Para tanto, 32 ratos Wistar (Rattus Novergicus) adultos foram aleatoriamente distribuídos em 4 grupos: Grupo 1 (n=08): DM+TO+PE; Grupo 2 (n=08): DM+TO; Grupo 3 (n=08): não-diabético+TO+PE; Grupo 4 (n=08): não diabético+TO. O DM foi induzido nos animais do G1 e G2 pela injeção intraperitoneal de 60 mg/kg de estreptozotocina (STZ). Dez dias após a indução do DM, os animais do G1 foram induzidos à PE pela colocação da ligadura de fio de seda no sulco dos primeiros molares inferiores do lado esquerdo (lado teste); o mesmo foi feito nos animais não diabéticos (G3). O molar contralateral (direito) de todos os animais dos 4 grupos serviu como controle. O início da indução do TO, pela cimentação do fio ortodôntico na oclusal dos molares já submetidos à PE, se deu 15 dias após a colocação da ligadura (G1 e G3). No mesmo dia o TO também foi induzido nos grupos sem ligadura (G2 e G4) permanecendo por 10 dias até a data do sacrifício. A avaliação intra-grupo mostrou haver diferença estatística significante de perda óssea alveolar quando comparado os dentes do lado teste (TO) ao lado controle (p<0,0001) em todos os grupos (G1, G2, G3 e G4). A avaliação intergrupo mostrou diferenças estatísticas significantes (p<0,05) entre G1 e G2 e entre G3 e G4, com reabsorção óssea mais severa para os grupos com TO+PE quando comparados aos grupos com TO. A média de reabsorção óssea do G1 (DM+ TO+PE) foi significantemente maior (p<0,05) do que a do G3 (TO+PE). Não foram detectadas diferenças estatísticas significantes (p>0,05) entre G2 (DM+TO) e G4 (TO). Dentro dos limites deste estudo pôde-se concluir que o diabetes mellitus agrava a perda óssea alveolar nos animais com trauma oclusal quando associado à periodontite experimental. / Abstract: The aim of this study was to evaluate the diabetes mellitus (DM) effects on alveolar bone resorption induced by occlusal trauma (OT) with or without experimental periodontites (EP) in rats. Thirty-two adult male rats (Rattus Novergicus) were randomly assigned to one of four groups: Group1 (n=08): DM+OT+EP; Group 2 (n=08): DM+OT: Group 3 (n=08): OT+EP: Group 4 (n=08): OT. Experimental DM was created by intraperitoneal injection of 60 mg/kg body weight streptozotocin (STZ) in G1 and G2. Ten days after diabetes induction, the animals in G1 received a silk ligature in the sulcular area of their mandible left first molar (test side (OT) to induce EP; the same was performed with the non-diabetics animals (G3). The contralateral molar (right side) was left as a control in all of the groups. G1 and G3 were submitted to OT induction (by orthodontic wire bonding in the occlusal molar surface) after fifteen days period. At the same day, the OT was also inducted in the groups (02 and 04) without ligature. The animals were sacrificed 10 days later. The intragroup analysis revealed significant difference between teeth in the test side (OT) and control side (p<0.05) among the 4 groups (G1. G2. G3. G4). The test side (OT) showed higher levels of bone resorption. The histometrics results showed significant difference (p< 0.05) between G1 and 02 and between 03 and G4, with more severe resorption for the groups with OT+EP compared do OT. For the histological analysis, 01 (DM+OT +EP) showed more bone loss when compared to G3 (OT +EP) (p<0.05). No difference was observed (p<0.05) between G2 (DM+OT) and G4 (OT). Within the limit of the study, it was concluded that diabetes mellitus enhances the alveolar bone loss in animals with occlusal trauma associate to experimental periodontitis. / Mestrado / Periodontia / Mestre em Clínica Odontológica

Modelo de doença de Alzheimer pela infusão intracerebroventricular de estreptozotocina : influência do tempo de administração e do sexo dos animais sobre parâmetros neurogliais

Muitos trabalhos têm demonstrado a correlação existente entre gênero e prevalência da doença de Alzheimer (DA), sendo que as mulheres compreendem dois terços dos pacientes afetados. A estreptozotocina (STZ) é uma substância que vem sendo muito utilizada, por administração intracerebroventricular, para produzir um modelo de doença de Alzheimer esporádica com características neuroquímicas e fisiopatológicas semelhantes à DA, em roedores. Dessa forma, essa tese teve como objetivo avaliar as alterações comportamentais cognitivas e parâmetros neuroquímicos (ChAT, S100B, GFAP, GSH e captação de glicose) em hipocampo e cerebelo de ratos e ratas expostas ao modelo de doença de Alzheimer por STZ. Para um melhor entendimento das alterações, as avaliações foram realizadas em três diferentes tempos após a indução do modelo: duas, quatro e oito semanas. Como resultado, encontramos declínio cognitivo e alterações neuroquímicas hipocampais e cerebelares. Interessantemente, machos e fêmeas apresentaram distintas respostas frente ao insulto por STZ, mostrando ser este um campo interessante para caracterização e aprofundamento do modelo que utilizamos. Este trabalho confirma o comprometimento cognitivo e as alterações neuroquímicas presentes no modelo e estes se somam às variações funcionais encontradas nos astrócitos. Tais alterações também estão presentes na DA, apontando a interação neuroglial como um importante alvo de estudo na doença e, consequentemente, na busca por alternativas terapêuticas. / Many studies have demonstrated the correlation between gender and prevalence of Alzheimer's disease (AD), with women comprising two thirds of the affected patients. Streptozotocin (STZ) is a substance that has been widely used by intracerebroventricular administration to produce a model of sporadic Alzheimer's disease with neurochemical and pathophysiological characteristics similar to AD, in rodents. Thus, this thesis aimed to evaluate cognitive behavioral changes and neurochemical parameters (ChAT, S100B, GFAP, GSH and glucose uptake) in the hippocampus and cerebellum of rats (males and females) exposed to the model of sporadic Alzheimer's disease by STZ. For the best comprehension of the changes, the evaluations were performed at three different times after the induction of the model: two, four and eight weeks. As a result, we found cognitive decline and hippocampal and cerebellar neurochemical changes. Estudo dos efeitos da curcumina no modelo de doença de Alzheimer esporádica induzido por injeção intracerebroventricular de estreptozotocina em ratos

Defesa: Curitiba, 18/08/2017 / Inclui referências : f. 88-99 / Resumo: Alterações precoces no metabolismo cerebral da glicose e nas vias de sinalização da insulina podem participar da patogênese da forma esporádica da doença de Alzheimer (DA). Injeções intracerebroventriculares (ICV) de estreptozotocina (STZ) são usadas para mimetizar essas alterações em roedores. A STZ causa prejuízos na sinalização cerebral da insulina, estresse oxidativo, neuroinflamação e disfunções na neurogênese, os quais resultam em declínio cognitivo e são características da DA esporádica. O objetivo da primeira parte do presente trabalho foi avaliar em conjunto a pouco estudada influência da injeção ICV de STZ (3 mg/kg) sobre a neuroinflamação, a neurogênese adulta e os consequentes déficits cognitivos em ratos Wistar. A STZ causou uma resposta inflamatória aguda e crônica, representada por astrogliose (GFAP) e microgliose (Iba-1) reativas, nas áreas peri-ventriculares e no hipocampo dorsal. Isto foi acompanhado por redução da proliferação celular (Ki-67) no giro denteado (GD) do hipocampo e na zona subventricular (ZSV). Verificou-se também uma diminuição no número de neurônios imaturos (Doublecortin - DCX) e na sobrevivência, diferenciação e maturação dos novos neurônios (BrdU/NeuN). Isto resultou em prejuízos na memória espacial de curto prazo no teste de localização de objetos (TLO) e no labirinto em Y, além de prejuízos na memória espacial de longo prazo no condicionamento de medo ao contexto. Esses resultados apoiam relatos prévios de um efeito negativo da neuroinflamação sobre a neurogênese adulta. Na segunda parte do presente trabalho foram avaliados os efeitos da curcumina no modelo induzido por STZ-ICV. A curcumina é um composto polifenólico natural com evidências de propriedades antioxidantes, anti-inflamatórias e neuroprotetoras. Evidências recentes também sugerem que a curcumina melhora o desempenho cognitivo em modelos animais de demência e este efeito estaria relacionado à sua capacidade de estimular a neurogênese adulta. Dessa forma, o objetivo da segunda parte do trabalho foi testar a hipótese de que o tratamento oral prolongado com curcumina seria capaz de preservar as funções cognitivas pelo aumento da neurogênese adulta e inibição da neuroinflamação no modelo de DA esporádica induzido por STZ-ICV. Os animais foram injetados com STZ-ICV ou veículo e o tratamento com curcumina (25, 50 e 100 mg/kg, gavage) foi realizado por 30 dias, iniciando 1 h antes da cirurgia. Após quatro semanas, os animais do grupo STZ exibiram prejuízos na memória espacial (TLO e labirinto em Y) e de reconhecimento de curto prazo (TRO), diminuição da proliferação celular (Ki-67) e de neurônios imaturos (DCX) na ZSV e no GD, além de aumento dos marcadores gliais GFAP e Iba-1. O tratamento com curcumina (50 e 100 mg/kg) preveniu os prejuízos na memória de reconhecimento no TRO, mas não na memória espacial no TLO e labirinto em Y. Todas as doses de curcumina testadas exerceram uma melhora limitada na neuroinflamação (reatividade glial), resultando em ausência de efeitos significativos sobre a neurogênese hipocampal e subventricular. Esses resultados sugerem um efeito positivo da curcumina na memória de reconhecimento de objetos de curto prazo que não está relacionado a uma melhora na neurogênese adulta. Estreptozotocina. / Abstract: Early impairments in cerebral glucose metabolism and insulin signaling pathways may participate in the pathogenesis of the sporadic form of Alzheimer's disease (sAD). Intracerebroventricular (ICV) injections of low doses of streptozotocin (STZ) are used to mimic sAD these alterations in rodents. STZ causes impairments in insulin signaling and has been reported to trigger several alterations in the brain, such as oxidative stress, neuroinflammation, and dysfunctions in adult neurogenesis, which may be involved in cognitive decline and are features of human AD. The aim of the first part of the present study was to assess the poorly characterized influence of ICV injection of STZ (3 mg/kg) on neuroinflammation, adult neurogenesis and the consequent cognitive deficits in Wistar rats. STZ caused an acute and chronic neuroinflammatory response, reflected by reactive microgliosis (Iba-1) and astrogliosis (GFAP) in periventricular areas and the dorsal hippocampus, accompanied by a marked reduction of the cellular proliferation (Ki-67) in the dentate gyrus (DG) of the hippocampus and subventricular zone (SVZ). It was also verified a reduction in immature neurons (Doublecortin - DCX) and reduced survival, differentiation, and maturation of newborn neurons (BrdU/NeuN), resulting in impairments in short-term spatial memory in the object location test (OLT) and in the Y maze, and in long-term spatial memory in the contextual fear conditioning. These results further support previous reports of a detrimental effect of neuroinflammation on adult neurogenesis. In the second part of the present work, the curcumin effects in the sAD model induced by STZ-ICV were assessed. Curcumin is a natural polyphenol with evidence of antioxidant, anti-inflammatory and neuroprotective properties. Recent evidence also suggests that curcumin increases cognitive performance in animal models of dementia, and this effect would be related to its capacity to enhance adult neurogenesis. The aim of the second part of this work was to test the hypothesis that oral prolonged curcumin treatment would be able to preserve cognitive functions by increasing adult neurogenesis and decreasing neuroinflammation in the sAD model induced STZ-ICV. The animals were injected with STZ-ICV or vehicle and curcumin treatment (25, 50 and 100 mg/kg, gavage) was performed for 30 days, starting 1 h before the stereotaxic surgery. Four weeks after surgery, STZ-lesioned animals exhibited impairments in short-term spatial (TLO and Y maze) and recognition memory (object recognition test - ORT), decreased cell proliferation (Ki-67) and immature neurons (DCX) in the SVZ and DG of hippocampus, and increased immunoreactivity for the glial markers GFAP and Iba-1. Curcumin treatment in the doses of 50 and 100 mg/kg prevented the deficits in short-term recognition memory in the ORT, but not in short-term spatial memory in the OLT and Y maze. All of the curcumin doses tested exerted only slight improvements in neuroinflammation (glial reactivity), resulting in absence of significant effects on hippocampal and subventricular neurogenesis. These results suggest a positive effect of curcumin in short-term object recognition memory which was not related with enhancement in adult neurogenesis. Keywords: Alzheimer's disease. Curcumin. O modelo da Doença de Alzheimer induzido por estreptozotocina altera a sinalização hipocampal : relevância da O-GlcNAcilação de proteínas e a inflamação mediada por calcineurina

A doença de Alzheimer (DA) é caracterizada por um quadro clínico complexo e multifatorial, que envolve perdas cognitivas, déficit de memória e prejuízo na convivência social, observadas mais precocemente nessa população. O tecido cerebral dos portadores apresenta perdas neurais, disfunções neurovasculares, processos inflamatórios, gliose e lesões características da doença como as placas neuríticas e os emaranhados neurofibrilares. Ademais, as evidências apontam para similaridade entre DA e a demência induzida por diabetes mellitus, visto haver uma relação direta que liga a resistência e a deficiência na sinalização da insulina, com as mudanças no metabolismo da glicose. Essas alterações resultam em redução no estado de O-glicosilação com N-acetil-glucosamina das proteínas intracelulares, ativação das células gliais e o desequilíbrio homeostático na relação entre cinase/fosfatase desencadeada pela redução de metabólitos endógenos e aumento de cálcio intracelular. A resposta direta dessas mudanças é caracterizada pela expressão de marcadores celulares e liberação de mediadores inflamatórios que contribuem para a progressão e severidade da doença. Ainda, interações entre variações metabólicas e a ativação inflamatória alteram o microambiente do sistema nervoso central, ativando aguda e cronicamente astrócitos e microglia. Neste estudo investigamos a hipótese de que a infusão intracerebroventricular da estreptozotocina (STZ) desencadeia alterações patofisiológicas e moleculares em hipocampos. Em nossos experimentos, foram avaliadas as mudanças promovidas pela infusão de STZ após 1 e 4 semanas. Nossos resultados demonstram que a STZ afeta elementos importantes na via de sinalização da insulina no hipocampo, declínio da funções cognitivas e alterações neuroquímicas. Demonstramos que o grupo STZ apresentou níveis elevados da serina 307 do IRS-1 e redução na captação de glicose no hipocampo, possivelmente, através da resistência local à insulina e por disfunções astrocíticas. Além disso, demonstramos o impacto da ativação excessiva da calcineurina e a sinalização do fator nuclear de ativação das células T (NFAT), têm consequências a curto e longo prazo na progressão dos processos neurodegenerativos observados no modelo de DA esporádico. / Alzheimer’s disease (AD) is characterized by a complex and multifactorial clinical presentation, which includes cognitive loss, memory deficits, and impairment in social coexistence, observed earlier in this population. The patients’ brain tissue presents neural losses, neurovascular dysfunctions, inflammatory processes, gliosis, and characteristic lesions of the disease, such as senile plaques and neurofibrillary tangles. In addition, the evidence indicates to similarity shared AD and dementia induced by diabetes mellitus by occur a direct relationship of insulin resistance and deficiency in insulin signaling with changes in glucose metabolism. These changes result in lower levels of protein modified by glycosylation with O-linked β-N-acetyl-glucosamine of intracellular, in activation of glial cells, and homeostatic imbalance in the kinase/phosphatase relationship triggered by the decrease of endogenous metabolites and increase of intracellular calcium. The direct response of these changes is characterized by the expression of cellular markers and the release of inflammatory mediators that contribute to the progression and severity of the disease. Furthermore, interactions among metabolic variations and inflammatory activation alter the microenvironment of the central nervous system, activating acute and chronic astrocytes and microglia. We hypothesized that intracerebroventricular infusion of streptozotocin (STZ) triggers pathophysiological and molecular changes in hippocampus. The changes promoted by the STZ infusion were evaluated after one and four weeks. Our results show that insulin STZ affects important elements of the insulin-signaling pathway in the hippocampus, cognitive decline and neurochemical changes. Here, we show that STZ group had elevated levels of serine 307 phosphorylation of IRS-1 and decrease in the uptake of glucose into the hippocampus, possibly, through local resistance to insulin and by astrocytic dysfunction. In addition, these results indicate that aberrant calcineurin activation and nuclear factor of activated T cells (NFAT) signaling develop earlier and later consequences in the progression of the neurodegenerative processes observed in the sporadic AD model.

Concentrações plasmáticas de fator von Willlebrand. 3. Valores séricos de colesterol total e frações e triglicérides. 4. Possíveis correlações entre valores plasmáticos de homocisteína, fator von Willebrand, colesterol total e frações e triglicérides. Métodos: 35 ratos (rattus norvegicus albinus), machos, adultos (peso 180-200g), com glicemias aferidas foram randomizados em três grupos: 1. Controle (n=10) - não receberam droga ou veículo; 2. “Sham” (n=10) - receberam solução (tampão citrato 0.1M, pH4.5) veículo da estreptozotocina e 3. Diabetes (n=15) - receberam estreptozotocina (Sigma® ) para indução do diabetes melito (60 mg/kg de peso) em dose única; via intraperitoneal, diluída em 0.3mL de solução veículo. Foram considerados diabéticos todos os ratos cujos valores de glicemia foram iguais ou superiores a 250mg/dL. Após 08 semanas de indução do diabetes melito os ratos foram pesados, as glicemias aferidas e anestesiados com ionembutal (Sigma® ) via intraperitoneal (50mg/kg peso). Colheu-se sangue da artéria aorta abdominal para determinação dos valores de homocisteína plasmática total seguindo metodologia descrita, através de ensaio HPLC (Shimadzu® ). O sangue restante foi centrifugado e o soro e o plasma aliquotados e congelados até determinação do fator von Willebrand (técnica de ELISA) e concentrações séricas de glicose de jejum final, colesterol total e frações HDL-, LDL- e VLDL-colesterol, triglicérides e creatinina. Os resultados foram expressos em média + desvio padrão. Para análise estatística utilizou-se a análise de variância (ANOVA), seguido do teste de comparações múltiplas de Tukey e quando necessário o teste Brown Forsythe, seguido do procedimento de comparações múltiplas de Dunnett. Para a construção dos diagramas de dispersão das variáveis calculou-se os coeficientes de correlação de Pearson. Resultados: O modelo foi reprodutível em 100% dos animais. A média dos valores de glicemia inicial foi: 88,7±5,9mg/dL (controle); 88,9±8,2mg/dL (“sham”) e 85,1±5,2mg/dL (diabetes). Através da ANOVA não houve diferença das médias de glicemia inicial entre os grupos (p=0,24). A média da glicemia final foi: 85,0±7,1mg/dL (controle); 80,9±5,0mg/dL (”sham”) e 353,5±98,2mg/dL (diabetes). Houve diferença estatisticamente significante entre o grupo diabetes e os demais (p<0,01). A média das concentrações plasmáticas de homocisteína foi: 7,9±2,3µmol/L (controle); 8,6±2,2µmol/L (“sham”) e 6,1±1,3µmol/L (diabetes), com diferença entre os grupos (p<0,01). A média dos valores do fator von Willebrand foi 0,15±0,3U/L (controle), 0,16±0,2U/L (“sham”) e 0,18±0,4U/L (diabetes), com diferença entre os grupos (p<0,05). Os valores de colesterol total tiveram médias de: 123,9±40,8mg/dL (controle); 107,0±38,6mg/dL (“sham”) e 87,5±5,9mg/dL (diabetes). A ANOVA mostrou diferença entre os grupos (p<0,05). No grupo diabetes houve correlação inversa entre glicemia final e ganho de peso, homocisteína e colesterol total, homocisteína e fração VLDL-colesterol e homocisteína e triglicérides. Conclusões: Neste estudo, utilizando como modelo biológico o diabetes melito induzido por estreptozotocina em ratos albinos, nas condições de experimento apresentadas, torna-se lícito concluir: 1. Os ratos diabéticos apresentaram valores menores de homocisteína. 2. O grupo diabetes apresentou valores maiores de fator von Willebrand. 3. Houve correlação inversa entre homocisteína e colesterol total, homocisteína e fração VLDL-colesterol e homocisteína e triglicérides. 4. Não houve correlação entre glicemia final e homocisteína, glicemia final e fator von Willebrand e homocisteína e fator von Willebrand nos ratos diabéticos. 5. Houve correlação inversa entre ganho de peso e glicemia final nos ratos diabéticos. / Purpose: To determine plasma levels of homocysteine, von Willebrand factor, triglycerides and total cholesterol and fractions, in rats with diabetes induced by streptozotocin and evaluate possible correlations among these parameters. Methods: Adult male norvegicus albino rats (n=35), weigh (180-200g) were randomized into three groups: treated group (n=15), with diabetes induced by streptozotocin; sham group (n=10), treated with saline solution and normal control group (n=10), no treated. Initial fasting glucose was determinated before diabetes induction. Diabetes was induced by a single bolus intraperitoneal injection of streptozotocin, 60mg/kg/dose, diluted in citrate buffer (0.1M, pH4.5). Diabetes was confirmed by blood glucose levels ≥250mg/dL. Eight weeks after diabetes induction, animals were weighted and blood samples were collected from abdominal aorta for plasma levels of total homocystein, von Willebrand factor, final fasting glucose, total cholesterol and fractions (HDL-, LDL- and VLDL-cholesterol), triglycerides and creatinin. The results were expressed as the mean+SD. Data were analyzed using analysis of variance (ANOVA) followed by Tukey test or Brown Forsythe test followed by Dunnett test. Pearson test was used to correlate the parameters. The level of significance was set at p<0.05. Results: Disease model reproducibility was observed in 100% of tested animals. Mean plasma levels of initial fasting glucose according to animal group were: 85.1±5.2mg/dL for treated group, 88.9±8.2mg/dL for sham and 88.7±5.9mg/dL for control group. Statistical analysis (ANOVA) showed no significant difference among the three groups (p=0.24). Mean plasma levels of final fasting glucose, according to animal group were: 353.5±98.2mg/dL for treated group, 80.9±5.0mg/dL for sham and 85.0±7.1mg/dL for normal control. Statistical analysis showed that the difference between the treated group and the two other groups was statistically significant (p<0.01). Mean plasma concentration of homocystein according to animal groups was: 6.1±1.3µmol/L for treated group, 8.6±2.2µmol/L for sham and 7.9±2.3µmol/L for control group. Statistical analysis showed a significant difference in the three groups (p<0.01). Mean values for von Willebrand factor were: 0.18±0.4U/L for treated group, 0.16±0.2U/L for sham group and 0.15±0.3U/L for control group. Significant statistical difference was found among the three groups (p<0.05). Mean plasma levels of total cholesterol according to animal groups were: 87.5±5.9mg/dL for treated group, 107.0±38.6mg/dL for sham and 123.9±40.8mg/dL for control group. Significant statistical difference was found among the three groups (p<0.05). There was no statistical significant difference in the mean values of cholesterol fraction (LDL, HDL, VLDL), triglycerides and creatinin among the three groups. A negative correlation between final plasma glucose and weight gain, homocystein and total cholesterol, homocystein and VLDL-cholesterol and homocystein and triglycerides was found in the treated (diabetes) group. Conclusions: In this study, results from streptozotocin-induced diabetes rats showed: 1. Lower levels of homocystein; 2. Higher levels of von Willebrand factor; 3. Negative correlation between homocystein and total cholesterol; 4. No correlations between final fasting glucose and homocystein, final fasting glucose and von Willebrand factor and homocystein and von Willebrand factor; 5. Perfil bioquímico e molecular das ectonucleotidases no sistema cardiovascular de ratos diabéticos por estreptozotocina

Ectonucleotidases constituem uma cascata enzimática altamente organizada para regulação da sinalização mediada por nucleotídeos, controlando a quantidade e a taxa de degradação destes, além da formação do respectivo nucleosídeo. Nucleotídeos/ nucleosídeos extracelulares são conhecidos por regularem diversas respostas fisiológicas, incluindo o tônus vascular, função cardíaca e homeostasia. Neste estudo nós analisamos a presença dos membros das famílias das E-NTPDases, E-NPPs e ecto-5'-nucleotidase em tecido de ventrículo esquerdo (VE) de ratos. A análise por PCR mostrou a expressão da NTPDase1, 2, 3,5,6, ecto-5'-NT/CD73, NPP2 e NPP3, excluindo a presença da NPP1 e NTPDase8. Através da análise por PCR em tempo real, encontramos o maior nível de expressão de mRNA para a NTPDase2 no ventrículo esquerdo. Também caracterizamos as propriedades bioquímicas e atividades enzimáticas em sinaptossomas de terminal nervoso (sinaptossoma cardíaco) em VE. No estudo das propriedades das E-NTPDases e ecto-5'-NT, observamos dependência de cátions divalentes, pH ótimo de 8.0 para as hidrólises de ATP e ADP e 9.5 para a hidrólise de AMP. Os valores de Km aparente são 40 µM, 90 µM, 39 µM e os valores de Vmax aparente são 537, 219 and 111 nmol Pi liberado/min/mg de proteína para as hidrólises de ATP, ADP e AMP, respectivamente. Oligomicina e azida, ambos inibidores de ATPases mitocondriais, inibiram somente a hidrólise de ATP. A hidrólise de AMP não foi afetada por levamisole e tetramisole, enquanto molibdato de amônio praticamente aboliu a atividade da ecto-5'-nucleotidase. Utilizando o p-nitrofenil-5'-timidina monofosfato (p-Nph-5'-TMP) como substrato marcador para E-NPPs na preparação de sinaptossoma cardíaco, observamos um pH alcalino, dependência de cátions divalentes e valores de Km de 91.42 ± 13.97 µM e Vmax calculado de 63.79 ± 3.59 nmol p-nitrofenol liberado/min/mg de proteína. Não houve alteração na hidrólise do substrato com adição de levamisole no pH de 8.9 (pH ótimo caracterizado), porém o suramin reduziu fortemente a hidrólise de p-Nph-5'-TMP. Para estudarmos se estas enzimas poderiam participar na patofisiologia das doenças diabéticas cardiovasculares, usamos um modelo de diabetes experimental por estreptozotocina (STZ) 65mg/kg. A injeção de STZ causou aumento da glicemia, perda de peso corporal, diminuição dos batimentos cardíacos e pressão sangüínea. Seis dias de tratamento com insulina foram capazes de reverter as alterações encontradas no diabetes. Houve uma inibição das hidrólises de ATP e ADP, mas não de AMP e p-Nph-5'-TMP em sinaptossoma cardíaco. O tratamento com insulina reverteu as hidrólises para os níveis do controle (citrato). A análise do metabolismo extracelular de ATP em sinaptossoma cardíaco por HPLC mostrou que o ATP é hidrolisado mais lentamente no grupo diabético. Dados preliminares de análise molecular sugerem a participação das NTPDases 1 e 2 nas hidrólises de nucleotídeos em sinaptossoma cardíaco. Com o intuito de analisar um material biológico de fácil obtenção, avaliamos as atividades das ectonucleotidases e o perfil extracelular de hidrólise de ATP em soro de ratos diabéticos e também a influência da glicemia nestas atividades. Trinta dias após a injeção de STZ, observamos um aumento nas hidrólises de ATP, ADP, AMP e p-Nph-5'-TMP, quando comparados ao grupo citrato e este aumento foi revertido com 6 dias de tratamento com insulina. Glicose (20 mM), in vitro, não afetou a atividade enzimática em soro de ratos controle, após 24 h de pré-incubação. A análise por HPLC mostrou uma rápida hidrólise de ATP em soro de animais diabéticos, diminuindo os níveis de ATP e ADP mais rapidamente do que os grupos controle e insulina. Mostramos uma alteração na hidrólise de nucleotídeos em sinaptossoma cardíaco e em soro de ratos diabéticos. A alteração encontrada em coração parece fazer parte da etiologia da doença cardiovascular diabética. No entando, a alteração encontrada no soro dos ratos diabéticos, diminuição de ADP (pró-agregante plaquetário) e aumento de adenosina (antiagregante plaquetário) parece participar de um sistema de proteção a possíveis danos vasculares decorrentes do diabetes. / Ectonucleotidases constitute a highly organized enzymatic cascade in the regulation of nucleotidemediated signaling, controlling the rate, amount and timing of nucleotide degradation and ultimately, the nucleoside formation. Extracellular nucleotides/nucleosides are known to regulate several physiological responses, including vascular tone, cardiac function and haemostasis. In this study, we have analyzed the presence of the E-NTPDase family members, ecto-5'-nucleotidase/CD73 and E-NPPs in rat heart left ventricle. RT-PCR analysis from left ventricle tissue demonstrated different levels of expression of NTPDase1, 2, 3, 5, 6, 5'-NT/CD73, NPP2 and NPP3, but excluded the presence of NPP1 and NTPDase8. By quantitative real-time PCR we identified the NTPDase2 as the enzyme with the highest mRNA expression in rat left ventricle. Moreover, we characterized the biochemical properties and enzyme activities from synaptosomes of the nerve terminal endings (cardiac synaptosomes) of heart left ventricle. In the E-NTPDases study, we observed divalent cation-dependent enzymes that presented optimum pH of 8.0 for ATP and ADP hydrolysis, and 9.5 for AMP hydrolysis. The apparent Km values are 40 µM, 90 µM and 39 µM and apparent Vmax values are 537, 219 and 111 nmol Pi released/min/mg of protein for ATP, ADP and AMP hydrolysis, respectively. Oligomycin and sodium azide, both mitochondrial ATPase inhibitors, inhibited only the ATP hydrolysis. AMP hydrolysis was not affected by levamisole and tetramisole, whereas ammonium molybdate practically abolished the ecto-5'-nucleotidase activity. Using p-nitrophenyl-5'- thymidine monophosphate (p-Nph-5'-TMP) as substrate for E-NPPs in rat cardiac synaptosomes, we observed an alkaline pH dependence, divalent cation dependence and the Km value corresponded to 91.42 ± 13.97 μM and Vmax value calculated was 63,79 ± 3,59 nmol p-nitrophenol released/min/mg of protein. Levamisole was ineffective as inhibitor of p-Nph-5'-TMP hydrolysis in pH 8.9 (optimum pH for the enzyme characterized). Suramin strongly reduced the hydrolysis of p- Nph-5'-TMP. In order to study if these enzymes could participate in the pathophysiology of diabetic cardiovascular diseases, we induced experimental diabetes with an injection of streptozotocin 65mg/kg to study diabetic complications. The STZ injection caused a rise in glucose levels, loss of body weight, lower heart rate (HR) and systolic blood pressure (SBP) when compared to citrate group (control). The 6 days insulin treatment was able to reverse these parameters. There was an inhibition in ATP and ADP, but not in AMP and p-Nph-5'-TMP hydrolysis in cardiac synaptosomes and the insulin treatment returned the hydrolysis to the citrate group levels. The analysis of extracellular ATP metabolism in cardiac synaptosomes by HPLC shows that ATP was hydrolyzed slower in diabetes group. Preliminar data from molecular analysis suggest the participação of NTPDases 1 and 2 on nucleotide hydrolysis in cardiac synaptosomes. In order to analyse a readily available biological material, we evaluated the ectonucleotidases activities and the profile of extracellular ATP metabolism in blood serum of streptozotocin-diabetic rats and the influence of glycemic status. The 30 days of STZ injection was associated with a raise in ATP, ADP, AMP, and 5'-TMP hydrolysis in blood serum, when compared to the citrate group. In vitro, 20 mM glucose added did not affect ectonucleotidases activities in normal blood serum of rats with 24 h of preincubation. HPLC analysis showed a rapid hydrolysis of extracellular ATP by diabetic animals, decreasing the ATP and ADP levels faster than in serum of control and insulin groups, leading to the formation of high levels of adenosine when compared with citrate and insulin groups. We showed alterations in the nucleotides hydrolysis in the cardiac synaptosomes and in serum from diabetic rats. The cardiac alteration seems to be related to the ethiology of cardiac problems. However, the serum hydrolysis alterations seems to be related to a vascular protection, with the diminution of ADP (pro-agregant) and an increase in adenosine (antiagregant), found in diabetic serum rats.

O modelo da Doença de Alzheimer induzido por estreptozotocina altera a sinalização hipocampal : relevância da O-GlcNAcilação de proteínas e a inflamação mediada por calcineurina

A doença de Alzheimer (DA) é caracterizada por um quadro clínico complexo e multifatorial, que envolve perdas cognitivas, déficit de memória e prejuízo na convivência social, observadas mais precocemente nessa população. O tecido cerebral dos portadores apresenta perdas neurais, disfunções neurovasculares, processos inflamatórios, gliose e lesões características da doença como as placas neuríticas e os emaranhados neurofibrilares. Ademais, as evidências apontam para similaridade entre DA e a demência induzida por diabetes mellitus, visto haver uma relação direta que liga a resistência e a deficiência na sinalização da insulina, com as mudanças no metabolismo da glicose. Essas alterações resultam em redução no estado de O-glicosilação com N-acetil-glucosamina das proteínas intracelulares, ativação das células gliais e o desequilíbrio homeostático na relação entre cinase/fosfatase desencadeada pela redução de metabólitos endógenos e aumento de cálcio intracelular. A resposta direta dessas mudanças é caracterizada pela expressão de marcadores celulares e liberação de mediadores inflamatórios que contribuem para a progressão e severidade da doença. Ainda, interações entre variações metabólicas e a ativação inflamatória alteram o microambiente do sistema nervoso central, ativando aguda e cronicamente astrócitos e microglia. Neste estudo investigamos a hipótese de que a infusão intracerebroventricular da estreptozotocina (STZ) desencadeia alterações patofisiológicas e moleculares em hipocampos. Em nossos experimentos, foram avaliadas as mudanças promovidas pela infusão de STZ após 1 e 4 semanas. Nossos resultados demonstram que a STZ afeta elementos importantes na via de sinalização da insulina no hipocampo, declínio da funções cognitivas e alterações neuroquímicas. Demonstramos que o grupo STZ apresentou níveis elevados da serina 307 do IRS-1 e redução na captação de glicose no hipocampo, possivelmente, através da resistência local à insulina e por disfunções astrocíticas. Além disso, demonstramos o impacto da ativação excessiva da calcineurina e a sinalização do fator nuclear de ativação das células T (NFAT), têm consequências a curto e longo prazo na progressão dos processos neurodegenerativos observados no modelo de DA esporádico. / Alzheimer’s disease (AD) is characterized by a complex and multifactorial clinical presentation, which includes cognitive loss, memory deficits, and impairment in social coexistence, observed earlier in this population. The patients’ brain tissue presents neural losses, neurovascular dysfunctions, inflammatory processes, gliosis, and characteristic lesions of the disease, such as senile plaques and neurofibrillary tangles. In addition, the evidence indicates to similarity shared AD and dementia induced by diabetes mellitus by occur a direct relationship of insulin resistance and deficiency in insulin signaling with changes in glucose metabolism. These changes result in lower levels of protein modified by glycosylation with O-linked β-N-acetyl-glucosamine of intracellular, in activation of glial cells, and homeostatic imbalance in the kinase/phosphatase relationship triggered by the decrease of endogenous metabolites and increase of intracellular calcium. The direct response of these changes is characterized by the expression of cellular markers and the release of inflammatory mediators that contribute to the progression and severity of the disease. Furthermore, interactions among metabolic variations and inflammatory activation alter the microenvironment of the central nervous system, activating acute and chronic astrocytes and microglia. We hypothesized that intracerebroventricular infusion of streptozotocin (STZ) triggers pathophysiological and molecular changes in hippocampus. The changes promoted by the STZ infusion were evaluated after one and four weeks. Our results show that insulin STZ affects important elements of the insulin-signaling pathway in the hippocampus, cognitive decline and neurochemical changes. Here, we show that STZ group had elevated levels of serine 307 phosphorylation of IRS-1 and decrease in the uptake of glucose into the hippocampus, possibly, through local resistance to insulin and by astrocytic dysfunction. In addition, these results indicate that aberrant calcineurin activation and nuclear factor of activated T cells (NFAT) signaling develop earlier and later consequences in the progression of the neurodegenerative processes observed in the sporadic AD model.

No entando, a alteração encontrada no soro dos ratos diabéticos, diminuição de ADP (pró-agregante plaquetário) e aumento de adenosina (antiagregante plaquetário) parece participar de um sistema de proteção a possíveis danos vasculares decorrentes do diabetes. / Ectonucleotidases constitute a highly organized enzymatic cascade in the regulation of nucleotidemediated signaling, controlling the rate, amount and timing of nucleotide degradation and ultimately, the nucleoside formation. Extracellular nucleotides/nucleosides are known to regulate several physiological responses, including vascular tone, cardiac function and haemostasis. In this study, we have analyzed the presence of the E-NTPDase family members, ecto-5'-nucleotidase/CD73 and E-NPPs in rat heart left ventricle. RT-PCR analysis from left ventricle tissue demonstrated different levels of expression of NTPDase1, 2, 3, 5, 6, 5'-NT/CD73, NPP2 and NPP3, but excluded the presence of NPP1 and NTPDase8. By quantitative real-time PCR we identified the NTPDase2 as the enzyme with the highest mRNA expression in rat left ventricle. Moreover, we characterized the biochemical properties and enzyme activities from synaptosomes of the nerve terminal endings (cardiac synaptosomes) of heart left ventricle. In the E-NTPDases study, we observed divalent cation-dependent enzymes that presented optimum pH of 8.0 for ATP and ADP hydrolysis, and 9.5 for AMP hydrolysis. The apparent Km values are 40 µM, 90 µM and 39 µM and apparent Vmax values are 537, 219 and 111 nmol Pi released/min/mg of protein for ATP, ADP and AMP hydrolysis, respectively. Oligomycin and sodium azide, both mitochondrial ATPase inhibitors, inhibited only the ATP hydrolysis. AMP hydrolysis was not affected by levamisole and tetramisole, whereas ammonium molybdate practically abolished the ecto-5'-nucleotidase activity. Using p-nitrophenyl-5'- thymidine monophosphate (p-Nph-5'-TMP) as substrate for E-NPPs in rat cardiac synaptosomes, we observed an alkaline pH dependence, divalent cation dependence and the Km value corresponded to 91.42 ± 13.97 μM and Vmax value calculated was 63,79 ± 3,59 nmol p-nitrophenol released/min/mg of protein. Levamisole was ineffective as inhibitor of p-Nph-5'-TMP hydrolysis in pH 8.9 (optimum pH for the enzyme characterized). Suramin strongly reduced the hydrolysis of p- Nph-5'-TMP. In order to study if these enzymes could participate in the pathophysiology of diabetic cardiovascular diseases, we induced experimental diabetes with an injection of streptozotocin 65mg/kg to study diabetic complications. The STZ injection caused a rise in glucose levels, loss of body weight, lower heart rate (HR) and systolic blood pressure (SBP) when compared to citrate group (control). The 6 days insulin treatment was able to reverse these parameters. There was an inhibition in ATP and ADP, but not in AMP and p-Nph-5'-TMP hydrolysis in cardiac synaptosomes and the insulin treatment returned the hydrolysis to the citrate group levels. The analysis of extracellular ATP metabolism in cardiac synaptosomes by HPLC shows that ATP was hydrolyzed slower in diabetes group. Preliminar data from molecular analysis suggest the participação of NTPDases 1 and 2 on nucleotide hydrolysis in cardiac synaptosomes. In order to analyse a readily available biological material, we evaluated the ectonucleotidases activities and the profile of extracellular ATP metabolism in blood serum of streptozotocin-diabetic rats and the influence of glycemic status. The 30 days of STZ injection was associated with a raise in ATP, ADP, AMP, and 5'-TMP hydrolysis in blood serum, when compared to the citrate group. In vitro, 20 mM glucose added did not affect ectonucleotidases activities in normal blood serum of rats with 24 h of preincubation. HPLC analysis showed a rapid hydrolysis of extracellular ATP by diabetic animals, decreasing the ATP and ADP levels faster than in serum of control and insulin groups, leading to the formation of high levels of adenosine when compared with citrate and insulin groups. We showed alterations in the nucleotides hydrolysis in the cardiac synaptosomes and in serum from diabetic rats. The cardiac alteration seems to be related to the ethiology of cardiac problems. However, the serum hydrolysis alterations seems to be related to a vascular protection, with the diminution of ADP (pro-agregant) and an increase in adenosine (antiagregant), found in diabetic serum rats.
