A Quantitative Description of the Interaction of Enhancement and Depression of Transmitter Release at the Neuromuscular Junction

Synaptic transmission alters the strength of the postsynaptic potential, through a process called short-term synaptic plasticity (STP). In this study, endplate potentials (EPPs) from the frog neuromuscular junction were used to resolve and quantify the presynaptic components involved in enhancement and depression of transmitter release during repetitive stimulation under normal quantal release conditions (2 mM Ca2+, 1mM Mg2+). During trains of stimulation given between 10 - 200 Hz, the amplitude of the EPPs first increased then decreased; a maximum increase of 77% was produced after 2-4 stimuli. EPP amplitudes began to increase at ~ 20 Hz, were maximal at ~ 55 Hz, and thereafter, decreased as the rate of stimulation increased. The integrated total release after 25 stimuli was little changed across frequencies between 10 - 100 Hz. EPPs ran down in two phases: a fast phase, attributed to the depletion of a readily releasable pool (RRP) of synaptic vesicles, followed by a slow phase, attributed to the depletion of vesicles from a depot pool (DP). Depletion of the readily releasable pool of synaptic vesicles (RRP) was determined by quantifying release under the fast and slow time rundowns and subtracting the number of vesicles associated with mobilization to the RRP from the total number of vesicles released during stimulation trains of 50 impulses. Impulses were delivered at 12 different rates ranging from 50 to 200 /s. Estimates of the number of vesicles released from the RRP increased with frequency of stimulation until maximal depletion levels of 5500 - 6000 vesicles were reached at stimulation rates between 90-130/s, assuming a control quantal content of 200 vesicles released per impulse. Depletion was less at lower frequencies when the number of stimuli delivered was identical. When the RRP maximally depleted, release was inversely related to stimulation rate, as would be expected if mobilization from the depot pool was the sole determinate of release during the slow phase. An equation constructed from four known components of enhancement and two components of depression - the depletion of vesicles from a readily releasable pool (RRP) and from the depot pool (DP) that refills the RRP, was used to fit and then simulate EPPs obtained during trains using different patterns of stimulation and varying amounts of extracellular Ca2+; the decay time constant parameters of enhancement, numerically derived from the observed data, were fixed at tau ~ 46, 220, 1600, and 20000 ms. The number of components of enhancement necessary to approximate the data decreased, from four in low (0.14 - 0.2mM) extracellular Ca2+, to one (tau ~ 46 ms) in 2.0 mM extracellular Ca2+, but four components of enhancement were necessary to fit the data when the amplitude of the EPP was not depressed below the control amplitude. This model was able to predict within ~ 3 % EPP amplitudes over a 10-fold range of frequency and Ca2+ concentration.

Developmental and reproductive regulation of NR5A genes in teleosts

In mammals sex chromosomes direct and initiate the development of male and female gonads and subsequently secondary sex characteristics. In most vertebrates each individual is pre-destined to either become male or female. The process by which this genetic decision is carried out takes place during the embryonic development and involves a wide range of genes. The fushi tarazu factor-1 (FTZ-F1) is a nuclear receptor and transcription factor, which in mammals has proven to be essential for gonad development and directs the differentiation of testicular Sertoli cells. A mammalian FTZ-F1 homologue subtype, steroidogenic factor-1 (SF-1), is a member of the nuclear receptor 5A1 (NR5A1) group and regulate several enzymes involved in steroid hormone synthesis. It also regulates the expression of the gonadotropin releasing hormone receptor GnRHr and the β-subunit of the luteinizing hormone (LH), indicating that it functions at all levels of the reproductive axis. Another mammalian FTZ-F1 subtype, NR5A2, is in contrast to SF-1, not linked to steroidogenesis or sex determination. Rather, NR5A2 is involved in cholesterol metabolism and bile acid synthesis in liver. Hormones and environmental factors such as temperature and pH can influence teleost development and reproductive traits, rendering them vulnerable to pollutants and climate changes. Very little is known about teleost FTZ-F1 expression, regulation and function. In this thesis, expression patterns of four zebrafish FTZ-F1 genes (ff1a, b, c and d) and two Arctic char genes (acFF1α and β) were studied during development, displaying complex embryonic expression patterns. Ff1a expression was in part congruent with expression of both mammalian NR5A1 and NR5A2 genes but also displayed novel expression domains. The complexity of the expression pattern of ff1a led to the conclusion that the gene may be involved in several developmental processes, including gonad development, which also was indicated by its transcriptional regulation via Sox9a. Two ff1a homologues were also cloned in Arctic char and were shown to be involved in the reproductive cycle, as the expression displayed seasonal cyclicity and preceded that of the down stream steroidogenic genes StAR and CYP11A. High levels were correlated to elevated plasma levels of 11-ketotestosterone (11KT) in males and 17β-estradiol (E2) in females respectively. Treatment with 11KT did not affect FTZ-F1 expression directly but was indicated to alter expression of CYP11A and 3β-hydroxysteroid dehydrogenase. E2 treatment was indicated to down-regulate the expression of testicular FTZ-F1, which may contribute to the feminising effect previously observed in E2 treated salmonids. Ff1d is a novel FTZ-F1 gene, expressed in pituitary and interrenal cells during development, suggesting steroidogenic functions. In adult testis and ovary ff1d was co-expressed with anti-Mullerian hormone (AMH), a gene connected to sex determination in mammals and previously not characterised in teleost fish. The co-expression between ff1d and AMH was found in Sertoli and granulosa cells, which is congruent with the co-expression of mammalian SF-1 and AMH. This suggests that ff1d and AMH may have similar functions in teleost sex differentiation and reproduction, as their mammalian homologues. In conclusion, this study present data that connects members of the teleost FTZ-F1 family to reproduction, cholesterol metabolism and sex determination and differentiation.

A Document Similarity Measure and Its Applications

In this paper, we propose a novel similarity measure for document data processing and apply it to text classification and clustering. For two documents, the proposed measure takes three cases into account: (a) The feature considered appears in both documents, (b) the feature considered appears in only one document, and (c) the feature considered appears in none of the documents. For the first case, we give a lower bound and decrease the similarity according to the difference between the feature values of the two documents. For the second case, we give a fixed value disregarding the magnitude of the feature value. For the last case, we ignore its effectiveness. We apply it to the similarity based single-label classifier k-NN and multi-label classifier ML-KNN, and adopt these properties to measure the similarity between a document and a specific set for document clustering, i.e., k-means like algorithm, to compare the effectiveness with other measures. Experimental results show that our proposed method can work more effectively than others.

Costos de movilidad intersectorial del empleo, shocks comerciales y tecnológicos: resultados para Argentina en base a un modelo estructural

A partir de un modelo de ajuste dinámico en el mercado laboral se estiman costos de movilidad intersectorial del empleo para Argentina durante el período 1996-2009, siguiendo la metodología desarrollada en Artuç, Chaudhuri and McLaren (2010). Considerando los parámetros estimados se computan simulaciones para evaluar el impacto potencial de shocks comerciales y tecnológicos sobre la asignación sectorial del empleo, los salarios sectoriales y el bienestar de los trabajadores. Nuestros resultados indican la presencia de altos costos de ajuste en el empleo, lo que implica un ajuste lento en el mercado de trabajo como respuesta a shocks tecnológicos y comerciales. Encontramos que los costos son mayores para los trabajadores no calificados respecto de los trabajadores semi calificados, no difieren entre el sector del país que comprende al Gran Buenos Aires y la Región pampeana respecto al resto del país y presentan una estructura sectorial similar a la hallada por Artuç et al. (2010) para Estados Unidos. / This paper estimates workers’ intersectoral switching costs based on a dynamic model of labor adjustment using data for Argentina (1996-2009). The estimated parameters are incorporated into a neoclassical model of trade to simulate the dynamic equilibrium impact (on welfare, wages and labor allocation) of trade shocks and technological changes. The approach used in this paper follows the method developed in Artuç, Chaudhuri and McLaren (2010). Our estimates show that Argentinian workers face high average intersectoral adjustment costs. This result suggests that the adjustment of the labor market in response to shocks is slow.

Evaluation of Supervised Machine LearningAlgorithms for Detecting Anomalies in Vehicle’s Off-Board Sensor Data

A diesel particulate filter (DPF) is designed to physically remove diesel particulate matter or soot from the exhaust gas of a diesel engine. Frequently replacing DPF is a waste of resource and waiting for full utilization is risky and very costly, so, what is the optimal time/milage to change DPF? Answering this question is very difficult without knowing when the DPF is changed in a vehicle. We are finding the answer with supervised machine learning algorithms for detecting anomalies in vehicles off-board sensor data (operational data of vehicles). Filter change is considered an anomaly because it is rare as compared to normal data. Non-sequential machine learning algorithms for anomaly detection like oneclass support vector machine (OC-SVM), k-nearest neighbor (K-NN), and random forest (RF) are applied for the first time on DPF dataset. The dataset is unbalanced, and accuracy is found misleading as a performance measure for the algorithms. Precision, recall, and F1-score are found good measure for the performance of the machine learning algorithms when the data is unbalanced. RF gave highest F1-score of 0.55 than K-NN (0.52) and OCSVM (0.51). It means that RF perform better than K-NN and OC-SVM but after further investigation it is concluded that the results are not satisfactory. However, a sequential approach should have been tried which could yield better result.

Appropriation de la lecture et de l'écriture du français au Cameroun : cas des lycées bilingues de Yaoundé et de Buea / Ownership of French Language Reading and Writing Skills in Cameroon : the case of Bilingual Grammar Schools in Yaounde and Buea

Dans un Cameroun, officiellement bilingue, le constat de la dégradation de l'apprentissage de la lecture et de l'écriture du français, langue seconde/étrangère (FLS/FLE), est clairement établi. Notre travail de recherche porte sur l'origine de ces carences, en utilisant des méthodes de sociolinguistique et de didactique. Notre étude, sur le terrain scolaire, a analysé l'appropriation du français dispensé par des enseignants à des élèves de première année de secondaire, en système anglophone et francophone camerounais. Dans cette première année de post primaire, de niveau équivalent pour les deux sous systèmes éducatifs, nous avons analysé le déroulement des apprentissages du français et son utilisation. Notre choix de lieu d'enquête s'est porté, dans les Lycées bilingues de Buea et de Yaoundé, en zones urbaines, sur deux classes de 6ème (francophones) et deux classes de F1(anglophones). Nous avons constaté, dans cette étude synchronique, que les recommandations officielles du domaine linguistique pour le français sont mal connues des enseignants, que les éléments de base, dispensés au cycle primaire, sont mal assimilées par les élèves, qu'il existe des incompétences dans la production de certains phonèmes, que les règles prosodiques du français ne sont pas suivies (absence ou mauvaises liaisons, perte ou insertion, ou déformation/confusion de certains sons, etc.), que la production de l'intonation est délétère. Des facteurs sociaux et didactiques ont aussi été identifiés comme des obstacles à l'appropriation de la lecture et de l'écriture (plurilinguisme, faible intérêt pour l'écrit, rareté et mauvais usage des manuels scolaires, effectifs pléthoriques, indiscipline chronique, projets pédagogiques absents ou mal structurés, méthodes et méthodologies non efficientes, etc.). Toutes ces pratiques et ces facteurs précédents placent les élèves du corpus d'étude dans une insécurité linguistique grandissante. Des pistes possibles de remédiation à la dégradation de ces apprentissages se dégagent de notre recherche. Il serait souhaitable d'instaurer des pratiques pouvant favoriser la construction de micro processus qui permettraient d'installer la compétence de lecture et d'écriture chez les élèves : étude de la phonologie, amélioration des pratiques d'oral, production et reproduction constantes d'écrits... / In Cameroon, with two official languages, findings clearly show that there is a drop in learning how to read and write French as a second/foreign language (FSL/FFL). This project, using the sociolinguistic and didactic method, looks at the origin of these gaps. The study which was carried out in schools analyzed the ownership of French taught by teachers to first year students in secondary schools in the English-speaking subsystem and French-speaking subsystem. In the first year of post primary education, at the equivalent levels for both subsystems of education, this study analyzed the learning and usage of French. The study was carried out in Bilingual High Schools in urban areas, Buea and Yaounde, in two classes of 6ème for French-speaking students and two classes of Form One for English-speaking students. The synchronic study made the following findings: the official linguistic recommendations for French Language are poorly understood by teachers; basic elements taught in the primary school are poorly assimilated by students; poor production of certain phonemes; students do not follow French prosodic rules (absence or poor liaison, loss or insertion, or deformation/confusion of some sounds, etc.); and production of intonation is poor. Social and didactic factors were also identified as obstacles to the ownership of reading and writing (multilingualism, low interest in writing, scarcity and misuse of textbooks, overstaffing, chronic indiscipline, lack or poorly structured teaching plans, inefficient methods and methodologies, etc.). All these factors and practices put the students in this study under growing linguistic insecurity. The study further makes recommendations to remedy the gap in the learning of French. It would be desirable to introduce practices that promote the building of micro processes that will help students acquire skills in reading and writing: study of phonology, improvement of oral practices, constant production and reproduction of writing.

An assessment of demographic parameters of African rhinoceros species (Diceros bicornis and Ceratotherium simum) and their significance to management in captivity

Captive-breeding has been identified as an integral part of the conservation of threatened species. The black rhinoceros (Diceros bicornis) as a species is currently listed by the IUCN as critically endangered (CR), while the white rhinoceros’s (Ceratotherium simum) current status is near-threatened (NT). Three African rhinoceros subspecies currently occur in captive populations in regional population management programmes, namely the South-central black rhinoceros (Diceros bicornis minor), the Eastern black rhinoceros (Diceros bicornis michaeli) and the Southern white rhinoceros (Ceratotherium simum simum). Concerns have been raised, however, that these populations are not self-sustainable. This study aims to analyze the demographic parameters that contribute to the growth rates of the global captive populations of African rhinoceros. The study sets out to determine if these global captive populations are currently self-sustaining, which demographic factors have the most influence on the population growth rates, and whether or not 50- and 100- year targets set for the captive African rhinoceros populations are attainable. Demographic data from 1 January 2010 until 31 December 2010 were analysed through population census, life table and age structure analyses. Two additional concerns, namely poor reproductive performance of the female F1 generation and male-biased birth sex ratios, were also assessed. The results indicate that the captive populations of D. b. minor and C. s. simum are not self-sustainable, with population growth rates (λ) of 0.98 and 0.99, respectively. Diceros bicornis michaeli is the only subspecies with a growing population, with a population growth rate of 1.02. Sensitivity analyses conclude that fecundity rates, and not mortality rates, are the limiting factor to population growth in all three subspecies. While lifetime reproductive success values for D. b. minor and C. s. simum captive-born females are far lower than those of the founder generation, several factors need further investigation to determine the true cause of this. Birth sex ratio analysis shows no significant difference from parity for both black rhinoceros subspecies, however, quadratic logit regression conducted on the white rhinoceros data indicated a statistically significant male-bias. In all three subspecies, no significant link was found between maternal age and the sex of the offspring. Overall, the results of this study show that the 50 year and 100 year targets set at the GCAP workshop in 1992 are achievable. However, D. b. minor will only be able to achieve the target population sizes with an increase in fecundity rate of around 170% or alternatively, additional supplementation from the wild. Inhibition of <em>Escherichia coli</em> ATP Synthase by Polyphenols and Their Derivatives.

We have studied the inhibitory effect of natural and structurally modified polyphenols on Escherichia coli ATP synthase to test (I) if the beneficial dietary effects of polyphenols are related to their inhibitory actions on ATP synthase, (II) if inhibitory effects of polyphenolic compound could be augmented through structural modifications, and (III) if they can act as antimicrobial agent through their actions on ATP synthesis. X-ray crystal structures of polyphenol binding sites suggested that polyphenols bind at a distinct polyphenol binding pocket, at the interface of α,β,γ-subunits. We found that both natural and modified polyphenols inhibit E. coli ATP synthase to varying degrees and structural modifications resulted in augmented inhibition. Inhibition was reversible in all cases. Both natural and modulated compounds inhibited E. coli cell growth to varying degrees. We conclude that dietary benefits of polyphenols may be in part due to the inhibition of ATP synthase.

The Effects of Parental Carbamazepine and Gemfibrozil Exposure on Sexual Differentiation in Zebrafish (Danio rerio)

Endocrine-disrupting compounds (EDCs) interfere with the physiology of hormone systems. Traditionally, steroidogenic pharmaceuticals have been studied as EDCs however there has been growing evidence that non-steroidogenic pharmaceuticals can alter sex steroid levels and impair reproductive functions in fish. This is of concern as pharmaceuticals are detected in surface waters at the ng L-1 to µg L-1 range. Zebrafish (Danio rerio) were exposed to 10 µg L-1 of the pharmaceuticals carbamazepine and gemfibrozil for 6 weeks. Male-biased sex ratios were observed in the sexually mature offspring after paternal exposure, suggesting that sexual differentiation may be impacted in juveniles. Currently, the ability of pharmaceuticals to interfere with sexual differentiation of parentally exposed offspring is unknown. This thesis examined the gonad histology of juvenile zebrafish to understand how sexual differentiation was affected in the offspring of exposed parents. Paternal, but not maternal, exposure to carbamazepine resulted in a significantly faster sexual differentiation of the gonads and led to a male-biased sex ratio; these effects were not observed when both parents were exposed. Combined paternal and maternal exposure to gemfibrozil resulted in significantly faster sexual differentiation and paternal, but not maternal, exposure to gemfibrozil led to male-biased sex ratios. Interestingly, sex ratios observed in the juveniles did not always reflect those found in the same lineage at sexual maturity, suggesting a sex reversal, including a male to female transition, occurred past the juvenile sexual differentiation period in some fish. This thesis demonstrates that pharmaceuticals have the ability to disrupt sexual differentiation in the F1 offspring of exposed parents and that paternal exposure is most relevant for offspring effects. / Thesis / Master of Science (MSc) / Parental exposure to the environmentally-relevant pharmaceuticals carbamazepine or gemfibrozil led to male-biased sex ratios in adult offspring of zebrafish (Danio rerio), a common model organism. Intelligent ECG Acquisition and Processing System for Improved Sudden Cardiac Arrest (SCA) Prediction

The survival rate for a suddent cardiac arrest (SCA) is incredibly low, with less than one in ten surviving; most SCAs occur outside of a hospital setting. There is a need to develop an effective and efficient system that can sense, communicate and remediate potential SCA situations on a near real-time basis. This research presents a novel Zeolite-PDMS-based optically unobtrusive flexible dry electrodes for biosignal acquisition from various subjects while at rest and in motion. Two zeolite crystals (4A and 13X) are used to fabricate the electrodes. Three different sizes and two different filler concentrations are compared to identify the better performing electrode suited for electrocardiogram (ECG) data acquisition. A low-power, low-noise amplifier with chopper modulation is designed and implemented using the standard 180nm CMOS process. A commercial off-the-shelf (COTS) based wireless system is designed for transmitting ECG signals. Further, this dissertation provides a framework for Machine Learning Classification algorithms on large, open-source Arrhythmia and SCA datasets. Supervised models with features as the input data and deep learning models with raw ECG as input are compared using different methods. The machine learning tool classifies the datasets within a few minutes, saving time and effort for the physicians. The experimental results show promising progress towards advancing the development of a wireless ECG recording system combined with efficient machine learning models that can positively impact SCA outcomes.
