Edição semidiplomática de corpus selecionado a partir do códice ?Ofícios do Vice-Rei para o Governador da Capitania 1793/98?, visando a fornecer fonte fidedigna para futuros estudos filológicos, lingüísticos, históricos e culturais do período. Trata-se também de um estudo mais detalhado desse códice catarinense que contextualiza historicamente os documentos que o compõem, assim como aborda aspectos paleográficos, codicológicos e diplomáticos com o propósito de responder a questionamentos e hipóteses surgidos durante a edição acerca de sua composição, criação e origem. Este trabalho descreve codicológica e paleograficamente o códice em geral, apresenta uma demonstração de diferentes punhos e procura levantar traços de natureza fonológica de um manuscrito dentro do códice. Consta também deste trabalho uma tentativa de identificação tipológica mais adequada através da composição de um verbete descritivo do corpus. / Semidiplomatic edition of the selected corpus from the codex ?Oficios do Vice Rei para o Governador da Capitania 1793/98? (Memorandums from the Vice-king to the Governor of the Province 1793/98?) seeking to provide a trustworthy source to further philological, linguistical, historical and cultural studies of such period. It is also a thorough study of this Catarinense (from Santa Catarina) codex that put its documents into a historical perspective. It touches its paleographic, codicological and diplomatic aspects aiming to address questionings and hypotheses raised during the editing process concerning its composition, how it was assembled, and its origin. This work describes the codex general paleography and codicology. It presents a demonstration of different handwritings and attempts to raise traces of phonological aspects from the codex. This project also proposes a more adequate typological identification of the documents, through descriptive summaries of each memo.

São muitas as parcerias no cinema, mas a que relaciona o diretor de cena ao diretor de fotografia é certamente a principal. A proposta desta dissertação de mestrado é analisar o estilo de fotografia criado por Eduard Tissé em três dos filmes que trabalhou com Eisenstein. A metodologia teórica da pesquisa baseia-se em Roland Barthes a partir da análise do artefato primeiro do cinema, que é o fotograma. As dimensões sociais ligadas ao materialismo dialético e suas aplicações ao filme, na forma como o espectador promove a leitura de sentidos, preocupavam o diretor soviético. Com isso evidencia-se o princípio conceitual de Eisenstein, desenvolvido pela célula de montagem, na visão da sua missão sócio-cultural em busca da fotografia do conflito. / There are many partnerships in the movie business, but that between director and director of photography is certainly the main one. The purpose of this dissertation is to analyze the style of photography created by Eduard Tissé in three of the movies he worked with Eisenstein. The theoretical research methodology is based on Roland Barthes from the analysis of the first film artifact: the frame. The social dimensions related to the dialectical materialism and its applications to the cinema - the way the audience reads the film sense - worried the soviet director. With that, the conceptual principle developed by Eisenstein is highlighted in search of the conflict cinematography.

Elucidating the role of the RNA editing enzyme ADAR1 in the innate immune response

The adenosine deaminase acting on RNA (ADAR) enzymes catalyse the hydrolytic deamination of adenosine (A) to inosine (I) in double stranded (ds) RNA. Mutations in ADAR1 underlie the autoimmune disorder Aicardi Goutiѐres syndrome (AGS). Patients with AGS display heightened levels of type I interferon (IFN) and IFN stimulated genes (ISGs). The first aim of my thesis was to determine whether the mutations found in the human ADAR1 gene affected RNA editing. I found that the ADAR1 mutants identified in the AGS patients have reduced editing activity. Interestingly, the mutations have a greater effect on the IFN-inducible cytoplasmic isoform, ADAR1p150 than on the constitutive ADAR1p110 isoform. These results imply that A-to-I editing plays a role in regulating the type I IFN response. The Adar1 null mouse dies by E12.5 with a type I IFN signature similar to that observed in the AGS patients. The second aim of my thesis was to characterize the immune signalling pathway aberrantly activated in the absence of Adar1. A colleague in our research group rescued the Adar1 null mouse to birth by blocking the cellular response to cytoplasmic dsRNA by generating a double mutant with the mitochondrial antiviral signalling adaptor, Mavs. In the Adar1-/-; Mavs-/- mutant I found that the aberrant immune response is rescued at E11.5. This indicates that MAVS is the downstream adaptor in the aberrant immune response that underlies the embryonic lethality in the Adar1-/- mouse. The third aim of my thesis was to determine if the lack of inosine modification within cellular RNA was triggering the aberrant immune response in the Adar1-/- mouse. To study this, Adar1-/-; p53 -/- mouse embryonic fibroblasts (MEFs) were generated. By reintroducing various ADAR isoforms into the Adar1-/-; p53 -/- MEFs I found that to rescue the aberrant immune response requires both catalytic activity and the location of an ADAR protein within the cytoplasm. Moreover, I demonstrated that transfecting inosine-containing dsRNA oligonucleotides into Adar1-/-; p53 -/- MEFs suppresses the aberrant immune response. Overall my results suggest that A-to-I editing by ADAR1 is an essential RNA modification that is required by the cell to distinguish between ‘self’ and ‘non-self’ RNA. Editing of cellular RNAs prevents an autoimmune response whereas editing of viral RNA may act to suppress a heightened antiviral immune response and prevent long-term damage to the cell.

Investigating gene expression patterns in the mammalian cardiovascular system

The cardiovascular system is an essential component of mammalian biology. It is a complex network of various tissues and structures with unique functions. The function of the cardiovascular system is to supply nutrients including oxygen to the various cells, tissues and organs within the body, and remove waste products from them. Given the importance of this role, it is not surprising that there are countless regulatory mechanisms at the molecular, cellular and tissue levels that are required to support this functional system. Perturbations in parts of this system are likely to lead to abnormalities, and thus give rise to cardiovascular-related diseases. Despite the currently expanding list of genes reported to be involved in a variety of cardiovascular-related diseases, including calcific aortic valve disease (CAVD), the functions and associated pathways of these factors in both normal and pathological physiology have yet to be fully understood, such as at the transcriptomic level. In this thesis, a genome-wide transcriptomic atlas of the healthy mammalian cardiovascular system was generated using the sheep as a large animal model. This atlas was generated using RNA-seq, with the aim of further understanding normal gene expression patterns in the context of the known physiology of healthy mammalian tissues. Through this work, I identified novel gene networks and detailed functional clustering of co-expressed genes with region-specific expression and specialised cardiovascular roles. One interesting cluster was highly expressed in the cardiac valves, and shared genes found in physiological bone development, such as bone morphogenetic protein 4 (BMP4), collagen type I alpha 2 (COL1A2), Sry homeobox 8 (SOX8) and bone gamma-carboxyglutamate protein (BGLAP), some of which have been implicated in vascular calcification. Further to this work, I studied the expression profiles of these key cardiovascular genes during development in the sheep from foetal to adult stages. In addition, I investigated the gene expression patterns of various key vascular calcification genes. These studies showed differential expression of genes in the different cardiovascular tissues, demonstrating transcriptional differences between these different tissues known to have different functions. CAVD involves progressive valve leaflet thickening and severe calcification, resulting in impaired leaflet motion. The in vitro calcification of primary rat, human, porcine and bovine aortic valve interstitial cells (VICs) is commonly employed to examine the mechanisms of CAVD. However, to date, no published studies have utilised cell lines to investigate this process Thus, in this project, I generated and evaluated the calcification potential of an immortalised cell line derived from sheep aortic VICs (SAVICs). This novel large animal in vitro model of CAVD was demonstrated to calcify under high calcium and phosphate conditions. Changes in the expression of key calcification genes during VIC calcification was also observed, including increased mRNA expression of bone markers Runt-related transcription factor 2 (RUNX2) and sodium-dependent phosphate transporter 1 (PiT1), and a concomitant decrease in matrix Gla protein (MGP) mRNA expression. In addition, the role of extracellular nucleotides and their receptors (P2 receptors), which have been previously shown to be important in bone and vascular calcification, were investigated using SAVICs in vitro. This study has shown that extracellular nucleotides, particularly adenosine 5’-triphosphate (ATP) and uridine 5’-triphosphate (UTP) and other agonists of P2 receptors, reduced VIC calcification in vitro. Moreover, the cutting-edge gene-editing technology, clustered regularly interspaced short palindromic repeats (CRISPR)/CRISPR-associated protein-9 nuclease (Cas9), was successfully applied to generate large animal models of cardiovascular-related diseases. In this project, I applied the CRISPR/Cas9 technology to edit ectonucleotide pyrophosphatase/phosphodiesterase 1 (ENPP1) and fibrillin 1 (FBN1) to generate two models of vascular calcification and Marfan Syndrome (MFS), respectively. In the ENPP1-edited animals, soft tissue calcification has been observed in the biallelic mutant and homozygous pigs. In this project, I have developed a range of novel in vitro and in vivo tools to advance the study of cardiovascular disease. These studies demonstrate that large animal models are highly valuable in the field of cardiovascular biology. The in vivo and in vitro experimental models described should facilitate detailed analysis of cardiovascular molecular biology and ultimately lead to therapies which will minimise the morbidity and mortality currently arising from cardiovascular pathology.

Impact de CD38 dans la leucémie à tricholeucocytes / Impact of CD38 in Hairy Cell Leukemia

La leucémie à tricholeucocytes (ou HCL pour Hairy Cell Leukemia) est un syndrôme lymphoprolifératif B rare du sujet âgé, caractérisé par une infiltration médullaire et splénique de cellules présentant des protrusions cytoplasmiques. Des thérapies de première ligne efficaces existent contre ce cancer et l’intérêt de la recherche biomédicale dans ce domaine réside désormais dans le développement de nouvelles molécules actives contre les cellules leucémiques réfractaires aux traitements de référence. Parmi les voies de signalisation dérégulées dans l’HCL, celle des Rho-GTPases influe sur les phénomènes de croissance cellulaire et d’organisation du cytosquelette d’actine, perturbés dans les tricholeucocytes. Les travaux précédemment menés au laboratoire ont montré la sous-expression dans l’HCL d’une Rho-GTPase atypique, RhoH, dont l’expression ectopique dans un modèle cellulaire d’HCL atténue la progression tumorale. Afin de déterminer les cibles moléculaires de RhoH dans cette leucémie, une étude transcriptomique a été réalisée et a montré la sous-expression du marqueur de surface CD38 lorsque RhoH est surexprimée.Plus qu’un marqueur de différenciation lymphocytaire, CD38 est une molécule à effets pléïotropiques (à la fois récepteur, enzyme et protéine d’adhérence cellulaire), importante dans le développement des lymphocytes B. CD38 a également été décrit comme un marqueur délétère dans la leucémie lymphoïde chronique et représente une cible thérapeutique dans le myélome multiple. Bien qu’exprimé par un tiers des patients porteurs de l’HCL, son rôle dans cette leucémie restait jusqu’alors inconnu.Les travaux présentés dans cette thèse décrivent, d’une part, l’étude de la régulation du gène CD38 par RhoH dans l’HCL, et d’autre part, l’impact de la protéine CD38 dans la progression de cette leucémie. Des données préliminaires sur l’activité de fragments de promoteur du gène CD38 semblent indiquer un rôle du facteur de transcription Smad1 dans la régulation de ce gène par RhoH. Grâce à une technique de genome editing, nous avons produit deux lignées cellulaires HCL knock out pour le gène CD38. Ces modèles nous ont permis de déterminer que CD38 promeut la survie ainsi que l’adhésion à l’endothélium des cellules HCL, et modifie également leurs propriétés migratoires in vitro. Nous avons également observé que CD38 favorisait la progression tumorale dans un modèle murin de xénogreffe de ces lignées cellulaires. Enfin, des données produites par nos collaborateurs ont montré que CD38 est un marqueur de mauvais pronostic pour la rechute des patients atteints d’HCL et qu’il constitue une cible thérapeutique potentielle pour les 30% de patients qui l’expriment.Mimer l’effet de RhoH dans l’HCL à des fins thérapeutiques s’avèrerait délicat. Le ciblage de CD38 semble donc une alternative de choix. En effet, la sous-expression de RhoH dans l’HCL favorise l’expression de cette protéine, dont l’effet est délétère pour les patients puisqu’elle participe à la progression de la leucémie. Les anticorps monoclonaux thérapeutiques dirigés contre CD38 étant déjà utilisés en clinique pour traiter d’autres leucémies, ce travail ouvre la voie à l’extension de leur utilisation dans le traitement de l’HCL réfractaire, pour les patients qui l’expriment. / Hairy Cell Leukemia (HCL) is a B-lymphoproliferative disorder of the elderly, which is characterized by medullar and splenic homing of “hairy” cells bearing cytoplasmic protrusions. Efficient first-line therapies against this cancer do exist and the real challenge in biomedical research is now to develop new molecules targeting leukemic cells which are resistant to these first-line treatments. Among some deregulated signaling pathways that have been described in HCL, Rho-GTPases are noteworthy, mediating proliferation and reorganization of actin cytoskeleton, being both disrupted in hairy cells. Former works from our laboratory have shown the underexpression in HCL cells of an atypical Rho-GTPase called RhoH, which reconstitution decreased malignant progression in both in vitro and in vivo models of HCL. In order to determine the molecular targets of RhoH, a microarray study was performed that showed underexpression of the cell surface marker CD38 while RhoH is overexpressed.Not only a differentiation marker of lymphocytes, CD38 is a pleiotropic molecule (being at the same time a receptor, an enzyme and an adhesion protein), which is important in B-cell development. It is also known as a bad prognosis marker in chronic lymphocytic leukemia and a therapeutic target in multiple myeloma. Its role in Hairy Cell Leukemia has not been studied yet, despite its expression in one third of HCL patients.The work presented in this thesis deals with, on the one hand, the study of the regulation of CD38 gene by RhoH in HCL, on the other hand, the impact of CD38 protein on HCL progression. Preliminary data seem to indicate a potential role of Smad1 transcription factor in this mechanism of regulation of CD38 by RhoH. Thanks to genome editing technology, we produced two HCL cell lines knock out for the CD38 gene. These models allowed us to prove that CD38 enhances hairy cells survival and adhesion to endothelium, and modulates their migratory features in vitro. We also showed that CD38 promotes disease progression in vivo in an HCL xenograft mouse model. Finally, data from our collaborators indicated that CD38 is a bad prognosis marker for HCL relapses and could be a potential therapeutic target.Mimicking RhoH effects for therapeutic purposes would be somewhat tricky. Targeting CD38 seems an interesting alternative, as RhoH underexpression favours CD38 expression, which is deleterious for patients by enhancing malignant progression. As therapeutic monoclonal antibodies targeting CD38 are already used in clinics to treat other leukemias, this work brings the evidence of their potential usefulness against refractory HCL cases expressing this marker.

Color harmony : experimental and computational modeling / Harmonie des couleurs : modélisation expérimentale et algorithmique

Comme la consommation de médias numériques a explosé ces dernières années, faire des photos esthétiques, avec ou sans expertise artistique, est plus que jamais un sujet de recherche. Plusieurs axes peuvent être explorés: la haute définition, la luminance ou contraste étendue, les gamut couleur étendus. En plus de ces propriétés intrinsèques de l'image, des connaissances perceptuelles et/ou artistiques seraient de grande valeur pour tout utilisateur manipulant le contenu des images. Cette thèse propose d'aborder le thème de l'harmonie des couleurs. La littérature en lien avec ce sujet se retrouve à travers diverses disciplines : la science des couleurs, le traitement d'image, la psychologie… Ces expériences menées en science des couleurs privilégient la mesure de patchs combinant deux ou trois couleurs, rendant l'extrapolation à des images naturelles impossibles. D'autres approches ont défini des lois empiriques dictant l'arrangement des couleurs sur la roue des teintes. Le cadre applicatif de ces modèles géométriques manque de rigueur quant à leur utilisation. Malgré cela, des algorithmes en traitement d'image employant ces modèles ont vus le jour. Si les résultats semblent qualitativement agréables, ces algorithmes méritent une validation plus quantitative et objective, faisant intervenir une base de données appropriée. Dans cette thèse, deux approches sont mises en perspective: un travail expérimental et une partie algorithmique. Une expérience a été menée à l'aide d'un oculomètre avec une tâche dédiée à l'analyse de l'harmonie des couleurs, permettant de mesurer des effets dans le déploiement de l'attention visuelle. A partir de ces données, une vérité terrain a été extrapolée, permettant la validation des méthodes algorithmiques ensuite proposées. En premier, nous avons amélioré l'état de l'art sur l'harmonisation automatique des images au travers de diverses contributions et avons démontré de façon exhaustive le gain de notre approche. En deuxième contribution algorithmique, nous avons introduit une nouvelle sorte de métrique de qualité qui combine les concepts de masquage visuel et d'harmonie des couleurs. Ainsi, nous pouvons prédire quelles zones de l'image seront perçues harmonieuses, au vue de leur voisinage et donc des effets de masquages potentiels. Enfin, une dernière contribution, nous a amené à dériver deux outils d'édition incorporant les deux techniques précédentes, permettant de rendre accessible les concepts d'harmonie des couleurs à travers une formulation cachée et intuitive. / Since the consumption of digital media exploded in the last decade, making aesthetic pictures quickly - with or without artistic expertise - is more than ever a research topic. Different axis of investigations remain possible: high resolution, high dynamic range or wide color gamut. Additionally to these objective image properties, more perceptual and artistic insights could be of benefit to any user manipulating pictures. In such context, this thesis deals with the topic of Color Harmony. The literature related to this topic is limited, but involves many different scientific areas: color science, image processing and psychology and so on. The validity of collected data is questionable due to their limitation to two- or three-colors patches. The models fitted from these data remain non-exploitable on natural pictures. Other models depicting rules or areas on color wheel lack scientific guidelines for their utilization. Nonetheless, some algorithms employing color harmony theory and models as a core concept showed up in the literature, but suffered from being quantitatively tested and validated. In this thesis, two views are put in perspective in order to respond to the previous statements: an experimental and a computational approaches. The conducted experiment allowed observing some effects with an eye-tracking protocol, never applied before with a task on color harmony assessment. From the collected data of our experimental work, we designed a method to generate a ground truth, which would serve to the validation of the two proposed computational methods. First, we improved an existing architecture for automatic color harmonization and demonstrated exhaustively the benefit of our approach. As a second computational contribution, a novel quality metric is introduced that integrates the concepts of visual masking and color harmony. Thus, we may predict which areas would be perceived harmonious regarding its neighborhood and then the potential masking effects. As a last contribution, two editing tools made accessible the color harmony theory through a hidden formulation of it and a user-friendly and intuitive interface.

In this sense, its main objectives were to map the stages and the actors engaged in the process of editing the Teacher's Guide currently developed in Brazilian publishing houses and to analyze the role played by editors in this process, observing the profile of these professionals, the functions they perform in editing the Teacher’s Guide and the internal and external factors that influence this work. The theoretical reference framework is located in the area of History of Education and, particularly, in the area of History of Curriculum and Textbooks. The methodological procedures were the application of a questionnaire and interviews with editors of the two largest Brazilian textbook publishing houses. The interpretation of the data collected in the field research was based on the theoretical framework presented above and suggests that the main functions performed by editors in the editing process of the Teacher's Guide are related to coordinating activities of other professionals, editing and organizing the original text and writing or rewriting this material. In addition, the analysis of the data collected indicates that editors recognize the Teacher's Guide as an important material for training teachers, presenting the prescribed curriculum, mastering contents and planning classes, although they do not have objective information on how teachers use this material. In this way, by analyzing the practices developed by editors in the editing process of the Teacher's Guide, the work contributes to broadening the understanding about the set of practices that define the curricular system and that, in this sense, directly interfere with the current pedagogical practice / Este trabalho possui um caráter exploratório e busca compreender o papel do editor no processo de edição da versão específica dos livros didáticos destinada ao professor, conhecida como Manual do Professor. Neste sentido, os seus principais objetivos foram mapear as etapas e os atores que compõem o processo de edição do Manual do Professor desenvolvido atualmente nas editoras brasileiras e analisar o papel dos editores neste processo, observando o perfil destes profissionais, as funções que desempenham na edição do Manual do Professor e os fatores internos e externos que influenciam este trabalho. O referencial teórico está localizado na área da História da Educação e, particularmente, na área da História do Currículo e do Livro Didático. Os procedimentos metodológicos foram a aplicação de questionário e a realização de entrevistas com editores das duas maiores editoras brasileiras de livros didáticos. A interpretação dos dados coletados na pesquisa de campo foi feita com base no referencial teórico acima apresentado e sugere que as principais funções desempenhadas pelos editores no processo de edição do Manual do Professor se relacionam com as atividades de coordenação do trabalho de outros profissionais, de edição e organização do texto original e de reescrita ou autoria deste material. Além disso, a análise dos dados levantados indica que os editores reconhecem o Manual do Professor como um material importante para a formação dos professores, para a apresentação do currículo prescrito aos docentes, para o domínio dos conteúdos a serem ensinados e para o planejamento das aulas, ainda que não possuam informações objetivas sobre a forma como os professores utilizam este material. Desta maneira, ao analisar as práticas desenvolvidas pelos editores no processo de edição do Manual do Professor, o trabalho contribui para ampliar a compreensão sobre o conjunto de práticas que configuram o sistema curricular e que, neste sentido, interferem diretamente na prática pedagógica vigente

A invenção do invisível tensiona, principalmente, conceitos como “rastro” e “imagem dialética” de Benjamin e “duração” de Bergson. Realizo a dissecação de quadros e cenas do filme O Eclipse (1962) de Michelangelo Antonioni e, por meio dos conceitos propostos: olhos ausentes, (re)quadros, pontos cegos e rotações - imaginados como uma constelação de imagens - busco rastrear formas de presença do invisível na montagem de cinema. Proponho uma metodologia do invisível para análise do corpus. O espectador, ao perceber a imagem enquadrada e montada pelo cineasta, sobrepõe a ela “imagens invisíveis”, da sua memória ou repertório cultural e técnico, que descontroem e transformam, assim, a imagem vista no presente da exibição do filme. / In this thesis I explore the tracks of the invisible in the perception and memory of film spectators. The invention of the invisible creates a tension mainly with the concepts of “track” and “dialectical image” proposed by Benjamin and of “duration” of Bergson. I dissect frames and scenes of the film. The Eclipse (1962) by Michelangelo Antonioni based on the proposal of the following concepts: absent eyes, (re-framings, blind points and rotations. Imagined as a constellation of images, these concepts are operational for tracking forms of presence of the invisible in film editing. I propose a methodology of the invisible for analyzing the corpus, understanding that upon perceiving images framed and edited by the film-maker, spectators overlap them with “invisible images” from their cultural and technical memories or repertoires, which deconstruct and transform the image seen in the present of the exhibition of the film.
