Determination of the time in which the sperm is still viable for preservation represents a benefit for future research and use in animal-assisted breeding programs. In addition, the sperm cryopreservation process results in the production of radical free by the metabolism of the cell causing damage. The objectives of this study were: 1) to determine the viable time interval for cryopreservation of the epididymis tail sperm in cattle, 2) to test the addition of two antioxidants in the freezing media, 3) to determine the viability of semen samples from the tail of the epididymis in the production of embryos by in vitro fertilization. To guarantee the quality of freezing, the parameters: motility, vigor and sperm pathology in the pre-freezing were analyzed. After these assessments the material was divided into three treatments: tris-egg yolk diluent (TTG), Tris-egg yolk diluent catalase (TCAT) and tris-egg yolk diluent trolox (TTROL) and frozen in determined curve by pre-freezing automated process. After thawing, the motility and vigor were evaluated using CASA. Analysis of plasma membrane integrity and acrosome, test resistance thermometer (TTR), thiobarbituric acid reactive substances (TBARS), oxidation of 3,3 'diaminobenzidine (DAB) and in vitro fertilization (IVF) were also evaluated. According to the movement patterns analyzed using CASA, the analysis of data generated by the study allow us to conclude the characteristics of sperm in each treatment and it change on the different times of cooling (0h, 24h, 48h and 72h). In the assessment of integrity (INT) and injured sperm (LS), no statistical differences between the moments or between treatments were observed. Rates of cleavage D7 and D8 were statistically different, at 72 hours the trolox showed negative action. The results shown indicate that this procedure is important for conservation of animals of high genetic value or animals endangered that die suddenly. Further studies should be performed testing associations of antioxidants directly in the middle of fertilization and embryo culture, especially in production systems of embryos with high oxygen tension, as used in this experiment. xxii / O interesse pela preservação de espécies em risco de extinção tem aumentado a atenção pela criopreservação de sêmen da cauda do epidídimo de animais que morrem subitamente. A determinação do momento até quando os espermatozóides estão viáveis para preservação representa um benefício para pesquisas futuras e sua utilização em programas de reprodução animal assistida. Além disso, o processo de criopreservação dos espermatozóides resulta na produção de radicais livres pelo próprio metabolismo desta célula e causando danos. Os objetivos deste estudo foram: 1) determinar o intervalo de viabilidade de resfriamento de espermatozóides da cauda do epidídimo de bovinos, 2) testar o efeito da adição de dois antioxidantes nos meios de congelação, 3) determinar a viabilidade das amostras de sêmen da cauda do epidídimo na produção de embriões por fertilização in vitro. Para garantia da qualidade da congelação foram realizadas análises de motilidade, vigor, patologia espermática, no momento pré-congelação. Após estas avaliações o material foi dividido em três tratamentos: diluente tris-gema-glicerol (TTG), diluente tris-gema-glicerol com catalase (TCAT) e diluente tris-gema-glicerol com trolox (TTROL) e congelados em curva pré-determinada por processo de congelação automatizada. Após a descongelação foram avaliadas a motilidade e vigor com auxílio do CASA, análises de integridade de membrana plasmática e acrossoma, teste de termorresistência (TTR), substâncias reativas ao ácido tiobarbitúrico (TBARs), oxidação da 3,3’ diaminobenzidina (DAB) e fertilização in vitro (FIV). De acordo com os padrões de movimentos observados pelo CASA, a análise dos dados gerados pelo estudo nos possibilitam concluir as características dos espermatozóides em cada tratamento e como se comportaram nos momentos de resfriamento logo após o abate, 24 horas, 48 horas e 72 horas após resfriamento à 5ºC ( 0h, 24h, 48h e 72h). O uso dos antioxidantes não garantiu melhor proteção e nem auxílio na funcionalidade dos espermatozóides do epidídimo nos diferentes períodos de resfriamento. lulas imóveis) não houve diferenças estatísticas. Nas taxas de clivagem D7 e D8 foram observadas diferenças estatísticas, o α-tocoferol no momento 72 h demonstrou ação negativa. Novos estudos devem ser realizados testando associações de antioxidantes ou o uso de antioxidantes diretamente no meio de fecundação e cultivo embrionário, especialmente em sistemas de produção de embriões com alta tensão de oxigênio, como o utilizado neste experimento.

Impactos socioambientais locais decorrentes de acesso aos recursos genéticos para fins comerciais / Socio-environmental impacts resulting from the access to genetic resources for commercial purposes

O presente estudo tem como objetivo analisar através de um estudo de caso os impactos socioambientais locais decorrentes de dois acordos comerciais para o acesso aos recursos genéticos e participação nos benefícios entre uma empresa de cosméticos e um grupo de pequenos produtores rurais da região do Vale do Ribeira-SP. Embora acordos entre empresas privadas e comunidades locais para a utilização comercial de componentes da biodiversidade venham sendo estimulados atualmente como forma de conciliar o uso sustentável dos recursos naturais com desenvolvimento local, alguns estudos têm demonstrado que, além de benefícios, tais acordos podem ter efeitos socioambientais negativos sobre as condições locais em que são estabelecidos. No Brasil, desde 2001, vigora a Medida Provisória 2.186-16/01 que regulamenta a Convenção sobre Diversidade Biológica (CDB) no país instituindo normas para o acesso a componentes do patrimônio genético e aos conhecimentos tradicionais associados. Tais normas baseiam-se, fundamentalmente, na anuência prévia e na repartição de benefícios junto a seus provedores. Apesar da existência desses instrumentos jurídicos, ainda pouco se sabe a respeito de sua aplicação prática e de seus impactos locais. A partir de técnicas qualitativas (entrevistas semiestruturadas, observação participante e análise documental) ao longo de um ano foram levantados dados a respeito do estabelecimento de dois acordos para a utilização de recursos genéticos para fins comerciais e de seus efeitos sobre: (i) a geração e distribuição de renda; (ii) a infraestrutura local e acesso à tecnologia; (iii) a organização social e; (iv) o uso dos recursos naturais locais (i.e. solo, recursos vegetais e hídricos). Os resultados mostraram impactos positivos diretos sobre a geração de renda e sobre o acesso à tecnologia e infraestrutura local. A respeito da organização social do grupo, a falta de uma plataforma para ação coletiva e seu reconhecimento jurídico são obstáculos à maior participação e à sua autonomia. Os impactos diretos e indiretos sobre uso dos recursos naturais observados foram: transformações no uso do solo através do aumento de espécies cultivadas, da redução de áreas com cultivos ou criações de animais para subsitência e da mudança no manejo e nos insumos empregados e; transformações no uso dos recursos vegetais através do aumento da extração da espécie alvo, no primeiro acordo, e do consumo de madeira para lenha, no segundo. Embora a realização de acordos comerciais para a utilização da biodiversidade tenha apresentado potencial para contribuir para o desenvolvimento local e para a conservação da biodiversidade, a realização de novos estudos em diferentes contextos se faz necessária para um maior entendimento de correlações e possíveis trade-offs entre fatores sociais e ambientais. / The aim of this study is to analyze local socio-environmental impacts resulting from two commercial agreements for the access to genetic resources and benefit sharing between a cosmetic company and a group of smallholders from Vale do Ribeira-SP region. Although agreements between private companies and local communities for the commercial use of biodiversity components have been encouraged today as a way to conciliate the sustainable use of natural assets with local development, some studies have demonstrate that beyond of its benefits, these agreements may have negative socio-environmental effects on local conditions where it was established. In Brazil, since 2001, the Provisional Executive Order 2.186-16/01 which regulates the Convention of Biological Diversity in country establishing rules for the access to genetic resources and traditional knowledge was released. These rules are fundamentally based on the prior acceptance and benefit sharing along with its providers. Despite the existence of those juridical tools, yet little is known about its practice enforcement and its local impacts. Qualitative techniques (semi-structured interviews, participant observation and documental analysis) were adopted to gather data about the establishment of two commercial agreements for the use of genetic resources and its effects on: (i) income generation and distribution; (ii) local infrastructure and access to technology; (iii) social organization and; (iv) use of local natural assets (i.e. soil, water and vegetable resources). The results show positive direct impacts on income generating, local infrastructure and access to technology. About the social organization of the involved group, the lack of one platform for collective action just as legal acknowledgement are both drawbacks to effective participation of its members and their greater autonomy. As regards environmental impacts it was observed: transformations in land use by increase of cultivating species, reduction of areas occupied by subsistence cultures or animal husbandry, changes in applied handling and raw materials and; transformations in the use of vegetable resources by the raise of target specie, in the first agreement, and by the increase of firewood consume, in second one. On the one hand, the realization of commercial agreements shows oneself to be a possible manner to contribute to local development and to biodiversity conservation. In the other hand, we still need new studies in different contexts for a better understanding of the correlations and trade-offs between social and environmental factors.

Cruzabilidade entre espécies silvestres de Arachis visando à introgressão de genes de resistência a doenças no amendoim cultivado. / Crossability among wild species of Arachis for introgression of disease resistance genes into cultivated groundnut.

O amendoim (Arachis hypogaea) é a quarta oleaginosa mais consumida no mundo. O Brasil produziu, em 2002, aproximadamente 190 mil toneladas, sendo que 80% da área plantada situa-se no Estado de São Paulo. O principal problema da cultura neste Estado e no mundo são as doenças fúngicas de parte aérea. Diversas espécies do gênero Arachis são consideradas resistentes a várias pragas e doenças. Os objetivos dessa pesquisa foram: 1) identificar espécies silvestres pertencentes à Secção Arachis resistentes à mancha castanha (Cercospora arachidicola), mancha preta (Cercosporidium personatum) e ferrugem (Puccinia arachidis); 2) realizar cruzamentos entre espécies de genoma "A" e "B" resistentes a, pelo menos, uma doença fúngica; 3) duplicar os cromossomos dos híbridos estéreis; 4) realizar cruzamentos entre A. hypogaea e os anfidiplóides sintéticos; 5) obter a geração F1 que tivesse 50% do genoma do amendoim cultivado, 50% do genoma das espécies silvestres. Os experimentos foram conduzidos, em condições de telado, no Departamento de Genética da Escola Superior de Agricultura "Luiz de Queiroz". Para a identificação de genótipos resistentes às doenças fúngicas, utilizou-se a técnica de folhas destacadas, em laboratório, com inoculação artificial, condições controladas de temperatura a 25°C e luz alternada (10h luz). Os cruzamentos entre espécies silvestres, de genoma "B" com as de genoma "A", resistentes a pelo menos uma doença foram realizados em condições de telado, com emasculação realizada ao final da tarde e polinização na manhã do dia seguinte. A duplicação de cromossomos de células somáticas de híbridos com genoma "AB", foi obtida mediante o tratamento de estacas com colchicina a 0,2%, por aproximadamente 12h, em condições de luz e com temperatura entre 25-30°C. Os cruzamentos entre A. hypogaea e os anfidiplóides sintéticos foram realizados em condições de telado, na Embrapa Recursos Genéticos e Biotecnologia. Observou-se que várias espécies silvestres foram altamente resistentes a, pelo menos, uma das três doenças estudadas. Foi possível selecionar, como genitores masculinos, 12 acessos de genoma "A" e, como femininos, seis acessos de genoma "B". A partir de 26 cruzamentos distintos (3633 hibridações), foi possível obter 17 híbridos interespecíficos distintos com genoma "AB". Após o tratamento com colchicina de todos os 17 tipos de híbridos, foram obtidas cinco combinações híbridas que produziram flores tetraplóides (A. hoehnei x A. helodes, A. ipaënsis x A. duranensis, A. hoehnei x A. cardenasii, A. aff. magna x A. villosa, A. aff. magna x A. aff. diogoi). Foram realizados 21 cruzamentos entre A. hypogaea e os anfidiplóides sintéticos. Foram obtidos 13 tipos de híbridos: A. hypogaea (cvs. IAC-Tatu-ST, Br-1, IAC-Caiapó, IAC-Runner) x [A. hoehnei x A. cardenasii]; A. hypogaea cv. Br-1 x [A. aff. magna x A. villosa] ; A. hypogaea (acessos V 12548, V 12549, Mdi 1560, Mdi 1538, cvs. BR-1, IAC-Tatu-ST, IAC-Caiapó, IAC-Runner) x [A. ipaënsis x A. duranensis]. Os resultados obtidos confirmam que é possível a introgressão de genes de resistência a partir de espécies silvestres no amendoim cultivado, via cruzamentos, expandindo-se a lista de espécies silvestres utilizadas e ampliando a variabilidade genética liberada para seleção nos programas de melhoramento de amendoim. / Groundnut (Arachis hypogaea) is one of the most important oil species in the world. In 2002, Brazil produced about 190 thousand tonelades, with 80 per cent of cultivated area concentrated in the State of São Paulo. The main problem in crop management in this State and in many other growing areas in the world is represented by fungal diseases. Several species of the genus Arachis are considered resistant to main pests and diseases. This research was developed to take advantage of the genetic variability present in the Arachis genus, and has the following objectives: 1) to identify accessions of wild species belonging to Section Arachis that are resistant to early leaf spot (Cercospora arachidicola), late leaf spot (Cercosporidium personatum) and rust (Puccinia arachidis); 2) to cross resistant species having B and A genomes; 3) to duplicate chromosomes of sterile hybrids; 4) to cross A. hypogaea with synthetic amphiploids; 5) to obtain an F1 generation with 50% of the cultivated groundnut genome and 50% of wild species genomes. Experiments were developed under greenhouse conditions, in the Department of Genetics, faculty of Agriculture "Luiz of Queiroz". Detached leaves technique was used, in laboratory conditions, with artificial inoculation, controlled temperature of 25°C and alternate light (10h light) for the identification of genotypes resistant to fungal diseases. Crosses among wild resistant species with B and A genome genotypes were carried out in greenhouse conditions. Emasculation were hand-made at the end of the afternoon and pollination was made in the following morning. Chromosome duplication of somatic cells in AB genome interspecific hybrids was obtained by treating cuttings with 0.2% colchicine for approximately 12h, in daylight conditions, maintaining the temperature in range of 25-30°C. Crosses between A. hypogaea and synthetic amphyploids were done in greenhouse conditions, at Embrapa Genetic Resources and Biotechnology. It was observed that several wild species were highly resistant to one or more of the studied diseases. Selection of 12 A genome accessions for use as male parents was possible as well as six B genome species as female parents. From 26 different combinations, it was possible to obtain 17 interspecific AB genome hybrids. After colchicine treatment of all 17 hybrid types, five hybrid combinations that produced tetraploid flowers were obtained (A. hoehnei x A. helodes, A. ipaënsis x A. duranensis, A. hoehnei x A. cardenasii, A. aff. magna x A. villosa, A. aff. magna x A. aff. diogoi). Twenty-one different crosses were done between A. hypogaea and synthetic amphyploids. Thirteen different hybrid types were obtained: A. hypogaea (cvs. IAC-Tatu-ST, Br-1, IAC-Caiapó, IAC-Runner) x [A. hoehnei x A. cardenasii]; A. hypogaea cv. BR-1 x [A. aff. magna x A. villosa]; A. hypogaea (accessions V 12548, V 12549, Mdi 1560, Mdi 1538, cvs. Br-1, IAC-Tatu-ST, IACCaiapó, IAC-Runner) x [A. ipaënsis x A. duranensis]. Results confirmed the possibility of introgression of resistance genes from wild species into cultivated groundnut, by manual crosses, increasing the number of wild species used, and thus to enhance the genetic variability released for applying selection in breeding groundnut programs.

The Nagoya protocol: a possible solution to the protection of traditional knowledge in biodiverse societies of Africa

<p>There is a growing interplay of competing realities facing the international community in the general areas of innovation, technological advancement and overall economic development. The highly industrialised wealthy nations, largely located on the Northern hemisphere are on the one hand undoubtedly at the forefront in global research, technology and infrastructure development. The developing and least developed countries on the other hand are mostly situated on the Southern hemisphere. They are not as wealthy or technologically advanced as their&nbsp / Northern counterparts, but are naturally endowed with unique variations of plant, animal and micro-organism species occurring in natural ecosystems, as well as the traditional knowledge on&nbsp / how to use these unique species. This knowledge has been adjudged to be responsible for the sustainable maintenance of the earth&rsquo / s biodiversity. Increasing exploitation of biodiversity,&nbsp / spurred on by the competing realities identified above, has left the earth in a present state of alarm with respect to the uncontrolled loss of biodiversity. The traditional knowledge of local&nbsp / peoples has significantly offered leads to research institutes from the North in developing major advancements in drugs, cosmetics and agriculture. Little or no compensation has however been seen to go back to the indigenous&nbsp / communities and countries that provide resources, and indicate various possibilities through their traditional knowledge to the use of such resources. Efforts by some biodiversity rich countries to&nbsp / ddress this trend through legislation developed in accordance with the principles of the Convention on Biological Diversity have been frustrated due to the inability to enforce their domestic laws outside their borders. Theft of genetic resources and its associated traditional knowledge&nbsp / from such countries has therefore remained a major challenge. Against this backdrop, and on the&nbsp / insistence of biodiversity-rich developing countries, an international regime on access and benefit sharing was negotiated and its final text adopted in 2010. This international regime is as&nbsp / contained in the Nagoya Protocol. This research sets out to examine whether the Nagoya Protocol offers a final solution to the protection of traditional knowledge associated with biodiversity in&nbsp / biodiverse countries. It further examines the importance of domestic legislation in achieving the objectives of the Protocol. The research has been tailored to African biodiverse countries, and&nbsp / seeks these answers within the context of Africa.<br /> &nbsp / </p>

La pertinence de l’obligation de divulguer l’origine des ressources génétiques et des savoirs traditionnels dans les demandes de brevets

Le développement fulgurant noté dans le domaine des biotechnologies peut être attribué, sinon essentiellement du moins partiellement, à l’utilisation des ressources génétiques (RG) et des savoirs traditionnels (ST) acquis sur ces ressources. Ces ressources et ces savoirs sont, notamment, utilisés dans le cadre d’inventions biotechnologiques qui peuvent s’avérer concluantes et faire l’objet de demande de protection par brevet. Ce développement ne s’est tout de même pas réalisé sans heurts majeurs, il l’a été au prix de tumultueuses oppositions. En effet, la découverte progressive de la valeur commerciale et scientifique de telles ressources et de tels savoirs a fait naître des intérêts et attisé des rivalités qui ont fini par opposer fournisseurs et utilisateurs de ces matériels. Force est de constater que parmi leurs divergences, celle qui se rapporte au partage des avantages fait l’objet de discussions des plus âpres qui soient dans le domaine. Une solution qui a été, aussi, envisagée a porté sur les régimes d’accès et de partage des avantages. Ce partage des avantages, les pays fournisseurs espèrent le réaliser par le biais de l’obligation de divulguer l’origine des RG et des ST dans les demandes de brevets. L’application d’une telle exigence connaît des limites en ce sens qu’elle est d’application territoriale. C’est sur la base d’un tel constat que les pays fournisseurs envisagent d’en faire une obligation reconnue et applicable à un niveau international. Dans le cadre de cette étude, nous essaierons de démontrer que l’obligation de divulguer l’origine des RG et des ST dans les demandes de brevets, telle qu’elle est actuellement appliquée, ne constitue pas un moyen pertinent qui permettrait d’en arriver à un partage juste et équitable des avantages. / The rapid development in the field of biotechnology can be attributed to a large degree to the innovative use of genetic resources (GR), a significant portion of which were based on traditional knowledge (TK). The biotechnological inventions resulting from these resources and knowledge are, for the most part, subject to patent protection. This legal protection is designed to allow creators of innovative inventions the possibility to recoup their investment by limiting the use of the resulting creation by people other than the inventor and his assignees. Indeed, the gradual recognition of the scientific and commercial value of such resources and associated knowledge has raised interest and fuelled rivalries that eventually led to conflict over the use and trade of such products between user and producing States. A possible solution to resolve this conflict is the use of access and benefit-sharing agreements. One requirement proposed by producing States is an obligation to disclose the origin of GR and TK in patent applications. However, since the issues in this area, generally, exceed the national sphere, producing countries are attempting to make this requirement recognized at an international level. Accordingly the most effective and efficient means of doing so would be via the Agreement on Trade Related Aspects of Intellectual Property Rights (TRIPS) in the World Trade Organization WTO. In this thesis, we will demonstrate that the obligation to disclose the origin of GR and TK in patent applications does not constitute an appropriate means that would lead to a fair and equitable sharing of benefits arising out of their use.

This thesis explores the location and nature of the power that is deepening and broadening the revolution in modern biotechnologies, and which is inherent in the global governance of one type of genetic resource — plant genetic resources. Plant genetic resources are of increasing importance within the global political economy and ecology because of the power/knowledge networks contributing to, and responding to developments in the biotechnology sector, and concerned with the rampant erosion of biological diversity. The thesis argues that transnational norms, values and knowledge are important aspects of power. Discursive power, and particularly the power inherent in discourses of sustainable development, security and human rights, are a central focus of the thesis. The thesis challenges realist, neo-realist and other structural analyses of power which focus on relative distributions of power at the level of individual states or at the global level.
