brk1 and dcd1 Act Synergistically in Subsidiary Cell Formation in Zea mays

Malhotra, Divya 08 1900 (has links)
Subsidiary mother cell (SMC) divisions during stomatal complex formation in Zea mays are asymmetric generating a small subsidiary cell (SC) and a larger epidermal cell. Mutants with a high number of abnormally shaped subsidiary cells include the brick1 (brk1) and discordia1 (dcd1) mutants. BRK1 is homologous to HSPC300, an ARP2/3 complex activator, and is involved in actin nucleation while DCD1 is a regulatory subunit of the PP2A phosphatase needed for microtubule generation (Frank and Smith, 2002; Wright et al. 2009). Possible causes of the abnormal SCs in brk1 mutants include a failure of the SMC nucleus to polarize in advance of mitosis, no actin patch, and transverse and/or no PPBs (Gallagher and Smith, 2000; Panteris et al 2006). The abnormal subsidiary mother cell division in dcd1 is due to correctly localized, but disorganized preprophase bands (PPBs; Wright et al. 2009). The observation that brk1 has defects in PPB formation and that the dcd1 phenotype is enhanced by the application of actin inhibitors led us to examine the dcd1; brk1 double mutant (Gallagher and Smith, 1999). We found that dcd1; brk1 double mutants demonstrate a higher percentage of aberrant SCs than the single mutants combined suggesting that these two mutations have a synergistic and additive effect on SC formation. Our observations and results are intriguing and the future step will be to quantitate the abnormal PPBs and phragmoplasts in the double and single mutants using immunolocalization of tubulin and actin as well as observations of live cells expressing tubulin-YFP.

O processo e o (des)controle da expansão urbana de São Carlos (1857-1977) / The process and the (un)control of the urban expansion in São Carlos (1857-1977)

Lima, Renata Priore 13 November 2007 (has links)
Urban expansion. This work consists in the study of São Carlos, SP urban expansion process, between 1857 and 1977 years, from the implanted land divisions into lots and from the urban expansion instruments of control and planning. It is analyzed the main changes and continuities of the national, state and municipal socioeconomic context; of land use and occupation forms; of the municipal administrative structure of planning, and also of the urban legislation constructed throughout the period. There are three well defined periods in the studied process. In the first one, between 1857 the 1929, during the São Paulo coffee cycle, the expansion occurred in a continuous and concentrated way and the control was made by the municipal council through the application of \'código de posturas\' determinations. The second period, between 1930 and 1959, during the quick São Paulo industrial development process, is characterized by a rude expansion into the highway direction. At this moment, the expansion control in São Carlos was made by the executive power, by using urban parameters to incentive urban expansion. And the third period, between 1960 the 1977, during the military regimen, the expansion occurred in a peripheral and discontinuous way, being the control carried through a new set of urban laws, such as the law of land divisions into lots, the law of zoning and the law of buildings. In this period it also increased the immovable enterpriser action power. The performance of the municipal structure of planning, as well as the legislation, oscillated between controlled and uncontrolled moments, followed by new interests and social demands. The original documents used in this research are maps, plants and aerial photos from São Carlos city; projects of implanted land divisions; municipal, state and federal urban laws and codes; contracts and registration cards of the land divisions plans implanted in the city; and also interviews with the municipal employees. The systematization of this material resulted into maps, graphs and tables showed along this work.

Att arbeta i grupp, är det en bra metod? : En kvalitativ studie av tre lärares syn på och erfarenheter från att arbeta med eleverna i grupp och med grupparbeten i grundskolan

Blom, Helena January 2009 (has links)

The purpose of this study is to discuss various approaches to group work and the importance of different approaches in relation to group work in schools. The study also looks at the results of the children working in different groups and constellations.

This study is based on what previous research, literature and the tree teachers say about the subject group work and group divisions. Issues: Is there any kind of group work that always works? Does the structure of the tasks have any effect on the outcome? Is there any good reason to choose the teamwork approach that? The method I chose is a qualitative study with a phenomenological approach to the analysis. Three teachers were interviewed; result is presented in themes and concludes with a brief summary. In the theory part is the writer and researcher Kjell Granström frequently referenced when he is an actual person in Sweden in the subject. Furthermore, educators Vygotsky and Gardner’s theories of how children learn and how teachers at school can work with these theories are also important for the study. The study findings demonstrate the importance of teacher's choice of work and an awareness of how different group classifications can affect students' work. This is consistent with previous research Granström and also Forslund Frykedal reached.

Profits, Prestige, and a Little Bit of Sunshine: American Independent Cinema Enters the Mainstream

Landay, David A 01 January 2010 (has links)
This thesis investigates the recent developments that have occurred in the independent film market after the major Hollywood studios created their own specialty divisions to produce and distribute independent films. Due to the involvement of the major studios, independent films became more mainstream while their various costs were also driven up due to the increased amount of independent films that were released by the studios. The financial crisis of 2008 forced the major studios to contract their production of independent films and this contraction allowed the prices of independent films to return to more stable levels.

A Study on the Legal System of Administrative Division in Local Districts¡ÐThe Case of Kaohsiung City

Wu, Mao-Shu 18 July 2012 (has links)
Abstract Administrative division contains a string of complex questions. It does not only contain the simple concept of the division of regions, but the formation of the ideology of residence towards regions and the fairness of national resource distributions. However, followed by administrative division, the original administrative district still may not develop well as expected due to the change of time and space. By the analysis of the 5 major frameworks of administrative law, I found out that since the merger of Kaohsiung city and county in 2010, the transition of the environment has diluted and allocated the resources inappropriately. It leads to an unbalanced regional development because the people can not be satisfied with inadequate resources. However, the problem is that the current administrative division can not tackle the problems. It¡¦s a great loss that a reasonable and appropriate plan has not even been conducted for the administrative division of Kaohsiung. Moreover, the scale that administrative division covers in local districts is so comprehensive that it de facto influences the right and obligations of people. Therefore, the legal system of administrative division should be improved and adjusted. By this study I found out that Kaohsiung City should consider the overall development and related factors to assess the whole situation with a profounder perspective. By the expansion that brought by the merger of administrative districts, the resources will be more adequate so as to meet the need of people. With the local features of each administrative district, transport communication can be extended, as well as the space greening and beautification. As a goal-oriented activist, the district governors should dedicated to increasing the service quality and efficiency of district offices for people. Followed by the balanced development, local features and the enhancement of overall development, Kaohsiung City can become the leading city in Taiwan with easy access to administration, balanced resources and limitless development.
