectivamente). O resultado com diclofenaco nas doses de 1 e 5 mg/kg tambÃm reduziu de forma significante, a perda Ãssea alveolar (41% e 54,5%, respectivamente). A anÃlise macroscÃpica dos estÃmagos mostrou que celecoxib e diclofenaco nÃo promoveram lesÃes gÃstricas de forma significante quando comparados aos animais nÃo tratados. O uso de celecoxib nÃo causou alteraÃÃes, de forma significante, no hemograma dos animais submetidos à periodontite. Com diclofenaco, verificou-se uma leucocitose em decorrÃncia do aumento do nÃmero de neutrÃfilos, com maior pico no perÃodo compreendido entre o 7 e 11 dia apÃs o procedimento cirÃrgico. A nÃo alteraÃÃo do nÃmero de plaquetas do sangue dos animais submetidos à periodontite, inclusive nos animais que receberam celecoxib ou diclofenaco pode sugerir que as doses utilizadas foram relativamente seguras sob o ponto de vista cardiovascular ou ainda, possivelmente, tais alteraÃÃes nÃo foram vistas por nÃo se tratar de um estudo prolongado. Tanto o celecoxib como o diclofenaco nÃo foram capazes de reverter, de forma significante, a perda de massa corpÃrea. As maiores doses de diclofenaco (10 e 25 mg/kg) causaram reduÃÃo significante da taxa de sobrevida dos animais a partir do primeiro dia pÃs-operatÃrio e atingindo 50% dos animais no terceiro dia. Os resultados mostraram que a induÃÃo da periodontite em ratos no grupo controle e no grupo tratado com celecoxib, nÃo alterou as funÃÃes renais ou hepÃticas avaliadas (urÃia e creatinina ou AST). Contudo, o uso de diclofenaco, tanto na dose de 1 mg/kg como tambÃm na dose de 5 mg/kg determinou alteraÃÃes em ambas as funÃÃes consideradas, induzindo um aumento significativo nos nÃveis sÃricos de creatinina e AST. O diclofenaco e o celecoxib apresentaram efeitos protetores semelhantes na perda Ãssea. O celecoxib, utilizado na periodontite induzida durante 11 dias, foi menos tÃxico que o diclofenaco, uma vez que este causou maior mortalidade de forma dose-dependente, e alteraÃÃes ao nÃvel de leucograma e das funÃÃes renal e hepÃtica / Periodontitis is an inflammatory disease of the tissue that supports the teeth. It is caused by microorganisms and is characterized by leukocyte infiltration, progressive destruction of the periodontal ligaments, and alveolar bone. The clear role of prostaglandins on periodontal bone resorption has contributed to the rational use of the cyclooxygenase inhibitors drugs available. In this sense, it becomes necessary to bare safety and efficacy studies of the non-steroidal anti-inflammatory agents, such as celecoxib and potassium diclofenac. For the present study, a foreign object induced periodontitis model in rats was used, such as described on specific literature, to evaluate the activity of celecoxib and potassium and diclofenac. A surgical insertion of a nylon tread induced significant alveolar bone loss after 11 days. Celecoxib, given daily at 3, 9 and 27 mg/kg, significantly reduced this loss in a dose-dependent manner (64%, 53% and 75.4%, respectively). Diclofenac produced a similar effect at 1 and 5 mg/kg, reducing the loss by 41% and 54.5%, respectively. Macroscopic analyses of stomachs indicated that neither celecoxib nor diclofenac promoted gastric lesions when compared with non-treated animals. Celecoxib-treated rats did not show significant hemogram parameters alterations when subjected to periodontitis. For diclofenac-treated animals, it was verified a leukocytosis due to the augmentation of neutrophil count, which peaked between the 7th and the 11th day post-surgery. Platelet number of periodontitis-subjected animals, including those that received celecoxib or diclofenac treatment, was not altered, which may suggest that the doses used are relatively safe from the cardiovascular point of view, or that this alterations was not seen due to the short period of the study. Celecoxib and diclofenac were not able to significantly reverse the loss of body mass. The higher doses of diclofenac (10 and 25 mg/kg/day) significantly reduced the survival rate since the first day after surgery, reaching 50% at day 3. The induction of periodontitis in control and celecoxib-treated rats did not alter renal or hepatic function according to the biochemical parameters evaluated (urea and creatinine or AST). However, diclofenac, at 1 and 5 mg/kg/day, determined alterations in both kidney and liver functions, with a significant increase of seric levels of creatinine and AST. Diclofenac and celecoxib presented similar effects on bone loss prevention. Celecoxib, used for 11 days to induce periodontitis, was less toxic than diclofenac, which caused a dose-dependent mortality and leukogram alterations along with disruption of renal and hepatic functions

A dexametasona e indometacina foram usados como drogas controle. De acordo com os resultados obtidos nos dois modelos de inflamação aguda induzida pela carragenina em ratos, o tratamento dos animais com os inibidores seletivos da COX-2, celecoxib e rofecoxib, ao contrário do que foi observado para a indometacina e a dexametasona, indicam que os inibidores da COX-2 estudados apesar de reduzirem significativamente os níveis de PGE2 do exsudato pleural, a formação de edema na pleura e na pata tratadas com carragenina, seus efeitos antiinflamatórios são limitados.

Schicksal von Mikrokernen bzw. mikrokernhaltigen Zellen und Bedeutung von Mikrokernen als Biomarker / Fate of micronuclei and micronucleated cells along with relevance of micronuclei as biomarker

Mikrokerne sind als wichtiger Biomarker in der Gentoxizitätsforschung seit langer Zeit etabliert und ihre Bildung ist mechanistisch gut verstanden, wohingegen das Mikrokernschicksal und die genaue Funktion von Mikrokernen in der Kanzerogenese unzureichend erforscht sind. Um das Schicksal von Mikrokernen und mikrokernhaltigen Zellen über einen längeren Zeitraum zu untersuchen, wurden HeLa-Zellen, die mit einem GFP-markierten Histon H2B transfiziert worden sind, mittels Lebendzellmikroskopie nach Behandlung mit verschiedenen gentoxischen Agenzien für 96 h untersucht. Parameter wie die Mitose- oder Zelltodrate wurden dabei ebenso wie das Schicksal der Mikrokerne dokumentiert. Während Persistenz und Reinkorporation von Mikrokernen häufig beobachtet wurden, waren Degradation und Auswurf von Mikrokernen selten bis gar nicht zu sehen. Auch konnte ein Teil der mikrokernhaltigen Zellen über mehrere Zellteilungen persistieren und proliferieren, wodurch die in Mikrokernen manifestierte chromosomale Instabilität unverändert bleiben kann. Ein eindeutiger Substanzeinfluss auf das Mikrokernschicksal konnte nicht ausgemacht werden. Extrusion sollte weiterhin durch Behandlung mit Hydroxyurea oder Cytochalasin B in Kombination mit gentoxischer Behandlung induziert werden, es wurde jedoch kein Effekt auf die Extrusionsrate beobachtet. Degradation wurde mittels γH2AX-Antikörperfärbung und transduziertem dsRed-markierten Autophagiemarker LC3B in HeLa-H2B-GFP-Zellen untersucht. Trotz erhöhter DNA-Degradation in Mikrokernen wurde nur selten eine Ko-Lokalisierung mit LC3B beobachtet. Dafür gab es in HeLa-H2B-GFP-Zellen, die zusätzlich mit dsRed markierten Kernmembranmarker Lamin B1 transduziert worden sind, Anzeichen für eine eingeschränkte Mikrokernmembranintegrität. Weiterhin wurden Zytokinese-Block Mikrokerntests nach Behandlung mit Thebain mit und ohne metabolische Aktivierung sowie Celecoxib und Celecoxibderivaten durchgeführt. Hierbei wurde nach Thebainbehandlung nur ohne metabolische Aktivierung und bei Anwesenheit von Zytotoxizität mehr Mikrokerne gefunden, während nach Behandlung mit Celecoxib und Celecoxibderivaten kein Anstieg beobachtet wurde. Zusätzlich wurde der Einfluss durch neurodegenerative Veränderungen auf Mundschleimhautzellen in zwei großen Kohorten untersucht, wobei keine Effekte auf die Häufigkeit von Mikrokernen oder mikrokernhaltigen Zellen zugeordnet werden konnten, während es teilweise bei Parametern, die auf Zytotoxizität hindeuten, zu Veränderungen kam. Es konnte insgesamt gezeigt werden, dass Mikrokerne und mikrokernhaltige Zellen zusätzlich zu ihrer Funktion als Biomarker über wenigstens mehrere Zellteilungen bestehen bleiben können. Auf diese Weise können sie z. B. über Chromothripsis zu einer beschleunigten Kanzerogenese führen, was zu einer schlechten Prognose für Krebspatienten führen kann. / Micronuclei have been established as biomarkers long ago in genotoxicity research and although formation is mechanistically well understood, the fate of micronuclei as well as micronuclei function in carcinogenesis are poorly investigated. To explore the long-term fate of micronuclei and micronucleated cells, HeLa cells transfected with GFP-tagged histone H2B were examined via live cell imaging after treatment with various genotoxic agents for 96 h. Parameters like mitosis or cell death rate as well as the development of micronuclei were analysed. While persistence and reincorporation of micronuclei were frequently observed, degradation and extrusion of micronuclei were found only rarely or never. A subset of micronucleated cells could persist and proliferate over multiple cell divisions, so that micronuclei as manifestation of chromosomal instability also persisted. No clear substance-related effect on micronucleus fate could be determined. Extrusion of micronuclei was furthermore investigated after treatment with hydroxyurea or after combination treatment with cytochalasin B and a genotoxic agent, but no effect on the extrusion rate was observed. Degradation of micronuclei was investigated in detail by γH2AX antibody staining and transduction of dsRed-tagged autophagy marker LC3B in HeLa-H2B- GFP-cells. Although enrichment of DNA degradation in micronuclei were identified, co-localisation with LC3B was observed only rarely. In HeLa-H2B-GFP-cells transduced with dsRed-tagged nuclear membrane marker lamin B1, evidence for impaired nuclear membrane integrity was found. In addition, cytokinesis-block micronucleus tests after treatment with thebaine with and without metabolic activation as well as celecoxib and celecoxib derivatives were conducted. After thebaine treatment, increased micronucleus frequency was only observed without metabolic activation and together with cytotoxicity, while treatment with celecoxib and celecoxib derivatives caused no effect on micronucleus frequency. Effects of neurodegenerative disease on DMA damage in buccal cells of two large cohorts were furthermore investigated, but no effect on the frequency of micronuclei and micronucleated cells was observed, while parameters indicating cytotoxicity showed alterations. All in all, it could be demonstrated, that micronuclei and micronucleated cells, apart from their role as biomarker, can persist for multiple cell divisions. This way, cancerogenesis can be accelerated e.g. via chromothripsis, potentially leading to poor prognosis for cancer patients.

Efeito do acetaminofeno e do celecoxib na distalização de incisivos e na ativação de regiões cerebrais relacionadas à nocicepção durante a movimentação ortodôntica em ratos / Effect of actaminophen and celecoxib in the incisor distalization and neuronal activation of nociceptive area of rat

Antiinflamatórios podem diminuir a movimentação dentária (MD) em pacientes ortodônticos, fato este que leva à existência de poucas alternativas terapêuticas para o tratamento da dor. Acetaminofeno tem sido a medicação de escolha, mas não é tolerado por alguns pacientes. Este trabalho pesquisou a influência do celecoxib e do acetaminofeno na separação ortodôntica dos incisivos centrais superiores de ratos, assim como na ativação de estruturas relacionadas à nocicepção no núcleo espinhal do trigêmio, por meio da expressão de c-fos nos subnúcleos caudalis, interpolaris e oralis. Num primeiro momento, o peso do animal e a força ortodôntica foram variados, quando foi possível encontrar o relacionamento ideal entre estas duas variáveis, o qual permitiu movimentação dentária sem expansão da sutura palatina mediana. A aplicação de 35 g de força em animais de 400 g de peso distalizou os incisivos sem expandir a sutura palatina mediana, como pôde ser verificado em radiografias da maxila dos animais. Sendo assim, trinta ratos Wistar, machos, pesando cerca de 400 g, foram pré-tratados através de sonda gástrica (1 ml) com acetaminofeno (80mg/ml), celecoxib (20mg/ml) ou veículo (carboximetilcelulose 0,4%). Após 30 minutos, foram submetidos à movimentação dentária (35 g de força) ou deixados como controles. Ração moída foi fornecida e a cada 12 horas eles receberam, na mesma dosagem anterior, um dos medicamentos ou veículo. Depois de 48 horas, foram novamente anestesiados, fotografados e perfundidos com paraformaldeído 4%. Na seqüência, a maxilla foi radiografada e os cérebros removidos e processados para a imunohistoquímica para c-fos. A movimentação dentária induziu a distalização dos incisivos (p< 0.05), mas a distância inter-incisal não foi modificada pelos medicamentos. Adicionalmente, a movimentação dentária e o tratamento com as drogas não causaram qualquer alteração no número de neurônios Fos positivos nos subnúcleos interpolaris e oralis. No subnúcleo caudalis, entretanto, as duas drogas reduziram o número de neurônios Fos positivos devidos ao estímulo ortodôntico (p<0,05). Concluímos que os dois medicamentos reduzem a ativação neuronal de região nociceptiva e não afetam a movimentação dentária, sendo que o celecoxib seria uma possível alternativa ao acetaminofeno para o alívio de dor em pacientes ortodônticos, nos casos em que este for contra indicado. / Anti-inflammatory drugs may slow dental movement in orthodontic patients, which leads to low therapeutic alternatives for pain control. Acetaminophen has been the first choice medication for the majority of patients, but it is not well tolerate for some. This work evaluated the influence of celecoxib and acetaminophen in the orthodontic separation of superior central incisor of rats, such as in the activation of nociceptive related structures in the spinal trigeminal nucleus, using c-fos expression in caudalis, interpolaris, and oralis sub nuclei. In a first moment, animal weigh and orthodontic force were varied, when it was possible to find the ideal relationship between these two variables, resulting in dental movement without median palate suture expansion. The application of 35 g of orthodontic force in animals weighing 400 g has been establish as the ideal experimental model, and this way, 30 male Wistar rats were pre-treated, by oral gavage (1 ml), with acetaminophen (80 mg/ml), celecoxib (20 mg/ml), or vehicle (carboxymethylcellulose 0,4%), and 30 minutes later, they were submitted or not to orthodontic stimulus. Animals received ground food, and one of the drugs or vehicle, every 12 hours. After 48 hours from the dental manipulation, they were anesthetized, photographed, and perfused with 4% paraformaldehyde. In the sequence, maxila was radiographed and brains were removed and processed for FOS immunocytochemistry. Dental movement induced incisor distalization (p<0,05), but interincisal gap was not modified by the drugs. Furthermore, no changes were found in FOS positive neurons in oralis and interpolaris subnuclei, following tooth movement, independently of whether medications were given or not. In caudalis subnucleus, however, both analgesics reduced the number of tooth movement induced FOS positive neurons. We conclude that neither celecoxib nor acetaminophen seems to affect tooth movement, but both may reduce pain caused by orthodontic appliance. Celecoxib, thus, may be a therapeutic alternative to acetaminophen when the latter was counter indicated.

Análise da expressão da proteína pAKT em cultura de células de carcinomas epidermoides de cabeça e pescoço tratadas com curcumina e celecoxib / Expression analysis of pAKT in cultured cells of head and neck squamous cell carcinomas treated with curcumin and celecoxib

Diversas alterações genéticas estão associadas à patogênese do carcinoma epidermoide (CE), a neoplasia maligna mais comum de cabeça e pescoço. Algumas dessas alterações comprometem proteínas pertencentes à via de sinalização do AKT, envolvida em diferentes fenômenos celulares. Este projeto teve como objetivo estudar a expressão da proteína pAKT em linhagens celulares de carcinomas epidermoides de cabeça e pescoço, de forma a verificar possíveis alterações na expressão dessas moléculas em células de CE tratadas com Curcumina e Celecoxib. Foram utilizadas duas linhagens celulares de CE de cabeça e pescoço e uma linhagem de queratinócitos que foram divididas em quatro grupos: a. grupo controle não tratado; b. células tratadas com Curcumina, c. células tratadas com Celecoxib d. células tratadas com Curcumina e Celecoxib. A proliferação celular foi monitorada através do teste de viabilidade celular. A análise da expressão da proteína foi realizada através da técnica do Western Blot. Desta forma, foi possível entender em maior profundidade a ação do Celecoxib, da Curcumina e sua associação na via do AKT em carcinomas epidermoides de cabeça e pescoço. A associação de Curcumina com Celecoxib foi o tratamento que apresentou melhores resultados na redução da viabilidade celular. A linhagem SCC9 não apresentou expressão de pAKT após o tratamento com Curcumina e com a associação desta com o Celecoxib. A linhagem HaCat apresentou maior estabilidade na expressão de pAKT nos dois tempos de tratamento no grupo tratado com a associação das substâncias. A combinaçãoo da Curcumina com o Celecoxib mostrou resultados satisfatórios e promissores. / Several genetic changes are associated with the squamous cell carcinoma\'s (SCC) pathogenesis, the most common malignant head and neck tumor. Some of these changes compromise proteins that belong to the AKT signaling pathway, involved in different cellular\'s phenomena. The aim of this project was to study the expression of pAKT in head and neck SCC cell lines in order to investigate possible alterations in these molecules\'s expression in cells treated with Curcumin and Celecoxib. Two head and neck SCC cell lines and a line of keratinocytes was divided into four groups: a. control group: untreated; b. cells treated with Curcumin, c. cells treated with Celecoxib d. cells treated with Curcumin and Celecoxib. Cell proliferation was monitored by cell viability test. Analysis of the protein\'s expression was performed using the Western blot technique. Then was possible to understand the action of Curcumin, Celecoxib and its association in the AKT pathway in head and neck SCC. Curcumin\'s association with Celecoxib was the treatment that showed best results in cell viability reduction. The SCC9 cell line not presented pAKT expression after the treatment with Curcumin and its association with Celecoxib. The cell line HaCat showed greater stability in pAkt expression during the treatment period with the association of substances. The association of Curcumin and Celecoxib showed satisfactory and promising results.

Efeito do acetaminofeno e do celecoxib na distalização de incisivos e na ativação de regiões cerebrais relacionadas à nocicepção durante a movimentação ortodôntica em ratos / Effect of actaminophen and celecoxib in the incisor distalization and neuronal activation of nociceptive area of rat

Antiinflamatórios podem diminuir a movimentação dentária (MD) em pacientes ortodônticos, fato este que leva à existência de poucas alternativas terapêuticas para o tratamento da dor. Acetaminofeno tem sido a medicação de escolha, mas não é tolerado por alguns pacientes. Este trabalho pesquisou a influência do celecoxib e do acetaminofeno na separação ortodôntica dos incisivos centrais superiores de ratos, assim como na ativação de estruturas relacionadas à nocicepção no núcleo espinhal do trigêmio, por meio da expressão de c-fos nos subnúcleos caudalis, interpolaris e oralis. Num primeiro momento, o peso do animal e a força ortodôntica foram variados, quando foi possível encontrar o relacionamento ideal entre estas duas variáveis, o qual permitiu movimentação dentária sem expansão da sutura palatina mediana. A aplicação de 35 g de força em animais de 400 g de peso distalizou os incisivos sem expandir a sutura palatina mediana, como pôde ser verificado em radiografias da maxila dos animais. Sendo assim, trinta ratos Wistar, machos, pesando cerca de 400 g, foram pré-tratados através de sonda gástrica (1 ml) com acetaminofeno (80mg/ml), celecoxib (20mg/ml) ou veículo (carboximetilcelulose 0,4%). Após 30 minutos, foram submetidos à movimentação dentária (35 g de força) ou deixados como controles. Ração moída foi fornecida e a cada 12 horas eles receberam, na mesma dosagem anterior, um dos medicamentos ou veículo. Depois de 48 horas, foram novamente anestesiados, fotografados e perfundidos com paraformaldeído 4%. Na seqüência, a maxilla foi radiografada e os cérebros removidos e processados para a imunohistoquímica para c-fos. A movimentação dentária induziu a distalização dos incisivos (p< 0.05), mas a distância inter-incisal não foi modificada pelos medicamentos. Adicionalmente, a movimentação dentária e o tratamento com as drogas não causaram qualquer alteração no número de neurônios Fos positivos nos subnúcleos interpolaris e oralis. No subnúcleo caudalis, entretanto, as duas drogas reduziram o número de neurônios Fos positivos devidos ao estímulo ortodôntico (p<0,05). Concluímos que os dois medicamentos reduzem a ativação neuronal de região nociceptiva e não afetam a movimentação dentária, sendo que o celecoxib seria uma possível alternativa ao acetaminofeno para o alívio de dor em pacientes ortodônticos, nos casos em que este for contra indicado. / Anti-inflammatory drugs may slow dental movement in orthodontic patients, which leads to low therapeutic alternatives for pain control. Acetaminophen has been the first choice medication for the majority of patients, but it is not well tolerate for some. This work evaluated the influence of celecoxib and acetaminophen in the orthodontic separation of superior central incisor of rats, such as in the activation of nociceptive related structures in the spinal trigeminal nucleus, using c-fos expression in caudalis, interpolaris, and oralis sub nuclei. In a first moment, animal weigh and orthodontic force were varied, when it was possible to find the ideal relationship between these two variables, resulting in dental movement without median palate suture expansion. The application of 35 g of orthodontic force in animals weighing 400 g has been establish as the ideal experimental model, and this way, 30 male Wistar rats were pre-treated, by oral gavage (1 ml), with acetaminophen (80 mg/ml), celecoxib (20 mg/ml), or vehicle (carboxymethylcellulose 0,4%), and 30 minutes later, they were submitted or not to orthodontic stimulus. Animals received ground food, and one of the drugs or vehicle, every 12 hours. After 48 hours from the dental manipulation, they were anesthetized, photographed, and perfused with 4% paraformaldehyde. In the sequence, maxila was radiographed and brains were removed and processed for FOS immunocytochemistry. Dental movement induced incisor distalization (p<0,05), but interincisal gap was not modified by the drugs. Furthermore, no changes were found in FOS positive neurons in oralis and interpolaris subnuclei, following tooth movement, independently of whether medications were given or not. In caudalis subnucleus, however, both analgesics reduced the number of tooth movement induced FOS positive neurons. We conclude that neither celecoxib nor acetaminophen seems to affect tooth movement, but both may reduce pain caused by orthodontic appliance. Celecoxib, thus, may be a therapeutic alternative to acetaminophen when the latter was counter indicated.
