A elevada densidade deste mosquito pode causar problemas secundários que têm impacto na saúde e qualidade de vida das pessoas expostas; por isso, o controle desta espécie é importante. O objetivo do trabalho foi identificar se existem e quais são os mecanismos de resistência a inseticidas químicos e a diversidade genética de populações naturais de Culex quinquefasciatus coletadas em PE. As avaliações foram feitas por meio de bioensaios, ensaios bioquímicos para mensurar a atividade das enzimas de detoxificação e ensaios moleculares para estimar a frequência da mutação presente no gene da acetilcolinesterase (ace-1), associada à resistência a organofosforados (OPs) e carbamatos (CMs). A diversidade genética das populações foi avaliada utilizando 16 marcadores microssatélites. Ovos de Culex foram coletados em seis municípios de PE: Olinda (Peixinhos e Alto da Conquista), Recife (Água Fria), Jaboatão dos Guararapes, Ipojuca, Glória do Goitá e Santa Cruz do Capibaribe. Os resultados mostraram que apenas a população proveniente de Santa Cruz do Capibaribe (SC) apresentou alteração na susceptibilidade (RR = 7,2 vezes), com nível de resistência moderado ao temephos, enquanto as outras populações analisadas não mostraram alteração na susceptibilidade ao inseticida. Quanto aos testes bioquímicos, apenas em SC foi observado um aumento na atividade de todas as esterases estudadas e indícios de insensibilidade da acetilcolinesterase. A frequência da mutação G119S no alelo de resistência ace-1 (G119S) foi de 0,11 em SC e zero nas outras populações. O número de alelos por locus variou entre 2 e 15. Alguns desses alelos foram exclusivos de algumas localidades. O percentual de variabilidade genética encontrado foi maior dentro das populações do que entre as populações e os resultados indicam excesso de heterozigotos. Estes dados mostram que embora não haja programas nacionais para o controle de Culex, esta espécie apresenta mecanismos de resistência que estão sendo mantidos em populações naturais como um resultado da exposição acidental ao inseticida, o que poderia prejudicar as ações de controle que podem ser implantadas no futuro.

And this study aims to identify the Culicidae fauna in a rural community in central Amazonia and their associations between human population density and deforestation , by checking the abundance and diversity in relation to the strata and collections environments. Collections were made with CDC light traps for six months between 2009-2010 to posteriore were made analysis on the Free Software R. were captured 3,113 specimens, the most abundant genera Culex species distributed with three eight mofótipos representing 2,408 individuals (79.8 % of total); Psorophora five species and 196 individuals (6.5% of total); in the farm environment (443 individuals) in DA_DA stratum; the second room with the largest number of mosquitoes caught was the forest (411 individuals) in DB_DB stratum.The diversity of species in each type of landscape showed that the stratum / environment collection with the highest diversity was DB_DBforest with (H '= 2.71), followed by DB_DAfarm (H' = 2.50) while the lowest rate was observed in stratum / DB_DAedge collection environment (H '= 1.64) and DA_DBedge (H' = 1.71). The results of this study showed that both deforestation, as human population density cause influence on the diversity and abundance of mosquitoes, and these effects may be higher in these populations when these two factors are associated. While deforestation can lead to a rapid increase in the number of specimens followed by a reduction in that number in human population density can produce favorable locations for maintenance cycle of several species of mosquitoes more adaptable. / Os mosquitos (Diptera: Culicidae), distribuídos por toda a zona tropical e parte das zonas temperadas, constituem-se num sério problema devido a sua atuação como vetores de diversos agentes etiológicos, causadores de doenças ao homem e aos animais, assim como grande complexidade no seu controle. E este estudo tem por objetivo Identificar a fauna de Culicídeos em uma comunidade rural da Amazônia central e respectivas associações entre a densidade populacional humana e desmatamento, através da verificação da abundância e diversidade em relação aos estratos e ambientes de coletas. Foram realizadas coletas com armadilhas luminosas CDC, durante seis meses entre 2009-2010 a posteriore foram feitas análise no software livre R. Foram capturados 3.113 espécimes, sendo os gêneros mais abundantes Culex distribuídos com três espécies e oito mofótipos representando 2.408 indivíduos (79,8% do total); Psorophora com cinco espécies e 196 indivíduos (6,5% do total); no ambiente roça (443 indivíduos) no estrato DA_DA; o segundo ambiente com o maior número de culicídeos capturados foi a floresta (411 indivíduos) no estrato DB_DB. A diversidade das espécies em cada tipo de paisagem mostrou que o estrato/ambiente de coleta que apresentou maior diversidade foi DB_DBfloresta com (H’=2,71), seguido pelo DB_DAroça (H’=2,50) enquanto o menor índice foi observado no estrato/ambiente de coleta DB_DAborda (H’=1,64) e DA_DBborda(H’= 1,71). Os resultados do presente trabalho mostraram que tanto o desmatamento, quanto a densidade populacional humana causam influência na diversidade e abundância de culicídeos, e esses efeitos podem ser maiores nessas populações quando esses dois fatores estão associados. Enquanto o desmatamento pode levar a um rápido aumento no número de espécimes seguido de uma redução nesse número, a densidade populacional humana pode produzir locais favoráveis para manutenção do ciclo de diversas espécies de culicídeos mais adaptáveis.

Assessing the Effect of Bacillus Thuringiensis Var. Israelensis on Nontarget Chironomidae Emergence

Bacillus-derived larvicides, which selectively target mosquito (Diptera: Nematocera: Culicidae) populations to reduce nuisance and health risks, were applied in the South March Highlands Conservation Forest near residential neighbourhoods in Ottawa, Ontario. The objective was to assess effects of application on the nontarget mosquito relative, Chironomidae (Diptera: Nematocera: Chironomidae), and other nontarget aquatic taxa captured using emergence traps. A secondary objective was to assess physicochemical variables that influence Chironomidae emergence. Study ponds received an application of Bacillus thuringiensis var. israeliensis, a subset also received an application of Bacillus sphaericus, and a group of control ponds were left untreated over 3 years (2016-2018). Weekly sampling included trap collections and measurements of water temperature, pH, water depth, conductivity, dissolved oxygen, ammonia, nitrate, and sulphate. Drought in 2016, high precipitation throughout 2017, and seasonal precipitation in 2018 influenced variable physicochemical conditions. Principal component analyses identified differences between sampling groups and between years. Redundancy analyses correlated insect emergence with pond pH, average water depth and water temperature and indicated a strong relationship between Chironomidae emergence and average water depth. Although significantly less Chironomidae annual emergence was observed at treated sites in 2017 and 2018, zero-inflated negative binomial generalized linear mixed modelling failed to detect a significant Bti treatment effect when controlling for within group variation. Rather, variations in pH, mean water depth and water temperature were identified as drivers of Chironomidae emergence. Culicidae emergence was reduced to zero briefly following treatment in 2017 and 2018. The model detected a marginal negative treatment effect on Culicidae in 2017 only, and a positive treatment effect in 2018 at the onset of a secondary hydroperiod, in the absence of treatment. Variations in pH and water temperature were also identified to be drivers of Culicidae emergence. Modelling failed to detect treatment effects on any of the nontarget taxa abundance, including Diptera, Lepidoptera, Ephemeroptera, Odonata, Coleoptera, Hymenoptera, and Arachnida. An inverse relationship between insectivore and prey taxa abundance was observed. In 2018, taxa richness increased between years and trended higher at treated sites and a positive relationship between insectivore and prey taxa richness was observed. In 2017, Shannon-Weiner index and Simpson’s index of diversity were higher at untreated sites, and in 2018 diversity indices were higher at treated sites, with taxa richness increasing between years and higher evenness trending at treated sites. Our data suggest that treatment effects were potentially shrouded by natural variability of physicochemical variables, especially due to the varying hydroperiod observed over the three years of sampling. Additional work is needed to capture average conditions and separate confounding variables from treatment effects. This study provides an inventory of the current wetland insect community in the South March Highlands Conservation Forest landscape that offers a reference for ongoing mosquito management.

Investigating Phylogenetic Relationships of Mosquito-Borne Avian Malaria in Mississippi

The vectors of avian malaria (Haemosporida) are an understudied component of wildlife disease ecology. Most studies of avian malaria have focused on the secondary bird hosts. This imbalance leaves a significant gap in our knowledge and understanding of the insect hosts. This study investigates the diversity of malaria parasites carried by mosquitoes (Diptera, Culicidae) in the state of Mississippi. Using PCR techniques, haemosporidian infection rates were determined and parasites were identified in a phylogenetic context to those previously annotated. A total of 27,157 female mosquitoes representing 15 species were captured. Five of those species tested positive for malaria parasites with an overall infection rate of 4 per 1000 mosquitoes infected. Mosquitoes were shown to harbor Plasmodium and Haemoproteus parasites. Surprisingly, a unique lineage of parasites was discovered in Anopheles mosquitoes potentially representing a new genus of haemosporidian parasites, reinforcing the need to continue investigating this diverse group of parasites.

Culicídeos associados às bromélias na Mata Atlântica do município de Cananéia, São Paulo, Brasil / Bromeliad associated mosquitoes from Atlantic Forest, in Cananéia, São Paulo, Brasil

Introdução: A Mata Atlântica não possui fisionomia uniforme, pois seu relevo determina diferentes condições ambientais que definem a composição de sua fauna e flora. Em uma mesma região, a diversidade de espécies pode variar de acordo com a heterogeneidade dos habitats. Objetivo: Caracterizar a fauna de Culicidae em fitotelmatas de bromélias (Bromeliaceae), no município de Cananéia, São Paulo, Brasil. Métodos: Larvas e pupas de culicídeos foram obtidas, mensalmente, do conteúdo aquático de bromélias de três ambientes: planície, encosta e morro. As coletas ocorreram entre julho de 2008 e junho de 2009. Para estimar a variedade de espécies foram utilizados os índices de Margalef e de Menhinick. O índice de Jaccard e o de Sorensen indicaram a semelhança entre bromélias de dois ambientes. Para avaliar a dominância, empregou-se o índice de Simpson e de Berger-Parker. A eqüidade na distribuição das espécies foi avaliada pelo índice de Pielou e a constância em cada ambiente pelo índice c. Os testes estatísticos empregados foram: análise de variância (ANOVA) com o teste de Fisher; teste Kruskal-Wallis e valor p; teste de associação qui-quadrado e regressão linear simples por meio do coeficiente de correlação de Pearson. Resultados: O teste Kruskal-Wallis mostrou diferenças significantes para o volume hídrico das bromélias de cada ambiente (p=0,002, IC= 95 por cento) mas não para o valor do pH (p= 0,775). Foram identificados 2024 mosquitos, 786 (38,83 por cento) na planície, 606 (29,94 por cento) na encosta e 632 (31,23 por cento) no morro, dentre eles: Culex (Microculex) (62,25 por cento), Culex ocellatus (21,20 por cento), Anopheles (Kerteszia) (15,17 por cento), Wyemoyia (Phoniomyia) (1,33 por cento) e Runchomyia (Runchomyia) (0,05 por cento). O índice de riqueza de Margalef foi maior na planície (d1=2,55) e menor no morro (d1=1,86), e o de Sorensen indicou encosta e morro como os ambientes mais similares (QS=0,79) e planície e morro como os menos similares (QS=0,65). O teste 2 não mostrou associação entre os ambientes e o tipo de bromélia (p=0,060). O coeficiente de Pearson não mostrou correlação significativa entre o número de imaturos e a temperatura (p=0,111) ou precipitação acumulada (p=0,828). Conclusões: Planície apresentou os maiores índices de diversidade. A prevalência de An. homunculus foi maior do que a encontrada em estudos anteriores. Culex ocellatus, Cx. (Mcx.) retrosus, Cx. (Mcx.) neglectus, Cx.(Mcx.) imitator e An. (Ker.) homunculus foram as espécies mais dominantes quando os três ambientes foram analisados conjuntamente / Introduction: Atlantic Forest is not uniform and its relief determines different environmental conditions that define the composition of its fauna and flora. Thus, in the same region, species diversity may vary according to habitat heterogeneity. Objective: To assess mosquito (Culicidae) fauna in bromeliads phytotelmate (Bromeliaceae), in Cananéia, São Paulo, Brazil. Methods: The larvae and pupae of mosquitoes were collected monthly in the water content of bromeliads from three regions: the lowland, hillside and hilltop. Sampling occurred between July 2008 and June 2009. To estimate richness, Margalef and Menhinick indices were used. Jaccard and Sorensen indices indicated the similarity between bromeliads from two regions. To assess dominance, we employed Simpson and Berger-Parker indices. Equity in the distribution of species was assessed by Pielou index and c index evaluated species constancy in each region. Statistical analyses applied were: ANOVA with Fisher\'s exact test, Kruskal-Wallis test and p value; Chi-square association test and linear regression using Pearson correlation coefficient. Results: Kruskal-Wallis test showed significant differences in water volume for bromeliads from each region (p=0,002, CI=95 per cent) but not for the pH value (p=0,775). In all, 2024 mosquitoes were identified, 786 (38,83 per cent) in the lowland, 606 (29,94 per cent) on the hillside and 632 (31,23 per cent) on the hilltop, among them: Culex (Microculex) (62,25 per cent), Culex ocellatus (21,20 per cent), Anopheles (Kerteszia) (15,17 per cent), Wyemoyia (Phoniomyia) (1,33 per cent) and Runchomyia (Runchomyia) (0,05 per cent). The prevalence of Anopheles homunculus was higher than that found in previous studies. Culex ocellatus, Cx. (Mcx.) retrosus, Cx. (Mcx.) neglectus, Cx. (Mcx.) imitator and An. (Ker.) homunculus were the most dominant species when the three regions were analyzed together

Entre os múltiplos grupos que compõem as Seções Spissipes e Melanoconion do subgênero Melanoconion de Culex, Atratus e Educator apresentam espécies que estão envolvidas na transmissão de arbovírus ao ser humano e animais em ambientes rurais e silvestres. Porém, esses grupos exibem grande dificuldade na identificação das espécies, seja devido à grande semelhança morfológica entre seus representantes ou à falta de chaves de identificação atualizadas. Diante disso, objetivou-se realizar a revisão taxonômica das espécies pertencentes aos Grupos Atratus e Educator da Seção Melanoconion de Culex (Melanoconion). Para isso, foi realizado o estudo morfológico detalhado das diversas fases de desenvolvimento dos espécimes de interesse, verificada a existência de espécies a serem nomeadas e/ou espécies válidas colocadas na sinonímia de outras. Como resultado do estudo morfológico das espécies do Grupo Atratus, foram redefinidas oito espécies. Destas, duas foram validadas, pois estavam na sinonímia de outras e seis espécies desconhecidas da ciência foram descritas e serão formalmente nomeadas. Para o Grupo Educator, oito espécies foram descritas, sendo uma removida da sinonímia e três espécies desconhecidas da ciência foram descritas e serão formalmente nomeadas. Chaves de identificação ilustradas, com caracteres usados para o reconhecimento das espécies, foram elaboradas para todas as formas de desenvolvimento das espécies de ambos os grupos. Foram construídos mapas de distribuição geográfica das espécies, foram atualizados dados bionômicos, elaboradas ilustrações e pranchas fotográficas para comparação de estruturas morfológicas externas das formas imaturas, adultos e genitália masculina. / Among the multiple groups that compose the Spissipes and Melanoconion Sections of the subgenus Melanoconion of Culex, the Atratus and Educator Groups present species that are involved in the transmission of arboviruses to man and animals in rural and wild environments. However, these groups present great difficulty in identifying the species, either because of the great morphological similarity between their representatives or the lack of updated identification keys. The aim of this study was to carry out a taxonomic review of the species belonging to the Atratus and Educator Groups of the Melanoconion Section of Culex (Melanoconion). Therefore, a detailed morphological study of the several stages of development of the specimens of interest was carried out, verifying the existence of species to be named and/or valid species placed in the synonymy of others. As a result of the morphological study of the Atratus Group species, eight species were redefined. Of these, two were resurrected from the synonymy of others and six species unknown to science were described and will be formally named. For the Educator Group, eight species have been described, one being removed from the synonymy and three species unknown to science have been described and will be formally named. Illustrated identification keys, with characters used for species recognition, have been created for all forms of development of species in both Groups. Geographic distribution maps of the species were created, illustrations and photographic plates for comparison of external morphological structures of the immatures forms, adults and male genitalia were provided, and bionomic data were updated.

Identificação de fonte sanguínea em dípteros da Família Culicidae, em áreas de epizootia da febre amarela silvestre / Identification of blood source in the family Culicidae flies, in areas of outbreak of jungle yellow fever

A importância em conhecer o padrão alimentar em mosquitos da Família Culicidae permite esclarecer alguns aspectos relacionados à transmissão de zoonoses e estimar o grau de contato humano-vetor que é fator relevante em estudos epidemiológicos. Com o objetivo de explorar o comportamento alimentar dessa Família, em área epizoótica de febre amarela silvestre, foram coletados exemplares nos municípios de Santo Antônio das Missões e Garruchos, Estado do Rio Grande do Sul. Fêmeas ingurgitadas foram obtidas por aspiração em ambiente de mata, no período de setembro de 2005 a abril de 2007 e identificadas segundo fonte de sangue ingerido através da técnica imunoenzimática ELISA de captura no sistema avidinabiotina. Foram testadas seis fontes de alimento: ave, bovino, eqüino, humano, macaco e rato. Os resultados obtidos mediante a padronização de anticorpos monoclonais possibilitaram demonstrar pela primeira vez o reconhecimento de sangue humano ingerido nesses mosquitos pelo emprego da subclasse IgG1 e comprovar a sensibilidade e especificidade da técnica ELISA de captura. No município de Santo Antônio das Missões, de um total de 190 amostras, 60,9% reagiram para sangue de boi, 23,6% para humano, 9,9% para ave, 1,9% para macaco e 3,7% para combinações de dois hospedeiros. Quanto às amostras referentes ao município de Garruchos, das 158 fêmeas capturadas na área Cachoeirinha pode-se observar reatividade para ave (16%), boi (29,6%), humano (36,8%), cavalo (4%), macaco (0,8%) e combinações de hospedeiros (12,8%), enquanto que para as 149 fêmeas pertencentes à área de São José, detectou-se sangue ingerido de boi em (51,5%), ave e humano (11,5%), macaco (6,2%), cavalo (0,8%) e mistos (18,5%). Aedes scapularis, Aedes crinifer, Culex (Culex) spp., Haemagogus leucocelaenus apresentaram maior número de fêmeas ingurgitadas nos dois municípios. Os resultados obtidos com Aedes scapularis sugerem ecletismo, conforme combinações detectadas em amostras de sangue de diferentes fontes. Haemagogus leucocelaenus apresentou a maior proporção de amostras contendo sangue humano em relação às demais fontes e essa característica traz implicações, por ser espécie incriminada na transmissão e por se tratar de área de ocorrência de epizootias de febre amarela. / The knowledge of mosquitoes Culicidae host feeding patterns permits to clarify some aspects related to zoonosis transmission and to estimate the degree of human-vector contact which is relevant in epidemiological studies. Aiming to explore the feeding behavior of these mosquitoes, specimens were collected in the municipalities of Santo Antônio das Missões and Garruchos, Rio Grande do Sul, an epizootic area of sylvatic yellow fever. Engorged females were collected by aspiration from forested areas from September 2005-April 2007 and their blood meals were identified using the avidin-biotin system of immunoenzymatic ELISA capture. Six blood meal sources were tested: bird, cattle, horse, human, monkey and rat. The result achieved with the species-specific IgG1 mAb was unprecedented for mosquito blood meal identification and reinforced the sensibility and specificity of the immunoenzymatic ELISA capture. Of the 190 samples from Santo Antônio das Missões, 60.9% reacted to cattle, 23.6% to human, 9.9% to bird, 1.9% to monkey and 3.7% to mixed blood meals. In Garruchos, of the 158 females collected in Cachoeirinha, 16.0% reacted to bird, 29.6% to cattle, 36.8% to human, 4.0% to horse, 0.8% to monkey and 12.8% to mixed blood, while of the 149 engorged females from São José, blood from cattle accounted for 51.5%, of blood identified, bird and human 11.5%, monkey 6.2%, horse 0.8% and mixed blood 18.5%. Blood engorged females of Aedes scapularis, Aedes crinifer, Culex (Culex) spp., Haemagogus leucocelaenus predominated in the two municipalities. The results obtained with Aedes scapularis suggests its eclecticism, according to the combinations of blood which were detected from different sources. Haemagogus leucocelaenus was found to have the highest proportion of samples containing human blood in comparison with other sources, which has implications, on account of being incriminated in the transmission and also for taking into consideration the outbreaks reported that underline the risk of yellow fever.

Introdução. A cidade de São Paulo vem adotando medidas no enfrentamento das inundações, como a implantação dos Reservatórios de Contenção de Cheias \"Piscinões\", que no período de estiagem, após as precipitações, propiciam a formação de criadouros de culicídeos, especialmente nos componentes hidráulicos. Objetivos. Avaliar os aspectos ecológicos da fauna de culicídeos nos Reservatórios de Contenção de Cheias Caguaçu e Inhumas, verificando a diversidade, dominância, similaridade e riqueza de espécies. Correlacionar fatores abióticos com a abundância numérica de mosquitos. Materiais e Métodos: Coletas mensais de culicídeos, no período de março de 2006 a fevereiro de 2007, empregando-se os métodos de conchas e aspirador à bateria. Em laboratório, após o desenvolvimento das larvas, os adultos foram identificados até a categoria de espécie. Resultados. Foram coletados 8.917 espécimes de culicídeos, dos quais 7.750 larvas e pupas e 1.167 adultos. Os espécimes identificados foram distribuídos em 4 gêneros e 13 espécies. Observou-se a dominância de Culex (Culex) quinquefasciatus, que representou 98% dos espécimes coletados no Inhumas e 92% no Caguaçu. A maior freqüência de mosquitos foi registrada no Piscinão Inhumas (6.378 larvas e 930 adultos) e maior riqueza de espécies no Caguaçu (11 espécies). Verificou-se correlação positiva entre a temperatura a e abundância numérica de imaturos nas duas áreas, correlação positiva no Piscinão Caguaçu e correlação negativa no Inhumas entre a precipitação e a abundância numérica de imaturos. As análises estatísticas demonstraram que os fatores abióticos analisados exercem pouca influência na população de adultos. Conclusões. A espécie Culex quinquefasciatus mostrou-se dominante e mais freqüente nos dois ambientes. Medidas de controle da espécie nos piscinções estudados se fazem necessárias tendo em vista seu potencial epidemiológico na transmissão de agentes patogênicos. / Introduction. The City of São Paulo has been adopting highly measures to enable it to face up to the problem of flooding, among them being the construction of Reservoirs for the Containment of Flood Waters, known as \"piscinões\". In the dry season, after the occasional rains, these reservoirs, especially their hydraulic machinery, serve as breeding places for the Culicidae. Objectives. The assessment of the ecological aspects of the Culicidae fauna in the Reservoirs for the Containment of Flood Waters at Caguaçu and Inhumas to establish their diversity, dominance, similarity and species richness and, further, to correlate a-biotic factors with the numerical abundance of the mosquitoes. Materials and Methods: Monthly collections of Culicidae were undertaken during the period from March 2006 to February 2007, using the shell and battery aspirator methods. Results. A total of 8,917 Culicidae specimens were collected, 7,750 of which were larvae and pupae and 1,167 adults. The specimens were identified as belonging to 4 genera and 13 species. Culex quinquefasciatus, accounting for 98% of the specimens collected in the Inhumas Reservoir and 92% of those collected in Caguaçu, was the dominant species. The greatest frequency of mosquitoes was registered in the Inhumas Reservoir (6,378 larvae and 930 adults) and the greatest species richness in the Caguaçu Reservoir (11 species). A positive correlation was found as between the temperature and the numerical abundance of immature forms of the two areas and a positive correlation in Caguaçu and a negative correlation in Inhumas between the rainfall the and numerical abundance of immature forms. The analyses demonstrated that the a-biotic factors analyzed exercised little influence on the adult population. Conclusions. The species Culex quinquefasciatus was the dominant and most frequent species in both environments. Methods for the control of the species in the Reservoirs studied are necessary in view of their epidemiological potential in terms of the transmission of pathogenic agents.

Identificação de fonte sanguínea em dípteros da Família Culicidae, em áreas de epizootia da febre amarela silvestre / Identification of blood source in the family Culicidae flies, in areas of outbreak of jungle yellow fever

A importância em conhecer o padrão alimentar em mosquitos da Família Culicidae permite esclarecer alguns aspectos relacionados à transmissão de zoonoses e estimar o grau de contato humano-vetor que é fator relevante em estudos epidemiológicos. Com o objetivo de explorar o comportamento alimentar dessa Família, em área epizoótica de febre amarela silvestre, foram coletados exemplares nos municípios de Santo Antônio das Missões e Garruchos, Estado do Rio Grande do Sul. Fêmeas ingurgitadas foram obtidas por aspiração em ambiente de mata, no período de setembro de 2005 a abril de 2007 e identificadas segundo fonte de sangue ingerido através da técnica imunoenzimática ELISA de captura no sistema avidinabiotina. Foram testadas seis fontes de alimento: ave, bovino, eqüino, humano, macaco e rato. Os resultados obtidos mediante a padronização de anticorpos monoclonais possibilitaram demonstrar pela primeira vez o reconhecimento de sangue humano ingerido nesses mosquitos pelo emprego da subclasse IgG1 e comprovar a sensibilidade e especificidade da técnica ELISA de captura. No município de Santo Antônio das Missões, de um total de 190 amostras, 60,9% reagiram para sangue de boi, 23,6% para humano, 9,9% para ave, 1,9% para macaco e 3,7% para combinações de dois hospedeiros. Quanto às amostras referentes ao município de Garruchos, das 158 fêmeas capturadas na área Cachoeirinha pode-se observar reatividade para ave (16%), boi (29,6%), humano (36,8%), cavalo (4%), macaco (0,8%) e combinações de hospedeiros (12,8%), enquanto que para as 149 fêmeas pertencentes à área de São José, detectou-se sangue ingerido de boi em (51,5%), ave e humano (11,5%), macaco (6,2%), cavalo (0,8%) e mistos (18,5%). Aedes scapularis, Aedes crinifer, Culex (Culex) spp., Haemagogus leucocelaenus apresentaram maior número de fêmeas ingurgitadas nos dois municípios. Os resultados obtidos com Aedes scapularis sugerem ecletismo, conforme combinações detectadas em amostras de sangue de diferentes fontes. Haemagogus leucocelaenus apresentou a maior proporção de amostras contendo sangue humano em relação às demais fontes e essa característica traz implicações, por ser espécie incriminada na transmissão e por se tratar de área de ocorrência de epizootias de febre amarela. / The knowledge of mosquitoes Culicidae host feeding patterns permits to clarify some aspects related to zoonosis transmission and to estimate the degree of human-vector contact which is relevant in epidemiological studies. Aiming to explore the feeding behavior of these mosquitoes, specimens were collected in the municipalities of Santo Antônio das Missões and Garruchos, Rio Grande do Sul, an epizootic area of sylvatic yellow fever. Engorged females were collected by aspiration from forested areas from September 2005-April 2007 and their blood meals were identified using the avidin-biotin system of immunoenzymatic ELISA capture. Six blood meal sources were tested: bird, cattle, horse, human, monkey and rat. The result achieved with the species-specific IgG1 mAb was unprecedented for mosquito blood meal identification and reinforced the sensibility and specificity of the immunoenzymatic ELISA capture. Culicídeos associados às bromélias na Mata Atlântica do município de Cananéia, São Paulo, Brasil / Bromeliad associated mosquitoes from Atlantic Forest, in Cananéia, São Paulo, Brasil

Introdução: A Mata Atlântica não possui fisionomia uniforme, pois seu relevo determina diferentes condições ambientais que definem a composição de sua fauna e flora. Em uma mesma região, a diversidade de espécies pode variar de acordo com a heterogeneidade dos habitats. Objetivo: Caracterizar a fauna de Culicidae em fitotelmatas de bromélias (Bromeliaceae), no município de Cananéia, São Paulo, Brasil. Métodos: Larvas e pupas de culicídeos foram obtidas, mensalmente, do conteúdo aquático de bromélias de três ambientes: planície, encosta e morro. As coletas ocorreram entre julho de 2008 e junho de 2009. Para estimar a variedade de espécies foram utilizados os índices de Margalef e de Menhinick. O índice de Jaccard e o de Sorensen indicaram a semelhança entre bromélias de dois ambientes. Para avaliar a dominância, empregou-se o índice de Simpson e de Berger-Parker. A eqüidade na distribuição das espécies foi avaliada pelo índice de Pielou e a constância em cada ambiente pelo índice c. Os testes estatísticos empregados foram: análise de variância (ANOVA) com o teste de Fisher; teste Kruskal-Wallis e valor p; teste de associação qui-quadrado e regressão linear simples por meio do coeficiente de correlação de Pearson. Resultados: O teste Kruskal-Wallis mostrou diferenças significantes para o volume hídrico das bromélias de cada ambiente (p=0,002, IC= 95 por cento) mas não para o valor do pH (p= 0,775). Foram identificados 2024 mosquitos, 786 (38,83 por cento) na planície, 606 (29,94 por cento) na encosta e 632 (31,23 por cento) no morro, dentre eles: Culex (Microculex) (62,25 por cento), Culex ocellatus (21,20 por cento), Anopheles (Kerteszia) (15,17 por cento), Wyemoyia (Phoniomyia) (1,33 por cento) e Runchomyia (Runchomyia) (0,05 por cento). O índice de riqueza de Margalef foi maior na planície (d1=2,55) e menor no morro (d1=1,86), e o de Sorensen indicou encosta e morro como os ambientes mais similares (QS=0,79) e planície e morro como os menos similares (QS=0,65). O teste 2 não mostrou associação entre os ambientes e o tipo de bromélia (p=0,060). O coeficiente de Pearson não mostrou correlação significativa entre o número de imaturos e a temperatura (p=0,111) ou precipitação acumulada (p=0,828). Conclusões: Planície apresentou os maiores índices de diversidade. A prevalência de An. homunculus foi maior do que a encontrada em estudos anteriores. Culex ocellatus, Cx. (Mcx.) retrosus, Cx. (Mcx.) neglectus, Cx.(Mcx.) imitator e An. (Ker.) homunculus foram as espécies mais dominantes quando os três ambientes foram analisados conjuntamente / Introduction: Atlantic Forest is not uniform and its relief determines different environmental conditions that define the composition of its fauna and flora. Thus, in the same region, species diversity may vary according to habitat heterogeneity. Objective: To assess mosquito (Culicidae) fauna in bromeliads phytotelmate (Bromeliaceae), in Cananéia, São Paulo, Brazil. Methods: The larvae and pupae of mosquitoes were collected monthly in the water content of bromeliads from three regions: the lowland, hillside and hilltop. Sampling occurred between July 2008 and June 2009. To estimate richness, Margalef and Menhinick indices were used. Jaccard and Sorensen indices indicated the similarity between bromeliads from two regions. To assess dominance, we employed Simpson and Berger-Parker indices. Equity in the distribution of species was assessed by Pielou index and c index evaluated species constancy in each region. Statistical analyses applied were: ANOVA with Fisher\'s exact test, Kruskal-Wallis test and p value; Chi-square association test and linear regression using Pearson correlation coefficient. Results: Kruskal-Wallis test showed significant differences in water volume for bromeliads from each region (p=0,002, CI=95 per cent) but not for the pH value (p=0,775). In all, 2024 mosquitoes were identified, 786 (38,83 per cent) in the lowland, 606 (29,94 per cent) on the hillside and 632 (31,23 per cent) on the hilltop, among them: Culex (Microculex) (62,25 per cent), Culex ocellatus (21,20 per cent), Anopheles (Kerteszia) (15,17 per cent), Wyemoyia (Phoniomyia) (1,33 per cent) and Runchomyia (Runchomyia) (0,05 per cent). Pearson coefficient showed no significant correlation between the number of immatures and temperature (p=0,111) or rainfall (p=0,828). Conclusions: Lowland region had the highest diversity index. The prevalence of Anopheles homunculus was higher than that found in previous studies. Culex ocellatus, Cx. (Mcx.) retrosus, Cx. (Mcx.) neglectus, Cx. (Mcx.) imitator and An. (Ker.) homunculus were the most dominant species when the three regions were analyzed together
