Modèles de dysfonction érectile chez le rat

Des modèles de dysfonction érectile (DE) ont été développés et étudiés chez le rat. Cependant, peu de choses sont connues dans certains modèles présentant des facteurs de risque de la DE, en l’occurrence le rat soumis à une diète riche en glucose, le rat avec une restriction de croissance intra-utérine (RCIU) et le rat avec l’infusion continue de l’angiotensine (Ang) II en ce qui concerne l’évaluation de la fonction érectile in vivo. C’est ainsi que nous nous sommes proposés cette étude. La fonction érectile a été mesurée par la stimulation électrique du nerf caverneux à des cohortes de rats mâles Sprague-Dawley de différents âges comme suit: 1) des rats jeunes de 12 semaines d’âge, adultes reproducteurs à la retraite d’environ 30 semaines d’âge et des rats de 27 et 29 semaines soumis à une diète riche en glucose par l’eau de breuvage (10%) à court (1 semaine), moyen (6 semaines) et long (12 ou 13 semaines) termes; 2) des rats jeunes âgés de 12 semaines recevant une infusion soit de l’Ang II (200ng/kg/min) soit de la saline par mini-pompes osmotiques sous-cutanées pendant 1 et 2 semaines; et 3) des rats adultes âgés de 36 semaines avec une RCIU. La fonction érectile a été représentée par des courbes de variations de pression intracaverneuse (PIC) et d’aire sous la courbe (ASC) normalisées avec la pression artérielle moyenne (PAM) en réponse à la stimulation nerveuse à différents volts. À la fin des expériences, les animaux ont été sacrifiés, le sang recueilli pour le dosage de la glycémie et de l’insulinémie; le pénis, l’aorte thoracique et le cœur prélevés à des fins d’analyses protéique et histologique. La diète riche en glucose à court terme a eu un effet néfaste sur la fonction érectile chez le jeune rat mais bénéfique chez le rat adulte reproducteur; de même que pour les études à long terme, la diète riche en glucose a amélioré ou renversé la DE associée au vieillissement. Les rats avec RCIU ont exhibé une baisse très significative de la fonction érectile. Ces détériorations ou améliorations de la fonction érectile avec le glucose ou dans la RCIU ont été principalement associées à des modifications de l’expression de l’Akt, en plus d’une augmentation de l’insuline sérique dans les groupes avec le glucose. L’Ang II a entraîné une baisse de la fonction érectile, statistiquement très significative après 1 semaine et maintenue après 2 semaines de traitement, sans causer des changements morphologiques au cœur et à l’aorte thoracique. En conclusion : l’atteinte du système érectile se fait précocement dans les complications cardiovasculaires liées à l’activation du SRA, l’implication de l’Akt dans la modulation de la fonction érectile dépend de type de vieillissement et de la diète, le RCIU est un facteur de risque pour la DE. / Rat is a most rodent used for erectile function studies, and many erectile dysfunction (ED) rat models have been developed and studied. However, according to our knowledge, erectile function has never been studied in high-glucose fed rat model, intrauterine growth restriction rat model (IUGR) and continuous angiotensin (Ang) II infusion rat model. In this context, we set up this study. We mesured simultaneously mean arterial pression (MAP), intracavernosal pression (ICP), aire under the curve (AUC); and erectile function was obtained by variations of the ratio ICP/MAP and AUC/MAP in response to electrical stimulation of cavernosal nerve (0.89 – 6.50 Volts) in male Sprague-Dawley rats in different protocols as follow: 1) young (12 week old), procreator adult (~30 week old) and adult (27 and 29 week old) rats randomized and fed with high-glucose diet in water (10%) or simple water for a short (1 week), middle (6 weeks) and long (12 and 13 weeks) terms; 2) adult (36 week old) IUGR rats; 3) young (12 week old) rats randomized and received a chronic infusion of AngII (200 ng/kg/min) or saline by osmotic minipumps for 1 and 2 weeks. At the end of procedures, blood for measuring serum glucose and insulin in rats with chronic high-glucose diet was obtained, the heart was excised and left ventricle was weighed. Aorta and penis were excised, cleaned and a section of each was formalin fixed for histological studies and the rest snap frozen for protein studies. The short term high-glucose diet impacted negatively in young rats but positively in procreator adult rats; also, the erectile function was improved or reversed by chronic high-glucose diet in adult rats. We also observed a diminished erectile function in RCIU. The improvement or deterioration of erectile function was associated with changes in Akt expression, besides an increased insulin serum in rats with high-glucose diet. A diminished erectile function was observed after 1 week continuous Ang II infusion and was maintained after 2 week infusion without detectable structural changes on heart and aorta. In conclusion: this study suggests that erectile system is a precoce cible of cardiovascular complications of renin-angiotensin system. High-glucose diet deteriores erectile function but maskes the ageing induced-ED, RCIU is associated with ED in later life. Akt involvement in erectile function modulation depends on aging type and the diet.

Flexibilidade cognitiva em roedores adultos expostos à restrição de crescimento intrauterino : investigação do papel do cuidado materno e da resposta neuroquímica à recompensa

A restrição do crescimento intrauterino (RCIU) está associada com preferências alimentares alteradas, assim como com um aumento do risco de obesidade na vida adulta. Enquanto os desfechos metabólicos adversos têm sido de certa forma bem caracterizados nesta condição, seus déficits neurocomportamentais e alterações em áreas específicas do sistema nervoso têm recebido significativamente menos atenção. Além disso, o papel do cuidado materno no estabelecimento destes desfechos também deve ser avaliado. O objetivo deste estudo foi investigar os efeitos da RCIU sobre a flexibilidade cognitiva dos animais na vida adulta, explorando o possível envolvimento do cuidado materno e da resposta neuroquímica à recompensa – através da medida do conteúdo de tirosina hidroxilase (TH) no córtex orbitofrontal (OFC) e núcleo accumbens ( NAcc ) – sobre este desfecho. Materiais e Métodos: A partir do dia 10 de gestação e durante toda a lactação, ratas Sprague-Dawley receberam uma dieta ad libitum (Contr), ou uma dieta restrita a 50% do consumo médio das ratas do grupo Contr (FR). No dia do nascimento, foi realizada a adoção cruzada, gerando os grupos Contr/Contr e FR/Contr (gestação/lactação). Do dia 2 ao dia 7 pós-parto, o cuidado materno foi registrado em cinco sessões diárias de 72 minutos cada. A flexibilidade cognitiva foi medida nos ratos na idade adulta utilizando o Attentional Set- Shifting Task (ASST) que utiliza um pellet de alimento doce como recompensa. Os animais também foram submetidos a jejum de 4 horas e em seguida expostos ao alimento doce durante 1 hora para verificar o consumo agudo de alimento palatável. O conteúdo de TH no córtex orbitofrontal e núcleo accumbens foi medido em jejum ou em resposta à ingestão de alimentos palatáveis. Resultados: Ratas FR mostraram menos comportamentos de lamber os filhotes que fêmeas do grupo Contr (p<0,01) sem diferença nos escores das posturas de amamentação e no tempo permanecido em contato com os filhotes. Filhotes de ratas FR apresentaram peso reduzido ao nascer (p<0,01). Aos 90 dias de idade, machos do grupo FR/Contr apresentaram maior consumo de alimento doce no teste de exposição aguda (p=0,04), enquanto que as fêmeas não diferiram na quantidade consumida (p=0,37). Quando comparadas às controles, fêmeas RCIU necessitaram menos trials para alcançar o critério na etapa de Reversão 2 do ASST (p=0,04), e apresentaram um aumento de TH no OFC em resposta à ingestão de alimentos doces (p=0,04). Não foram observadas diferenças para o sexo masculino em relação à performance no ASST ou nos níveis de TH no OFC. No NAcc , houve um aumento de TH no estado de jejum tanto nos machos RCIU (p=0,03) quanto nas fêmeas RCIU (p=0,02). Discussão: O protocolo utilizado foi eficiente em induzir RCIU nos filhotes. Caracterizar os cuidado materno em diferentes protocolos é crucial para entender melhor as relações entre ambientes precoces adversos e seus coincidentes desfechos adversos na vida adulta. A melhora no desempenho do ASST e concomitante aumento de TH no OFC após consumo de doce sugere que as pistas relacionadas aos alimentos palatáveis sejam mais fortes para as fêmeas expostas à RCIU. Além disso, as alterações encontradas no Nacc em ambos os sexos sugere que a RCIU induz modificações na resposta central para as pistas de alimentos palatáveis e no consumo, afetando a liberação de dopamina em algumas estruturas do circuito encefálico de recompensa. / Intrauterine growth restriction (IUGR) is associated with altered food preferences as well as increased risk for obesity in adult life. However, while the adverse metabolic outcomes have been better characterized in this condition, the neurobehavioral disabilities and particular abnormalities in specific areas of the brain have received less attention. Moreover, the role of maternal care in the stablishment of these outcomes should be evaluated. The aim of this study was to investigate the effects of IUGR on the cognitive flexibility of adult animals, exploring the putative involvement of maternal care and the neurochemical response to reward – through the measurement of tyrosine hydroxylase (TH) content in the orbitofrontal (OF) cortex and nucleus accumbens (nacc) - on this behavioral outcome. Materials and methods: From day 10 of gestation and through lactation, Sprague-Dawley rats received either an ad libitum (AdLib), or a 50% food restricted (FR) diet. At birth, pups were cross-fostered, generating AdLib/AdLib and FR/AdLib groups (pregnancy/lactation). From day 2 to 7 postpartum maternal care was recorded in a five daily sessions. Cognitive flexibility was measured using the Attentional Set-Shifting Task (ASST) with a sweet pellet as a reward. Animals were also submitted to 4 hours of fasting and then exposed to a sweet food for 1 hour to verify the palatable food consumption. More precisely, these FR dams showed less licking/grooming than AdLib dams (p<0.01), without difference in the nursing postures and time spent out of the nest. Pups of FR dams had reduced birth weight (p<0.01). At 90 days of age, FR/AdLib males showed increased intake of sweet food in the acute exposure (p=0.04), while females did not differ in the amount consumed (p=0.37). When compared to Controls, IUGR females needed fewer trials to reach criterion in the ASST(p=0.04) and had an increase in TH in response to sweet food intake in the OFC (p=0.04), but not at baseline. No differences were seen in males (p=0.51). In the nacc, there was an increase in TH at baseline in IUGR males (p=0.03) and females (p=0.02). Discussion: The protocol used was efficient in inducing IUGR in the pups. To characterize maternal care in different protocols is crucial to better understand the relationships between early adverse environment and their coincident adverse outcomes in adult life. The improvement in performance of FR females in ASST and concomitant increase in OFC TH in respone to sweet food consumption suggests that palatable food cues are stronger for intrauterine restricted females. Besides, alterations found in the Nacc in both sexes suggest that IUGR induces modifications in the central response to palatable food cues and its intake, affecting dopamine release in select structures of the brain reward system.

O conteúdo de TH no córtex orbitofrontal e núcleo accumbens foi medido em jejum ou em resposta à ingestão de alimentos palatáveis. Resultados: Ratas FR mostraram menos comportamentos de lamber os filhotes que fêmeas do grupo Contr (p<0,01) sem diferença nos escores das posturas de amamentação e no tempo permanecido em contato com os filhotes. Filhotes de ratas FR apresentaram peso reduzido ao nascer (p<0,01). Aos 90 dias de idade, machos do grupo FR/Contr apresentaram maior consumo de alimento doce no teste de exposição aguda (p=0,04), enquanto que as fêmeas não diferiram na quantidade consumida (p=0,37). Quando comparadas às controles, fêmeas RCIU necessitaram menos trials para alcançar o critério na etapa de Reversão 2 do ASST (p=0,04), e apresentaram um aumento de TH no OFC em resposta à ingestão de alimentos doces (p=0,04). Não foram observadas diferenças para o sexo masculino em relação à performance no ASST ou nos níveis de TH no OFC. No NAcc , houve um aumento de TH no estado de jejum tanto nos machos RCIU (p=0,03) quanto nas fêmeas RCIU (p=0,02). Discussão: O protocolo utilizado foi eficiente em induzir RCIU nos filhotes. Caracterizar os cuidado materno em diferentes protocolos é crucial para entender melhor as relações entre ambientes precoces adversos e seus coincidentes desfechos adversos na vida adulta. A melhora no desempenho do ASST e concomitante aumento de TH no OFC após consumo de doce sugere que as pistas relacionadas aos alimentos palatáveis sejam mais fortes para as fêmeas expostas à RCIU. Além disso, as alterações encontradas no Nacc em ambos os sexos sugere que a RCIU induz modificações na resposta central para as pistas de alimentos palatáveis e no consumo, afetando a liberação de dopamina em algumas estruturas do circuito encefálico de recompensa. / Intrauterine growth restriction (IUGR) is associated with altered food preferences as well as increased risk for obesity in adult life. However, while the adverse metabolic outcomes have been better characterized in this condition, the neurobehavioral disabilities and particular abnormalities in specific areas of the brain have received less attention. Moreover, the role of maternal care in the stablishment of these outcomes should be evaluated. The aim of this study was to investigate the effects of IUGR on the cognitive flexibility of adult animals, exploring the putative involvement of maternal care and the neurochemical response to reward – through the measurement of tyrosine hydroxylase (TH) content in the orbitofrontal (OF) cortex and nucleus accumbens (nacc) - on this behavioral outcome. Materials and methods: From day 10 of gestation and through lactation, Sprague-Dawley rats received either an ad libitum (AdLib), or a 50% food restricted (FR) diet. At birth, pups were cross-fostered, generating AdLib/AdLib and FR/AdLib groups (pregnancy/lactation). From day 2 to 7 postpartum maternal care was recorded in a five daily sessions. Cognitive flexibility was measured using the Attentional Set-Shifting Task (ASST) with a sweet pellet as a reward. Animals were also submitted to 4 hours of fasting and then exposed to a sweet food for 1 hour to verify the palatable food consumption. TH content in the OF cortex and Nacc was measured at baseline or in response to palatable food intake. Results: Dams of FR group showed less care toward her pups. More precisely, these FR dams showed less licking/grooming than AdLib dams (p<0.01), without difference in the nursing postures and time spent out of the nest. Pups of FR dams had reduced birth weight (p<0.01). At 90 days of age, FR/AdLib males showed increased intake of sweet food in the acute exposure (p=0.04), while females did not differ in the amount consumed (p=0.37). When compared to Controls, IUGR females needed fewer trials to reach criterion in the ASST(p=0.04) and had an increase in TH in response to sweet food intake in the OFC (p=0.04), but not at baseline. No differences were seen in males (p=0.51). In the nacc, there was an increase in TH at baseline in IUGR males (p=0.03) and females (p=0.02). Discussion: The protocol used was efficient in inducing IUGR in the pups. To characterize maternal care in different protocols is crucial to better understand the relationships between early adverse environment and their coincident adverse outcomes in adult life. The improvement in performance of FR females in ASST and concomitant increase in OFC TH in respone to sweet food consumption suggests that palatable food cues are stronger for intrauterine restricted females. Besides, alterations found in the Nacc in both sexes suggest that IUGR induces modifications in the central response to palatable food cues and its intake, affecting dopamine release in select structures of the brain reward system.

This thesis will be organized into two chapters discussing the role of hypoxia in the human placenta. The goal of this thesis is to characterize pyruvate kinase M2, mammalian target of rapamycin, mitochondrial function, and cell invasion in hypoxic conditions in the trophoblast. Understanding the mechanisms of placental metabolism can lead to further treatments for placental diseases. Chapter one covers the background of intrauterine growth restriction, hypoxia, placental metabolism, and pyruvate kinase M2 (PKM2). Little is currently understood about the role of the mitochondria in placental diseases. Expression of PKM2, trophoblast cell invasion, and mitochondrial function is shown to be inhibited by hypoxia. PKM2 inhibition decreases trophoblast cell invasion and nuclear expression of PKM2, but increases mitochondrial function. Studying how hypoxia affects the placenta during placental diseases can help clarify the mechanisms by which these diseases occur. Chapter two further characterizes the background of intrauterine growth restriction and hypoxia. It also covers the background of mammalian target of rapamycin. The objective of this chapter was to assess activated mTOR in the trophoblast in hypoxia. Decreased placental and fetal weights, as well as trophoblast cell invasion were observed in hypoxia. A decrease in the activation of mTOR was also found in the hypoxic placenta. This study could provide insight into the physiological relevance of the pathways and could be targeted to help alleviate placental diseases.

Quantification de la vascularisation de l'unité utéro-placentaire par Doppler 3D : évaluation clinique et expérimentale / Quantification of utero-placental unit vascularization by 3D Doppler : Clinical and experimental evaluation

Les pathologies vasculaires placentaires (PVP), prééclampsie (PE) et retard de croissance intra-utérin (RCIU), sont une des causes majeures de morbidité et mortalité maternelles et périnatales dans les pays développés. Un défaut de vascularisation utéro-placentaire est présent dans ces deux pathologies. Le Doppler énergie 3D (3DPD) est un outil facile à utiliser en clinique, les indices quantitatifs calculés à partir de volume utéro-placentaire semblent prometteurs pour dépistage de la PE. Le but principal de cette thèse est d’évaluer cette technique sur le plan de sa valeur discriminative entre situation physiologique et pathologique, ainsi que sur le plan de ses limites techniques en clinique et sur modèles animaux. La performance des indices 3DPD au 1er trimestre pour dépister la PE et/ou RCIU a été étudiée dans une étude prospective multicentrique. Une évaluation de la corrélation entre les indices 3DPD et les indices morphométrique a été ensuite réalisée chez les femmes enceintes à la fin de grossesse. Une corrélation entre le phénotype des villosités placentaire et la qualité de la perfusion placentaire a été mise en évidence. À la fin, l’intérêt de la technique a été démontré dans un modèle de RCIU chez brebis gestante. / Placental vascular pathologies (PVP), preeclampsia (PE) and intra uterine growth restriction (IUGR), are a major cause of maternal and perinatal morbidity and mortality in developed countries. A default of utero-placental vascularization is present in these two pathologies. 3D Power Doppler (3DPD) is an easy tool to use in clinic, its quantitative indices calculated from obtained utero-placental volume seem promising for PE prediction. The major objective of this thesis was to evaluate the discriminative value of this technique between physiological and pathological situations, as well as its limitations in clinical and animal models. The screening performance of PE and/or IUGR by 3DPD indices in the first trimester was studied in a prospective multicenter study. An evaluation of the correlation between 3DPD indices and morphometric indices was then carried out among pregnant women at term. At the end, the interest of the technique was demonstrated in an ewe model of IUGR (treated with testosterone).

Prädiktive Wertigkeit von Parametern des oxidativen Status bei Präeklampsie und intrauteriner Wachstumsretardierung

Eine Dysbalance zwischen Sauerstoffradikalen und der plasmatischen antioxidativen Kapazität sowie eine gesteigerte Aktivität der neutrophilen Granulozyten werden als mögliche pathophysiologische Faktoren der Schwangerschaftserkrankungen Präeklampsie (PE) und intrauterine Wachstumsretardierung (IUGR) diskutiert. Das Ziel der Studie war, zu prüfen, ob der plasmatische antioxidative Status und die Plasmaspiegel der neutrophilen Granulozyten Myeloperoxidase und PMN Elastase bei Schwangerschaften mit uteriner Perfusionsstörung, die ein Hochrisikokollektiv für die oben genannten Schwangerschaftskomplikationen darstellen, verändert sind. Die Messungen mit dem immundiagnostischen ELISA zur Bestimmung der antioxidativen Kapazität (ImAnOx) ergaben, dass eine signifikante Erniedrigung der Totalen Antioxidativen Kapazität im Hochrisikokollektiv vorlag. Eine Verbesserung der Risikobeurteilung war durch den ImAnOx-Test nicht möglich. Die Berechnung des Antioxidativen Quotienten durch die zusätzliche Bestimmung des Harnsäurespiegels konnte die Voraussage nicht verbessern. Zur inflammatorischen Komponente mit Hilfe der Enzymspiegelmessungen zeigten die Ergebnisse weder einen Zusammenhang mit der uterinen Perfusionsstörung noch verbesserten sie die Einschätzung des Risikos. Anhand der Ergebnisse wird deutlich, dass mit der uterinen Dopplersonographie insbesondere für das fetale Geburtsgewicht eine Kalkulation möglich ist.

Its theoretical part summarizes the specifics of the development of these individuals and defines the problems of terminology and classification. It deals with the risks associated with early postnatal periods and long-term complications with metabolic and neurological impacts. The target of this study was to find out whether motor disorders are more common in children with a history of SGA (IUGR) than those of general population. The test group and the general population group underwent kinesiological examination by aspects and a battery of MABC-2 tests. This hypothesis has not been proven. In addition, the thesis aimed to evaluate the possible link between the motor manifestation at the early age and the age of two and three years.
