L’affect de dégoût dans une perspective transmédiale : le cas de The walking dead

Lorsque l’on s’intéresse au dégoût comme forme d’affect, on remarque que son application abonde au niveau des études cinématographiques et des arts visuels. Par contre, peu de chercheurs se sont intéressés à son analyse à travers les médias dont l’institutionnalisation académique est plus récente, comme la bande dessinée, la télésérie et le jeu vidéo. Ce mémoire a pour objectif d’étendre la pratique des études du dégoût comme affect sur ces médias, en s’attardant sur l’analyse de certaines composantes de la franchise transmédiatique The Walking Dead. De plus, comme ce corpus est marqué par une transmédialité qui dépasse les simples récurrences narratives, ce mémoire veut également produire un modèle d’analyse capable de déceler les structures génératives du dégoût qui tendent à migrer ou être partagées entre les médias de la franchise. Ce modèle sera conçu dans un premier temps par l’établissement d’un dialogue entre les études de l’affect appliquées au cinéma et aux arts visuels et les études sur l’intermédialité. La fonctionnalité de ce modèle sera ensuite testée à travers son application sur la bande dessinée, la télésérie et l’un des jeux vidéo de la franchise The Walking Dead. / When we seek for works that focus on disgust as a form of affect, we note that its application abounds in film and visual art studies. However, few researchers dedicated its analysis through media whose academic institutionalization is newer, such as comics, TV series, and video games. This thesis aims to extend the studies of disgust as an affect’s practice on these media, focusing on the analysis of certain components from the transmedia franchise The Walking Dead. Moreover, because this body of media is marked by a transmediality that exceeds simple narrative recurrences, this thesis also wants to produce an analytical model capable of detecting disgust’s generative structures that tend to migrate or be shared between the media of the franchise. This model will be developed initially by the establishment of a dialogue between affect studies applied to cinema and visual arts, and intermedial studies. Then, this model’s functionality will be tested through its application within the comics, the television series and one of the video games of The Walking Dead franchise.

The aim was to provide an overview of the development of justified title achievement, especially in the Pyramid Texts, Coffin Texts and Book of the Dead. The conclusion is that the motive of justification in ancient Egyptian religion evolved along with the concept of Judgment of the Dead with whom he was associated.

presente trabalho tem como objetivo compreender as cenas típicas de heróis caídos em sequência na Ilíada, tendo por base as revisões mais recentes da Teoria Oral cuja ênfase é na tradicionalidade dos poemas homéricos e em sua recepção. Em particular, cotejou-se a estrutura dessa cena típica com a dos catálogos épicos, seguindo a análise estabalecida pelo artigo seminal de Beye (1964). Um dos resultados mais influentes de seu artigo é a demonstração de que a estrutura do Catálogo das Naus e da cena típica dos heróis tombados é a mesma, razão pela qual costuma-se falar de um \"catálogo de mortos\" na Ilíada. Porém, uma discussão da noção de catálogo na poesia épica e se é realmente possível classificar todas as sequências de guerreiros derrotados como tal ainda não foi realizada. Para atingir esse objetivo, esta dissertação fez uso dos recentes desenvolvimentos da Teoria Oral, responsáveis por renovar a interpretação dos catálogos homéricos pelo viés da performance e da oralidade através de múltiplas abordagens. Assim, catálogos podem ser considerados grandes feitos de memória que causam admiração no público, ou como recursos que auxiliam o aedo no ato da composição, ou até mesmo como o princípio unificador de toda a poética homérica. Aplicou-se cada uma dessas abordagens com o objetivo de responder em que medida é possível falar de um catálogo de mortos na Ilíada. / The present work aims to understand the typical scene of multiple fallen heroes in the Iliad, having as its basis recent revisions of the Oral Theory whose enphasis is in the traditionality of the Homeric poems and its reception. Particularly, it contrasts the structure of this typical scene with the structure of the epic catalogue, following the analysis established by the seminal article of Beye (1964). One of the most influential results of his article was the demonstration that the structure of the Catalogue of Ships and the typical scene of fallen heroes is the same, which is the reason why it is common to speak of a \"catalogue of the dead\" in the Iliad. However, a discussion of what is a catalogue in epic poetry and if it\'s really possible to classify all the sequences of defeated heroes as such has not happened yet. Seeking to fufill that purpose, this dissertation uses more recent developments of the Oral Theory. They are responsible for renovating the interpretation of the Homeric catalogues by using multiple approaches that centers around performance and orality. A catalogue may be perceived as a great feat of memory that astonishes the audience, a tool that help the singer in the act of composition and even as the unifying principle of the Homeric poetics. Each one of this approaches were used with the objective of answering to what extent it is possible to talk about a catalogue of the dead in the Iliad.

Characterizing the Role of the DEAD-box Protein Dbp2 in RNA Structure Remodeling and Pre-mRNA Processing

RNA helicases are found in all kingdoms of life, functioning in all aspects of RNA biology mainly through modulating structures of RNA and ribonucleoprotein (RNP) complex. RNA structures have fundamental impacts on steps in gene expression, including transcription, pre-mRNA processing, and translation. However, the precise roles and regulatory mechanisms of RNA structures in co- and post-transcriptional processes remain elusive. By probing genome-wide RNA structures in vivo, a recent study suggested that ATP-dependent factors, such as RNA helicases, maintain the actively unfolded state of RNAs. Among all RNA helicases, DEAD-box proteins form the largest family in eukaryotes, and have been shown to remodel RNA/RNP structures both in vitro and in vivo. Nevertheless, for the majority of these enzymes, it is largely unclear what RNAs are targeted and where they modulate RNA/RNP structures to regulate co-transcriptional processes. To fill the gap, my research focused on identification of the RNAs and structures targeted by the DEAD-box protein Dbp2 in S. cerevisiae to uncover the cellular processes that Dbp2 is involved in.<br><div><div>My studies revealed a role of Dbp2 in transcriptional termination. Dbp2 binds to ~34% of yeast mRNAs and all snoRNAs, and loss of DBP2 leads to a termination defect as evidenced by RNA polymerase II (RNAPII) accumulation at 3’ ends of these genes. In addition, the binding pattern of Dbp2 in mRNAs is highly similar to Nrd1 and Nab3 in the Nrd1-Nab3-Sen1 (NNS) termination complex, and deletion of DBP2 leads to reduced recruitment of Nrd1 to its target genomic loci. In Dbp2 and NNS targeted 3’ UTRs, RNA structural changes resulted from DBP2 deletion also overlap polyadenylation elements and correlate with inefficient termination, and loss of stable structure in the 3’ UTR bypasses the requirement for Dbp2. These findings lead to a model that Dbp2 promotes efficient termination of transcription through RNA structure remodeling.</div><div>Interestingly, my research also revealed the requirement of DBP2 for efficient splicing, as loss of DBP2 leads to accumulation of unspliced pre-mRNAs. Moreover, this function is dependent on the helicase activity of Dbp2. Further studies are needed to characterize the molecular mechanism of how Dbp2 facilitates splicing in cells. Overall, my research demonstrated that DEAD-box RNA helicases remodel mRNA structure in vivo and that structural alteration can be essential for proper gene expression.</div></div>

O objetivo desse estudo foi verificar os efeitos do treinamento resistido agudo (única sessão - TA) e de 5 semanas de exercício crônico (TC) sobre a apoptose de neutrófilos e linfócitos de camundongos controle e nocaute para TNFR1. Determinamos: viabilidade, externalização de fosfatidilserina, fragmentação do DNA e potencial de membrana mitocondrial. O ensaio do conteúdo de lipídeos neutros foi realizado em neutrófilos e a proliferação em linfócitos. TA e TC não alteraram os parâmetros avaliados nos neutrófilos. Não houve alteração nos linfócitos dos camundongos que realizaram TA. Porém, o TC aumentou a porcentagem de linfócitos com externalização de fosfatidilserina e diminuiu a proliferação dessa células nos camundongos selvagens. O exercício crônico aumentou a proporção de linfócitos em apoptose e diminuiu a proliferação nos camundongos nocaute para TNFR1. Concluímos então que a via do TNFR1 está envolvida neste processo. / The aim of this study was to investigate the acute or chronic effects of resistance exercise training on apoptosis of neutrophils and lymphocytes from knockout mice for TNF-<font face=\"Symbol\">a receptor 1 and control mice. The following parameters were determined viability, externalization of phosphatidylserine on the plasma membrane, DNA fragmentation and mitochondrial membrane potential. The measurement of the content of neutral lipids was performed in neutrophils only and the proliferation assay in lymphocytes. The acute and chronic exercise did not alter the parameters evaluated in neutrophils. Acute exercised also did not affect lymphocyte function. However, chronic resistance exercise increased the percentage of lymphocytes with phosphatidylserine externalization and decreased lymphocyte proliferation capacity of wild type mice. Chronic exercise raised the proportion of lymphocytes in apoptosis and decreased lymphocyte proliferation in TNFR1 knockout mice. We concluded that the effect of chronic exercise on lymphocyte death involves the TNFR1 pathway.

Estudaram-se as perdas fetais ocorridas no distri to de São Paulo, no período de dois anos (1 de junho de 1968 a 31 de maio de 1970) e cuja residência da mãe se localizasse também no distrito de São Paulo. O trabalho objetivou o estudo das mesmas segundo as variáveis fornecidas pelos atestados dos nascidos mortos, que se constituiram em seu material básico. O ponto de vista social foi focalizado através dos aspectos referentes à cor, filiação, naturalidade e profissão dos pais. Os itens relativos a sexo, duração da gestação, local onde ocorreu o evento, história obstétrica, idade da mãe e causa da perda constituiram o conjunto de dados para análise do ponto de vista médico-estatístico. Quanto ao problema da causa, é de se destacar que esta é a primeira vez que tal aspecto é analisado entre nós. Utilizando dados obtidos atravês da \"Investigação Interamericana de Mortalidade na Infância\", foi possível obter o coeficiente corrigido de mortalidade perinatal. A pesquisa visou ainda ao estudo das implicações que as perdas fetais acarretam no campo Jurídico. Quanto aos resultados, devem ser destacados o problema do registro de perdas por local de ocorrência e não de residência, a maior proporção de ilegitimidade nas perdas fetais precoces e a maior proporção de pais naturais da região leste do Brasil. A razão de masculinidade se mostrou maior quanto menor fosse o tempo de gestação. Alguns dados permitiram inferir que existe uma sub-enumeração de perdas precoces e intermediárias. Verificou-se ainda que, aproximadamente, 86% das perdas ocorreram em hospitais,que a quarta parte dos casos estudados foi produto de primeira gestação e que em 85% das perdas o parto foi normal. Relativamente à idade da mãe, foi verificado que o risco de vir a ter uma perda - excetuando-se o grupo de mulheres de menos de vinte anos - aumenta com a idade materna. Em face de erros e imperfeições quanto ao preenchimento do atestado e da ausência de alguns dados de grande interesse para a Saúde Pública, foi sugerida a adoção de novo modelo de atestado de óbitos perinatais. / Foetal loss of resident women of the district of Sao Paulo that occurred in this area, were studied for a two years period, from June 1st, 1968 to May 31st, 1970. The objectives of this study were: - analysis of the foetal mortality differentials according to the available information in the certificates of stillbirths; - analysis of legal involvements pertinent to the foetal deaths. The variables considered were: - social: color, legitimacy, parents birth place and their occupation; - medical-demographic: sex, lenght of pregnancy, birth-order, single or plural deliveries, age of mother, and cause of death. Data available in the Inter American Investigation in Childhood permitted the calculation of the Perinatal Mortality rate. The conclusions were: - a large proportion of illegitimacy observed among the early foetal deaths - the sex-ratio (x 1000 women) was higher in the early pregnancy\'s foetal loss; - there is an under-registration of foetal loss occurred before 28 weeks of gestation; - 86% of the foetal loss occurred in hospitals; - 25% of foetal loss occurred in women pregnant for the first time; - 11% of the foetal loss were delivered by Caesarean section; - the risk of having a foetal loss increases with the increasing of the mother\'s age. The recommendations were: - the tabulation of the stillbirth certificate data should be done by local of residence instead of by occurrence; - a new model of Perinatal death certificate was proposed in order to provide more complete data that are important from the point of view of Public Health.

Planejamento probabilístico com becos sem saída / Probabilistic planning with dead-ends

Planejamento probabilístico lida com a tomada de decisão sequencial em ambientes estocásticos e geralmente é modelado por um Processo de Decisão Markoviano (Markovian Decision Process - MDP). Um MDP modela a interação entre um agente e o seu ambiente: em cada estágio, o agente decide executar uma ação, com efeitos probabilísticos e um certo custo, que irá produzir um estado futuro. O objetivo do agente MDP é minimizar o custo esperado ao longo de uma sequência de escolhas de ação. O número de estágios que o agente atua no ambiente é chamado de horizonte, o qual pode ser finito, infinito ou indefinido. Um exemplo de MDP com horizonte indefinido é o Stochastic Shortest Path MDP (SSP MDP), que estende a definição de MDP adicionando um conjunto de estados meta (o agente para de agir ao alcançar um estado meta). Num SSP MDP é feita a suposição de que é sempre possível alcançar um estado meta a partir de qualquer estado do mundo. No entanto, essa é uma suposição muito forte e que não pode ser garantida em aplicações práticas. Estados a partir dos quais é impossível atingir a meta são chamados de becos-sem-saída. Um beco-sem-saída pode ser evitável ou inevitável (se nenhuma política leva do estado inicial para a meta com probabilidade um). Em trabalhos recentes foram propostas extensões para SSP MDP que permitem a existência de diferentes tipos de beco-sem-saída, bem como algoritmos para resolvê-los. No entanto, a detecção de becos-sem-saída é feita utilizando: (i) heurísticas que podem falhar para becos-sem-saída implícitos ou (ii) métodos mais confiáveis, mas que demandam alto custo computacional. Neste projeto fazemos uma caracterização formal de modelos de planejamento probabilístico com becos-sem-saída. Além disso, propomos uma nova técnica para detecção de becos-sem-saída baseada nessa caracterização e adaptamos algoritmos de planejamento probabilístico para utilizarem esse novo método de detecção. Os resultados empíricos mostram que o método proposto é capaz de detectar todos os becos-sem-saída de um dado conjunto de estados e, quando usado com planejadores probabilísticos, pode tornar esses planejadores mais eficientes em domínios com becos-sem-saída difíceis de serem detectados / Probabilistic planning deals with sequential decision making in stochastic environments and is modeled by a Markovian Decision Process (MDP). An MDP models the interaction between an agent and its environment: at each stage, the agent decides to execute an action, with probabilistic effects and a certain cost which produces a future state. The purpose of the MDP agent is to minimize the expected cost along a sequence of choices. The number of stages that the agent acts in the environment is called horizon, which can be finite, infinite or undefined. An example of MDP with undefined horizon is the Stochastic Shortest Path MDP, which extends the definition of MDP by adding a set of goal states (the agent stops acting after reaching a goal state). In an SSP MDP the assumption is made that it is always possible to achieve a goal state from every state of the world. However, this is a very strong assumption and cannot be guaranteed in practical applications. States from which it is impossible to reach the goal are called dead-ends. A dead-end may be avoidable or unavoidable (when no policy leads from the initial state to the goal with probability one). Recent work has proposed extensions to SSP MDP that allow the existence of different types of dead-ends as well as algorithms to solve them. However, the detection of dead-end is done using: (i) heuristics that may fail to detect implicitly dead-ends or (ii) more reliable methods that require a high computational cost. In this project we make a formal characterization of probabilistic planning models with dead-ends. In addition, we propose a new technique for dead-end detection based on this characterization and we adapt probabilistic planning algorithms to use this new detection method. The empirical results show that the proposed method is able to detect all dead-ends of a given set of states and, when used withprobabilistic planners, can make these planners more efficient in domains with difficult to detect dead-ends.
