T-Cell Immunogenicity and Dysfunction in Cancer and Viral DiseasesJanuary 2017 (has links)
abstract: CD8+ T-lymphocytes (CTLs) are central to the immunologic control of infections and are currently at the forefront of strategies that enhance immune based treatment of a variety of tumors. Effective T-cell based vaccines and immunotherapies fundamentally rely on the interaction of CTLs with peptide-human leukocyte antigen class I (HLA-I) complexes on the infected/malignant cell surface. However, how CTLs are able to respond to antigenic peptides with high specificity is largely unknown. Also unknown, are the different mechanisms underlying tumor immune evasion from CTL-mediated cytotoxicity. In this dissertation, I investigate the immunogenicity and dysfunction of CTLs for the development of novel T-cell therapies. Project 1 explores the biochemical hallmarks associated with HLA-I binding peptides that result in a CTL-immune response. The results reveal amino acid hydrophobicity of T-cell receptor (TCR) contact residues within immunogenic CTL-epitopes as a critical parameter for CTL-self/nonself discrimination. Project 2 develops a bioinformatic and experimental methodology for the identification of CTL-epitopes from low frequency T-cells against tumor antigens and chronic viruses. This methodology is employed in Project 3 to identify novel immunogenic CTL-epitopes from human papillomavirus (HPV)-associated head and neck cancer patients. In Project 3, I further study the mechanisms of HPV-specific T-cell dysfunction, and I demonstrate that combination inhibition of Indoleamine 2, 3-dioxygenase (IDO-1) and programmed cell death protein (PD-1) can be a potential immunotherapy against HPV+ head and neck cancers. Lastly, in Project 4, I develop a single-cell assay for high-throughput identification of antigens targeted by CTLs from whole pathogenome libraries. Thus, this dissertation contributes to fundamental T-cell immunobiology by identifying rules of T-cell immunogenicity and dysfunction, as well as to translational immunology by identifying novel CTL-epitopes, and therapeutic targets for T-cell immunotherapy. / Dissertation/Thesis / Doctoral Dissertation Biological Design 2017

Desenvolvimento de peptídeos recombinantes epítopos específicos do Rhipicephalus (Boophilus) microplus selecionados por bibliotecas de Phage Display / Development of epitope specific recombinant peptides of the Rhipicephalus (Boophilus) microplus selected by Phage Display librariesPrudencio, Carlos Roberto 27 May 2008 (has links)
Ticks are implicated in serious economic losses to animal production worldwide in
the order of billions of dollars. Although many antigens have been in vaccine tests,
none has been effective in the control of ticks, which justifies the development of
new antigens as well as new vaccine strategies. Phage Display techniques have
been widely employed to map epitope structures, which have served as the basis
for developing molecular vaccines. In the present study, we have applied this
technique to map specific epitopes of Rhipichephalus (Boophilus) microplus and to
validate the peptides as potential immunogens, we have adopted a process of
selection prior to final field tests. This strategy was established in order to reduce
the number of clones to be tested in the field. Six Phage -displayed random
peptide libraries were selected with seven different strategies against the purified
hyperimmune serum of chickens (IgY) that were immunized with total proteins of
larvae and adults of Rhipicephalus (Boophilus) microplus. The selected Phage
clones were sequenced, translated and analyzed through bioinformatics. To
evaluate the potential of these phagotopes as effective candidate vaccines, ELISA
assays, dot-blot, mice immunization (MI), humoral immune response (HIR) of
cattle against the clones and a cutaneous hypersensitivity tests in cattle were
performed. The selected Phage clones were analyzed through bioinformatics,
generating 107 different peptides in a total of 281 sequences. Some selected
phagotopes showed excellent matches with linear sequences of known proteins of
the Rhipicephalus (Boophilus) microplus. Peptides that did not present significant
matches with known proteins, but shared extensive homology among each other,
were clustered and classified as conformational epitopes of Rhipicephalus
(Boophilus) microplus, or considered as mimotopes of unknown proteic antigens.
Among all clones tested by dot-blots, the 40 most reactive ones were further
screened by HIR and MI. The mice sera raised against the Phage clones clearly
recognized tick proteins indicating that the phagotope-induced immune responses
were antigen-specific, but not all could be identified by the sera of naturally infested cattle (Bos taurus and Bos indicus). Four clones were then selected to go
through the cutaneous hypersensitivity tests, and apparently all of them were
effectives at diferent degrees. The stringent selection processes coupled with this
schemes of validation assays allowed us to narrow down the number of peptides
that should be tested in the field. One of the advantages of using whole
recombinant M13 virus, carrying the recombinant peptide in fusion with the pIII
protein of the virus, is that its capsid acts as adjuvant and could be tested directly
without any further mixture. Additionally, we have finally used anergy skin tests
(cutaneous hypersensitivity tests) once they are sometimes used to guide clinical
decisions. We have assumed that cutaneous hypersensitivity tests measure a
common property of cellular immune function relevant to the outcome, which
together with the humoral response in naturally infested animals may provide us
with sequences of peptides that may be relevant as vaccines. Finally, we have
used those sequences to produce multiple antigen peptide system (MAPS), which
will be used in future field tests. The present work demonstrates that the whole
epitope profile can obtained through screening the Phage Displayed peptide
libraries with the hyperimmune serum and reveals the potential of using epitopedisplaying
Phage s as peptide vaccines against ticks. / Os carrapatos tem provocado significativas perdas econômicas na
produção animal mundial na ordem de bilhões de dólares anuais. Embora vários
antígenos específicos tenham sido empregados em testes vacinais, nenhum tem
propiciado controle eficaz, sendo necessária a identificação de novos antígenos,
bem como de novas estratégias vacinais. A tecnologia de Phage Display tem sido
amplamente empregada no mapeamento de estruturas antigênicas as quais tem
servido como base para o desenvolvimento de vacinas moleculares e representa
uma alternativa as metodologias tradicionais. No presente trabalho, utilizou-se a
tecnologia para o mapeamento de epítopos do Rhipicephalus (Boophilus)
microplus e, para validar os peptídeos selecionados como potencial imunógenos,
foi adotado um processo com múltiplas etapas de caracterização dos clones
previamente aos ensaios clínicos. Esta estratégia foi estabelecida com o objetivo
de reduzir o número de clones a ser testados em bovinos como imunógenos
candidatos. Foram utilizadas para a seleção seis bibliotecas distintas de
peptídeos randômicos expressados em bacteriófagos filamentosos e sete
estratégias de seleção contra as imunoglobulinas (IgY) purificadas de soro
hiperimune de galinhas imunizadas com proteínas totais de larvas e adultos de
Rhipicephalus (Boophilus) microplus. As seqüências do DNA correspondente aos
insertos dos fagos selecionados foram seqüenciados, traduzidos e analisados por
análises in silico. Foram realizados testes de ELISA, Dot-Blot, imunização em
camundongos, determinação da resposta imune humoral de bovinos naturalmente
infestados, e a medição da hipersensibilidade cutânea com o objetivo de evoluir o
potencial destes fagotopos como candidatos efetivos a novos imunógenos. Foram
gerados 107 peptídeos distintos em um total de 281 seqüências, sendo alguns
destes similares às seqüências protéicas lineares do Rhipicephalus (Boophilus)
microplus. Os peptídeos sem similaridades significativas com proteínas
conhecidas, mas que apresentaram seqüências consenso entre os peptídeos
obtidos, foram agrupados e classificados como epítopos conformacionais ou
considerados como mimetopos de antígenos protéicos desconhecidos do
Rhipicephalus (Boophilus) microplus. Entre os clones testados por Dot-Blot, os 40
mais reativos foram posteriormente testados em ensaios de imunização em camundongos e também submetidos ao reconhecimento humoral de bovinos
naturalmente infestados com carrapatos. Adicionalmente, utilizou-se o teste
cutâneo de hipersensibilidade para a análise da imunidade celular. Os soros de
camundongos imunizados com os clones de fagos foram reativos as proteínas de
carrapato indicando que os fagotopos foram capazes de gerar resposta imune
antígeno específica, embora nem todos os fagos fossem reconhecidos pelo soro
de animais naturalmente infestados (Bos taurus e Bos indicus). Após a
caracterização e validação, quatro fagotopos foram escolhidos para a realização
dos testes cutâneos, os quais apresentaram graus variados de reatividade. Uma
das vantagens da utilização do vírus M13 recombinante, contendo o peptídeo
recombinante fusionado na proteína PIII do vírus, é que o próprio capsídio atua
como adjuvante e pode ser testado diretamente após a seleção sem
procedimentos adicionais de preparo do antígeno. Finalmente, as seqüências
determinadas foram utilizadas para a produção e expressão de um sistema
recombinante de múltiplos antígenos peptídicos (MAPS). Foram escolhidos 10
peptideos para a expressão na forma de multi-epitopos, os quais serão
futuramente utilizados em ensaios clínicos. O presente trabalho demonstrou que
um perfil geral dos epítopos pode ser obtido através da utilização de bibliotecas
de peptídeos randômicos expressados em bacteriófagos filamentosos contra
anticorpos obtidos de soro hiperimune e revela o potencial da seleção e utilização
peptídeos epítopos específicos como potenciais vacinas recombinantes multiantigênicas

Previous issue date: 2008 / Resumo: Este trabalho visou efetuar a identificação dos epitopos das prottnas recombinantes CB-22 e CB-104 do capsídeo do Cítrus tristeza vírus (CTV), reconhecidos pelos anticorpos monoclonais (MAb) produzidos por Stach-Machado e colaboradores (2000). Assim, os epitopos reconhecidos pelos MAbs 30.G.02 e 37.G.ll, correspondem às seqüências de aminoácidos NLHIDPTLI e TQQNAALNRDLF, localizadas nas posições 32 a 40 e 50 a 61 das proteínas recombinantes CB-22 e CB-104. O epitopo reconhecido pelo MAb
39.07 que apresenta especificidade apenas para a CB-104, homóloga aos isolados severos do CTV corresponde a seqüência TDVVFNSKGIGN, localizados na posição 120 a 131 da proteína. Estes dados corroboram com os ensaios de ELlSA, confirmando o MAb 30.G.02 como um anticorpo universal, atuando quer como anticorpo de captura ou de detecção em ELlSA Sandwich, uma vez que, seu epitopo é conservado em 87.2% dos isolados de CTV. O MAb 37.G.ll deve ser adotado apenas como anticorpo de detecção pois reconhece uma seqüência encontrada em 62,6% dos isolados, permitindo uma identificação ampla, sem efetuar a diferenciação de estirpes. O MAb 39.07 foi classificado como "forte", pois seu epitopo está presente em apenas 19,7% das seqüências, sendo estas provenientes de isolados fortes do CTV de todo o mundo. A utilização de peptídeos lineares não permitiu a identificação e caracterização do epitopo do MAb 1C.04-12, confirmando dados preliminares de nosso laboratório que permitiram inferir que o mesmo reconhece um epítopo conformacional. O conhecimento das características estruturais da proteína do capsídeo viral possibilita obtenção de informações relevantes quanto às suas propriedades biológicas, como a participação na organização e montagem da partícula viral, proteção do material genético, interação com o inseto-vetor e com a planta hospedeira e também, pela "movimentação" da partícula viral no processo de infecção na planta. Considerando que até a presente data não existem dados disponíveis no Protein Data Bank (PDB) sobre a estrutura tridimensional da proteína do capsídeo do CTV, efetuamos o estudo estrutural da CB-22, utilizando Dicroísmo Circular (CD) e Difração de Raios X a Baixo Ângulo (SAXS). A análise dos dados de CD mostrou um espectro característico de proteínas cuja estrutura secundária é predominantemente formada por a-hélices, sendo este resultado coerente com a estrutura secundária predita pelo software PSIPRED. Os estudos de SAXS revelaram uma proteína altamente oligomerizada com estruturas formadas por subunidades, que variam de acordo com a concentração da mesma em solução. Este resultado, embora tenha impossibilitado a resolução e modelagem da estrutura secundária da CB-22, demonstrou que esta proteína recombinante mantém a função da proteína nativa, responsável pela formação do capsídeo vira!. Assim sendo, podemos postular a sua utilização como uma molécula carreadora, podendo ser aplicada em protocolos de vacinação ou no transporte dirigido de ácidos nucléicos ou proteínas. Por fim, este trabalho estabeleceu diagnósticos imunomoleculares como Imunocaptura-PCR (IC-PCR) e Imuno-PCR (I-PCR) para a detecção da bactéria gramnegativa Xylella jastid[osa, agente etiológico da Cvc. Os ensaios imunomoleculares apresentaram o limite de detecção muito superior aos diagnósticos utilizados na rotina como ELlSA e PCR, sendo o limite de detecção do IC-PCR e do I-PCR de 103 a 101 células, respectivamente. Portanto, estes métodos são uma alternativa viável para efetuar o diagnóstico da CVC e também em estudos epidemiológicos, com a vantagem de eliminar as etapas de extração e concentração do material genético bacteriano, permitindo um diagnóstico acurado dessa doença / Abstract: This work aims to carry out the identification of epitopes of the Citrus tristeza virus (CTV) recombinant coat protein (CB-22 and CB-l04) which are recognized by four monoclonal antíbodies (MAb) produced by Stach-Machado et aI. (2000). MAb 1C.04-12 recognizes CB-22 protein, homologous to CTV isolates that induce weak symptoms in indicators plants; while MAb 39.07 reacts to CB-l04 protein, homologous to severe CTV isolates and, MAbs 30.G.02 and 37.G.ll recognize both proteins. MAb 30.G.02 recognized the sequence formed by amino acids NLHIDPTLI, located at positions 32 to 40, while MAb 37.G.ll recognized the amino acids from 50 to 61, whose sequence is TQQNAALNRDLF. On the other hand, the MAb 39.07 recognized the sequence TDVVFNSKGIGN, located at positions 120 to 131 in CB-l04 protein. MAb 30.G.02 was considered a Universal antibody, once its epitope is conserved in 87.2% of CTV isolates, and can be applied as a coating antibody or even as a detection antibody in ELlSA Sandwich assays. The epitope of MAb 37.G.ll is conserved in 62.6% of sequences and, can be applied as detection antibody, allowing a quick and wide range screening but without strains differentiation. Finally, MAb 39.07 was classified as "Severe" antibody, once its epitope is present at only 19.7% of CTV sequences, which were derived frem severe worldwide CTV isolates. The technique of linear peptides did not allow the epitope determination of MAb 1C.04-12, since this antibody may recognize a conformational epitope in the CB-22 tridimensional structure, according to the early data from our laboratory showing that this MAb did not recognize the CB-22 under denaturating conditions, confirming the conformational nature of its eptiope. The knowledge about structural features of viral coat protein enables acquiring relevant information about biological properties of this protein. The coat protein plays an essential role in the organization and assembly of viral particle, acts in genetic material protection and, probably is involved in the interaction with insect-vector and with host plant, being responsible for the movement of viral particle in the infection processo However, there are not any data in Protein Data Bank (PDB) about the secondary and tertiary structures of CTV coat protein, and so, this work carried out a structural study about CB-22 by Circular Oichroism Spectroscopy (CO) and Small-Angle X-ray Scattering (SAXS). The CO data analysis demonstrated a spectrum characteristic of proteins whose secondary structure is predominantly formed by a-helixes, and these results are coherent of predicted structure by PSIPREO software. The SAXS data revealed a highly oligomeric protein comprising many subunits whose quantities are dependent and vary according to the protein concentration in solution. These data, although had impaired the modeling and resolving of CB-22 secondary structure, demonstrated that this recombinant protein maintain the function of native protein, responsible for the assembly of CTV capsid, and this structure without bacterial ONA is also called virus-/ike, which allows to postulate its usefulness as nucleic acid ca rrier in vaccination protocols. Finally, the third objective of this work was the establishment of immunomolecular diagnosis to gram-negative bacterium, Xy/ella fastidiosa, which causes Citrus Variegated Chlorosis in Brazil. We carried out two immunomolecular assays, like !mmmuno,Çapture PCR and !mmuno-PCR for direct detection of X. fastidiosa without ONA isolation. Whereas the reactivity of ELlSA and PCR ranged from 106 to 104 bacterial cells, the IC-PCR sensitivity was up to 103 and the detection limit of I-PCR was up to 101 bacterial cells. A persistência de células T de memória funcionais é importante para garantir uma imunidade protetora na infecção pelo Vírus da Imunodeficiência Humana (HIV). As células T de memória têm sido subdivididas em memória central (TCM), memória efetora (TEM) e memória efetora altamente diferenciada (TEMRA) com base na expressão de moléculas de superfície como CCR7 e CD45RA, e na capacidade de produzir citocinas e proliferar. Recentemente, identificamos 18 peptídeos derivados de seqüências do consenso B do HIV-1, ligadores de múltiplas moléculas HLA-DR e amplamente reconhecidos por linfócitos T de sangue periférico de pacientes infectados pelo HIV. Diante disso e considerando a importância das células T de memória na manutenção da resposta imune específica, nosso objetivo foi caracterizar fenotípica e funcionalmente as subpopulações de células T de memória de indivíduos infectados pelo HIV envolvidas no reconhecimento in vitro desses epitopos. Foram incluídos 14 indivíduos controles sadios e 61 pacientes HIV+ com contagem de linfócitos T CD4+ maior que 250 células/mm3. Os pacientes HIV+ foram divididos em seis diferentes grupos clínicos de acordo com o estágio da infecção, carga viral (CV) plasmática e uso de terapia anti-retroviral (ART): não progressores por longo tempo (LTNP), avirêmicos em uso de ART (AV-ART), virêmicos em uso de ART (VI-ART), virêmicos sem uso de ART (VI sem ART), virêmicos recéminfectados sem uso de ART (VI-RI) e controladores. Células mononucleares do sangue periférico dos indivíduos do estudo foram estimuladas com o conjunto de peptídeos do HIV-1 e com um conjunto de peptídeos do Citomegalovírus (CMV). A freqüência de células de memória produtoras de IFN- e IL-2 e a proliferação celular antígeno-específica foram detectadas por citometria de fluxo de multiparâmetros. Nossos resultados mostraram que o conjunto de peptídeos do HIV-1 foi capaz de ativar subpopulações funcionais de memória TCM, TEM e TEMRA secretoras de IFN- e IL-2 em 100% dos pacientes HIV+ dos diferentes grupos clínicos. O conjunto de peptídeos do HIV-1 também induziu proliferação das subpopulações de linfócitos T de memória. As freqüências de TEMRA CD4+IFN-+, TEMRA CD4+IFN-+ total, TCM CD8+IFN-+, TCM CD8+IFN-+ total, TEM CD8+IFN-+, TEM CD8+IFN-+ total e TEMRA CD8+IFN-+ correlacionaram-se negativamente com a carga viral do HIV em pacientes virêmicos. Esses dados sugerem que essas subpopulações de memória funcionais são importantes no controle da viremia. Comparando as respostas HIV e CMVespecíficas observamos freqüências mais elevadas de células T de memória produtoras de IL-2, IFN-/IL-2 e IFN- em respostas ao pool de peptídeos do HIV. Esses dados sugerem que esse conjunto de peptídeos derivados de seqüências do HIV-1 ativa respostas polifuncionais de subpopulações de linfócitos T de memória. Nossos resultados mostraram que o conjunto de peptídeos do HIV-1 foi capaz de estimular diferentes subpopulações distintas de linfócitos T de memória produtores de IFN-, IFN-,/IL-2 e IL-2 de indivíduos em diferentes estágios da infecção pelo HIV e sugerem o envolvimento de subpopulações de memória funcionais no controle da viremia. Estes achados fortalecem a possibilidade de uso desses peptídeos em uma formulação vacinal bem-sucedida em humanos / The persistence of functional memory T cell is important to ensure a protective immunity to Human Immunodeficiency Virus (HIV) infection. Memory T cells have been subdivided into central memory (TCM), effector memory (TEM) and highly differentiated effector memory (TEMRA) based on the expression of surface molecules such as CCR7 and CD45RA, and the ability to produce cytokines and proliferate. Recently, we identified 18 peptides derived from B consensus sequences of HIV-1 that bind to multiple HLA-DR molecules and are widely recognized by peripheral blood T lymphocytes from HIV-infected patients. Given this and considering the importance of memory T cells in the maintenance of specific immune response, our objective was to characterize phenotypic and functionally memory T cell subsets from HIV-infected individuals involved in the recognition of these epitopes in vitro. The study included 14 healthy control subjects and 61 HIV+ patients with CD4+ lymphocytes counts higher than 250 cells/mm3. The HIV+ patients were divided into six different clinical groups according to the stage of infection, plasma viral load (VL) and antiretroviral therapy use (ART): long-term non-progressors (LTNP), aviremic under ART (AV-ART), viremic under ART (VI-ART), viremic without using ART (VI without ART), recently infected viremic without using ART (VI-RI) and controllers. Peripheral blood mononuclear cells from study subjects were stimulated with HIV-1 peptide pool and with a cytomegalovirus (CMV) peptide pool. The frequencies of IFN- and IL-2 producing memory cells and antigenspecific cell proliferation were detected by multiparametric flow cytometry. Our results showed that the HIV-1 set of peptides was able to activate TCM, TEM and TEMRA functional memory subsets that secrete IFN- and IL-2 in 100% of the HIV patients from the different clinical groups. The HIV-1 set of peptides also induced memory T lymphocyte subsets proliferation. TEMRA CD4+IFN-+, total TEMRA CD4+IFN-+, TCM CD8+IFN-+, total TCM CD8+IFN-+, total TEM CD8+IFN-+, TEM CD8+IFN-+ and TEMRA CD8+IFN- + frequencies negatively correlated with HIV viral load in viremic patients. These data suggest that these functional memory subsets are important to control the viremia. When comparing the HIV and CMV-specific responses we observed higher frequencies of IL-2, IFN-/IL-2 and IFN- producing memory T cells in response to HIV peptide pool. These data suggest that this set of HIV sequence derived peptides activates polyfunctional response of memory T lymphocyte subsets. Our results showed that the HIV-1 peptide set was able to stimulate different IFN-, IFN-/IL-2 e IL-2 producing memory T lymphocytes from individuals in different stages of HIV infection and suggest the involvement of functional memory subsets in the control of viremia. These findings strengthen the possibility of using these peptides in a successful vaccine formulation in humans

Previous issue date: 2012 / Resumo: Tratamento térmico, alta pressão ou hidrólise enzimática, utilizados em conjunto ou separadamente, podem alterar determinantes antigênicos (epítopos) presentes nas proteínas, e têm sido estudados como estratégia para obtenção de produtos com menor potencial alergênico, porém ainda com sucesso limitado. A enzima transglutaminase (TG), que catalisa a reação de ligação cruzada inter ou intramolecular em diversas proteínas, também tem sido utilizada com este propósito. A associação da hidrólise enzimática com a polimerização por TG é uma estratégia ainda pouco explorada, mas que oferece boas perspectivas em relação à redução da antigenicidade de proteínas. Diante disso, o presente estudo teve como objetivos obter e caracterizar a ß-Lactoglobulina (ß-Lg) polimerizada por TG e a ß-Lg polimerizada pré e pós hidrólise enzimática com as proteases Alcalase, Neutrase ou bromelina, e avaliar o efeito destes processos na antigenicidade da proteína. A melhor condição de hidrólise da ß-Lg, com cada uma das proteases estudadas foi definida a partir de um delineamento composto central rotacional (DCCR), tendo como variáveis independentes a concentração de proteína (2,2 -7,8% p/v) e relação enzima : substrato (E:S), 6,9 - 28 U g-1 proteína e, como variável dependente o grau de hidrólise (GH), determinado pelo método pH-stat. A condição selecionada, 3% de proteína e E:S 25 U g-1, foi a utilizada para a hidrólise da ß-Lg pré ou pós polimerização com a enzima TG. As condições experimentais foram: (1) hidrólise da ß-Lg com as três enzimas, seguida de polimerização com TG (10 ou 25 U g-1) ou (2) polimerização da ß-Lg (7%p/v) com TG (10U TG g-1) realizada pós tratamento térmico ou na presença de agente redutor Cys (0,1 mol L-1), seguido por hidrólise com as diferentes proteases. As amostras hidrolisadas e as polimerizadas pré ou pós hidrólise foram caracterizadas por eletroforese SDS-PAGE/tricina, cromatografia líquida de alta eficiência de fase reversa (CLAE-FR), cromatografia líquida acoplada à espectrometria de massas (MS) e ensaios imunoquímicos (ELISA e Imunoblote). Os hidrolisados obtidos com Alcalase apresentaram 12,7% de GH e com Neutrase e bromelina aproximadamente 4%. Os perfis eletroforéticos da ß-Lg polimerizada pós tratamento térmico (ß-Lg TT TG) ou na presença de Cys (ß-Lg Cys TG), apresentaram bandas de massa molecular (MM) alta, porém a polimerização pós hidrólise, avaliada por eletroforese e cromatografia, não resultou em aumento da MM aparente dos hidrolisados. A ligação isopeptídica e-(?-Gln)Lys, catalisada pela TG, foi detectada em concentração mais elevada no material polimerizado pós hidrólise com alcalase, seguido do tratamento de polimerização da presença de Cys. No estudo da polimerização pré hidrólise, a alcalase foi a enzima que hidrolisou mais eficientemente tanto a ß-Lg como seus polímeros. Os polimerizados pre ou pós hidrólise com neutrase ou bromelina apresentaram ß-Lg residual, observada no perfil eletroforético e cromatográfico, sendo que para os hidrolisados com neutrase, a fração correspondente à ß-Lg residual apresentou maior intensidade. O tratamento isolado de hidrólise, principalmente com alcalase ou bromelina, ou a associação deste processo com a TG foram capazes de reduzir significativamente a resposta antigênica da ß-Lg. Os hidrolisados da ß-Lg com neutrase apresentaram reação antígeno-anticorpo apenas na região correspondente ao monômero da ß-Lg, sendo que a reação foi menos intensa após a polimerização do hidrolisado com a TG. As amostras foram submetidas à digestão in vitro e avaliadas por espectrometria de massas (MS/MS) quanto à identificação de epítopos lineares. Verificou-se que tanto a ß-Lg polimerizada após tratamento térmico ou na presença de Cys como a amostras polimerizadas pré ou pós hidrólise com bromelina apresentaram alguns fragmentos correspondentes aos epítopos, embora em menor número quando comparados aos epítopos identificados na ß-Lg nativa. Para o tratamento com a alcalase, isolado ou associado à TG, não foram identificados fragmentos correspondentes aos epítopos após digestão gastrointestinal das amostras. Os resultados obtidos indicaram que a hidrólise, principalmente com alcalase, associada ou não à polimerização com TG, foi capaz de reduzir a antigenicidade da ß-Lg, mostrando-se uma alternativa para a produção de alimentos hipoalergênicos / Abstract: Heat treatment, high pressure and enzymatic hydrolysis have been studied aiming at obtaining products with low allergenic potential since they can alter antigenic determinants (epitopes) in the protein. The transglutaminase (TG), an enzyme that catalyzes inter or intramolecular cross-link in different proteins has also been used to reduce the protein antigenicity. The association of enzymatic hydrolysis and polymerization by TG is another strategy for reducing the antigenicity of food proteins, promising that needs further research. Therefore, the present study aimed to obtain and characterize the ß-Lg polymerized by TG before or after enzymatic hydrolysis with the proteases alcalase, neutrase or bromelain and evaluate the effect of these processes on the protein antigenic response. The optimum conditions for hydrolysis of ß-Lg, obtained with the three different enzymes, were defined using a central composite rotatable design (CCRD), where the independent variables were protein concentration (2.2% -7.8 w / v) and enzyme: substrate ratio (E:S), 6.9 to 28 U g-1 protein, and the dependent variable (response) was the degree of hydrolysis (DH) determined by the pH-stat method. The hydrolysis condition defined by CCRD was 3% of protein (w/v) and E:S ratio 25 U g-1. The experimental conditions were: (1) hydrolysis of ß-Lg with the three enzymes used separately, followed by polymerization by TG (10 or 25 U g-1) or (2) polymerization of ß-Lg (7% w/v) by TG (10U g TG-1) after heat-treatment (80 °C/60 min) or in presence of reducing agent Cys (0.1 mol L-1), followed by hydrolysis. The samples were characterized by SDS-PAGE/tricine, reverse phase high-performance liquid chromatography (RP-HPLC), liquid chromatography coupled to mass spectrometry (HPLC-MS) and by immunochemical assays (ELISA and Imunoblote). The highest DH of the ß-Lg was obtained with alcalase (12.6%), while with neutrase or bromelain a DH of 4% approximately was achieved. The electrophoresis profiles of ß-Lg polymerized after heat treatment (ß-Lg TT TG) or in the presence of Cys (ß-Lg Cys TG) showed bands with high molecular mass (MM). On the other hand, the post-hydrolysis polymerization, analyzed by SDS-PAGE and RP-HPLC, resulted in no increase in the MM of the hydrolysates. The highest concentration of the cross-link, by formation of the isopeptide bound e-(?-Gln) Lys, was found in the sample polymerized after hydrolysis with alcalase, followed by the sample ß-Lg Cys TG. The polymerized pre or pos hydrolysis with neutrase or with bromelain showed residual ß-Lg, observed in electrophoretic and chromatographic profiles, and for hydrolysates with neutrase, the band corresponding to ß-Lg showed higher residual intensity. Alcalase was the enzyme which hydrolysed more efficiently both untreated and polymerized ß-Lg. Using neutrase or bromelain to hydrolyse polymerized ß-Lg, a residual ß-Lg (18.3 kDa) was observed in the electrophoretic and chromatographic profiles. The avaluation of antigenicity showed that hydrolysis treatment used alone, especially with alcalase or bromelain, or the combination of this process with TG was capable to reduce significantly the antigenic response of ß-Lg. The ß-Lg hydrolysed with neutrase showed antigen-antibody reaction only in the region corresponding to the ß-Lg monomer, and this reaction was less intense after the association of hydrolysis with neutrase and polymerization by TG. The samples were submitted to in vitro digestion and then analyzed by mass spectrometry (MS / MS) to identification of linear epitopes in the protein. It was verified that the polymerized samples (ß-Lg TT TG or ß-Lg Cys TG) as well as those polymerized before or after hydrolysis with bromelain, retained some fragments corresponding to the epitopes, although to a lesser extent when compared to the epitopes identified in the untreated ß-Lg. Interestingly that for the treatment with alcalase, associated or not with the TG, no epitopes were identified after gastrointestinal digestion. These results indicated that the hydrolysis, especially with alcalase, and the association of hydrolysis with polymerization, were capable to reduce the ß-Lg antigenicity and could be an alternative for elaboration of hypoallergenic products / Doutorado / Nutrição Experimental e Aplicada à Tecnologia de Alimentos / Doutor em Alimentos e Nutrição

Uma das estratégias mais promissoras para a biotecnologia de vacinas é o desenvolvimento de linhagens precisamente atenuadas, e que possam ser usadas como carregadoras de antígenos heterólogos. Mutantes de <i}>Salmonella Typhimurium têm sido extensivamente utilizados com essa fmalidade. A flagelina, monômero constituinte do filamento flagelar, vem sendo empregada como carregadora de antígenos heterólogos, inseridos na região central, hipervariável (região IV). Inserções nessa região são freqüentemente funcionais, e levam à exposição do epitopo na superfície do filamento. O presente trabalho explora o potencial de outras regiões da molécula para a inserção de epitopos. Nós inserimos a mesma seqüência usada anteriomente (epitopo da proteína M de S. pyogenes, Tipo 5) em regiões com diferentes níveis de homologia (III e VI), e em região totalmente conservada (VIII). Também foram feitas inserções duplas em regiões que se mostraram toleráveis (III e IV; IV e VI). Todas as proteínas híbridas foram sintetizadas pela Salmonella, como demonstrado em imunoblots, usando anticorpo contra a flagelina e contra o peptídeo. Todas as regiões, exceto a VIII, aceitaram a inserção sem perda de motilidade, apesar de, em alguns casos, ela ter sido extremamente reduzida. A imunogenicidade foi avaliada pela imunização de camundongos com bactérias vivas, inativadas ou, quando possível, flagelina purificada. Os resultados foram similares aos descritos na literatura para inserções envolvendo a região IV, obtendo-se um elevado título de anticorpos contra flagelina. Um baixo nível de anticorpo contra o peptídeo também foi detectado para todas as novas linhagens testadas. Nossos resultados com imunização de bactérias vivas sugerem uma resposta levemente melhor ao peptídeo quando duas cópias estão presentes, mas os dados não são conclusivos. / One of the most promising strategies for the biotechnology of vaccines is the development of precisely attenuated strains, which could be used as carriers of heterologous antigens. Mutants of Salmonella Typhimurium have been extensively explored to this effect, since the infection ofmice by S. Typhimurium mimics the infection of humans by S. Typhi, and the genetics of the species is extremely well known, making it easy the obtention of defined mutants with reduced pathogenicity. Mutants with auxotrofy in genes of the aromatic pathway are particularly attractive, since they need PABA and DHB to grow, and these compounds are unavailable in mammalian tissues. Flagellin, the monomer which constitutes the flagellar filament, has been used as a carrier for heterologous epitopes, inserted in a central, hypervariable region (region IV). Insertions in this region are often functional, and lead to exposition of the epitope at the filament\' s surface. The present work explored the potential of the other regions ofthe molecule for the insertion of epitopes. We inserted the same reporter sequence (MS epitope from S. pyogenes M protein) in regions with different levels of homology (III and VI), and totally conserved (VIII). We also made double insertions in regions shown to be permissive (III and IV; IV and VI). All hybrid proteins were synthesized by Salmonella, as demonstrated by immunoblots using antibody against flagellin and against the synthetic peptide. All regions, except the highly conserved region VIII, accepted the insertions without loss of motility, albeit, in some cases, motility was seriously reduced. Immunogenicity of the hydrids was evaluated by immunization with live bacteria, killed bacteria, and purified flagellin (when possible). Results obtained with the new constructs were similar to the ones published for insertions involving region IV, in the sense that antibody titers to the carrier protein were very high. A low level of antibody to the inserted peptide was also detected in all groups of animals. Our results with live immunization suggest a slightly better response to the peptide when two copies are present, but the data are not conclusive.

An investigation of the biology and chemistry of the Chinese medicinal plant, Amorphophallus konjacYee, Melinda Chua Fui January 2011 (has links)
Konjac glucomannan (KGM), the main biologically active constituent of konjac flour extracted from corms of Amorphophallus konjac (konjac), can be used to prepare functional foods and may also have potential as a pharmaceutical product to combat obesity. The current study employed three experimental approaches to study the biology and chemistry of konjac, namely (1) glasshouse experiments to study the morphogenesis, growth and productivity of konjac plants, (2) a histological and immunocytochemical investigation of the localisation and developmental regulation of the deposition and metabolism of KGM in developing corm tissues, and (3) a comparative study of methodologies for the extraction and analysis of KGM. The current data demonstrated a morphological and functional separation between the ventral and dorsal regions of corms. The ventral region appeared to function as a source during the initial period of shoot development, while the dorsal region appeared to operate as a sink after the development of mature canopy. Once the corm reached maturity, both an inflorescence and a leaf were produced within a single season. It has also been demonstrated that the age of the ‘mother’ corm is an important factor affecting the quality of offsets produced. An anti-mannan antiserum detected a temporally regulated pattern of mannan epitope production within glucomannan idioblasts in developing corm tissues, with increased expression as the corm approached maturity/dormancy. The current observations also suggest that the mobilization of KGM initiates at the periphery of the corm and proceeds inwards towards the centre of the corm. Compositional analysis showed that the purified konjac flour (PKF) produced using a modified extraction procedure contained 92% glucomannan, with a weight average molecular weight (Mw), polydispersity index (PDI) and degree of acetylation (DA) of 9.5 ± 0.6 x 105 gmol-1, 1.2 and 2.8 wt. %. These data, plus Fourier-transform infrared spectral (FTIR) and zero shear viscosity analyses of the extract (PKF) were all consistent with the literature. Comparison of three existing methodologies for the quantitative analysis of the KGM content, namely 3,5-dinitrosalicylic acid (3,5-DNS), phenol-sulphuric acid and enzymatic colorimetric assays; indicated that the 3,5-DNS colorimetric assay was the most reproducible and accurate method, with a linear correlation coefficient of 0.997 and recoveries between 97% and 103% across three spiking levels of starch. In summary, this study has provided a better understanding of aspects of the biology and cultivation of A. konjac and has also produced methodologies which can be used as the basis for an improved good laboratory practice (GLP) for the commercial extraction and analysis of this multifunctional natural polymer.
