Fonction des aquaporines de la membrane plasmique dans les mouvements stomatiques chez Arabidopsis thaliana / Function of plasma membrane aquaporins in stomatal movements in Arabidopsis thaliana

Les mouvements stomatiques sont dus à des changements importants du volume des cellules de garde. Un rôle des canaux hydriques membranaires (aquaporines) dans ces processus a été proposé mais n'a jamais été démontré. Des analyses transcriptomiques indiquent que plusieurs aquaporines de la membrane plasmique (PIPs), dont AtPIP1;2 et AtPIP2;1, sont exprimées dans la cellule de garde. Dans ce travail, nous avons étudié la fonction de ces deux aquaporines dans les mouvements stomatiques. Les stomates des mutants d'insertion pip1;2 et pip2;1 montrent une ouverture normale à la lumière et aux faibles concentrations en CO2, une fermeture normale en réponse à l'obscurité et aux fortes concentrations en CO2, mais sont quasiment insensibles à la fermeture induite par l'ABA. Le rôle d'AtPIP1;2 et d'AtPIP2;1 dans le transport d'eau a été testé en mesurant la perméabilité hydrique de protoplastes (Pf) à l'obscurité, à la lumière ou à la lumière et en présence d'ABA. Le Pf des protoplastes de cellules de garde de pip2;1 est réduit de façon significative, spécifiquement en présence d'ABA. Comme la production extracellulaire de peroxyde d'hydrogène (H2O2) est essentielle pour la signalisation intracellulaire de l'ABA, nous avons également étudié la possibilité qu'AtPIP1;2 et AtPIP2;1 facilitent la diffusion de l'H2O2 à l'intérieur des cellules de garde en réponse à l'ABA. La cinétique d'accumulation d'espèces réactives de l'oxygène est abolie dans les cellules de garde chez pip2;1, mais pas chez pip1;2. Par ailleurs, dans d'autres types d'approches, nous avons étudié la régulation de ces deux aquaporines après production en levure Pichia pastoris et reconstitution fonctionnelle en protéoliposomes. Une analyse structure fonction a été réalisée avec succès pour AtPIP2;1. Elle montre que le résidu His199 est impliqué dans l'inhibition de l'activité de cette isoforme par les protons et les cations divalents. A l'inverse, le résidu Arg124 rend AtPIP2;1 complètement insensible au calcium, mais n'altère pas sa sensibilité aux protons. La surexpression de formes dérégulées d'AtPIP1;2 et d'AtPIP2;1 dans, respectivement, les mutants pip1;2 et pip2;1, indique que les régulations de l'activité intrinsèque de ces isoformes sont importantes pour leurs fonctions stomatiques. L'ensemble des résultats suggère qu'AtPIP1;2 et AtPIP2;1 ont des fonctions majeures et distinctes dans le transport d'eau et d'H2O2 lors de la fermeture stomatique induite par l'ABA chez Arabidopsis thaliana. / Stomatal movements are mediated by drastic changes in guard cell volume. A role in these processes for water channel proteins named aquaporins has been proposed but not demonstrated. Transcriptome analyses have indicated that several plasma membrane aquaporins (PIPs) including AtPIP1;2 and AtPIP2;1 are expressed in Arabidopsis thaliana guard cells. In the present work, we investigated the function of these two aquaporins in stomatal movements. The stomata of pip1;2 and pip2;1 knock-out mutants showed a normal opening response to light and low CO2, a normal closing response to darkness and high CO2, but were almost insensitive to abscisic acid (ABA)-induced stomatal closure. A direct role of AtPIP1;2 and AtPIP2;1 in water transport was investigated by measurement of guard cell protoplast water permeability (Pf) under darkness, light and light with ABA. The Pf of pip2;1 guard cell protoplasts was significantly reduced, specifically in the presence of ABA. As extracellular hydrogen peroxyde (H2O2) production is essential for intracellular ABA signalling, we also investigated the possibility that AtPIP1;2 and AtPIP2;1 facilitate the diffusion of H2O2 through the guard cell plasma membrane. Time-dependent accumulation of reactive oxygen species in response to ABA was abolished in pip2;1 but not pip1;2 guard cells. In another type of approach, the regulation of the two aquaporins was investigated after production in the yeast Pichia pastoris and functional reconstitution in proteoliposomes. Structure-function analysis was achieved in the case of AtPIP2;1, showing that cytoplasmic residue His199 is involved in both divalent-cation- and proton-mediated gating. In contrast, mutation of Arg124 rendered AtPIP2;1 largely insensitive to calcium while remaining fully sensitive to protons. Over-expression of deregulated forms of AtPIP1;2 and AtPIP2;1 in pip1;2 and pip2;1, respectively, indicated that the gating regulation of these proteins may play an important role in their guard cells functions. Altogether, our results suggest that AtPIP2;1, and to a lesser extent AtPIP1;2, play important and distinct roles in water and H2O2 fluxes during ABA-induced stomatal closure in Arabidopsis thaliana.

The thesis is primarily focusing on wild game and its ownership, rights and duties of hunting guards, hunting management and importance of hunting management plan and issues relating to overpopulation of specific types of wild cloven-hoofed game which is connected with a liability for damage caused by game.

It is a sometimes humorous, but often tragic attempt to make peace and to make sense of the places, comrades and enemies that graced and plagued a career. First names and nicknames were used to protect the privacy of a few and render respect for the surviving children of a fallen two.

From Containing Communism to Fighting Floods: The Louisiana Army National Guard in the Cold War, 1946-1965

In the decades following World War II, the Louisiana National Guard evolved due to world, national, and local events. In response to the United States’ Cold War policies to contain Communism, the Guard expanded, professionalized, and was occasionally called to federal service. In conjunction with Cold War fears of external attack and internal subversion, a civil defense mission brought coordination between federal, state and local response agencies. Despite the lack of large scale war service or an attack on the U.S. homeland , the skills and responsibilities acquired by the Louisiana Guard during this time period resulted in an enhanced ability to respond to Louisiana’s biggest practical threat, i.e. natural disasters.

A presente dissertação trata das narrativas de experiências pessoais de velhos sambistas. O corpus documental é constituído de sambas e narrativas feitos por participantes dos conjuntos musicais chamados Velha Guarda da Portela e Velha Guarda do Império Serrano. As histórias de vida de sambistas e pastoras, produzidas adotando os procedimentos da história oral, expõem temas polêmicos afeitos à tradição e à modernização dos desfiles. Sambas gravados por essas duas Velhas Guardas funcionam como apoio e complemento, tomados como documentos que permitem acessar o discurso mnemônico vinculado a um discurso dominante sobre o samba no contexto cultural carioca dos anos 2000. A partir das vivências e das visões dessas pessoas sobre a evolução das escolas de samba, procura-se entender o papel dos grupos aos quais pertencem, localizados simbolicamente entre as raízes e os desafios da atualização das agremiações carnavalescas. / This dissertation deals with personal experiences narrated by old sambistas. Its document corpus consists of sambas and narratives made by participants in musical ensembles called Velha Guarda da Portela and Velha Guarda do Império Serrano. The life stories of samba men and women, which were produced following oral history procedures, expose controversial issues that relate to the tradition and modernization of samba school parades. The songs that were recorded by those two samba Old Guards are regarded as complementary documents, that allow access to a mnemonic speech linked to a dominant discourse about samba in Rio de Janeiros cultural context of the 2000s. By observing the life experiences and visions of those people about the evolution of samba schools, the purpose is to understand the role of the groups to which they belong, symbolically positioned between the roots and the challenges of the adjustment of carnival groups to new times.

Design Guidelines for the use of Curbs and Curb/Guardrail Combinations Along High-Speed Roadways

"The potential hazard of using curbs on high-speed roadways has been a concern for highway designers for almost half a century. Curbs extend 75-200 mm above the road surface for appreciable distances and are located very near the edge of the traveled way, thus, they constitute a continuous hazard for motorist. Curbs are sometimes used in combination with guardrails or other roadside safety barriers. Full-scale crash testing has demonstrated that inadequate design and placement of these systems can result in vehicles vaulting, underriding or rupturing a strong-post guardrail system though the mechanisms for these failures are not well understood. For these reasons, the use of curbs has generally been discouraged on high-speed roadways. Curbs are often essential, however, because of restricted right-of-way, drainage considerations, access control, delineation and other curb functions. Thus, there is a need for nationally recognized guidelines for the design and use of curbs. The primary purpose of this study was to develop design guidelines for the use of curbs and curb-barrier combinations on roadways with operating speeds greater than 60 km/hr. The research presented herein identifies common types of curbs that can be used safely and effectively on high-speed roadways and also identifies the proper combination and placement of curbs and barriers that will allow the traffic barriers to safely contain and redirect an impacting vehicle. Finite element models of curbs and curb-guardrail systems were developed, and the finite element program, LS-DYNA, was used to investigate the event of a vehicle traversing several curb types. Finite element analysis was also used in the analysis of a vehicle impacting a number of curb-guardrail combinations. The results obtained from these analyses were synthesized with the results of previous studies, which involved full-scale crash testing, computer simulation, and other methods. The combined information was then used to develop a set of guidelines for using curbs and curb-barrier combinations on high-speed roadways."

<p>The market for security is one filled with great opportunities. The security services companies are numerous and diverse, the Swedish market is dominated by two large companies: Securitas Bevakning AB and Falck Security AB. In this study we have focused on the bigger of the two, Securitas. </p><p>This report describes and analysis the security service branch and its surroundings in an involved manner. The target group of the result is mainly people within the security branch. However, by making a wide theoretical description the person unacquainted with the business will be able to understand the concept of this interesting trade. Further, those who are familiar with this field will be enabled to broaden their minds. The aim with this project is to create a foundation upon which Securitas can further continue their work. We have not presented any direct solutions, instead we have displayed concrete problems and presented guidelines for further exploration. </p><p>The purpose of the investigation is to discover how modern technology can improve today’s guard services. Moreover, the aim is to show how a change of attitude needs to be present while developing future security services. This change can also be applied to other security services than the time-sharing guarding. </p><p>The methodology used to meet our goals was a qualitative investigation based on interviews and literary studies. In order to form a robust overview of the study area the objects were carefully selected according to set preferences. These were, among others, which function the object has in society, how long they have been customers with Securitas and what kind of business they run. </p><p>In the discussion and analysis four important areas is to be taken into consideration: The change of the market, the adaptation of the service to the needs of the customer, the adaptation of technology, and the assertion to the customer of the profound effects following these amendments. These four areas represent the foundation for the presented model. </p><p>A solid cooperation between the customer and the supplier of the security service is vital. In contemporary society the customers have become more specialised and thereby the delivering company must aim to meet the increasing demands expected of this specialised service. Therefore the demand on the competence of the guard is getting higher. Another conclusion is also that one should introduce a quality measurement system that together with a statistical basis makes reporting and the analysing of the effects of the guarding easier to the customer. </p><p>Technology is the most important corner stone in an effective alarm system. The time for an emergency call response must be reduced. Further qualities that the customer demands are that the security companies possess good knowledge of the object and its surroundings. The effectiveness in the guarding rounds and the learning time for a new object can be reduced by multimedia teaching aids. </p><p>The customer satisfaction is not likely to be long term if they cannot see the results and high quality that they ask for. This is why it will be very important to measure, calculate and point out what both the guard and the security company can contribute with to please the customer. To enhance the dialogue the guard will be presenting solutions rather than static reports, and an acting counsellor strengthen the feeling of professionalism. Any change from the customers that affects the actual risk picture has to be identified promptly by the security service company.</p>

The National Guard, the National Board for the Promotion of Rifle Practice, and the National Rifle Association: Public Institutions and the Rise of a Lobby for Private Gun Ownership

Today, the strength of the National Rifle Association (NRA) is understood by the general public and many scholars to be dependent on the Second Amendment of the U.S. Constitution, the right of individuals to own firearms. This dissertation challenges that understanding by focusing on three organizations, the NRA, the National Guard and the National Board for the Promotion of Rifle Practice (NBPRP). While each organization appears in today's world to be distinct and independent, this dissertation reveals how they garnered strength from a symbiotic relationship. The NRA was founded in 1871, originally as a marksmanship organization. The National Guard, which grew from the nation's militia, was formally established in the early twentieth century. The NBPRP was a small organization that was established in 1903 within the War Department at the encouragement of the NRA. Following passage in 1903 of legislation bringing state militia units under federal control, the newly formed National Guard became dependent on the NRA, which in turn leveraged that dependence to create a nationwide grassroots organization. The NBPRP was headed by the Assistant Secretary of War until 1916 when the position of the Director of Civilian Marksmanship was created, to be held by a U.S. Army or U.S. Marine Corps officer. The NRA acted as the surrogate of the NBPRB outside of the halls of government. At the same time, the NBRPB provided the NRA with a voice within those same halls that aided in the development of federal policy and budget positions related to firearms acquisition, competition, and training. The purpose of this dissertation is to reveal how the NRA was able to employ these two organizations to develop an exceptionally powerful grassroots organization that today is recognized as one of the most powerful special interest groups in America. Understanding how this powerful organization grew offers one perspective of how the bureaucracy that has been developed to support America's federal system of government is uniquely susceptible to special interest influence.
