Syntheses and Characterization of Novel Materials for Efficacious Anticancer Drug Delivery and Selective Sensing of Bioanalytes

The thesis entitled “Syntheses and Characterization of Novel Materials for Efficacious Anticancer Drug Delivery and Selective Sensing of Bioanalytes” encompasses the syntheses and characterization of various novel materials those are primarily used for efficacious pH-targeted chemotherapy, selective sensing and quantification of ATP inside a single living cell and also for specific sensing of female sex pheromone of certain agriculturally important pests. In recent era of cancer research, pH guided anticancer drug delivery is an emerging field by which not only the drug-sensitive, but also the drug-resistant cancer cell lines can be targeted efficiently. Scientists have paid lot of attentions to this area of research to design biocompatible, pH-responsive drug delivery vehicles, where most of the literatures are end up with complex, elaborated synthetic procedures and use of expensive chemicals. There are only a few reports in the literature on small molecule based drug delivery vehicles, which is not well explored. Herein some of the biocompatible, pH-sensitive lipid and short peptide sequences are synthesized in easy and short synthetic procedures and successfully tested for their efficacious anticancer drug delivery properties by various biophysical and biological techniques. A pH and reduction dual bio-responsive short peptide sequences are also generated in simple steps for the same cause. The formation of different nanostructures from the self-assembly of these short peptides is probed from high level of theoretical calculations and ultimately a well known chemotherapeutic drug, doxorubicin, has been delivered efficiently both to the drug-sensitive and drug-resistant cancer cell lines. In a particular case, in vivo study has also been performed to establish the drug delivery efficacy of those serum-stable vehicles that led to proficient reduction of tumour volume as compared to the free drug. On the other hand, a few of the molecules are synthesized and characterized by various analytical means for the selective sensing and quantification of adenosine 5’-triphosphate (ATP) inside a single living cell. Unique surface functionalized templates are also fabricated over MEMS devices for specific sensing of female sex pheromone of Helicoverpa armigera and Bactocera oleae pest in an agricultural field to detect the early pest infestation. Toward this end, an extensive study on the design, syntheses and characterization of different novel materials is presented below.

Desenvolvimento e fabricação de filmes ultra-­finos, obtidos pela técnica layer-by-layer, para aplicações na entrega direcionada de fármacos e na captura seletiva de bio-­marcadores / Development and fabrication of ultrathin films obtained by layer­-by­-layer, aiming targeted drug delivery applications and the selective capture of biomarkers

O objetivo geral deste trabalho foi explorar a versatilidade de filmes multicamadas de polieletrólitos (PEM) e suas aplicações em sistemas de entrega de drogas e como filmes funcionais para aplicações biomédicas. Filmes PEM montados pela técnica de camada por camada (layer­-by­-layer, LbL), foram explorados em três aplicações principais. Na primeira, foi explorado o desenvolvimento de um protocolo de funcionalização em filmes de poli(alilamina)/poli (estireno sulfonato), PAH/SPS. Os parâmetros de construção do filme para biotinilação dos grupamentos amina do PAH foram otimizados e aplicados na captura e detecção do antígeno específico da próstata (PSA), na concentração de 100 a 0,1 ng/mL, usando pontos quânticos (Qdots). Em comparação com outros trabalhos, este sistema apresentou uma boa sensibilidade na detecção de PSA, dentro do limite de detecção clínica de 0,4 a 0,1 ng/mL. A segunda aplicação envolveu o desenvolvimento de filmes de sacrifício baseados nas interações naturais da mucina submandibular bovina e da lectina, jacalina (BSM/JAC). Filmes de BSM/JAC apresentaram estabilidade quando submetidos a uma ampla faixa de pH (pH 3-­-9) e em solução de alta força iônica (5 M NaCl). A dissolução dos filmes BSM/JAC pôde ser seletivamente desencadeada mediante à incubação em solução de melibiose, 37 °C, pH 7,4, sem apresentar citotoxicidade às células. Na última parte deste trabalho, a incorporação de lipossomos ecogênicos (ELIP) em mochilas celulares foi investigada. Mochilas celulares são \"patches\" de 7­-10 µm de diâmetro que podem ser fabricados por meio de deposição alternada de polímeros utilizando-­-se a técnica de LbL, sobre uma matriz pré­-moldada obtida por fotolitografia, a fim de criar um sistema composto por três multicamadas estratificadas: uma região de liberação, para promover o destacamento do substrato, uma região de carga de droga, e uma região adesiva às células. O uso de ELIP permitiu incorporação de até 9x mais doxorrubicina (DOX) se comparado com o fármaco livre em solução absorvido pelos dos filmes. A liberação de DOX pelos filmes foi monitorado por 25 dias. Mochilas contendo ELIP-­DOX foram então aderidos a monócitos, e sua viabilidade monitorados por 72h. Mochilas vazias mostraram diminuir a proliferação de monócitos ao longo das 72 horas, enquanto mochilas carregadas com ELIP-­DOX mostraram uma diminuição dramática na população celular, apontando uma potencialização dos efeitos da droga pela sua proximidade com as células. / The overall goal of this thesis was to exploit the versatility of polyelectrolite multilayers (PEM) to be applied in drug delivery systems and biofunctionalizable films for biomedical applications. PEM films assembled by the layer-by­-layer technique were explored in three main applications. In the first part of this work, the development of a functionalization protocol of poly(allylamine)/poly(styrene sulfonate), PAH/SPS was explored. The optimal film parameters to the use of biotinylated multilayers were applied for the capture and detection of prostate specific antigen (PSA) protein in the range of 100 to 0.1 ng/mL, by using quantum dots. Compared to previous work, this system presented a good sensitivity for PSA detection that is within the clinical limit range of 0.4 to 0.1 ng/mL. The second application involved the creation of a novel sacrificial multilayer film. Films based in natural interactions of bovine submaxillary mucin and the lectin jacalin, BSM/JAC were assembled. BSM/JAC films showed stability when underwent a wide rage of pH (pH 3 to 9) and high ionic strength (5 M NaCl) solutions. BSM/JAC dissolution could be triggered released by incubation in melibiose at 37 °C in pH 7.4 buffer, without cytotoxicity. In the last part of this work the incorporation of echogenic liposomes (ELIP) into cell backpacks was investigated. Cell backpacks are 7-10 µm diameter patches that can be fabricated through LbL polymer deposition onto a photopatterned array to create a stacked composite of three stratified multilayer systems: a releasable region for easy detachment from the substrate, a drug payload region, and a cell adhesive region. The use of ELIP allowed up to 9x more doxorubicin (DOX) loading when compared to free drug in solution adsorbed through the films. DOX release from films was monitored for over 25 days. ELIP­-DOX backpacks were then attached to mouse monocytes and their viability monitored by 72h. Empty backpacks showed to decrease monocytes proliferation over the course of 72h, while ELIP­-DOX backpacks showed a dramatic decrease in cell population, showing that DOX effects were enhancement in drug potency by its proximity.

Expressão de grupos de genes como marcadores moleculares preditivos de resposta à quimioterapia neoadjuvante com doxorrubicina e ciclofosfamida em pacientes com câncer de mama / Expression of gene groups as predictive molecular markers response to neoadjuvant chemotherapy with doxorubicin and cyclophosphamide in breast cancer patients

Pacientes com câncer de mama localmente avançado são submetidas à quimioterapia neoadjuvante na tentativa de reduzir a dimensão do tumor e aumentar a possibilidade da realização de uma cirurgia conservadora. Nosso grupo identificou previamente através da tecnologia de cDNA microarray, trios de genes, incluindo BZRP, CLPTM1, MTSS1, NOTCH1, NUP210, PRSS11, RPL37A, SMYD2 e XLHSRF-1, cuja expressão era capaz de predizer a resposta à quimioterapia neoadjuvante com doxorrubicina e ciclofosfamida em pacientes com câncer de mama. No presente estudo, avaliamos se a expressão destes genes é reprodutível na identificação de pacientes responsivas e não-responsivas através de RT-PCR em tempo real, que representa uma técnica mais acessível. Avaliamos inicialmente amostras de 28 pacientes anteriormente estudadas (grupo de validação técnica = 23 responsivas e cinco não-responsivas) e a seguir um grupo de 14 novas pacientes (grupo de validação biológica = 11 responsivas e três não-responsivas). Dentre os trios de genes inicialmente identificados, a expressão de RPL37A + XLHSRF-1 + NOTCH1 e RPL37A + XLHSRF-1 + NUP210 classificou corretamente 86% (24/28) das amostras do grupo de validação técnica e 71% (10/14) das amostras do grupo de validação biológica, através de análise de classificação discriminante. Desse modo, esses trios não demonstraram a mesma precisão em comparação com resultados de cDNA microarray. Uma nova análise combinatória foi realizada na procura do melhor modelo preditivo utilizando valores de expressão obtidos por RT-PCR em tempo real. Identificamos então um novo trio, composto pelos genes RPL37A, SMYD2 e MTSS1, cuja expressão classificou corretamente 93% das amostras do grupo de validação técnica (22/23 responsivas e 4/5 não-responsivas) e 79% do grupo de validação biológica (8/11 responsivas e 3/3 não-responsivas). Portanto, o teste apresentou 88% de sensibilidade e especificidade em detectar pacientes responsivas para o total de amostras analisadas. Ao verificarmos o poder de classificação do mesmo grupo de genes, utilizando os valores de expressão pela análise de cDNA microarray, observamos um resultado semelhante (91% de sensibilidade e especificidade em reconhecer as amostras responsivas). Dessa forma, demonstramos que o perfil de expressão gênica obtido com cDNA microarray é reprodutível através do uso de RT-PCR em tempo real. Um estudo integrando um maior número de pacientes e uma plataforma de cDNA microarray mais abrangente pode auxiliar na identificação de um modelo preditivo baseado em grupos de genes mais acurado para antever a resposta ao tratamento com quimioterapia baseada em doxorrubicina. / Patients with locally advanced breast cancer are submitted to primary chemotherapy as an attempt to reduce tumor dimension and increase breast conserving surgery rates. Our group has previously identified through cDNA microarray technology gene trios, including BZRP, CLPTM1, MTSS1, NOTCH1, NUP210, PRSS11, RPL37A, SMYD2 and XLHSRF-1, whose expression was capable of predicting response to neoadjuvant chemotherapy with doxorubicin and cyclophosphamide in breast cancer patients. In the current study, it was evaluated whether expression of these genes is reproducible in the identification of responsive and non-responsive patients by real time RT-PCR, which represents a more accessible technique. We initially evaluated samples from 28 patients earlier studied (technical validation group = 23 responsive and 5 non-responsive) and subsequent to a new 14 patients set (biological validation group = 11 responsive and three non-responsive). Among the initially identified gene trios, RPL37A + XLHSRF-1 + NOTCH1 and RPL37A + XLHSRF-1 + NUP210 expression correctly classify 86% (24/28) samples from the technical validation group and 71% (10/14) samples from the biological validation group, through discriminant classification analysis. Therefore, these trios didnt demonstrate the same precision as compared with cDNA microarray results. A new combinatorial analysis was also performed in search of the best predictive model using real time RT-PCR expression values. A new trio was identified, represented by RPL37A, SMYD2 and MTSS1 genes, whose expression correctly classified 93% samples from technical validation group (22/23 responsive and 4/5 non-responsive) and 79% samples from biological validation group (8/11 responsive samples and 3/3 non-responsive samples). Therefore, the test presented 88% sensibility and specificity in identifying responsive patients for all samples analyzed. By means of verifying the classification strength of the same gene group, using cDNA microarray expression values, we observed a similar result (91% sensibility and specificity in recognizing responsive samples). Thus, we demonstrated that gene expression profile obtained by cDNA microarray is reproducible through real time RT-PCR. A study integrating a larger number of patients and a more comprehensive cDNA microarray platform may help the identification of a more accurate predictive model based on gene groups to foresee response to doxorubicin-based chemotherapy treatment.

Patched, une nouvelle cible thérapeutique pour le cancer de la corticosurrénale / Patched as a new target for adrenocortical carcinoma treatment

Nous avons récemment démontré que le récepteur du morphogène Hedgehog, Patched, qui est exprimé dans de nombreux cancers, est un transporteur de multiples drogues qui contribue à la résistance des cellules cancéreuses à la chimiothérapie. Le criblage d'une banque de molécules nous a permis d'identifier deux molécules qui inhibent l'activité d'efflux de doxorubicine de Patched. Nous avons montré que ces molécules renforcent les effets cytotoxiques, proapoptotiques, antiprolifératifs et anticlonogéniques de la doxorubicine sur les cellules de cancer de la glande surrénale (surrénalome) qui expriment de façon endogène Patched. De plus, nous avons observé que l’ajout de la molécule P375 au traitement à la doxorubicine inhibe le développement des tumeurs chez des souris ayant reçu une xénogreffe de cellules de surrénalome de façon plus significative que la doxorubicine seule. Nos résultats suggèrent que l'utilisation d'un inhibiteur de l'activité d'efflux de drogues de Patched en association avec la doxorubicine est une option thérapeutique prometteuse pour le surrénalome, et très probablement pour d'autres cancers exprimant Patched. Nous avons découvert qu'une petite fraction seulement des cellules de la lignée de surrénalome exprime Patched au niveau de la membrane plasmique (cellules PM-Patched). Les cellules PM-Patched sont plus résistantes à la doxorubicine, et présentent une expression plus élevée de Patched mais aussi de la protéine ABCG2. ABCG2 étant un marqueur de cellules souches cancéreuses (CSC), nous pensons que les cellules PM-Patched pourraient être des CSC.D'autres expériences sont nécessaires pour valider cette hypothèse. / We recently demonstrated that the Hedgehog receptor Patched, which is expressed in many recurrent and metastatic cancers, is a multidrug transporter contributing to chemotherapy resistance. The screening of a chemical library allowed us identifying two molecules which inhibit the doxorubicin efflux activity of Patched. We showed that these molecules enhance the cytotoxic, proapoptotic, antiproliferative and anticlonogenic effects of doxorubicin on adrenocortical carcinoma (ACC) cells which endogenously express Patched. Moreover, we reported that the addition of the drug-like molecule P375 to doxorubicin treatment prevents the development of xenograft ACC tumours in mice much more significantly than the doxorubicin alone. Our results suggest that the use of an inhibitor of Patched drug efflux activity in combination with doxorubicin is a promising therapeutic option for ACC and most likely for other Patched-expressing cancers. We discovered that only a small fraction of the ACC cell line expressed Patched at the plasma membrane (PMPatched cells). We observed that these cells are more resistant to doxorubicin treatment than ACC cells that express Patched only in intracellular compartments. Moreover, we estimated that PMPatched cells have higher expression of Patched but also of ABCG2/BCRP proteins. Based on the fact ABCG2/BCRP is a cancer stem cell (CSC) marker and that Hedgehog signaling is involved in maintenance of CSC, we think that PM-Patched cells could be CSCs. More experiments are needed to confirm this hypothesis.

Despite advances in cancer diagnosis and therapy, cancer is the second leading cause of death globally. The improvements of cancer treatment are the major challenge in this research. The aim of the thesis was studying of effects of two anticancer drugs ellipticine (Elli) and doxorubicin (DOX) on some cancer and healthy cell lines. Specific consideration was given to expand current knowledge about the metabolism and cytostatic effects of Elli in neuroblastoma cell lines. Another part of this study was focused on mechanisms contributing to the development of ellipticine-resistance in cancer cells and influence of histone deacetylase inhibitors on anticancer therapy was investigated. Moreover, the aim was to develop apoferritin (Apo) nanocarrier suitable for the active transport of cytostatics to cancer cells. Several essential data were found in this doctoral thesis. Anticancer efficiency of Elli depends on the CYP3A4-mediated metabolism in cancer. The CYP3A4 enzyme encapsulated into two nanoparticle forms, liposomes and SupersomesTM , was tested to activate ellipticine to its reactive species forming covalent DNA adducts. Des aptamères pour améliorer l’encapsulation et la libération contrôlée des liposomes

Mémoire par article / Hypothèse : L’incorporation dans des liposomes d’aptamères spécifiques à un principe actif permet d’obtenir une encapsulation active du principe actif et de modifier les profils de libération sans diminuer l’efficacité thérapeutique. Méthode : Une série d’aptamères d’affinité variable a été incorporée dans la préparation de liposomes cationiques. Ces lipoplexes ainsi formés ont été caractérisés en taille par diffusion dynamique de la lumière et par la mesure du potentiel de surface zêta. Ils ont été optimisés en matière de complexation maximale des aptamères, puis incubés avec la doxorubicine, choisie comme principe actif modèle. L’efficacité d’encapsulation de la doxorubicine a été comparée avec et sans aptamères, et contre la méthode d’encapsulation active offerte commercialement. Les meilleures formulations ont été étudiées sur le plan de la cinétique de libération et l’efficacité de celles-ci a été évaluée pour leur cytotoxicité sur des cellules cancéreuses de type HeLa. Résultats : Les vecteurs cationiques optimisés permettent la complexation d’au moins 94% des aptamères. Trois des quatre aptamères ont démontré de l’encapsulation active de la doxorubicine, avec des efficacités d’encapsulation allant jusqu’à 85%. De ces trois formulations, différents profils de libération ont été obtenus, permettant tous une libération plus importante qu’une formulation ressemblant aux liposomes commerciaux de doxorubicine (Doxil®). L’efficacité des trois formulations testées sur les cellules HeLa s'est avérée équivalente ou supérieure au standard similaire du Doxil®. / Hypothesis : Incorporation in some liposomes of specific aptamers for a drug allowed the obtaining of active encapsulation of that drug and allowed the modification of their drug release profiles, without negatively impacting its therapeutic efficacy. Methods : A series of aptamers of various affinity were incorporated in the preparation of cationic liposomes. The resulting lipoplexes were characterized for their size by dynamic light scattering method and their surface potential were analysed by zeta potential measurement. Lipoplexes were optimized in terms of highest aptamer complexation and they were incubated with doxorubicin, a model drug that was chosen. The encapsulation efficiency of doxorubicin was compared with and without the presence of aptamers, and with the commercially available active loading method. The best formulations were studied for their doxorubicin’s release kinetic, with the different aptamers, and they were tested for their cytotoxicity on HeLa cancer cells. Results : Cationic liposomes were optimized to allow a minimum aptamer complexation of 94%. Three out of the four tested aptamers were able to demonstrate active loading capabilities, with up to 85% encapsulation efficiencies. Out of these three formulations, very different release profiles were found, all allowing more initial content release of the commercially-like available doxorubicin’s liposomes (Doxil®). The efficacy of those three formulations on HeLa cancer cells demonstrated equivalent or higher cytotoxicity than the similar standard of Doxil®.

Study on Self-Assembly of Fullerenes and Biopolymers

The understanding of self-assembly processes is important for fabrication of well-defined structures with new functionalities for applications in the area of biomedical sciences, material sciences and electronics. In this thesis, two types of self-assembly processes are described: (1) self-assembly of fullerene derivatives in water and (2) self-assembly on surfaces using layer-by-layer (LbL) approach. The various interactions and parameters involved in the self-assembly are detailed in the introductory chapter 1. The various internal parameters like molecular geometry, intramolecular and intermolecular forces that guides the self-assembly process of amphiphiles in water are discussed. The experimental procedures used in the present thesis for the fabrication of nanostructures via self-assembly approach are also described. In the later part of the chapter, the LbL technique for fabrication of thin films and microcapsules is reviewed where various interactions involved in the growth of LbL assembly are discussed. The effect of ionic strength and pH on the growth and property of LbL assemblies is elaborated. A brief discussion of the materials used in the thesis ‒ fullerene, bovine serum albumin (BSA) and nanocrystalline cellulose (NCC) is also provided The self-assembly behaviour of amphiphilic fullerene derivatives are described in chapter 2. Fullerene is anisotropically substituted with five polar hydroxyl groups using organo-copper reagent. The derivative can interact in water via the van der Waals and hydrophobic interactions of the fullerene moiety as well as the intermolecular hydrogen bonding among the hydroxyl groups and also with water. The penta-hydroxy fullerene derivative self-assembles in water as vesicular structures. The size of these vesicles can be varied by modifying the kinetics of self-assembly which was done by changing the rate of addition of non-solvent (water) to the solution of the fullerene derivative. In the second derivative, the hydroxyl groups are substituted with less polar methoxy groups. The penta-methoxy fullerene derivative cannot participate in inter-molecular hydrogen bonding formation unlike the penta-hydroxy derivative but there is possibility of hydrogen bond formation with water where oxygens on methoxy group can act as hydrogen bond acceptor. The penta-methoxy fullerene does not show any vesicle formation in water. The computational simulation studies were carried out on the two fullerene derivatives to understand the self-assembly behaviour of these two derivatives. Furthermore, the vesicle structures formed by the penta-hydroxy fullerene derivative are used for entrapment of hydrophobic polymer, poly[2-methoxy-5-(2-ethylhexyloxy)-1,4-phenylenevinylene] (MEH-PPV) and also hydrophilic dye, Rhodamine B. In both the cases, fluorescence quenching is observed due to electron transfer reaction with fullerene and hence these fullerene vesicles can be used to study the effect of confinement on electron transfer reactions and other chemical dynamics. The layer-by-layer self-assembly approach for the fabrication of biopolymeric thin films and microcapsules is discussed in the chapters 3 to 6. The biocompatible nanoparticles and nanofibers were used as the components of the assembly. In chapter 3, we have described fabrication of thin film of bovine serum albumin (BSA) nanoparticles via LbL approach using biopolymer chitosan as the complementary polymer. The driving force for the assembly growth of the assembly was the electrostatic and complementary hydrogen bond formation between the two components. The idea of incorporating nanoparticles in the thin film was that the nanoparticles can act as reservoirs for functional materials. The films were loaded with anticancer drug doxorubicin and show pH dependent release of the drug. The various interactions involved in the LbL assembly of BSA nanoparticles and polymers were investigated towards understanding the growth mechanism of the assembly in chapter 4. The understanding of the interactions involved in the assembly formation is important in order to modify the conditions of the assembly for enhancing the growth. It is inferred from the study reported in this chapter that not only the interaction of nanoparticles with polymers but also the inter-particle interactions are important factors in determining the growth of LbL assembly of nanoparticles/polymers. The growth of the assembly is enhanced on minimizing the inter-particle repulsions, which was achieved in case of BSA nanoparticles by modifying the pH of the assembly. We also utilized the LbL self-assembly approach for the delivery of lipophilic drugs. The lipophilic drugs are difficult to administer in the body due to their poor water solubility and hence show poor pharmacokinetic profile. The methods for incorporating hydrophobic drugs in LbL assembled thin films and microcapsules are described in chapters 5 and 6. In chapter 5, hydrophobic molecules binding property of albumin has been exploited for solubilisation of a water-insoluble molecule, pyrene (model drug) and hydrophobic drug, curcumin, by preparation of non-covalent conjugates with BSA. The interaction with BSA provided negative zeta potential to the previously uncharged molecules and hence they can be incorporated in the LbL assembled thin films and microcapsules using electrostatic as well as hydrogen bonding interaction with biopolymer, chitosan. The fabrication of protein encapsulated stable microcapsules with hydrophobic molecules incorporated in the shell of the microcapsules has also been demonstrated. The microcapsules were further capable of loading hydrophilic molecules like Rhodamine B. Thus, this approach can be employed for fabrication of multi-agent carrier for hydrophobic and hydrophilic drugs as well as therapeutic macromolecules. In chapter 6, we have incorporated nanocrystalline cellulose (NCC) LbL assembled thin films and microcapsules. The assembly formed was porous in nature due to the nano-fibrous morphology of NCC. The nanoassemblies can act as potential drug delivery carrier, which has been demonstrated by loading anticancer drug doxorubicin, and a lipophilic drug, curcumin. Doxorubicin hydrochloride, the salt form of the drug, doxorubicin, has good water solubility and hence can be postloaded in the assembly by diffusion from its aqueous solution. In the case of curcumin, which has limited solubility in water, a stable aqueous dispersion of the drug was prepared via noncovalent interaction with NCC prior to incorporation in the LbL assembly. The interaction of various other lipophilic drugs with NCC was analysed computationally.

The effects of various combinations of different classes of anticancer drugs and tyrosine kinase inhibitors on the human MCF-7 breast carcinoma cell line

Magister Scientiae (Medical Bioscience) - MSc(MBS) / This study investigated the effects of TKIs on the growth and proliferation of MCF-7 breast carcinoma cells in culture. MCF-7 cells were exposed to different concentrations of TKIs alone and in combination with each other. Inhibition of cell growth by TKIs used individually occurred in a dose- and time-dependent manner. When EGFR Inhibitor I, EGFR Inhibitor II/BIBX1382 and the multi-specific EGFR/ErbB-2/ErB-4 Inhibitor were used in combination with each other at equimolar log dose concentrations, the combined effects on cell growth was significantly different to inhibitors used individually as reflected in a decreased EC50 (IC50) during combination treatments. Generally, for the combinations with DOX, CPL and the TKIs, synergistic as well as antagonistic effects were observed at isoeffective concentrations with resultant decreases in dose reduction indices (DRIs) implying greater efficacies with the respective combinations. In this study, conventional PCR was used to detect and illustrate the presence of the EGFR gene in the samples, while RT-qPCR was used to determine the mRNA expression levels of this gene in MCF-7 breast carcinoma cells

<p>Danas, u raznim analitičkim laboratorijama postoji veći broj analitičkih protokola,<br />zasnovanih bilo na izuzetno sofisticiranim ili jednostavnijim tehnikama, koji služe za<br />određivanje različitih ciljnih analita od farmakolo&scaron;kog značaja. Među tim grupama ciljnih analita pripadaju i antibiotici koji predstavljaju veliko otkriće u oblasti medicine i zahvaljujući njima spa&scaron;eno je vi&scaron;e od sedam miliona života, ali pored navedenih koristi, antibiotici mogu da izazovu&nbsp; veliki broj neželjenih efekata i žučne kiseline zajedno sa svojim derivatima, koji su fiziolo&scaron;ki deterdženti, mogu biti citotoksične za organizam ako se njihova koncentracija ne kontroli&scaron;e. U ovoj doktorskoj disertaciji prikazan je razvoj analitičkih metoda pre svega voltametrijskihmetoda u kombinaciji sa jednostavnim i savremenim elektrodama/senzorima za određivanje&nbsp; odabranih analita kao &scaron;to je antraciklični antibiotik doksorubicin (DOX), makrolidni antibiotici<br />eritromicin-etilsukcinata (EES), azitromicina (AZI), klaritromicina (CLA) i roksitromicina&nbsp; (ROX) i 3-dehidro-deoksiholne kiseline.</p><p>Voltametrijska karakterizacija i određivanje gore navedenih antibiotika primenom obnovljive srebro-amalgam film elektrode (Hg(Ag)FE)rađena je direktnom katodnom&nbsp; voltametrijom sa pravougaonim talasima (SWV) i visoko osetljivom adsorptivnom voltametrijom sa pravougaonim talasima (SW-AdSV) u Briton-Robinson puferu, kao pomoćnom elektrolitu, obuhvatajući &scaron;irok opseg pH vrednosti. Odgovor DOX-a primenom&nbsp; Hg(Ag)FE&nbsp;praćen je u intervalu potencijala od -0,20 do -0,80 V.&nbsp; Za analizu tragova, optimizacija metode ukazuje da su optimalni parametri za analitički pik na potencijalu (Ep ) -0,57 V u odnosu na zasićenu kalomelovu elektrodu (ZKE): pH 6,0, potencijal&nbsp; akumulacije -0,20 V i vreme&nbsp; akumulacije 140 s. U model rastvoru, DOX je određivan u koncentracionom opsegu 4,99-59,64&nbsp; ng mL<sup>-1</sup>. Razvijena SW-AdSV metoda je primenjena za određivanje DOX-a u obogaćenom uzorku humanog urina. Niža koncentracija DOX-a 9,89ng mL<sup>-1&nbsp;</sup>u voltametrijskoj&nbsp; ćeliji je određivana sa relativnom standardnom devijacijom (RSD) manjom od 6,0%. &Scaron;to se ispitivanih makrolida tiče oni su pokazali redukcione signale u dalekoj negativnoj oblasti potencijala. Ispitivanja direktnom katodnom SWV rađena su u opsegu potencijala od -0,75 V do -2,00 V u odnosu na ZKE, pri čemu su dobijena jedan ili dva redukciona pika u opsegu potencijala od -1,5 V do -1,9 V. Oblik i intenzitet signala zavisi od primenjene pH vrednosti u &scaron;irokoj pH oblasti. Za analitičke svrhe, radi razvoja direktne katodne SWV i adsorptivne inverzne/striping SWV metode, pogodnim su se pokazale neutralna i slabo alkalna sredina tj. pH 7,0 sa&nbsp; E<sub>p</sub> na -1,67 V u odnosu na ZKE za ROX i EES i pH 7,2 sa E<sub>p</sub> na -1,85 V u odnosu na ZKE za AZI i pH 7,4 sa E<sub>p</sub> na -1,64 V u odnosu na ZKE za CLA. Na osnovu snimljenih cikličnih voltamograma na&nbsp; optimalnim pH vrednostima, može se predložiti adsorptivno-kontrolisan kinetički proces na elektrodi u slučaju sva&nbsp; četiri ispitivana jedinjenja. Takođe,&nbsp; <sup>1</sup>H NMR merenja uz potiskivanje&nbsp; signala vode u pH oblasti između pH 6,0 i 10,5 ukazuju na to da su makrolidni molekuli pri optimalnim analitičkim uslovima predominantno u protonovanoj formi preko tercijerne amino grupe &scaron;to potpomaže, u sva&nbsp; četiri slučaja, njihovu adsorpciju na odgovarajuće polarizovanoj Hg(Ag)FE. Optimizovane direktne katodne SWV metode&nbsp; pokazuju dobru linearnost u koncentracionom opsegu 4,81-23,3&nbsp; &micro;g mL<sup>-1</sup> , 4,53-29,8&nbsp; &micro;g mL<sup>-1</sup> , 1,96-28,6&nbsp; &micro;g mL<sup>-1</sup> i 1,48-25,9 &micro;g mL<sup>-1</sup> za AZI, EES, CLA odnosno ROX. Razvijene SW-AdSV metode rezultiraju u linearnom odgovoru pri nižim koncentracionim intervalima 1,0-2,46 &micro;g mL<sup>-1</sup> ,&nbsp; 0,69-2,44&nbsp; &micro;g mL<sup>-1</sup>, 0,05-0,99 &micro;g mL<sup>-1</sup> i 0,10-0,99&nbsp; &micro;g mL<sup>-1</sup> , za AZI, EES, CLA i ROX. RSD za sve razvijene metode nije veća od 1,5% izuzev SWV metode u slučaju AZI-a gde je 4,5%. Direktna katodna SWV metoda&nbsp; je uspe&scaron;no primenjena za određivanje EES-a u farmaceutskom proizvodu Eritromicin<sup>&reg;</sup> dok SW-AdSV metoda je primenjena u slučaju određivanja EES-a u obogaćenom uzorku humanog urina i&nbsp;za određivanje ROX-a u farmaceutskom proizvodu Runac<sup>&reg;</sup> . U svim pomenutim slučajevima, primenjena je metoda standardnog dodatka. Pouzdanost i tačnost elaboriranih procedura u slučaju određivanja EES-a u model sistemu i&nbsp; farmaceutskom proizvodu Eritromicin<sup>&reg;</sup> su potvrđena poređenjem sa rezultatima dobijenim primenom HPLC-DAD metode.</p><p>Nakon preliminarnih studija 3-dehidro-deoksiholne&nbsp; kiseline/3-dehidro-deoksiholata primenom elektrode od staklastog ugljenika (GCE), gde je uočeno da ne dolazi do formiranja redukcionog signala u Briton-Robinson puferu između pH 5,0 i 11,8 primenom direktne katodne SWV, bizmut-film je izdvojen&nbsp; <em>ex situ</em> na povr&scaron;ini iste elektrode od staklastog ugljenika (BiF-GCE) iz uobičajeno kori&scaron;ćenog rastvora za elektrodepoziciju (0,02 mol L<sup>-1</sup> Bi(NO<sub>3</sub>)<sub>3</sub>, 1,0 mol L<sup>-1</sup> HCl i 0,5 mol L<sup>-1</sup> KBr) i tako pripremljena elektroda je primenjena za karakterizaciju i određivanje pomenutog jedinjenja u alkalnoj sredini. Redukcioni signal ispitivanog analita od analitičkog značaja je uočen jedino primenom BiF-GCE u Briton-Robinson puferusa pH vrednostima između 9,5 i 11,8 u režimu adsorptivne inverzne/stripingvoltametrije sa pravougaonim talasima, dok u slučaju direktnih katodnih SWV eksperimentalnih uslova uočen je slab redukcioni pik sa niskom strujom maksimuma pika. Optimizovani eksperimentalni uslovi za određivanje 3-dehidro-deoksiholata obuhvataju odgovarajuće kondicioniranje elektrode uključujući kondicioniranje&nbsp; <em>ex situ</em> pripremljene BiF-GCE u Briton-Robinson pomoćnom elektrolitu pH 11,8 do stabilizacije struje bazne linije elektrohemijskim cikliranjem potencijala radne elektrode u potencijalskom opsegu između -1,00 i -2,00 V u odnosu na ZKE (blizu 15 puta) i primenu dva ključna parametara adsorptivne voltametrije sa pravougaonim talasima: vreme akumulacije od 30 s i potencijal akumulacije&nbsp; -1,00 V u odnosu na ZKE. Zbog relativne asimetričnosti dobijenih redukcionih signala ispitivanog analita sa&nbsp; E<sub>p</sub> na -1,35 V u odnosu na ZKE, &scaron;to je takođe prisutno i u slučaju primene SW-AdSV, određivanje ispitivanog analita je zasnovano na linearnoj zavisnosti između povr&scaron;ine pika redukcionog signala&nbsp; spitivanog analita i njegove odgovarajuće koncentracije i postignuta granica detekcije je 1,43 &micro;g mL<sup>-1</sup> sa dva linearna opsega kalibracione krive od 4,76 &micro;g mL<sup>-1</sup> do 13,0 &micro;g mL<sup>-1</sup> i od 13,0 &micro;g mL<sup>-1</sup> do 23,1 &micro;g mL<sup>-1</sup> za razvoj analitičke metode. RSD metode je 3,22%. Dodatni eksperimenti, elektroliza ispitivanog analita na potencijalu -1,55 V (blizu maksimuma pika ciljnog analita) u odnosu na ZKE su rađeni primenom GCE u obliku ploče (povr&scaron;ina 33,52 cm 2 ) modifikovane sa&nbsp; <em>ex situ&nbsp;</em>pripremljenim bizmut-filmom. Rastvor od interesa uzorkovan je na početku eksperimenta, nakon 2,5 h i nakon 4,5 h tretmana. Ovakvi uzorci su analizirani primenom <sup>1</sup>H NMR merenja uz potiskivanje signala vode u puferskom rastvoru pH 11,8. Može se pretpostaviti da tokom elektrolize 3-dehidro-deoksiholata dolazi do redukcije keto grupe prisutne u strukturi ispitivanog analita.</p><p>Na osnovu literaturnih podataka da neki od ciljnihmakrolidnih antibiotika kao &scaron;to je npr. azitromicin pokazuju oksidativno pona&scaron;anje na elektrodi od ugljenične paste i elektrodi od zlata deteljna karakterizacija i određivanje&nbsp; četiri makrolidna antibiotika rađena je primenom&nbsp; asične&nbsp;elektrode od ugljenične paste (CPE) koja se sastoji samo od grafitnog praha i parafinskog ulja sa optimizovanih direktnih anodnih SWV metoda. U slučaju EES-a i AZI-a diferencijalna pulsna voltametrija (DPV) je testirana za iste svrhe. Ključni&nbsp; parametar u slučaju razvoja analitičkih voltametrijskih metoda je odabir pH vrednosti pomoćnog elektrolita gde je oblik/simetričnost i intenzitet oskidacionog pika glavni kriterijum prilikom odabira. Kao odgovarajuće pH vrednosti za voltametrijsko određivanje EES-a primenom SWV metode odabrana je pH 8,0 sa E<sub>p</sub> na 0,83<br />V u odnosu na ZKE, dok u slučaju DPV metode pH 12,0 sa&nbsp; E<sub>p</sub> na 0,55 V u odnosu na ZKE je bila najpogodnija za analitičke svrhe. Za određivanje AZI-a, u slučaju obe SWV i DPV metode pH 7,0 se pokazala najpogodnijom sa E<sub>p</sub> analitičkog signala na 0,85 V odnosno 0,80 V u odnosu na ZKE, dok u slučaju CLA i ROX koji su ispitivani samo primenom SWV metode za analitičke svrhe pH 12,0 je bila najpogodnija sredina sa E<sub>p</sub> analitičkog signala na 0,65 V odnosno na 0,63 V&nbsp;u odnosu na ZKE. Postignute granice detkcije primenom nemodifikovane CPE i direktne anodne SWV su uglavnom u submikrogramskom koncentracionom opsegu 0,17 &micro;g mL<sup>-1</sup> , 0,32&nbsp; &micro;g mL<sup>-1</sup> i 0,30&nbsp; &micro;g mL<sup>-1</sup>, u slučaju EES-a, AZI-a i ROX-a i u niskom mikrogramskom koncentracionom opsegu 1,43&nbsp; &micro;g mL<sup>-1</sup> za CLA. Razvijena SWV metoda sa jednostavnom CPE pokazala se pogodnom za određivanje ROX-a u komercijalnom proizvodu Runac<sup>&reg;</sup> tableti. U slučaju optimizovanih DPV metoda postignute granice detekcije za EES i AZI su u niskom mikrogramskom&nbsp; koncentracionom opsegu 1,03&nbsp; &micro;g mL<sup>-1</sup> odnosno 1,53&nbsp; &micro;g&nbsp; mL<sup>-1</sup> . U želji da se&nbsp;postigne niža granica detekcije za AZI, DPV metoda&nbsp; je testirana u kombinaciji sa&nbsp; CPE radnom elektrodom povr&scaron;inski modifikovanom sa zlatnim nanočesticama&nbsp; prečnika 10 nm (Au-CPE) i&nbsp; postignuta granica detekcije je 0,95&nbsp; &micro;g mL<sup>-1</sup> sa&nbsp; E<sub>p</sub> analitičkog signala na 0,80 V u odnosu na&nbsp; ZKE. RSD metode u slučaju Au-CPE je 3,5%, dok je u slučaju nemodifikovane CPE 6,0%.&nbsp; Linearnost analitičke metode zasnovane na primeni Au-CPE je dva puta &scaron;ira nego u slučaju&nbsp; primene nemodifikovane CPE.</p><p>Na osnovu dobijenih rezultata može se zaključiti da&nbsp; odgovarajuće kombinacije&nbsp; optimizovanih voltametrijskih tehnika sa ekolo&scaron;ki prihvatljivim i lako primenljivim radnim&nbsp; elektrodama, kao &scaron;to su Hg(Ag)FE, BiF-GCE i CPE zajedno sa Au-CPE, rezultuju razvojem&nbsp; pouzdanih analitičkih metoda, kako u oksidacionim tako i u redukcionim proučavanjima, koje&nbsp; često omogućuju&nbsp; određivanje tragova analita od farmakolo&scaron;kog značaja u jednostavnim, a u&nbsp;nekim slučajevima i u složenim sistemima.&nbsp;</p> / <p>Nowadays in different analytical laboratories there is the increasing number of analytical protocols, either based on highly sophisticated or simpler measurements techniques, which serving for determination of different target analytes of pharmacological importance. Among such target groups of the analyte belongs the antibiotics which present a great discovery in the field of medicine and thanks to them were saved more than seven million people but beside to the mentioned great benefits, antibiotics can cause a large number of side effects and bile acids together with their derivatives which are physiological detergents but if their concentration is not<br />controlled they can be cytotoxic to the body. In the present doctoral dissertation the development of analytical methods, primarily analytical voltammetric methods in combination with simple and contemporary electrodes/sensors, for the determination of selected analytes as antracycline antibiotic doxorubicin (DOX), macrolide antibiotics erythromycin ethylsuccinate (EES), azithromycin (AZI), clarithromycin (CLA) and roxithromycin (ROX) and 3-dehydrodeoxycholic acid were performed.</p><p>Voltammetric characterization and determination of the above mentioned antibiotics using a renewable silver-amalgam film electrode (Hg(Ag)FE) was performed by direct cathodic square-wave voltammetry (SWV) and by highly sensitive adsorptive square-wave voltammetry (SW-AdSV) in aqueous Britton-Robinson buffer solutions as supporting electrolyte covering the wider pH range. The Hg(Ag)FE response of DOX was monitored in the potential range between -0.20 and -0.80 V. For the trace level analysis the method optimization showed that the optimal conditions for the analytical peak with peak potential (E<sub>p</sub>) at -0.57 V vs. SCE were: the pH 6.0, the accumulation potential -0.20 V, and the accumulation time 140 s. In the model solutions, DOX was determined in the concentration range of 4.99-59.64 ng mL<sup>-1</sup>. The developed SWAdSV method was applied for the determination of DOX in spiked human urine sample. The lowest concentration of DOX of 9.89 ng mL<sup>-1</sup> in voltammetric vessel was determined with the relative standard deviation (RSD) less than 6%. As for the investigated macrolides, they showed reduction signals in fairly negative potential range. During direct cathodic SWV investigations conducted over the potential range from -0.75 V to -2.00 V vs. SCE, either one or two reduction peaks were obtained in the&nbsp; potential range from -1.5 to -1.9 V. For analytical purposes concerning the development of direct cathodic SWV and adsorptive stripping SWV methods the neutral&nbsp; and slightly alkaline media were suitable as pH 7.0 with E<sub>p</sub> at -1.67 V vs.&nbsp; SCE for ROX and EES and pH 7.2 and pH 7.4 with E<sub>p</sub> at -1.85 V and -1.64 V vs. SCE for AZI and CLA, respectively. Based on the cyclic voltammograms recorded at these pH values, adsorptioncontrolled electrode kinetics process can be proposed for all four investigated compounds. The water suppressed <sup>1</sup>H NMR measurements in the pH range between 6.0 and 10.5 indicated that the macrolide molecules at the optimal analytical conditions are predominantly in protonated form via their tertiary amino groups which supported in all four cases their adsorption on the appropriately polarized Hg(Ag)FE electrode. The optimized direct cathodic SWV methods showed good linearity in concentration ranges 4.81-23.3 &mu;g mL<sup>-1</sup>, 4.53-29.8 &mu;g mL<sup>-1</sup>, 1.96-28.6&nbsp; &mu;g mL<sup>-1</sup>, and 1.48-25.9 &mu;g mL<sup>-1</sup> for AZI, EES, CLA and ROX, respectively. The SW-AdSV methods resulted in the linear responses at lower concentration ranges as 1.0-2.46 &mu;g mL<sup>-1</sup>, 0.69- 2.44 &mu;g mL<sup>-1</sup>, 0.05-0.99 &mu;g mL<sup>-1</sup> and 0.10-0.99 &mu;g mL<sup>-1</sup>, for AZI, EES, CLA and ROX, respectively. The RSD for all developed methods was not higher than 1.5% except the SWV method for AZI with 4.7%. The direct cathodic SWV method was successfully applied for the determination of EES in the pharmaceutical preparation Eritromicin<sup>&reg;</sup>, while SW-AdSV was tested in the case of the spiked urine sample and for determination of ROX in pharmaceutical preparation Runac<sup>&reg;</sup>. In all above cases, the standard addition method was used. The reliability and accuracy of the above procedures in the case of EES determination in model system and pharmaceutical preparation Eritromicin<sup>&reg;</sup> were validated by comparing them with those obtained by means of HPLC-DAD measurements.</p><p>After initial study of 3-dehydro-deoxycholic acid/3-dehydro-deoxycholate by glassy carbon electrode, where the absence of any reduction peak was observed in the Britton-Robinson buffer solutions between pH 5.0 and 11.8 by direct cathodic SWV, a bismuth-film was electrodeposited ex situ on the same glassy carbon electrode surface (BiF-GCE) from the usually used plating solution (0.02 mol L<sup>-1</sup> Bi(NO<sub>3</sub>)<sub>3</sub>, 1.0 mol L<sup>-1</sup> HCl and 0.5 mol L<sup>-1</sup> KBr) and such prepared film-electrode was applied for the characterization and determination of the the target analyte in alkaline media. The reduction signal of analytical importance was observed only by BiF-GCE in Britton-Robinson buffer solutions with pH values between 9.5 and 11.8 in adsorptive stripping square-wave voltammetry working regime, while in the case of the direct cathodic SWV experimental protocol only a very poor reduction peak was obtained. The optimized experimental conditions for the 3-dehydro-deoxycholate determination consist of the optimized electrode conditioning including the electrochemical cycling of the <em>ex situ </em>prepared BiF-GCE potentials in the potential span between -1.0 and -2.0 V vs. SCE (nearly 15 times) in the Britton-Robinson supporting electrolyte pH 11.8 till the stabilization of the baseline current, and the application of two key parameters of the adsorptive square-wave voltammetric protocol: the accumulation time as 30 s and accumulation potential as -1.0 V vs. SCE. Because of the relative asymmetry of the obtained reduction signals of the target analyte with peak E<sub>p</sub> at -1.35 V vs. SCE, which is still present in the case of the SW-AdSV, the quantification of the target analyte was based on the linear correlation between peak area of the reduction signal and its appropriate concentrations, and reached limit of detection is 1.43 &mu;g mL<sup>-1</sup> and with two linear ranges of calibration curve from 4,76 &mu;g mL<sup>-1</sup> to 13.0 &mu;g mL<sup>-1</sup> and from 13,0 &mu;g mL<sup>-1</sup> to 23,1 &mu;g mL<sup>-1</sup> for the development of analytical method. The RSD of the method&nbsp; was 3.22%. Additional experiments were performed applying GCE with rectangular form (area 35.32 cm<sup>2</sup>) modified with ex situ prepared bismuth-film for the electrolysis of the target analyte which was performed at the potential -1.55 V (nearly the peak maxima of the target analyte) vs. SCE. The solution of interest was sampled at the beginning of the experiment, after 2.5 h and after 4.5 h of treatment. Such samples were analysed by simply water suppressing <sup>1</sup>H NMR measurements in the buffered solution at pH 11.8. It can be assumed that during electrolysis of 3-dehydrodeoxycholate the reduction of the keto group present in the structure of the target analyte can be occurred.</p><p>Driven by earlier literature data about the fact that some of the target macrolide antibiotics as e.g. azithromycin showed oxidation behavior at a carbon paste and gold working electrodes detailed characterization and determination of four target macrolide antibiotics were performed on classical carbon paste electrode (CPE) constituted only from graphite powder and paraffin oil with optimized direct anodic SWV methods. In the cases of EES and AZI differential pulse voltammetric (DPV) methods were tested for the same purpose as well. The key parameter in the case of the development of the analytical voltammetric methods is the selection of the pH value of the supporting electrolyte where the shape/simmetry and intensity of the oxidation peak were the criteria. As the appropriate pH value for determination of EES by SWV method the pH 8.0 was selected with E<sub>p</sub> at 0.83 V vs. SCE while in the case of the DPV method the pH 12.0 with E<sub>p</sub> at 0.55 V vs. SCE was the most suitable for analytical&nbsp; purpose. As for AZI determination, in the case of both SWV and DPV methods the pH 7.0 was the most appropriate supporting electrolyte with the Ep of analytical signal at 0.85 V and 0.80 V vs. SCE, respectively, while in the case of CLA and ROX which were investigated only with SWV method for the analytical purposes the pH 12.0 was the most suitable with E<sub>p</sub> at 0.65 V and at 0.63 V vs. SCE. The obtained detection limits applying the bare CPE and the direct anodic SWV are mainly&nbsp; in submicrogram concentration range as 0.17 &mu;g mL<sup>-1</sup>; 0.32 &mu;g mL<sup>-1</sup> and 0.30 &mu;g mL<sup>-1</sup> for EES, AZI, and ROX and in the low microgram concentration range as 1.43 &mu;g mL<sup>-1</sup> for the CLA, respectively. The developed method succesfully tested for the determination of ROX in the commercial formulation, Runac<sup>&reg;</sup> tablet. In the case of the optimized DPV methods the obtained detection limits for EES and AZI are in the low microgram concentration range 1.03 &mu;g mL<sup>-1</sup> and 1.53 &mu;g mL<sup>-1</sup>, respectively. For the improvement of the sensitivity for AZI the DPV method was tested in combination with a&nbsp; CPE working electrode surface modified with gold nanoparticles with diameter of 10 nm (Au-CPE) and reached the limit of detection was 0.95 &mu;g mL<sup>-1</sup> at E<sub>p</sub> of 0.80 V vs. SCE. The RSD of the method in the case of the Au-CPE is 3.5% while in the case of the native CPE 6.0%. The linearity of the Au-CPE based analytical method is twice wider then it is case with the bare CPE applying protocol.</p><p>Based on the obtained results it can be conclude that the appropriate combination of the optimized voltammetric pulse techniques and the environmentally friendly and easy to use working electrodes as Hg(Ag)FE, BiF-GCE and CPE together with Au-CPE resulted in the development of reliable analytical method either in the oxidation or reduction studies, often allowing trace level determination of pharmacological importance target analytes in simpler and in some case complexes systems.</p>
