A regulatory role for N-acylethanolamine metabolism in Arabidopsis thaliana seeds and seedlings.

N-Acylethanolamines (NAEs) are bioactive acylamides that are present in a wide range of organisms. Because NAE levels in seeds decline during imbibition similar to ABA, a physiological role was predicted for these metabolites in Arabidopsis thaliana seed germination and seedling development. There is also a corresponding increase of AtFAAH (fatty acid amide hydrolase), transcript levels and activity, which metabolizes NAE to ethanolamine and free fatty acids. Based on whole genome microarray studies it was determined that a number of up-regulated genes that were responsive to NAE were also ABA responsive. NAE induced gene expression in these ABA responsive genes without elevating endogenous levels of ABA. It was also determined that many of these NAE/ABA responsive genes were associated with an ABA induced secondary growth arrest, including ABI3. ABI3 is a transcription factor that regulates the transition from embryo to seedling growth, the analysis of transcript levels in NAE treated seedlings revealed a dose dependent, inverse relationship between ABI3 transcript levels and growth, high ABI3 transcript levels were associated with growth inhibition. Similar to ABA, NAE negatively regulated seedling growth within a narrow window of early seedling establishment. When seedlings are exposed to NAE or ABA within the window of sensitivity, the induction of genes normally associated with the ungerminated desiccation tolerant state resumed. The NAE tolerant FAAH overexpressor and the NAE sensitive FAAH knockout both had a NAE/ABA sensitive window similar to the wild type A. thaliana. The abi3-1 ABA insensitive mutant does not undergo growth arrest upon exposure to ABA, but NAE did induce growth arrest when treated within the sensitivity window. This evidence showed that although NAE functions within an ABA dependent pathway, it also functions in an ABA independent signaling pathway. The FAAH overexpressor is tolerant to NAE through its ability to quickly metabolize NAE from the growth media, yet it is hypersensitive to ABA. The FAAH overexpressor also displayed hypersensitivity to GA, which improved its delayed germination in non-stratified seed, while the FAAH knock out showed GA insensitivity. Overall, these results showed that NAE functions as a negative regulator of germinating seed and seedling growth in ABA dependent and independent signaling pathways, and that altered NAE metabolism may interfere with ABA/GA perception in germinating seed.

Considerando o grande potencial das enzimas como biocatalisadores e apesar de já existerem no mercado algumas aplicações definidas, existe necessidade de estudo e aprimoramento dos processos enzimáticos nesta área, principalmente para estabelecer os processos em escala industrial. Neste trabalho foi estudada a síntese de quatro ésteres de poliglicóis, com propriedade surfactante e antiespumante, utilizando lipases e esterases como biocatalisadores. Foram avaliadas nove enzimas comerciais de diferentes fornecedores e selecionado três; as quais se apresentaram mais adequadas para os ésteres a serem obtidos e nas condições de processo que possibilitem a produção dos mesmos em escala industrial. A seleção das enzimas e condições reacionais foram baseadas nos resultados obtidos das 227 reações realizadas em laboratório e 2 reações em escala piloto. Três ésteres foram obtidos com sucesso por biocatálise, sendo dois com propriedades de surfactante e um com propriedades de antiespumante, igual ou superior aos produtos obtidos por processo químico. Todos os produtos obtidos por biocatálise apresentaram coloração mais clara, indicando menor oxidação, quando comparados com os produtos obtidos pelo processo químico, no qual é utilizado catalisador ácido e altas temperaturas. Nos estudos realizados foi possível comprovar a seletividade da esterase avaliada como biocatalisador, ao alterar um dos componentes do substrato. As esterificações foram realizadas em diferentes temperaturas, concentrações de enzimas e tempos de reação, visando obter condições de processo que permitam a produção dos ésteres em escala industrial. O acompanhamento das reações foi realizado através de análise titulométrica e espectrometria no infravermelho. A identidade dos ésteres obtidos por biocatálise frente aos ésteres obtidos por processo químico, foi realizada através das técnicas de espectrometria no infravermelho, cromatografia em camada delgada e cromatografia líquida de alta eficiência. A faixa de temperatura ideal para a obtenção dos ésteres estudados foi de 70 ¿ 75º C, com tempo de reação entre 8 e 10 horas e porcentagem de esterificação de até 95%. Os resultados obtidos comprovam que lipases e esterases podem ser utilizadas, na síntese de ésteres de poliglicóis, em substituição aos catalisadores utilizados nos processos químicos, permitindo a redução da temperatura e pressão do processo. Com isso, aumenta a segurança do processo e reduz os impactos ambientais, pela redução do consumo de energia e uso de catalisadores menos agressivos ao meio ambiente. Salientamos que as enzimas são produzidas com insumos renováveis e processos que causam mínimos impactos ao meio ambiente / Abstract: The ester biocatalyst synthesis is becoming technical and economically viable, based on the associated benefits by the use of these catalyst, the constant investiments in enzymes development with high yield, stability, enzymatic activity and new immobilized enzymes that allow multiple uses of the same material. Considering the great potential of enzymes as biocatalysts and although there is already in the market some defined applications, there is still the need to study and improve the enzymatic processes in this area, mainly to establish processes on industrial scale. In this paper, the synthesis of four polyglycol esters with defoamer and surfactant properties, using lipases and esterases as biocalyst, were studied. Nine commercial enzymes coming from different suppliers were evaluated and three were selected; which showed to be more adequate for the esters to be obtained and for the conditions of the process to allow their use on industrial scale. The selection of enzymes and the reacting conditions were based on the results obtained from the 227 reactions runned in the laboratory and 2 reactions in pilot scale. Three esters were successfully obtained by biocatalysis, two with surfactant properties and one with defoamer properties, with the same or better performance than the products obtained by the chemical process. All products obtained by biocatalyst process presented light color, indicating less oxidation, when compared with products obtained by chemical process, where an acid catalyst and high temperatures are used. Esterase biocatalyst selectivity was demonstrated when one component of the substrate was changed. Esterification was carried out in different temperatures, enzymes concentrations, and reaction times, in order to obtain adequate process conditions that allowed the production of esters on industrial scale. Reaction tracking was done using titration analysis and infrared spectrometry. The identity of esters obtained by biocatalysis in relation to esters obtained by chemical process was established through infrared spectrometry techniques, thin layer chromatography, and high performance liquid chromatography. The ideal temperature range to obtain the esters studied was 70 ¿ 75º C, with a reaction time between 8 to 10 hours and esterification percentage up to 95%. The results obtained prove that lipases and esterases can be used in the synthesis of polyglycol esters in substitution to catalysts used in chemical processes, allowing temperature and pressure reduction in the process. Thus, the process safety increases and environmental impacts are reduced, by the reduction of energy consumption and the use of less aggressive catalyst to the environment. A mais recente hipótese para explicar tal fato situa os adipócitos e suas funções autócrina, parácrina e endócrina no centro do cenário, através da relação das adipocinas, por ele secretadas, com a obesidade e o câncer de mama. Objetivo: Artigo 1- Comparar o padrão de expressão imunoistoquímica da adiponectina (APN) e dos seus receptores tipos 1 e 2 (adipoR1/R2) no carcinoma invasor (CDI), carcinoma ductal in situ (CDIS) e lesões benignas da mama (BE) e correlacioná-los com parâmetros clínicos e histológicos. Artigo 2- Avaliar a expressão protéica da FABP4 nos tecidos epiteliais e adiposos mamário de portadoras de CDI, CDIS e lesões benignas da mama. Material e Métodos: Foram incluídos os blocos de parafina de 223 mulheres sendo 69 com CDI, 73 com CDIS e 81 com biópsias negativas para câncer de mama, tratadas no CAISM/UNICAMP de janeiro de 2008 a dezembro de 2011, e preparadas lâminas de Tissue Microarray (TMA). A expressão de APN e Adipo R1/R2 foi avaliada no tecido tumoral nos casos CDI e CDIS e no tecido epitelial e nos casos benignos. A expressão de FABP4 foi avaliada no tecido tumoral, na gordura peritumoral (GP) e na gordura mamária distante (GD) nos casos de CDI e CDIS, e no tecido epitelial e gorduras mamários nos casos benignos. Para avaliar uma possível relação entre a expressão dos marcadores entre si e com parâmetros antropométricos, clínicos e histopatológicos, foram utilizados os testes qui-quadrado ou exato de Fisher, Mann-Whitney, Kruskal-Wallis e correlação de Spearman. As determinações foram calculadas considerando o valor de ?=0,05 (p<0,05). Resultados: Artigo 1 - A APN mostrou-se expressa em 65% dos CDI, 48% dos CDIS e 12% das BE e AdipoR1 em 98%, 94% e 71%, respectivamente. Todos os casos de CDI e CDIS expressaram AdipoR2 contra 81% de BE. Nos CDI e CDIS observou-se associação entre maior expressão de APN e tumores RE negativo. No CDIS esta associação foi também observada com RP negativo. Artigo 2 - Observou-se expressão protéica da FABP4 no tecido epitelial em 90% dos CDI, 40% dos CDIS e 28% em BE. Considerando-se a GP e GD esta expressão foi maior nas BE que nos CDI, diferenças consideradas significativas. Nas pacientes com CDI a expressão da FABP4 foi maior quando o diagnóstico ocorreu até 50 anos de idade. A totalidade dos casos expressou moderada a intensamente este marcador no tecido gorduroso periepitelial e distante. Conclusões: As diferenças de expressões protéicas da adiponectina e dos seus receptores AdipoR1/R2 observadas em diferentes diagnósticos mamários sugerem sua participação no complexo mecanismo etiológico destas diferentes condições. Os resultados deste estudo indicam, ainda, que existe uma correlação direta entre expressão protéica da FABP4, câncer de mama e obesidade / Abstract: Introduction: Obesity has been shown to be responsible for a 30 to 50% increase in new breast cancer cases, in particular in the postmenopausal period. The most recent hypothesis that explains this fact places adipocytes and its autocrine, paracrine and endocrine functions at center stage, linking adipokines secreted by adipocytes to obesity and breast cancer. Objective: Article 1- to compare immunohistochemistry expression pattern of adiponectin (APN) and its receptors types 1 and 2 (adipoR1/R2) in invasive carcinoma (IDC), ductal carcinoma in situ (CDIS) and benign breast lesions (BE), correlated with clinical and histological parameters. Article 2- To assess FABP4 protein expression in epithelial and adipose breast tissue in women diagnosed with IDC, DCIS and benign breast lesions. Material and Methods: Paraffin-embedded blocks from 223 women were included. Of the total number of women, 69 had IDC CDI, 73 had CDIS and 81 had biopsies negative for breast cancer. The patients have been treated at CAISM/Unicamp from January 2008 to December 2011 and Tissue Microarray (TMA) slides were constructed. Expression of APN and Adipo R1/R2 was assessed in tumor tissue in cases of IDC and DCIS and in epithelial tissue in benign cases. FABP4 expression was evaluated in tumor tissue, peritumoral fat tissue (PF) and distant fat breast tissue (DF) in cases of IDC and DCIS and in the epithelial tissue and breast fat tissue in benign cases. To assess a possible relationship between marker expression and anthropometric, clinical and histopathological parameters, the chi-square test or Fisher's exact test, Mann-Whitney test, Kruskal-Wallis test and Spearman's correlation were used. Determinations were calculated, considering a value ?=0.05 (p<0.05) as significant. Results: Article 1 - APN was shown to be expressed in 65% of IDC, 48% of DCIS and 12% of BE and AdipoR1 in 98%, 94% and 71%, respectively. All IDC and DCIS cases expressed AdipoR2 versus 81% of BE. In IDC and DCIS, an association between a higher level of APN expression and ER-negative tumors was observed. In DCIS, this association was also observed with PR-negative tumors. Article 2 - FABP4 protein expression was observed in epithelial tissue in 90% of CDI, 40% of DCIS and 28% of BE. Considering PF and DF, FABP4 expression had a higher level in BE than in IDC, a difference that was considered significant. In patients with IDC, FABP4 expression was higher when diagnosis was made in patients aged up to 50 years. In all cases, this marker was moderately to intensely expressed in the peri-epithelial and distant fat tissue. Conclusions: Discrepancies in protein expression of adiponectin and its receptors AdipoR1/R2 observed in different breast diagnoses suggest its participation in the complex etiologic mechanism of these different conditions. Produção de gases, síntese microbiana pelo radiofósforo e digestibilidade do babaçu e mofumbo em dietas de ovinos / Gas production, microbial synthesis by radio phosphorus and digestibility of Babassu and Mofumbo in sheep diets

Quando a escassez de alimentos em pastagens naturais compromete a alimentação animal, os pequenos ruminantes podem incorporar em suas dietas as folhas de outras plantas, como árvores e arbustos, muitas delas ricas em metabólitos secundários como taninos e que ainda carecem de estudos acerca de seus efeitos na produtividade animal. Com o objetivo de verificar a possibilidade de utilização das folhas de Orbignya phalerata (Babaçu) e Combretum leprosum (Mofumbo) na alimentação e avaliar o efeito da inclusão na produção de ovinos, dois estudos foram conduzidos no Laboratório de Nutrição Animal do Centro de Energia Nuclear na Agricultura da Universidade de São Paulo, Piracicaba (LANA/CENA-USP). Um primeiro estudo avaliou as variáveis de desempenho, parâmetros bioquímicos e hematológicos, estimativas de síntese de proteína microbiana, digestibilidade aparente dos nutrientes e produção entérica de metano (CH4) dos animais. O segundo estudo avaliou as características de carcaça, coloração e perfil de ácidos graxos da carne dos ovinos machos utilizados no primeiro estudo. Os tratamentos experimentais testados foram dietas, com relação concentrado:volumoso de 50:50, elaboradas com base na utilização das folhas das plantas como substituição de 30 % do feno de Cynodon dactylon (Tifton-85), resultando em três tratamentos: Controle (sem substituição do feno), Babaçu e Mofumbo. No primeiro estudo, foram utilizados 24 ovinos Santa Inês, em um delineamento experimental casualizado com oito repetições para cada tratamento e 48 dias de período experimental avaliando o desempenho dos ovinos. Também nesse período, um ensaio in vitro de síntese de proteína microbiana foi realizado utilizando o radiofósforo em cinco inóculos de cada tratamento. Após o experimento de desempenho, durante nove dias, seis animais de cada tratamento foram alocados em gaiolas metabólicas para a determinação da digestibilidade aparente dos nutrientes, a síntese de proteína microbiana e o balanço de nitrogênio. Simultaneamente foi quantificada a produção de CH4 entérico in vivo. Os ovinos do tratamento Controle apresentaram maior (P < 0,05) digestibilidade aparente da fibra em detergente ácido. A produção entérica de CH4 dos ovinos do tratamento Mofumbo não diferiu do tratamento Controle e foi menor (P < 0,05) que a dos ovinos do tratamento Babaçu. No segundo estudo, os cinco animais machos de cada tratamento foram encaminhados para abate e procedeu-se a avaliação da carcaça, dos componentes não carcaça e a coloração e perfil de ácidos graxos na carne. Não foram observadas diferenças (P > 0,10) nos resultados da avaliação da carcaça, dos componentes não carcaça, coloração e total de ácidos graxos. O tratamento Mofumbo apresentou maiores (P < 0,10) valores da estimativa de atividade da enzima ?9-dessaturase C16. Efeito linear significativo (P = 0,01) foi observado quando se analisou a estimativa de atividade da enzima ?9-dessaturase C18 e os teores de TC nas dietas. A inclusão das folhas do Babaçu e Mofumbo não apresentou efeitos deletérios à saúde dos animais, não comprometeu o desempenho, potencial de produção e qualidade da carne dos ovinos, tendo o Mofumbo inclusive apresentado potencial mitigador de CH4, indicando que podem ser utilizadas como ingredientes na composição de dietas para ovinos / When food shortages in natural pastures is committed to animal nutrition, small ruminants can incorporate into their diets the leaves of other plants, such as trees and shrubs, many of them rich in secondary metabolites such as tannins and which still lack of studies about its effect on animal productivity. In order to verify the possibility of using leaves of Orbignya phalerata (Babassu) and Combretum leprosum (Mofumbo) in feed and to evaluate the effect of their inclusion in the sheep production system, two studies were conducted at the Animal Nutrition Laboratory of Centro de Energia Nuclear na Agricultura, Universidade de São Paulo, Piracicaba (LANA/CENA-USP). The first study evaluated the performance variables, biochemical and hematological parameters and also determined the microbial protein synthesis, nutrient apparent digestibility and enteric production of methane (CH4). The second study assessed the carcass characteristics, fatty acid profile and meat color of male sheep used in the first study. The experimental treatments were diets with forages to concentrate rate of 50:50, drawn up on the basis of using the leaves of the experimental plants replacing 30% of the Cynodon dactylon (Tifton-85) hay, resulting in three treatments: Control (no hay replacement), Babassu and Mofumbo. In the first study, there were used 24 Santa Inês sheep, in a randomized experimental design with eight repetitions for each treatment and 48 days of trial period. Also during this period, an in vitro microbial protein synthesis was performed using the radio phosphorus using five different inocula of each studied treatment. After this period, for nine days, six animals from each treatment were allocated in metabolic cages for determining the nutrient apparent digestibility, microbial protein synthesis and nitrogen balance. Simultaneously it was quantified the enteric CH4 production in vivo. The Control group showed greater (P < 0.05) apparent digestibility of acid detergent fiber. Enteric CH4 production of sheep fed with Mofumbo leaves did not differ from the Control group but was lower (P < 0.05) than the sheep fed Babassu leaves. In the second study, the five male animals of each treatment were sent to slaughter and to precede the assessment of carcass, not carcass components and color and fatty acid profile in the meat. The results of the assessment of carcass, not carcass components, color and overall fatty acids showed no differences between the treatments. Mofumbo treated sheep showed greater (P < 0.10) values of the ?9-desaturase C16 enzyme activity. Significant linear effect (P = 0.01) was observed when it analyzed the enzyme activity estimation ?9-desaturase C18 and TC levels in the diets. The inclusion of Babassu and Mofumbo leaves shown no negative effects on animal health, did not compromise the performance, production potencial or meat quality of the animals, having Mofumbo also presented CH4 mitigating potencial, indicating that those plants can be used as ingredients in the composition of sheep diets

Utilização do grão de soja cru integral na dieta de bovinos de corte confinados / Whole raw soybean grain in the beef cattle diet in feedlot

Objetivou-se avaliar o uso do grão de soja cru integral na dieta de bovinos de corte sobre o consumo e digestibilidade da matéria seca e nutrientes, fermentação ruminal, desempenho produtivo, síntese de proteína microbiana, características de carcaça qualidade da carne, perfil de ácidos graxos da carne, concentrações de parâmetros sanguíneos, excreção e composição do grão de soja presente nas fezes. Foram realizados dois experimentos, sendo que no primeiro, foram utilizados 12 bovinos Nelore, castrados, canulados no rúmen, agrupados em três quadrados latinos 4x4. No segundo experimento foram utilizados 52 bovinos Nelore, inteiros, confinados por 84 dias, em delineamento inteiramente casualizado. A dieta empregada foi a mesma, para ambos os experimentos, constituída de quatro rações, com relação volumoso/concentrado de 60/40, a qual foi composta de milho moído, farelo de soja, grão de soja cru integral, núcleo e silagem de milho. As dietas foram: G0: dieta controle sem a inclusão do grão de soja; G8, G16 e G24, com 8, 16 e 24%, respectivamente de grão de soja cru integral na ração, na matéria seca. No Experimento 1, amostras de fezes e sobras foram coletadas nos 11o, 12o 13o dia experimento, amostras de sangue e urina foram colhidas e avaliado o peso vivo. A digestibilidade foi determinada por meio de indicador interno FDAi. No Experimento 2, a cada 28 dias, os animais foram pesados e amostras de sangue foram coletadas. Os animais foram abatidos ao 85o dia e foi avaliado o peso do fígado (PFi) e peso de carcaça quente (PCQ). Vinte e quatro horas após o abate, foram mensurados o pH, e rendimento de carcaça (RC). Na desossa foi avaliada a área de olho de lombo (AOL) e a espessura de gordura subcutânea (EGS) no músculo Longissimus. Foram retiradas amostras do músculo Longissimus, para determinação do perfil de ácidos graxos na carne (PAG) extrato etéreo (EE), maciez objetiva e sensorial da carne maturada por 14 dias. Houve redução no consumo de matéria seca, matéria orgânica, de carboidratos totais e carboidratos não fibrosos nos animais alimentados com a ração G24. Foi observado efeito linear crescente no consumo de EE e efeito quadrático no consumo de proteína bruta. Houve diferença na digestibilidade aparente total do EE e carboidratos totais com a adição do grão de soja na dieta. Na fermentação ruminal, houve queda linear no valor de pH ruminal e a concentração de nitrogênio amoniacal ruminal foi maior para os animais que receberam a dieta controle em relação às dietas com grão de soja, havendo um maior valor de ácidos graxos de cadeia curta para o grupo controle. O teor de colesterol sanguíneo aumentou linearmente com a inclusão do grão e no primeiro experimento também houve aumento do colesterol HDL. Para desempenho, atributos de carcaça e medidas de ultrassonografia não foi observado efeito de acordo com a dieta. A utilização de diferentes níveis de grão de soja nas rações alterou o consumo, digestibilidade, metabolismo e características de qualidade da carne. / The objective of this study was to evaluate the use of whole raw soybean grain in the diet of beef cattle on consumption and digestibility of dry matter and nutrients, ruminal fermentation, growth performance, microbial protein synthesis, carcass traits, meat quality, profile of fatty acids in meat, concentrations of blood parameters, excretion and composition of soybean in the faeces. Two experiments were conducted and in the first one were used 12 Nellore, castrated, cannulated in the rumen, grouped in three 4x4 Latin squares. In the second experiment were used 52 Nellore, not castrated, confined for 84 days in a completely randomized delineation. The diet used was the same for both experiments, consisting of four rations, with the forage / concentrate ratio of 60/40, which was composed of ground corn, soybean meal, whole raw soybean grain, nucleos and corn silage. The diets were: G0: control diet without the inclusion of soybean, G8, G16 and G24, with 8, 16 and 24% respectively of whole raw soybean grain in ration, in the dry matter. In the first experiment, fecal samples and remains were collected on the 11th, 12th and 13th day of experiment, blood and urine samples were collected and the body weight was analyzed. The digestibility was determined by an internal indicator iADF. In Experiment 2, every 28 days, the animals were weighed and blood samples were collected. The animals were slaughtered at 85 days and were weighed the liver (PFi) and hot carcass weight (HCW). Twenty-four hours after the slaughter, the pH was measured, likewise the carcass yield (CY). Deboning was evaluated in rib eye area (REA) and subcutaneous fat thickness (SFT) in the Longissimus muscle. Samples were taken from the Longissimus for determining the fatty acid profile in meat (PAG) in ether extract (EE), objective tenderness and sensorial in aged beef for 14 days. There was a reduction in the dry matter intake, organic matter, total carbohydrates and non-fibrous carbohydrates in animals fed with feed with ration G24. It was observed an increasing linear effect on the EE intake and a quadratic effect on crude protein. Also difference in the apparent digestibility of total carbohydrates and total EE. On ruminal fermentation, there was a linear decline in ruminal pH and ruminal ammonia concentration was higher for animals that received the control diet compared to diets with soybean, with a value greater short-chain fatty acids for the group control. The level of blood cholesterol increased linearly with the addition of grain and in the first experiment there was also increase of HDL cholesterol. For performance, attributes of carcass and measures of ultrasound effect were not observed according to the diet. There was a quadratic effect for shear force (FC), with higher value for the meat of animals that received 8% of the soybean. There was a slight improvement in lipid profile for animals receiving the grain and the inclusion in the sensory analysis of the grain at 24% of diet DM improved the texture of the meat. The use of different levels of whole raw soybean grain in the beef cattle diet changed the intake, digestibility, metabolism and meat quality characteristics.

Effect of maternal diet on mother’s own milk pH and preterm infant intestinal inflammation

BACKGROUND: Quantitative evidence of direct links between dietary intake during pregnancy and maternal and infant outcomes in the preterm population is lacking in the literature. However, studies are starting to investigate relationships between them, and data shows that many of the adverse effects of poor maternal diet are linked to inflammatory response and dysbiosis of the microbiome in both the mother and her offspring. OBJECTIVE: Our objective with this study was to analyze the relationships between sociodemographic factors, maternal diet, pH of breast milk, and infant inflammation. We aimed to assess the dietary pattern of our population of mothers who delivered preterm in comparison to other populations, as well as explore the possibility of using pH of mother’s own milk in future research. METHODS: We reviewed the Electronic Medical Records (EMRs) of participants to gather clinical and demographic characteristics (infants n = 53; mothers n = 45). Maternal participants also completed the Dietary Screener Questionnaire (DSQ). Spearman’s rank correlation and raw unadjusted linear regression analyses were used to investigate relationships between maternal diet and characteristics, mother’s own milk pH, and infant urinary intestinal fatty acid binding protein (I-FABP). Kruskal Wallis analysis was used to analyze between group differences of maternal comorbidities. RESULTS: None of our maternal participants met the guidelines for dietary recommendations by the United States Department of Agriculture (USDA) for pregnant women. This follows the trend in national data for women who are pregnant. The greatest number of women met the recommendation for fiber intake (n = 12). Maternal intake of fiber and whole grains was negatively correlated with pH of mother’s own milk (p <0.5). We did not find any significant correlations between maternal characteristics and maternal diet or pH of mother’s own milk. However, meeting the guidelines for added sugars differed by race (p = 0.03). We found no statistically significant correlations between urinary I-FABP and pH of mother’s own milk or maternal dietary intake. Urinary I-FABP values differed by infant sex (p = 0.03) and infant feeding status (> 50% formula or donor milk vs. < 50% formula or donor milk, p = 0.03). Analysis by groups showed statistically significant differences (p = 0.04). Preeclamptic participants had a higher intake of whole grains (0.97 oz) than women without preeclampsia (0.69 oz). Mothers with gestational diabetes had a lower intake of sugar (14.4 tsp) compared to women without diabetes (17.9 tsp) (p = 0.01). We found no other statistically significant results between groups for maternal diet, milk pH, or urinary I-FABP. CONCLUSION: Our findings suggest that additional research on mother’s own milk pH may be warranted, and that continued education on the importance of a healthy diet and its benefits during pregnancy is needed. Areas of planned future research include fat intake calculations and inflammatory measures of the maternal dietary data.

Activity and mRNA abundance of enzymes for fatty acid synthesis and desaturation in mammary cell cultures

The effect of exogenous unsaturated fatty acids on cellular fatty acid biosynthesis in mammary cells was examined. Under normal situations, even though the diet of a dairy cow contains considerable amounts of unsaturated fatty acids, viz. oleic acid (18:1) and linoleic acid (18:2), the major 18-carbon fatty acid that enters the circulation post-ruminally for delivery to the mammary gland is saturated fatty acid, viz. stearic acid (18:0). This is due to extensive ruminal biohydrogenation of unsaturated fatty acids. Studies have indicated that saturated fatty acids such as 18:0 are enhancers and that certain unsaturated fatty acids are inhibitors of de novo fatty acid synthesis in tissues such as the liver and adipose tissue. The present study investigated the effect of cis and trans isomers of 18:1 and 18:2 on de novo fatty acid synthesis and desaturation in mouse and bovine mammary epithelial cell cultures, and compared it with the effect caused by 18:0. In the first experiment 12.5, 25, 50 or 100 micromoles stearic acid (SA), oleic acid (OA), elaidic acid (EA), trans-vaccenic acid (TVA), linoleic acid (LA) or conjugated linoleic acid (CLA) were supplemented in the media of mouse mammary epithelial (MME) cells that were grown to confluence in Dulbecco's modified Eagle's medium (DMEM). As indicated by cellular palmitic acid (16:0) content and fatty acid synthetase (FAS) activity, when compared with SA all unsaturated fatty acid treatments inhibited de novo fatty acid synthesis in MME cells. In addition, OA at all concentrations and LA and CLA at 50 and 100 micromoles inhibited cellular stearoyl-CoA desaturase (SCD) activity and mRNA abundance. However, EA and TVA, when compared with SA, enhanced SCD activity and mRNA abundance at 12.5 and 25 micromoles. In the second experiment 25, 50 or 100 micromoles SA, OA, TVA, LA or CLA were supplemented in the media of bovine mammary epithelial cells that were grown to confluence in DMEM. As indicated by cellular 16:0 content, acetyl-CoA carboxylase (ACC) activity and FAS activity, treatment with the unsaturated fatty acids inhibited de novo fatty acid synthesis at all concentrations, when compared with SA. Unsaturated fatty acid treatments also reduced the abundance of ACC and FAS mRNA in the cells. When compared with SA at all treatment-concentrations, OA and LA inhibited whereas TVA and CLA enhanced cellular SCD activity and mRNA abundance in the bovine cells. In both cell types, CLA and TVA appeared to be the most potent inhibitors of saturated fatty acid biosynthesis. / Ph. D.

Quantitation of Formaldehyde Resulting from Oxidative Deterioration of Unsaturated Fatty Acids

Derivatives of 3-methyl-1-2-benzothiazolinone hydrazone were formed from the oxidation products of three oxidized unsaturated fatty acids. Aeration with oxygen and heat (65C) was used as a means of accelerating deterioration of lipid samples. Formaldehyde was recovered from oxidized pure standards of oleic, linoleic and linolenic acid methyl esters , and identified using gas chromatography. Formaldehyde was also recovered from oxidized lipid extracts of mechanic ally deboned turkey meat and turkey cluster fat. Gas chromatography was used to measure concentrations of unsaturated fatty acids during the oxidation of extracted lipids. Linoleic and linolenic fatty acids significantly decreased (p < .005) in oxidized lipid extracts from turkey cluster fat stored frozen for six months. Measurement of the amounts of formaldehyde which accumulates as a result of the oxidation of unsaturated fatty acids, may be the most reliable method of assessing the extent of oxidative deterioration in foods.
