La Sardaigne, en raison de sa position géographique au cœur de la Méditerranée occidentale, constitue une zone clé pour la compréhension de la diffusion du Néolithique dans le domaine nord-tyrrhénien. La deuxième moitié du Ve millénaire cal BC marque l’essor et l’aboutissement du processus de diffusion de l’économie néolithique dans l’île, sous l’égide des groupes humains « San Ciriaco ». Des fouilles récentes dans la Sardaigne centre-occidentale, zone cruciale pour cette entité culturelle régionale, ont décelé des contextes inédits dont l’étude est fondamentale pour la connaissance de ces sociétés. Cette thèse se propose d’explorer le rôle des poteries dans les systèmes techniques et les comportements symboliques des groupes humains San Ciriaco en Sardaigne centre-occidentale, afin d’obtenir des indices économiques sur les stratégies de gestion des ressources et sur les modes de vie de ces populations. L’approche interdisciplinaire suivie combine l’analyse morphométrique des récipients, l’observation des traces d’usure et l’analyse chimique des résidus organiques (en CPG et CPG/SM) piégés dans la matrice céramique. Les résultats révèlent une structuration des assemblages céramiques en plusieurs gammes fonctionnelles, variablement représentées sur des sites à destination différente (habitat, sépultures, « dépôts » de vases). Des comportements techniques et symboliques liés à la gestion des récipients tout au long de leur cycle d’utilisation sont mis en lumière. Ces résultats offrent une première contribution à la plus large problématique de l’évolution de la fonction des poteries, tout au long du Néolithique, au sein du domaine méditerranéen occidental. / The isle of Sardinia (Italy) is a key zone in understanding the spread and the diffusion of the Neolithic in North Tyrrhenian region, due to its position in the middle of western Mediterranean Sea. Here, the rise and culmination of Neolithic economy is carried by San Ciriaco populations in the second half of V millennium cal BC. Recent archaeological excavations in central-western Sardinia revealed new contexts the study of which is fundamental in increasing our knowledge of these Neolithic societies.The aim of this work is to investigate the role of pottery in technical systems and symbolical behaviours of San Ciriaco societies in central-western Sardinia, in order to find evidence of strategies of use and explore their way(s) of life. The interdisciplinary approach in this study combines morphometrical analysis of vessels, use-wear observation and GC / GC-MS chemical analysis of organic residues absorbed in pottery.This research revealed a structuration of pottery assemblages into several functional categories which are differently represented in sites with a different function (settlements, burials, vessels “deposits”). The results contribute to highlight technical and symbolical behaviors linked to the use of pottery during its life cycle phases. These data offer a contribution to the issue of the evolution of pottery function during the Neolithic period in Western Mediterranean. / La Sardegna, in virtù della sua posizione centrale nel Mediterraneo occidentale, rappresenta una zona cruciale per la comprensione del processo di propagazione del Neolitico e della sua evoluzione nell’area nord-tirrenica. Il pieno sviluppo dell’economia neolitica nell’isola è segnato dalla formazione e diffusione di un’entità culturale regionale originale, denominata “cultura di San Ciriaco”, durante la seconda metà del V millennio cal BC (Neolitico Medio B). Alcuni recenti scavi nella Sardegna centro-occidentale, scenario fondamentale nello sviluppo di tale entità culturale, hanno portato alla luce dei contesti inediti, il cui studio si rivela essenziale per la conoscenza di queste società neolitiche.L’obiettivo di questa ricerca è specificamente quello di indagare il ruolo dei recipienti ceramici nei sistemi tecnici e nei comportamenti simbolici dei gruppi umani San Ciriaco della Sardegna centro-occidentale, nell’intento di acquisire degli indizi sulle strategie di gestione delle risorse naturali e sulle modalità di sussistenza delle popolazioni durante il Neolitico medio B.L’approccio interdisciplinare seguito associa l’analisi morfometrica dei recipienti all’osservazione delle tracce d’uso e all’analisi chimica dei residui organici assorbiti nella matrice ceramica, mediante la tecnica della gascromatografia (GC) e della gascromatografia/spettrometria di massa (GC/MS).I risultati ottenuti rivelano una strutturazione delle collezioni ceramiche in molteplici gamme funzionali distinte e variabilmente rappresentate in siti con destinazione funzionale differente (abitato, sepolture, “depositi” di recipienti). L’analisi di tali contesti ha permesso di evidenziare alcuni specifici comportamenti tecnici e simbolici legati alla gestione dei recipienti durante tutto il loro ciclo d’utilizzo.L’interpretazione storica dei dati acquisiti con questa ricerca offre un contributo alla più vasta questione dell’evoluzione della funzione dei recipienti ceramici, nel corso del Neolitico, nell’area del Mediterraneo occidentale.

Suivi physico-chimique, microbiologique et écotoxicologique du compostage de boues de STEP mélangées à des déchets de palmier : validation de nouveaux indices de maturité / physico-chemical microbiological and ecotoxicological monitoring during composting of sewage sludge-date palme waste : validation of new maturity index

Le traitement de boues de station d'épuration (Boues activées de Marrakech) en mélange avec déchets verts (Palmier dattier) selon deux mélanges dont les proportions : A (1/3 Boues + 2/3 Déchets de palmier) et B (1/2 Boues + 1/2 Déchets de palmier), a été effectué par la filière du compostage, pendant six mois. Au cours du co-compostage, l’intense activité microbienne s’est traduite par une augmentation de température (autour 65 C°) au cours des premières semaines (phase thermophile) et un taux de décomposition final de l’ordre de 40%. Après six mois de co-compostage, le compost final est caractérisé par un rapport C/N voisin de 10, un rapport de NH4+/NO3- < 1, un pH autour de la neutralité signe de maturité des composts. L’analyse FTIR a montré une diminution de l'absorbance des bandes aliphatiques et l'augmentation de la structure de bandes d'absorbance aromatiques reflètent l'état d'avancement du processus d'humification. Le taux d’abattement des lipides totaux est de l’ordre de 43%. Les esters méthyliques d’acide gras (FAMEs) des Gram-positif (i,C15 :0) ont augmenté durant la phase thermophile. Les FAMEs d’origine non spécifique (C6 :0, C14 :0) ont connu une intense diminution, les FAMEs de bactéries non spécifiques représentent une grande teneur durant la phase thermophile. L’indice CPI a augmenté à la fin du co-compostage indiquant l’enrichissement du compost en FAMEs d’origine végétale. Les principaux composés ligneux identifiés, au cours du co-compostage, par Py-GC-MS, sont classés en deux groupes. Le premier est constitué de 7 composés dont la teneur diminue au cours du co-compostage, parmi lesquels : Toluène, 2,4-diméthylbenzène, éthylbenzène, Styrène, 1-éthyl-2-méthylbenzène, 4-méthylphénol et 2-méthylnaphthalène. Le deuxième groupe est constitué de 4 composés qui augmentent au cours du co-compostage : phénol, benzofuran, éthylméthoxyphénol et diméthoxyphénol. Les principaux stéroïdes identifiés sont les C27-C29 sterènes, stanols, 5β-cholesta-3-one, cholesta-3,5-diène et 2 thiostéranes. A l’exception des thiosteranes et quelques composés de C27-C29 cholestenes la concentration relative des stéroides diminue au cours du processus suit à leur attaque microbien. L’abattement total de la teneur des stéroïdes est corrélé positivement avec les indicateurs de maturité du compost (C/N et NH4+/NO3-) ce qui ouvre la voie à une éventuelle utilisation des stéroïdes comme indicateur de dépollution et de maturité de compost. 12 isolats d’actinomycètes ayant une activité antimicrobienne vis-à-vis d’un large spectre des germes pathogènes ont été isolés sur le milieu sélectif CTEA. Le degré d’hygiénisation est confirmé par la diminution de la concentration des coliformes fécaux et totaux, et l’abattement des œufs d’helminthes identifiés (Ascaris sp., Capillaria sp., et Trichuris sp.) vers la fin du co-compostage. La phytotoxicité, déterminée par l’effet des extraits hydrosolubles à différents stades de co-compostage sur la germination et la croissance des radicules (Navet, Cresson, Laitue, Luzerne), a diminué et l’indice de germination dépasse 100%, après six mois de co-compostage. La génotoxicité du chrome hexavalent (Cr(VI)) du substrat de co-compostage est corrélée positivement avec la fréquence des micronoyaux (MN). Après six mois de co-compostage le taux des MN diminue avec un taux d’abattement de 70,4% et 77,2% avec l’abattement de la concentration du Cr(VI) avec 58 et 58,6% respectivement pour le mélange A et B. Ceci ouvrira la voie d’utilisation de cet indice comme un indice de maturité des composts. La diminution de la phytotoxicité et la génotoxicté au cours du co-compostage confirme l’état de stabilisation et de la maturité des co-composts, ce qui pemettra l’épandage de ces composts en tant qu’amendement organique des sols sans risque de contamination du système sol-plante. / The co-composting of activated sludge and lignocellulose waste (palm tree waste) was monitored to study the behaviour of two mixtures, referred to as A (1/3 sludge + 2/3 palm waste) and B (1/2 sludge + 1/2 waste palm) for 6 months. The biotransformation was evaluated by physicochemical and spectroscopic analyses. The thermophilic phase is characterized by a rise in temperature, which peaked at 65°C. This is the result of intense microbial activities. The final composts exhibited a higher degree of decomposition than the controls as shown by a decomposition rate of about 40%, decrease of C/N ratio to around 10 and NH4 +/NO3 - ratio below 1. The decrease of aliphatic absorbance bands and the increase of aromatic absorbance bands follow the progress of the humification process. Total extractable lipid was decreased by 43%. The fatty acid methyl esters (FAMEs) from Gram-positive bacteria (i, C15: 0) increased during the thermophilic phase. FAMEs from non-spécific origin (C6 :0, C14 :0) exhibited a decrease toward the end of co-compostin, linear FAMEs from non-specific bacteria underwent a decrease during co-composting. The CPI index thus increased at the end of the composting process, indicating that the final product was proportionally richer in fatty acids of plant origin. Two lignin groups were distinguished by Py-GC-MS. Group 1 contained toluene, 2,4-dimethylbenzene, ethylbenzene, styrene, 1-ethyl-2-methylbenzene, 4-methylphenol and 2-methylnaphthalene; their relative proportions decreased during co-composting. A second group of 4 components showed concentrations that increased with co-composting time: phenol, benzofuran, ethylmethoxyphenol and dimethoxyphenol. The main steroids identified were C27-C29 sterenes and stanols, 5β-cholesta-3-one, cholesta-3,5-diene and 2 thiosteranes. Except for thiosteranes and some of the C27-C29 cholestenes, the relative concentrations decreased during co-composting due to microbial degradation. The changes in steroids during co-composting, was positively correlated with the physico-chemical parameters of mature compost, especially C/N and NH4 +/NO3 - ratios, opening the way for the use of steroids as indicators of pollution and compost maturity. On the selective CTEA medium, 12 active strains of isolated actinobacteria presented a suppressive action against various pathogens. This may justify that a biotic factor is also an important factor contributing to making co-composting substrates hygienic. The degree hygiene reached is confirmed by the reduction in the faecal and total coliforms, and by the abatement of identified helminth eggs (Ascaris sp., Capillaria sp., and Trichuris sp.) towards the end of the process. The phytotoxicity determined by the effect of aqueous extract, at various stages of the co-composting, performed by monitoring the number of germinated seeds and the rootlets growth of turnip, watercress, alfalfa, and lettuce was decreased, and the growth of radicals that have a germination index that exceeds 100% after six months of co-composting. Hexavalent chromium (Cr(VI)) genotoxicity showed a positive correlation with micronucleus (MN) frequency. After six months of co-composting, the MN rate decreased significantly by 70.4 and 77.2% with decreasing Cr(VI) concentration with 58 and 58.6%, for mixtures A and B respectively. That indicates their suitability for use as a maturity index. During co-composting the abatement rate of phytotoxicity and genotoxicity confirme the maturation and stabilization degree of co-composting end products which encourages their recycling in agriculture as a fertilizer for the soil without any contamination of the soil-plant system.

Réactivité chimique en phase gazeuse de molécules organiques d’intérêt atmosphérique et astrophysique / Chemical reactivity in gaz-phase of organic molécules of atmospherical and astrophysical interests

Cette thèse reporte l'étude expérimentale de la cinétique en phase gazeuse de plusieurs réactions d'intérêt atmosphérique et astrophysique. La connaissance des constantes de vitesse de ces réactions est utile à la compréhension des processus de synthèse et de destruction des molécules dans l'atmosphère terrestre et dans le milieu interstellaire. Sur le plan atmosphérique, nous avons étudié les réactions d'une série d'hydroxycétones, une sous-famille de COV carbonylés, (4-hydroxy-2-butanone, 3-hydroxy-3-méthyl-2-butanone and 4-hydroxy-4-méthyl-2-pentanone) avec le radical OH et l'atome de Chlore Cl afin de définir leur devenir atmosphérique. Nous avons déterminé les constantes de vitesse des réactions de ces hydroxycétones avec le radical OH en absolue à température ambiante et en fonction de la pression en utilisant la cellule cryogénique couplée à la technique PLP-LIF (Photolyse à Laser Pulsé – Fluorescence Induite par Laser). Les constantes de vitesse des réactions de ces hydroxycétones avec Cl ont été mesurées en relative à température ambiante et à pression atmosphérique en utilisant les chambres de simulation atmosphérique couplées aux techniques analytiques de détection : FTIR et GC-MS. En plus, une étude mécanistique a été menée également afin d'identifier et quantifier les produits issus de ces réactions. L'ensemble des résultats obtenus est discuté et nous a permis d'évaluer les implications atmosphériques de ces hydroxycétones (durée de vie et impact sur l'environnement). Le plan astrophysique, la cinétique des réactions impliquant le radical CN et une série de molécules azotées (méthyamine, diméthylamine, triméthylamine et acétonitrile) ont été étudiés sur une large gamme de température (23 K – 354 K) en utilisant la technique CRESU (Cinétique de Réaction en Écoulement Supersonique Uniforme) et la cellule cryogénique couplées à la technique de détection PLP-LIF. Ces études expérimentales ont été accompagnées par des études théoriques afin de mieux comprendre les mécanismes réactionnels. Les éventuelles implications astrophysiques de l'ensemble de ces résultats ont fait l'objet d'une discussion détaillée. / This thesis reports the experimental kinetic study of the gas phase reactions of atmospheric and astrophysical interests. The knowledge of the reactions rate constants is useful to understand the mechanisms of formation and destruction of molecules in the Earth’s atmosphere and in the interstellar medium. On the atmospheric side, we have studied the reactions of a series of hydroxyketones (4-hydroxy-2-butanone, 3-hydroxy-3-methyl-2-butanone and 4-hydroxy-4-methyl-2-pentanone), a large category of hydroxycarbonyls with OH radicals and chlorine atoms Cl in order to determine their atmospheric fate. We have measured the absolute rate constants of the reactions of hydroxyketones with OH radicals by using the cryogenic cell coupled to PLP-LIF technique (Pulsed Laser Photolysis - Laser Induced Fluorescence) at room temperature and as function of pressure. The relative rate constants of the reactions of the compounds investigated with Cl atoms were measured at room temperature and atmospheric pressure by using the atmospheric simulation chamber coupled to the analytical detection techniques: FTIR and GC-MS. In addition, a mechanistic study was also conducted in order to identify and quantify the products formed from these reactions. The whole results are discussed in order to assess the atmospheric implications of these hydroxyketones (lifetimes and environmental impact). On the astrophysical side, the kinetic of the reactions involving CN radical and a series of nitrogen-containing molecules (methylamine, dimethylamine, trimethylamine and acetonitrile) was studied over a wide range of temperature (23 K - 354 K) using CRESU technique (a French acronym standing for Cinétique de Réaction en Ecoulement Supersonique Uniforme) and the cryogenic cell coupled to the detection technique PLP-LIF. These experimental studies were accompanied by theoretical studies to better understand the reaction mechanisms. Possible astrophysical implications of the whole results have been discussed in this study.

<p>Hemijska karakterizacija ekstrakata&nbsp; <em>T. pratense&nbsp;</em>L.&nbsp; određena je LC-MS-MS, a etarskih ulja GC-MS analizom, pri čemu je&nbsp; identifikovano vi&scaron;e&nbsp;bioaktivnih jedinjenja.&nbsp; Spektrofotometrijskim&nbsp;metodama ispitan je antioksidantni potencijal&nbsp;ekstrakata i etarskih ulja. Hepatoprotektivni&nbsp;efekat određen je&nbsp; na&nbsp; homogenatu i hemolizatu&nbsp;jetre laboratorijskih mi&scaron;eva.&nbsp; Budući da&nbsp; ova&nbsp;biljka ima visok sadržaj izoflavona, određena je&nbsp;<br />estrogena, antiestrogena aktivnost, kao i&nbsp;citotoksičnost ekstrakata. Antimikrobna&nbsp;<br />aktivnosta ekstrakata i etarskih ulja ispitana je&nbsp;na&nbsp; &scaron;est bakterijskih sojeva.&nbsp; Ekstrakti i etarska&nbsp;ulja su bili umereno aktivni u pogledu biolo&scaron;kih&nbsp;aktivnosti, osim u slučaju antimikrobne&nbsp;aktivnosti koju nije pokazao nijedan ispitivani&nbsp;uzorak.</p> / <p>The chemical characterization of&nbsp; <em>T.&nbsp; pratense</em> extracts was determined by LC-MS-MS, and the essential oils of the GC-MS analysis, wherein the&nbsp; various&nbsp; bioactive compounds were identified.&nbsp; Antioxidant potential of extracts and essential oils was tested&nbsp; by spectrophotometric methods.&nbsp; Hepatoprotective&nbsp; effect&nbsp; was determined&nbsp; in&nbsp; haemolysate and liver homogenate of&nbsp; laboratory&nbsp; mice.&nbsp; Estrogen, antiestrogen activity, as well&nbsp; as the cytotoxicity of the extracts were&nbsp; determined&nbsp; due to the high&nbsp;content of isoflavones.&nbsp; Antimicrobial activity of extracts and essential oils was&nbsp; investigatied&nbsp; on six bacterial strains. Essential oils and extracts were moderately active in terms of biological activity, except&nbsp; in the case of&nbsp;&nbsp; antimicrobial activity&nbsp; where none of the extracts were active against tested bacterial strains.</p>

Isolierung und Charakterisierung der Chitin-basierten Skelette der marinen Schwämme Aplysina cavernicola und Ianthella basta

Die Schwammskelette der Ordnung Verongida zeichnen sich durch das Fehlen mineralischer Komponenten aus. Stattdessen bestehen sie aus Spongin, einem kollagenartigen Protein, und Chitin. Im Rahmen der vorliegenden Arbeit wurden die aus solch einem Chitin-Protein-Komplex bestehenden Skelette der Schwammspezies Aplysina cavernicola und Ianthella basta aus der Ordnung Verongida untersucht. Aufgrund ihrer morphologischen Unterschiede wurde für jede Schwammart eine eigene Methode zur Isolierung der Schwammskelette entwickelt. Die isolierten Skelette konnten anschließend mit verschiedenen Methoden wie REM, ATR-FTIR-Spektroskopie und NMR-Spektroskopie charakterisiert werden. Weiterhin wurde eine Methode zur Extraktion und Analyse der in den Skeletten befindlichen Aminosäuren mittels GC-MS entwickelt. Die Untersuchungen zeigten deutlich, dass es sich bei Spongin um ein kollagenartiges und halogeniertes Protein handelt, welches je nach Schwammart Unterschiede in der Aminosäurezusammensetzung aufweist. Darüber hinaus gelang es zum ersten Mal das Chitin aus dem Chitin-Protein-Komplex mittels Phosphorsäure zu entfernen. Aus den gewonnenen Erkenntnissen konnte abschließend ein Modell zum Aufbau des Chitin-Protein-Komplexes in der Schwammspezies Aplysina cavernicola entwickelt werden.

Entwicklung einer Multimethode zur Probenaufarbeitung und Bestimmung von gas- und flüssigkeitschromatographisch erfassbaren Pestiziden in Hühnereiern

Die Rückstandsanalytik tierischer Lebensmittel ist eine anspruchsvolle Aufgabe aufgrund des hohen Lipidanteils der Proben sowie des sich stetig vergrößernden Wirkstoffspektrums. Heutzutage werden für die Probenaufarbeitung die DFG S 19 Methode, mit der vorrangig unpolare Analyten nachgewiesen werden und zunehmend die QuEChERS Methode eingesetzt, die insbesondere auf die Erfassung polarer Pestizide abzielt. In dieser Arbeit wurde eine moderne Multirückstandsmethode für Hühnereier entwickelt, um sowohl gas- als auch flüssigkeitschromatographisch (GC, LC) erfassbare Wirkstoffe zu analysieren. Dazu gehören unpolare PCBs, Pyrethroide und Organochlorpestizide, aber auch polarere Organophosphate, Triazole und Carbamate. Das Verfahren basiert auf der Extraktion mittels Matrix Solid Phase Dispersion, der Reinigung auf Grundlage einer modifizierten Gelpermeationschromatographie (GPC) und zwei verschiedenen Festphasenextraktionen (SPEs) für GC- und LC-erfassbare Pestizide sowie der Quantifizierung mittels GC- und LC-MS/MS. Dünnschichtchromatographisch wurde die effektive Entfernung hochmolekularer Lipide durch die modifizierte GPC und niedrigmolekularer Fette durch die SPEs belegt. Laut der für Ei durchgeführten Validierung erfüllten 164 der 172 untersuchten Pestizide und alle sechs PCBs die Leistungskriterien für die amtliche Rückstandskontrolle - zumeist am niedrigsten validierten Level (5 µg/kg bzw. 0,5 µg/kg). Ausnahmen bildeten sehr polare LC-Pestizide (z.B. Aminopyralid, Clopyralid, MCPA, Quinmerac) und pH-Wert-abhängige GC-Analyten (Nicotin, Tolylfluanid, Dichlofluanid), die auch mit den etablierten Verfahren schwierig zu analysieren sind. Weiterhin verdeutlichte die erfolgreiche Untersuchung von verschiedenen Ringversuchsmaterialien, dass die ursprünglich für Eier entwickelte Methode auch für mageres Geflügelfleisch und Sahne genutzt werden kann. Gegenüber den etablierten Verfahren wies die neue Methode deutliche Vorzüge auf. So belegte die Dünnschichtchromatographie, dass mit der neuen Methode Cholesterin, aber auch freie Fettsäuren besser abgetrennt werden als mit den etablierten Verfahren. Die neue Methode verbrauchte im Vergleich zur DFG S 19 Methode 46 % weniger Lösungsmittel und ermöglichte eine Verdopplung des Probendurchsatzes innerhalb von 8 h. Zudem eignete sich das entwickelte Verfahren laut den Validierungsdaten für GC-Analyten deutlich besser als die QuEChERS Methode und etwas besser als die DFG S 19 Methode (v.a. für Pyrethroide). Hinsichtlich der LC-Analyten unterschieden sich die neue und die QuEChERS Methode nur bei wenigen Analyten. Complexes pinceurs de type POCOP de Nickel (II) : synthèse, caractérisation, réactivité et applications catalytiques

Ce mémoire décrit la synthèse, la caractérisation spectroscopique et l’étude de la réactivité catalytique d’une nouvelle série de complexes pinceurs de Ni(II) formés à partir du ligand POCOPPh (P,C,P-2,6-{Ph2PO}2C6H4), très peu étudié dans le cas du nickel. Les études décrites dans ce mémoire examinent l’effet des substituants des phosphines sur les propriétés spectroscopiques et électrochimiques ainsi que les activités catalytiques. La synthèse du ligand a été améliorée par rapport à la procédure connue dans la littérature en diminuant le temps de réaction à 30 min et la température jusqu'à température ambiante. Les composés pinceur (P,C,P-2,6-{Ph2PO}2C6H3)NiX ont été obtenus avec des rendements variant entre 60% et 88%. Le premier complexe a été synthétisé en faisant réagir le précurseur NiBr2(NCCH3)x avec le ligand POCOPPh pour donner (POCOPPh)NiBr. Ce dernier réagit par la suite avec les sels d’argent et de potassium pour donner 4 nouveaux complexes soient : (POCOPPh)NiCN, (POCOPPh)NiOTf, (POCOPPh)NiOAc et (POCOPPh)NiONO2 (OTf = triflate et OAc = acetate). Vu la réactivité limitée du dérivé bromure, le dérivé (POCOPPh)NiOTf a été utilisé pour la préparation du composé (POCOPPh)NiCCPh. Le dérivé Ni-OTf a été utilisé également pour la synthèse des complexes (POCOPPh)NiR qui ont été détectés par RMN. Ces complexes (POCOPPh)NiR ont montré une stabilité trop faible et donnent des nouveaux complexes de type (POCOPPh)NiX en échangeant l’halogène avec le Mg ou de type (POCOPPh)NiOH en s’hydrolysant. Les espèces cationiques [(POCOPPh)NiNCR][OTf] (R= Me, CHCH2, CHCHMe, C(Me)CH2, NCCH2CH2N(Ph)H) ont été obtenues facilement et avec des bon rendements à partir du (POCOPPh)NiOTf. Tous les composés obtenus ont été caractérisés par la spectroscopie RMN (1H, 13C{1H}, 31P{1H}, 19F{1H}), la spectroscopie IR et la spectroscopie UV-vis. L’analyse élémentaire et l’analyse par la diffraction des rayons X, dont le but est de résoudre la structure à l’état solide, ont été utilisées pour la plupart des complexes. Des études de voltampérométrie cyclique ont été menées pour déterminer la densité électronique des centres métalliques et l’effet des phosphines sur cette propriété électrochimique. Dans le but de déterminer l’effet des substituants des phosphines sur l’activité catalytique des complexes, nous avons évalué les réactivités catalytiques des deux complexes (POCOPPh)NiOTf et (POCOPi-Pr)NiOTf dans la réaction d’hydroamination des oléfines activés et plus spécifiquement l’acrylonitrile. Après optimisation des conditions expérimentales, on a constaté que la réactivité des deux composés sont similaires mais une grande différence apparaît après l’ajout des additifs. En effet, le complexe (POCOPi-Pr)NiOTf donne une bonne activité catalytique en présence de la triéthylamine, tandis que cette activité diminue considérablement en présence d’eau, contrairement au complexe (POCOPPh)NiOTf qui est plus actif en présence d’eau. Dans le cas du complexe (POCOPPh)NiOTf, on a pu montrer que la base se coordonne au nickel dans le produit formé après la réaction d’hydroamination, ce qui diminue l’activité de ce complexe dans certains cas. Également on a exploré la réaction de l’addition du lien O-H sur l’acrylonitrile, et étonnamment le complexe (POCOPPh)NiOTf est beaucoup plus actif que son homologue (POCOPi-Pr)NiOTf dans le cas des alcools aromatiques. Par contre, les alcools aliphatiques restent un défi majeur pour ce genre de complexe. Le mécanisme de cette réaction qui a été proposé montre que l’alcoolyse passe par les deux intermédiaires (POCOPPh)NiOAr et [(POCOPPh)NiOAr][HOAr] mais l’isolation de ces intermédiaires observés par RMN semble être difficile. / This thesis describes the synthesis, spectroscopic characterization and the catalytic activities of a new family of pincer complexes of Ni (II) starting from the ligand POCOPPh (P,C,P-2,6-{Ph2PO}2C6H4) for which very few nickel complexes have been reported previsouly. We discuss the influence of P-substituents on the spectroscopic, electrochemical and catalytic activities of these complexes. The synthesis of POCOPPh has been improved comparatively to the procedure reported in the literature by reducing the reaction time to 30 minutes and the temperature to room temperature. The complex (P,C,P-2,6-{Ph2PO}2C6H3)NiBr was obtained with 88% yield by reacting the precursor NiBr2(NCCH3)x with POCOPPh . This complex was then reacted with various silver and potassium salts to give the following complexes (POCOPPh)NiCN, (POCOPPh)NiOTf, (POCOPPh)NiOAc and (POCOPPh)NiONO2 (OTf = triflate et OAc = acetate). The limited reactivity of the bromo derivative led us to use (POCOPPh)NiOTf for the preparation of some of the desired derivatives, such as (POCOPPh)NiCCPh. Attempts to prepare the desired alkyl derivatives (POCOPPh)NiR were not successful, but we were able to detect these derivatives using NMR. The thermal instability of (POCOPPh)NiR led to formation of new (POCOPPh)NiX complexes by halogen exchange with MgX2 or (POCOPPh)NiOH by hydrolysis. The cationic species [(POCOPPh)NiNCR][OTf] (R = Me, CHCH2, CHCHMe, C(Me)CH2, NCCH2CH2N(Ph)H) also were obtained easily from the (POCOPPh)NiOTf with good yields. All these complexes were characterized by elemental analysis, NMR spectroscopy (1H, 13C{1H} 31P{1H}, 19F{1H}), IR spectroscopy and UV-vis spectroscopy. For most complexes analysis by X-ray diffraction allowed us to establish their solid state structures. A few studies by cyclic voltammetry have been done to determine the electronic density of the metal center and the P-substituent influence on this characteristic. In order to investigate the effect of phosphine substituents on the catalytic activities of this type of complexes, catalytic studies were undertaken with the following two complexes (POCOPPh)NiOTf and (POCOPi-Pr)NiOTf in hydroamination of activated olefins specifically acrylonitrile. After optimization of experimental conditions, it was found that both complexes have similar activities but what makes a huge difference is the use of additives. Indeed, (POCOPi-Pr)NiOTf showed good catalytic activity in the presence of triethylamine as base but this activity decreased significantly in the presence of water. The opposite was observed with (POCOPPh)NiOTf complex: it was shown that triethylamine coordinates to the nickel center in this complex and hence reduces its activity in some cases. We Also explored other reactions such as the addition of the O-H bond in aromatic alcohols to acrylonitrile, and it was surprising that (POCOPPh)NiOTf is much more active than its homologous (POCOPi-Pr)-NiOTf. However aliphatic alcohols remain a major challenge for this kind of complex. Mechanistic studies suggest that this reaction passes through the following intermediates (POCOPPh)NiOAr and [(POCOPPh)NiOAr][HOAr]. These species were observed by NMR but not isolated.

Desenvolvimento e aplicação de uma nova fase para extração por sorção em barra de agitação (SBSE) / Development and application of a new phase for stir bar sorptive extraction (SBSE)

Uma fase polimérica inédita de polidimetilsiloxano/carvão ativado (PDMS-ACB) é proposta como fase extratora para extração por sorção em barras de agitação (SBSE). A barra de PDMS-ACB foi desenvolvida no laboratório usando um molde de teflon simples, demonstrando estabilidade e resistência aos solventes orgânicos. Utilizando uma única barra de PDMS-ACB mais de 150 extrações foram realizadas sem qualquer dano. A barra de SBSE contém aproximadamente 100 &micro;L de revestimento polimérico, 2,36 mm de diâmetro e 2,2 cm de comprimento. A barra PDMS-ACB foi aplicada na extração de seis pesticidas (ametrina, atrazina, bifentrina, carbofurano, metribuzim e tebutiurom), com propriedades predominantemente polares, comumente aplicados em plantações de cana-de-açúcar. A barra PDMS-ACB foi utilizada na determinação de pesticidas em garapa através de cromatografia gasosa hifenada à espectrometria de massa (GC-MS). O planejamento experimental foi empregado na etapa de otimização da extração SBSE, um planejamento fatorial fracionado avaliou a influência dos principais parâmetros envolvidos. Posteriormente, o planejamento composto central (CCD) com conformação estrela foi explorado para otimizar os fatores significativos na extração. Apenas 200 &micro;L de acetato de etila foram utilizados como solvente na dessorção líquida (LD). Para os compostos avaliados, o método apresentou limite de quantificação (LOQ) no intervalo de 0,5 - 40 &micro;g L-1, as recuperações variaram entre 0,18 - 49,50 % e a precisão intra-dia variou de 0,072 - 8,40 %. Concluída a etapa de validação, o método foi aplicado em amostras reais de garapa disponíveis comercialmente em São Carlos-SP. / A novel polydimethylsiloxane/activated carbon (PDMS-ACB) is proposed as new polymeric phases for stir bar sorptive extraction (SBSE). The PDMS-ACB was developed in lab using simple teflon\'s mold, demonstrating remarkable stability and resistance to organic solvents, over 150 extractions without any damage. SBSE bar contained 100 &micro;L of polymeric coating, a diameter of 2.36 mm and a length of 2.2 cm. It was applied to the determination of pesticides (ametryn, atrazine, bifenthrin, carbofuran, metribuzine and tebuthiuron) having predominantly polar properties, applied in sugarcane crops. PDMS-ACB was employed in the determination of pesticides in sugarcane juice using gas chromatography coupled to mass spectrometry (GC-MS). Experimental design was employed in the optimization step, a fractional factorial evaluated the main parameters involved in the extraction procedure. Afterwards, central composite design (CCD) was exploited to optimize the significant factors on the extraction. About 200 &micro;L of ethyl acetate were employed as solvent in the liquid desorption (LD). The method presented limit of quantification (LOQ) from 0.5 to 40 &micro;g L-1, recoveries varied 0.18 - 49.50% and precision intra-day 0.072 - 8.40%. Hence, the method was applied to the analysis of real sugarcane juice samples commercially available in São Carlos-SP.

Composição química e capacidade sequestrante de espécies reativas de oxigênio e nitrogênio de mel orgânico brasileiro / Chemical composition and Nitrogen and Oxygen Reactive Species scavenging activity of Brazilian organic honey

O Brasil apresenta grande potencial para a exploração da apicultura, dado ao seu vasto território e flora diversificada, o que permite diferentes variedades de méis com propriedades únicas. O estado do Paraná é um dos maiores produtores de méis do país e o investimento em produção que atenda aos mercados mais exigentes estimulou a produção do mel orgânico. O conhecimento desde a antiguidade sobre os efeitos benéficos à saúde pelo mel vem estimulando a pesquisa deste alimento nobre. Assim, este trabalho teve por objetivo estudar méis orgânicos brasileiros certificados (MO) para a caracterização do perfil fenólico, volátil, além da avaliação da capacidade de sequestro das espécies reativas de oxigênio e nitrogênio. Os méis foram coletados nos apiários de apicultores com certificação orgânica de dois municípios do sul do Paraná, General Carneiro e Turvo-PR. Nos ensaios foram utilizados extratos fenólicos dos méis, obtidos por meio da utilização da resina Amberlite® XAD®2, bem como méis brutos in natura. Os extratos apresentaram conteúdo de compostos fenólicos significativo, sendo o melato (MO5), de General Carneiro, o de maior teor (117,68± 4,40 mg EAG/g). Para as análises de sequestro das espécies reativas de oxigênio e nitrogênio, os extratos fenólicos foram sempre superiores aos méis brutos in natura. Os extratos fenólicos, de maneira geral, apresentaram alta capacidade de sequestro para o radical peroxila (ROOo), ácido hipocloroso (HOCl) e óxido nítrico (NOo). Em relação aos melatos, o extrato MO7 apresentou alta capacidade para o sequestro do HOCl (EC50= 4,83 ± 0,13 ?g/mL), enquanto que o MO5 foi melhor para o sequestro do NOo (EC50=2,16 ± 0,18 ?g/mL). Pelo método HPLC-ABTS on-line foi possível identificar e quantificar a contribuição para a atividade antioxidante do ácido ferúlico no extrato (MO1) e do flavonoide kanferol na amostra (MO4). O ácido ascórbico foi identificado e quantificado por HPLC somente nos melatos (MO3, MO5 e MO7). Pela técnica de LC-MS/MS foram identificados a presença dos seguintes compostos fenólicos: ácido caféico, rutina e hesperidina em todos os extratos. A análise de compostos voláteis por SPME-CG/EM mostrou a presença de dois compostos, encontrados apenas nos melatos, que foram o terpineno-4-ol, que possui ação antifúngica, antiparasitológica e anti-inflamatória; e o 3,4-dimetil-1-deceno, podendo assim serem utilizados como marcadores químicos destes méis. O conhecimento da composição química destes méis, bem como a composição fenólica bioativa, contribui para o fornecimento de antioxidantes naturais para a dieta, atenuando assim os efeitos negativos dos radicais livres / Brazil has a great potential to explore beekeeping due to its vast territory and diversified flora, what allows different varieties of honeys with unique characteristics. Parana state is one of the largest honey producers and the investment in production that meets the most demanding markets stimulated the organic honey production. The knowledge since early in history regarding the beneficial health effects promoted by honey is stimulating the scientific research of this noble food. Thus, this paper aimed to study certified Brazilian organic honeys (MO) in order to determine the phenolic and volatile profiles, and also the evaluation of radical scavenging capacity against Nitrogen and Oxygen Reactive Species (RNS and ROS, respectively). The honeys were collected from apiaries from beekeepers with the organic certification from two municipalities of southern Parana, General Carneiro and Turvo, PR. In the essays, phenolic extracts were obtained from honeys by using Amberlite® XAD®2 resin, as well as crude in natura honeys. The extracts showed a significant content in phenolic compounds, with honeydew (MO5), from General Carneiro, showing the highest content (117,68 ± 4,40 mg AGE/g). For the analyzes to determine the radical scavenging capacity against RNS and ROS, the phenolic extracts always showed up superior results in comparison to crude in natura honeys. Phenolic extracts showed, in general, great capacity to scavenge peroxyl radical (ROOo), hypochlorous acid (HOCl) and nitric oxide (NOo). In relation to honeydews MO7 extract showed the highest capacity to scavenge HOCl (IC50= 4,83 ± 0,13 ?g/mL) while MO5 was the sample with better capacity to scavenge NOo (IC50=2,16 ± 0,18 ?g/mL). By using HPLC-ABTS on-line method it was possible to identify and to quantify the ferulic acid in MO1 extract, a compound with an important contribution to the antioxidant activity of this sample, as well as the flavonoid kaempferol in MO4 sample. Ascorbic acid was identified and quantified by HPLC only in the honeydew samples (MO3, MO5 and MO7). The analyzes developed by LC-MS/MS techniques indicated the presence of the phenolic compounds caffeic acid, rutin and hesperidin in all the extracts. The analysis of volatile substances developed by SPME-GC/MS promoted the identification of two compounds found only in the honeydew samples. The compounds were the terpinen-4-ol, which has antifungal, antiparasitological and anti-inflammatory activities; and 3,4-dimethyl-1-decene. Both compounds can be used as chemical markers of these honeys. The knowledge of the chemical composition of the studied honeys, as well as their bioactive phenolic composition, contributes to supply natural antioxidants to human diet, thus attenuating the negative effects of free radicals

Caracterização das substâncias húmicas extraídas do solo do manguezal de Pai Matos (Cananéia, SP, BR) e de marismas da Espanha (Galícia e Valência) / Characterization of the humic substances extracted from the soil of the Pai Matos mangrove forest (Cananéia, SP, BR) and Spanish salt marshes (Galicia and Valencia)

Os objetivos deste estudo foram: 1) avaliar por meio da análise elementar (C, N, H, S, O), Ressonância Paramagnética Eletrônica (RPE), e Espectroscopia de Infravermelho com Transformada de Fourier (FTIR) às características dos ácidos húmicos (AHs) extraídos do solo do manguezal de Pai Matos (Cananéia-SP-BR) sob diferentes tipos de vegetação (Rhizophora mangle, Avicennia schaueriana, Laguncularia racemosa, Spartina alterniflora) e condições de redução, bem como às características dos AHs extraídos do solo de marismas espanholas também sob diferentes tipos de vegetação (Galícia: Spartina maritima, Juncus maritimus e Phragmites australis; Valência: Phragmites australis e Scirpus maritimus) e condições de redução; e 2) avaliar por meio da Pirólise-associada à cromatografia gasosa/espectrometria de massas (Pi-CG/EM) às características da matéria orgânica (MO) extraída do solo destes ecossistemas sob os mesmos tipos de vegetação e condições e redução. O carbono orgânico total, o nitrogênio total, o enxofre total e os valores de Eh e pH dos solos estudados também foram analisados. Os resultados mostraram que o ambiente predominantemente redutor dos solos estudados ocasionou uma maior incorporação de N e S e uma menor concentração de radicais livres semiquinona (CRLS) em relação aos AHs derivados de solos de ecossistemas terrestres, sugerindo desta forma, a presença de uma matéria orgânica (MO) menos humificada no solo das áreas de estudo; entretanto, somente os AHs extraídos do solo do manguezal apresentaram uma maior razão atômica H/C e O/C em relação aos AHs derivados de solos terrestres. A CRLS também foi maior nos AHs das camadas subsuperficiais dos solos estudados, sugerindo que o ambiente mais anóxico ocasionou um maior grau de humificação da MOS destas camadas, entretanto, como os manguezais e as marismas são ambientes de sedimentação, a contribuição de uma matéria orgânica mais humificada de origem terrestre previamente depositada nas áreas de estudo também pode ter ocasionado este resultado. Entre as diferentes vegetações estudadas, foi observada uma maior relação C/N e CRLS dos AHs extraídos do solo sob Rhizophora e Avicennia (manguezal) e uma maior relação C/N dos AHs extraídos do solo da marisma de Valência-ES, sugerindo uma maior recalcitrância da MOS sob estas vegetações. Os espectros de FTIR não permitiram verificar variações significativas na composição química e estrutural dos AHs obtidos dos solos sob as diferentes vegetações e condições de redução; entretanto, observo-se através da Pi-CG/EM uma maior concentração de compostos lignificados na MO extraída do solo sob Avicennia e Rhizophora (manguezal) e da marisma de Valência-ES, também sugerindo uma maior recalcitrância e menor humificação da MOS destas áreas de estudo. Além disso, observou-se que a maior decomposição da MOS ocasionou o decréscimo no conteúdo de lignina com o acréscimo relativo de compostos alifáticos; o acréscimo dos compostos alifáticos de cadeia curta em detrimento ao acúmulo dos de cadeia longa; a perda da dominância dos alcanos/alquenos de cadeias ímpares em relação aos de cadeias pares; e o acréscimo de compostos orgânicos de origem microbiana nos solos estudados. / The goals of this study were: 1) evaluate by elemental analysis (C, N, H, S, O), Electronic Paramagnetic Resonance (EPR), and Fourier transformed infrared spectroscopy (FTIR) the characteristics of the humic acids (HA) extracted from the soil of Pai Matos mangrove forest (Cananéia, SP, BR) under different vegetation types (Rhizophora mangle, Avicennia schaueriana, Laguncularia racemosa, Spartina alterniflora) and soil reduction conditions, as well as the HA extracted from Spanish salt marsh soils (Galícia and Valência) under different vegetation types (Galicia: Spartina maritima, Juncus maritimus, and Phragmites australis; Valencia: Phragmites australis e Scirpus maritimus) and soil reduction conditions; and 2) evaluate by Pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS) technique the characteristics of the SOM extracted from the these study areas under the same vegetation types and reduction condition. The total organic carbon, the total nitrogen, the total sulfur, and the soil pH and Eh in the field also were analyzed. The results showed that the predominantly anoxic soil condition caused the higher N and S content in the HA of both study areas than the HA derived from terrestrial soils and marine environments, and that the studied HA also showed a lower semiquinone-type free radical (SFR) content than terrestrial soils, thus suggesting a lower humification degree of the SOM derived from the study areas; however, only the HA derived from de mangrove forest soil showed a higher H/C and O/C atomic ratio than the HA derived from the terrestrial soils and marine environments. In addition, the SFR content also was lower in the surface soil layers, suggesting the input of a fresh and labile plant-derived organic matter as responsible by the lower humification degree of the SOM derived from these layers; however, as mangroves and salt marshes are sedimentation environments, it is likely that besides the more anoxic soil condition, a more humified organic matter (OM) derived from terrestrial soils also may cause the highest SFR content observed in the subsurface soil layers. Among the different vegetation types, also was observed a higher C/N ratio of the soil and HA derived from Valencian salt marsh, as well as both a higher C/N ratio and SFR content in the HA extracted from the soil under Rizhophora and Avicennia (mangrove), suggesting a more recalcitrant SOM in these sites. The FTIR spectras did not show significant variation among vegetation types and soil depths, and finally, by the Py-GC/MS technique was observed that the OM derived from the Valencian salt marsh soils and from the soil under Avicennia and Rhizophora (mangrove) showed a more recalcitrant SOM (higher lignin content) than the SOM derived from the soil under the other vegetation types studied. In addition, it was also observed that the SOM decomposition caused a decrease in lignin contents and a relative increase in aliphatics; an increase in short-chain aliphatics at the expense of longer ones; a loss of odd-over-even dominance in the n-alkanes and n-alkenes; and an increase in microbial products in the studied soils.
