I metaphorically link disparate pieces of information to the human condition in order to present multiple readings through juxtaposition. I assemble both abstract and figurative subject matter in a collage-like manner through methods of cropping and fragmentation to symbolize the disjuncture I feel is indicative of how we experience the world through short-lived thoughts, feelings, and actions.

Spatial and Temporal Bacterial Community Dynamics in Constructed Wetland MesocosmsWeber, Kela January 2009 (has links)
The objective of this work was to understand microbial population density and diversity, both spatially and temporally, in wetland mesocosms to gain a better fundamental understanding for use in the optimization and design of constructed wetlands (CWs).
A standardized community level physiological profiling (CLPP) data analysis protocol was adapted and utilized for CW mesocosms. A new one-dimensional metric was developed to track community divergence using BIOLOGTM ECO plate data. The method proved easy to use, did not require a background in multivariate statistics, and accurately described community divergence in mesocosm systems.
To study mesocosm biofilm-bound bacterial communities an appropriate detachment protocol was required. Various shaking protocols were evaluated for their effectiveness in the detachment of bacteria from mesocosm pea gravel, with a focus on detachment of viable and representative bacterial communities. A protocol based on mechanical shaking with buffer and enzymes was identified as an optimal approach and used further in this study. The bacterial communities associated with the interstitial water, pea gravel media, and rhizospheric regions from both planted and unplanted CW mesocosms were profiled using the CLPP method and compared. Vertical community stratification was observed for all mesocosm systems. Rhizospheric communities were found to be significantly more active than their gravel-associated counterparts, suggesting that although rhizospheric bacteria were less abundant in the mesocosms they may play a more significant role in the removal and fate of water born contaminants.
The start-up dynamics of CW mesocosms was investigated using the CLPP and standard CW characterization methods over an eight month period. All mesocosms showed a steep increase in interstitial community divergence until day 75-100, at which point a steady-state was reached. The interstitial communities were also characterized in terms of similarity based on experimental design treatments (planted/unplanted and origin of seeding inoculum). Four stages were identified during the start-up consisting of an initial stage where mesocosm communities were differentiated based on origin of the inoculum, a period where adjustments and shifts occurred in all mesocosm, a time where all mesocosm communities were quite similar, and a final state where community differentiations were made based plant presence in the mesocosms.

Reflecting on a period of change in a governmental development agency : understanding management as the patterning of interaction and politicsMukubvu, Luke January 2012 (has links)
Management was once described as the art of getting things done through the efforts of oneself and other people (Follett, 1941) and is functionalised through acts of planning, organising, leading and controlling tasks and people for pre-defined objectives. These four cardinal pillars of management are translated into various models, tools and techniques of best practice of how to manage. While acknowledging that the substance of the current management models, tools and techniques have for years broadly contributed to how organisations are run, my research sheds more light on the shortcomings underlying some of the assumptions and ways of thinking behind these models and tools. My research findings based on my experience in working for the Department for International Development suggests that management practice and organisational change occur in the context of human power relationships in which people constrain and enable each other on the basis of human attributes such as identities, attitudes, values, perceptions, emotions, fears, expectations, motives and interests. I argue that these human attributes, human power relations and the totality of human emotions arise in the social, and understanding the ways in which these attributes shape local interaction and daily human relating is critical in making sense of the reality of organisational change and management. I suggest that management practice occurs in the context of everyday politics of human relating. It is that type of politics that takes place within families, groups of people, organisations, communities, and indeed throughout all units of society around the distribution of power, wealth, resources, thoughts and ideas. This way of thinking has enormous implications for the way we conceptualise management theory and practice. I am suggesting that managers do not solely determine, nor do employees freely choose their identities, attitudes, values, perceptions, emotions, fears, expectations and motives. These human dimensions arise from social relationships and personal experiences. As such, it is simply not for a manager to decide or force other employees on which of these human attributes to influence their behaviour. I am arguing that the social nature of management practice and role of human agents is inherently complex and cannot, in the scientific sense, be adequately reduced to discrete, systematic, complete and predictive models, tools and techniques without losing some meaning of what we do in management.

Tectonic evolution of Aegean metamorphic core complexes, Andros and Tinos Islands, GreeceShin, Timothy Andrew 10 October 2014 (has links)
The Aegean is a classic setting for studying exhumation of high-pressure (HP) metamorphic rocks. Two end-member models are proposed to explain the uplift of these rocks: core-complex style extension along low-angle normal faults and extrusion-wedge uplift. Extrusion-wedge underplating is the mechanism that exhumed HP rocks on Evia whereas Tinos hosts several detachments varying in age from 30-9 Ma. Andros, situated between them, may be the geological manifestation of the interplay of these processes and provides an opportunity to test these models. Detachments on NW Tinos and on Andros and the enigmatic low-angle Makrotantalon Unit contact on Andros were insufficiently dated prior to this study. Geo- and thermochronometrycombined with structural observations from sampling transects in the transport direction from (1) lower plate Cycladic Blueschist Unit on Andros and Tinos, (2) middle plate Makrotantalon Unit on Andros, and (3) hanging wall Upper Unit address these issues.
Maximum depositional ages from detrital zircon U-Pb geochronometry and structures reveal Paleocene-Eocene syn-HP metamorphism thrusting resulted in an inversed-age relationship between the Permian Makrotantalon Unit and the underlying Triassic-Eocene Cycladic Blueschist Unit on Andros. The Makrotantalon Unit has an internal inversed stratigraphy whereas the Cycladic Blueschist Unit on Andros and Tinos appear stratigraphically intact. Structures and zircon and apatite (U-Th)/He ages in transects from NW Tinos (~12-8 Ma) and central Andros Cycladic Blueschist Unit (~13-7 Ma) indicate rapid cooling due to exhumation associated with the Livada Detachment. Older cooling ages (~16-10 Ma) and structures in the Makrotantalon Unit indicate later brittle strain localization on the Makrotantalon Thrust contact is accommodated by rheologically weaker serpentinites and calc-schists, resulting in slivering of the footwall under the Livada Detachment on Andros.
Estimated mean cooling slip rates of the Livada Detachment on Andros of ~3.8 (+1.2/-1.3) km/Myr and 2.1 (+0.2/-0.2) km/Myr on NW Tinos resulted in minimum vertical exhumations of 15 km and 4 km, respectively. The NCDS here accommodated ~12-25% of 60 km of HP-rock exhumation from ~30-7 Ma. We present a tectonic model to elucidate the evolution of the Makrotantalon Unit and the magnitude, temporal, and spatial variability of exhumation via detachments on these islands.

L’objectif général de ce travail doctoral était d’identifier des leviers d’action permettant de préserver la santé psychologique des travailleurs, de réduire certaines de leurs attitudes et comportements néfastes pour les organisations de santé, et de comprendre les mécanismes explicatifs de ces relations. La première étude a démontré que le climat de sécurité psychosociale (i.e., PSC) était négativement associé au conflit travail-famille (i.e., WFC) et aux intentions de turnover des soignants via la frustration des besoins psychologiques. La seconde étude a mis en évidence que le PSC, via son influence sur la frustration des besoins, était associé à une réduction du burnout des soignants trois mois plus tard qui expliquait à son tour une augmentation du WFC et des intentions de turnover. La troisième étude a démontré que la dissonance émotionnelle et les ressources organisationnelles influençaient le manque de détachement psychologique et l’engagement au travail des professionnels d’encadrement au terme d’une période de trois mois, via leurs effets sur la frustration et la satisfaction des besoins. Ces trois études enrichissent la littérature scientifique sur la théorie de l’autodétermination en contexte de travail et alimentent la réflexion sur les pratiques organisationnelles susceptibles de modifier durablement l’expérience professionnelle des salariés du secteur / The general purpose of this doctoral dissertation was to contribute to identify triggers to preserve workers’ psychological health and reduce some of their negative attitudes and behaviors which appear to be harmful to healthcare organizations. Moreover, we aimed to understand the mechanisms underlying such effects. Study 1 used a cross-sectional design to demonstrate that psychosocial safety climate (i.e., PSC) was negatively related to nursing staff’s work-family conflict (i.e., WFC) and turnover intentions, through the mediation of psychological need thwarting. Study 2 developed these results by showing how PSC, through its influence on psychological need thwarting, related to a decrease in burnout three months later. Burnout itself explained an increase in WFC and turnover intentions. Study 3 focused on managers’ psychological health and found that emotional dissonance and organizational resources influenced managers lack of psychological detachment and work engagement three months later, through psychological need thwarting and satisfaction. Altogether, these studies contribute to self-determination theory in the work setting and offer some perspectives on organizational practices that could lastingly alter healthcare employees’ professional experience

L’architecture crustale et l’évolution de la partie profonde des marges passives peu-magmatiques sont encore mal comprises. En prenant comme chantier principal les marges Australie-Antarctique, cette thèse montre que l’enregistrement de la déformation dans les sédiments met en évidence une évolution polyphasée des marges distales. Cette évolution polyphasée implique le développement de multiples systèmes de détachement présentant une organisation hors-séquence et menant à une architecture finale symétrique des domaines exhumés. Cette organisation des systèmes de failles est liée à des cycles de délocalisation / relocalisation de la déformation influencés par l’apport magmatique, par un niveau de découplage et par la remontée asthénosphérique. L’interaction faille / magma apparait particulièrement importante dans l’évolution des marges distales. Cette étude a permis de mieux caractériser la rupture lithosphérique, qui peut se définir comme un événement tectono-magmatique progressif. / The crustal architecture and evolution of the deepest part of magma-poor rifted margins is currently not well understood. Taking the Australia-Antarctica margins as main study area, this thesis shows that the record of the deformation in sediments highlights a clear polyphase evolution of distal margins. This polyphase evolution implies the development of multiple detachment systems presenting an out-of-sequence organization and leading to a final symmetric architecture of the exhumed domains. This organization of fault systems is linked to cycles of delocalisation / re-localisation of the deformation influenced by the magmatic supply, by a decoupling level and by the asthenospheric uplift. Fault / magma interaction appears particularly important during the evolution of distal margins. This study allowed a better characterization of the lithospheric breakup, which can be defined as a progressive tectono-magmatic event.

Cette thèse est centrée sur la caractérisation des traceurs des fluides qui interagissent avec les roches du socle et les roches sédimentaires dans les systèmes riftés hyper-amincis exposés dans la Téthys alpine, les Pyrénées et Ibérie-Terre Neuve. L’étude de ces fluides est basée sur les observations géologiques, les analyses géochimiques et les données géophysiques. Deux types de fluides ont été identifiés : les fluides associés à la croûte continentale, avec une signature caractérisée par Si et Ca, ainsi que les fluides liés au manteau en exhumation, avec une signature caractérisée par Si, Mg, Fe, Mn, Ca, Ni, Cr et V. La percolation des fluides est fortement liée à la formation des failles de détachement et à l’évolution des systèmes hyper-amincis. Le flux de fluides dans ces systèmes a des implications importantes pour les changements rhéologiques, pour la nature des sédiments et pour les modifications chimiques des réservoirs de la Terre. / This thesis focus in the identification of geochemical tracers and effects of fluid that interact with basement and sedimentary rocks in hyperextended systems. The investigation of such fluids is based on geological observation, geochemical analyses and geophysical data from fossil hyperextended rift systems exposed in the Alps and in the West Pyrenees, and the present-day distal margins of Iberia and Newfoundland. Two types of fluids were identified during this study. The first type, referred to as continental crust-related fluids, has a signature of Si and Ca. The second type, referred to as mantle-related fluids, has a signature of Si, Mg, Fe, Mn, Ca, Ni, Cr and V. The fluid percolation is strongly related to the formation of extensional detachment faults and the evolution of hyperextended systems. Fluid flow in these systems has major implications for the nature of sediments, rheological changes and chemical modifications of the Earth’s reservoirs throughout its evolution.

Role of AMP-Activated Protein Kinase in Cancer Cell Survival under Matrix-Deprived ConditionsSaha, Manipa January 2015 (has links) (PDF)
Cancer progression is a multi-step process requiring cells to acquire specific properties that aid the neoplastic growth. One such property is the ability to survive in the absence of matrix-attachment, a critical necessity for cells to traverse in circulation and seed metastases. Therefore, understanding the signalling mechanisms that protect cells from undergoing death in matrix-deprived condition, termed as anoikis, is important. We have used two systems to study this, one involving experimental transformation model, and another involving cancer cell lines.
In the in vitro transformation model system involving the serial introduction of oncogenes, the ability to survive in anchorage-independent condition and generate spheres/colonies was dependent on the presence of the Simian Virus Small T antigen, SV40 ST. We identified that the viral antigen mediates its effects, at least in part, by activating the master metabolic regulator and cellular stress kinase AMP-activated protein kinase (AMPK) leading to maintenance of energy homeostasis.
Consistent with this, our lab has previously identified both activation of AMPK upon matrix-deprivation in breast cells, as well as its requirement for survival under these conditions. However, a pathway often associated with survival under matrix-deprivation is the PI3K/Akt pathway. Surprisingly, we observed an AMPK-dependent decrease in Akt activity under conditions of matrix-detachment. Since this was contrary to the general notion, we probed deeper into a possible crosstalk between these two kinases. Our work revealed that AMPK activation in suspension inhibits Akt via upregulation of a known Akt phosphatase, pleckstrin homology domain leucinrich repeat protein phosphatise (PHLPP). We further show that the AMPK-PHLPP-Akt signalling axis is important for anoikis-resistance and metastasis. In addition, our results point to a yet unidentified protumorigenic role of PHLPP in breast cancer progression.
With an aim to identify cellular proteins differentially regulated upon AMPK activation in breast cancer cells, we undertook a proteomics approach. Using 2-dimensional gel electrophoresis followed by mass spectrometric analysis, we identified some candidate proteins. We have validated the increase in levels of one of these proteins, annexin A2, in cancer cells upon AMPK activation.
In summary, the present study unveils novel oncogenic functions of AMPK in cancer cells under the stress of matrix-deprivation. Furthermore, our results elucidate a double-negative feedback loop between two critical cellular kinases AMPK and Akt, and also identify a novel pro-tumorigenic role of PHLPP in breast cancer. In addition, we identify PHLPP and annexin A2 as novel proteins upregulated by AMPK in cancer cells. Thus, our results begin to identify pathways utilised by cancer cells to aid anchorage-independent growth, a critical step for cancer metastasis. Based on our results, inhibition of AMPK or perturbation of signalling axes involving AMPK, and PHLPP or annexin A2 might be considered as novel therapeutic approaches to combat cancer progression

Abstract
The tube flow fractionation method used in this thesis is one of the field flow separation processes, which can be used to fractionate particles over the entire size range of pulp particles (1–5000 µm) with high selectivity. The classification of particles in different size categories enables the fractional analysis of samples. According to this thesis, particle length is the most significant individual shape factor in particle classification for different size categories using tube flow fractionation.
The fractional approach investigated in this thesis offers an interesting tool for investigating different pulp types more thoroughly than many other conventional laboratory methods such as the Bauer-McNett classification of pulp or the hyperwashing of DIP samples. The tube flow fractionator can be used to replace the Bauer-McNett device to determine the mass percentages of pulp sample fractions. Furthermore, pulp fractionation can be done quickly with the tube flow fractionation method and the results achieved are also comparable to the Bauer-McNett classification.
The utilisation of recycled raw material in paper production is likely to continue to increase in the near future. This fact leads to even more heterogeneous raw material becoming one of the major challenges in deinked pulp production. Changes in the quality of raw material such as age or paper type have a notable effect on the deinkability of pulp. This creates a need to develop new methods for measuring DIP properties; in particular, information about the ink-releasing rate and ink particle size would be useful in controlling the deinking process. If DIP is fractionated with tube flow fractionation, it is possible to study the ink size distribution and ink content inside each fraction, thus obtaining fractional information on the ink release rate and ink particle size.
A comparison of different DIP raw materials and process solutions can be performed with the fractional analysis presented in this thesis. It was noticed that aging had a negative effect on the slushing rate of LWC and SC raw material, but not on ONP. The ink detachment of LWC and SC was poorer when furnishes aged. However, the aging of LWC and SC did not result in a significant fragmentation of ink particles. In contrast, strong ink fragmentation is typical of aged ONP furnishes. The fractional approach gives new insight into deinking mechanisms, and could be essential for finding optimal deinking process conditions, for example in a pulping process or flotation environment. The method would also enable enhanced control of different DIP process stages and would contribute to creating a more efficient process with better waste management and improved final product quality. / Tiivistelmä
Tässä työssä käytetyllä putkivirtausfraktioinnilla, joka on eräs kenttävirtausfraktioinnin sovellus, voidaan luokitella hyvin selektiivisesti erikokoisia (1–5000 µm) partikkeleita. Partikkeleiden luokittelu partikkelikooltaan erikokoisiin jakeisiin mahdollistaa näytteiden fraktionaalisen analyysin. Tässä työssä todettiin, että partikkelin pituus on tärkein yksittäinen muototekijä, joka vaikuttaa partikkelien fraktioitumiseen eri kokoluokkiin putkivirtausfraktioinnissa.
Työssä tutkittu fraktionaalinen menetelmä mahdollistaa paljon syvällisemmän paperisulppujen analyysin kuin monet perinteiset laboratorioanalyysit, kuten Bauer McNett -luokittelu tai siistausmassojen hyperpesu. Putkivirtausfraktionaattorilla voidaan korvata Bauer McNett -luokittelu massasulppujen eri fraktioiden massaosuuksien määrityksessä. Lisäksi massojen fraktiointi putkivirtausfraktionaattorilla on nopea ja luotettava menetelmä, ja saadut tulokset ovat vertailukelpoisia Bauer McNett -luokittelulla saatujen tulosten kanssa.
Kierrätyspaperiraaka-aineen kasvava hyödyntäminen paperin tuotannossa jatkunee lähivuosina. Tämä tarkoittaa, että siistausprosessissa hyödynnettävän raaka-aineen laatuvaihtelut aiheuttavat entistä suurempia haasteita. Muutokset raaka-aineen laadussa, kuten paperin ikä tai käytetty paperityyppi vaikuttavat huomattavasti massan siistattavuuteen. Näistä tekijöistä johtuen on tarvetta kehittää uusia mittausmenetelmiä määrittää siistausmassojen ominaisuuksia. Erityisesti musteen irtoaminen ja mustepartikkeleiden kokojen määritys voisi auttaa musteenpoistoprosessin ohjauksessa. Jos siistausmassa fraktioidaan putkivirtausfraktionaattorilla, on mahdollista tutkia mustekokojakaumia ja mustepitoisuuksia eri fraktioissa ja näin saadaan tietoa mustepartikkeleiden irtoamisesta ja mustepartikkelien koosta.
Fraktionaalisella analyysilla on tässä työssä tutkittu eri siistausmassojen raaka-aineita ja prosessiratkaisuja. Työssä huomattiin, että LWC- ja SC-paperin ikääntymisellä on negatiivinen vaikutus paperin sulputtumisnopeuteen pulpperissa, mutta sanomalehtipaperilla vastaavaa ilmiötä ei huomattu. Musteen irtoaminen oli huonompaa LWC- ja SC-paperin ikääntyessä. Kuitenkaan ikääntyminen ei vaikuttanut LWC- ja SC-paperin mustepartikkeleiden pilkkoutumiseen merkittävästi. Sen sijaan sanomalehtipaperilla havaittiin voimakasta mustepartikkeleiden pilkkoutumista raaka-aineen ikääntyessä.
Fraktionaalinen tutkiminen antaa uuden näkökulman musteenpoistomekanismeihin ja voisi olla hyödyllinen, kun etsitään optimaalisia musteenpoisto-olosuhteita esimerkiksi pulpperointiin tai flotaatioon. Menetelmä voisi mahdollistaa myös eri siistausprosessin osaprosessien säätämisen. Lisäsi menetelmän avulla voitaisiin saada tehostettua prosesseja ja parannettua laatua sekä saada prosesseista ympäristöystävällisempiä.
