The purpose of making possible the use of the branchoneta as an alternative food source for fish and crustaceans. The research consisted of two treatments (inoculation and without inoculation of D. brasiliensis cysts) blocked in time (May and October), with two replicates for each treatment / block combination. For this, four semi-excavated nurseries of 2,000 m2 each were used. Two nurseries were inoculated with 2g of cysts and the other two nurseries were not inoculated and only hatching of nauplii occurred due to the natural production of this microcrustacean previously existing in these nurseries. After 15 days of cultivation, the treatment with inoculation produced 20.75 ± 2.31 g of cysts, being higher than that without inoculation, which produced only 7.75 ± 2.31 g, thus occurring a significant difference (P < 0.05) according to the analysis of variance and the mean comparison test (Tukey at 5%). In this way, an average of 2.075g / ha / year of cysts was produced. Regarding the biomass production, the statistical analysis showed that the average obtained with inoculation of 2 g of cysts per nursery, of 18.63 ± 0.74 kg was superior to 8.00 ± 0.75 kg of the treatment without inoculation. Thus it is possible to produce a biomass of 1,863 kg / ha / year. / Este trabalho teve o objetivo de desenvolver uma metodologia para a produção de cistos e biomassa de “branchoneta” Dendrocephalus brasiliensis, na Estação de Piscicultura de Paulo Afonso (09º22’38” S e 38º13’58” W), Paulo Afonso, BA, com a finalidade de viabilizar a utilização da branchoneta como fonte alternativa de alimento para peixes e crustáceos. A pesquisa constou de dois tratamentos (com inoculação e sem inoculação de cistos de D. brasiliensis) bloqueados no tempo (maio e outubro), com duas repetições para cada combinação tratamento/bloco. Para isso, foram utilizados quatro viveiros semi-escavados de 2.000 m2 cada. Dois viveiros foram inoculados com 2g de cistos e os outros dois viveiros não foram inoculados, tendo ocorrido apenas a eclosão de náuplios, em virtude da produção natural desse microcrustáceo existente anteriormente nestes viveiros. Após 15 dias de cultivo, o tratamento com inoculação produziu 20,75 ± 2,31 g de cistos, sendo portanto superior aquele sem inoculação, que produziu apenas 7,75 ± 2,31 g, ocorrendo dessa forma uma diferença significativa (P< 0,05) conforme a análise de variância e o teste de comparação de média (Tukey a 5%). Desta forma, foi produzida em média 2.075g/ha/ano de cistos. Quanto à produção de biomassa, a análise estatística mostrou que a média obtida com inoculação de 2g de cistos por viveiro, de 18,63 ± 0,74 Kg foi superior a 8,00 ± 0,75 Kg do tratamento sem inoculação. Assim é possível produzir uma biomassa de 1.863 Kg/ha/ano.

Desempenho reprodutivo da égua crioula / Reproductive performance of the Crioulo mare

O objetivo deste trabalho foi avaliar alguns aspectos reprodutivos de éguas Crioulas a fim de verificar a influência destes nos índices de prenhez: idade das éguas, status reprodutivo, uso do cio do potro, presença de líquido intra-uterino (LIU), eficiência dos tratamentos pós-cobertura, presença de cistos endometriais, realização de vulvoplastia, incidência de ovulações duplas e gestações gemelares. Foram utilizados 517 ciclos de 406 éguas Crioulas de um rebanho comercial, com idades entre dois e vinte e dois anos. O índice de prenhez por ciclo foi de 80,5% aos 12 dias, 76,5% aos 42 dias e a mortalidade embrionária foi de 3,9%. O índice de prenhez foi influenciado pela idade das éguas (P < 0,01). O índice de prenhez no cio do potro foi de 85,4%, melhor que o índice obtido pelas éguas em que não se utilizou este cio e que foram cobertas no subseqüente (P = 0,07). O método de cobertura influenciou a prenhez, sendo a inseminação superior à monta natural (P = 0,01). A idade influenciou a presença de LIU sendo a incidência em éguas com mais de 12 anos maior que nas mais jovens (P < 0,01). A presença de cistos endometriais influenciou negativamente a taxa de prenhez aos 12 e aos 42 dias (P = 0,00). A correção cirúrgica da região perineal melhorou os índices de prenhez aos 12 e aos 42 dias (P < 0,05). A taxa de ovulações duplas foi de 4,84% e não influenciou os índices de prenhez aos 12 e aos 42 dias (P > 0,5). Em 6 ciclos (1,16%) de 5 éguas solteiras foram observadas gestações gemelares. Concluímos que a fertilidade das éguas Crioulas é influenciada pela idade das éguas, pela conformação vulvar, pela presença de cistos endometriais e pelo status reprodutivo. O acúmulo de LIU é um fator importante na reprodução da égua Crioula e deve ser observado e devidamente tratado para a obtenção de melhores índices de prenhez. / The aim of this study was to evaluate the reproductive aspects of Crioulo mares to verify the influence of the following aspects on the pregnancy rates: mare’s age, reproductive status, use of foal heat, presence of intrauterine fluid (IUF), efficiency of post-breeding treatments, presence of endometrial cysts, incidence of double ovulations and twin pregnancies. Pregnancy rate on foal heat was 85.4%, better than the rate obtained by the mares which foal heat was not used and the subsequent cycle was (P = 0.07). The breeding method influenced pregnancy, being artificial insemination superior to natural breeding (P = 0.01). Age influenced the presence of IUF, once the incidence was higher in mares with more than 12 years old than in younger mares (P < 0.01). The presence of endometrial cysts negatively influenced the pregnancy rate at 12 and 42 days (P = 0.00). Surgical repair of perineal region improved the pregnancy rates at 12 and 42 days (P < 0.05). The rate of double ovulations was of 4.84% and did not influence the pregnancy rates either at 12 and 42 days (P > 0.5). In 6 cycles (1.16%) of 5 single mares there were observed twin pregnancies. We concluded that the fertility of Crioulo mares is influenced by the age of the mares, vulvar conformation, presence of endometrial cysts and reproductive status. The accumulation of IUF is an important factor on the reproduction of Crioulo mares and must be observed and correctly treated in order to obtain better pregnancy rates.

Role of CD4+CD25+ Regulatory T Lymphocytes in Experimental Toxoplasmosis

Toxoplasmosis is an important cause of congenital disease, and it is one of the most common opportunistic infections in patients with acquired immunodeficiency syndrome. The need for a reliable experimental model of this infection is crucial not only for achieving a better understanding of the patho-physiology of the disease, but also for developing better methods for evaluating new therapeutic regimens. The purpose of the present study was to investigate the role of CD4+CD25+ T regulatory lymphocytes in mice infected with Toxoplasma gondii. T regulatory (Treg) cells have been shown to play an important role in our immune system in controlling the activity of other T lymphocytes. These cells are differentiated from other T lymphocyte populations based on the co-expression of CD4 and CD25 and expression of the Foxp3 gene. The results of several recent studies have suggested that certain pathogens may be able to increase their survival in the host by exploiting T reg cell activity. T regulatory cells have been shown to control the persistence of the protozoan parasite, Leishmania major, in mice; however, this population of cells plays only a limited role during murine infection with Trypanosoma cruzi. In the present study we have investigated the role of Treg cells during murine infection with the ME49 strain of T. gondii. In vivo depletion of Treg cells was accomplished by injecting mice with a monoclonal antibody (Mab) isolated from the 7D4 rat hybridoma cell line. This Mab is specific for the interleukin-2 receptor chain (also known as CD25). Female Swiss Webster mice of approximately 6-7 weeks of age were depleted of Treg cells by intraperitoneal injection of 400µg of Mab, mice were injected once 7days prior to infection, and a second time 1 day prior to infection, with 20 tissue cysts of T. gondii. Mouse weight and tissue cyst numbers were monitored to evaluate the impact of Treg depletion on the outcome of infection. Our results suggest that depletion of Treg cells has little measurable impact during the acute stage of infection with the ME49 strain of T. gondii. Further studies will be required to determine what role, if any, these cells play in the chronic stage of murine toxoplasmosis.

Δραστηριότητα λυοσωματικών ενζύμων στο περιτοναϊκό υγρό γυναικολογικών καρκίνων, πυελικών φλεγμονών και υγρού καλοήθων κύστεων ωοθηκών

Η δραστικότητα των λυοσωματικών ενζύμων είναι αυξημένη στο εξωκυττάριο υγρό ασθενών με μηνιγγίτιδα και περιτονίτιδα μικροβιακής αιτιολογίας. Ο σκοπός της παρούσας μελέτης ήταν να ερευνήσουμε αν η δραστικότητα αυτών των ενζύμων ήταν αυξημένη στο περιτοναϊκό υγρό ασθενών με πυελική φλεγμονή και γυναικολογικό καρκίνο. Η δραστικότητα της β-γλυκουρονιδάσης, β-γαλακτοσιδάσης και α-μαννοσιδάσης μετρήθηκε στο περιτοναϊκό υγρό 5 ασθενών με PID, 10 ασθενών με γυναικολογικό καρκίνο, 10 ασθενών που χρησιμοποιήθηκαν σαν σημείο αναφοράς και το υγρό 7 ασθενών με καλοήθεις κύστεις ωοθηκών. Η μέση τιμή της +/-SD της δραστικότητας της β- γλυκουρονιδάσης , β-γαλακτοσιδάσης, και της α- μανοσιδάσης στην PID ήταν 148+/-82, 278+/ -112, και 291+/-140 nmol 4 -methylummbelliferone/ ml / h αντίστοιχα. Στα δείγματα αναφοράς ήταν 22+/-9, 48+/-10 και80+/-23 , αντίστοιχα (p<=0,003 - 0.00001). Στο γυναικολογικό καρκίνο η δραστικότητα ήταν 113+/-35, 210+/- 82, και 243+/-123 αντίστοιχα ( διαφορά από τα δείγματα αναφοράς p<=0,0006-0,000001). Υπήρξε θετική συσχέτιση μεταξύ της δραστικότητας της β- γλυκουρονιδάσης και του σταδίου του καρκίνου. Η μέτρηση της δραστικότητας των ενζύμων στο υγρό των ωοθηκικών κύστεων δεν διέφερε σημαντικά σε σχέση με αυτή των δειγμάτων αναφοράς. Η δραστικότητα των λυοσωματικών ενζύμων είναι αυξημένη στο περιτοναϊκό υγρό ασθενών με (PID), πυελική φλεγμονή και γυναικολογικό καρκίνο. Η απουσία επικάλυψης των μετρηθέντων τιμών δραστικότητας των ανωτέρω ενζύμων μεταξύ ασθενών με γυναικολογικό καρκίνο και PID σε σχέση με τα δείγματα αναφοράς δείχνει οτι κάποιες μετρήσεις πρέπει να αρχίζουν να εφαρμόζονται για διαγνωστικούς σκοπούς. / The activity of lysosomal enzymes is increased in extracellular fluids οf patients with bacterial maningitis and peritonitis. Our objective was to investigate whether the activity of these enzymes is increased in the peritoneal fluid of pelvic inflammatory disease (PID) and gynecologic cancers. The activity of β-glucuronidase, β-galactosidase and α-mannosidase was measured in the peritoneal fluid of 5 patients with PID, 10 with gynecologic cancer, 10 control subjects, and the flouid of 7 benign ovarian cysts. The mean +/- SD β-glucoronidase, β-galactosidase, and α-mannosidase activity in PID was 148+/-82, 278+/-112, and 291+/-140 nmol 4- methylumbelliferone/ ml/h, respectively; in the controls it was 22+/-9, 48+/-10 and 80+/-23, respectively ( p<=0,003-0,00001). In the gynecologic cancers the activity was 113+/-35, 210+/-82, and 243+/-123, respectively (difference from controls p<=0.0006-0.000001). There was a positive correlaton between β-glucoronidase activity and stage of cancer.The activity of the ovarian cysts fluid did not differ significantly from the controls. The lysosomal enzyme activity is increased in the peritoneal fluid of PID and gynecologic cancers. The absence of overlapping values between patients and controls indicates that such measurements may be applied for diagnostic purposes.

These processes in odontogenic tumors are considered rare. This study aimed to conduct a study to assess the presence and biological significance of clear cells in the epithelial lining of a series of odontogenic cystic lesions. A total of 48 histological slides of these lesions were examined by two calibrated examiners. The paraffin blocks corresponding to the lesions that were observed in clear cell were re-processed and 22 slides were stained by the periodic acid Schiff (PAS) after diastase digestion, and evaluated for the presence of mucous cells. It was performed a descriptive statistical analysis of data and these were represented as graphs and tables. Mucous cells were found in 41,66% cases of radicular cysts, 50% of keratocystic odontogenic tumors, 100% of dentigerous cysts and 100% of unicystic ameloblastomas. The results of this research may suggest that degenerative and metaplastic phenomena usually occur in the epithelium of odontogenic cysts and odontogenic cystic tumors, which may represent a phenomenon associated with the chronic inflammatory process that is usually observed in these lesions, or be indicative of the multipotentiality odontogenic epithelium. / Alterações degenerativas e metaplásicas são usualmente observadas no revestimento epitelial dos cistos odontogênicos. Nos tumores odontogênicos estes processos são considerados mais raros. Esta pesquisa objetivou realizar um estudo para avaliar a presença e o significado biológico das células claras no revestimento epitelial de uma série de lesões odontogênicas císticas. Um total de 48 lâminas histológicas das referidas lesões foram examinadas por dois avaliadores previamente calibrados. Os blocos parafinados correspondentes às lesões em que foram observadas células claras foram novamente processados e 22 lâminas foram coradas pela técnica do ácido periódico de Schiff (PAS) após digestão pela diástase e avaliadas quanto à presença de células mucosas. Foi realizada a análise estatística descritiva dos dados e estes foram representados na forma de gráficos e tabelas. Células mucosas foram observadas em 41,66% dos casos de cistos radiculares, 50% dos tumores odontogênicos ceratocísticos, 100% dos cistos dentígeros e 100% dos ameloblastomas unicísticos. Os resultados dessa pesquisa podem sugerir que fenômenos degenerativos e metaplásicos são de ocorrência usual no epitélio de cistos odontogênicos e de tumores odontogênicos císticos, podendo representar um fenômeno associado ao processo inflamatório crônico que, comumente, é observado nessas lesões, ou ser indicativo da multipotencialidade do epitélio odontogênico.

Estudo da terapêutica cirúrgica dos tumores odontogênicos queratocísticos associados ou não à síndrome do carcinoma nevóide de células basais e análise do tempo livre de recorrência / Study of surgical treatment of keratocystic odontogenic tumors associated or not to the nevoid basal cell carcinoma syndrome and analysis of recurrence-free period

O tumor odontogênico queratocístico (TOQ) é uma neoplasia cística benigna originada de remanescentes epiteliais da odontogênese, que se destaca pela alta recorrência e eventual associação com a síndrome do carcinoma nevóide de células basais (SCNCB). O presente estudo buscou responder questionamentos relacionados à sua terapêutica cirúrgica e suprir a carência de pesquisas atuais sobre as lesões associadas à SCNCB, que são ainda mais recorrentes, analisando uma amostra formada por 40 TOQs primários. As variáveis de interesse determinaram grupos amostrais, que receberam análise dos tempos livres de recorrência por função Kaplan-Meier, comparação desses tempos pelo teste log-rank com nível de significância de 5% (p < 0,05) e determinação do risco acumulado para o evento nos primeiros cinco anos. Vinte e sete lesões foram tratadas por exérese (GE) e 13 receberam terapêutica descompressiva (GD). Tratamentos complementares ocorreram em 38 (95%) lesões, sendo ostectomia periférica isolada em 10 (GO) e combinada à solução de Carnoy em 28 (GC). Treze eram associadas à SCNCB (GS) e as demais (n = 27) representaram lesões não sindrômicas (GnS). No período de acompanhamento médio de 43,5 meses (12 102 meses), seis (15%) recorrências foram diagnosticadas. Não houve diferença significativa entre os tempos livres de recorrência nos grupos comparados (p > 0,05), nem expressividade do risco acumulado para o evento na terapêutica descompressiva (15,4%) e na associação com a SCNCB (12,5%). A aplicação da solução de Carnoy não aumentou a efetividade da ostectomia periférica, mas se relacionou com risco acumulado de recorrência de 0% ao final do quinto ano de acompanhamento pós-exérese nas lesões sindrômicas. Portanto: 1) a terapêutica descompressiva não aumentou o risco para recorrências; 2) a ostectomia periférica mostrou efetividade similar quando combinada à solução de Carnoy, pelo menos nas lesões não associadas à SCNCB; 3) o risco de recorrência das lesões associadas à SCNCB também foi controlado por tratamentos complementares. / The keratocystic odontogenic tumor (KOT) is a benign cystic neoplasm originating from odontogenic epithelial remnants which is highlighted by its high recurrence rate and occasional association with the nevoid basal cell carcinoma syndrome (NBCCS). The present study aimed to answer questions related to its surgical therapy and to fulfill the lack of recent researches over the lesions associated to the NBCCS, which are even more recurrent, analyzing a sample ok 40 primary KOTs. Variables of interest determined sample groups that underwent recurrence-free period analyses by Kaplan-Meier function, comparing these results by log-rank test with a significance level of 5% (p < 0.05) and determination of cumulative risk for the recurrence event within the first five years. Twenty seven lesions were treated by exeresis (GE) and 13 underwent decompressive therapeutic (GD). Complementary treatment occurred in 38 (95%) lesions being as isolated peripheral ostectomy in 10 (GO) and combined with Carnoys solution in 28 (GC). Thirteen were associated to NBCCS (GS) and the others (n = 27) represented non syndromic lesions (GnS). For the mean follow-up period of 43.5 months (12 102 months), six (15%) recurrences were diagnosed. There was no significant difference among recurrence-free periods for the compared groups (p > 0.05) or cumulative risk expression for the decompressive therapeutic (15.4%) or for the association with NBCCS (12.5%). Carnoys solution application did not increase the efficacy of the peripheral ostectomy but was related to the cumulative recurrence risk of 0% at the end of the fifth year of post-exeresis follow-up for syndromic lesions. Therefore: 1) the decompressive therapeutic did not increase the recurrence risk; 2) peripheral oostectomy demonstrated similar efficacy when combined to Carnoys solution, at least for lesions not associated to NBCCS; 3) the recurrence risk of lesions associated to NBCCS was also controlled by complementary treatments.

Measuring Viability of the Red-Tide Dinoflagellate Lingulodinium polyedra Following Treatment with Ultraviolet (UV) Light

Harmful algae blooms (HABs) have caused millions dollars in annual losses to the aquaculture industry, inhibited beach recreation, and have threatened marine and human health. HABs and red tides can develop suddenly and their frequency, geographic range, and intensity have increased over the past decade. A possible source for spreading and seeding new areas expanding the geographic range of HABs is ballast water. The process of ballast water discharge has been identified as a primary vector for the translocation of non-indigenous species (NIS) and invasive species. National and international efforts are currently underway to address the impact of NIS and invasive species. Policy is being developed detailing stringent rules to kill, remove, or otherwise inactive organisms in ballast water prior to or upon discharge. Currently, vendors are developing technologies to treat ballast water and U.S. and international facilities are testing these technologies to verify their efficacy. Ultraviolet (UV) radiation is commonly employed in ballast water treatment technologies. Previous studies have shown that UV light is effective for disinfecting drinking water, but the response of non-pathogenic and marine organisms is largely unknown. The purpose of this research was to measure the viability of the durable red-tide forming dinoflagellate, Lingulodinium polyedra following UV treatment. Two methods were used to measure the viability signal; manual epifluorescence microscopy with correlated viability stains and Pulse Amplitude Modulated (PAM) fluorometry to measure the physiological state of the organism following UV treatment. The number of cysts was also enumerated. The results showed that there was a significant decrease in the number of living L. polyedra cells following a UV treatment of more than 100 mWs cm-2. The results also have showed a significant increase in the number of L. polyedra cysts following UV treatment as low as 50 mWs cm-2.

Rab35 GTPase and initiation of apico-basal polarity in 3D renal cysts / Rab35 GTPase et initiation de la polarité apico-basal dans un modèle cellulaire 3D

L'établissement de la polarité apico-basale dans les tissus épithéliaux est étroitement lié à la division cellulaire, mais les mécanismes moléculaires sous-jacents n'ont pas encore été établis. A l'aide d'un modèle de culture en 3 dimensions de cellules rénales (MDCK), j'ai montré que lors du développement d'un cyst, la GTPase Rab35 joue un rôle majeur dans l'établissement de la polarité et le positionnement du lumen pendant la première division cellulaire. Au niveau moléculaire, Rab35 permet de coupler l'initiation de la polarité apico-basale avec la cytocinèse via l'attachement au sillon de clivage de vésicules intracellulaires contenant des déterminants clé de l'établissement de la polarité. Ces vésicules transportent notamment les protéines aPKC, Cdc42, Crumbs3 ainsi que le facteur d'ouverture de la lumière Podocalyxin. De plus, l'attachement de ces vésicules au sillon de clivage dépend de l'interaction directe entre Rab35 et la queue cytoplasmique de Podocalyxin. Par conséquence, l'inactivation de Rab35 entraine une inversion complète de la polarité apico-basale des kystes 3D. J'ai mis en évidence un nouveau mécanisme de ciblage des vésicules intracellulaire au site de clivage dépendant de la protéine Rab35 impliqué à la fois dans l'initiation de la polarité apico-basale et dans l'ouverture de la lumière au centre du cyst. / Establishment and maintenance of apico-basal polarity in epithelial organs must be tightly coupled with cell division, but the underlying molecular mechanisms are largely unknown. Using 3D cultures of renal MDCK cells (cysts), I found that the Rab35 GTPase plays a crucial role in polarity initiation and apical lumen positioning during the first cell division of cyst development. At the molecular level, Rab35 physically couples cytokinesis with the initiation of apico-basal polarity by tethering intracellular vesicles containing key apical determinants at the cleavage site. These vesicles transport aPKC, Cdc42, Crumbs3 and the lumen promoting factor Podocalyxin, and are tethered through a direct interaction between Rab35 and the cytoplasmic tail of Podocalyxin. Consequently, Rab35 inactivation leads to complete inversion of apico-basal polarity in 3D cysts. This novel and unconventional mode of Rab-dependent vesicle targeting provides a simple mechanism for triggering both initiation of apico-basal polarity and lumen opening at the centre of cysts.
