Factors causing feed intake depression in lambs infected by gastrointestinal parasitesDynes, Robyn A. January 1993 (has links)
A reduction in voluntary feed intake is a major factor in the lost productivity of grazing lambs infected by gastrointestinal parasites yet the mechanisms involved are poorly understood. Potential pathways involved in parasite-induced feed intake depression were investigated in lambs with minimal previous exposure to parasites and artificially infected by the small intestinal parasite Trichostrongylus colubriformis. Six in vivo experiments were conducted on lambs housed in individual pens or metabolism crates with similar feeding and experimental procedures. In Experiment 1 (Chapter 4) the effect of T. colubriformis infection on short term feed intake in lambs and of some pharmacological agents on feed intake depression were investigated. Prior to and for the duration of infection, lambs were fed once per day and feed intake recorded at regular intervals over the day (8 h). Following the onset of feed intake depression in the infected group (9 weeks after commencing dosing), all animals were treated with an analgesic (codeine phosphate per os), an anti-inflammatory agent (indomethacin per os), a CCK antagonist (L364-718 by subcutaneous injection) or saline (control) in a replicated Latin square design (n = 8). Although the pattern of feed consumption was similar in infected and non-infected lambs, average daily intake was reduced 32 % and short term intake (recorded at 10 minute intervals for the first hour of feeding, 15 minute intervals for the second hour and hourly for the next 6 hours of feeding) reduced 40 % by infection. This identified the key component by which intake was depressed and enabled the use of a short term intake model and short duration of action compounds to identify the pathways involved in intake depression in this sequence of experiments. None of the pharmacological treatments increased intake in the infected group. These results suggest a reduction in the rate of consumption due to reduced hunger signals, rather than change of meal eating patterns, is the major cause of feed intake depression. Specific conclusions about the pathways investigated using the pharmacological agents could not be obtained. Experiment 2 (Chapter 5) was designed to investigate the roles of pain and osmolality on feed intake depression. Digesta samples collected prior to and during parasite infection and before and after feeding had similar osmolalities (240-260 mosmol/l) which indicated that feeding or infection had no effect on osmolality of digesta. Following the onset of feed intake depression in infected animals, all animals were treated in a Latin square design (n = 4) with no treatment, saline, local anaesthetic (xylocaine) or analgesic (codeine phosphate) solution 15 minutes before feeding, by slow injection into the duodenum. There was no effect of these treatments on food intake. In the second part of the experiment, hyperosmotic solutions (mannitol and NaCI) markedly depressed short term intake in non-infected animals, suggesting a role for osmoreceptors in intake regulation. However these effects were not blocked by local anaesthetic so the depressed intake may have resulted from generalised malaise rather than from specific osmoreceptor effects. In Experiment 3 (Chapter 6) the role of peripheral CCK on intake depression was examined by a dose-response study utilising the CCK antagonist, loxiglumide. Intravenous injection of 5, 10 or 20 mg/kg LW of loxiglumide to infected lambs 10-15 minutes before feeding (n = 6) had no effect on feed intake at any of the dose levels. In experiment 4 (Chapter 7) loxiglumide was infused intravenously for 10 minutes (30 mg/kg/h) before feeding and for the first 2 h (10 mg/kg/h) after feed was offered to minimise any effect of the rate of clearance of loxiglumide on the lack of feed intake response. As well, the rate of marker disappearance from the abomasum was recorded in both infected and non-infected animals. Continuous infusion of loxiglumide did not attenuate parasite induced intake depression nor did it have any effect on abomasal emptying. Abomasal volume was reduced by infection (66.3 vs 162 ml) as was the fractional outflow rate (2.2 vs 2.8 ml/min) but these differences were accounted for by the lower level of feed intake in the infected animals. In Experiment 5 (Chapter 8) brotizolam, a benzodiazepine appetite stimulant, thought to act on the hypothalamus, was administered in a dose-response study to infected and non-infected animals (n = 4) immediately prior to feeding or following termination of the first meal (45 minutes after feeding) and the feed intake response recorded. Brotizolam elevated both the short term (0-0.75 h), daily (22 h) intake and all time intervals in the first 5 h after feeding in infected and non-infected animals when administered after the first meal but when administered prior to feeding elevated intake only over the first 6 h of feeding. In both cases the magnitude of the response was greater in infected animals than in non-infected animals. Brotizolam appeared to increase the rate of eating without having a major impact on meal eating patterns when administered before feeding. Where administration was after the first meal, the effect was due to an "extra" meal being consumed. These findings showed that infected animals can respond to central stimulators of intake although the mechanism of the response is not known. Opioids were implicated in intake depression as the rate of intake rather than meal patterns appeared to be the major parameter depressed under parasitism. This was examined in experiment 6 (Chapter 9) where animals (n = 6) were fasted for 26 h or not fasted, then treated with saline (control), brotizolam (intake stimulant) or naloxone (opioid antagonist) immediately prior to feeding. Fasting stimulated feed intake in the short term (100 % increase in 75 min) and over the day (12 % increase) in both infected and non-infected animals. Following fasting, infected animals ate a similar amount of feed to the non-infected, fasted animals and more than the non-infected, non-fasted animals. The signals resulting from a one day fast were sufficient in the short term to override parasite induced mechanisms causing feed intake depression. Naloxone suppressed the intake stimulatory effects of a 26 h fast in both infected and non-infected animals, which supports a role for endogenous opioids as hunger signals. Where animals were not fasted, naloxone reduced intake only in the non-infected animals which suggested endogenous opioid levels may be lower in infected animals than in non-infected animals. In the final experiment (Experiment 7, Chapter 10) the role of central hunger and satiety mechanism were investigated. Infected and non-infected animals (n = 6) were treated with naloxone or saline by intravenous injection, or saline and met-enkephalinamide (an opioid analogue) by intracerebral infusion, or naloxone and the opioid analogue simultaneously to investigate the role of central opioids in feed intake depression. To determine the role of CCK induced satiety signals on feed intake at a central level, loxiglumide and CCK were infused separately and in combination for 30 minutes prior to feeding and for the first 60 minutes of feed on offer, into a lateral cerebral ventricle of the brain of infected and control animals (n = 6). The opioid analogue tended to increase intake in infected animals but the effect was not significant probably because the dose used was too low to elicit a response in sheep. Naloxone depressed intake only in the infected animals, which conflicted with the results of Experiment 4. As a consequence these results were inconclusive because of the single low dose of opioid analogue used and the conflicting naloxone responses. CCK alone depressed intake by 39-52 % only in infected animals and this effect of the 90 minute infusion was evident over the 8 h short term recording period. Loxiglumide attenuated the feed intake depressive effects of CCK in the infected animals to the extent that intake was elevated above control levels. Loxiglumide alone was an intake stimulant in both infected and non-infected animals. Intake was increased over the entire 8 h but mostly in the second hour when intake was increased by 188 % in infected animals and by 16 % in the non-infected animals and resulted in almost continuous eating. These results showed loxiglumide will temporarily block the effect of parasite infection on feed intake in sheep when administered centrally and the fact that it blocked the effects of exogenous CCK on intake indicated that the effect is mediated via CCK receptors. In conclusion GIT parasite infection reduced both short term and daily feed intake apparently by a change in rate of intake rather than any alteration in meal patterns. It was further suggested that anyone of a number of potential peripheral pathways, including changes to osmolality, gut emptying, pain and inflammation of the gut, alone is not involved in anorexia in sofar as the compounds used could block these factors and the results support the idea that intake depression is mediated via a central mechanism. Intake in infected animals responded to a much greater extent when fasting, i.c.v. loxiglumide or brotizolam were employed. Feed intake thus appears to be regulated through the same mechanisms in infected and non-infected animals. The results from compounds affecting the central mechanism suggest central CCK receptors are important in parasite induced anorexia, possibly by changing the onset of satiety or by interacting with endogenous opioids to reduce the rate of feed intake. Secondly reduced endogenous opioids may be causing the reduction in the rate of feed consumption alone or as a result of other interactions. It was concluded that intake in parasitised animals could be increased to that of control animals by employing procedures and compounds thought to act on the hypothalamus.

Somatotropina bovina recombinante (rbST) e grupo racial sobre o desempenho, características de carcaça e qualidade da carne de caprinos em crescimento /Rodrigues, Luciana. January 2006 (has links)
Orientador: Heraldo César Gonçalves / Banca: André Mendes Jorge / Banca: Francisco de Assis Fonseca de Macedo / Resumo: Foram utilizados 43 animais de três grupos raciais (GR), sendo 13 Alpino (8 machos e 5 fêmeas), 9 u Boer + u Alpino (4 machos e 5 fêmeas) e 21 Boer + o Alpino (11 machos e 10 fêmeas), dos quais 21 receberam rbST (4 machos e 3 fêmeas do grupo Alpino (A); 2 machos e 2 fêmeas do u BA e 6 machos e 4 fêmeas do BA) e 22 placebo (4 machos e 2 fêmeas A; 2 machos e 3 fêmeas u BA e 5 machos e 6 fêmeas BA). O hormônio de crescimento utilizado foi a somatotropina recombinante bovina (rbST) e os animais do tratamento 1 receberam o hormônio na quantidade de 0,3 mg/kg de peso vivo, a partir dos 45 dias, ajustada em intervalos de 14 dias. Os animais do tratamento 2 (controle) receberam solução salina na mesma dosagem e intervalo. Não foi observada nenhuma influência da somatotropina bovina recombinante nas características de desempenho avaliadas. Os machos foram superiores às fêmeas no ganho em peso do início da aplicação do hormônio aos 120 dias de idade, assim como no ganho médio diário de peso dos 60 aos 90, 90 aos 120 e do início da aplicação aos 120 dias de idade. O peso aos 90 e 120 dias de idade das fêmeas foram inferiores em relação aos machos. As fêmeas BA apresentaram maiores ganho em peso e ganho médio diário de peso dos 150 aos 180 dias em relação às demais. Não foi observada influência do hormônio de crescimento e do grupo racial para o peso ao cio, idade ao cio e idade para atingir 30 kg de peso vivo. / Abstract: It was used 43 animals of three breed groups, wich 13 Alpine (8 males and 5 females), 9 u Boer + u Alpine (4 males and 5 females) e 21 Boer + o Alpine (11 males and 10 females), wich 21 received rbST (4 males and 3 females of Alpine group (A); 2 males and 2 females of u BA and 6 males and 4 females of BA) and 22 control (4 males and 2 females of A; 2 males and 3 females of u BA and 5 males and 6 females of BA).The growth hormone used was the bovine recombinant somatotropin (rbST) and the animals of treatment 1 received the hormone in the adjusted amount from 0,3 mg/kg of live weight in intervals of 14 days. The animals of treatment 2 (control) had received saline solution in the same dosage and interval. There was no influence of the recombinant bovine somatotropin in the evaluated characteristics of performance. The males had been bigger to the females in the weight gain of the application of the hormone to the 120 days of age, as well as in the daily average weight gain of the 60 to the 90, 90 to the 120 and of the application to the 120 days of age. The weight to the 90 and 120 days of age of the females had been smaler in relation to the males. The females BA had presented bigger weight gain and daily average weight gain of the 150 to the 180 days in relation to others. It was not observed influence of growth hormone and the breed group for weight to oestrus, age to oestrus and age to reach 30 kg live weight. / Mestre

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / The experiment was conducted to evaluate the effects of particle size of the limestone and the use of artificial light on feeding behavior, performance, egg quality and bone of laying hens. A total of 300 Hisex White hens at 18 weeks of age, distributed in a completely randomized design in factorial 5 x 2, with 5 replicates of 6 birds. The factors studied were 5 granulometries obtained by increasing the proportion of coarse limestone (0, 25, 50, 75 and 100%) compared to 2 types of thin and light, with and without artificial light. No significant interaction between factors (size x lighting program) for all parameters evaluated on performance, egg quality and bone. There was also no significant effect on the level of coarse limestone in diets on performance and egg quality, however had an effect on bone quality. Among the light programs, showed that the use of artificial light showed eggs with higher Haugh Units compared to birds that received only natural light. With respect to feeding behavior no significant interaction between factors limestone, light and time. However, significant effects of the interaction (light and time), there was a higher consumption in birds without artificial light in relation to artificial light during the hours of 6am to 9h40min and 12:40 to 17:40 and the interaction effect (limestone and time) , it was observed that at the time of 6am to 9h40min, increased consumption, corresponded to the gradual increase in the ration of coarse limestone. For laying hens in the first laying cycle, created in geographic coordinates that developed in the research, changes in particle size of limestone provided an improvement in bone quality of the birds and the use of artificial light may favor albumen quality as Haugh unit. / O experimento foi realizado com o objetivo de avaliar os efeitos da granulometria do calcÃrio e do uso da luz artificial sobre o comportamento alimentar, desempenho, qualidade do ovo e Ãssea de poedeiras comerciais. Foram utilizadas 300 poedeiras Hisex White com 18 semanas de idade, distribuÃdas em um delineamento inteiramente casualizado, em esquema fatorial 5 x 2, com 5 repetiÃÃes de 6 aves. Os fatores estudados foram 5 granulometrias obtidas pelo aumento da proporÃÃo de calcÃrio grosso (0, 25, 50, 75 e 100%) em relaÃÃo ao fino e 2 tipos de iluminaÃÃo, com e sem luz artificial. NÃo houve interaÃÃo significativa entre os fatores (granulometria x programa de luz) para os parÃmetros avaliados sobre desempenho, qualidade do ovo e Ãssea. TambÃm nÃo houve efeito significativo do nÃvel de calcÃrio grosso na raÃÃo sobre o desempenho e qualidade do ovo, entretanto apresentou efeito sobre a qualidade Ãssea. Entre os programas de luz, observou-se que o uso de luz artificial apresentou ovos com maiores valores de unidade Haugh em relaÃÃo Ãs aves que receberam apenas luz natural. Com relaÃÃo ao comportamento alimentar nÃo houve interaÃÃo significativa entre os fatores calcÃrio, luz e horÃrio. Contudo, houve efeito significativo entre a interaÃÃo (luz e horÃrio), observou-se um maior consumo nas aves sem luz artificial em relaÃÃo aos com luz artificial nos horÃrios de 6h Ãs 9h40min e 12h40min Ãs 17h40min e efeito na interaÃÃo (calcÃrio e horÃrio), observou-se que no horÃrio de 6h Ãs 9h40min, o aumento do consumo, correspondeu ao aumento gradativo do calcÃrio grosso na raÃÃo. Para poedeiras comerciais no primeiro ciclo de postura, criadas nas coordenadas geogrÃficas em que se desenvolveu a pesquisa, as mudanÃas na granulometria do calcÃrio proporcionou uma melhora na qualidade Ãssea das aves e a utilizaÃÃo da luz artificial pode favorecer a qualidade do albÃmen medida em unidade Haugh.

Caracterização do perfil de expressão gênica hepática global associada à eficiência alimentar em bovinos Nelore / Characterization of global hepatic gene expression profile associated with feed efficiency in Nelore cattlePâmela Almeida Alexandre 30 March 2015 (has links)
A seleção de bovinos de corte para eficiência alimentar (EA) é bastante vantajosa não só do ponto de vista produtivo e econômico como também por ajudar a diminuir o impacto ambiental da produção pecuária. Por ser uma característica multifatorial e de mensuração onerosa, objetivou-se com esse trabalho, a partir da expressão gênica global hepática de animais com alta eficiência alimentar (AEA) e baixa eficiência alimentar (BEA), identificar novos genes, funções biológicas e genes reguladores associados a esse fenótipo. Para isso, 98 bovinos Nelore foram avaliados quanto ao desempenho em confinamento, medidas de EA, ultrassonografia de carcaça, bioquímica sérica, biometria, rendimento de carcaça, maciez de carne e avaliação de líquido ruminal. Além disso, 8 animais de AEA e 8 animais de BEA, selecionados pela medida de consumo e ganho residual ao final do confinamento, tiveram dados de perfil transcriptômico hepático analisados por 3 abordagens: expressão gênica diferencial, co-expressão e co-expressão diferencial. Foi observado que os grupos de AEA e BEA apresentaram desempenho igual quanto ao peso inicial e final, ganho de peso, rendimento de carcaça e área de olho de lombo. Por outro lado, os animais de BEA tiveram maior consumo de alimentos e maior deposição de gordura subcutânea e visceral. Além disso, os animais de BEA apresentaram níveis elevados de colesterol e de GGT e a análise de perfil transcriptômico mostrou estar relacionada à resposta imunológica, inflamação e metabolismo de lipídeos. Baseado nesses resultados e em pesquisas em humanos criou-se a hipótese de que os animais de BEA são mais susceptíveis a infecção bacteriana no fígado em resposta à mudança da dieta a pasto para a dieta de confinamento. / The selection of beef cattle to feed efficiency (FE) traits is very important not only from the productive and economic perspective but also for helping to reduce the environmental impact of livestock production. Being a multifactorial and expensive to measurement trait, the aim of this work was to analyze the liver global gene expression profile of animals with high feed efficiency (HFE) and low feed efficiency (LFE), to identify new genes, biological functions and regulatory genes associated to this phenotype. For this purpose, 98 Nellore cattle were evaluated regarding feedlot performance, FE measures, carcass ultrasound, serum biochemistry, biometric measures, carcass evaluation, meat tenderness and evaluation of ruminal fluid. In addition, 8 animals of HFE and 8 animals of LFE, selected by the measure of residual intake and body weight gain at the end of feeding trial, had liver transcriptomic profile data analyzed by three approaches: differential gene expression, co-expression and differential co-expression. It was observed that HFE and LFE groups showed equal performance for initial and final weight, weight gain, carcass yield and rib eye area. On the other hand, the animals of LFE had higher feed intake and increased deposition of subcutaneous and visceral fat. In addition, animals of LFE showed higher levels of cholesterol and GGT and transcriptomic profile analysis showed to be related to immune response, inflammation and lipid metabolism. Based on these results and research in humans we created the hypothesis that the LFE animals are more susceptible to bacterial infection in the liver in response to the change of pasture diet to feedlot diet.

O consumo alimentar residual (CAR) é uma medida de eficiência alimentar independente do crescimento e do peso à maturidade. O melhoramento genético para CAR pode reduzir o custo de alimentação de bovinos, porém uma melhor compreensão dos processos biológicos relacionados ao CAR é necessária. Além disso, associações entre CAR e qualidade de carcaça têm sido pouco investigadas em raças zebuínas. Desta forma, o objetivo com este estudo foi avaliar o metabolismo protéico, a composição corporal, as características de carcaça e a qualidade de carne em bovinos zebuínos com alto e baixo CAR. Adicionalmente, foi testada a hipótese da existência de interações entre CAR e peso vivo ao abate para características de carcaça e composição corporal. Setenta e dois novilhos da raça Nelore (16 a 21 meses de idade, 334±19 kg de peso vivo inicial [PV]) foram mantidos em confinamento e alimentados ad libitum (74,5% NDT; 14,3%PB) por 70 dias. O consumo de matéria seca (CMS) e o ganho médio de peso (GMD) diários foram medidos individualmente. Os 12 novilhos com maior CAR e os 12 com menor CAR foram classificados como grupos de alto e baixo CAR, respectivamente (fase de seleção) e foram alimentados até quando alcançassem pesos vivo ao abate de 460, 490, 520 e 550 kg (fase de terminação). Antes do abate, foi realizada colheita total de urina para a determinação da excreção diária de 3-metil-histidina e da taxa fracional de degradação miofibrilar. A composição química corporal foi estimada pelo método de diluição isotópica utilizando óxido de deutério. A maciez objetiva da carne e a atividade de proteases cálcio-dependentes foram determinadas no músculo Longissimus. Na fase de seleção, novilhos com baixo CAR tiveram menores CMS, conversão alimentar, CAR e ganho de gordura sobre a garupa que novilhos com alto CAR, mas nenhuma diferença foi observada no GMD, no PV final, na gordura subcutânea e na área do Longissimus. Na fase de terminação, nenhuma interação foi observada entre CAR e PV ao abate. Não houve diferenças entre animais mais e menos eficientes quanto ao peso e rendimento de carcaça, gordura renal, pélvica e inguinal, vísceras, área de Longissimus, gordura subcutânea, marmorização, aparas e porção comestível. Novilhos com baixo CAR apresentaram menos gordura sobre o trato gastrintestinal (TGI) que novilhos com alto CAR. Não foram observadas diferenças quanto ao índice de fragmentação miofibrilar, força de cisalhamento e atividade do sistema calpaína. As taxas fracionais de degradação, síntese e acréscimo protéico foram similares entre os grupos de CAR. Novilhos Nelore com baixo CAR depositaram menos gordura subcutânea na carcaça em pesos vivos entre 340 e 460 kg. Em pesos mais elevados (460-550 kg), as características de carcaça e a composição corporal não foram influenciadas pelo CAR, mas indivíduos menos eficientes apresentaram maior massa de gordura visceral. A seleção de bovinos zebuínos para baixo CAR pode diminuir a ingestão de alimentos e melhorar sua eficiência alimentar, sem comprometer a qualidade da carne. / Residual feed intake (RFI) is a feed efficiency trait that is independent of growth rate and mature weight. Genetic improvement in RFI may reduce the costs of feeding cattle, however a better understanding of biological processes underlying variation in RFI is necessary. Moreover, associations between RFI and carcass quality have been poorly investigated in Zebu breeds. Therefore, this study aimed to evaluate protein metabolism, body composition, carcass traits and meat quality in high- and low-RFI Zebu cattle. In addition, the hypothesis that there are interactions between RFI and harvest body weight for carcass traits and body composition was investigated. Seventy-two Nellore steers (16 to 21 month-old, 334±19 kg initial body weight [BW]) were fed a finishing ration (74.5% TDN, 14.3%CP) on an ad libitum basis, for 70 days. Daily dry matter intake (DMI) and body weight gain (ADG) were measured individually. The 12 lowest and the 12 highest RFI steers were classed as low- and high-RFI groups, respectively (selection phase), and were fed until reaching slaughter BW of 460, 490, 520 and 550 kg (finishing phase). Before slaughter, total urine was collected for determination of daily 3-methylhistidine excretion and myofibrillar protein breakdown rates. Body chemical composition was estimated by the isotope dilution method using deuterium oxide. Objective tenderness and Ca+2-dependent protease activities were measured on Longissimus muscle. In the selection phase, low-RFI steers had lower DMI, feed:gain, RFI, and ultrasound rump fat thickness gain than high-RFI cattle, but no differences were observed for ADG, final BW, ultrasound fat thickness and Longissimus area. In the finishing phase, no interactions were observed between RFI and slaughter BW. No differences between more and less efficient cattle were observed for hot carcass weight, dressing percentage, kidney, pelvic and inguinal fat, visceral mass, Longissimus area, backfat thickness, marbling score, trimmings and retail product yield. Low-RFI steers presented less fat on the gastrointestinal tract (GIT) than high-RFI cattle. No differences were observed for myofibrillar fragmentation index, Warner-Bratzler shear force and calpain system activities. Fractional rates of protein degradation, synthesis and accretion were similar between high- and low-RFI cattle. Low-RFI Nellore steers may store less subcutaneous carcass fat at body weights ranging from 340 and 460 kg. At higher body weights (460-550 kg), carcass traits and body composition are not affected by RFI, but least efficient cattle present greater visceral fat mass. Breeding zebu cattle for improved RFI may decrease feed intake and improve feed efficiency without compromising meat quality.

O consumo alimentar residual (CAR) é uma medida de eficiência alimentar independente da taxa de crescimento e do peso adulto. Pouco se sabe sobre associações entre o CAR e as características de produtividade dos animais selecionados para este parâmetro, bem como os mecanismos relacionados ao aumento na eficiência alimentar dos animais selecionados para o CAR. O objetivo deste estudo foi avaliar as associações entre CAR, desempenho, crescimento dos componentes da carcaça e digestão do amido em bovinos Nelore, em confinamento, e avaliar a utilização dos parâmetros fecais, (porcentagem de amido, proteína, matéria seca e pH), como indicadores da digestão do amido. Noventa e seis touros da raça Nelore selecionados como elite ou superior na prova de ganho de peso a pasto e classificados quanto ao seu temperamento, medido pela distância mínima de aproximação, foram confinados e alimentado ad libitum (68% NDT, 13,5% PB, 28,1% amido) durante 68 dias com peso vivo médio inicial de 408±33 e idade de 645±52 dias. A ingestão de matéria seca (IMS) e o ganho médio diário (GMD) foram medidos individualmente. As pesagens e as mensurações da espessura de gordura subcutânea (EGS), espessura de gordura na picanha (EGP) e a área de olho de lombo (AOL) foram realizadas a cada 21 dias. Os animais foram classificados como sendo de alto CAR (DP>0,5), baixo CAR (DP<0,5) e intermediário. O amido das amostras da ração e da sobra coletadas durante sete dias consecutivos, no final do experimento, foi determinado por análise laboratorial e a quantificação do amido fecal foi feita através do NIRS e a digestão do amido estimada. Durante o confinamento, novilhos com baixo CAR tiveram menores IMS e conversão alimentar que novilhos com alto CAR, mas nenhuma diferença foi observada no GMD, no PV final, AOL, EGP, EGS, temperamento e ganho de peso durante a prova de ganho de peso. Animais mais eficientes também não diferiram dos demais quanto à digestão do amido, no entanto, animais mais eficientes tiveram menores quantidades de proteína fecal. O CAR foi positivamente correlacionado com a IMS, consumo de amido, proteína fecal e eficiência alimentar e negativamente correlacionado com a conversão alimentar, no entanto não houve correlação entre o CAR e o GMD na prova de ganho de peso a pasto e temperamento. A digestão do amido foi positivamente associada com a PB fecal e negativamente correlacionada com a MS e o amido fecal. Não houve associação entre o CAR, o GMD a pasto e o temperamento, talvez porque são filhos de vacas selecionadas para essas características e também foram os que tiveram os melhores desempenhos na prova a pasto. / Residual feed intake (RIF) is a trait of efficiency independent of growth rate and mature weight. Little is known about the association between RFI and productivity and the mechanisms associate on improve efficiency in RFI. This work aimed to study the association between RFI, performance, growth of carcass traits and starch digestion in Nellore bulls and to investigate the utilization of fecal parameters like crude protein, dry matter and pH, as indicators of starch digestion. Ninety six Nellore bulls, with 645±52 days, 408±33kg initial body weight, previously selected as elite or superior in performance test on pasture and classified according to their temperament, were fed a finishing ration (68% TDN, 13,5%CP and 28,1% starch) on an ad libitum basis, for 68 days. Daily dry matter intake (DMI) and body weight gain (BWG) were measured individually. All animals were weighed and had ultrasonic measurements of Longissimus muscle area (REA) and backfat thickness (BF) on the 12th and 13th ribs obtained every 21 days. RFI was calculated as the difference between actual DMI and predicted DMI for a common mid-test BW0.75 and daily BWG and the animals were classified as high (SD>0,5), low (SD<0,5) and intermediary RFI. The starch of the diet and orts was determinate by laboratory analyses and the fecal starch was determinate with NIRS, and the starch digestion estimated. Low RFI bulls had low DMI and feed:gain than high RFI bulls, but no difference were observe in daily BWG, final BW, REA, BF, temperament and daily gain in the pasture performance test. More efficient bulls had no difference in starch digestion, however low RFI bulls had lower crude protein in the feces. RFI was positively correlated with DMI, starch intake and feed efficiency, but RFI was not correlated with daily BWG in the pasture performance test and temperament. Starch digestion was positively associated with Fecal CP and negatively associated with DM and starch. There was no association between RFI and daily BWG on pasture and temperament, perhaps because these animals belonged to a herd that was selected for these characteristics and also were the animals that had the best performance on pasture test.

Feed efficiency traits in Santa Inês sheep under genomic approaches / Eficiência alimentar em ovinos da raça Santa Inês sob abordagem genômicaAmanda Botelho Alvarenga 28 September 2017 (has links)
The selection on genetic values predicted from markers could substantially increase the rate of genetic gain in animals by increasing accuracy of prediction and reducing generation interval, especially for difficult to measure traits, such as feed efficiency. Feed efficiency is the most important trait in animal production due to its impacts on cost of production and environmental factors. Many metrics measure the feed efficiency, such as ratio of gain to feed (FER), the ratio of feed to gain (FCR) and residual feed intake (RFI). Nevertheless, in ovine, no study with the aim of understand the genetic variants or the accuracy of genomic estimated breeding value (GEBV) for feed efficiency traits was published yet. Moreover, before to apply the genomic information, it is necessary to understand and characterized the population structure, for instance, by linkage disequilibrium (LD). Both genome-wide association studies (GWAS) and genomic selection (GS) leverage LD between marker and causal mutation. Based on the above considerations, the aim of this study was to map LD in ovine, characterized by Brazilian Santa Inês sheep; to search genetic variants for feed efficiency traits (FER, FCR and RFI) through GWAS; and to verify the accuracy of GEBV for RFI. In total, 396 samples (animals) of Longissimus dorsi muscle were collect. A high-density panel of SNP (Illumina High-Density Ovine SNP BeadChip®) comprising 54,241 SNPs was used to obtain the genotyping data. The phenotype data was comprised of 387 animals. The average LD between adjacent markers for two LD metrics, r² and |D\'|, were 0.166 and 0.617, respectively. The degree of LD estimated was lower than reported in other species and it was characterized by short haplotype blocks. Consequently, for genomic analyses, high-density panels of marker are recommended. Many markers were associated to feed efficiency traits in GWAS, mainly to RFI trait. Few candidate genes were reported in this study, highlighting NRF-1 (nuclear respiratory factor 1), which controls mitochondrial biosynthesis, the most important process responsible by a great fraction of the produced energy. Finally, we verified the accuracy of GEBV for RFI using few Bayesian regression models, and we found low accuracy, ranging from 0.033 (BayesB with π=0.9912) to 0.036 (BayesA), which might be explained by the low relationship among animals and small training population. / A seleção com base nos valores genéticos genômicos preditos pode aumentar substancialmente a taxa de ganho genético em animais por meio do aumento da acurácia de predição e redução do intervalo de gerações, especialmente para características de difícil e/ou onerosa mensuração, como eficiência alimentar. A eficiência alimentar é uma das características mais importantes na produção animal devido principalmente aos seus impactos econômicos e ambientais. Muitas métricas representam a eficiência alimentar, por exemplo: a relação do ganho de peso e consumo alimentar (EA), a proporção do consumo alimentar e ganho de peso (CA) e o consumo alimentar residual (CAR). Em ovinos, nenhum estudo com o objetivo de buscar variantes genéticas ou verificar a acurácia do valor genético genômico estimado para eficiência alimentar foi publicado. Adicionalmente, antes de aplicar a informação genômica, é necessário compreender e caracterizar a estrutura da população, como por meio do desequilíbrio de ligação (LD). O estudo de associação genômica (GWAS) e seleção genômica (GS) consideram o LD entre marcador e a mutação causal. Com base nas considerações acima, o objetivo deste estudo foi mapear o LD em ovinos, caracterizado pela raça ovina Santa Inês; localizar variantes genéticas para as características de eficiência alimentar (EA, CA e CAR) utilizando a abordagem GWAS; e verificar a acurácia da estimação dos valores genéticos genômico para o CAR. No total, foram coletadas 396 amostras (animais) do músculo Longissimus dorsi, para posterior genotipagem utilizando o painel de alta densidade (Illumina High-Density Ovine SNP BeadChip®), compreendendo 54.241 SNPs. O banco fenotípico é composto por 387 animais. O LD médio entre marcadores adjacentes para duas métricas de LD, r² e |D\'|, foram 0,166 e 0,617, respectivamente. O grau de LD estimado foi menor que o relatado em outras espécies e foi caracterizado por blocos de haplótipos curtos. Consequentemente, para as análises genômicas são recomendados painéis de marcadores de alta densidade. No GWAS, foram encontrados muitos marcadores associados aos fenótipos, em especial, à característica CAR. Alguns genes candidatos foram relatados neste estudo, destacando-se o NRF-1 (fator respiratório nuclear 1), que controla a biossíntese mitocondrial, o processo mais importante responsável por grande parte da produção de energia. Finalmente, verificamos a acurácia do valor genético genômico estimado para o CAR usando modelos de regressão Bayesiana, e encontramos baixos valores para acurácia (0,033 a 0,036) o que pode ser explicado pelo baixo grau de relacionamento entre os indivíduos e tamanho reduzido da população de treinamento.
